Four German isolates (FS1, SR2, SAW1 and DEG2) of Bremia lactucae originating from lettuce cultivars with R‐factors R18 and Dm6 + R36 were used for detailed characterization of virulence factors (v‐factors) and for the study of the resistance efficiency in wild Lactuca spp. germplasm. The highest complexity of v‐phenotype was recognized in isolate DEG2, which overcomes resistance in cv. Mariska (R18) and line CS‐RL (L. serriola × L. sativa, R18 + ?), until now known as resistant to all known races of B. lactucae in Europe. However, some sparse sporulation also occurred on cv. Titan (Dm6 + R36). The isolates SR2 and SAW1 overcome the resistance based on the gene R36, but are avirulent to R18. The v‐phenotype of SR2 is highly complex with the most important v‐factors being present except for v14 and v18. The isolate FS1 is the first in Germany originating from a cultivar with R18 (cv. Samourai). The search for efficient sources of resistance in 64 accessions of 11 wild Lactuca spp. and primitive forms of L. sativa showed broad variation in accession–isolate interactions. Expression of race‐specific resistance in wild Lactuca spp. (L. serriola, L. viminea, L. virosa) was recorded frequently. Lactuca indica and L. saligna could be considered as the most efficient sources of resistance against isolates FS1, SR2 and SAW1. The isolate DEG2 showed the highest level of virulence. On seedlings of L. saligna, which is considered as a most important source of resistance against B. lactucae, there was frequently recorded limited sporulation, however this response cannot be considered as a susceptible. Except for some L. saligna accessions (CGN 05310 and CGN 05315), the resistance to all studied isolates was only observed in one accession of L. serriola (PI 253467).  相似文献

In the period 1996–2001 the natural occurrence of Bremia lactucae (lettuce downy mildew) on Asteraceae plants was studied in the Czech Republic. Lactuca serriola (prickly lettuce) is the most common naturally growing host species of B. lactucae. Infection of plants was recorded during the whole vegetation season with the first occurrence in April and last in November. Bremia lactucae was found on host plants in all developmental stages. High percentages of naturally infected populations of L. serriola were recorded. Host plants exhibited broad variation in phenotypic expression of disease symptoms and degree of infection, however, the intensity of infection was rather low in the majority of populations. Geographic distribution of B. lactucae was studied in the two main parts of Czech Republic, central and southern Moravia, and eastern, northern and central Bohemia. Bremia lactucae was recorded in all these areas. Nevertheless, in the warmest parts of the Czech Republic (southern Moravia) only sporadic occurrence of the pathogen was recorded. Bremia lactucae infection on L. serriola and disease severity was judged also in relation to the type of habitat, and the size and density of host plant populations. However, no substantial differences among various habitats were found; only host plants growing in urban areas were frequently free of infection and the degree of infection was very low. Nevertheless, these plants were commonly infected with powdery mildew (Golovinomyces cichoracearum), which is most aggressive pathogen of this type of habitat.  相似文献

The use of surfactin and mycosubtilin as an eco-friendly alternative to control lettuce downy mildew caused by the obligate pathogen Bremia lactucae was investigated. Preliminary ecotoxicity evaluations obtained from three different tests revealed the rather low toxicity of these lipopeptides separately or in combination. The EC50 (concentration estimated to cause a 50 % response by the exposed test organisms) was about 100 mg L?1 in Microtox assays and 6 mg L?1 in Daphnia magna immobilization tests for mycosubtilin and 125 mg L?1 and 25 mg L?1 for surfactin, respectively. The toxicity of the mixture mycosubtilin/surfactin (1:1, w/w) was close to that obtained with mycosubtilin alone. In addition, the very low phytotoxic effect of these lipopeptides has been observed on germination and root growth of garden cress Lepidium sativum L. While a surfactin treatment did not influence the development of B. lactucae on lettuce plantlets, treatment with 100 mg L?1 of mycosubtilin produced about seven times more healthy plantlets than the control samples, indicating that mycosubtilin strongly reduced the development of B. lactucae. The mixture mycosubtilin/surfactin (50:50 mg L?1) gave the same result on B. lactucae development as 100 mg L?1 of mycosubtilin. The results of ecotoxicity as well as those obtained in biocontrol experiments indicated that the presence of surfactin enhances the biological activities of mycosubtilin. Mycosubtilin and surfactin were thus found to be efficient compounds against lettuce downy mildew, with low toxicity compared to the toxicity values of chemical pesticides. This is the first time that Bacillus lipopeptides have been tested in vivo against an obligate pathogen and that ecotoxic values have been given for surfactin and mycosubtilin.  相似文献

Nigrosporin A and B, two new phytotoxic and antibacterial metabolites were isolated from a culture filtrate of Nigrospora oryzae.

The active principles were absorbed on XAD-2 resin and purified by successive ODS-HPLC. The structures were identified by spectroscopic and derivatization analysis as naphthoquinone derivatives. The substances showed phytotoxic activities, such as root elongation inhibition, necrotic effects, oxygen evolution inhibition, starch synthesis inhibition, and CO2 fixation inhibition at concentrations of 10–100 ppm. They also showed growth inhibition activity against Bacillus subtilis in a disc diffusion assay as well as when compared with streptomycin.  相似文献

The systemic activity of thirty-three N-substituted S-esters derived from dithiocarbamic acid was investigated by assessing their ability to reduce the infection of broad-bean seedlings by Botrytis fabae and of wheat seedlings by Erysiphe graminis, following application to the roots or cut shoots of the host. Marked systemic activity against mildews was shown by the N-carboxymethyl dithiocarbamates, by S-carboxymethyl-N, N-dimethyl-dithiocarbamate (G33) and by procaine and 6-azauracil. The effect was not very host-specific since most compounds showing high activity in wheat also showed activity in pea, cucumber and, to a smaller extent, apple. All the materials tested showed a much lower level of systemic activity in broad-bean seedlings against B. fabae. Measurements were also made of the uptake, translocation, phytotoxicity and the stability within plant tissues of some of these compounds. The degree of systemic activity which they show is discussed in relation to these and other properties of the compounds.  相似文献

Oxazolomycin diacetate, dipropionate, monobutyrate and dibutyrate were derived from oxazolomycin, a product of Streptomyces sp. KBFP-2025. These esters were potent inhibitors of crown gall formation on potato tuber disks upon infection with Agrobacterium tumefaciens. They showed neither antibacterial nor phytotoxic activity, whereas oxazolomycin showed both antibacterial and phytotoxic activities. Further, they had no inhibitory activity against A. tumefaciens on the potato tuber disk. The inhibitory activity of these esters against crown gall formation seems to be due to specific inhibition of the transformation of plants with A. tumefaciens.  相似文献

The results of the first detailed screening of a resistance to Bremia lactucae in naturally growing populations of Lactuca saligna are presented here. In total, 146 accessions from 25 populations of L. saligna originating in Israel (N = 136), France (N = 8), Jordan (N = 1) and Turkey (N = 1) were tested at seedling stage for their resistance to 10 highly virulent isolates (races) of B. lactucae from Lactuca sativa (DEG2, Bl:5, Bl:15, Bl:16, Bl:17, Bl:18, Bl:21, Bl:22, Bl:24 and Bl:25). Our study strongly supports the suggestion that L. saligna is indeed generally highly resistant to B. lactucae. However, our results provide evidence that at least at a seedling stage L. saligna may not be a non‐host plant for B. lactucae, as was hypothesised for approximately the last 30 years. Some accessions expressed a differential (i.e. race‐specific) response, which accords with other recently published data for this Lactuca species. Furthermore, some geographical differences in race‐specific resistance were observed, too. Tests performed at an adult‐plant stage, however, did not prove race‐specificity of the respective accessions. To summarise, what is behind the race‐specific character of the responses observed at a seedling stage is still uncertain, as is its comparability with the race‐specific resistance of some other Lactuca species such as L. sativa or L. serriola. The presence of plant stage‐dependent resistance, governed by a combined effect of different quantitative trait loci in young and adult plants of L. saligna, is discussed.  相似文献

Bremia lactucae Regel (Chromista, Peronosporaceae) is an economically destructive pathogen, which causes downy mildew disease on lettuce (Lactuca sativa L.) worldwide. The ribosomal internal transcribed spacer (ITS) of Bremia lactucae isolates was analyzed for the first time. The ITS region of lettuce downy mildew was observed to have a size of 2458 bp; thereby, having one of the longest ITS sizes recorded to date. The majority of the extremely large sized ITS2 length of 2086 was attributed to the additional presences of nine repetitive elements with lengths of 179–194 bp, which between them shared the low homology of 48–69%. Comparison of the ITS2 sequences with the B. lactucae isolates from other host plants showed that isolates present on Lactuca sativa were distinct from those on L. indica var. laciniata, as well as Hemistepta and Youngia. We suggest the high degree of sequence heterogeneity exhibited in the ITS2 region of B. lactucae may warrant the specific detection and diagnosis of this destructive pathogen or its division into several distinct species.  相似文献

Host‐induced gene silencing (HIGS) is an RNA interference‐based approach in which small interfering RNAs (siRNAs) are produced in the host plant and subsequently move into the pathogen to silence pathogen genes. As a proof‐of‐concept, we generated stable transgenic lettuce plants expressing siRNAs targeting potentially vital genes of Bremia lactucae, a biotrophic oomycete that causes downy mildew, the most important disease of lettuce worldwide. Transgenic plants, expressing inverted repeats of fragments of either the Highly Abundant Message #34 (HAM34) or Cellulose Synthase (CES1) genes of B. lactucae, specifically suppressed expression of these genes, resulting in greatly reduced growth and inhibition of sporulation of B. lactucae. This demonstrates that HIGS can provide effective control of B. lactucae in lettuce; such control does not rely on ephemeral resistance conferred by major resistance genes and therefore offers new opportunities for durable control of diverse diseases in numerous crops.  相似文献

Two novel ligand?metal complexes were prepared through the reaction of Zn(II) and Sn(II) with moxifloxacin (MOX) in the presence of glycine (Gly) to investigate their biological activities. IR, UV/VIS and 1H‐NMR analysis have been carried out for insuring the chelation process. Results suggested that MOX and Gly react with the metal ions through the carbonyl oxygen atom and the oxygen atom of the carboxylic group of MOX and Gly. The antimicrobial activity was carried out against some common bacterial and fungal pathogens and the radical scavenging activity (RSA%) was evaluated using DPPH and ABTS methods. Phytotoxic effect of the prepared complexes was evaluated in vitro against Raphanus raphanistrum and Lepidium sativum. Hemolytic activity was tested against cell membrane of erythrocytes. Results showed that the two prepared complexes exhibited high antimicrobial activity against all tested phytopathogens and no significant phytotoxic effect has been observed. Only MOX?Zn(II) complex showed moderate hemolysis at 100 % concentration.  相似文献

Summary Previously undetected race-specific resistance to Bremia lactucae (downy mildew) was located in many lettuce cultivars hitherto considered to be universally susceptible to this disease. This resistance factor(s) may also be widely distributed in other cultivars known to carry combinations of already recognised factors R1 to R11. Specific virulence to match this resistance is almost invariably present in pathogen collections. This situation may be either a relic of the evolutionary history of the B. lactucaeL. sativa asssociation or may reflect a rare mutation in B. lactucae for avirulence on all but a few specialised L. sativa genotypes.  相似文献

Verticillium chlamydosporium produced in submers culture several antifungal and/or phytotoxic compounds which were detected in a bioassay by using the pathogen-host system Puccinia coronata and oat seedlings. The antifungal compounds were also tested against P. recondita on wheat and P. sorghi on corn seedlings. The production of the active metabolic compounds highly depended on the nutrient solution (peptone-Czapek [PC] and malt extract [ME]) and on the fermentation times. Cell-free filtrates of PC-cultures of the fungus were highly phytotoxic; the fungitoxic and phytotoxic compounds were heat-labile and dialyzable. The ethyl acetate extracts of the PC-culture filtrates contained only the antifungal active substances. The antifungal compounds in ME-culture filtrates proved to be heat-stable, could be dialyzed and extracted with ethyl acetate. Ethyl acetate extracts of PC- and ME-culture filtrates at concentrations of 500 μg/ml reduced rust disease incidence by up to 80 % compared to the control treatment. Further studies with extracts of ME-culture filtrates displayed a distinct protective but no systemic activity. The extract interfered with the development of several infection structures of the rust fungi, mostly with the growth of germ tubes as well as with the formation of the aappressoria and haustorial mother cells. Three rust-active fractions were obtained by preparative layer chromatography on silica gel. One of these fractions exhibited phytotoxic activity. The most active antifungal fraction is identical with the macrolid antibiotic monorden which caused a desorientated spiral growth in P. coronata germlings on oat leaves.  相似文献

Plant-pathogenic oomycetes secrete effector proteins to suppress host immune responses. Resistance proteins may recognize effectors and activate immunity, which is often associated with a hypersensitive response (HR). Transient expression of effectors in plant germplasm and screening for HR has proven to be a powerful tool in the identification of new resistance genes. In this study, 14 effectors from the lettuce downy mildew Bremia lactucae race Bl:24 were screened for HR induction in over 150 lettuce accessions. Three effectors—BLN06, BLR38 and BLR40—were recognized in specific lettuce lines. The recognition of effector BLR38 in Lactuca serriola LS102 did not co-segregate with resistance against race Bl:24, but was linked to resistance against multiple other B. lactucae races. Two unlinked loci are both required for effector recognition and are located near known major resistance clusters. Gene dosage affects the intensity of the BLR38-triggered HR, but is of minor importance for disease resistance.  相似文献

The chemical profile and phytotoxic action of Hibiscus trionum essential oil (EO) was studied. In total 17 compounds were identified via GC/MS, representing 94.18 % of the entire oil, with phytol (40.37 %) being the dominant constituent. Bioassay revealed that the EO inhibited root elongation of Medicago sativa and Amaranthus retroflexus by 32.66 % and 61.86 % at 5 mg/mL, respectively; meanwhile, the major component phytol also exhibited significant phytotoxic activity, suppressing radical elongation of Pennisetum alopecuroides, M. sativa and A. retroflexus by 26.08 %, 27.55 % and 43.96 % at 1 mg/mL, respectively. The fact that the EO showed weaker activity than phytol implied that some constituents might trigger antagonistic action to decrease the oil's activity. Our study is the first on the chemical profile and phytotoxic effect of H. trionum EO.  相似文献

Bremia lactucae is recorded for the first time causing downy mildew on common sowthistle (Sonchus oleraceus) and spiny sowthistle (Sonchus asper) in Brazil. The disease and etiological agent are described. Pathogencity of sporangia obtained from S. oleraceus was tested on 12 species belonging to the Asteraceae, already recorded in the literature as hosts of B. lactucae, and four commonly cultivated varieties of lettuce. All four cultivars of lettuce, S. oleraceus and S. asper showed symptoms of the disease and sporulation of the pathogen 5 days after inoculation. It has been observed that the disease occurs on Sonchus spp. throughout the year in Viçosa (state of Minas Gerais), being more common on these two hosts than on lettuce. Both weeds are very common invaders of vegetable growing areas in Brazil. This indicates that those two hosts for the fungus may be important inoculum reservoirs for disease occurring in lettuce, highlighting the need for control of these weeds as part of the procedures aimed at controlling this disease. These results are also in agreement with the views that B. lactucae is split into host‐specific infra‐specific taxa. Lettuce and sowthistles are regarded as closely related, belonging to the same subfamily and tribe in the Asteraceae (Subfamily Cichorioideae: Tribe Lactuceae).  相似文献

On screening the leaf extracts of some higher plants for their volatile antifungal activity against the test organism Aspergillus flavus, the extract of Ocimum adscendens exhibited the strongest fungitoxicity. The leaves showed maximum, fungitoxicity as compared with other plant parts. The volatile fungitoxic fraction obtained as an essential oil was standardized by various physico-chemical properties. The oil showed its fungicidal nature and broad range of activity at its minimum inhibitory concentration. The oil was thermostable and the toxicity remained unchanged even on autoclaving and on storage for up to 360 days. Moreover, the oil proved more active than some prevalent synthetic fungicides and exhibited no phytotoxic effect onVigna radiata.  相似文献

A stock of lettuce cv. Bourguignonne Grosse Blonde d'Hiver used in previous studies was shown to carry the race specific resistance factor R1 and not R9 as originally thought. Investigations into a commercial stock of cv. Bourguignonne which exhibited a heterogeneous reaction to some B. lactucae isolates showed that it was a mixture of two lines. Each line carried one of two R-factors (referred to as R9A and R9B) in a homozygous condition, although their identification was made difficult by the presence of R5/8. No plants were found which carried both R9A and R9B.  相似文献

Studies of the phytotoxic effects between plants can be a crucial tool in the discovery of innovative compounds with herbicide potential. In this sense, we can highlight ruzigrass (Urochloa ruziziensis), which is traditionally used in the crop rotation system in order to reduce weed emergence. The aim of this work was to characterize the secondary metabolites of ruzigrass and to evaluate its phytotoxic effects. In total, eight compounds were isolated: friedelin, oleanolic acid, α‐amyrin, 1‐dehydrodiosgenone, sitosterol and stigmasterol glycosides, tricin and p‐coumaric acid. Phytotoxic effects of the crude methanolic extract and fractions of ruzigrass were assessed using germination rate, initial seedling growth, and biomass of Bidens pilosa, Euphorbia heterophylla and Ipomoea grandifolia. Chemometric analysis discriminated the weed species into three groups, and B. pilosa was the most affected by fractions of ruzigrass. The phytotoxic activities of 1‐dehydrodiosgenone, tricin, and p‐coumaric acid are also reported, and p‐coumaric acid and 1‐dehydrodiosgenone were active against B. pilosa.  相似文献

Infected leaves of the noxious weed johnsongrass were collected in a field with a view to the isolation of toxigenic phytopathogens. Exserohilum turcicum (Pass.) Leonard et Suggs [≡ Drechslera turcica (Pass.) Subramanian et Jain] was isolated from leaf lesions and grown in a liquid culture to yield the known fungal metabolite monocerin (1). This compound was shown to possess (non-specific) phytotoxic activity. Monocerin inhibited seedling growth both of johnsongrass and, to a lesser extent, cucumber. The phytotoxic nature of this compound has not been described previously. Its activity against leaf tissue of two other species, from different plant families, was also demonstrated.  相似文献
