Two novel alkaloids 3β,4α,13α-trihydroxylupanine and 3β,4α-dihydroxy 13α-O-(2'-pyrrolylcarbonyl)-lupanine (calpaurine) have been isolated from the leaves of Ethiopian Calpurnia aurea subsp. aurea. In addition, lupinine and epilupinine (both new for the genus), calpurmenine and calpurmenine pyrrolecarboxylic acid ester (previously found in subsp. sylvatica but not in subsp. aruea) have been isolated together with 13-hydroxylupanine, its tiglate and pyrrolecarboxylic acid esters (calpumine), virgiline and virgiline pyrrolecarboxylic acid ester, alkaloids which have been reported previously from subsp. aruea.  相似文献

A new lupin alkaloid, (−)-3β,13α-dihydroxylupanine was isolated from Cytisus scoparius together with five known sparteine-type lupin alkaloids and tyramine. The absolute structure of the new alkaloid was confirmed by comparison of the natural product with the synthetic sample derived from (+)-13-hydroxylupanine. It was also shown that the alkaloid constituents of C. scoparius differed considerably in the aerial parts, flowers and seeds.  相似文献

The menace caused by ticks and tick-borne diseases is a major limitation to the livestock industry in Africa. The high costs and non-availability of synthetic, chemical acaricides to resource-limited farmers, resistance of ticks to available acaricides and residue problems in meat and milk consumed by humans further complicate matters. The use of plant extracts as a possible source of new acaricides has received much interest in the last decade. In our endeavour to discover natural acaricidal compounds, tick toxicant bioassays were conducted and the chloroform fraction of Calpurnia aurea ethanol leaf extract had good acaricidal activity. Further purification of the fraction revealed two flavonoids, isolated from C. aurea for the first time. These flavonoids were characterized as apigenin-7-O-β-d-glycoside and isorhoifolin by means of NMR spectroscopic and mass spectrometry analysis. Isorhoifolin was the most potent compound (LC50?=?0.65 mg/ml), was not cytotoxic and should be further investigated for its potential as an acaricidal agent.  相似文献

Background aimsOutcomes for patients with glioblastoma remain poor despite aggressive multimodal therapy. Immunotherapy with genetically modified T cells expressing chimeric antigen receptors (CARs) targeting interleukin (IL)13Rα2, human epidermal growth factor receptor 2, epidermal growth factor variant III or erythropoietin-producing hepatocellular carcinoma A2 has shown promise for the treatment of glioma in preclinical models. On the basis of IL13Rα2 immunotoxins that contain IL13 molecules with one or two amino acid substitutions (IL13 muteins) to confer specificity to IL13Rα2, investigators have constructed CARS with IL13 muteins as antigen-binding domains. Whereas the specificity of IL13 muteins in the context of immunotoxins is well characterized, limited information is available for CAR T cells.MethodsWe constructed four second-generation CARs with IL13 muteins with one or two amino acid substitutions, and evaluated the effector function of IL13-mutein CAR T cells in vitro and in vivo.ResultsT cells expressing all four CARs recognized IL13Rα1 or IL13Rα2 recombinant protein in contrast to control protein (IL4R) as judged by interferon-γ production. IL13 protein produced significantly more IL2, indicating that IL13 mutein–CAR T cells have a higher affinity to IL13Rα2 than to IL13Rα1. In cytotoxicity assays, CAR T cells killed IL13Rα1- and/or IL13Rα2-positive cells in contrast to IL13Rα1- and IL13Rα2-negative controls. Although we observed no significant differences between IL13 mutein–CAR T cells in vitro, only T cells expressing IL13 mutein–CARs with an E13K amino acid substitution had anti-tumor activity in vivo that resulted in a survival advantage of treated animals.ConclusionsOur study highlights that the specificity/avidity of ligands is context-dependent and that evaluating CAR T cells in preclinical animal model is critical to assess their potential benefit.  相似文献

Two cDNAs encoding taxoid-O-acetyl transferases (TAX 9 and TAX 14) were obtained from a previously isolated family of Taxus acyl/aroyl transferase cDNA clones. The recombinant enzymes catalyze the acetylation of taxadien-5α,13α-diacetoxy-9α,10β-diol to generate taxadien-5α,10β,13α-tri-acetoxy-9α-ol and taxadien-5α,9α,13α-triacetoxy-10β-ol, respectively, both of which then serve as substrates for a final acetylation step to yield taxusin, a prominent side-route metabolite of Taxus. Neither enzyme acetylate the 5α- or the 13α-hydroxyls of taxoid polyols, indicating that prior acylations is required for efficient peracetylation to taxusin. Both enzymes were kinetically characterized, and the regioselectivity of acetylation was shown to vary with pH. Sequence comparison with other taxoid acyl transferases confirmed that primary structure of this enzyme type reveals little about function in taxoid metabolism. Unlike previously identified acetyl transferases involved in Taxol production, these two enzymes appear to act exclusively on partially acetylated taxoid polyols to divert the Taxol pathway to side-route metabolites.  相似文献

Genlisea aurea A.St.-Hil. is a carnivorous plant endemic species to Brazil in the Lentibulariaceae family. Very few studies have addressed the genetic structure and conservation status of G. aurea and the Lentibulariaceae. Microsatellites markers are advantageous tools that can be employed to predict the vulnerability of Lentibulariaceae species. Therefore, the development of molecular markers focusing the population analyses of Genlisea for future genetic studies and conservation actions are essential. Thus, we developed simple sequence repeats (SSRs) based on in silico analyses of G. aurea draft genome assembly. We characterized 40 individuals from several populations and identified 12 loci that were polymorphic, with heterozygosity between 0.123 and 0.650. We demonstrated that the G. aurea SSR markers work cross-species in Genlisea filiformis, G. repens, G. tuberosa and G. violacea. These markers will be important for future population, phylogeographic and conservation studies in G. aurea and other Genlisea species.  相似文献

A brief pulse of red light accelerates chlorophyll accumulation upon subsequent transfer of dark-grown tomato (Lycopersicon esculentum) seedlings to continuous white light. Such potentiation of greening was compared in wild type and an aurea mutant W616. This mutant has been the subject of recent studies of phytochrome phototransduction; its dark-grown seedlings are deficient in phytochrome, and light-grown plants have yellow-green leaves. The rate of greening was slower in the mutant, but the extent (relative to the dark control) of potentiation by the red pulse was similar to that in the wild type. In the wild type, the fluence-response curve for potentiation of greening indicates substantial components in the VLF (very low fluence) and LF (low fluence) ranges. Far-red light could only partially reverse the effect of red. In the aurea mutant, only red light in the LF range was effective, and the effect of red was completely reversed by far-red light. When grown in total darkness, aurea seedlings are also deficient in photoconvertible PChl(ide). Upon transfer to white light, the aurea mutant was defective in both the abundance and light regulation of the light-harvesting chlorophyll a/b binding polypeptide(s) [LHC(II)]. The results are consistent with the VLF response in greening being mediated by phytochrome. Furthermore, the data support the hypothesis that light modulates LHC(II) levels through its control of the synthesis of both chlorophyll and its LHC(II) apoproteins. Some, but not all, aspects of the aurea phenotype can be accounted for by the deficiency in photoreception by phytochrome.  相似文献

The diurnal trends of gas exchange and chlorophyll fluorescence parameters in four Lycoris species (L. houdyshelii, L. aurea, L. radiata var. pumila and L. albiflora) were determined and compared with a portable photosynthesis analysis system. Our study revealed that L. houdyshelii had the lowest light compensation point (LCP), while the other three species had higher LCP (12.37–14.99 μmol m?2 s?1); L. aurea had the highest light saturation point (LSP) (1,189 μmol m?2 s?1), and L. houdyshelii and L. albiflora had lower LSP with the values being 322 and 345 μmol m?2 s?1, respectively, and L. radiata var. pumila showed the intermediate LSP. Both the species L. houdyshelii and L. albiflora exhibited a typical and obvious decline in net photosynthetic rate (P N) during midday, which was not observed in L. aurea. This indicated a possible photoinhibition in L. houdyshelii and L. albiflora as the ratio of variable to maximum fluorescence (Fv/Fm) values were higher in these two species. The minimal fluorescence (F0) values were lower in L. aurea and L. radiata var. pumila. The diurnal changes of transpiration rate (E) in all four species presented only one peak, appearing between 11:00 h or 13:00 h. By using simple correlation analyses, it was observed that the environmental factors affecting P N were different among four species and the main factors were photosynthetic photon flux density (PPFD) and relative humidity especially for L. aurea and L. radiata. The results of studying indicated that the four species could be divided into two groups. The species L. radiata var. pumila and L. aurea were more adapted to a relatively high irradiance, and L. houdyshelii and L. albiflora could be grown in moderate-shade environment in order to scale up their growth and productivity.  相似文献

The GC/MS detection is reported of over 30 compounds, in extracts of the endosperm and embryos from seeds of Cucurbita maxima. The compounds which were identified from reference spectra include: cis,trans-ABA; trans,trans-ABA; dihydrophaseic acid; IAA; GA4; GA12; GA13; GA25; GA39; GA43; GA49; ent-13-hydroxy-, ent-6α,7α-and ent-7α,13-dihydroxy-, and ent-6α,7α,13-trihydroxykaur-16-en-19-oic acids; ent-7α,16,17-trihydroxy- and ent-6α,7α,16,17-tetrahydroxy-kauran-19-oic acids, ent-6,7-seco-7-oxokauren-6,19-dioic acid and/or ent-6,7-secokauren-6,7,19-trioic acid, and 7β,12α-dihydroxykaurenolide. New compounds, the structures of which were deduced from GC/MS data, include: the 12α-hydroxy-derivatives of GA12, GA14, GA37 and GA4, and the 12β-hydroxy-derivatives of ent-7α-hydroxy- and ent-6α,7α-dihydroxykaurenoic acids.  相似文献

If determination of functional groups, the elementary units of functional diversity, is essential in order to understand how an ecosystem functions, it is also necessary to take into account the specific variability within each group. Within a group the different species' impact on ecosystem functioning can in fact vary widely. Laboratory experiments carried out with two species of bivalve molluscs, Ruditapes decussatus and Venerupis aurea, using paniculate luminescent tracers, show similar sediment reworking modes for the two species, a biodiffusive mode, but with different intensities. Estimation of the mixing parameters using a biodiffusor model gives a biological diffusion rate more than twice as high for Ruditapes decussatus (Rd* =14 10−3 cm2 d−1) as for Venerupis aurea (Rd* = 6.25 10−3 cm2 d−1) and an inverse trend for the output to the water column rates (f*=0.0135cm d−1 for R. decussatus and f*= 0.028 cm d−1 for V. aurea). As R. decussatus buries more of the organic matter which settles at the sediment surface and more rapidly, its effect on the early diagenesis processes and on organic matter bioavailabiliry for the other benthic species in the sediment column differs from that of V. aurea.  相似文献

Plant architecture and phenotypic plasticity under natural conditions remain little known for many rhizomatous species. This study evaluates, in situ, the plastic responses of Alstroemeria aurea plants from three Patagonian populations to flower or flowering-shoot removal. The size and architecture of treated and untreated plants were assessed. Nutrient contents (N, P and K) were evaluated for rhizomes and roots developed in two successive years. Those plants that were deprived of their inflorescences developed, on average, a heavier rhizome than both control plants and plants from which flowering shoots had been removed. Neither of the two treatments applied altered the number of metamers or the branching pattern of the rhizomes. The contents of N, P and K were higher in rhizomes than in roots. In summer, nutrients were more concentrated in inflorescences and the new rhizome segment than in the rhizome segment developed in the previous year. The idea that fruiting failure in A. aurea promotes resource re-assignment from aerial shoots to rhizomes without altering the architecture of plants is supported. The development of the underground portion of aerial shoots in late summer-autumn allows A. aurea plants to take full advantage of short growth periods, but would impose a limit to plasticity.  相似文献

Cardiac metabolism affects systemic energetic balance. Previously, we showed that Krüppel-like factor (KLF)-5 regulates cardiomyocyte PPARα and fatty acid oxidation-related gene expression in diabetes. We surprisingly found that cardiomyocyte-specific KLF5 knockout mice (αMHC-KLF5?/?) have accelerated diet-induced obesity, associated with increased white adipose tissue (WAT). Alterations in cardiac expression of the mediator complex subunit 13 (Med13) modulates obesity. αMHC-KLF5?/? mice had reduced cardiac Med13 expression likely because KLF5 upregulates Med13 expression in cardiomyocytes. We then investigated potential mechanisms that mediate cross-talk between cardiomyocytes and WAT. High fat diet-fed αMHC-KLF5?/? mice had increased levels of cardiac and plasma FGF21, while food intake, activity, plasma leptin, and natriuretic peptides expression were unchanged. Consistent with studies reporting that FGF21 signaling in WAT decreases sumoylation-driven PPARγ inactivation, αMHC-KLF5?/? mice had less SUMO-PPARγ in WAT. Increased diet-induced obesity found in αMHC-KLF5?/? mice was absent in αMHC-[KLF5?/?;FGF21?/?] double knockout mice, as well as in αMHC-FGF21?/? mice that we generated. Thus, cardiomyocyte-derived FGF21 is a component of pro-adipogenic crosstalk between heart and WAT.  相似文献

In this study, we investigated the in vitro antiviral activity of the mycelia of higher mushrooms against influenza virus type A(serotype H1N1) and herpes simplex virus type 2(HSV-2), strain BH. All 10 investigated mushroom species inhibited the reproduction of influenza virus strain A/FM/1/47(H1N1) in MDCK cells reducing the infectious titer by 2.0–6.0 lg ID50. Four species, Pleurotus ostreatus, Fomes fomentarius, Auriporia aurea, and Trametes versicolor, were also determined to be effective against HSV-2 strain BH in RK-13 cells, with similar levels of inhibition as for influenza. For some of the investigated mushroom species—Pleurotus eryngii, Lyophyllum shimeji, and Flammulina velutipes—this is the first report of an anti-influenza effect. This study also reports the first data on the medicinal properties of A. aurea, including anti-influenza and antiherpetic activities. T. versicolor 353 mycelium was found to have a high therapeutic index(324.67), and may be a promising material for the pharmaceutical industry as an anti-influenza and antiherpetic agent with low toxicity. Mycelia with antiviral activity were obtained in our investigation by bioconversion of agricultural wastes(amaranth flour after CO2 extraction), which would reduce the cost of the final product and solve some ecological problems.  相似文献
