目的:探讨抑制LRP16的表达对宫颈癌Siha细胞的化疗药物敏感性的影响。方法:将抑制LRP16表达的小干扰RNA:negativecontrol-si RNA(NC)、si RNA-374(si374)转染入Siha宫颈鳞癌细胞系中,通过顺铂(DDP)和紫杉醇(TAX)的处理后,采用CCK-8检测不同浓度紫杉醇、顺铂作用宫颈癌细胞系Siha48 h后,计算出细胞被抑制一半时顺铂、紫杉醇的药物浓度(IC50);使用Hoechst33342染色观察细胞凋亡,采用流式细胞仪检测顺铂IC50作用Siha细胞48小时后的细胞凋亡情况,紫杉醇IC50作用Siha细胞之后的细胞周期分布情况。结果:CCK-8检测转染的Siha细胞增殖活性受到抑制,Hoechst33342染色观察转染的Siha细胞凋亡明显增加,流式细胞仪检测凋亡显示,si374+顺铂的早期凋亡率22.15±2.24,NC+顺铂12.45±2.72,流式细胞仪检测周期显示G2/M(%),si374+紫杉醇29.94±1.87,NC+紫杉醇17.66±2.32。结论:LRP16基因表达下调之后,抑制Siha细胞的增殖、促进其凋亡,使细胞周期滞留于G2/M期,从而提高Siha细胞的化疗敏感性。  相似文献

探讨ErbB- 4受体在卵巢癌细胞增殖、侵袭等生物学行为中的作用.免疫组化检测ErbB- 4受体在卵巢癌细胞株OVCAR-3表达;噻唑蓝比色法(MTT)测定不同浓度的ErbB- 4单克隆抗体(Ab-3)对体外培养的卵巢癌OVCAR-3细胞增殖的影响;流式细胞仪检测细胞周期和凋亡;侵袭实验观察Ab-3对OVCAR-3侵袭能力的影响.结果表明,ErbB- 4在OVCAR-3细胞株中呈阳性表达;不同浓度(0.625~10 μg/ml)的Ab-3均能抑制OVCAR-3细胞生长,且具有浓度和时间依赖性,其作用72 h的中效浓度为4.48 μg/ml.流式细胞仪检测发现Ab-3(2.0 μg/ml)作用24、48及72 h后,OVCAR-3细胞出现S期阻滞,肿瘤细胞的凋亡率逐渐增加;抗体处理组穿透侵袭小室滤膜的细胞数(78.0±6.1)明显少于对照组(132.0±17.2)(P<0.01).因此,Ab-3可抑制内源性ErbB- 4表达的OVCAR-3细胞株的增殖、侵袭能力,诱导细胞凋亡.  相似文献

目的:研究片仔癀对人卵巢癌细胞株OVCAR-3增殖抑制作用,及其对细胞周期、细胞凋亡的影响。方法:采用MTT法观察片仔癀对人卵巢癌细胞株OVCAR-3细胞的增殖抑制率,流式细胞仪检测细胞凋亡及细胞周期,western-blot检测相关蛋白的表达。结果:片仔癀以剂量依赖式抑制人卵巢癌细胞株OVCAR-3细胞增殖,片仔癀250、500、1000μg·mL-1作用于OVCAR-3细胞24 h后,其早期凋亡率分别为6.6%、30.9%、43.2%,而对照组为0%,其诱导凋亡作用呈现剂量依赖性;细胞积聚在G0/G1期,同时S期细胞比例减少;Akt、PARP、CDK6表达下调。结论:片仔癀可以抑制OVCAR-3细胞增殖及诱导细胞凋亡作用,并能阻滞细胞于G0/G1期,有望成为卵巢癌治疗药。  相似文献

目的:探讨Genistein增加顺铂诱导的耐药卵巢癌细胞SKOV-3凋亡的可能作用机制.方法:倒置相差显微镜下观察药物处理后细胞形态学的变化;MTT比色法检测不同药物处理后对SKOV-3细胞增殖的影响;流式细胞仪检测药物处理后细胞的凋亡情况;流式细胞仪和荧光显微镜检测细胞内活性氧(ROS)的水平.结果:10ug/ml的Genistein和2.5ug/ml的顺铂联用24h后,引起了细胞内ROS的增加,细胞的凋亡率也显著增高,与单用顺铂组相比差异有显著性(P<0.05);用NAC预处理细胞2h后,有效抑制了ROS的产生,并增加了细胞的活性,降低了细胞的凋亡率,与未加NAC组相比差异有显著性(P<0.05).结论:Genistein增加顺铂诱导的耐药卵巢癌细胞SKOV一3的凋亡与细胞内ROS水平的升高有关,这可能是Genistein增加顺铂诱导的耐药卵巢癌细胞SKOV-3凋亡的作用机制之一.  相似文献

利用纳秒脉冲电场(nanosecond pulsed electric fields,ns PEFs)治疗肿瘤具有潜在优势。该研究探讨了纳秒脉冲电场对体外培养的人卵巢癌顺铂敏感细胞COC1及顺铂耐药细胞COC1/DDP的生长抑制、凋亡诱导效应及其机制。CCK-8法分析显示,纳秒脉冲电场降低COC1及COC1/DDP细胞存活率,而且抑制效应呈明显时间依赖效应。流式细胞术检测结果结合Hoechst 33342荧光染色结果表明,纳秒脉冲电场可诱导COC1及COC1/DDP细胞发生凋亡。JC-1荧光染色及caspase-3蛋白酶活性检测表明,纳秒脉冲电场处理可引起COC1及COC1/DDP细胞线粒体膜电位降低及caspase-3活性增强。流式细胞术检测细胞周期显示,纳秒脉冲电场可将COC1细胞周期阻滞于S期。以上研究结果揭示,纳秒脉冲电场可以抑制COC1及COC1/DDP细胞增殖,诱导细胞凋亡,线粒体途径凋亡为其可能机制。  相似文献

以地塞米松(DEX)诱导小鼠胸腺细胞凋亡;利用PI和AnneXin V/PI流式细胞术分别检测细胞晚期和早期凋亡;利用JC-1和DiOC_6(3)/PI在细胞水平检测凋亡中线粒体膜电势(△ψm)变化:抽提线粒体,利用JC-1直接染色技术检测现存线粒体△ψm情况。实验结果显示,DEX显著诱导胸腺细胞早期和晚期凋亡,凋亡细胞主要来自G_0/G_1期;细胞水平可见DEX介导与△ψm相关的J-aggregate和DiOC_6(3)可染性降低,同时介导线粒体数量显著降低,6h细胞膜完整性无显著变化:单纯线粒体检测结果显示,多数线粒体维持正常△ψm。提示,DEX介导胸腺细胞凋亡中线粒体数量降低,现存线粒体多保持着正常△ψm以维持凋亡过程细胞能量供给。  相似文献

用核酶技术阻断或降低抗凋亡蛋白 Bcl- 2的表达以促进化疗药物紫杉醇诱导的食管癌细胞凋亡 ,探索克服耐药、提高紫杉醇疗效的新途径 .将特异性切割 Bcl- 2 m RNA的核酶克隆至含MTII启动子并可为 Zn SO4 诱导表达的真核表达载体中 ,通过脂质体转入食管癌鳞状上皮细胞系Eca 1 0 9中 ,经 G41 8筛选得到稳定抗性细胞株 X1 0 9R,挑取单细胞株扩大培养 ,1 40μmol/L Zn SO4诱导 3d,用 Northern- blot、免疫荧光、流式细胞仪鉴定核酶及 Bcl- 2蛋白表达情况 ,用 TUNEL标记及流式细胞术检测凋亡细胞的比例 .bcl- 2核酶在不同单细胞株中有不同程度的表达 ,其中一株X1 0 9R1 4表达最高 .测定其中 Bcl- 2蛋白含量 ,发现 Bcl- 2蛋白表达大为降低 .加入紫杉醇后 ,TUNEL标记及凋亡峰测定结果都表明同一条件下凋亡率升高 .结果提示 ,转入特异性切割 bcl- 2m RNA的核酶可有效地阻断 Bcl- 2蛋白合成 .Bcl- 2蛋白表达降低可明显促进紫杉醇诱导的细胞凋亡 .说明 Bcl- 2蛋白在细胞产生耐药过程中起着重要作用  相似文献

目的观察PTCH1在卵巢癌组织中的表达,探讨奈达铂抑制卵巢癌OVCAR-3细胞的增殖以及对Hedgehog信号通路相关蛋白表达的影响。方法应用免疫组织化学染色法检测正常卵巢组织及卵巢癌标本各80例中PTCH1的表达,并观察卵巢癌组织标本中PTCH1表达的规律及其与病理类型、病理分级、淋巴结转移、临床分期、是否行新辅助化疗之间的关系。常规培养卵巢癌细胞OVCAR-3,奈达铂干预后Western blot观察细胞中PTCH1、cyclin D1及Gli1的表达变化情况,CCK-8法检测细胞增殖能力的变化。结果 PTCH1在卵巢癌标本表达率明显高于正常卵巢组织,PTCH1的表达与病理类型、病理分级、淋巴结转移、临床分期无明确联系,但与含奈达铂方案的新辅助化疗的执行相关;奈达铂干预的卵巢癌细胞中PTCH1、cyclin D1及Gli1的表达较未干预组低,其增殖受到明显抑制。结论奈达铂可以通过干扰卵巢癌中Hedgehog通路关键因子从而影响卵巢癌的发生发展。  相似文献

目的:探讨组蛋白去乙酰化酶抑制剂(HDIs)trichostatinA(TSA)对非小细胞肺癌(NSCLC)A549细胞放疗敏感性的影响.方法:以5Gy γ-射线照射细胞,或同时以TSA处理细胞.采用MTT法检测细胞存活率,Armexin V-PI染色检测细胞凋亡,流式细胞仪检测caspase-3活性.结果:SGy γ-射线可轻度降低细胞存活率,仅有少量细胞发生凋亡,但同时以TSA和γ-射线处理细胞,细胞存活率明显下降,凋亡细胞显著增多,且明显提高活化的caspase-3水平.结论:TSA通过促进caspase-3激活增强A549细胞对γ-射线的敏感性.  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

The levels of endogenous phytohormones and respiratory rate in nine sorts of flowers such as Cymbidium faberi Rolfe, Nopalxochia ackermannii Kunth and others were investigated both at full bloom and senescence and meanwhile the effect of exogenous phytohormones on prolonging the blossoms and promoting ethylene production were tested. There is a high content of endogenous ethylene in all the long-lived flowere, about 3–16 folds higer than the short-lived ones. There is a high level of ABA at full blooming flowers of short-lived flowers, in which there is no or only some cytokinins in it, but the ratio of CTK (6BA+zeatin)/ABA is smaller(l.7). The endogenous ABA reached a much higher level at senescence in all nine sorts of flowers, so it is reasonable to consider that it is ABA which plays an important role of regulation in controlling flower's senescence. There is a much higher level of GA3 and zeatin in the long-lived flowers which is not demonstrated in the shortlived ones. The respiratory rate is one of the factors controtling the longevity of flowers, but it does not play a decided role. Application of 6BA and zeatin prolongs distinctly orchid’s longevity, however exogenous IAA through the promotive action on ethylene production, evidently extends the longevity of the flowers of the Nopalxochia ackermannii Kunth.  相似文献

Summary Anthers were cultured from two sets of seven lines of hexaploid wheat (Triticum aestivum L.) with different cytoplasms, the euplasmic nucleus donors, Siete Cerros 66 and Penjamo 62, as well as their six alloplasmic lines derived from wild relative species of the genera Triticum and Aegilops. Significant cytoplasmic and nuclear effects but no cytoplasmic-nuclear interaction were found for embryogenic anther response, with the best performance of Penjamo 62 in Ae. kotschyi cytoplasm. Plant regeneration was not affected significantly by the cytoplasmic background of the lines cultured. The possible genetic implications of the observed cytoplasmic and nuclear influences on the in vitro haploid induction of wheat are discussed.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

Few comparative studies of spatial patterns at different scales have examined several species in the same habitat or the same species over a range of habitats. Therefore, variability in patterns among species or among habitats has seldom been documented. This study quantifies spatial patterns of a suite of intertidal snails and a species of barnacle using a range of statistical techniques. Variability in densities was quantified from the scale of adjacent quadrats (over a distance of centimeters) to tens of kilometers. Significant differences in abundances occurred primarily at two spatial scales. Small-scale differences were found at the scales of centimeters or 1–2 m and, for many species on many shores, these accounted for most of the variability in abundances from place to place. These are likely to be determined by behavioural responses to small-scale patches of microhabitat. Large-scale differences in abundance were also found in most species at the scale of hundreds of meters alongshore. These are likely to be due to variation in recruitment (and/or mortality) because of limited dispersal by adults of these species. There was little or no additional variation among shores, separated by tens of kilometers, than was shown among patches of shore separated by hundreds of meters. Identification of the scale(s) at which significant differences in abundance are found focus attention on the processes (and the scales at which these processes operate) that influence patterns of distribution and abundance. Some of the advantages and disadvantages of various procedures are discussed.  相似文献
