Role of ethylene in auxin-induced flower bud formation in tobacco explants   总被引:1,自引:0,他引:1  
The effect of ethytene on in vitro flower bud formation in thin-layer explants from tobacco pendicels ( Nicotiana tabacum L. cv. Samsun) was studied Endogenous ethylene production was stimulated by l-minocyclopropanc-l-carhoxylic acid (ACT), and inhibited by aminoethoxyviny lglycine (AVG). resulting in higher and lower ethylene accumulation. respectively. In the presence of an elevated ethylene concentration, the number of flower buds formed after 7 days of culture in explants was increased, compared with the control. Treatment with AVG or with AgNO3 which blocks ethylene action resulted in decreased bud numbers after 7 days of culture. A different effect of ethylene was visible after 14 days of culture, when regeneration was complete. Treatment with AgNO3 led to more bud regeneration, and increasing ethylene concentrations to lower bud numbers. The endogenous production of ethylene was enhanced by high concentrations of 1-naphthaleneacetic acid (NAA).
The inhibitory effect of applied ethylene was almost 100% in explants cultured at low concentrations of NAA (below 1 μ M ). but hardly visible at high concentrations (4.5 μ M ). As a consequence, the optimal NAA concentration shifted to a higher value in the presence of ethylene. These results are interpreted as a reduction in tissue sensitivity to auxin and in regenerative capability by ethylene. The effect of ethylene on auxin action is not exerted at the level of hormone concentration. Neither NAA uptake nor conversion to conjugates was effected by ethylene.  相似文献

Explants from flower stalks of Nicotiana tabacum L. were cultured on different cytokinins to induce flower bud formation. All cytokinins tested except zeatin and zeatin-riboside induced the same maximal number of flower buds. Benzyladenine, benzyladenosine, and dihydrozeatin were the most active compounds whereas isopentenyladenosine and isopentenyladenine acted at a 20-fold higher concentration. These data suggest that the active cytokinins bind to the same receptor with different affinities. The presence of benzyladenine in the medium was necessary only during the first 2 days of culture (initiation period). The equilibrium between benzyladenine and its conjugates (the riboside, glucoside, and nucleotides) after a 4-day pulse was independent of the benzyladenine concentration whether it was inductive or noninductive for bud formation. The level of all derivatives was proportional to the benzyladenine concentration in the medium. Isopentenyladenine was used as a competitive inhibitor of benzyladenine conjugation. Isopentenyladenine concentrations that were too low for bud formation led to a synergistic increase in bud number when applied together with benzyladenine. Isopentenyladenine decreased benzyladenine uptake and conjugation. In spite of the lower uptake, the concentration of free benzyladenine inside the explants was higher in the presence of isopentenyladenine than in its absence whereas the concentration of the 7-glucoside of benzyladenine was lower. It was concluded that the free cytokinin base is the main active compound.  相似文献

Calli were induced in cacao cotyledon explants on a half-strength Murashige and Skoog medium containing 6 × 10-2 g m-3 saccharose and various combinations of 2,4-dichlorophenoxyacetic acid (2,4-D) with kinetin (kin), benzylaminopurine (BAP) or 2-isopentenylphosphate (2-iP). Experiments were carried out on two clones of cacao differing in their susceptibility to black pod disease. The highest percentage of explants forming callus and the most rapid callus development were obtained with 10-6 g m-3 2,4-D and 0.5× 10-6 g m-3 kin. Somatic embryogenesis and rhizogenesis were induced by transferring 3-week-old callus in a half strength Murashige and Skoog medium containing 3 × 10-2 g m-3 saccharose and NAA or IBA in the 0 to 5 × 10-6 g m-3 concentration range. No differentiation could be observed when the medium was supplemented with kin or BAP. The conversion of callus into somatic embryos and roots was accompanied by a drop in phenol content and an increase in peroxidase and IAA-oxidase activities. Moreover, cell differentiation was characterized by the persistence in the callus of one acidic soluble isoperoxidase which was not detected in nondifferentiating callus. Although some differences were noticed between the clones, alterations responsible for cell differentiation were the same in both genotypes. Shoots of poplar (Populus tremula × P. tremuloïdes) were multiplied in vitro and rooted on a rooting medium in the presence of NAA. No rooting occurred in the absence of exogenous auxin. A peak of soluble peroxidase activity, which corresponded to a decrease in the free IAA level in the shoots, preceded rooting These events were considered as corresponding to the initiative phase of rooting. They are preceded by a peak in free IAA activity which might initiate the inductive phase of the rooting process. A burst of ethylene production was measured in both rooting and non-rooting shoots, but the ethylene peak from rooting shoots appeared earlier and was higher. The effects of two inhibitors of polyamine (spermidine and spermine) biosynthesis, cyclohexylamine (CHA; 5 and 10 mM) and methylglyoxal(bis-guanylhydrazone) (MGBG; 0.1, 0.5 and 1 mM), on the organogenic response in vegetative bud-forming tobacco (Nicotiana tabacum L. cv. Samsun) thin layer explants were evaluated micro- and macroscopically at different times during culture. The final number of buds formed and the percentage of organogenic explants was significantly reduced by both inhibitors, but much more so by MGBG than CHA. This inhibitory effect was already evident in MGBG-treated explants on day 5, in terms of the number of meristemoids per explant. On the contrary, in the presence of CHA, the number of meristemoids on day 5 was higher than that in the controls. Between days 9 and 13, meristemoid formation slowed down considerably in inhibitor-treated explants compared with controls. On day 13, the number of bud primordia was similar in control and CHA-treated explants, but significantly lower in MGBG-treated explants. This inhibitor also induced peculiar cytohistological events, such as a reduced formation of oval-shaped cell aggregates on the explant surface and more frequent cases of nucleolar extrusion, while CHA led to the appearance of hypertrophic epidermal cells; callus formation at the basal end of the explant and xylogenesis were also affected by the inhibitors. Ethylene biosynthesis, measured as [ C]methionine incorporation, was stimulated 2- (day 2) to 3-fold (15 h) by 0.5 mM MGBG, whereas CHA (10 mM) had little effect and aminoethoxyvinylglycine (AVG; 0.1 μM), an ethylene synthesis inhibitor, was strongly inhibitory. In control explants, the incorporation of labelled methionine into ethylene and spermidine followed an inverse trend up to day 8. In these explants, free putrescine increased 32-fold and spermidine increased about 10-fold between days 0 and 8. Trichloroacetic acid (TCA)-soluble conjugated putrescine also accumulated dramatically during culture. While CHA provoked a decline in spermidine levels, MGBG caused an unexpected increase in free spermidine and spermine titres; however, its most conspicuous effect was on the further enhancement of putrescine conjugate accumulation, while CHA and AVG had the opposite effect. Results are discussed in view of establishing a putative link between MGBG-enhanced ethylene synthesis, increased conjugate titres and inhibition of meristemoid formation.  相似文献

In vitro flower bud initiation and development depend on the presence of two hormones in the culture medium—auxin (NAA) and cytokinin (BAP). The uptake of both NAA and BAP by the explants was shown to be proportional to the concentrations supplied in the medium over a period of 4 days after the onset of culture. However, when supplied at equal concentrations for 24 h, the NAA uptake was up to 10-fold higher than the BAP uptake. Both hormones are rapidly metabolized by the explants. Nevertheless, the concentrations of free hormones inside the explants appeared to be high and in the case of NAA exceeded the concentration in the medium by more than 1 order of magnitude within 24 h. Apparently flower bud initiation in tobacco explants requires relatively high concentrations free NAA and BAP in the tissue maintained by a continuous supply in the medium. There are at present no indications that the products of hormone metabolism are directly involved in bud formation.  相似文献

Phenolic content and IAA-oxidase (IAA-o) activity have been assayed in cells and medium of tobacco crown gall suspension culture in several stages of culture cycle. The highest content of total phenolics in the cells were found prior to cell division and in the middle stage of intensive growth. The beginning of intensive growth is accompanied by temporary reduction in phenolic level in the cells as well as their intensive secretion to the medium. In the second part of the culture cycle, when the phenolic production was weaker, the majority of these compounds were maintained in the cells. The highest activity of IAA-o in the cells was detected in the middle stage of intensive growth, simultaneously with high phenolic content; following it a considerable decrease of IAA-o activity is correlated with maximum of chlorogenic acid (ChA) content (at reduced amount of total phenolics). IAA-o activity increased again at the end of the stage of intensive growth when the level of phenolics was low including ChA. These data suggest that IAA-o in relation to phenolic level determines cell growth in the culture. In the culture medium — fairly distinct negative correlation between IAA-o activity and phenolic content suggests that the latter participates in enzyme activity regulation. During intensive growth IAA-o activity is strongly inhibited. The results prove that phenolic level. IAA-o activity and auxin level are closely correlated and may constitute essential elements of a mechanism of regulation crown gall cell growth in culture.  相似文献

Pith segments isolated from haploid and diploid Nicotiana tabacum cv. IAC-70 plants from five different heights in the stem, were cultured in vitro on MS-62 medium supplied with 0.5 mg/l IAA and 0.02 mg/l kinetin. Pith tissues of haploid plants showed greater growth potential than those of diploids; peroxidase activity was higher in the calluses of diploid tissues whereas protein content was higher in those of haploids. After three subcultures the growth pattern was inversed, as were the results for peroxidase activity and protein content. After the onset of culture, peroxidase activity in haploid explants dropped sharply, reaching values lower than in diploid tissues 40 d later, but after three subcultures these values were higher than those for diploid calluses. The results are discussed from the viewpoint of a possible relationship between greater polyploidization in haploid tissues on the one hand and growth decrease and peroxidase activity increase on the other.  相似文献

A comparative analysis of IAA and total cytokinin contents at various developmental stages of floral morphogenesis (I–XII) in transgenic tobacco (Nicotiana tabacum L.) plants with mutant phenotypes was performed. Disturbances in flower structure were correlated with changes in the content and dynamics of these phytohormones. In plants with the phenotype “wavy corolla”, IAA content increased by three times as compared with control at stage VIII; in plants with the phenotype “longostyly”, IAA content increased gradually from VIII to XII stage and sharply (by nine times) at stage XII. In plants with the phenotype “wavy corolla”, total content of cytokinins was lower than in control at all developmental stages (except for stage I), whereas their dynamics coincided with that in control flowers. In plants with the phenotype “longostyly”, cytokinin content was twice lower than in control at early developmental stages, whereas it oscillated at later stages, approaching to control value at stage XII. It is supposed that differences observed could result from T-DNA insertion into plant own genes, which led to changes in the content and dynamics of phytohormones in developing flowers and production of anomalies in the flower structure.  相似文献

The rooting recalcitrant rac Nicotiana tabacum cv Xanthi mutant has been multiplied in vitro under the form of shoots in parallel to wild-type. rac Shoots grew at a lower rate and did not root whatever the treatments when compared to those of wild-type shoots. They were characterized by a higher lignin level, a higher total specific peroxidase activity with higher activity of both acidic and basic isoperoxidases (although missing and supernumerary isoenzymes were observed), and higher ethylene production. These observations might be causally related to growth inhibitions as similar incidences have been noted in different stress-induced growth limitation, through cell wall rigidification and auxin catabolism. The relationship between these aspects and rooting recalcitrance remains to be explored.  相似文献

It has been established that not only the higher primates, such as chimpanzees and orangutans, but also the lower ones—macaques, capuchins, squirrel monkeys—have the “mathematical capabilities.” The detailed study of formation of quantitative concepts in primates of different levels of phylogenesis allows disclosing mechanisms of formation of this function in human.  相似文献

We analyzed low molecular mass phenolics, lignin content and both soluble and cell wall bound peroxidase activity in the needles of three Picea omorika (Pancic) Purkyne lines grown in the generative seed orchard. The highest values of the total phenol content as well as of catechine, caffeic acid, coniferyl alcohol, isoferulic acid and lignin concentration were detected in B5 line (“semidichotomy” line). The soluble guaiacol peroxidase activity was the highest in A3 line (line “borealis”). The highest activity of cell wall bound peroxidases was measured in B5 line, and it was in correlation with lignin content.  相似文献

The application of gibberellic acid via the stem of intact preclimacteric carnation flowers inhibited the climacteric surge of ethylene evolution by the flowers. Gibberellic acid also inhibited the rate of ethylene production by all individual floral parts during both the early preclimacteric (low basal level of ethylene production) and the later climacteric stages of flower development. The extent of inhibition did however, vary from one floral part to another. The most pronounced inhibition was recorded in the petal bases between the preclimacteric and senescing stages. This suggests that the petal base is an important regulatory site for ethylene production and therefore may be involved in controlling the onset and degree of petal inrolling. In all floral parts endogenous levels of ACC were reduced with GA3 treatment, being more pronounced in the petal bases. In A. angustifolia seeds the highest values of freeIAA occurred in the embryonic axis, at the initial phases of development. Thesevalues decreased sharply coincident with the increase of IAA with thedifferentiation of cotyledons and seed elongation. During seed development,tryptophan concentrations varied inversely with free IAA and directly withconjugated IAA. An increase in peroxidase activity was followed by a decreaseinfree IAA in the embryo axis, and in conjugated IAA in the megagametophyte.Megagametophyte tissues did not exhibit significant variation in free IAAduringseed development. A previous paper (Galaud et al. 1993. Physiol. Plant. 87: 25–30) reported a rapid demethylation of DNA after mechanical rubbing of young Bryonia dioica internodes, leading to growth inhibition. In order to assess an eventual causative role of DNA demethylation in the mechanisms underlying thigmomorphogenesis, the effects of the anti-DNA methylation drugs, 5-azacytosine (5-azacyt) and 5-azacytidine (5-azacyd) were investigated. Treatment with 5-azacyd induced a decrease of the methylated DNA level. This was accompanied by an increase of peroxidase activity and of ethylene production. On the contrary, 5-azacyt had no significant effect on growth, peroxidase activity and ethylene production. The use of 5-azacyd as an inducer of DNA demethylation is discussed in relation to the control of gene expression  相似文献

Sequence analysis of five gene families that were isolated from tobacco thin cell layer explants initiating floral development [Meeks-Wagner et al. (1989). Plant Cell 1, 25-35] showed that two encode the pathogenesis-related proteins basic chitinase and basic beta-1,3-glucanase, while a third encodes the cell wall protein extensin, which also accumulates during pathogen attack. Another sequence family encodes the water stress-induced protein osmotin [Singh et al. (1989). Plant Physiol. 90, 1096-1101]. We found that osmotin was also induced by viral infection and wounding and, hence, could be considered a pathogenesis-related protein. These genes, which were highly expressed in explants during de novo flower formation but not in explants forming vegetative shoots [Meeks-Wagner et al. (1989). Plant Cell 1, 25-35], were also regulated developmentally in day-neutral and photoresponsive tobacco plants with high expression levels in the roots and moderate- to low-level expression in other plant organs including flowers. An unidentified gene family, FB7-4, had its highest level of expression in the basal internodes. Our findings indicate that these genes, some of which are conventionally considered to encode pathogen-related proteins, also have a complex association with normal developmental processes, including the floral response, in healthy plants.  相似文献

This study examined the impacts of elevated CO2 or O3 on indole-3-acetic acid (IAA) content, activities of IAA oxidase (IAAO) and peroxidase (POD) in Ginkgo biloba leaves. Plants grown in open-top chambers were exposed to ambient atmosphere (control; C), elevated CO2 and elevated O3 from 1 June to 30 September. An increase in IAA content and decrease in IAAO and POD activities were observed in plants exposed to elevated CO2 compared with C. Elevated O3 had no significant effect on IAA content and IAAO activity, but increased POD activity during the early days. When trees pre-exposed to elevated CO2 were transferred to elevated O3 or C, the increase in IAAO activity resulted in the decrease in IAA content. When trees pre-exposed to elevated O3 were transferred to elevated CO2 or C, IAA content, IAAO and POD activities showed no significant changes. The influence of POD activity on the IAA activity was low.  相似文献

Relationship of bud production (axillary and terminally) of annual shoot (1Y) and/or the content of bud-derived indol-3-acetic acid (IAA) to branching of the 1Y was studied in common walnut (Juglans regia L.), cvs. Franquette and Lara. Cultivar-related branch architecture was determined. Lara tended to branch more densely than Franquette (53 vs. 42%). Significantly more fruiting off-spring shoots (FO) than vegetative ones (VO) grew-out per 1Y in both cultivars, whereas the ratio FO/VO of Lara exceeded that of Franquette by four times. An acrotonic branching pattern was more strongly expressed in Lara compared to Franquette. Bud-derived IAA was influenced by the cultivar (Franquette had 3.6 times more cumulative IAA along the 1Y than Lara), and by the relative position (terminal, subterminal, medial and basal) of the buds along 1Y. An opposite relationship between branching density and cumulative IAA content was established in both cultivars. At the 1Y relative position level, the opposite ratio between branching density and IAA content was clearly shown only on the basal position of the bud along 1Y in the Lara cultivar. Such an inconsistent linkage between bud production and the IAA spatial distribution along the 1Y illustrated that hormonal factors probably weakly affect the branching of Franquette and Lara. The length of the parent 1Y, the position of the buds along the 1Y-length, and the fate of the buds seemed to have a stronger influence on the bud out-growth and further development of the off-springs. In further analyses, seasonal fluctuations of the IAA, and the following activity of the buds should be investigated in order to improve the understanding of a complex branching phenomenon in walnut.  相似文献
