The effect of temperature on filtration and ingestion rates ofSalpa fusiformis Cuvier was determined over the temperature range 13–22°C at a constant algal concentration. Filtration and ingestion rates increased with temperature up to an optimum (19.7 to 20.2°C) beyond which they decreased. Food assimilation for this salp species was determined while fed onPhaeodactylum tricornutum Bohlin orHymenomonas elongata Stein at different algal concentrations. The percent assimilation varied between 28 and 39% (mean: 32%) with the diatomP. tricornutum as food, and between 39 to 81% (mean: 64%) with the flagellateH. elongata. Assimilation did not seem to depend on the algal concentration but it was greater with the flagellate than with the diatom, the latter having a higher ash content.  相似文献

We investigated the influence of CO2/HCO3 -depletion and of the presence of acetate and formate on the in vivo photosynthetic electron transport in the two green algae Chlamydobotrys stellata and Chlamydomonas reinhardtii by means of thermoluminescence technique and mathematical glow curve analysis. The main effects of the removal of CO2 from the algal cultures was: (1) A shift of the glow curve peak position to lower temperatures resulting from a decrease of the B band and an increase of the Q band. (2) Treatment of CO2-deficient Chl. stellata with DCMU yielded two thermoluminescence bands in the Q band region peaking at around +12°C and +5°C; in case of Chl. reinhardtii DCMU treatment induced only one band with an emission maximum at +5°C. The presence of acetate or formate in CO2-depleted algal cultures lowered the intensities of all of the individual TL bands but that of a HT band (TL+37). Functional and structural characteristics of the photosynthetic apparatus were studied in the diatom Stephanodiscus neoastraea and the cyanobacterium Planktothrix agardhii which were grown semi-continuously under constant irradiance or under simulated natural light fluctuations. The light fluctuations consisted of 24 oscillations of exponentially increasing and decreasing irradiance over a 12-h light period. Maximum irradiance was 1100 μmol photons m−2 s−1 with the ratio of maximum to minimum intensities being 100, simulating Langmuir circulations with a ratio of euphotic to mixing depth of 1. S. neoastraea acclimated to the light fluctuations by doubling the number and halving the size of photosynthetic units (PS II) while the amount of chlorophylls and carotenoids remained unchanged. The chlorophyll-specific maximum photosynthetic rate was enhanced while the slope of the photosynthesis versus irradiance curves was not influenced by the light fluctuations. Acclimation of P. agardhii was mainly characterized by an increase in chlorophyll content. Both photosystems showed only little changes in number and size. Maximum photosynthetic rate, saturating irradiance and initial slope of the photosynthesis versus irradiance curves did not vary. Although both high and low light were contained in the fluctuating light, an analogy to low or high light acclimation was not found for the diatom nor for the cyanobacterium acclimated to light fluctuations. We suggest that the acclimation to fluctuating light is a response type outside the known scheme of low and high light acclimation. Benthic diatoms live in photoautotrophic/heterotrophic biofilm communities embedded in a matrix of secreted extracellular polymeric substances. Closely associated bacteria influence their growth, aggregation, and secretion of exopolymers. We have studied a diatom/bacteria model community, in which a marine Roseobacter strain is able to grow with secreted diatom exopolymers as a sole source of carbon. The strain influences the aggregation of Phaeodactylum tricornutum by inducing a morphotypic transition from planktonic, fusiform cells to benthic, oval cells. Analysis of the extracellular soluble proteome of P. tricornutum in the presence and absence of bacteria revealed constitutively expressed newly identified proteins with mucin‐like domains that appear to be typical for extracellular diatom proteins. In contrast to mucins, the proline‐, serine‐, threonine‐rich (PST) domains in these proteins were also found in combination with protease‐, glucosidase‐ and leucine‐rich repeat‐domains. Bioinformatic functional predictions indicate that several of these newly identified diatom‐specific proteins may be involved in algal defense, intercellular signaling, and aggregation.  相似文献

When photosynthesis of the blue-green alga Anacystis nidulans was measured as 14CO2-fixation, the inhibitory effect of DCMU at low concentrations was greatest when mainly Photosystem 1 (PS 1) (excitation at 446 or 687 nm) was operative. At concentrations above 10-6M the inhibition on 14CO2-fixation was greatest when mainly Photosystem 2 (PS 2) was operative (excitation at 619). During excitation of PS 1, the excretion of glycolate was stimulated at low concentrations of DCMU (5 × 10-8M and lower), while higher concentrations inhibited excretion. All concentrations of DCMU inhibited glycolate excretion when mainly PS 2 was excited. The curves showing the relative effect of DCMU on the two photosystems, measured as PS 1/PS 2, had opposite shapes for 14CO2-fixation and glycolate excretion. An increase in 14CO2-fixation coincided with a decrease in glycolate excretion and vice versa. It appears that the increased rate of photosynthesis when mainly PS 1 was operative relative to that when mainly PS 2 was excited, increases the consumption of glycolate in an oxidation process associated with the excitation of PS 1, resulting in less excretion of glycolate to the medium. The influence of DCMU inhibition on labelled amino acid pools connected to the glycolate pathway (glycine-serine) is quite similar to that for 14CO2-fixation. At concentrations below 10-6M DCMU, inhibition of 14CO2- incorporation into the amino acids was greatest when PS 1 was excited, while at the higher concentrations tested, inhibition was greater when PS 2 was excited. We conclude that the metabolism of glycine and serine is closely connected to the rate of photosynthesis.  相似文献

Nutritional insufficiency and toxicity are deleterious effects of phytoplankton on grazers. We hypothesize that toxic food is likely to have stronger evolutionary selective effects on grazers than nutritionally insufficient food. We explore this hypothesis in comparative studies of egg production and egg hatching of the copepod Acartia hudsonica challenged with both a toxic and a nutritionally insufficient alga. Experiments lasting 6 days, in which mixtures of different proportions of the suspect and a control alga were offered as food to female copepods, showed that the dinoflagellate Alexandrium fundyense, which bears paralytic shellfish toxins, was toxic to A. hudsonica. In contrast, the diatom Phaeodactylum tricornutum was nutritionally insufficient to A. hudsonica. In another set of experiments, the effects of A. fundyense and P. tricornutum, respectively, as sole foods on egg production and egg hatching success of two geographically separated populations (Maine and Connecticut) of the copepod A. hudsonica were examined in common-environment experiments, after being raised under identical conditions for two generations. The location in Maine regularly experiences toxic blooms of Alexandrium sp. whereas the location in Connecticut does not. During a 6-day period, A. fundyense reduced the egg production rates of the Connecticut copepod population, but not of the Maine population. In contrast, the diatom P. tricornutum reduced the egg production of both populations. These results of this study are consistent with the hypothesis of local adaptation to toxic food, but not to nutritionally insufficient food.  相似文献

Intact cells of diatoms are characterized by a rapid diatoxanthin epoxidation during low light periods following high light illumination while epoxidation is severely restricted in phases of complete darkness. The present study shows that rapid diatoxanthin epoxidation is dependent on the availability of the cofactor of diatoxanthin epoxidase, NADPH, which cannot be generated in darkness due to the inactivity of PSI. In the diatom Phaeodactylum tricornutum, NADPH production during low light is dependent on PSII activity, and addition of DCMU consequently abolishes diatoxanthin epoxidation. In contrast to P. tricornutum, DCMU does not affect diatoxanthin epoxidation in Cyclotella meneghiniana, which shows the same rapid epoxidation in low light both in the absence or presence of DCMU. Measurements of the reduction state of the PQ pool and PSI activity indicate that, in the presence of DCMU, NADPH production in C. meneghiniana occurs via alternative electron transport, which includes electron donation from the chloroplast stroma to the PQ pool and, in a second step, from PQ to PSI. Similar electron flow to PQ is also observed during high light illumination of DCMU-treated P. tricornutum cells. In contrast to C. meneghiniana, the electrons are not directed to PSI, but most likely to a plastoquinone oxidase. This chlororespiratory electron transport leads to the establishment of an uncoupler-sensitive proton gradient in the presence of DCMU, which induces diadinoxanthin de-epoxidation and NPQ. In C. meneghiniana, electron flow to the plastoquinone oxidase is restricted, and consequently, diadinoxanthin de-epoxidation and NPQ is not observed after addition of DCMU.  相似文献

A multiple-strain algal biosensor was constructed for the detection of herbicides inhibiting photosynthesis. Nine different microalgal strains were immobilised on an array biochip using permeable membranes. The biosensor allowed on-line measurements of aqueous solutions passing through a flow cell using chlorophyll fluorescence as the biosensor response signal. The herbicides atrazine, simazine, diuron, isoproturon and paraquat were detectable within minutes at minimal LOEC (Lowest Observed Effect Concentration) ranging from 0.5 to 100μgL−1, depending on the herbicide and algal strain. The most sensitive strains in terms of EC50 values were Tetraselmis cordiformis and Scherffelia dubia. Less sensitive species were Chlorella vulgaris, Chlamydomonas sp. and Pseudokirchneriella subcapitata, but for most of the strains no general sensitivity or resistance was found. The different responses of algal strains to the five herbicides constituted a complex response pattern (RP), which was analysed for herbicide specificity within the linear dose-response relationship. Comparisons of herbicide-specific RP to reference RPs of the five herbicides always showed the lowest deviation of the herbicide-specific RP tested with the reference RP of the same herbicide for the triazine and phenylurea herbicides. We therefore conclude that, in principle, identification of a specific herbicide is possible employing the algal sensor chip.  相似文献

Diatoms are prominent and highly diverse microalgae in aquatic environments. Compared with other diatom species, Phaeodactylum tricornutum is an “atypical diatom” displaying three different morphotypes and lacking the usual silica shell. Despite being of limited ecological relevance, its ease of growth in the laboratory and well-known physiology, alongside the steady increase in genome-enabled information coupled with effective tools for manipulating gene expression, have meant it has gained increased recognition as a powerful experimental model for molecular research on diatoms. We here present a brief overview of how over the last 25 years P. tricornutum has contributed to the unveiling of fundamental aspects of diatom biology, while also emerging as a new tool for algal process engineering and synthetic biology.  相似文献

We hypothesize that algae with different cell compositions are differently perceived by their predators and consequently subjected to selective grazing. Five populations of the diatom Phaeodactylum tricornutum that differed in organic and elemental composition, but were otherwise identical, were generated by acclimation to distinct growth regimes. The different populations were then mixed in pairs and subjected to predation by either the rotifer Brachionus plicatilis or the copepod Acartia tonsa. The presence of rotifers had no impact on the ratio between any two algal populations. The presence of copepods, however, affected the ratio between algae previously acclimated to a medium containing 1 mM NH4+ and algae acclimated to 0.5 mM NO3?, and to either a lower irradiance or a higher CO2 concentration. We discuss the possible reason for the influence of different nutritional histories on the vulnerability of algae to predators. The differential impact of grazers on the growth of algae with different nutritional histories may result from direct selective grazing (i.e., grazers can detect algae with the most palatable cell composition), alone or combined to an asymmetric utilization of the nutrients regenerated after predation by co‐existing algal populations. Our results strongly suggest that the nutritional history of algae can influence the relationships between phytoplankton and grazers and hint at the possibility that algal cell composition is potentially subject to natural selection, because it influences the probability that algae survive predation.  相似文献

Air-grown cells of the marine diatom Phaeodactylum tricornutum showed only 10% of the carbonic-anhydrase activity of air-grown Chlamydomonas reinhardtii. Measurement of carbonic-anhydrase activity using intact cells and cell extracts showed all activity was intracellular in Phaeodactylum. Photosynthetic oxygen evolution at constant inorganic-carbon concentration but varying pH showed that exogenous CO2 was poorly utilized by the cells. Sodium ions increased the affinity of Phaeodactylum for HCO 3 - and even at high HCO 3 - concentrations sodium ions enhanced HCO 3 - utilization. The internal inorganic-carbon pool (HCO 3 - +CO2] was measured using a silicone-oil-layer centrifugal filtering technique. The internal [HCO 3 - +CO2] concentration never exceeded 15% of the external [HCO 3 - +CO2] concentration even at the lowest external concentrations tested. Diatoms are ubiquitous marine photosynthetic eukaryotes that are responsible for about 20% of global photosynthesis. Nevertheless, little is known about the redox-based mechanisms that mediate diatom sensing and acclimation to environmental stress. Here we used a redox-sensitive green fluorescent protein sensor targeted to various subcellular organelles in the marine diatom Phaeodactylum tricornutum, to map the spatial and temporal oxidation patterns in response to environmental stresses. Specific organelle oxidation patterns were found in response to various stress conditions such as oxidative stress, nutrient limitation and exposure to diatom-derived infochemicals. We found a strong correlation between the mitochondrial glutathione (GSH) redox potential (EGSH) and subsequent induction of cell death in response to the diatom-derived unsaturated aldehyde 2E,4E/Z-decadienal (DD), and a volatile halocarbon (BrCN) that mediate trophic-level interactions in marine diatoms. Induction of cell death in response to DD was mediated by oxidation of mitochondrial EGSH and was reversible by application of GSH only within a narrow time frame. We found that cell fate can be accurately predicted by a distinct life-death threshold of mitochondrial EGSH (−335 mV). We propose that compartmentalized redox-based signaling can integrate the input of diverse environmental cues and will determine cell fate decisions as part of algal acclimation to stress conditions.  相似文献

Diatoms (Bacillarophyceae) are photosynthetic unicellular microalgae that have risen to ecological prominence in oceans over the past 30 million years. They are of interest as potential feedstocks for sustainable biofuels. Maximizing production of these feedstocks will require genetic modifications and an understanding of algal metabolism. These processes may benefit from genome‐scale models, which predict intracellular fluxes and theoretical yields, as well as the viability of knockout and knock‐in transformants. Here we present a genome‐scale metabolic model of a fully sequenced and transformable diatom: Phaeodactylum tricornutum. The metabolic network was constructed using the P. tricornutum genome, biochemical literature, and online bioinformatic databases. Intracellular fluxes in P. tricornutum were calculated for autotrophic, mixotrophic and heterotrophic growth conditions, as well as knockout conditions that explore the in silico role of glycolytic enzymes in the mitochondrion. The flux distribution for lower glycolysis in the mitochondrion depended on which transporters for TCA cycle metabolites were included in the model. The growth rate predictions were validated against experimental data obtained using chemostats. Two published studies on this organism were used to validate model predictions for cyclic electron flow under autotrophic conditions, and fluxes through the phosphoketolase, glycine and serine synthesis pathways under mixotrophic conditions. Several gaps in annotation were also identified. The model also explored unusual features of diatom metabolism, such as the presence of lower glycolysis pathways in the mitochondrion, as well as differences between P. tricornutum and other photosynthetic organisms.  相似文献

Extensive beds of benthic, non-calcareous macroalgae are associated with reef formations in oligotrophic coastal waters of Northeastern Brazil. Large amounts of these algae constantly and naturally detach and decay. Part of them is deposited on the beach, remains exposed during low tides, and decomposes. Field experiments on decomposition were carried out in a tidal pool (temperatures fluctuating from 29 to 43°C salinity from 6 to 35 at Ponta do Seixas (Paraiba, Brazil), and laboratory studies were made in aerobic and anaerobic conditions. High values of NO 3 (up to 148.92 μg at 1−1), NO 2 (up to 3.14 μg at 1−1) and PO 3 4− (up to 22.95 μg at 1−1) were released during algal degradation. Nitrogen values were higher in the tidal pool than in aerobic and anaerobic experiments, as opposed to phosphorus where the opposite phenomenon occurred. The diatom Phaeodactylum tricornutum inoculated in seawater enriched with decomposition products of algae showed intense growth, sometimes similar to that in complete algal culture medium. Inhibition and death of this diatom was observed in some other experiments. The importance of drift seaweed as a source of nutrients for the local ecosystem is stressed.  相似文献

Diatoms are unicellular photoautotrophic algae, which can be found in any aquatic habitat. The main storage carbohydrate of diatoms is chrysolaminarin, a nonlinear β‐glucan, consisting of a linear 1,3‐β‐chain with 1,6‐β‐branches, which is stored in cytoplasmic vacuoles. The metabolic pathways of chrysolaminarin synthesis in diatoms are poorly investigated, therefore we studied two potential 1,6‐β‐transglycosylases (TGS) of the diatom Phaeodactylum tricornutum which are similar to yeast Kre6 proteins and which potentially are involved in the branching of 1,3‐β‐glucan chains by adding d ‐glucose as 1,6‐side chains. We genetically fused the full‐length diatom TGS proteins to GFP and expressed these constructs in P. tricornutum, demonstrating that the enzymes are apparently located in the vacuoles, which indicates that branching of chrysolaminarin may occur in these organelles. Furthermore, we demonstrated the functionality of the diatom enzymes by expressing TGS1 and 2 proteins in yeast, which resulted in a partial complementation of growth deficiencies of a transglycosylase‐deficient ?kre6 yeast strain.  相似文献

The acclimation depression of capacity of photon utilisation in photochemical reactions of photosystem 2 (PS2) can develop already after three months of cultivation of the Norway spruces (Picea abies [L.] Karst.) under elevated concentrations of CO2 (i.e., ambient, AC, + 350 µmol(CO2) mol–1 = EC) in glass domes with adjustable windows. To examine the role that duration of EC plays in acclimation response, we determined pigment contents, rate of photosynthesis, and parameters of chlorophyll a fluorescence for sun and shade needles after three seasons of EC exposure. We found responses of shaded and exposed needles to EC. Whereas the shaded needles still profited from the EC and revealed stimulated electron transport, for the exposed needles the stimulation of both electron transport activity and irradiance saturated rate of CO2 assimilation (P Nmax) under EC already disappeared. No signs of the PS2 impairment were observed as judged from high values of potential quantum yield of PS2 photochemistry (FV/FM) and uniform kinetics of QA reoxidation for all variants. Therefore, the long-term acclimation of the sun-exposed needles to EC is not necessarily accompanied with the damage to the PS2 reaction centres. The eco-physiological significance of the reported differentiation between the responses of shaded and sun exposed needles to prolonged EC may be in changed contribution of the upper and lower crown layers to the production activity of the tree. Whereas for the AC spruces, P Nmax of shaded needles was only less than 25 % compared to exposed ones, for the EC spruces the P Nmax of shaded needles reached nearly 40 % of that estimated for the exposed ones. Thus, the lower shaded part of the crown may become an effective consumer of CO2.  相似文献

In this study the effects of potassium iodide on the growth and metabolite accumulation of Nitzschia closterium (Ehr.) W. Smith and Phaedactylum tricornutum Bolin were investigated to assess its possible application to the mass culture of the two diatoms in open environment, extensive systems. The results indicated that supplementation of potassium iodide at a concentration of 1000 mg L−1 resulted in a reduction of the induction phase in cultures of N. closterium and P. tricornutum and led to an increase in the accumulation of biomass and extracellular polymeric substances. Conversely, the addition of potassium iodide, at all concentrations tested, showed no obvious effect on the fatty acid profiles of the two diatoms, particularly in the content of eicosapentaenoic and decosahexaenoic acid. Potassium iodide was also found to inhibit the growth of Dunaliella salina, Cryptomonas sp. and Chlorella sp. at minimum inhibitory concentrations of 356.8, 475.9 and 696.2 mg L−1, respectively. It also inhibited bacteria, including species isolated from the two diatom cultures, at a minimum concentration of 400 mg L−1. These results suggest that potassium iodide is an effective agent for inhibiting the proliferation of certain flagellate and non-flagellate algae, and bacteria, thus forming a favorable environment for diatoms to proliferate and consequently improving accumulation of biomass and EPS. These properties of potassium iodide provide a possible solution for preventing contamination from flagellate and non-flagellate algae in mass culture of the two diatoms without causing significant changes in their fatty acid composition.  相似文献

SYNOPSIS. Symbiosis between Chlamydomonas hedleyi (Lee, Crockett, Hagen & Stone) and Archais angulatus (Fichtel & Moll) was examined during laboratory studies of primary production and light-enhanced calcification. Photosynthesis and calcification are directly proportional to light intensity in the range of 0–200 μEinsteins m-2 sec-1. Calcification in the light is directly proportional to photosynthesis and proceeds at rates that are 2–3 times that observed in the dark. The herbicide 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU), in concentrations of 1–100 μM, completely inhibits photosynthesis and light-enhanced calcification. Calcification of the foraminiferan test is therefore due to the photosynthetic activity of the symbiote. Calcification rates for foraminifers incubated in the dark or with DCMU are not significantly different from the calcification rates obtained for dead foraminifers. Rates of calcification obtained with 45Ca are twice that obtained with 14C.  相似文献

The adsorption of plutonium-237, added from an acid solution, by two species of marine phytoplankton, Monochrysis lutheri Droop, a flagellated chrysomonad, and Phaeodactylum tricornutum Bohlin, a diatom, is governed by a passive mechanism. This is clearly indicated by the observation that the rapid rates of sorption were not significantly different for live and heat-killed cells incubated at 24°C and 0°C. The Q10 values for the chrysomonad and the diatom of 0.95 and 1.18, respectively, are indicative of passive processes. The average ratio of (activity on algae/activity in filtrate) per unit volume at 24 h, the time when maximum levels of algal radioactivity were attained, of (32 ± 4) × 103 was not significantly different for the diatom or the chrysomonad. On a per cell basis, maximum levels of radioactivity were attained within 2 h. Less than 20% of the adsorbed 237Pu desorbed after 24 h.  相似文献

Biosurfactants have been suggested as a method to control harmful algal blooms (HABs), but warrant further and more in-depth investigation. Here we have investigated the algicidal effect of a biosurfactant produced by the bacterium Pseudomonas aeruginosa on five diverse marine and freshwater HAB species that have not been tested previously. These include Alexandrium minutum (Dinophycaee), Karenia brevis (Dinophyceae), Pseudonitzschia sp. (Bacillariophyceae), in marine ecosystems, and Gonyostomum semen (Raphidophyceae) and Microcystis aeruginosa (Cyanophyecae) in freshwater. We examined not only lethal but also sub-lethal effects of the biosurfactant. In addition, the effect of the biosurfactant on Daphnia was tested. Our conclusions were that very low biosurfactant concentrations (5 μg mL−1) decreased both the photosynthesis efficiency and the cell viability and that higher concentrations (50 μg mL−1) had lethal effects in four of the five HAB species tested. The low concentrations employed in this study and the diversity of HAB genera tested suggest that biosurfactants may be used to either control initial algal blooms without causing negative side effect to the ecosystem, or to provoke lethal effects when necessary.  相似文献
