中国板栗EST-SSR分子标记在栲树中的通用性分析   总被引:3,自引:0,他引:3

简单重复序列也称为微卫星分子标记,不仅在同属近缘种间具有良好的通用性,甚至在近缘属间也具有一定的通用性。本研究利用壳斗科基因组信息数据库中公布的中国板栗124对多态的EST-SSR引物在栲树中进行跨属(栗属到栲属)通用性研究,结果显示中国板栗EST-SSR引物在栲树中通用性和多态性分别为42.7%和56.6%;使用19对多态的EST-SSR引物对4个栲树自然居群的遗传多样性进行初步分析,结果显示栲树自然居群具有较高的遗传多样性(Na=6.105,Ho=0.563,He=0.621)。这些引物为栲树群体遗传学的深入研究提供了有力工具。  相似文献

广西武宣濠江流域普通野生稻居群遗传多样性及保护研究   总被引:16,自引:4,他引:12  
选用平均分布于水稻基因组的24对SSR引物,对沿河分布最长的广西武宣濠江流域的12个普通野生稻居群343份材料的遗传结构进行研究.结果表明:(1)该地普通野生稻遗传多样性丰富.24个位点共检测到206个等位变异,平均等位变异数A=8.7083,有效等位变异数Ae=3.7117;(2)该地普通野生稻居群具有较高的遗传分化和一定频率的基因流.群体遗传分化系数Gst=0.2659,基因流Nm=0.6901,表明26.59%的遗传变异存在于居群间;(3)SSR标记使普通野生稻居群中一些稀有等位变异得以显现.206个等位变异中,65个等位变异仅出现在1个或2个居群中,且频率较低,其中12个等位变异只出现在居群B中;(4)通过聚类分析和主坐标分析(PCO),下游居群A和B遗传关系较近,中游居群C比较独特,单独成为一类,中游居群D、E、F和G遗传关系较近,中游居群H、I和J及上游居群K和L遗传关系较近.根据上述分析结果,建议对濠江下游和中游具有代表性的居群(即居群B、D和H)的普通野生稻进行重点保护.  相似文献

We analyzed genetic structure and diversity among eight populations of popcorn, using SSR loci as genetic markers. Our objectives were to select SSR loci that could be used to estimate genetic diversity within popcorn populations, and to analyze the genetic structure of promising populations with high levels of heterozygosity that could be used in breeding programs. Fifty-seven alleles (3.7 alleles per locus) were detected; the highest effective number of alleles (4.21) and the highest gene diversity (0.763) were found for the Umc2226 locus. A very high level of population differentiation was found (F(ST) = 0.3664), with F(ST) for each locus ranging from 0.1029 (Umc1664) to 0.6010 (Umc2350). This analysis allowed us to identify SSR loci with high levels of heterozygosity and heterozygous varieties, which could be selected for production of inbred lines and for developing new cultivars.  相似文献

利用5个DNA片段及12个微卫星标记(SSR)对姜状三七(Panax zingiberensis C.Y.Wu et K.W.Feng)的遗传多样性和遗传分化进行分析。结果显示,与其他人参属(Panax)植物相比:姜状三七具有相对较高物种水平的核苷酸多态性和等位基因数;在居群水平上,景谷居群具有最高的核苷酸多态性和等位基因数,而江城居群最低。AMOVA分析结果表明,姜状三七在物种水平上具有一定程度的遗传分化,但不显著;在居群水平,江城居群与其他居群间的分化程度最高,其他居群间遗传分化不明显。Structure分析结果也显示江城居群与其他居群被聚类到不同分组,且形态特征相近的样本并未聚到一起,同一分布点的样本遗传成分更相似。  相似文献

利用32对SSR引物对来自全部7个自然居群的217份广东高州普通野生稻(简称“高野”)材料进行遗传结构、多样性和遗传聚类分析。结果表明, 高野各居群因遗传结构存在差异而相对独立, 但各居群之间由于存在基因渗透又具有一定的相似性。高野总体多样性指数(Ht)为0.65, 居群内的多样性(HS=0.431)略大于居群间的多样性(DS=0.392), 二者差异并不显著。居群间的遗传分化系数(GST)为0.611, 说明高野群体的遗传差异是由居群内和居群间的遗传分化共同作用的结果。其中A、B、E居群间, D、F、G居群间遗传相似性较高, C居群与其它居群之间存在较大差异。根据7个居群的遗传结构, 结合其地理分布状况, 认为遗传多样性最大的B和E居群以及遗传分化最小的C居群应作为重点对象进行保护。  相似文献

利用改良FIASCO法(Fast Isolation by AFLP Sequences COntaining repeats)开发出的9对多态性SSR引物评价了薇菜(Osmunda japonica Thunb.)2个野生居群(庐山和恩施)、1个栽培居群(恩施)的遗传多样性和遗传分化水平。结果显示,9个SSR标记在3个薇菜居群中共检测到47个等位基因,每个SSR位点的平均等位基因数为5.222个,观测杂合度和期望杂合度分别为0.000~0.944和0.577~0.834,香农指数为0.962~1.860,表明各SSR位点多态性较高;各居群的平均期望杂合度均大于平均观测杂合度且种内近交系数均为正值,说明3个薇菜居群中都存在非随机交配现象;对各居群的相关遗传多样性参数分析表明,恩施野生居群遗传多样性最高,而其栽培居群最低;庐山野生居群与恩施野生居群间遗传分化系数为0.092,说明两地野生薇菜居群的遗传分化程度较低,AMOVA分析也表明遗传变异主要存在于野生居群内部。  相似文献

采用基因组DNA富集法FIASCO对我国典型常绿阔叶林建群种栲树(Castanopsis fargesii Franch.)进行了微卫星分子标记的开发。从栲树基因组中分离和筛选了15个微卫星位点,并对江西九连山栲树自然分布居群的遗传多样性进行了分析。结果表明,栲树具有较高水平的遗传多样性,每个位点在28株栲树个体上的平均等位基因数(A)为6.7(4~8个),平均观察杂合度(HO)为0.690(0.250~1.000),平均预期杂合度(HE)为0.698(0.293~0.867)。每个位点的第一排除概率值Pr(Ex1)为0.043~0.527,位点综合值为0.9972。单个位点的第二排除概率Pr(EX2)为0.159~0.694,位点综合值为0.9999。这些微卫星标记可为研究栲树的遗传多样性及居群遗传结构提供有效的遗传工具。  相似文献

利用RAPD分子标记对 5个栲树 (CastanopsisfargesiiFranch .)天然群体共计 188个个体的遗传多样性和群体遗传结构进行了分析。 4 1个随机寡核苷酸引物共检测到 385个位点 ,其中多态位点 15 7个 ,占 4 0 .78%。物种水平的Shannon多样性指数I=0 .4 5 97,Nei基因多样度h =0 .2 96。遗传变异分析表明 ,栲树群体的遗传变异主要存在于群体内 ,利用Shannon多样性指数估算的分化 (Hsp_Hpop) /Hsp=0 .0 4 76 ,遗传分化系数Gst =0 .0 4 2 9,分子方差分析 (AMOVA)也证实了这一结论 ,群体内的变异组分占了 94 .97% ,群体间变异只占 5 .0 3%。AMOVA分析结果的显著性检验也表明 ,群体间及群体内个体间均呈现出显著分化 (P <0 .0 0 1)。  相似文献

This study examined the genetic diversity in 20 rice landrace populations from parts of traditional farming areas of the Indian Himalayas using 11 mapped simple sequence repeats (SSR) loci. Twenty‐four individuals sampled from each of the 20 landraces (480 individuals), which were collected from farmers from Northwest to Northeast Himalaya, showed that all landraces showed within population variation and none were homogeneous. The number of polymorphic loci in a landrace population ranged from 5 to 11. A total of 71 alleles were recorded of which 58 were common and 13 were rare. Of the 71 alleles, 46 were common to both Northwest and Northeast regions, whereas 9 were unique to the former and 16 were unique to the latter. The mean number of alleles per locus was 6.45 and for landrace populations from Northwest and Northeast regions were 5.0 and 5.64, respectively. Population differentiation, as shown by a high FST value (0.61), was greater for Northeast populations. The unweighted pair group method with arithmetic mean (UPGMA) dendrogram classified the populations into three major clusters: cluster I comprised seven populations from the Northwest region, cluster II comprised seven populations from the Northeast region and cluster III comprised populations from both regions. Investigating the population genetic structure can help monitor change in diversity over time and space, and also help devise a rational plan for management of crop landraces on‐farm under farmer management.  相似文献

Wild sunflower Helianthus annuus originates from North America and has naturalised in Argentina where it is considered invasive. The present study attempts to assess the genetic diversity using two different molecular marker systems to study the wild genetic patterns and to provide data applicable to conservation and breeding uses. Ten natural populations sampled throughout the wild range and six inbred lines were studied using inter‐simple sequence repeat (ISSR) and simple sequence repeats (SSR) markers. A total of 64 ISSR bands and 29 SSR alleles were produced from 106 wild and cultivated plants. We found 9 ISSR private bands and 21 SSR private alleles in wild accessions, but no private bands/alleles were found in cultivated sunflowers. Molecular variability in wild populations was approximately 60% higher than in inbred lines. Local wild sunflowers kept considerable diversity levels in comparison with populations in the centre of origin (approximately 70%) and therefore they might possess a potential for adaptive evolutionary change. Analysis of molecular variance (AMOVA) indicated population structure with nearly 20% of genetic variability attributable to between‐population differentiation. Principal coordinate analyses (PCO) grouped wild populations from different geographic locations, and a Mantel test showed low congruence between genetic distance (GD) and geographic distances (GGD); hence, molecular data could not rule out multiple wild introduction events. Low correlations were found between ISSR and SSR GD at individual and population levels; thus, divergent evolutionary groups were not evident in local wild sunflowers. Several genetic diversity criteria were utilised to assign conservation value and certain wild populations emerged as interesting sites for more extensive sampling.  相似文献

猕猴桃野生居群的SSR分析初报   总被引:11,自引:0,他引:11  
采用SSR分子标记技术对我国猕猴桃的2个商业栽培物种——中华猕猴桃和美味猕猴桃的9个天然居群(共221个样)的遗传多样性进行了初步分析。通过对14对猕猴桃引物的筛选,8对重现性好的引物扩增结果表现出良好的多态性。在8个多态性位点上共获得222个等位基因。居群等位基因平均数A=17.3,多态位点百分率P-100,多态信息指数PIC为0.87~0.96,显示出我国的猕猴桃野生居群具有极高的遗传多样性。中华猕猴桃和美味猕猴桃野生居群拥有高比例的共同等位基因,反映出二者的亲缘关系极近。  相似文献

A large collection of genebank accessions of the hexaploid outcrossing forage grass species timothy (Phleum pratense L.) was for the first time analysed for SSR diversity on individual, population and regional level. Timothy is the most important forage grass species in the Nordic countries. Eighty-eight timothy accessions from Nordic countries and eight accessions around Europe were analysed with recently developed simple sequence repeat (SSR) markers. Timothy proved to be very polymorphic: the 13 selected SSRs amplified a total of 499 polymorphic alleles, the number of alleles per SSR locus varying from 15 to 74. Taking all SSR alleles together, the observed number in each accession ranged from 95 to 203. Levels of diversity were found to be significantly different between countries, vegetation zones and different cultivar types. However, the differentiation between accessions was low: most of the variation (94%) in the studied timothy material was due to variation within accessions and only 5% was between accessions and 1% between countries. Lack of geographical differentiation may reflect the outcrossing and hexaploid nature of timothy. Our results showed that neutral SSR markers are suitable for demonstrating levels of diversity but not alone adequate to resolve population structure in timothy. Nordic timothy material seems to be diverse enough for breeding purposes and no decline in the level of diversity was observed in varieties compared to wild timothy populations. Challenges in analysing SSR marker data in a hexaploid outcrosser were discussed.  相似文献

本文利用13对微卫星标记,对我国3个核心种源地(巴山、秦岭、川西高原)圈养林麝种群进行遗传多样性和遗传结构分析。在167份样品中共检测到142个等位基因(Na),每个位点等位基因数介于7~16,均值为10.92,平均有效等位基因数(Ne)为6.3730,期望杂合度(He)和观测杂合度(Ho)均值分别为0.8302和0.3897。这些圈养林麝种群遗传多样性水平较高,但较低的观测杂合度表明圈养群体存在近交现象。两两群体间的Fst 值和AMOVA分析结果均表明种群之间分化程度不明显。群体遗传结构分析显示,全部样本聚为3个遗传簇(最佳K值=3),其主体与3个地理来源相符,但种群间存在基因渗透现象。本研究中的秦岭种群遗传变异最为丰富,可以作为种质改良的基因池。  相似文献

Microsatellite (simple sequence repeats, SSR) and mitochondrial DNA markers were used to assess the structure of European codling moth populations showing different levels of susceptibility towards one of the most important biocontrol agents used in apple production, the Cydia pomonella granulovirus CpGV-M. In 638 C. pomonella individuals from 33 different populations a total of 92 different alleles were scored using six SSR loci. The global estimate of genetic differentiation for all 33 populations was not significantly different from zero, thus indicating a lack of genetic differentiation. AMOVA analysis revealed a very weak but significant variance among C. pomonella populations from different geographic regions, however, no significant variation was evident between CpGV-M resistant or susceptible C. pomonella populations. Sequence analysis of a fragment of the cytochrome oxidase subunit 1 in eight C. pomonella populations resulted in 27 haplotypes, which were grouped in two distinct clusters. Again, no genetic differentiation between CpGV-M resistant and susceptible codling moth populations was detectable. In addition, Structure analysis using microsatellites and association tests with mtDNA haplotypes found neither population-level nor individual correlations associated with CpGV-M resistance. Accordingly, this lack of population structure does not allow discriminating between one or several, separate origins of CpGV-M resistance.  相似文献

选用8对多态的微卫星引物, 研究了毛红椿(Toona ciliata var. pubescens)核心居群和边缘居群的遗传多样性。研究结果显示, 边缘居群的观察等位基因数和有效等位基因数并不比核心居群低, 甚至更高。普通广布和稀有地方等位基因在所有居群中均有分布, 而普通地方和稀有广布等位基因分别仅在5个和3个居群中有分布。从4种类型等位基因总数上看, 边缘居群高于核心居群, 特别是边缘居群的稀有地方等位基因数量明显多于核心居群。边缘居群的平均期望杂合度和观察杂合度都高于核心居群。核心居群间的基因分化系数为0.1520, 边缘居群间为0.3045, 边缘居群与核心居群的遗传分化差异达到显著水平。核心居群间基因流大于1, 而边缘居群间基因流小于1, 说明核心居群间基因交流频繁, 边缘居群由于片段化和地形影响, 基因流较小。Mantel检验结果显示, 居群间遗传距离与地理距离的相关性不显著, 说明地理距离对毛红椿居群遗传分化的影响不显著。  相似文献

The population genetic structure of plant pathogenic fungus Pyrenophora tritici-repentis was examined using microsatellite (SSR) markers. According to the geographical origin of the pathogen populations, they were designated as North Caucasian (S, 33 isolates), northwest (Nw, 39), and Omsk (Om, 43). The populations were analyzed at the nine most polymorphic SSR loci, at which 75 alleles were identified. To characterize the genetic variation within and between populations, the AMOVA algorithm as implemented in the Arlequin v. 3.5 software program was used. The number of alleles per locus ranged from 5 to 12 and their sizes varied within the range from 180 to 400 bp. The mean gene diversity at SSR loci was high for all populations (H = 0.58–0.75). The populations were considerably different in the frequencies of individual alleles of the SSR loci. Most isolates in the populations were represented by unique haplotypes. The within-population variation of the isolates at molecular markers was 86.4%; among the populations, 13.6%. Substantial interpopulation differences were found between the Om and S (Fst = 0.16) and between the Om and Nw (Fst = 0.20) populations, whereas between the S and Nw populations, these differences were small (Fst = 0.05). Thus, it was demonstrated that the population of P. tritici-repentis from Omsk oblast had the independent status of the geographical population; northwest and North Caucasian populations differed in the allelic diversity of SSR loci, and despite the low Fst value (0.05), they also belonged to independent geographical populations.  相似文献

为研究青藏高原海拔变化与藏川杨遗传多样性和群体遗传结构的关系,以青藏高原东南部色季拉山分布的藏川杨群体为材料,用24对微卫星标记对其进行遗传分析。在469个个体中共检测到126个等位基因,每个位点的平均等位基因数(Na)为5.25;多态性位点百分率(PPL)为100%,期望杂合度(He)在高、低海拔的群体中都处于较高水平,分别为0.48和0.49;分子方差分析(AMOVA)的结果表明,种群间分化占总变异的6.38%,遗传变异主要集中在群体内不同个体之间;基因分化系数(FST)为0.02,也证实群体分化处于较低水平,而检测到群体间的基因流动(Nm)处于较高水平,为9.89。上述结果表明,海拔因素未对生长在色季拉山高低海拔的藏川杨群体造成地理隔离,从而产生种群分化;在此地区藏川杨遗传背景一致,藏川杨种质资源的保存无需考虑海拔的差异。本研究结果为研究高海拔适应机制材料的选择提供了很大的便利,并为藏川杨的可持续利用与保护提供了一定的理论依据。  相似文献
