Five strains of Thiobacillus ferrooxidans, which included three recent isolates from a uranium mine, possessed flagella. Three of the strains had several pili per cell. The dimensions, fine structure, and orientation of the flagella were different. Both polar and peritrichous flagella were observed, indicating strain-dependent ultrastructural variation in acidophilic thiobacilli. Neither flagella nor pili were detected in eight other strains of T. ferrooxidans and two strains of Thiobacillus acidophilus by electron microscopy, although all of the cultures contained motile cells.  相似文献

Zooplankton species diversity and selected chemical parameters were investigated at three stations in Lake St. Clair, Ontario, Canada, from 15 June–26 August, 1971. Primary productivity and zooplankton species diversity were greatest at stations 1 and 2 which were enriched by the Thames River drainage. No significant correlation between total zooplankton diversity and chlorophyll a was found, however, within the Cladocera and Copepoda, positive correlations with chlorophyll a, reactive silicate and nitrate were shown at stations 2 and 3. Rotifer species diversity showed negative correlation with chlorophyll a, nitrate and reactive silicate.  相似文献

The Bay of Quinte receives drainage from several large river systems, including the Moira River which carried sediment from mines into the Bay from the 1880s to the 1960s. We are investigating possible metal contamination of submerged weed beds and marsh biota which may contribute to the low diversity and biomass of macrophyte beds and Typha marshes in the Bay. In 1987, sediment, macrophytes and snails were sampled in wetlands close to the Moira River and at Hay Bay (part of the Bay of Quinte presumably unaffected by mine effluents) located 20 km from the Moira. Some element concentrations in sediment and biota were determined by neutron activation analysis (NAA) including Al, As, Br, Ca, Co, Cl, Cr, Cs, Fe, Hf, K, La, Na, Mg, Sb, Sc, Rb, Ta, Th, Ti, U, V and Zn. Other elements were analysed by acid dissolution and atomic absorption spectrophotometry (AAS) including Ag, As, Cu, Hg, Ni, Pb, and Zn. Levels of As in sediments and plants were higher close to the Moira River, whereas Cu and Ni showed the opposite pattern in sediments. The usefulness of species as bioassays differed: Stagnicola elodes Say accumulated significantly higher levels of Cu (35 vs 18 ppm) and V (l. l vs 0.5 ppm) than Planorbella trivolvis Say collected from the same sites. The macrophyte, Myriophyllum spicatum L. acted as an accumulator of Pb (up to 9.6 ppm), whereas Pb in Vallisneria americana Michx. at the same sites was undetectable.  相似文献

To determine if different algal viruses ( Phycodnaviridae ) share common patterns of seasonal abundance, quantitative PCR methods were developed and applied to monitor the abundances of three different viruses in Lake Ontario, Canada over 13 months. Throughout the year, the abundances of two different phycodnavirus polB gene fragments (LO1b-49 and LO1a-68) varied by more than two orders of magnitude, peaked during the autumn months, and were lowest during the summer. The seasonal abundance patterns of these two virus genes were similar and both were detected in almost every sample, but LO1b-49 was consistently an order of magnitude more abundant than LO1a-68. LO1b-49 reached a maximum abundance of 5413 ± 312 genes ml−1, whereas LO1a-68's abundance peaked at only 881 ± 113 genes ml−1. Another phycodnavirus polB fragment that was monitored (LO1b-16) was detected in only a few samples, but reached a higher maximum concentration (6771 ± 879 genes ml−1) than either LO1b-49 or LO1a-68. The results of this year-long investigation of virus gene abundances suggests that Lake Ontario's phycodnavirus community is composed of persistent viruses detectable throughout the year and transient viruses present in only a few sporadic samples. The results also suggest that some persistent algal viruses are able to survive at relatively low abundances through several seasons.  相似文献

A study of the horizontal and vertical distribution of heterotrophic bacteria in brackish Lake Shira in summer periods showed that mesophilic bacteria dominated in all areas of the lake, whereas psychrotolerant bacteria dominated in the metalimnion and hypolimnion of its central part. Nonhalophilic bacteria were mostly mesophilic and dominated in coastal waters. Most psychrotolerant bacteria were able to grow in the presence of 5–10% NaCl. Heterotrophic bacteria isolated in different regions of the lake were identified to a generic level. The isolates were classified into autochthonous and allochthonous microorganisms on the bases of their distribution pattern in the lake water, halotolerance, and ability to grow at low temperatures.  相似文献

Microbially mediated reduction and immobilization of U(VI) to U(IV) plays a role in both natural attenuation and accelerated bioremediation of uranium-contaminated sites. To realize bioremediation potential and accurately predict natural attenuation, it is important to first understand the microbial diversity of such sites. In this paper, the distribution of sulfate-reducing bacteria (SRB) in contaminated groundwater associated with a uranium mill tailings disposal site at Shiprock, N.Mex., was investigated. Two culture-independent analyses were employed: sequencing of clone libraries of PCR-amplified dissimilatory sulfite reductase (DSR) gene fragments and phospholipid fatty acid (PLFA) biomarker analysis. A remarkable diversity among the DSR sequences was revealed, including sequences from δ-Proteobacteria, gram-positive organisms, and the Nitrospira division. PLFA analysis detected at least 52 different mid-chain-branched saturate PLFA and included a high proportion of 10me16:0. Desulfotomaculum and Desulfotomaculum-like sequences were the most dominant DSR genes detected. Those belonging to SRB within δ-Proteobacteria were mainly recovered from low-uranium (≤302 ppb) samples. One Desulfotomaculum-like sequence cluster overwhelmingly dominated high-U (>1,500 ppb) sites. Logistic regression showed a significant influence of uranium concentration over the dominance of this cluster of sequences (P = 0.0001). This strong association indicates that Desulfotomaculum has remarkable tolerance and adaptation to high levels of uranium and suggests the organism's possible involvement in natural attenuation of uranium. The in situ activity level of Desulfotomaculum in uranium-contaminated environments and its comparison to the activities of other SRB and other functional groups should be an important area for future research.  相似文献

Samples of muscle from 4,773 specimens of 18 species of wild mammals from Ontario were examined for Trichinella. One of 12 mink (Mustela vison), 83 of 1,821 fisher (Martes pennanti) and 68 of 1,980 marten (Martes americana) had T. spiralis. Prevalences of infections by Trichinella were determined for fisher and marten from the Algonquin region, over a 10-yr period. Prevalences ranged from 0.9-9.2% in fisher and 1.3-8.7% in marten indicating that the parasite is well-established in the region. Prevalences of Trichinella increased with age of both fisher and marten. Intensities determined for the 1981-1982 sample ranged from 0.4-15.8 larvae/g for fisher and 22.4-159.7 larvae/g for marten. Higher intensities were not correlated with older hosts. Fisher and marten appeared to be the key hosts maintaining Trichinella in the Algonquin region, but transmission dynamics were unclear. Transmission may include an unidentified small rodent or other host and natural carrion-feeding.  相似文献

Our study examines the relative importance of the causal linkages between exogenous total phosphorus (TP) loading and internal nutrient recycling with the water quality conditions in Lake Simcoe, Ontario, Canada. We enhance the mechanistic foundation of a simple TP mass-balance model, originally developed to guide the eutrophication management in the system. The structural improvements include the incorporation of macrophyte dynamics, the explicit representation of the role of dreissenids in the system, and the improved portrayal of the interplay between water column and sediments. Our model provides good agreement with the observed TP variability in the system during the study period (1999–2007). Consistent with empirical evidence, our model predicts that macrophyte uptake from the interstitial waters is responsible for a significant loss of P from the sediments. Our model also suggests that dreissenids filter a considerable amount of particulate P from the water column, but the effective clearance rate is significantly lower with a substantial amount of the filtered particles (> 85%) returned into the water column as faeces, pseudofeces or other metabolic excreta. P diffusive fluxes from the sediments account for about 30–35% of the exogenous P loading in Lake Simcoe. The sediments in the main basin are mostly driven by fast diagenetic processes of settling organic matter from the epilimnion, suggesting an internal P loading of 9.2 tonnes yr 1. Finally, our study attempts to explain the lack of distinct decreasing trends in ice-free TP concentrations after the invasion of dreissenid mussels, suggesting that the presence of active nutrient recycling pathways, potentially magnified by the particular morphological features and hydrodynamic patterns of Lake Simcoe, could counterbalance the direct effects of dreissenid filtration.  相似文献

A technique was developed for measuring (14)CO(2) uptake by chemolithotrophic bacteria directly in pyritic materials associated with coal and coal refuse. There was good correlation between (14)CO(2) uptake, as determined by this technique, and the most probable number of iron-oxidizing bacteria. Maximal (14)CO(2) uptake occurred in coal refuse material 2 to 3 years old, and only slight incorporation was demonstrated in fresh material or material 40 years old. Samples taken from the surface of the coal refuse pile always demonstrated maximal (14)CO(2) uptake, and in most samples, only slight activity was demonstrated at depths below 8 to 10 cm. Optimal uptake of (14)CO(2) by natural samples occurred at 20 to 30 C and at a moisture content of between 23 and 35%. In addition to chemolithotrophic bacteria, heterotrophic fungi and yeasts were also routinely isolated in high numbers from acidic coal refuse. In contrast, acidophilic, heterotrophic bacteria were either absent or present in low numbers in such acidic samples.  相似文献

Synopsis The bulk of Lake Opinicon fish biomass is concentrated in the physically diverse inshore areas. Quantitative analysis of the community compositions of the various inshore habitat types (weedy inlets, sandy shallows, rock shelf, gravel, etc.) showed that each supported a characteristic assemblage of fishes with a relatively constant species and year class composition. This was maintained throughout the season despite a drop in fish biomass in late summer when there was a progressive movement into the offshore waters.Weedbed areas supported the highest biomass, and greatest species and year class diversity. Thereafter, in declining order of richness, were rocky and sandy areas. A few fish species were restricted to single habitat types, e.g.Notropis heterodon to inshore weedbeds.Lepomis macrochirus, the commonest species in the lake was, by contrast, versatile and occurred in all inshore habitats. Commonly the numbers of a species in a habitat differed between day and night. Significant diel movements between habitats characterized two nocturnal feedersPomoxis nigromaculatus andIctalurus nebulosus.  相似文献

Using quantitative PCR, the abundances of six phytoplankton viruses DNA polymerase (polB) gene fragments were estimated in water samples collected from Lake Ontario, Canada over 26 months. Four of the polB fragments were most related to marine prasinoviruses, while the other two were most closely related to cultivated chloroviruses. Two Prasinovirus-related genes reached peak abundances of >1000 copies ml−1 and were considered ‘high abundance'', whereas the other two Prasinovirus-related genes peaked at abundances <1000 copies ml−1 and were considered ‘low abundance''. Of the genes related to chloroviruses, one peaked at ca 1600 copies ml−1, whereas the other reached only ca 300 copies ml−1. Despite these differences in peak abundance, the abundances of all genes monitored were lowest during the late fall, winter and early spring; during these months the high abundance genes persisted at 100–1000 copies ml−1 while the low abundance Prasinovirus- and Chlorovirus-related genes persisted at fewer than ca 100 copies ml−1. Clone libraries of psbA genes from Lake Ontario revealed numerous Chlorella-like algae and two prasinophytes demonstrating the presence of candidate hosts for all types of viruses monitored. Our results corroborate recent metagenomic analyses that suggest that aquatic virus communities are composed of only a few abundant populations and many low abundance populations. Thus, we speculate that an ecologically important characteristic of phycodnavirus communities is seed-bank populations with members that can become numerically dominant when their host abundances reach appropriate levels.  相似文献

Like Bythotrephes, Bosmina (Eubosmina) coregoni coregoni Baird appears to have invaded North America from Eurasia, since there are no records from before 1965. All previously known North American populations have a well-rounded ventrocaudal corner of the carapace, suggesting that the founding race is one of the rotunda type, the most common eubosminid in European lakes. In Lake Muskoka, immediately to the east of Georgian Bay, Ontario, eubosminids were found that shared features of B. (E.) longicornis kessleri Ulianine. In contrast to other North America populations of B.(E. )c. coregoni, these specimens have a short, broadly based mucro. Morphological features suggest that these are interspecific F1-hybrids between B. (E.) longispina Leydig and B. Fish introductions are one of the most widespread anthropogenic perturbations to aquatic ecosystems. Paradoxically, the effects of these introductions on aquatic ecosystems are typically poorly documented. This project studied the effect of fish introductions on Lake Opeongo, an oligotrophic lake in Algonquin Provincial Park, Ontario, Canada (45° 42′ N, 78° 22′ W), using the remains of algae (diatoms) and zooplankton (cladocerans) preserved in the sediments. It was hypothesized that the introduction of cisco or lake herring (Coregonus artedii Lesueur) in 1948, which filled the underutilized pelagic forage fish niche, should have altered nutrient availability for phytoplankton. Prior to cisco introduction, the diatom community of Lake Opeongo reflected a relatively stable oligotrophic state established before European settlement, and consisted of the Cyclotella stelligera complex with subdominants Tabellaria flocculosa IIIp and the Aulacoseira distans complex. No marked changes occurred until ca. 1962 when the diatom community shifted to an assemblage with increased total phosphorus preferences, consisting of Asterionella formosa and lesser amounts of Cyclotella bodanica var lemanica, the C. stelligera complex, Fragilaria crotonensis and T. flocculosa IIIp. The dominant cladoceran Bosmina longirostris increased significantly in relative abundance since the introduction of cisco. The most likely cause of this shift was increased nutrient recycling and/or trophic level changes caused by human manipulation of the fish community of the lake.  相似文献

This study identified two scales of pattern in the assemblages of Chydoridae (Crustacea, Branchiopoda, Anomopoda) in the shallow (<2 m) littoral zone of Plastic Lake, Ontario, Canada in the autumn of 1987. Twenty chydorid species were collected in 15 over-night sets of funnel traps in each of four habitat types. Analysis of variance multivariate analysis of variance, and discriminant function analysis revealed that assemblages differed among the habitats. Alona intermedia, Alona quadrangularis and Chydorus bicornutus were particularly abundant in the most structurally diverse habitat type – muddy, rock-strewn areas with approximately 40% bottom cover by the pipewort, Eriocaulon septangulare. In contrast, Anchistropus cf. minor was caught most often on bare shelves of rock. A second set of analyses demonstrated that chydorid assemblages also differed at a smaller scale, i.e. with local patchiness in bottom cover by the dominant macrophyte (E. septangulare). The abundance of Alona affinis was positively correlated with cover by E. septangulare, whereas Anchistropus cf. minor was caught mainly in microhabitats without vegetation. Alona intermedia and A. quadrangularis were most abundant in microhabitats with intermediate amounts of vegetation, suggesting their abundance is influenced by habitat factors other than vegetation.  相似文献

In previous studies, three different strains (BrG1, BrG2, and BrG3) of ferrous iron-oxidizing, nitrate-reducing bacteria were obtained from freshwater sediments. All three strains were facultative anaerobes and utilized a variety of organic substrates and molecular hydrogen with nitrate as electron acceptor. In this study, analyses of 16S rDNA sequences showed that strain BrG1 was affiliated with the genus Acidovorax, strain BrG2 with the genus Aquabacterium, and strain BrG3 with the genus Thermomonas. Previously, bacteria similar to these three strains were detected with molecular techniques in MPN dilution series for ferrous iron-oxidizing, nitrate-reducing bacteria inoculated with different freshwater sediment samples. In the present study, further molecular analyses of these MPN cultures indicated that the ability to oxidize ferrous iron with nitrate is widespread amongst the Proteobacteria and may also be found among the Gram-positive bacteria with high GC content of DNA. Nitrate-reducing bacteria oxidized ferrous iron to poorly crystallized ferrihydrite that was suitable as an electron acceptor for ferric iron-reducing bacteria. Biologically produced ferrihydrite and synthetically produced ferrihydrite were both well suited as electron acceptors in MPN dilution cultures. Repeated anaerobic cycling of iron was shown in a coculture of ferrous iron-oxidizing bacteria and the ferric iron-reducing bacterium Geobacter bremensis. The results indicate that iron can be cycled between its oxidation states +II and +III by microbial activities in anoxic sediments.  相似文献

When grown on iron-salt medium supplemented with the bisulfite ion, Leptospirillum ferrooxidans was much more sensitive to the ion than was Thiobacillus ferrooxidans. The causes of the sensitivity of L. ferrooxidans to the bisulfite ion were studied. The bisulfite ion completely inhibited the iron-oxidizing activities of L. ferrooxidans and T. ferrooxidans at 0.02 and 0.2 mM, respectively. A trapping reagent for the bisulfite ion, formaldehyde, completely reversed the inhibition. The treatment of intact cells with 1.0 mM bisulfite ion for 1 h and washing the bisulfite ion from the cells had no harmful effects on the iron-oxidizing activity of T. ferrooxidans. However, the treatment of L. ferrooxidans with 0.1 mM bisulfite ion for 1 h completely destroyed the iron-oxidizing activity. T. ferrooxidans had sulfite:ferric ion oxidoreductase activity. In contrast, a quite low level of sulfite:ferric ion oxidoreductase activity was found in L. ferrooxidans, suggesting that it is much more difficult for L. ferrooxidans to oxidize the bisulfite ion to the less harmful sulfate than it is for T. ferrooxidans. These results suggest that the sensitivity of L. ferrooxidans to the bisulfite ion is due to a lack of an active sulfite:ferric ion oxidoreductase and the sensitivity of its iron oxidase to bisulfite ion.  相似文献

Heart Lake, a small dimictic lake in Southern Ontario shows classical characteristics of eutrophy: high phytoplankton populations, blooms of blue-greens, predominently Aphanizomenon flos-aquae, in summer, a shallow photic zone decreasing seasonally in response to dense phytoplankton crops and a hypolomnion which is deoxygenated from early summer to fall turnover. Large decreases occur in TDP and SRS in particular, but also in TDN from spring to summer.The distribution of individual species of algae with depth in the epilimnion was seldom uniform but, more commonly conformed to a definite pattern. The simultaneous existence of different patterns in different species provides evidence of spatial heterogeneity (niche diversification) in the epilimnion, which could lessen any interspecific competition.Diatom stratigraphy in a 70 cm core revealed that the lake has been eutrophic throughout but may have gone through 3 periods; of these, the most recent one was the most eutrophic and was preceeded by a period of unusually low water levels.  相似文献

Anaerobic, nitrate-dependent microbial oxidation of ferrous iron was recently recognized as a new type of metabolism. In order to study the occurrence of three novel groups of ferrous iron-oxidizing, nitrate-reducing bacteria (represented by strains BrG1, BrG2, and BrG3), 16S rRNA-targeted oligonucleotide probes were developed. In pure-culture experiments, these probes were shown to be suitable for fluorescent in situ hybridization, as well as for hybridization analysis of denaturing gradient gel electrophoresis (DGGE) patterns. However, neither enumeration by in situ hybridization nor detection by the DGGE-hybridization approach was feasible with sediment samples. Therefore, the DGGE-hybridization approach was combined with microbiological methods. Freshwater sediment samples from different European locations were used for enrichment cultures and most-probable-number (MPN) determinations. Bacteria with the ability to oxidize ferrous iron under nitrate-reducing conditions were detected in all of the sediment samples investigated. At least one of the previously described types of bacteria was detected in each enrichment culture. MPN studies showed that sediments contained from 1 × 105 to 5 × 108 ferrous iron-oxidizing, nitrate-reducing bacteria per g (dry weight) of sediment, which accounted for at most 0.8% of the nitrate-reducing bacteria growing with acetate. Type BrG1, BrG2, and BrG3 bacteria accounted for an even smaller fraction (0.2% or less) of the ferrous iron-oxidizing, nitrate-reducing community. The DGGE patterns of MPN cultures suggested that more organisms than those isolated thus far are able to oxidize ferrous iron with nitrate. A comparison showed that among the anoxygenic phototrophic bacteria, organisms that have the ability to oxidize ferrous iron also account for only a minor fraction of the population.  相似文献

In another paper (SCHMIDT, 1969b) the relations between the numbers of bacteria and phytoplankton in the surface layers of some waters near Manaus (Amazonas) were reported. As a stratification of phytoplankton is not possible, due to the strong turbulence in the Rio Solimões (Amazon River), and since current was also present in the Rio Negro off the bays and littoral regions, the vertical distribution of bacteria and algae was of special interest in the Lago do Castanho, the third of the waterbodies investigated in the paper mentioned above. In the present study, two important fractions of bacteria, their total number, and the fraction of saprophyta (estimated by the number of bacterial colonies), were also differentiated.  相似文献
