Low soil temperatures and low water potentials reduce and delay the seed germination of canola (Brassica rapa L., B. napus L.) in western Canada. Germination is also very sensitive to the salinity effects of nitrogen fertiliser placed with the seed, especially when the seed bed is relatively dry. The effects of pre-hydration and re-drying treatment on canola (Brassica rapa L. cv. Tobin) seed germination and seedling emergence at 10°C subjected to either a water or salt stress were determined. Low water potentials, induced by polyethylene glycol (PEG 8000), low soil moisture, or high concentrations of salts, reduced both germination and seedling emergence, and increased the time to 50% germination and emergence of seeds at 10°C. At equal osmotic potentials, Na2SO4 was less inhibitory on low temperature germination than either NaCl or PEG, suggesting that the sulphate ion partially alleviated the inhibitory effects of low water potential. Solutions of NaCI produced more abnormal seedlings compared to Na2SO4, suggesting that NaCl was more toxic than Na2SO4 during seedling development. Pre-hydration and re-drying partially overcame the inhibitory effects of both low water potential and salts on seed germination and seedling emergence at 10°C. The seed treatment increased the germination rate in Petri dishes and seedling emergence from a sandy loam soil. Water potentials or soil water contents required to inhibit 50% germination or emergence at 10°C were lower for treated seeds compared to control seeds. Salt concentrations inhibiting 50% emergence were higher for treated seeds than control seeds. Neither treated nor control seeds produced seedlings which emerged if the soil water content was lower than 9% or when the soil was continuously irrigated with salt solutions of 100 mmol kg-1 of NaCl or 50 mmol kg-1 of Na2SO4. These results suggest that the pre-hydration and re-drying treatment did not lower the base water potentials at which seedling emergence could occur. Abnormal seedlings were observed in both treated and control seeds, particularly if the soil was watered with NaCl solutions; however, the seed treatment reduced the number of abnormal seedlings.  相似文献

研究了盐旱互作对潮间带和盐碱地生境盐地碱蓬棕色种子萌发、地上部生长和离子积累的影响。不同盐浓度预处理后,未萌发的种子风干后复水,其萌发率与对照相比没有降低,说明两种生境盐地碱蓬种子萌发期间都耐干湿交替。400mmol/LNaCl溶液浇灌的潮间带生境盐地碱蓬幼苗在第3次干旱处理后萎蔫幼苗的百分比高于盐碱地生境的,而复水后正常幼苗的百分比却相反。400mmol/LNaCl溶液处理下,第3次干旱处理复水后,盐碱地生境盐地碱蓬幼苗地上部离子含量(主要是Na+和Cl-)高于潮间带生境的。表明在干旱情况下,盐碱地生境盐地碱蓬幼苗能积累更多的无机离子,降低渗透势,提高根系吸收水分的能力。上述结果说明,盐碱地生境盐地碱蓬幼苗较潮间带生境盐地碱蓬幼苗更耐盐与干旱的交互作用。  相似文献

Cadmium (Cd2+) is one of the most toxic heavy metal pollutants in nature. Mesophyll cells from the leaf of radish seedlings exposed to 0.25 and 1.0 mM of CdCl2 during 24 h exhibited structural changes of chloroplasts, mitochondria and nuclei when compared to non-treated control plants. Chloroplasts from Cd2+-exposed samples exhibited changes in the organelle shape, an increase in the stroma volume and a deposition of electron-dense material in the double membrane. The changes in the chloroplast membranes were not very drastic, however and reorganization of the thylakoids and stroma was observed. In contrast, the breakdown of the nuclear envelope of the plant cells treated with Cd2+ was very clear. The accumulation of electron-dense granules was also observed in mitochondria. No alterations were observed in the vacuoles of radish seedlings grown at different Cd2+ concentrations for the periods tested.  相似文献

Summary The release of chromaffin granular content from staphylococcal enterotoxin B (SEB)-treated and-untreated PC12 cells was studied by electron microscopy. The treatment of the cells with SEB at the concentration of 20 μg/ml caused marked increase of the chromaffin granules that either bound to the plasma membrane by the characteristic rods, measuring 15 to 20 nm in length and showing a tubular structure, or budded off at the free cell surface, surrounded by a layer of rod-containing cytoplasm and enclosed by the plasma membrane. The binding between the granular and plasma membranes by the rods did not lead to membrane fusion and exocytosis of the granular content. Many of the bound granules showed vesiculation with loss of the electron-dense core material; at the same time, some of the binding rods contained intraluminal electron-dense material similar to the granular core material. These findings suggested that the electron-dense material (i.e., norepinephrine) of the bound granules was released extracellularly through channels within the rods. Although the granules were bound to the plasma membrane with equal frequency at the free and contiguous cell surfaces, the granular budding occurred only at the free cell surface, indicating that it occurred incidentally to some granules bound at the free cell surfaces. On the basis of the morphological observations, it is postulated that the electron-dense material of the bound granule is selectively released extracellularly through the rods, leaving the vesiculated granules behind in the cytoplasm. The same mode of release of the granular content was observed, though less frequently, in the untreated control cells. No morphological evidence that indicated that the granular content was released extracellularly by exocytosis was found in the treated and control cells. The present observations indicated that the SEB treatment of PC12 cells stimulated the binding of chromaffin granules to the plasma membrane by the rods and the budding of the bound granules at the free cell surface.  相似文献

Amaranthus retroflexus L. and Chenopodium glaucum L. are two widely distributed destructive weeds. Their strong adaptability and massive seed production make them the hardest weeds to deal with. This present study intended to investigate the effect of leachate from Eupatorium adenophorum on the growth of these weeds and explore the potential to develop an environmental friendly strategy to use the leachate to control the weeds. Seeds of A. retroflexus L. and C. glaucum L. were soaked in solutions containing 0%, 0.6%, 1.25%, 2.5%, and 5% leachate from E. adenophorum leaves. A. retroflexus and C. glaucum seedlings grown in pots were sprayed with leachate solutions in the same concentration range. The effects of these leachate solutions on membrane permeability and germination of seeds, and growth and physiological characteristics of the seedlings were investigated. The highest concentration of leachate (5%) caused significant damage to the cell membrane of seeds of both weed species, whereas lower concentrations (0.6%) promoted repair of the membrane system, as reflected by higher and lower than control in relative conductivity (RC), respectively. Different concentrations of leachate showed distinct allelopathic inhibitory effects on the two weed species; lower concentrations showed weak inhibitory or even positive effects, whereas higher concentrations showed stronger inhibitory effects. Higher concentrations of leachate (2.5% and 5%) delayed germination and significantly decreased the emergence rate of the seeds, survival rate, and dry matter accumulation of the seedlings. When treated by 5% leachate, the emergence date of A. retroflexus was delayed by 3.6 d, emergence rate of the seeds and survival rate was 69.1% and 70.6% of the control, respectively, seedling dry matter was 48.6% less than the control; In the case of C. glaucum, the emergence date was delayed by 2.7 d, emergence rate of the seeds and survival rate was 45.1% and 58.6% of the control, respectively, seedling dry matter was 44.7% less than the control. There were significant interactions among the different concentrations of leachate and the length of treatment period with respect to activities of antioxidant enzymes, malondialdehyde (MDA) contents, and chlorophyll contents. Seedlings treated with 0.6%, 1.25%, or 2.5% leachate solution for 24–72 h showed increased superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) activities. When seedlings were treated with leachate solutions for 96 h, antioxidant enzyme activities and chlorophyll content decreased in A. retroflexus, but only CAT activity decreased in C. glaucum. When seedlings of the two weed species were treated with 5% leachate solution, CAT activity and chlorophyll content decreased and MDA content gradually increased with longer treatment times (from 24 to 96 h). The two weed species showed different allelopathic responses to E. adenophorum; A. retroflexus was more sensitive than C. glaucum. Based on the investigation, it could be speculated that the delayed germination and low germination rate of the weeds after treatment by leachate could be due to the fact that leachate damaged the membrane system of the seeds. By delaying germination, lowering the germination rate of the weeds and inhibiting seedling growth, leachate from E. adenophorum could provide an effective way of controlling the weeds.  相似文献

Aims We investigated the impact of salinity on seed germination, chlorophyll content, chloroplast structure and photosynthesis of the green embryos in desiccated seeds of the xerophyte Haloxylon persicum, xero-halophyte Haloxylon ammodendron and euhalophyte Suaeda physophora.Methods Seeds of H. persicum, H. ammodendron and S. physophora were collected from natural environment in Fukang, Xinjiang province. Pretreatment with 700 mM NaCl was carried out to stimulate the natural 'seed priming'; we analyzed the joint effect of salinity and different species on germination physiology and cotyledonal structure and photosynthetic function changes during germination and recovery stage.Important findings We found that seeds did not suffer ion toxicity for the two halophytes H. ammodendron and S. physophora, as evidenced by the high final germination after ungerminated seeds pretreated with 700 mM NaCl were transferred to distilled water, but the final germination of the xerophyte H. persicum was significantly lower than that of control. The Na + concentration in embryos increased under salinity for all species, while K + concentration decreased by salinity only for H. persicum and H. ammodendron, i.e. the concentration of K + in embryos of H. persicum and H. ammodendron decreased by 36% and 46%, respectively. For all species, whether dry intact seeds or cotyledons of dry seeds imbibed in deionized water and NaCl solution, had high chlorophyll content. Treatment with NaCl also caused chloroplast thylakoids to swell and chlorophyll content to decrease in seeds of H. persicum, but no significant change was observed in the more salt-tolerant species S. physophora and H. ammodendron. Fluorescence measurement showed that 700 mM NaCl decreased the Fv/Fm ratio of cotyledons in seeds for all species, especially for H. persicum and H. ammodendron. Photosynthetic oxygen releasing was detected from the seeds that were moistened with distilled water and 700 mM NaCl for 6 or 24 h and from the seeds that were initially moistened with 700 mM NaCl in darkness for 10 days, then transferred to distilled water for another 6 and 24 h. The results indicated that the chlorophyll in cotyledon of desiccated seed had photosynthetic function in early germination stage, even under high-saline condition. In addition, the photosynthesis of chlorophyll in the embryonic cotyledons of desiccated seeds during germination was similar to that in leaves of young seedlings for all species. In conclusion, the chloroplasts of the two halophytes were more salt resistant compared with the xerophyte H. persicum. The photosynthetic function of chlorophyll in cotyledons of mature seeds may be ecologically important for seedling development in early stage for plants growing in extremely saline or arid environments.  相似文献

Salinity and high temperature stresses adversely affect growthand development of rice plants. To investigate the responseof rice cells to these stresses, we have analysed short-termstress-induced subcellular alterations in undifferentiated leafcells of rice seedlings by transmission electron microscopy.Perturbations noted particularly with respect to plasma membrane,mitochondrial membranes, endoplasmic reticulum, polyribosomesand dictyosomes are highlighted. The subcellular changes evokedby both stresses after 4 h were lysis of the cytoplasm, accumulationof electron-dense granules in the cytoplasm, distension in theER membranes, enhanced association of ribosomes with the endoplasmicreticulum, reduction in the number of mitochondrial cristae,as well as disorganization of cell wall fibrillar material.Certain changes were found to be unique to either the salinityor high temperature stress. 采用N +、C +离子注入拟南芥 (Arabidopsisthaliana)种子 ,统计了种子的发芽指数 (发芽率和发芽势 ) ;用改良的RAPD技术对N+离子注入种子植株的DNA进行11个引物的随机片段多态性扩增。结果表明 ,合适剂量的N+、C+离子注入可使种子发芽率提高 ,两种离子注入种子的发芽率峰值 (分别为92.3 %和74.4 % )都在5×1014ions/cm2;分析N +离子注入材料发现 ,在1×1013 -1×1016ions/cm2剂量范围内 ,基因组DNA的变异率与发芽指数的变化趋势基本一致 ,变异率峰值 (9.0 % )在1×1015 ions/cm2。结果提示 ,分析低能N+离子诱变效应的最佳注入剂量在1×1014-5×1015 ions/cm2。对N+、C+离子注入的比较发现 ,一定范围内同等剂量C+离子注入的诱变率高于N +离子注入  相似文献

Quaternary ammonium salts inhibited the growth of yeast especially at pH higher (pH 8) than optimal. It was postulated that compounds integrate with the cell membrane and interfere with its functions. The yeast cell ultrastructure investigated under an electron microscope confirms this hypothesis. A relatively high percentage of cells treated at pH 6 with the quaternary ammonium salt of alanine derivative (DMALM-12) at the minimal inhibitory concentration showed an irregularity in the cell shape. No such irregularity was observed in the control. Besides, in the cells treated with the drug, practically no lipid droplets were seen at all. Inside the control cells, electron-dense round bodies were clearly seen and interpreted as vacuoles. These bodies were absent in the cells treated with DMALM-12. Although the yeast cells growing at pH 8 showed a more or less normal shape, they seemed to have difficulty in budding - no fully developed buds were found in the preparations. Only some convexities of the cell wall were seen that could be the beginning of budding which stopped early after the start. Some changes in the round bodies interpreted as vacuoles were visible: they were less dense and full of granules.  相似文献

Controlled conditions were used to investigate the relationship between ion distribution in developing seeds of two Suaeda salsa populations and seed germination and seedling emergence. Seeds were harvested from S. salsa plants that had been treated with 1 or 400 mM NaCl for 122 (saline inland population) or 135 days (intertidal zone population) in a glasshouse. Germination and seedling emergence were evaluated under salinity. In both populations, more ions were accumulated in the pericarps of plants cultured in 400 mM NaCl than in 1 mM NaCl. Pericarps accumulated much higher ion concentrations in the intertidal zone population than in the saline inland population, while the opposite trend occurred for ion accumulation in the embryos. Seeds of plants from the intertidal zone population germinated more rapidly than those from plants of the saline inland population, regardless of the NaCl concentration during seed germination. However, seedling emergence under high salinity was lower with seeds from the intertidal zone population than with seeds from the saline inland population. In conclusion, S. salsa in the intertidal zone employs superior control of ion compartmentalization in the pericarps to tolerate salinity but requires a minimal level of ions in embryos to ensure seedling establishment in highly saline environments. This indicates that euhalophytes require salts during the mature seed stage to maintain seed viability and to ensure seedling emergence and population establishment.  相似文献

用不同浓度的氯化钠溶液处理罗布麻种子,试验结果表明,氯化钠浓度低于0.8%时,可促进罗布麻种子萌芽,高于0.8%小于2.0%时抑制萌芽,超过2.0%时极显著抑制种子萌芽。氯化钠浓度高低均对罗布麻幼苗生长有抑制作用,当浓度大于0.4%时,出叶速度减慢,植株矮化、相对抑制率超过60%,鲜重与对照相比降低了50%以上。  相似文献

This study evaluated changes in DNA methylation in Arabidopsis thaliana plants grown from seeds implanted with low-energy N(+) and Ar(+) ions. Methylation-sensitive amplified polymorphism (MSAP) testing revealed altered DNA methylation patterns after ion implantation at doses of 1 × 10(14) to 1 × 10(16) ions/cm(2). Comparison of the MSAP electrophoretic profiles revealed nine types of polymorphisms in ion-implanted seedlings relative to control seedlings, among which four represented methylation events, three represented demethylation events, and the methylation status of two was uncertain. The diversity of plant DNA methylation was increased by low-energy ion implantation. At the same time, total genomic DNA methylation levels at CCGG sites were unchanged by ion implantation. Moreover, a comparison of polymorphisms seen in N(+) ion-implanted, Ar(+) ion-implanted, and control DNA demonstrated that the species of incident ion influenced the resulting DNA methylation pattern. Sequencing of eight isolated fragments that showed different changing patterns in implanted plants allowed their mapping onto variable regions on one or more of the five Arabidopsis chromosomes; these segments included protein-coding genes, transposon and repeat DNA sequence. A further sodium bisulfite sequencing of three fragments also displayed alterations in methylation among either different types or doses of incident ions. Possible causes for the changes in methylation are discussed.  相似文献

The cell wall is a fundamental component in the response of plants to environmental changes. To directly assess the role of the cell wall we have increased the expression and activity of a cell wall associated peroxidase (TPX2), an enzyme involved in modifying cell wall architecture. Overexpression of TPX2 had no effect on wild-type development, but greatly increased the germination rate under high salt or osmotic stress. Differential scanning calorimetry showed that transgenic seeds were able to retain more water available for germination than wild-type seeds. Thermoporometry calculations indicated that this could be due to a lower mean pore size in the walls of transgenic seeds. Therefore, the higher capacity of transgenic seeds in retaining water could result in higher germination rates in conditions where the availability of water is restricted.  相似文献

Abstract Current interest in seed hydration treatments for the improvement of level, rate and uniformity of germination or field emergence has revealed how little is known of the physiology of germination control under water stress. This review surveys briefly the responses of seeds held at different hydration levels, from normal germination in a free water supply to seed activation without germination under slight moisture stress, seed deterioration at greater moisture stress and the damage that can be caused to seeds in very dry conditions, as well as the responses to subsequent dehydration. Inhibition of germination, though not of seed activation, at certain levels of water stress is likened to various forms of dormancy, and the mechanism governing the initiation of cell elongation is suggested as the possible key to control over germination. Several lines of evidence on cell membrane integrity and action, and their responses to external factors, point to the role that the membrane may play in cell elongation (and hence germination) control, and membrane integrity may also be associated with the transition between seed deterioration at one hydration level and seed activation and repair at slightly higher hydration levels. Seed activation without germination is also considered in an ecological context.  相似文献

通过3%KNO3溶液浸泡和室内光照培养对美引茄冠(Solanum melongena L.)在Ca(NO3)2胁迫下的种子萌发特性和幼苗耐盐性的影响进行研究.结果表明,在Ca(NO3)2胁迫下,经过KNO3引发处理的种子发芽率、发芽势、发芽指数、活力指数和幼苗干鲜重均显著高于对照;与对照相比,引发处理显著提高了幼苗叶片超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)和抗坏血酸过氧化物酶(APX)的活性,而丙二醛(MDA)含量则显著降低,膜脂过氧化程度较轻;引发处理植株叶片游离脯氨酸和可溶性蛋白质含量均显著高于对照.结果表明,经KNO3引发处理显著提高了茄子种子的活力并增强了其幼苗的耐盐性.  相似文献

本文研究了用25keV N+注入丰豆103的种子后,N+离子对其种子的活力及子叶伸展后48小时和96小时的幼苗内蛋白质含量、谷胱甘肽巯基转移酶(GSH-Ts)、谷胱甘肽过氧化酶(GSH-Px)、抗坏血酸过氧化物酶(ASA-POD)活性的影响.结果表明当N+注量在2.6×1016N+/cm2~5.2×1016N+/cm2时,种子的发芽率、发芽指数、活力指数都明显提高;幼苗的可溶性蛋白含量高于对照.在6.5×1016N+/cm2~10.4×1016N+/cm2注量时,幼苗的可溶性蛋白含量低于对照,96小时幼苗可溶性蛋白的含量高于48小时,说明辐射引起的损伤可随生长时间的增加而有所恢复.高注量可引起幼苗内一些抗氧化酶活性的升高,且随注量的增加酶的活性升高也越明显,96小时幼苗的GSH-Px和ASA-POD活性高于48小时幼苗,GSH-Ts活性略有下降.而低注量(1.3×1016N+/cm2~5.2×1016N+/cm2)的上述酶指标升幅不大.说明经N+离子处理后可通过诱导这些抗氧化酶活性的升高起到减轻伤害的作用.  相似文献
