Mitosis and microtubule organizational changes in rice root-tip cells   总被引:1,自引:0,他引:1  
The pattern of change of the microtubule cytoskeleton of the root-tip cells of rice during mitosis was studied using immunofluorescence technic and confocal laser scanning microscopy. All the major stages of ceil division including preprophase, prophase, metaphase, anaphase and telophase were observed. The most significant finding was that in the preprophase cells microtubules radiating from the nuclear surface to the cortex were frequently seen. During development these microtubules became closely associated with the preprophase band and prophase spindie indicating that the microtubules radiating from the nuclear surface, the preprophase band and the prophazc spindle were structurally and functionally closely related to each other. Granule-like anchorage sites for the radiating microtubules at the muclear surface were often seen and the possibility that these gramle-like anchorage sites might represent the microtubule organizing centres was discussed.  相似文献

Chilling stability of microtubules in root-tip cells of cucumber   总被引:3,自引:0,他引:3  
The chilling stability of microtubules was investigated in root-tip cells of cucumber (Cucumis sativus L.) using immunofluorescence labeling and a confocal laser scanning microscope. After low temperature (4°C) treatment for 5 h, we found not only chilling-stable cortical microtubules under the plasma membrane of the root-tip cells, but also chilling-stable punctate microtubules in the cytoplasm, which might be the microtubules associated with organelles. 6-Dimethylaminopurine (6-DMAP), an inhibitor of protein kinase, reduced the chilling stability of the cortical microtubules in the cells. When the root-tip cells were restored to normal growth temperature (28°C), the recovery of the microtubules seemed to be closely associated with the microtubules still remaining in these cells after chilling treatment.Abbreviations 6-DMAP 6-Dimethylaminopurine - DMSO Dimethyl sulfoxide - FITC Fluorescein isothiocyanate - MTOCs Microtubule-organizing centers - PPB Pre-prophase bandCommunicated by H. Aluminum-induced cell death in root-tip cells of barley   总被引:1,自引:0,他引:1  
Aluminum-induced cell death was investigated in root-tip cells of barley (Hordeum vulgare). The growth of roots in 0.1-50 mM Al treatments was inhibited after 8 h treatments, and could not be recovered after 24 h recovery culture without Al. Viable detection with fluorescein diacetate-propidium iodide (FDA-PI) staining shows that most of the root-tip cells have lost viability. These results suggest that the irreversible inhibition of root growth after 8 h Al treatments or 24 h recovery culture is mainly caused by cell death. DNA ladders occurred in root tips only after 8 h Al treatments (0.1-1.0 mM), but no apoptotic bodies in root tips were observed. Thus, the cell death caused by Al stress is likely to be Al-induced programmed cell death (PCD). The reactive oxygen species (ROS) in root-tip cells measured by ultraweak luminescence indicated that the oxidation status in root-tip cells basically ceased after exposure to 10-50 mM Al for 24 h, but was very violent in the root-tip cells treated with 0.1-1.0 mM for 24 h. Exposure to 0.1-1.0 mM Al for 3-12 h led to ROS burst. Therefore, our results suggest that 0.1-1.0 mM Al treatments for 8 h induce cell death (Al-induced PCD) possibly via a ROS-activated signal transduction pathway, whereas 10-50 mM Al treatments may cause necrosis in the root-tip cells. These results have an important role for further studies on the mechanism of Al toxicity in plants.  相似文献

Some species of Psychotria (Rubiaceae) are important in herbal medicine, where their extracts are used internally for infections of the female reproductive system, bronchitis, gastrointestinal disturbances, skin irritations, tumors, ulcers, and eye disturbances. The antiproliferative and genotoxic effects of Psychotria myriantha Mull. Arg. and P. leiocarpa Cham. et Schlecht infusions on the Allium cepa L. cell cycle were evaluated. The teas were prepared by infusing the leaves in distilled water, in two concentrations: 1.13 mg/mL and 6.78 mg/mL. Three groups of four bulbs were used for each Psychotria species. After the bulbs rooted in distilled water, they were transferred to the teas for 24 hours, except for the control that stayed in water. The rootlets were then collected, fixed in ethanol-acetic acid (3:1) for 24 hours, and stored in 70% ethanol. For each group of bulbs, 4000 cells were analyzed, calculating the mitotic indexes, submitting them to statistic analysis, using the χ 2 test (p = 0.05). The results showed a decrease in mitotic index with an increase in tea concentration in both species. In P. leiocarpa, the mitotic index values differed significantly between the control and concentration of 6.78 mg/mL (χ 2 = 9.863). For P. myriantha, the values referring to the mitotic index differed greatly between the control and the treatments (χ 2 = 124.8). With this study, it was determined that P. myriantha and P. leiocarpa infusions possess antiproliferative effects on the A. cepa cell cycle, and teas of P. myriantha also have genotoxic activity.  相似文献

Physical and chemical properties of the rat liver lysosomes with single Triton WR 1339 overloading were studied during the administration of a detergent to intact rats and those with acute toxic hepatitis. Administration of the latter to intact animals was accompanied by a reduction of the floating density of the particles, solubilization of the lysosome enzymes and by increased fragility of the particles in the hypotonic medium. Lysosomes of the hepatocytes in rats with toxic hepatitis also displayed signs of overloading of the vacuolar apparatus with the preparation administered. The most pronounced solubilization of the lysosomal enzymes beta-galactosidase, acid RNA-ase, cathepsin D--was noted in case of combined action of CCl4 and Triton WR 1339 24, 48, 72 hours and 7 days after the CCl4 poisoning. Possible consequences of overloading of the vacuolar apparatus of the rat hepatocytes are discussed.  相似文献

In dividing root-tip cells of Pinus brutia Ten., immunolocalization of the luminal endoplasmic reticulum (ER) proteins, which have the C-terminal HDEL sequence, reveals that the ER is reorganized during the preprophase/prophase stage. Portions of ER were arrayed into a ring-like structure at the site of the microtubule preprophase band (Mt-PPB). This preprophase ER band (ER-PPB) resembles that of the Mt-PPB. The former undergoes a maturation process closely similar to that of the latter. Our data show that the PPB region has a more complex organization than is currently believed. The probable function(s) of the ER-PPB is discussed.  相似文献

Summary The carotid bifurcation areas of 25 human fetuses aged from 8 to 22 weeks were studied using the formaldehyde-induced fluorescence method. A long process from the sympathetic trunk reached the area at the age of 8 weeks contacting with the carotid body primordium. Brightly fluorescent cells can be seen both in the carotid body and in the ganglionic process. Migration of these cells from the sympathetic trunk to the carotid body is suggested. The connection from the sympathetic trunk to the carotid body totally disappeared after the tenth week, leaving no fluorescent elements between these two. Control electron microscopy and light microscopy were performed to identify the fluorescent and nonfluorescent components of the human fetal carotid body.  相似文献

Summary Nuclear and microtubular cycles were studied in large heterophasic multinuclear cells induced in root tips ofTriticum turgidum by caffeine treatment. Multinuclear cells and cells with polyploid nuclei exhibited various configurations of multiple and complex preprophase microtubule (Mt) bands (PPBs), including helical ones. The developmental stages of PPBs in some heterophasic cells did not comply with the cell cycle stages of the associated nuclei, a fact indicating that these events are not directly controlled by the associated nuclei. The heterophasic cells exhibited asynchronous nuclei at different stages of mitosis. In cells displaying prophase and interphase nuclei, the prophase spindle was either absent or developed around both of them or developed around the prophase nuclei earlier than around the interphase ones. During prometaphase-metaphase of the advanced nuclei the lagging interphase nuclei were induced to form prematurely condensed chromosomes (PCCs) along with spindle formation around them. These observations suggest that the mitotic transition in heterophasic cells is delayed but is ultimately achieved due to the effect of the advanced nuclei, which induces a premature mitotic entry of the lagging nuclei. Although kinetochore Mt bundles were found associated with PCCs, their metaphase and anaphase spindles were abnormal resulting in abnormal or abortive anaphases. In some heterophasic cells, metaphase-anaphase transition did not take place simultaneously in different chromosome groups, signifying that the cells do not exit from the mitotic state after anaphase initiation of the advanced nuclei. Asynchronous pace of mitosis of different chromosome groups was also observed during anaphase and telophase. The probable involvement of phospholipase D (PLD)/phosphatidic acid (PA) signalling in the hyperosmotic stress response of Triticum turgidum root cells was investigated by examining the effects of butanol-1, butanol-2, phosphatidylbutanol (PtdBut), N-acylethanolamine (NAE) and PA on the hyperosmotic response, the organization of the tubulin cytoskeleton and the accumulation of a phosphorylated p38-like mitogen-activated protein (MAP) kinase (phospho-p46) in plasmolysed root cells. The effects of all the treatments were assessed by differential interference contrast (DIC) microscopy of living cells, tubulin immunofluorescence, conventional transmission electron microscopy (TEM), tubulin immunogold localization, protoplast volume measurements and western blot analysis. Butanol-1 and NAE compromised the viability of plasmolysed cells, induced a marked reduction in the plasmolysed protoplast volume, and inhibited hyperosmotically induced tubulin macrotubule formation and the accumulation of phospho-p46. Exogenous PA reinforced the hyperosmotic response of T. turgidum root cells and positively affected tubulin macrotubule formation. Additionally, PA reduced the effects of butanol-1 in plasmolysed cells. Taken together, the data suggest that PLD-mediated PA synthesis occurs upstream of the accumulation of phospho-p46 to regulate hyperosmotically induced macrotubule formation in plasmolysed T. turgidum root cells.  相似文献
