The unusual morphology of the coccoid, neustonic alga Nautococcus mammilatus Korschikov prompted its study with the electron microscope. Ultrastructural features including a fibrous cell wall with definite periodicity and an external tomentum were observed. These and other features of the alga are compared with those of related, species, and the use of cell wall characteristics in taxonomy is discussed.  相似文献

Observations on the ultrastructure of Friedmannia israelensis Chantanachat & Bold revealed the presence of a phycoplast and zoospores with cruciate rootlets. During mitosis, the nuclear envelope partially disintegrates and the basal bodies remain at the cell surface on either side of the developing cleavage furrow. The events during mitosis and cleavage in Friedmannia resemble those reported in the other green algae, Platymonas and Pleurastrum.  相似文献

Strains of Chlamydomonas with cross-reacting sporangium wall autolysins also correspond in morphology, reproduction, development, and physiological properties. We therefore undertook a revision of the systematics of the genus based on this correlation. Isolates of Chlamydomonas available in culture collections were re-examined microscopically, preferably with respect to their reproduction and development. In particular all authentic clones, on which earlier species diagnoses had been founded, were included. Their detectable autolysins were tested crosswise in bioassays. We intend to complete and extend species diagnoses in order to separate species more precisely. - At first 7 strains with a common sporangium autolysin have been examined, four of which are the authentic cultures of the species C. aggregate Deason et Bold, C. akinetos Deason et Bold, C. applanata Pringsheim, and C. humicola Lucksch. They have been brought together under the name of the species first described, C. applanata Pringsheim, due to their apparent similarities in morphology and development. An amended species diagnosis is presented. The significance of authentic cultures as important sources of further information is discussed.  相似文献

Ultrastructural studies revealed that Chlorosarcina stigmatica Deason differs from C. brevispinosa and C. longispinosa Chantanachat and Bold because the vegetative cell possess a pyrenoid, and the zoospores have chlorophycean characteristics. Zoospores of the latter species exhibit pleurastrophycean characteristics including counterclockwise absolute orientation of the flagellar apparatus components. Nuclear division in C. longispinosa, like that of Friedmannia israelensis Chantanachat and Bold, is metacentric. It is suggested that a new genus, Desmotetra, be erected for Chlorosarcina stigmatica, and that it, along with Chlorosarcinopsis Herndon, be placed in the Chlorococcales, Chlorophyceae. Chlorosarcina brevispinosa and C. longispinosa belong in the Pleurastrales, Pleurastrophyceae.  相似文献

The effects of incubation time and deficiencies of calcium and nitrate on zoospore production were investigated in Botrydiopsis arhiza Borzi, B. intercedens Vischer et Pascher, Bumilleriopsis peterseniana Vischer, Pseudobumilleriopsis pyrenoidosa Deason et Bold, and Ophiocytium maius Naegeli. Release of zoospores occurred after the transfer of stationary phase, vegetative cells into fresh Bold's basal medium. Maximum yields of zoospores were produced on the second day after transfer, 0·5–1·0 h after the start of the light period of a 12–12 h light-dark cycle. Lack of calcium or nitrate reduced the yield of zoospores in each taxon. Only O. maius produced large numbers of motile cells in nitrate-free medium.  相似文献

Eight complete nuclear-encoded small-subunit ribosomal RNA (18S rRNA) gene sequences were determined for four genera of the Microthamniales (Pleurastrophyceae) and for Gloeotilopsis planctonica Iyengar & Philipose (Ulvophyceae, Ulotrichales) to investigate evolutionary relationships within the Microthamniales and the taxonomic position of this order within the green algae. Phylogenies inferred from these data revealed specific relationships at the level of genera and species that disagree with those inferred from vegetative cell morphology but agree with those inferred from motile cell characters. The rRNA phylogenies provide even better resolution than that gained from morphology alone. The coccoid lichen alga Trebouxia spp. is specifically related to other coccoid lichen and soil algae (i.e. Myrmecia biatorellae Boye-Petersen and Friedmannia israelensis Chantanachat & Bold), forming the “Lichen Algae Group,” an evolutionarily distinct lineage within the Microthamniales. Trebouxia is a paraphyletic and Pleurastrum a polyphyletic genus in rRNA phylogenies. In contrast to previous hypotheses based on morphology, Pleurastrum terrestre Fritsch & John is not closely related to Trebouxia but occupies an isolated position within the Microthamniales. The filamentous alga Microthamnion kuetzingianum is not ancestral to coccoid members of the Microthamniales but is closely related with the coccoid Fusochloris perforata (Lee & Bold) Floyd, Watanabe & Deason. The Microthamniales are inferred to be an array of independent lineages that radiate nearly simultaneously and may also include some autosporic coccoid taxa previously classified in the Chlorophyceae. Monophyly of the Microthamniales could not be demonstrated unequivocally. In contrast to a hypothesis based on ultrastructure, the Microthamniales are evolutionarily distinct from the Ulotrichales. The latter are ancestral to the radiation of the Microthamniales and the Chlorophyceae in the rRNA phylogenies.  相似文献

The influence of increasing salinity stress on plant growth, antioxidant enzymes and proline metabolism in two cultivars of Vigna radiata L. (cv. Pusa Bold and cv. CO 4) was investigated. Salt stress was imposed on 30-days-old cultivars with four different concentrations of NaCl (0, 100, 200 and 300 mM). The roots and shoots of CO 4 showed greater reduction in fresh weight, dry weight and water content when compared to Pusa Bold with increasing salt stress. Under salinity stress, the roots and shoots of CO 4 exhibited higher Na+: K+ ratio than Pusa Bold. The activities of reactive oxygen species (ROS) scavenging enzymes and reduced glutathione (GSH) concentration were found to be higher in the leaves of Pusa Bold than in CO 4, whereas oxidized glutathione (GSSG) concentration was found to be higher in the leaves of CO 4 compared to those in Pusa Bold. Our studies on oxidative damage in two Vigna cultivars showed lower levels of lipid peroxidation and H2O2 concentration in Pusa Bold than in CO 4 under salt stress conditions. High accumulation of proline and glycine betaine under salt stress was also observed in Pusa Bold when compared to CO 4. The activities of proline biosynthetic enzymes were significantly high in Pusa Bold. However, under salinity stress, Pusa Bold showed a greater decline in proline dehydrogenase (ProDH) activity compared to CO 4. Our data in this investigation demonstrate that oxidative stress plays a major role in salt-stressed Vigna cultivars and Pusa Bold has efficient antioxidative characteristics which could provide better protection against oxidative damage in leaves under salt-stressed conditions.  相似文献

Chlorophyll a:b ratios for Chlorella capsulata Guillard, Bold & MacEntee, Chlorosarcinopsis halophila Guillard, Bold,& MacEntee, Nannochloris atomus Butcher and Micromonas pusilla (Butcher) Manton & Parke were found to be 1.74, 2.46, 1.89, and 1.53, respectively. Compared to previously published values in marine planktonic algae, these ratios are similar to the average value (2.17, SD = 1.12) reported for 25 species in 15 genera. Both the new data and previously published data support the hypothesis that marine algae have lower chlorophyll a:b ratios than do other chlorophyll b containing plants.  相似文献

Before cytokinesis, the identically constructed chromatophores ofHantzschia amphioxys andAchnanthes coarctata are transformed into less effigurated bodies. In normal cytokinesis, the course of mitosis, chromatophore division, and cleavage furrowing are exactly synchronized. The division of the chromatophore appears as a passive process, i.e. intersection by the cleavage furrow. In inequal cell divisions before the formation of inner valves cytokinesis can take place without chromatophore division. Once chromatophore division without mitosis and cytokinesis was observed. InHantzschia there are three types of inner valve formation, inAchnanthes coarctata only two. The inner valves develop under unfavorable growth conditions, the cells possessing them, however, are not resting spores as in some other diatoms. InHantzschia, auxospore formation is suppressed under the cultural conditions used, the cells multiply intensely without diminution.

Vegetative cells and zoospores of Hormotilopsis gelatinosa Trainor & Bold, H. tetravacuolaris Arce & Bold, Planophila terrestris Groover & Hofstetter, and Phyllogloea fimbriata (Korchikov) Silva were examined by transmission electron microscopy. All cells had pyrenoids traversed by cytoplasmic channels. Zoospores were quadriflagellate and had essentially cruciate flagellar apparatuses. Scales were present on free-swimming zoospores. These features are essentially identical to those of Chaetopeltis sp. and are dissimilar to those of other described green algae. The new order Chaetopeltidales is created to accommodate the genera Chaetopeltis, Hormotilopsis, Planophila sensu Groover & Hofstetter, Phyllogloea, Dicranochaete, and Schizochlamys, organisms previously scattered among the orders Tetrasporales, Chloro-coccales, Chlorosarcinales, and Chaetophorales. Members of the order are closely related to the ancestral chlorophycean flagellate genus Hafniomonas, may be ancestral with respect to other Chlorophyceae, and may also be closely related to the ulvophycean order Ulotrichales.  相似文献

The Deason House is located in the heart of the Piney Woods region in the town of Ellisville, Mississippi. During the American Civil War, the Deason House became the epicenter for an event that started a rebellion against the Confederate government. On 5 October 1863, a Confederate deserter named Newton Knight allegedly shot and killed Confederate Major Amos McLemore in the Deason House, which eventually lead to the formation of the Free State of Jones. Despite the historical importance of the house, oral accounts and written documents have failed to provide accurate construction history of the structure. We coupled techniques of dendrochronology and geophysical survey to better understand the construction history of the Deason House and cultural activities of its inhabitants. We extracted 35 Pinus palustris (Mill.) timbers along the west-facing exterior weatherboards and from various logs within the second-floor attic, 25 of which were successfully crossdated against a P. palustris reference chronology located 15 km south in De Soto National Forest (1742–2013). The Deason House chronology anchored against the De Soto time series during the period 1742–1875 (r = 0.55, t = 4.01, p < 0.0001) and extended the reference chronology back to the year 1680 CE. Clustering of cutting- and near-cutting dates from timbers revealed 3 possible stages of construction. First, the house likely existed as a single-pen structure built in the winter of 1835/1836, based on two timbers with wane/bark that were affixed to the original chimney made of clay bricks fired on the homestead. Second, based on the clustering of 1855 cutting dates, we suggest this was the most likely time the original structure was expanded with a vestibule, porch, and larger 4-room house. Finally, six timbers revealed a back addition was constructed in the year 1866. Ground penetrating radar (GPR) helped provide evidence mentioned in written records that the back addition (1866) existed as the detached kitchen then was affixed to the structure ca. 1890. The GPR data revealed geophysical anomalies that indicated the original sites of the detached kitchen, water well, livestock pen, privies, and trash pit. Through the use of dendrochronological analyses and geophysical survey, we were able to provide an updated history of the Deason House construction dynamics and cultural activities of its inhabitants beyond the historical record.  相似文献

An ultrastructure study of mitosis and cytokinesis in the sessile sporangium of Trentepohlia aurea (L.) Mart, was made to clarify the phylogenetic position of the alga. Mitosis was closed and centric at late anphase with cytokinesis involving the production of cleavage membranes by dictyasames between the numerous, well-separated daughter nuclei. Neither phycoplast nor phragmoplast microtubules were observed during cytokinesis. The lack of phycoplast microtubules and the presence of multilayered structures in flagellated cells suggest Trentepohlia is phylogenetically related to those green algae thought to have given rise to the land plants. The primitive type of mitosis and the lack of microbodies suggest that the ancestors of Trentepohlia may have branched off from this line relatively early.  相似文献

A restriction endonuclease with a novel site-specificity has been isolated from the Escherichia coli strain RFL31. The nucleotide sequences around a single Eco31I cut on pBR322 DNA and two cuts of λ DNA have been compared. A common 5′GAGACC3′CTCTGG sequence occurs near each cleavage site. Precise mapping of the cleavages in both DNA strands places the cuts five nucleotides to the left of the upper sequence and one nucleotide to the left of the lower sequence. Abstract The rehabilitation of native plant communities in urban bushland remnants is an increasingly important activity requiring the collection of large amounts of seed. Best practice generally identifies that local seed are best, but how far does the local provenance extend? Using the DNA fingerprinting technique amplified fragment length polymorphism, we assessed genetic differentiation between potential seed source populations and the target population, Bold Park, a large and significant bushland remnant in Perth, Western Australia. For each of 15 species, analysis of molecular variance was used to partition genetic variation within and among populations. Genetic differentiation between Bold Park and potential seed source populations was assessed by non‐metric multidimensional scaling ordination, and statistically by Fisher’s exact tests. The partitioning of variation among populations (ΦST) varied from 0.66 for Santalum acuminatum to 0.04 for Mesomelaena pseudostygia. For eight of 15 species, Bold Park plants were completely or largely non‐overlapping with other populations in ordinations, suggesting genetic differentiation and a narrow provenance. Five species showed overlap between Bold Park and some other, but not all, populations sampled, with geographically closest populations generally undifferentiated. Only two species, Acanthocarpus preissii and Mesomeleana pseudostygia, showed little genetic differentiation between Bold Park and all other populations, suggesting a regional genetic provenance. These species can be classified into three broad provenance classes – narrow, local and regional – to help guide decisions about appropriate seed‐collection zones for the rehabilitation of urban bushland remnants.  相似文献

Embryo-like structures (ELS) with clearly defined cotyledons and radicles have been regenerated fromLupinus albus L. microspores. ELS induction from microspores in liquid medium was successful, with a maximum of over 3,500 ELS per anther being produced from microspores predominantly at the early binucleate stage of development. Cytological analysis revealed that first pollen grain mitosis occurred in closed buds with maximum ELS production being obtained from buds at the point of first petal emergence. Generally there was a lack of synchronisation of microspores within anthers from the less mature bud germination from ELS has not been observed; recurrent somatic embryogenesis occurred following internal cleavage within the ELS or on the surface of the ELS. Laser scanning confocal microscopy and TEM were used to study the morphology of secondary plastids in algae of the genus Mallomonas (Synurophyceae). At interphase, Mallomonas splendens (G. S. West) Playfair, M. rasilis Dürrschm., M. striata Asmund, and M. adamas K. Harris et W. H. Bradley contained a single H‐shaped plastid consisting of two large lobes connected by a narrow isthmus. Labeling of DNA revealed a necklace‐like arrangement of plastid nucleoids at the periphery of the M. splendens plastid and a less‐patterned array in M. rasilis. The TEM of M. splendens and M. rasilis showed an electron‐dense belt surrounding the plastid isthmus in interphase cells; this putative plastid‐dividing ring (PD ring) was adpressed to the inner pair of the four plastid membranes, suggesting that it is homologous to the PD ring of green and red plastids. The PD ring did not contain actin (indicated by lack of staining with phalloidin) and displayed filaments or tubules of 5–10 nm in diameter that may be homologous to the tubules described in red algal PD rings. Confocal microscopy of chl autofluorescence from M. splendens showed that the plastid isthmus was severed as mitosis began, giving rise to two single‐lobed daughter plastids, which, as mitosis and cell division progressed, separated from one another and then each constricted to form the H‐shaped plastids of daughter cells. Similar plastid division cycles were observed in M. rasilis and M. adamas; however, the plastid isthmus of M. striata was retained throughout most of cell division and was eventually severed by the cell cleavage furrow.  相似文献
