Incubation and field experiments were conducted on the influence of thiourea in inhibiting nitrification of urea N, and subsequently on reducing over-winter losses of fallapplied N. Under incubation, most of the added urea placed in bands was nitritified within five or six weeks. However, thiourea when pelleted with urea (21 urea to thiourea by weight) reduced the amount of nitrification to less than one-half during the same period.In two uncropped field experiments in an early dry fall, the application of pelleted urea+thiourea (21) in bands resulted in almost complete inhibition of nitrification of urea for four weeks. In two other uncropped field experiments begun in June with the same fertilizer in bands, half or less of applied N appeared as nitrate after eight weeks. In 10 cropped field experiments with 56 kg N ha–1, urea+thiourea placed in bands depressed nitrification of fall-applied urea over the winter. By early May, the urea mixed into the soil in the previous fall was nearly all nitrified, while only one-half of the banded urea+thiourea was nitrified. The loss of mineral N by early May was 38% with urea mixed into the soil, but only 18% with bands of urea+thiourea.The 10 sites were cropped to spring barley. The increase in yield of grain or the increase in %N uptake from fertilier N was approximately only one-half as much with fall-applied urea mixed into the soil as compared to spring-applied urea added in the same way. Specifically, fall-applied mixed urea produced 930 kg ha–1 less grain yield and 32% less N uptake from fertilizer N than did mixed urea in spring. On fall-application there was some benefit from banding of urea or with mixing urea+thiourea pellets into the soil, but the banding of urea+thiourea pellets gave more benefit. Among the fall applications, banded urea+thiourea pellets produced 670 kg ha–1 more grain yield and 26% more N uptake in grain from fertilizer N than did urea mixed into the soil.  相似文献

Summary Using vertically isolated micro-plots the isotopic recovery technique was tested for assessing the extent of ammonia volatilization loss from N15-labelled urea applied on the forest floor (Pinus silvestris L.). The size of the ammonia loss was obtained as a difference between the amounts of labelled urea N added and the amounts of labelled N recovered in the soil profile after 13, 31, and 39 days's exposure, respectively. Urea materials of two different pellet size were used: granulated small pellets (280 pellets per gram) and tablets (2.06 g each). The nitrogen application rate was 200 kg N per hectare. The recovery data for 13 days' exposure indicated a volatilization loss, which for the small pellet urea was 24.9 per cent and for the tabletted urea 12.1 per cent. The corresponding figures for the 31 days' exposure, during which the total amount of precipitation was 14 mm, were 15.1 and 26.9 per cent, respectively. The pattern of labelled N distribution in the soil profile examined showed that during the period of exposure in question a leaching loss of labelled N was rather unlikely. It was demonstrated, furthermore, that nitrogen from the tabletted urea had diffused to a greater depth of the soil than that from the small-pellet urea. Nitrogen from the small-pellet urea was to a large extent recovered in the litter layer. On exposure to heavy rain the tabletted urea was subjected to the highest leaching loss. An addition of 10 per cent (w/w) of metaphosphoric acid or sublimed sulphur to the tabletted urea did not result in any further reduction of the volatilization loss. The merits and limits of the isotopic recovery technique are discussed.  相似文献

Pellet coating is traditionally carried out using the Wurster coater. This study investigated the feasibility of pellet coating in a newly developed coater built with a unique airflow system, the Supercell™ coater (GEA Pharma Systems, UK). A full factorial design study was carried out to evaluate the influences of the spray rate of the coating dispersion, batch size of the pellet load, pellet size fraction and plenum pressure of the fluidizing air on the color coating of pellets in the Supercell™ coater. Results showed that pellets could be successfully coated using the Supercell™ coater. Higher plenum pressures and lower spray rates were found to minimize pellet agglomeration during coating. Although coating efficiencies were comparable amongst the different pellet size fractions, larger batch sizes of pellets were coated with higher efficiencies. Process optimization was carried out for each pellet size fraction and a large batch size (120 g) in combination with a high plenum pressure (1,500 mm WC) were deemed optimal. Optimal spray rates differed according to pellet size fraction and a lower spray rate was required for smaller pellets. Pellet flow patterns observed during coating were dependent on the pressure drop across the fluidized load. Isolates of aflatoxin-producing strains of Aspergillus grow on autoclaved and field-grown (lesser extent) Glycine max beans. Both mixed and aflatoxin B1 inhibit G. max, cv. Essex bean germination and elongation of either attached or excised cultured roots. Because B1 impairs the latter roots' ability to intracellularize [14C]-leucine, it may alter plasmalemma structure and/or function. To determine whether incubation of excised roots for 18 hours in toxin-containing medium could affect cellular membrane chemical content, organelles were isolated by differential centrifugation (1 000, 40 000, and 80 000 xg) of homogenates and characterized chemically. Statistically significant differences between treated and untreated roots in acid insoluble protein but not either sterol or lipid phosphorus levels were observed for both 40,000 and 80,000 xg pellets. Protein and sterol recoveries were 81 (treated) and 84 (untreated) % for the former and 77 (treated) and 79 (untreated) % for the latter. Lipid phosphorus recoveries were 87.3 (treated) and 136 (untreated) % with and 96 (treated) and 83 (untreated) without membrane stabilization. Protein:sterol:lipid phosphorus were 35.74.51 (1 000 xg), 18.93.61 (40000 xg), 26.34.61 (80 000 xg) and 1,010291 (80 000 xg supernatant) for untreated and 36.93.31 (1,000 xg), 23.13.81 (40 000 xg), 36.24.81 (80 000 xg) and 1,05321.71 (80 000 xg supernatant) for treated roots. Significant differences in RNA content between treated and untreated roots were found for both 1 000 and 40 000 xg pellets but not for the 80 000 xg pellet and its supernatant. Whereas a significant increase in the 1 000 xg pellet occurred upon treatment, a decrease was noted for the 40 000 xg pellet but not for the 80 000 xg pellet and its supernatant. Similar pH 6 (plasmalemma marker enzyme) and 9 (mitochondrial marker enzyme) K+-stimulated ATPase activities were demonstrated for 40 000 and 80 000 xg pellets. The 1 000 xg pellet contained greater than 50% of the NADH-cytochrome c-reductase activity (endoplasmic reticulum marker enzyme) recovered from fractions examined for this activity which was absent from the 40 000 xg pellet. Both the 80 000 xg pellet and its supernatant possessed equivalent reductase activities. Inosine diphosphatase activity (dictyosome marker enzyme) was not present in 1 000 xg pellets obtained from either treated or untreated roots but was in both 40 000 and 80 000 xg pellets. Summary Urea hydrolysis was measured in two separate sets of experiments. (1) Nine soil (0–15 cm) samples were treated with 200 g of urea-N g–1 dry soil and incubated (at 37°C) at 50 per cent of the water holding capacity. Samples were periodically analysed for the remaining urea-N. The urease activity (time in hoursrequired to hydrolyse half the applied urea-N) was determined to be 5.8 to 15.2 hours in the various soils, which appeared to associate principally with the organic carbon content of the soils (r=–0.80**). (2) Three soils were treated with 25 to 2000 g urea-N g–1 dry soil amounting to 0.9 to 72.0 mM urea in 11 soil: solution. The system was buffered at pH 7.2 and agitated for 5h when the remaining urea-N was determined. The values of Km and Vmax were computed by two methods (i) from the integrated form of the Michaelis-Menten equation based on the results of the first study, and (ii) from the Michaelis-Menton equation based on urea hydrolysis in the second study. The integrated method appeared to be more suitable for enzyme kinetic studies in soil systems where the Km and Vmax values bore close relationship (r=–0.88**).  相似文献

The suitability of the fluorescein diacetate hydrolyzing activity (FDA) assay for determining the biological potential (ie fungal biomass produced per unit of substrate) of solid pelleted fungal inoculum intended for use in the bioaugmentation of contaminated soils with white-rot fungi, was evaluated. FDA activity of the white-rot fungusPhanerochaete chrysosporium grown on pelleted substrates and on agar was found to be proportional to quantities of fungal ergesterol and fungal dry matter, respectively. Inoculum biological potential was found to be greatly influenced by substrate formulation and structure, and temperature. Biological potential and the type of carrier influenced the ability ofP. chrysosporium to tolerate pentachlorophenol (PCP).Phanerochaete chrysosporium andTrametes versicolor introduced into PCP-contaminated soil on pellets with higher biological potential and higher nitrogen content (C:N ratio of 501), did not remove PCP more efficiently than when the fungi were introduced on pellets with a lower biological potential (C:N ratio of 3091). However, under the latter conditions most of the PCP was transformed to pentachloroanisole (PCA). In soil inoculated withT. versicolor on pellets with high biological potential, higher manganese peroxidase activity was detected compared to soil inoculated with pellets with a lower biological potential.  相似文献

Generalizing results from fungal fermentations is difficult due to their high sensitivity toward slight variation in starting conditions, poor reproducibility, and difference in strains. In this study a mathematical model is presented in which oxygen transfer, agitation intensity, dissolved oxygen tension, pellet size, formation of mycelia, the fraction of mycelia in the total biomass, carbohydrate source consumption, and biomass growth are taken into account. Two parameters were estimated from simulation, whereas all others are based on measurements or were taken from literature. Experimental data are obtained from the fermentations in both 2 L and 100 L fermentors at various conditions. Comparison of the simulation with experiments shows that the model can fairly well describe the time course of fungal growth (such as biomass and carbohydrate source concentrations) and fungal morphology (such as pellet size and the fraction of pellets in the total biomass). The model predicts that a stronger agitation intensity leads to a smaller pellet size and a lower fraction of pellets in the total biomass. At the same agitation intensity, pellet size is hardly affected by the dissolved oxygen tension, whereas the fraction of mycelia decreases slightly with an increase of the dissolved oxygen tension in the bulk. All of these are in line with observations at the corresponding conditions.  相似文献

Rainfall is a driving force for the transport of environmental contaminants from agricultural soils to surficial water bodies via surface runoff. The objective of this study was to characterize the effects of antecedent soil moisture content on the fate and transport of surface applied commercial urea, a common form of nitrogen (N) fertilizer, following a rainfall event that occurs within 24 hr after fertilizer application. Although urea is assumed to be readily hydrolyzed to ammonium and therefore not often available for transport, recent studies suggest that urea can be transported from agricultural soils to coastal waters where it is implicated in harmful algal blooms. A rainfall simulator was used to apply a consistent rate of uniform rainfall across packed soil boxes that had been prewetted to different soil moisture contents. By controlling rainfall and soil physical characteristics, the effects of antecedent soil moisture on urea loss were isolated. Wetter soils exhibited shorter time from rainfall initiation to runoff initiation, greater total volume of runoff, higher urea concentrations in runoff, and greater mass loadings of urea in runoff. These results also demonstrate the importance of controlling for antecedent soil moisture content in studies designed to isolate other variables, such as soil physical or chemical characteristics, slope, soil cover, management, or rainfall characteristics. Because rainfall simulators are designed to deliver raindrops of similar size and velocity as natural rainfall, studies conducted under a standardized protocol can yield valuable data that, in turn, can be used to develop models for predicting the fate and transport of pollutants in runoff.  相似文献

RNase-sensitive DNA polymerase activity (RSDP) was tested in different cell fractions of Neurospora crassa cell types and its morphological mutants. This RSDP was found localized in the microsomal pellet fraction and absent in the purified nuclear pellets isolated from different N. crassa cell types: conidia, germinated conidia, and mycelia. This enzyme is capable of synthesizing a DNA product only in the presence of all four deoxyribonucleoside-5-triphosphates and Mg2+. Removal of RNA from the pellet fraction by RNase strongly inhibited the DNA synthesis. The endogenous synthesis of DNA in the microsomal pellet fraction was associated with the formation of an RNA:DNA hybrid as analyzed by Cs2SO4 equilibrium density gradient centrifugation. The DNA product after alkali hydrolysis hybridizes with the RNA isolated from the same pellet fraction, as analyzed by elution from hydroxylapatite column at 60 C. This DNA product did not hybridize with poly(A). A few mutants tested showed this RNase-sensitive DNA polymerase activity.This work was supported in part by a contract with the U.S. Department of Energy and a grant from the U.S. Naval Research.  相似文献

The responses of second-stage juveniles (J2) of Meloidogyne incognita race 3 to calcium alginate pellets containing hyphae of the nematophagous fungi Monacrosporiura cionopagum, M. ellipsosporum, and Hirsutella rhossiliensis were examined using cylinders (38-mm-diam., 40 or 72 mm long) of sand (94% <250-μm particle size). Sand was wetted with a synthetic soil solution (10% moisture, 0.06 bar water potential). A layer of 10 or 20 pellets was placed 4 or 20 mm from one end of the cylinder. After 3, 5, or 13 days, J2 were put on both ends, on one end, or in the center; J2 were extracted from 8-ram-thick sections 1 or 2 days later. All three fungal pellets were repellent; pellets without fungi were not. Aqueous extracts of all pellets and of sand in which fungal pellets had been incubated were repellent, but acetone extracts redissolved in water were not. Injection of CO₂ (20 μl/minute) into the pellet layer attracted J2 and increased fungal-induced mortality. In vials containing four randomly positioned pellets and 17 cm³ of sand or loamy sand, the three fungi suppressed the invasion of cabbage roots by M. javanica J2. Counts of healthy and parasitized nematodes observed in roots or extracted from soil indicated that, in the vial assay, the failure of J2 to penetrate roots resulted primarily from parasitism rather than repulsion. Data were similar whether fungal inoculum consisted of pelletized hyphae or fungal-colonized Steinernema glaseri. Thus, the results indicate that nematode attractants and repellents can have major or negligible effects on the biological control efficacy of pelletized nematophagous fungi. Factors that might influence the importance of substances released by the pellets include the strength, geometry, and duration of gradients; pellet degradation by soil microflora; the nematode species involved; and attractants released by roots.  相似文献

The effects of dissolved oxygen tension and mechanical forces on fungal morphology were both studied in the submerged fermentation of Aspergillus awamori. Pellet size, the hairy length of pellets, and the free filamentous mycelial fraction in the total biomass were found to be a function of the mechanical force intensity and to be independent of the dissolved oxygen tension provided that the dissolved oxygen tension was neither too low (5%) nor too high (330%). When the dissolved oxygen concentration was close to the saturation concentration corresponding to pure oxygen gas, A. awamori formed denser pellets and the free filamentous mycelial fraction was almost zero for a power input of about 1 W/kg. In the case of very low dissolved oxygen tension, the pellets were rather weak and fluffy so that they showed a very different appearance. The amount of biomass per pellet surface area appeared to be affected only by the dissolved oxygen tension and was proportional to the average dissolved oxygen tension to the power of 0.33. From this it was concluded that molecular diffusion was the dominant mechanism for oxygen transfer in the pellets and that convection and turbulent flow in the pellets were negligible in submerged fermentations. The biomass per wet pellet volume increased with the dissolved oxygen tension and decreased with the size of the pellets. This means that the smaller pellets formed under a higher dissolved oxygen tension had a higher intrinsic strength. Correspondingly, the porosity of the pellets was a function of the dissolved oxygen tension and the size of pellets. Within the studied range, the void fraction in the pellets was high and always much more than 50%.  相似文献

Actinomycetes, especially members of the genus Streptomyces, are responsible for producing the majority of known antibiotics. The production of antibiotics by filamentous organisms is often dependent on the morphology and size distribution of the pellet population within the culture. Particle interaction and subsequent pellet formation are primarily dependent on the rate of collision of particles in culture, which is in turn, a function of fluid turbulence. The microbial polysaccharide xanthan gum was used to artificially regulate the apparent viscosity (a) of S. hygroscopicus fermentation broths with the aim of controlling particle interaction, aggregation and hence pellet formation. An increase in both pellet count and biomass concentration from approximately 2,000 to 8,000 pellets ml–1 and 0.9–2.1 g l–1 dry weight of biomass, as well a decrease in the mean pellet volume from 0.014 to 0.004 mm3 was observed in cultures supplemented with 3 g l–1 xanthan gum. The addition of xanthan gum significantly alters fluid rheology by increasing the a. Counter-intuitively, an increase in the a within the experimental range examined resulted in an increase in the rate of gas–liquid mass transfer. This was attributed to the predominantly diffusive nature of oxygen transfer in shake flask cultures.  相似文献

Cyanobacteria play a major role as direct producers of biofuels, such as ethanol and butanol, with the aid of genetic engineering. However, development of a new harvesting-technology is essential to achieve economic viability of biofuel production from cyanobacteria. In this study, we demonstrated the feasibility of harvesting the unicellular cyanobacterium Synechocystis sp. PCC 6803 using pre-made filamentous fungal pellets and investigated key factors affecting efficiency of harvest, including fungal strain, pellet quantity (number of pellets), initial pH, and organic carbon source. Synechocystis sp. PCC 6803 cells attached to Aspergillus oryzae pellets, indicating that this fungal pellet had a desirable harvesting effect, while Rhizopus oryzae pellets had no effect on harvesting. Increasing pellet quantity and adding organic carbon sources, such as glucose and xylose, improved the harvesting efficiency of Aspergillus oryzae pellet; efficiency was not affected by the initial pH.  相似文献

Combined torrefaction and pelletization is used to increase the fuel value of biomass by increasing its energy density and improving its handling and combustion properties. In the present study, a single-pellet press tool was used to screen for the effects of pellet die temperature, moisture content, additive addition, and the degree of torrefaction on the pelletizing properties and pellet quality, i.e., density, static friction, and pellet strength. Results were compared with pellet production using a bench-scale pelletizer. The results indicate that friction is the key factor when scaling up from single-pellet press to bench-scale pelletizer. Tuning moisture content or increasing the die temperature did not ease the pellet production of torrefied wood chips significantly. The addition of rapeseed oil as a lubricant reduced the static friction by half and stabilized pellet production; however, the pellet quality, strength, and density were negatively affected. The pellets produced from pine wood torrefied at 250 and 280 °C were shorter than pellets produced from untreated wood and their quality did not match conventional wood pellet standards. However, the heating value of the torrefied pellets was higher and the particle size distribution after grinding the pellets was more uniform compared to conventional wood pellets.  相似文献

Summary Four-year-old Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) saplings planted in pots with a sand and peat mix (11) were fertilized at the rate of 200 kg N/ha of (15NH2)2CO (U-15),15NH4NO3 (A-15) and NH4 15NO3(An-15). They were placed in a shadehouse and watered regularly to maintain soil moisture at field capacity over periods of one and two years. Quantity of15N in foliage generally increased from old to current growth, irrespective of the nitrogen source. Utilization of15N fertilizers by saplings after the first and second growing seasons following fertilization was greatest with nitrate labelled ammonium nitrate AN-15, and nearly equal for urea U-15 and ammonium labelled ammonium nitrate A-15. The soil immobilized more fertilizer nitrogen-15 from U-15 and A-15 than from AN-15. Data from the present study, in which leaching losses of fertilizer were minimized, demonstrated that in terms of nitrogen uptake by the saplings the nitrate fertilizer was superior to ammonium fertilizer.  相似文献

Summary Using diluted phosphoric acid- and glycerol-treated polyurethane plastic foam discs as static ammonia absorbers, the extent of ammonia loss from applied urea was determined on forest soil under field conditions. The investigation, which primarily involved a comparison of urea materials of two different pellet sizes (2–4 mg and 2060 mg urea per pellet, respectively), extended over a period of 28 days. The urea was applied to the soil surface at a rate of 200 kg N per hectare.It was found that the gaseous ammonia loss from the large-pellet urea (tablets) was markedly lower than that from the small-pellet urea during the first two weeks of exposure. On extending the observation period to four weeks, this difference in cumulative ammonia loss decreased successively. After 28 days' exposure, at a daily mean temperature of 13°C, the volatile loss amounted to 20 and 22 per cent, respectively. An addition of 5% (w/w) of a solution of concentrated orthophosphoric acid, or a fine-crystalline ortho-boric acid to the large-pellet urea, resulted in a reduction of the loss to half the value. Summary A study of the effects of malathion and parathion applied at 10 and 50 g/g of soil on transformations of urea and (NH4)2SO4–N in a sandy loam showed that the insecticides retarded urea hydrolysis as well as nitrification of urea and (NH4)2SO4–N. At 50 parts/106 rate of the insecticides, inhibition of urea hydrolysis ranged from 44 to 61% after 0.5 week and from 7 to 21% after 3 weeks of application. The insecticides inhibited the conversion of NH4 + to NO2 without appreciably affecting the subsequent oxidation of NO2 to NO3 –N. This resulted in accumulation of higher amounts of NH4 +–N in soil samples treated with ammonium sulfate or urea N.The results suggest that transformations of urea and NH4 + fertilizers in soils may be influenced by the amount of organophosphorus insecticide present and this may affect plant nutrition and fertilizer use.  相似文献

Adult male hamsters bearing either a blank beeswax, 6-methoxy-2-benzoxazolinone (6-MBOA), or melatonin pellet were exposed to 8 weeks (Oct. 6–Dec. 6) of natural autumn decreasing photoperiod (<11 h light) and temperature conditions (mean 10°C for last 4 weeks) or to a 14 h light/10 h dark (14L10D) photoperiod and controlled temperature (20°C). Melatonin but not 6-MBOA pellets partially prevented the combined effects of short photoperiod and cold temperatures on the testes and accessory organs. However, both 6-MBOA-and melatonin-treated hamsters maintained outdoors had significantly higher pituitary follicle stimulating hormone (FSH) values compared to their respective indoor-treated controls or to the animals kept outdoors and treated with a blank beeswax pellet. When one compares the various effects of 6-MBOA and melatonin (2 mg/month) on the reproductive system of the male hamster, 6-MBOA is not as effective as melatonin in altering reproductive responses to short photoperiod and cool temperatures at the dose administered.  相似文献

Florida has more than 14 million ha of acidic soils and most of these acidic soils are very sandy. Transport of phosphorus from these soils to surface water is a great concern in Florida. One of the promising approaches is to develop slow release P fertilizers to replace the regular water soluble fertilizers. Dolomite phosphate rock (DPR) is an ideal material for making this type of fertilizer for acidic sandy soils, which are wide-spread in Florida. The effects of soil moisture, particle size, and source of DPR on the extent and rate of DPR dissolution in a representative acidic sandy soil were examined in an incubation study. DPR dissolution estimated by the NaOH extraction method was strongly dependent on the mineralogy and chemistry of the DPR source materials. Decreasing particle size increased the percentage dissolution of the DPR in soil. Soil moisture affected the initial dissolution rate of the DPR and the time to reach equilibrium. Soil extractable P (Olsen-P) generally increased with DPR dissolution, but the ratios of Olsen-P to dissolved DPR-P (NaOH-P) were significantly different between the two DPR sources. Dissolution of DPR in the acidic sandy soil was well described by the Langmuir and Elovich kinetic equations, with the Langmuir equation being better. Constant A derived from the Elovich equation and potential maximum dissolution (PMDPR) from the Langmuir equation were closely correlated with the increases in pH and exchange (Ca+Mg) after DPR application. The two parameters appeared useful in evaluating DPR dissolution and timing of DPR application in acidic sandy soils.  相似文献
