The striped bass (Morone saxatilis) and its relatives (genus Morone) are of great importance to fisheries and aquaculture in North America. As part of a collaborative effort to employ molecular genetics technologies in striped bass breeding programs, we previously developed nearly 500 microsatellite markers. The objectives of this study were to construct a microsatellite linkage map of striped bass and to examine conserved synteny between striped bass and three-spined stickleback (Gasterosteus aculeatus). Of 480 microsatellite markers screened for polymorphism, 289 informative markers were identified and used to genotype two half-sib mapping families. Twenty-six linkage groups were assembled, and only two markers remain unlinked. The sex-averaged map spans 1,623.8 cM with an average marker density of 5.78 cM per marker. Among 287 striped bass microsatellite markers assigned to linkage groups, 169 (58.9%) showed homology to sequences on stickleback chromosomes or scaffolds. Comparison between the stickleback genome and the striped bass linkage map revealed conserved synteny between these two species. This is the first linkage map for any of the Morone species. This map will be useful for molecular mapping and marker-assisted selection of genes of interest in striped bass breeding programs. The conserved synteny between striped bass and stickleback will facilitate fine mapping of genome regions of interest and will serve as a new resource for comparative mapping with other Perciform fishes such as European sea bass (Dicentrarchus labrax), gilthead sea bream (Sparus aurata), and tilapia (Oreochromis ssp.).  相似文献

Average age and maximum life span of breeding adult three-spined sticklebacks (Gasterosteus aculeatus) were determined in eight Fennoscandian localities with the aid of skeletochronology. The average age varied from 1.8 to 3.6 years, and maximum life span from three to six years depending on the locality. On average, fish from marine populations were significantly older than those from freshwater populations, but variation within habitat types was large. We also found significant differences in mean body size among different habitat types and populations, but only the population differences remained significant after accounting for variation due to age effects. These results show that generation length and longevity in three-spined sticklebacks can vary significantly from one locality to another, and that population differences in mean body size cannot be explained as a simple consequence of differences in population age structure. We also describe a nanistic population from northern Finland exhibiting long life span and small body size.  相似文献

The mutational mechanism underlying the striking diversity in MHC (major histocompatibility complex) genes in vertebrates is still controversial. In order to evaluate the role of inter- and intragenic recombination in MHC gene diversification, we examined patterns of nucleotide polymorphism across an exon/intron boundary in a sample of 31 MHC class IIB sequences of three-spined stickleback (Gasterosteus aculeatus). MHC class IIB genes of G. aculeatus were previously shown to be under diversifying (positive) selection in mate choice and pathogen selection experiments. Based on recoding of alignment gaps, complete intron 2 sequences were grouped into three clusters using maximum-parsimony analysis. Two of these groups had >90% bootstrap support and were tentatively assigned single locus status. Intron nucleotide diversity within and among loci was low (p-distance within and among groups = 0.016 and 0.019, respectively) and fourfold lower than the rate of silent mutations in exon 2, suggesting that noncoding regions are homogenized by frequent interlocus recombination. A substitution analysis using GENECONV revealed as many intergenic conversion events as intragenic ones. Recombination between loci may explain the occurrence of sequence variants that are particularly divergent, as is the case in three-spined stickleback, with nucleotide diversity attaining dN = 0.39 (peptide-binding residues only). For both MHC class II loci we also estimated the amount of intragenic recombination as population rate (4Ner) under the coalescent and found it to be approximately three times higher compared to point mutations (Watterson estimate per gene, 4Neμ). Nonindependence of molecular evolution across loci and frequent recombination suggest that MHC class II genes of bony fish may follow different evolutionary dynamics than those of mammals. Abstract The morphology of the lateral-line system of the nine-spined stickleback (Pungitius pungitius) and the three-spined stickleback (Gasterosteus aculeatus) has been studied. In the nine-spined stickleback, a preopercular, infraorbital, supraorbital, postotic and peduncular canal can be identified on both sides of the body. Replacement lines are found as a continuation of the preopercular and infraorbital canal. In addition, lines of free neuromasts are found on the mandible and trunk. An accessory line is present above and below the peduncular canal. The presence of both canals and accessory lines on the peduncle suggests that the peduncle in this species has important sensory functions. No canals are found in the three-spined stickleback. Instead, replacement lines corresponding to the canals can be identified on the head. Accordingly, the lateral-line system of the three-spined and the nine-spined stickleback has a different structure. The lateral-line system of both species shows signs of specialization but the three-spined stickleback has a more specialized lateral-line system than the nine-spined stickleback.  相似文献

Three-spined sticklebacks from the northern Baltic proper, caught in 1968 and 1969, were examined with regard to number of plates, body length, head length in relation to body length, and the percental proportions of the forms trachurus, leiurus (= gymnurus) , and semiarmatus. A total number of 1791 specimens were examined. It is shown that the number of asymmetric specimens, regarding the number of plates on the right and left side of the body, is higher than numbers shown earlier, and that there is a significant difference in relative head length between females and males, and, that smaller fish displayed proportionally longer heads than adults. Semiarmatus specimens accounted for about 80 per cent of the samples and only 2 leiurus specimens were found.  相似文献

The nature of gene flow in parasites with complex life cycles is poorly understood, particularly when intermediate and definitive hosts have contrasting movement potential. We examined whether the fine-scale population genetic structure of the diphyllobothriidean cestode Schistocephalus solidus reflects the habits of intermediate threespine stickleback hosts or those of its definitive hosts, semi-aquatic piscivorous birds, to better understand complex host-parasite interactions. Seventeen lakes in the Cook Inlet region of south-central Alaska were sampled, including ten in the Matanuska-Susitna Valley, five on the Kenai Peninsula, and two in the Bristol Bay drainage. We analyzed sequence variation across a 759 bp region of the mitochondrial DNA (mtDNA) cytochrome oxidase I region for 1,026 S. solidus individuals sampled from 2009-2012. We also analyzed allelic variation at 8 microsatellite loci for 1,243 individuals. Analysis of mtDNA haplotype and microsatellite genotype variation recovered evidence of significant population genetic structure within S. solidus. Host, location, and year were factors in structuring observed genetic variation. Pairwise measures revealed significant differentiation among lakes, including a pattern of isolation-by-distance. Bayesian analysis identified three distinct genotypic clusters in the study region, little admixture within hosts and lakes, and a shift in genotype frequencies over time. Evidence of fine-scale population structure in S. solidus indicates that movement of its vagile, definitive avian hosts has less influence on gene flow than expected based solely on movement potential. Observed patterns of genetic variation may reflect genetic drift, behaviors of definitive hosts that constrain dispersal, life history of intermediate hosts, and adaptive specificity of S. solidus to intermediate host genotype.  相似文献

Spatial heterogeneity in diversity and intensity of parasitism is a typical feature of most host-parasite interactions, but understanding of the evolutionary implications of such variation is limited. One possible outcome of infection heterogeneities is parasite-mediated divergent selection between host populations, ecotypes or species which may facilitate the process of ecological speciation. However, very few studies have described infections in population-pairs along the speciation continuum from low to moderate or high degree of genetic differentiation that would address the possibility of parasite-mediated divergent selection in the early stages of the speciation process. Here we provide an example of divergent parasitism in freshwater fish ecotypes by examining macroparasite infections in threespine stickleback (Gasterosteus aculeatus) of four Swiss lake systems each harbouring parapatric lake-stream ecotype pairs. We demonstrate significant differences in infections within and between the pairs that are driven particularly by the parasite taxa transmitted to fish from benthic invertebrates. The magnitude of the differences tended to correlate positively with the extent of neutral genetic differentiation between the parapatric lake and stream populations of stickleback, whereas no such correlation was found among allopatric populations from similar or contrasting habitats. This suggests that genetic differentiation is unrelated to the magnitude of parasite infection contrasts when gene flow is constrained by geographical barriers while in the absence of physical barriers, genetic differentiation and the magnitude of differences in infections tend to be positively correlated.  相似文献

Inter-population differences in stickleback anti-predator behaviour in responses to predation risks are now well established (HUNTINGFORD 1982; GILES & HUNTINGFORD 1984). This paper reanalyses this data in search of individual behavioural differences in the exploration of a novel environment and escape strategy shown by sticklebacks when stalked by a hunting pike (Esox lucius). 8 factors (accounting for 70% of the variance in the original data) emerge from the analysis and significant sex and population effects are described. Factors 1 and 2 correspond to predator investigation and the level of boldness shown towards pike. Factors 3, 4 and 5 separate the escape response into three distinct tactics adopted by sticklebacks when being stalked by the pike. These behaviour groupings are independent of the activity of the stickleback before the presence of the pike is detected. Factors 6, 7 and 8 were comprised of activities (use of open water, of weed beds and of the cover provided by the bottom of the tank) which load heavily for both the periods before and after the sticklebacks reacted to the predator. These groupings may represent different anti-predator tactics adopted by individual sticklebacks.  相似文献

The purpose of this study was to seek evidence for the influence of sexual selection on both male and female three-spined sticklebacks, a species in which the ♂ alone has parental responsibilities. We studied the intrasexual relationships of ♂♂ and ♀♀ prior to courtship, and the intrasexual and intersexual relationships of both sexes during courtship. Results demonstrated that both ♂♂ and ♀♀ compete intrasexually, and suggested that both sexes are discriminative in mate selection. These results are compatible with Trivers' (1972) model of the role of parental investment in sexual selection.  相似文献

Our previous research showed that territorial threespine sticklebacks are more aggressive toward a male conspecific placed in their territory if they have been housed adjacent to a gravid female rather than a male or a nongravid female. This study replicated the condition of a territorial male with an adjacently housed male and compared the results with isolated territorial males. This permitted us to contrast explanations for the increased aggression when the neighbor was a gravid female. If the social context determines the level of aggression, then males with no neighbors (isolates) might be less aggressive because there would be no unique reproductive resource to protect nor any neighbor to protect resources from. Alternatively, if aggression is higher in the isolated group it might be because there was no male neighbor to redirect aggression toward. We found that isolated males were more aggressive toward an intruded male stimulus than males with a male neighbor. The study also provided evidence that sensitization produced by the appearance of an intruding male energizes other aggressive behavior, but not those related to feeding, nor does it cause increase in general activation as measured by increased locomotion.  相似文献

Synopsis Freshwater and marine threespine stickleback, Gasterosteus aculeatus, differ remarkably in armour plate number and body shape, although differences in other morphological characters are also common. Most freshwater populations have apparently evolved after isolation of marine sticklebacks in freshwater. After colonisation of freshwater habitats, they show rapid morphological changes and associated genetic isolation within as few as eight generations. I transferred fish from marine tide pools to two isolated freshwater ponds, differing in habitat characteristics, at the beginning of the breeding season, when females had ripe ovaries and males had breeding coloration. The first generation fish that I sampled from the ponds had significantly fewer armour plates than their marine ancestors and differed in shape. I also found some significant differences between fish sampled from the larger pond and those from a smaller, adjacent pond. This extremely rapid morphological divergence suggests that either the marine sticklebacks were highly phenotypically plastic or that there was very strong natural selection acting on the first generation within freshwater habitats.  相似文献

The ability to discriminate between related and unrelated individuals has been demonstrated in many species. The mechanisms behind this ability might be manifold and depend on the ecological context in which the species lives. In brood‐caring species, both familiarity and phenotype matching are known to be used in kin recognition. However, results of studies disentangling these two phenomena have proved contradictory. We aimed to broaden our knowledge about the mechanisms of kin recognition using shoaling preferences of three‐spined stickleback (Gasterosteus aculeatus) as a model behavior. In our first experiment, focal fish had the choice to shoal either with kin or unfamiliar non‐kin. In half of the trials, kin groups were composed of familiar individuals, while they were unfamiliar in the other half. Focal fish significantly preferred kin as shoaling partner, a result which was not reinforced by familiarity. In our second experiment, focal fish were given the choice between a shoal of familiar kin and a shoal of unfamiliar kin. Here, focal fish did not show any significant preference. These results indicate that familiarity does not impact stickleback's ability to recognize kin. Furthermore, they show that familiarity does not overrule recognition based on phenotype matching or innate recognition, underlining the importance of these mechanisms. Finally, our results lead to the assumption that individual recognition might play a minor role also in non‐kin‐based preferences for familiars.  相似文献

Sex-biased dispersal is capable of generating population structure in nonisolated populations and may affect adaptation processes when selective conditions differ among populations. Intrasexual competition for local resources and/or mating opportunities predicts a male-biased dispersal in polygynous species and a female bias in monogamous species. The patterns of sex-biased dispersal in birds and mammals are well explained by their respective mating systems, but the picture emerging from fish studies is still mixed. Using neutral genetic markers, we investigated whether there is any evidence for sex-biased dispersal among Baltic Sea populations of the three-spined stickleback ( Gasterosteus aculeatus ). The null hypothesis of non sex-biased dispersal was rejected in favour of male-biased dispersal in this species. As the three-spined stickleback has a polygynous mating system, the observed male bias in dispersal is consistent with the hypothesis that local mate competition might drive the observed pattern. Although more research both on the proximate and ultimate causes behind the observed pattern is needed, our results serve as a first step towards understanding patterns of sex-biased dispersal in this species.  相似文献

18 non-breeding three-spined sticklebacks from a heavily predated site a further 24 fish from a predator-free site were screened for their responses to a realistic model pike. The behaviour of the same fish when confronted with a territorial intruder during the breeding season was also recorded, the two tests being carried out in random sequence and with an interval of at least 6 weeks. Multivariate analyses identified major behavioural dimensions in the pike test and in the intruder test, summarized by the labels ‘boldness’ and ‘aggression’ respectively. Sticklebacks from the heavily predated site were both less bold and less aggressive than those from the unpredated site. Levels of aggression for individuals within each population were positively correlated with their boldness scores, but only for the period when the subjects were recovering from a simulated predatory attack.  相似文献

Electrophoretic variation of 13 presumptive enzymatic loci was studied in sticklebacks (Gasterosteus aculeatus) at 6 marine (Baltic) and 13 freshwater sites from Poland. Marine samples are significantly more variable (proportion of polymorphic loci, p # 0.38) than freshwater Samles (p # 0.12). Also the mean number of alleles per locus is significantly higher in the marine samples (n # 1.54) than in freshwater samples (n # 1.14). There is however no significant difference in values of mean heterozygosity between marine and freshwater sticklebacks. Some alleles are found in Baltic sticklebacks only, but all these occur in very low frequencies. Genetic distance between samples is low ranging from D = 0.000053 to 0.031410. However D is significant smaller between marine samples than between freshwater Samles. We suggest than this probaily is the result of larger genetic subdivision of freshwater populations. The low genetic divergence of marine and freshwater populations of the stickleback from poland might be expfained by the recent postglacial colonization of this area from a common refugium.  相似文献
