Cytokinins in a genic male sterile line of Brassica napus   总被引:4,自引:0,他引:4  
Endogenous cytokinin levels were analyzed in four different organs viz., root, stem, leaf and mature flower, of the wild type (cv. Westar) and a genic male sterile (GMS) line of Brassica napus . Various cytokinins viz., bases (Z, DZ), ribosides (ZR, DZR), glucosides (OGZ + OGZR, OGDZ + OGDZR) and nucleotides (ZNT, DZNT) were quantified using Amaranthus betacyanin bioassay and ELISA. The major cytokinin in all the organs was DZ. In general, leaves had the highest levels of cytokinins as compared to the other organs. Root, stem and mature flowers of the wild type plants had higher levels of cytokinins and their metabolites than the GMS line. However. levaves of the GMS line had greater amount of various cytokinins as compared to the wild type. The major cytokinin quantitatively different between the two lines was DZ. The results suggest the possible involvement of cytokinins in the expression of genic male sterility in Brassica napus .  相似文献

Purification and characterization of a nitrilase from Brassica napus   总被引:1,自引:0,他引:1  
In germinating seedlings of Brassica napus glucosinolate levels decrease and are potentially degraded to nitriles by a myrosinase. Little is known about the metabolism of glucosinolate aglycone products and the objective of this work was to investigate nitrilase activity and carry out a purification of the enzyme from seedlings of B. napus . A nitrilase capable of converting phenylpropionitrile to phenylpropionic acid was purified to apparent homogeneity from seedlings of B. napus . The protein has a molecular mass of approximately 420 kDa made up of 38 kDa subunits. The pI of the native protein was found to be 4.6. Under denaturing conditions on an isoelectric focusing (IEF) gel a major and minor protein was observed with pI in the range of 5.4-5.9, suggesting the presence of isoforms. Apart from the potential role of the nitrilase in indole-3-acetic acid (IAA) synthesis a developmental study with seedlings indicates that the increase in activity observed may be linked to the in vivo degradation of glucosinolates.  相似文献

Cruciferin (12S globulin) is a large, neutral, oligomeric protein synthesized in rapeseed ( Brassica napus ) during the seed development. It is composed of six subunit pairs. Each pair consists of one heavy α chain (30 kDa) and one light β chain (20 kDa). Four different subunit pairs exist. In contrast to earlier studies, our investigations using two-dimensional electrophoresis showed, that the majority of α and β chains of each subunit are disulfide-linked. Analysis of subunit composition of cruciferin hexamers by ion-exchange chromatography suggested that a large array of hexamers exist, composed of mixed combinations of the four subunits.  相似文献

以油菜品种085(抗菌核病)与湘油13号杂交得到的F2代作为作图群体,以RAPD标记来构建油菜连锁图谱。通过对300个10-mer随机引物的筛选,共获得200个多态性RAPD位点。经Mapmaker/EXP3.0软件处理,构建了1张含193个标记位点、19个连锁群、覆盖长度为1324cM的连锁图谱。在此基础上利用Mapmaker/QTL 1.1将抗菌核病基因定位在第四、八和十四3个连锁群上,即:S  相似文献

Fitness of hybrids between genetically modified (GM) crops and wild relatives influences the likelihood of ecological harm. We measured fitness components in spontaneous (non-GM) rapeseed x Brassica rapa hybrids in natural populations. The F1 hybrids yielded 46.9% seed output of B. rapa, were 16.9% as effective as males on B. rapa and exhibited increased self-pollination. Assuming 100% GM rapeseed cultivation, we conservatively predict < 7000 second-generation transgenic hybrids annually in the United Kingdom (i.e. approximately 20% of F1 hybrids). Conversely, whilst reduced hybrid fitness improves feasibility of bio-containment, stage projection matrices suggests broad scope for some transgenes to offset this effect by enhancing fitness.  相似文献

质粒pBN119的3.2kb BamHI片段的PvuⅡ-BglⅡ片段全顺序长为840bp,其中含油菜叶绿体16S rRNA基因5′端的140bp。通过寻找GTTC顺序,发现在395至468位核苷酸之间是tRNA~(Val)基因;在73至118位核苷酸之间是一个蛋白阅读框。和已发表的玉米叶绿体16S rRNA前导顺序进行比较,同样存在三个相应的大肠杆菌RNA聚合酶的结合位点。和大肠杆菌的启动子及相应基因作比较,表明叶绿体基因组具有很明显的原核性,但其tRNA~(Val)基因没有CCA3′顺序。在16S rRNA基因、tRNA~(Val)基因及蛋白阅读框的5′端附近均能找到一个比较稳定的茎环结构。我们推测这些茎环结构可能和位于反问重复顺序上的某些基因的转录调节有关。  相似文献

Abstract

A study was made on the changes observed in the protein, starch and soluble sugar content during development of siliquas and seeds of rapeseed grown in central Italy.

Concentration of starch and soluble sugars in the seed increases to 75 per cent dry matter during the first few weeks of pod development and then drops to minimum values. The protein increases steadily until maturity, when a level of 0.85 mg per seed is reached, equivalent to 18 per cent dry matter. The protein and starch in the hull? decrease continuously during development, while in the initial stages the soluble sugars are accumulated until they account for 33 per cent dry matter, after which they decline towards maturity.  相似文献

The donor-recipient fusion method was used to combine the cytoplasm of Brassica lournefortii with the nucleus of B. napus for the production of cytoplasmic male sterile (CMS) plants. X-ray-irradiated mesophyll protoplasts of B. tournefortii were fused with iodoacetamide (lOA)-inactivated hypocotye protoplasts of B. napus . Selective conditions of IOA concentrations and X-ray doses were determined, which resulted in recovery of fusion products and inhibition of further growth of unfused parental cells. In total, 54 plants were obtained from different fusion experiments, of which 25 were verified as cybrids or partial hybrids. Mitochondrial DNA (mtDNA) analyses using 5 mitochondrial gene probes revealed that 20 of the 25 fusion-derived plants had mtDNA either identical, or with varying degrees of similarity, to B. lournefortii . These plants were classified into four groups on the basis of pollen viability and number. Seven plants were categorised as male sterile since they did not produce pollen or had non-viable pollen. Of the male sterile plants, five had a mtDNA pattern identical to B. tournefortii and a nuclear DNA content corresponding to B. napus . The nuclear-mito-chondrial constitution of these plants thus indicates that the combination of B. tournefortii cytoplasm with the B. napus nucleus results in CMS. Furthermore, mtDNA analysis of the two additional male sterile plants which displayed a rearranged mtDNA, revealed that the only mtDNA similarity shared among all male sterile plants was specific for B. tournefortii atp6 pattern. This indicates that the atp6 region of B. tournefortii may be involved in the expression of CMS.  相似文献

Transgenic oilseed rape ( Brassica napus ) plants may remain as 'volunteer' weeds in following crops, complicating cultivation and contaminating crop yield. Volunteers can become feral as well as act as a genetic bridge for the transfer of transgenes to weedy relatives. Transgenic mitigation using genes that are positive or neutral to the crop, but deleterious to weeds, should prevent volunteer establishment, as previously intimated using a tobacco ( Nicotiana tabacum ) model. A transgenically mitigated (TM), dwarf, herbicide-resistant construct using a gibberellic acid-insensitive (Δ gai ) gene in the B. napus crop was effective in offsetting the risks of transgene establishment in volunteer populations of B. napus . This may be useful in the absence of herbicide, e.g. when wheat is rotated with oilseed rape. The TM dwarf B. napus plants grown alone had a much higher yield than the non-transgenics, but were exceedingly unfit in competition with non-transgenic tall cohorts. The reproductive fitness of TM B. napus was 0% at 2.5-cm and 4% at 5-cm spacing between glasshouse-grown plants relative to non-transgenic B. napus . Under screen-house conditions, the reproductive fitness of TM B. napus relative to non-transgenic B. napus was less than 12%, and the harvest index of the TM plants was less than 40% of that of the non-transgenic competitors. The data clearly indicate that the Δ gai gene greatly enhances the yield in a weed-free transgenic crop, but the dwarf plants can be eliminated when competing with non-transgenic cohorts (and presumably other species) when the selective herbicide is not used.  相似文献

Abstract

Brassica crops are able to hybridize with closely related wild and weedy species such as Brassica rapa and therefore these species could become recipients of transgenes from GM brassica crops, such as oilseed rape. Transgenes which protect against herbivory, such as a gene conferring the production of a Bt toxin, could increase the recipient's fitness and therefore enhance its competitiveness. We used microbial Bt and several other pesticides to exclude several guilds of herbivores and thus simulate the effect of transgene introgression on the performance of wild B. rapa. There were only minor negative effects of excluding insect herbivores on the performance of B. rapa and it appears that vertebrate herbivory has a more significant effect. The advantages and disadvantages of simulating the risks associated with GM plants are discussed. If we apply this simulation to GM risk assessment and make inferences from these findings, the implications of this for gene flow from insect-resistant transgenic plants are that it will be of little significance should it occur.  相似文献

为探讨低磷胁迫对油菜(Brassica napus)幼苗叶片蛋白酶二维酶谱的影响,以低磷胁迫下的甘蓝型油菜鼎油杂4号为试材,采用优化的非变性双向电泳技术结合蛋白酶谱检测技术获得清晰的蛋白酶分离图谱,通过图像分析获得磷胁迫下油菜叶片蛋白酶差异表达图谱。结果显示,低磷胁迫下共有6个油菜叶片蛋白酶点表达水平具有显著差异(表达量差异在2倍以上),其中3个表达水平显著上调,3个表达水平显著下调。结果提示有不同的油菜叶片蛋白酶类参与到油菜抗(耐)低磷的应答途径中,为进一步解析不同蛋白酶(同工酶)响应低磷胁迫研究打下基础。文中基于非变性等电聚焦的二维酶谱方法亦为其他逆境胁迫下不同蛋白酶及其同工酶二维酶谱响应研究提供技术参考。  相似文献

以生长在非控温控光下的4个冬性甘蓝型油菜(BrassicanapusL.)品种为供体,进行游离小孢子培养。研究发现,多数品种在开花3-7天取材最为适宜。在甘蓝型油菜小孢子培养中只有单核晚期的小孢子才可能发育成胚状体,而花药培养时处于单核早期的小孢子易于发育成胚状体。在适当花期选取发育比较一致的单核晚期小孢子培养,经数小时后,部分小孢子便开始膨大,这是小孢子发育成胚的最早标志,膨大的小孢子中,有部分形成多细胞球并进一步发育成胚。用春性甘蓝型油菜为材料进行蔗糖浓度的实验结果表明:培养3天后,在16%蔗糖培养基中存活的小孢子最多,达16.13%;培养30天后,胚状体诱导频率则以13%蔗糖浓度为最高,每花蕾可达144个胚状体。如果在16%蔗糖培养基中培养3天后,添加等体积的13%蔗糖培养基,能够大大提高胚状体的诱导频率,为仅用13%蔗糖培养基培养的3.7倍。这一实验体系正在用于抗菌核病的诱变与筛选,并作为外源基因导入的实验体系。  相似文献

通过对新选育的甘蓝型油菜高温敏感生态型不育系417S与30份中外品种(系)进行测交、杂交和回交,研究其温敏不育性的遗传特点.结果表明:(1)30份品种(系)可不同程度的恢复417S的育性,其中24个品种(系)可完全恢复417S育性,占所配组合的80.0%;5个品种(系)可高度恢复417S的不育性,占所配组合的17.2%.(2)自然条件下同一组合正反交F2群体育性分离结果不同,人工控温条件下不同组合F2分离群体中可育与不育呈现15∶1分离,回交群体中出现3∶1分离,表明417S温敏不育性受细胞质和2对隐性重叠核基因共同控制.(3)以417S与1521C构成的回交一代群体为试材,采用分离群体分组分析法(BSA法)对回交一代群体构成的不育集团和可育集团筛选育性基因的RAPD标记,从247个供试引物中筛选出2个引物BA392(5′-AGTCACTCCC-3′)、S113(5′-GACGCCACAC-3′)在不育群体和可育群体间扩增出多态性产物.经进一步对F2分离群体的单株检测,引物BA392扩增出了得到重复验证和可靠的多态性扩增产物,其产生的特异条带BA392-400bp与417S温敏雄性不育的恢复基因连锁,遗传图距为6.0cM,可作为417S温敏不育恢复基因的连锁标记.  相似文献

Recently, it has been reported that a gene (PEN1) in Arabidopsis thaliana is highly resistant to Plutella xylostella. We screened all the homologous genes of PEN1 in Arabidopsis thaliana and found that the motif of these genes was very conserved. At present, few insect resistance genes have been identified and characterized in Brassica napus. Therefore, we screened all the homologous genes containing this motif in the Brassica napus genome and systematically analyzed the basic information, conserved domain, evolutionary relationship, chromosomal localization and expression analysis of these genes. In this study, 12 PEN1 homologous genes were identified in the Brassica napus genome, which is more than the number in Arabidopsis thaliana. These genes are unevenly distributed on the 12 chromosomes in Brassica napus. Furthermore, all the PEN1 homologous genes contained light responsiveness elements, and most of the genes contained gibberellin-responsive elements, meJA-responsive elements and abscisic-acid-responsive elements. The results will provide a theoretical basis for screening insect resistance genes from the genome of Brassica napus and analyzing the molecular mechanism of insect resistance in Brassica napus.  相似文献

本研究在NLN-16和NLN-13的培养基中分别加入0.1mg/L 6-BA和0.05%的活性碳,结果表明6-BA对小孢子再生胚有明显地促进作用,再生胚的频率比对照增加26枚/皿,经分析达到显著水平;而0.05%的活性碳对小孢子再生胚促进作用不显著。对甘蓝型油菜小孢子培养再生植株的染色体加倍及移栽的研究结果表明,在小孢子培养初期加50mg/L秋水仙碱加倍效率最佳,加倍率达到67.6%。小孢子培养的再生苗移栽至大田后,采用遮阳网覆盖小苗,移栽成活率达到87.6%。 Abstract:The application of microspore culture technique was restricted because of its low frequency of embryogenesis and chromosome doubling.Two methods of enhancing the frequency of embryogenesis were employed in the study,namely,activated charcoal treatment in NLN-13 media and 6-BA treatment in NLN-16 media.The treatment with 0.05% activated charcoal produced 24 embryos per plate,which increased 1.7 embryos per plate,as compared with the treatment without activated charcoal.However,the analysis of T-test showed that it was not significant.After adding 0.1mg/L 6-BA in NLN-16 media,the frequency of embryogeny was 38.3 embryos per plate,and it was 26 embryos more per plate than that of CK.Analysis of T-test is significant.This indicates that 6-BA promotes embryogeny in microspore culture.Adding 50mg/L colchicines in NLN-16 media,the doubling frequency was 67.6%.The plantlets transplanted into field with two methods of light-covered net and plastic films were investigated.A survival rate of 87.6% was obtained using light-covered method whereas 57.7% survived using plastic film method.  相似文献

雌性不育突变体FS-M1是从甘蓝型油菜(Brassica napus)品种宁油10号中发现的.为了从细胞学角度研究FS-M1的雌性不育机理,利用荧光显微镜、扫描和透射电子显微镜观察分析了FS-M1柱头乳突细胞的授粉行为和超微结构.结果表明:花粉粒能在FS-M1乳突细胞上附着和萌发形成花粉管,但花粉管无法穿越柱头乳突细胞;开花后的FS-M1乳突细胞迅速衰退而呈干瘪萎蔫状,在衰退过程中,FS-M1柱头乳突细胞的细胞器数量减少,细胞液泡化明显,高尔基体、内质网和线粒体等一些细胞器结构被逐渐破坏.因此,推测FS-M1的雌性不育性是由于柱头乳突细胞发育异常造成的.  相似文献
