The preparation and molecular and crystal structure of the complex [(ethylenediamine)bis(7,9,-dimethylhypoxanthine)platinum(II)] hexafluorophosphate, [Pt(C2H8N2)(C7H8N4O)2] (PF6)2, are reported. The complex crystallizes in the monoclinic system, space group C2/c, with a = 12.334(2)Å, b = 10.256(2)Å, c = 22.339(3)Å, β = 101.31(1)°, V = 2771.0Å3, Z = 4, Dmeasd = 2.087(3) g cm?3, Dcalc = 2.094 g cm?3. Intensities for 3992 symmetry-averaged reflections were collected in the θ-2o scan mode on an automated diffractometer employing graphite-monochromatized MoKα radiation. The structure was solved by standard heavy-atom Patterson and Fourier methods. Full matrix least-squares refinement led to a final R value of 0.051. Both the ethylenediamine chelate and the PF6? anion are disordered. The primary coordination sphere about the Pt(II) center is approximately square planar with the bidentate ethylenediamine ligand and the N(1) atoms [Pt(II) ? N(1) = 2.020(5)Å] of two 7,9-dimethylhypoxanthine bases (related by a crystallographic twofold axis of symmetry) occupying the four coordination sites. The exocyclic O(6) carbonyl oxygen atoms of the two 7,9-dimethylhypoxanthine ligands participate in intracomplex hydrogen bonding with the amino groups of the ethylenediamine chelate [N(ethylenediamine) ? O(6) = 2.89( )Å]. The observed Pt ? O(6) intramolecular distances of 3.074(6)Å are similar to those found in other Pt(II) N(1)-bound 6-oxopurine complexes and in several Pt(II) N(3)-bound cytosine systems.  相似文献

Reaction of platinum(II) salts with 5-ferrocenylpyrimidine (FcPM) afforded cis-[Pt(NH3)2(FcPM)2](PF6)2 (1), trans-[Pt(NH3)2(FcPM)2](PF6)2 (2), cis-[PtCl2(FcPM)2] (3), and cis-[PtCl2(DMSO)(FcPM)] (4): their spectroscopic and electrochemical properties were investigated. A variety of Group 6 mono bipyridine (bpy) complexes were prepared, and substitution reactions of [(bpy)(MeIm)M(CO)2(NO)]PF6 complexes (MeIm = 1-methylimidazole, M = W or Mo) were investigated. Nitrosylation of complexes having the general formula (bpy)(L)M(CO)3 (L = a variable ligand) gave cationic complexes of the form [(bpy)(L)M(CO)2(NO)]PF6. The structure of [(bpy)(MeIm)W(CO)2(NO)]PF6 was confirmed by single-crystal X-ray diffractometry. [(bpy)(MeIm)M(CO)2(NO)]PF6 complexes undergo facile substitutions with mono-, tri- and tetra-dentate ligands, yielding di- or mono-carbonyl mononitrosyl complexes. The structures of [(bpy)(PMe3)2W(CO)(NO)]PF6 and [(dien)(PMe3)W(CO)(NO)]PF6 (dien = diethylenetriamine) were determined by X-ray diffraction.  相似文献

From the reaction between dihydroxoplatinum(II) and l-ascorbic acid, two types of platinum(II) ascorbate complexes were obtained and structurally characterized with ethylenediamine (en), N,N-dimethylethylenediamine (dmen) and N,N,N′-trimethylethylenediamine (trimen) as stabilizing ligands. In [Pt(en)(asc-C,O)] (1), [Pt(dmen)(asc-C,O)] (2) and [Pt(trimen)(asc-C,O)] (4), the ascorbate dianion forms a five-membered chelate ring, coordinating to the Pt(II) ion at the 2-carbon and the 5-oxygen atoms (C,O-chelate). From the same mother solution, crystals of [Pt(trimen)(asc-O,O′)] (3) were obtained during the precipitation of 4; in 3 the ascorbate is bound to the Pt at the 2- and 3-oxygen atoms (O,O′-chelate). Compounds 3 and 4 are the first well-characterized linkage isomers among the transition-metal ascorbate complexes. The O,O′-chelated 3 slowly changes to the C,O-chelated 4 in an aqueous solution. Bulkiness of the stabilizing ligand, i.e. en, dmen and trimen has an influence on the formation of the C,O-chelated species, 1, 2 and 4.  相似文献

A complex containing a protonated and N3-platinated cytosine (C), [CH][Cl3Pt(C)] (1a) has been prepared, converted into its K[Cl3Pt(C)] (1b) and NH4[Cl3]Pt(C)]·H2O (1c) analogs, and structurally characterized (X-ray, Raman, NMR). Reaction of 1b with L = 1-methylcytosine and with L = Me2SO gave the neutral mixed-ligand complexes cis-Cl2Pt(C)L. Excess NH3 was used to convert the anion of 1b into the cation [(NH3)3Pt(C)]2+ (3a). The pKa of the N(1)H proton in 3a is 9.4, as determined by UV spectroscopy. The N(1)H is displaced by Pt(II) electrophiles even at neutral pH to give N3,N1-diplatinated cytosinato complexes, as shown by 1H NMR (3J coupling or 195Pt at N(1) with H6, 29 Hz, and 4J coupling of 195Pt at N(3) with H5, 14Hz). The results of the X-ray structure determination of 1a (R = 0.031, Rw = 0.034) are of relevance in that they permit a direct comparison of the effect of a proton as opposed to that of a Pt electrophile on the nucleobase geometry. Moreover, the expected decrease in CO(2) bond length as a consequence of Pt binding is observed.  相似文献

Synthesis of the heterobimetallic platinum(II)-palladium(II) complexes with poly fluorinated benzenethiolates as intermetallic bridges, [(dppe)Pd(μ-SRF)2Pt(dppe)](SO3CF3)2 with SRp-SC6F4(CF3) (1), SC6F5 (2), p-SC6HF4 (3) and o-SC6H4(CF3) (4), have been accomplished either by a redistribution reaction in mixtures of the homonuclear bimetallic species, [(dppe)Pd(μ-SRF)2Pd(dppe)]2+/[(dppe)Pt(μ-SRF)2Pt(dppe)]2+ or by assembling the monometallic building blocks [(dppe)M(μ-SRF)2]/[(dppe)M′(solvent)2]2+, M, M′ = Pd or Pt. Both experimental systems reach an equilibrium state which is independent of the temperature within the probed range, −90 °C to +50 °C. A single crystal of the heterobimetallic compound [(dppe)Pd(μ-SC6F5)2Pt(dppe)](SO3CF3)2(acetone)2 (2) was isolated and analyzed by X-ray diffraction. Comparison with the corresponding structures exhibited by the homobimetallic analogous, [Pd2(μ-SC6F5)2(dppe)2](SO3CF3)2(acetone)2 (5) and [Pt2(μ-SC6F5)2(dppe)2](SO3CF3)2(acetone)2 (6) shows that all three structures are isostructural in space group . All three compounds exhibit a centrosymmetric planar [M2(μ-S)2] ring in which the sulfur substituents are arranged in an anti configuration.  相似文献

A series of water-soluble N-substituted iminodiacetato (diammine)platinum(II) complexes [Pt(NRIDA)(NH3)2] have been synthesized and characterized by measurement of physical properties (conductivity and pH) and by various spectroscopic techniques (infrared, 1H and 13C{1H} nuclear magnetic resonance). The iminodiacetate ligand is coordinated to platinum through an O,N linkage. The results obtained suggest that these complexes are relatively stable for more than 24 h in aqueous solution. Preliminary in vitro and in vivo screening test for antitumor activity of these complexes against L1210 murine leukemia were performed. Many of complexes had acceptable in vitro cytotoxicity, but none displayed a significant level of in vivo antitumor efficacy.  相似文献

 The interaction of the new antitumor-active platinum organoamide complexes [Pt{N(p–HC6F4)CH2}2(py)2] and [Pt{N(C6F5)CH2}2(py)2] (py = pyridine) with small G-containing (oligo)nucleotides [GMP, d(GpG)] has been studied to establish whether or not these compounds can bind to DNA in an analogous manner to cisplatin. The reaction products have been analyzed by 1H, 19F and 31P NMR spectroscopy. From the NMR data it is concluded that the {Pt(py)2}2+ moiety binds to the N7 position of the G base, analogously to cisplatin, with the organoamide ligand acting as the leaving group. For the GG-N7,N7 adduct, structural differences are found for the sugar conformation, compared with cisplatin. These differences may account for the activity of these new compounds in tumor cell lines resistant to cisplatin. Received: 25 September 1995 / Accepted: 7 March 1996  相似文献

Two new heterobimetallic complexes of rhenium(I) and ruthenium(II) [(CO)3(NN)Re(4,4′-bpy)Ru(NN)2Cl](PF6)2 and already known monometallic complexes [Cl(NN)2Ru(4,4′-bpy)](PF6) and [(CO)3(NN)Re(4,4′-bpy)](PF6) and bimetallic complexes [Cl(NN)2Ru(4,4′-bpy)Ru(NN)2Cl](PF6)2, [(CO)3(NN)Re(4,4′-bpy)Re(NN)(CO)3](PF6)2 (NN = 2,2′-bipyridine, 1,10-phenanthroline; 4,4′-bpy = 4,4′-bipyridine) are synthesized and characterized by spectral techniques. The photophysical properties of all the complexes are studied. It is found that attachment of rhenium(I) altered the photophysical characteristics of ruthenium(II). Excited state energy transfer from the rhenium(I) chromophore to the ruthenium(II) is observed upon excitation at 355 nm.  相似文献

A variety of platinum(II) complexes of methimazole (2-mercapto-1-methylimidazole; HImS = neutral form and ImS = thiolate form), coordinated in both thione and thiolate forms, have been isolated by reacting methimazole with [PtCl(terpy)]Cl (terpy = 2,2′:6′,2″ terpyridine), [PtCl2(bipy)] (bipy = bipyridine), [PtCl2(o-phen)] (o-phen = o-phenanthroline), [PtCl2(CH3CN)2] and [PtCl2(COD)] (COD = 1,5-cyclooctadiene). These complexes were characterized by electronic absorption, IR and NMR (1H, 13C, 195Pt) spectroscopies. Molecular structure of [Pt(bipy)(HImS)2]Cl2·3H2O (3a·3H2O) has been established by single crystal X-ray crystallography. Platinum thiolate complex, [Pt(ImS)2(HImS)2] (5), could be obtained by treatment of [Pt(HImS)4]Cl2 with sodium methoxide in methanol. The solution of 5 in organic solvents yielded bi- and tri-nuclear platinum complexes. The effect of diimine ligands on oxidation of methimazole moiety in the complexes has been studied by electrochemical oxidation and pulse radiolytic oxidation employing specific one-electron oxidant, radical.  相似文献

Reported herein are studies of the concentration and temperature dependent interactions with DNA of the stereochemically defined mixed-metal supramolecular complexes, [(tpy)Ru(tppz)PtCl](PF6)3 and [ClPt(tppz)Ru(tppz)PtCl](PF6)4 (tpy = 2,2′:6′,2′′-terpyridine and tppz = 2,3,5,6-tetrakis(2-pyridyl)pyrazine). These metal complexes couple a ruthenium based light absorber (LA) to the bioactive platinum sites (BAS) using a tridentate bridging ligand (BL). The complexes exhibit intense Ru → tppz(π∗) metal to ligand charge transfer (MLCT) transitions in the visible region and adopt a square planar geometry around the Pt(II) center. The effect of incubating these metal complexes with DNA on the subsequent migration of DNA through an agarose gel was found to be more dramatic than that observed for the well known anticancer drug, cis-[Pt(NH3)2Cl2] (cisplatin). This effect was enhanced with increased incubation temperature. Unwinding of supercoiled plasmid DNA was found to be more pronounced for the trimetallic complex, [ClPt(tppz)Ru(tppz)PtCl](PF6)4, than for the bimetallic complex, [(tpy)Ru(tppz)PtCl](PF6)3.  相似文献

Two dinuclear palladium(II) complexes, [{Pd(en)Cl}2(μ-pz)](NO3)2 and [{Pd(en)Cl}2(μ-pydz)](NO3)2, have been synthesized and characterized by elemental microanalysis and spectroscopic (1H and 13C NMR, IR and UV–vis) techniques (en is ethylenediamine; pz is pyrazine and pydz is pyridazine). The square planar geometry of palladium(II) metal centers in these complexes has been predicted by DFT calculations. The chlorido complexes were converted into the corresponding aqua complexes, [{Pd(en)(H2O)}2(μ-pz)]4+ and [{Pd(en)(H2O)}2(μ-pydz)]4+, and their reactions with N-acetylated l-histidylglycine (Ac–l–His–Gly) and l-methionylglycine (Ac–l–Met–Gly) were studied by 1H NMR spectroscopy. The palladium(II)-aqua complexes and dipeptides were reacted in 1:1 M ratio, and all reactions performed in the pH range 2.0 < pH < 2.5 in D2O solvent and at 37 °C. In the reactions of these complexes with Ac–l–His–Gly and Ac–l–Met–Gly dipeptides, the hydrolysis of the amide bonds involving the carboxylic group of both histidine and methionine amino acids occurs. The catalytic activities of the palladium(II)-aqua complexes were compared with those previously reported in the literature for the analogues platinum(II)-aqua complexes, [{Pt(en)(H2O)}2(μ-pz)]4+ and [{Pt(en)(H2O)}2(μ-pydz)]4+.  相似文献

Using the semi-empirical all-valence method (GRINDOL) (recently modified and extended to transition series elements), electronic structure and intermolecular interactions of the model antitumor Pt(II) compounds with guanine and thioguanine have been calculated. Several possible models of antitumor action of platinum compounds are discussed. It is concluded that cis-Pt(II) complexes with guanine form stable intrastrand N7N7 cross-links (but chelation to the O6 atom is also possible). The trans-isomers of platinum(II) exclusively form interstrand cross-links, but the cis-Pt(II) complexes with thioguanine form almost entirely the N7S five-membered chelates.  相似文献

 The compound [Pt(lysine)Cl2] (Kplatin) was previously identified in a study of platinum amino acid complexes as a potential antitumor drug candidate. The DNA binding properties, high mobility group (HMG)-domain protein affinity for the platinated DNA, and cytotoxicity against HeLa cells of Kplatin and three related (N,O) chelated platinum(II) amino acid complexes, [Pt(arginine)Cl2] (Rplatin), K[Pt(Ne-acetyllysine)Cl2] (NacKplatin), and K[Pt(norleucine)Cl2] (Norplatin), are reported. The four complexes have identical PtCl2(N,O) coordination environments. A new solid phase screening methodology was devised in which platinated DNA probes are covalently attached to a nylon support and tested for their ability to bind a fluorescently labeled HMG-domain protein. The fluorescent HMG-domain protein was generated by expressing a fusion of the green fluorescent protein (GFP) with recombinant rat HMG1. Binding revealed by the solid phase method correlated well with the results of gel mobility shift and HeLa cytotoxicity assays. These results suggest that the net charge on the complex, rather than the nature of the side chain, is the most important factor underlying the DNA binding properties and toxicity of amino acid (N,O) chelated platinum complexes. This property explains why Kplatin was previously selected from the pool of platinum amino acid complexes based on the ability of its DNA adducts to bind HMG1. Ring-substituted diaqua(1,2-diphenylethylenediamine)platinum(II) sulfate shows unusual kinetics in its reaction with salmon testis DNA. The mechanism for diaqua[meso-1,2-bis(2,6-dichloro-4- hydroxyphenyl)ethylenediamine]platinum(II) sulfate, [Pt(H2O)2(meso-6)]2+SO4(2-), a representative of this series, has been investigated and compared with that for cis-[Pt(NH3)2(H2O)2]2+. Reactions were followed by atomic absorption, analytical HPLC of Pt-DNA digests, arrest of enzymatic DNA synthesis/degradation, ultraviolet and fluorescence spectrophotometry. Except for the formation of monofunctional DNA adducts, the kinetics of the platinum(II) complexes are comparable. The pseudo-first-order rate constant for the attack of DNA by [Pt(H2O)2(meso-6)]2+ follows the concentration of DNA in a hyperbolic fashion, which is in contrast to the linear dependence for cis-[Pt(NH3)2(H2O)2]2+. The hyperbolic dependence is typical for a dissociable DNA/drug complex preceding the coordination reaction. By studying the binding of free ligand to DNA, and by correlating ligand structures and electrostatic charges with effects on adduct formation, both the phenyl residues and the positive charge of the platinum(II) complex are shown to be crucial for the stability of the dissociable complex. A non-intercalative mode of binding to the DNA backbone is suggested. At the high concentrations of DNA found in cell nuclei, the reaction of the dissociable complex can, principally, become rate-limiting in the attack of DNA and thus reduce the cytotoxic efficiency of a drug.  相似文献

Three new complexes [Pt(dpop)(Cl)2], [(Cl)2Pt(dpop)Pt(Cl)2] and [(bpy)2Ru(dpop)Pt(Cl)2](PF6)2 (dpop = dipyrido(2,3-a:3′,2′-h)phenazine) were prepared and studied. The electronic absorption spectra of the complexes display Pt dπ → dpop π* and Ru dπ → dpop π* MLCT transitions at longer wavelengths than for previously reported similar complexes. Results of cyclic voltammograms show reversible dpop centered reductions while for the mixed metal [(bpy)2Ru(dpop)Pt(Cl)2]2+ an irreversible Pt(II) oxidative wave precedes the Ru(II) oxidation/reduction couple. Spectroelectrochemical results show that all oxidative and reductive processes are completely reversible. The [(Cl)2Pt(dpop)Pt(Cl)2] complex cleaves in solution with pseudo-first order kinetics resulting in loss of the Pt dπ → dpop π* MLCT transition at 545 nm.  相似文献

The reactions of the platinum(II) complexes, [Pt(dien)(H(2)O)](2+), [PtCl(dien)](+) and [PtBr(dien)](+) (dien is diethylenetriamine) with some biologically relevant ligands such as inosine (INO), inosine-5'-monophosphate (5'-IMP), guanosine-5'-monophosphate (5'-GMP), glutathione (GSH) and l-methionine (S-meth), have been studied by UV-Visible spectrophotometry and (1)H NMR spectroscopy. Kinetic and thermodynamic parameters of these reactions were determined. Competitive reactions of [PtCl(dien)](+) with l-methionine and 5'-GMP demonstrated initially rapid formation of [Pt(dien)(S-meth)](2+) followed by displacement of l-methionine by 5'-GMP. In the later stages the concentration of [Pt(dien)(N7-GMP)](2+) is predominant. The results are analyzed in reference to the anti-tumour activity of Pt(II) complexes.  相似文献

The products obtained from the reaction of Pt(IV)Cl4(LL) compounds (LL denotes the chelating ligands ethylenediamine (en) and 2,2-dimethyl-1,3-diaminopropane (dmdap), or two cis- or trans-coordinated ammines) with 9-methylhypoxanthine (mHyp) at high temperature (80°C) have been characterized by proton NMR spectroscopy. It appeared that both platinum(II) and platinum(IV) adducts were present in the reaction mixtures. After cation-exchange chromatography, the Pt(II) compound could be characterized as Pt(II)(LL)(mHyp)2, whereas the Pt(TV) fractions appeared to contain mainly one or two adducts for the chelating diamine compound but more adducts for the ammine compounds. A 3J(195Pt-1H) coupling was observed for the Pt(IV), but not for the Pt(II) compounds at the used spectrometer frequency. This supplies a useful tool to discriminate between these two types of platinum adducts.  相似文献

A series of flexible multidentate ligands containing N,P-donor, 2-[N-(diphenylphosphino)methyl]amino-pyridine (L1), 2-[N-bi-(diphenylphosphino) methyl]amino-pyridine (L2), 2-[N-(diphenylphosphino)methyl]amino-7-methyl-1,8-naphthyridine (L3) and 4-[(N-diphenylphosphino)methyl]amino-pyridine) (L4) have been synthesized. The mono- and dinuclear cyclometalated platinum(II) complexes [Pt(C^N^N)L1]ClO4 (HC^N^N = 6-phenyl-2,2′-bipyridine), [Pt2(C^N^N)2L1](ClO4)2, [Pt2(C^N^N)2L2](ClO4)2, [Pt(C^N^N)L3]ClO4 and [Pt2(C^N^N)2L4](ClO4)2 were prepared and their structures determined by X-ray crystal analysis. These complexes exhibit long-lived bright orange emissions ranging from 560 to 610 nm in the solid state at room temperature. In solution, dinuclear complexes have emissions with higher quantum yields than mononuclear complexes. This can be attributed to intramolecular interaction of free functional group with Pt(II) at axial position, resulting in the quenching of phosphorescence for platinum(II) complexes in the 3MLCT excited state.  相似文献

The preparation is reported of [(NH3)3Pt(9- MeA)] X2 (9-MeA = 9-methyladenine) with XCl (1a) and XClO4 (1b) and of trans-[(OH)2Pt(NH3)3- (9-MeA)]X2 with XCl (2a) and XClO4 (2b), and the crystal structure of 1b. [(NH3)3Pt(C6H7N5)](ClO4)2 crystallizes in space group P21/n with a = 20.810(7) Å, b = 7.697(3) Å, c = 10.567(4) Å, β = 91.57(6)°, Z = 4. The structure was refined to R = 0.054, Rw = 0.063. In all four compounds Pt coordination is through N7 of 9-MeA, as is evident from 3J coupling between H8 of the adenine ring and 195Pt. Pt(II) and Pt(IV) complexes can be differentiated on the basis of different 3J values, larger for Pt(II) than for Pt(IV) by a factor of 1.57 (av). In Me2SO-d6, hydrogen bonding occurs between Cl? and C(8)H of 9-MeA as weil as between Cl? and the NH3 groups in the case of the Pt(II) complex 1a. Protonation of the 9-MeA ligands was followed using 1H NMR spectroscopy and pKa values for the N1 protonated 9-MeA ligands were determined in D2O. They are 1.9 for 1a and 1.8 for 2a, which compares with 4.5 for the non-platinated 9-MeA. Possible consequences for hydrogen bonding with the complementary bases thymine or uracil are discussed briefly. Protonation of the OH groups in the Pt(IV) complexes has been shown not to occur above pH 1.  相似文献
