The primary reactants in photosynthesis are defined as the chemical entities on which charges are generated and stabilized after capture of a photon by the photochemical trap: PIX hv P * IX P + I X P + IX , where P stands for the primary electron donor, P * for its excited singlet state, I for the first (ESR-detectable) electron acceptor and X for the secondary acceptor complex. The ESR and ENDOR experiments which have played a rÔle in the identification and characterization of P, I, and X in the bacterial and plant photosystems are comprehensively reviewed. The structural and kinetic information obtained with magnetic resonance techniques are integrated with results obtained with optical spectroscopy to give a unified picture of the pathway of primary photochemistry in photosynthesis. Nomenclature of Primary Reactants In the interest of uniformity this review introduces a nomenclature of the primary reactants that deviates in some respects from the commonly used labels. The nametags used here and listed below are abbreviations of the molecules that are identified as primary reactants, with the exception of the donors, for which I have retained the commonly accepted designation. Photosystems: PS 1, photosystem 1 of plants; PS 2, photosystem 2 of plants; pBPS, the photosystem of purple bacteria; gBPS, ditto of green bacteria. P: Primary donors: P700 (PS 1), P680 (PS 2), P860 (generic label for BChl a containing purple bacteria), P960 (generic label for BChl b containing purple bacteria), P840 (generic name for green bacteria). I: First acceptors: Chl a (PS 1), Ph a (PS 2), BPh a,b (pBPS). X: Secondary acceptors: F x (PS 1), pQ 1 (PS 2), uQ 1 or mQ 1 (pBPS), B (gBPS). A simple analytical method is described for calculating transient EPR spectra of the primary reactants of photosynthesis. Polarization is generated in the primary radical pair P+I and passed to the acceptor X by electron transfer. Satisfactory agreement between theoretical and experimental spectra is obtained by including a dipolar interaction between P+ and I and an anisotropic g tensor for X.  相似文献

Abstract. The photosynthetic characteristics of Elodea nuttallii grown in wastewater in continuous flow reactors in a greenhouse were investigated. The diurnal changes in dissolved inorganic carbon (DIC), dissolved oxygen (DO) and pH were monitored. Photosynthesis removed both CO2(aq) and HCO3? from the reactors. A stoichiometry of 1.19:1 was observed between HCO3? removal during photosynthesis and OH? production during photosynthesis, consistent with theories regarding direct bicarbonate utilization. In laboratory experiments, the light compensation points (гPPFD) were similar (31–35μmol m?2 s?1) to reported values for other macrophytes; however, the light saturation level was high (1100μmol m?2 s?1) and similar to values reported for aerial portions Of heterophyllous macrophytes. The kinetics of photosynthetic oxygen evolution (Km (CO2) = 96mmol m?3; Vmax= 133mmol g?1 Chl h?1) and the CO2 compensation point (г= 44cm3 m?3) suggested an adaptive, low photorespiratory state in response to low carbon concentrations. Photosynthetic Vmax values were slightly, but significantly higher (P 0.001) at pH 8.0 compared to pH 4.5. While CO2 utilization at pH 8 could account for most of the observed phototsynthetic rates, an HCO3? component was present, suggesting two separate transport systems for HCO3? and CO2(aq) in E. nuttallii. The activity of RUBISCO (160.3 mmol g?1 Chl h?1 was one of the highest reported values for aquatic macrophytes. Compared to RUBISCO, we observed lower activities of the β-carboxylating enzymes phopho enolpyruvate carboyxlase (PEPcase), 24.1 mmol g?1 Chl h?1; phosphor enol pyruvate carboxykinase (PEPCKase), 14 mmol g?1 Chl h?1. This suggests that the potential light-independent fixation of carbon in E. nuttallii was much less than RUBISCO-dependent fixation. The RUBISCO/PEPcase ratio was 6.6, indicating that E. nuttallii was similar to Myriophyllum sp. in possessing a physiological adaptation to low CO2 levels which is hypothesized to include carbonic anhydrase (CA) and an active transport system for HCO3?. Many laboratories in different countries are involved in the study of the mechanism of conversion of light energy into chemical energy, namely photosynthesis. As is evident from the literature, the initial phases of photosynthesis, which determine the character of this process, proceed at time intervals of 10(-8) and 10(-13) s. They are associated with absorption of light quanta and energy transfer from the molecules of light-harvesting antenna (LHA) chlorophyll and accesory pigments to the reaction centers (RC), where the key reaction of photosynthesis occurs: photo-induced charge separation. Evidently it is of importance to study experimentally the process that occurs within the 10(-8) -10(-13)s time domain.  相似文献

The triplet state EPR spectra of magnetically aligned whole cells of Rhodopseudomonas viridis and Rhodopseudomonas palustris display a marked dependence on the orientation of the static EPR field with respect to the alignment field direction. This observation implies that the primary donor species on which the triplets are localized are ordered within the membranes. We have developed a theoretical model for the system to enable calculation of the orientation of the magnetic axes of the primary donor species with respect to the membranes in which they reside. The triplet state spectra are generated by an ensemble of partially ordered magnetic systems and a computer simulation of the experimental results. The triplet orientation is very similar for the two organisms studied, where one axis lies predominantly in the plane of the membrane and the other two axes have approximately equal projections onto the normal to the membrane.  相似文献

Phosphatase cytochemistry was used to distinguish between the Golgi apparatus and GERL (considered as a specialized region of endoplasmic reticulum [ER] at the inner [trans] aspect of the Golgi stack) in pancreatic exocrine cells of guinea pig, rat, rabbit, and hamster. The trans element of the Golgi stack exhibits thiamine pyrophosphatase (TPPase) but no acid phosphatase (AcPase) activity. In contrast, GERL shows AcPase but no TPPase activity. The nascent secretory granules, or condensing vacuoles, are expanded cisternal portions of GERL. Continuities of condensing vacuoles with rough ER are suggested, and it is proposed that some secretory components may have direct access to the condensing vacuoles from ER. Connections of Golgi apparatus with GERL were not seen.  相似文献
