A few smell deficient mutants of Drosophila have been characterized recently. In a series of experiments, we have compared the slow electrical response of the antenna (EAG) to calibrated concentrations of butanol and aliphatic n-acetates of olf C mutants with those of wild flies. This mutation was found to affect the dose-response curves specifically. Our data can be explained by assuming that this mutation affects the peripheral olfactory membrane at several levels.  相似文献

Cell extracts from shake cultures of the wild type and six mutant strains of Phycomyces converted [2-14C] MVA into carotenes, squalene and prenyl phosphates. Oxygen was required for the desaturation of phytoene. When compared with the wild type, cells extracts of carB and carR mutants are much less effective in phytoene dehydrogenation and lycopene cyclization, respectively. This confirms previous conclusions about the biochemical functions of the carB and carR genes, which were based on genetic and in vivo studies. CarA strain mutants accumulate, in vivo, much less β-carotene than the wild type. This correlates with a 10-fold decrease in carotenogenesis in vitro. The addition of retinol to incubations of cell extracts of the wild type and C2 strains stimulated β-carotene formation. Both carB and carR mutants show enhanced total carotenogenic activities in vitro and the carS mutant shows a higher β-carotene-synthesizing activity than the wild type. It is suggested that the feed-back regulatory mechanism known to control this pathway operates at the level of enzyme synthesis.  相似文献

Pseudomonas aeruginosa mucoid strains as well as mutants defective in pili, flagella, lipopolysaccharide, and proteases were isolated and tested for their virulence for the larvae of Galleria mellonella. Of all of the mutants, only the lipopolysaccharide-deficient (rough) strain showed a major decrease in virulence when compared to the wild type. The LD50 of the rough strain was about 30,000 bacteria/larva or roughly 10,000-fold higher than the wild type, suggesting an important role for the lipopolysaccharide in P. aeruginosa infections of G. mellonella larvae.  相似文献

Thermal stability and pH optima of NADH-nitrate reductase-associated cytochrome c reductase and FMNH2-nitrate reductase from wild type, cv Steptoe or Winer, and mutants nar 1d, nar 1g, nar 1h, Xno 18 and Xno 19 were compared to determine if structural differences in the nitrate reductase protein could be detected. Also, the nitrate reductase-associated cytochrome c reductase from nar 1d was purified and compared with the wild type by peptide mapping. The pH optimum for FMNH2-nitrate reductase from Steptoe and nar 1h, and for NADH-cytochrome c reductase from Steptoe, nar 1d, nar 1g and nar 2a was 7.5. Thermal stabilities of the nitrate reductase-associated activities (FMNH2-nitrate reductase or NADH-cytochrome c reductase) from nar mutants were less than the Steptoe wild type, while Xno mutants were equal to the Winer wild type. Cleveland peptide maps of nar 1d NADH-cytochrome c reductase and Steptoe nitrate reductase were identicalwhen digested with endoprotease lys-C but were distinctly different in one peptide when digested with Staphylococcus aureus endoprotease V8. These results provide evidence that nar 1 gene codes for the nitrate reductase polypeptide.  相似文献

Background and Aims Barley (Hordeum vulgare) double mutants Hv-Hd/tw2, formed by hybridization, are characterized by inherited phenotypic instability and by several new features, such as bract/leaf-like structures, long naked gaps in the spike, and a wide spectrum of variations in the basic and ectopic flowers, which are absent in single mutants. Several of these features resemble those of mutations in auxin distribution, and thus the aim of this study was to determine whether auxin imbalances are related to phenotypic variations and instability. The effects of auxin inhibitors and 2,4-D (2,4-dichlorophenoxyacetic acid) on variation in basic and ectopic flowers were therefore examined, together with the effects of 2,4-D on spike structure.Methods The character of phenotypic instability and the effects of auxin inhibitors and 2,4-D were compared in callus cultures and intact plants of single homeotic Hv-tw2 and Hv-Hooded/Kap (in the BKn3 gene) mutants and alternative double mutant lines: offspring from individual plants in distal hybrid generations (F9F10) that all had the same BKn3 allele as determined by DNA sequencing. For intact plants, two auxin inhibitors, 9-hydroxyfluorene-9-carboxylic acid (HFCA) and p-chlorophenoxyisobutyric acid (PCIB), were used.Key Results Callus growth and flower/spike structures of the Hv-tw2 mutant differed in their responses to HFCA and PCIB. An increase in normal basic flowers after exposure to auxin inhibitors and a decrease in their frequencies caused by 2,4-D were observed, and there were also modifications in the spectra of ectopic flowers, especially those with sexual organs, but the effects depended on the genotype. Exposure to 2,4-D decreased the frequency of short gaps and lodicule transformations in Hv-tw2 and of long naked gaps in double mutants.Conclusions The effects of auxin inhibitors and 2,4-D suggest that ectopic auxin maxima or deficiencies arise in various regions of the inflorescence/flower primordia. Based on the phenotypic instability observed, definite trends in the development of ectopic flower structures may be detected, from insignificant outgrowths on awns to flowers with sterile organs. Phenotypically unstable barley double mutants provide a highly promising genetic system for the investigation of gene expression modules and trend orders.  相似文献

The white and topaz eye color mutants of L. cuprina are defective in the production of the brown screening pigment xanthommatin. Both white and topaz mutants were found to be unable to accumulate xanthommatin precursors in the larval malpighian tubules, correlating with their reduced early pupal level of this metabolite. In addition, white mutants showed reduced rates of accumulation of kynurenine and 3-hydroxykynurenine in the adult eyes. Another mutant strain, grape, was also defective in its ability to accumulate these xanthommatin precursors in the eyes, although accumulation was normal in the larval tubules. In contrast, the topaz mutants were found to be normal in eye accumulation, although tubule accumulation was markedly abnormal. These properties of the white and topaz mutants of L. cuprina are compared with those of the white and scarlet mutants of D. melanogaster, and it seems likely that in the two species these genes are involved with the uptake or storage of xanthommatin precursors in specific tissues.This work was supported by Grant D2 75/15248 from the Australian Research Grants Committee.  相似文献

Two mutants with different conidiogenesis were isolated from wild phatotypes of Hirsutella thompsonii var. thompsonii collected from the citrus rust mite in Florida. One mutant (WSI) produces more typical single-neck phialides and conidia than the wild type. The other mutant (SS) was developmentally altered in producing condiogenous cells earlier than the wild type as well as phialides with multiple neck capable of conidiogenesis. The mutant (SS) appeared more virulent than mutant (WSI) and the wild type when bioassayed using the citrus rust mite as a host.  相似文献

Toxoplasma gondii, strain RH, produced plaques in human fibroblast tissue cultures over the temperatures 30–41 C. Muta?enesis with N-methyl-N′-nitro-N-nitrosoguanidine yielded seven temperature-sensitive mutants that had lost the ability to form plaques at 40 C but still grew well at 33 C. No spontaneous mutants were detected. The temperature-sensitive mutants were not markedly thermolabile and adsorbed normally to tissue culture cells at 40 C. Three mutants differed from one another in their temperatures for optimal growth, and in their ability to remain infectious within cells incubated at 40 C. Both mutants that were tested were found to be markedly less virulent for mice than was the wild type RH strain.  相似文献

Cell extracts capable of converting MVA-[2-14C] into isoprenoids were obtained from the yellow C115-mad-107(−) and red C9-carR21(−) mutants of Phycomyces blakesleeanus. Neither air nor light was essential for carotene biosynthesis. The specific activities of the terpenoid-synthesizing enzymes varied with the age of the cultures although the formation of lycopene (ψ,ψ-carotene) in the C9 and of β-carotene (β,β-carotene) in the C115 'mutants. respectively, followed the increase in the dry weight yield of the cultures. The significance of these results to the biosynthesis of carotenes and to the classification of these compounds as secondary metabolites is discussed.  相似文献

Wild-type Drosophila melanogaster and the learning mutants dunce, amnesiac and rutabaga, were tested using a new operant conditioning paradigm for single flies. All strains are able to learn to different extents, but no evidence of memory was found in the mutants amnesiac and rutabaga, while dunce has a reduced but extended memory. The relationship between this characteristic and cAMP levels are discussed. The three mutants have previously been shown, using classical conditioning paradigms to be deficient in olfactory learning and/or memory, and show reduced visual learning. The variability of the response of the mutants in the different paradigms is discussed in relation to the generality of the Aplysia model of the cellular mechanism underlying learning. In the operant conditioning paradigm described here, 93% of the wild-type flies learned to criterion. The performance of individual flies was consistent.  相似文献

Individual phospholipids were assayed in exponentially growing and G1-arrested temperature-sensitive cell division cycle (cdc) mutants of Saccharomyces cerevisiae. It was observed that cdc28 cells which are known to arrest at ‘start’ when shifted to their non-permissive temperature, resulted in a 40% decrease in phosphatidylinositol (PI) level while the phosphatidylserine (PS) content was doubled in these cells. The reduced level of PI was restored in cdc4 and cdc7 mutants which are known to arrest past the ‘start’. The increase in PS level in cdc8 mutant which was probably to compensate the intrinsic charging of membrane environment, was also reduced in cdc4 and cdc7 mutants. Our results demonstrate that PI may play a role in yeast cell division and growth that the abnormalities of cdc28 could also be related to PI decrease.  相似文献

Eight mutants of Drosophila m. have been isolated that fail to display longlasting modification of “choice tube phototaxis”. Two different classes of stocks defective in plasticity of phototaxis (dipp) can be discriminated: (1) Mutants which are affected in the stability of the Prolonged Depolarising Afterpotential (PDA) in the peripheral retinula cells R1–R6. (2) Mutants with normal PDA but abnormalities in fast escape phototaxis especially under conditions where only receptor cells R7–R8 serve as input for this behavioural trait.  相似文献

Actin is a key cytoskeletal protein with multiple roles in cellular processes such as polarized growth, cytokinesis, endocytosis, and cell migration. Actin is present in all eukaryotes as highly dynamic filamentous structures, such as linear cables and branched filaments. Detailed investigation of the molecular role of actin in various processes has been hampered due to the multifunctionality of the protein and the lack of alleles defective in specific processes. The actin cytoskeleton of the fission yeast, Schizosaccharomyces pombe, has been extensively characterized and contains structures analogous to those in other cell types. In this study, primarily with the view to uncover actin function in cytokinesis, we generated a large bank of fission yeast actin mutants that affect the organization of distinct actin structures and/or discrete physiological functions of actin. Our screen identified 17 mutants with specific defects in cytokinesis. Some of these cytokinesis mutants helped in dissecting the function of specific actin structures during ring assembly. Further genetic analysis of some of these actin mutants revealed multiple genetic interactions with mutants previously known to affect the actomyosin ring assembly. We also characterize a mutant allele of actin that is suppressed upon overexpression of Cdc8p-tropomyosin, underscoring the utility of this mutant bank. Another 22 mutant alleles, defective in polarized growth and/or other functions of actin obtained from this screen, are also described in this article. This mutant bank should be a valuable resource to study the physiological and biochemical functions of actin.  相似文献

A forced heterocaryon was established between two auxotrophic conidial color mutants of Metarhizium anisopliae. From the heterocaryon, a prototrophic somatic diploid was selected which, in turn, yielded somatic segregants. The virulence of the original mutants, the somatic diploid, and the somatic segregants was evaluated on three species of mosquitoes as well as on Ostrinia nubilalis larvae. The virulence of the somatic diploid was comparable to that of the wild-type parental strain while the auxotrophic somatic segregants exhibited virulence approximately equal to that of the auxotrophic components of the heterocaryon. Putative somatic diploids were obtained between morphological mutants of the two species varieties (M. anisopliae var. minor and var. major). The presumptive diploids were avirulent for the insect species to which the parental strains exhibited virulence.  相似文献

Lotus japonicus hypernodulating mutants, Ljsym78-1 and Ljsym78-2, by the arbuscular mycorrhizal fungus Glomus sp. was characterized. The mutants are defective in systemic autoregulation of nodulation and nitrate inhibition, and form an excess of nodules and lateral roots. The percent root length colonized by the arbuscular mycorrhizal fungi was significantly higher for the mutant than wild-type roots. Detailed assessment of the colonization indicated that the percentage of colonization by arbuscules was increased, but that by external hyphae, internal hyphae and vesicles was decreased, in the mutant roots compared with the wild-type. The succinate dehydrogenase activity of arbuscules, external hyphae and internal hyphae showed similar trends. In addition, the majority of individual arbuscules that formed on the mutant roots had a well-developed and seemingly tough morphology. The results suggest that mutation at the Ljsym78 locus positively stimulates the growth and activity of arbuscules, but leads to reduced growth and activity of hyphae. We report the first identification of Lotus japonicus mutants that show significantly increased arbuscule formation and termed these mutants Arb++. We used histology, optical measurements of photoreceptor integrity and electrophysiology to characterize rdgB mutants of Drosophila which have hereditary retinal degeneration. We studied different mutant alleles of rdgB which had been reported to have differing receptor involvement in degeneration. However, we found similar profiles of degeneration: compound eye receptors R1-6 degenerate structurally over one week in a diurnal lighting cycle while R7/8 are preserved. We confirmed earlier findings that norpA transduction mutants inhibit degeneration in rdgB. Our micrographs show degeneration in the lamina (first synaptic neuropil) but none in the medulla (second synaptic neuropil).  相似文献

We studied rdgB, a retinal degeneration mutant, and trp, a phototransduction mutant, separately and in combination in Drosophila. First we showed that trp did not block degeneration in white-eyed rdgB mutants. Thus, rdgB was useful in determining the defects which trp caused in the compound eye receptors R7 and R8; this is because rdgB selectively eliminates R1-6 photoreceptors which would, if present, dominate the compound eye responses. R7 and R8 both express the trptransient receptor potential phenotype in trp mutants. The trp mutation does not change receptor spectral sensitivities, nor does it alter the dark stability of R1-6's and R7's metarhodopsins as judged by dark adaptation studies. The dark adaptation is not significantly affected by trp. However, trp slows the dark adaptation of R8 considerably and seems to make the blue-induced inactivation of R1-6 less stable.  相似文献
