Acrocomia aculeata is an oil producing tropical palm tree with exceptional potential for producing biofuel. As the propagation of this species is often difficult because of its pronounced seed dormancy, the present work examined the morphology and the anatomy of zygotic embryos and seedlings during in vitro germination. Embryos were put in MS media supplemented with organic compounds and cultivated in the dark at 30°C for 20 days. The dry weights, lengths, and diameters of the cotyledonary petioles, haustoria, roots, ligules, and leaf sheaths of embryos obtained from mature seeds and seedlings removed from culture were measured every 2 days; anatomical and histochemical evaluations were performed on embryos and seedlings removed from culture after 2, 5, 8, 10, 12, and 15 days. Elongation of the embryo axis was observed to initiate after 2 days. Elongation of the cotyledonary petiole was observed starting on the fifth day; this is a morphological indication of germination that is associated with the formation of starch and raphides as well as the differentiation of tracheary elements. The growth of the cotyledon is due to increases in cell volumes as well as the development of a meristematic band peripheral to the haustorium. In spite of the fact that the radicle is less differentiated than the plumule, radicular development is precocious and the root emerges first, indicating the absence of morphological dormancy. Atrophy of the haustorium and the accumulation of phenolic compounds in subepidermal cell layers occur due to culturing conditions.  相似文献

BACKGROUND AND AIMS: The shrub Viburnum tinus is widely distributed in mattoral vegetation of the Mediterranean basin. The purpose of the present study was to classify the seed dormancy type and examine the requirements for embryo growth, root protrusion and shoot emergence. METHODS: Overwintered fruits were collected in western Spain in April 2001 and prepared in three ways: entire pericarp was removed, exocarp and mesocarp were removed or fruits were left intact. Fruits treated in these three ways were subjected to artificial annual temperature cycles or to constant temperature regimes for 1.5 years. KEY RESULTS: Removal of exocarp and mesocarp was necessary for embryo growth and germination. High temperature favoured dormancy alleviation and embryo growth, intermediate to low temperatures favoured root protrusion, and intermediate temperature shoot emergence. There was substantial germination at constant temperature regimes, indicating an overlap between temperature intervals suitable for the different stages of embryo and seedling development. Functionally, V. tinus has the same root and shoot emergence pattern that is described for other Viburnum species considered to have epicotyl dormancy. However, the requirement for high and low temperatures for radicle protrusion and epicotyl emergence, respectively, was missing in V. tinus; these characters are the foundation for the epicotyl dormancy classification. CONCLUSIONS: It is concluded that V. tinus does not have epicotyl dormancy. Instead, there is a combination of a weak morphophysiological dormancy and a slow germination process, where different temperatures during an annual cycle favour different development stages. The present study suggests that the first complete seedlings would emerge in the field 1.5 years after fruit maturation in October, i.e. seed dispersal during winter, embryo growth during the first summer, root protrusion and establishment during the second autumn and winter, and cotyledon emergence during the second spring.  相似文献

Butia capitata is a palm tree endemic to the Cerrado biome of Brazil and has significant potential for ornamental and food uses. In this work, we characterized the structures of the seeds and seedlings of this species to identify anatomical aspects related to its pronounced dormancy and determine the processes involved in reserve mobilization. Intact seeds, and seeds from which the operculum had been removed, were allowed to germinate and their morphology, physiology, anatomy, and histochemistry, together with those of the seedlings, were followed for 30 days. The seed coat was found to be rich in phenolic compounds and not lignified. The endosperm contains abundant protein and lipidic reserves, and the embryo has additional starch reserves. Germination occurred only in seeds with their opercula removed and involved the elongation of the cotyledon cells and meristematic activity in the “M zone” located between the embryonic axis and the proximal extremity of the embryo. The mobilization of embryonic reserves initiates during the first phase of imbibition, while the mobilization of endosperm reserves represents a post-germination event associated with the formation of a secretory epidermis and aerenchyma and the vascularization of the haustorium. Seeds with intact opercula did not germinate, but demonstrated embryonic reserve mobilization and cell elongation, indicating that dormancy in B. capitata is related to the incapacity of the embryo to dislocate the operculum.  相似文献

Aristolochiaceae have been described as having seeds with underdeveloped embryos and morphological or morphophysiological dormancy. Aristolochia galeata is a native climber found in the Cerrado biome, associated with road and gallery forest edges. The aims of this study were to investigate: embryo growth rate, morphology and seed germination parameters under different treatments. Embryos were excised to obtain embryo length at four stages: initial, seeds after coat rupture, radicle tip protrusion and cotyledon emergence from the seed coat. Germination tests were conducted at 30 °C under three nitrate concentrations (1, 10 and 20 mM), fluctuating temperature (27/20 °C) and light and dark conditions. We found that seeds have underdeveloped embryos, which take about 301 ± 178 h (±SD) to achieve seed coat rupture, another 205 ± 126 h to reach radicle protrusion and 176 ± 76 h more to the final stage of cotyledon emergence. Germinability was above 52% in all treatments, except in the dark (15%). For all treatments, average germination time was above 290 ± 123 h. Potassium nitrate increased germinability to >87%. No particular treatment was required for embryo development, but seeds in the population that continued to germinate after 1 month were probably in various states of non-deep, simple morphophysiological dormancy. Increased germinability in nitrate treatments and light requirement for germination could prevent germination under unsuitable environmental conditions and be a strategy to increase seedling establishment in the cerrado.  相似文献

Studies in reproductive ecology were made in indigenous, western American plants in the genus Marah (Cucurbitaceae), with particular attention given plants of M. oreganus occurring in the Berkeley Hills near San Francisco Bay in California. These tuberous perennials produce capsular fruits on their annual aboveground shoots; the fruits dehisce in early summer, each one exposing about three large seeds with an average seed weight of 1.05 g. The embryo of a M. oreganus seed has two thick and fleshy cotyledons packed with protein granules. The embryonic axis, with shoot and root apices, is ca. 0.5-1.0 mm long, roughly ½0 or less the length of the seed. In the Berkeley Hills dispersal of the seeds is accomplished by nocturnal rodents, after which germination begins with the fall rains and cooler temperatures of November and December. Instead of a radicle emerging first from the seed at germination, the minute radicle and epicotyl are pushed or carried far out of the seed, down into the soil, by the elongating bases of the cotyledons. These cotyledon bases, or petioles, are fused, and as they elongate they form a hollow tube that bears the embryonic axis at its extreme tip. The cotyledonary petiole tube ceases elongation by January, when it may be 5-25 or more cm long in a seedling of M. oreganus. Then, from its tip, the radicle grows downward and the epicotyl upward—up the hollow petiole tube. The green shoot (epicotyl) reaches the soil surface by early March in this area, completes the first season's growth, and dries up by late May, when the arid summer season is beginning. But even before the epicotyl grows out of the petiole tube and above ground, the seedling's hypocotyl begins to enlarge, forming a tuber. The fleshy cotyledon blades remain in the seed coat below ground, and some food from the blades is transferred to the tuber that produces shoots in the following growing seasons. This pattern of germination and seedling establishment is now known for species of Marah and for a very few other dicotyledonous plants, all of them growing mainly in areas of hot and dry habitat that are generally referred to as having Mediterranean climate. This elongation of the fused hypogeal cotyledons is considered a complex adaptation in dicotyledons that helps ensure fast and successful seedling establishment in seasonally arid areas such as “Mediterranean” California.  相似文献

It is not known how embryos of seeds of the Pinaceae protrude from their enclosing tissues to complete germination. Prior to protrusion of the radicle there is an increase in endo-β-1,4-mannanase (EC 3.2.1.78) activity associated with weakening of the micropylar megagametophyte/nucellus from seeds of white spruce ( Picea glauca [Moench.] Voss). Mannanase activity is present as three isoforms (pI values 5.0, 4.8, 4.7) in both the embryo and surrounding structures (megagametophyte and nucellus) prior to and during imbibition. Activity of all the isoforms increases in the chalazal and micropylar megagametophyte during germination. Activity then declines after the testa splits, typically 1 day prior to radicle protrusion, due partially to its leaching from the seed into the surrounding water. Activity increases in the cotyledons and axis as the embryo commences elongation. Seeds from dormant seedlots exhibit a lower germination percentage, relative to seeds from nondormant seedlots, and the force necessary for the embryo to puncture the surrounding structures tends to be greater. Although similar mannanase activities are present in unimbibed seeds of dormant and nondormant seedlots, during germination, enzyme activity in seeds of dormant seedlots is lower. Moist chilling alleviates dormancy in the seeds of the Pinaceae and, during 3 weeks of this treatment, mannanase activity slowly increases. After 3 weeks of moist chilling and regardless of whether the seedlot was dormant or not prior to moist chilling, the force necessary to puncture the micropylar megagametophyte and nucellus is lower, and the speed of germination greater. Seeds from previously dormant seedlots also complete germination to a greater percentage, relative to unchilled seeds from dormant seedlots. Upon transfer to 25°C, mannanase activity in moist-chilled seeds decreases during germination of all seedlots regardless of their previous dormancy status.  相似文献

Little is known about the control exerted by hormones in specific tissues during germination and post‐germinative development in monocot seeds, whose embryos have complex structures and can remain dormant for long periods of time. Here the tissue‐specific hormonal profile of macaw palm (Acrocomia aculeata) seeds overcoming dormancy and seedling during initial development was examined. Endogenous hormonal concentrations were determined in the cotyledonary petiole, haustorium, operculum, endosperm adjacent to the embryo and peripheral endosperm of dry dormant seeds, imbibed seeds trapped in phase I of germination, and germinating (phase 2 and phase 3) seeds 2, 5, 10 and 15 days after sowing. Evaluations were performed on seeds treated for overcoming dormancy by removal of the operculum and by immersion in a gibberellic acid (GA3) solution. Removal of the operculum effectively helped in overcoming dormancy, which was associated with the synthesis of active gibberellins (GAs) and cytokinins (CKs), as well as reductions of abscisic acid (ABA) in the cotyledonary petiole. In imbibed seeds trapped in phase I of germination, exogenous GA3 caused an increase in active GAs in the cotyledonary petiole and operculum and reduction in ABA in the operculum. Initial seedling development was associated with increases in the CK/auxin ratio in the haustorium and GA levels in the endosperm which is possibly related to the mobilization of metabolic reserves. Increases in salicylic acid (SA) and jasmonic acid (JA) concentrations were associated with the development of the vegetative axis. Hormones play a crucial tissue‐specific role in the control of dormancy, germination and initial development of seedlings in macaw palm, including a central role not only for GAs and ABA, but also for CKs and other hormones.  相似文献

The seed germination characteristics of Maianthemum dilatatum were investigated in a laboratory experiment and the results compared with those of other species in the subfamily Asparagoideae, LILIACEAE (Engler's system). M. dilatatum seeds mature in late September to October in montane to subalpine areas across Japan. Germination percentages and rates were low for fresh seeds at 10 to 30°C. Seeds cold stratified for 4 months or longer showed increased germination percentages and rates. The seeds lost germinability with decreasing moisture content. The seeds germinated well in dark conditions. The process of germination from the cotyledonary sheath/petiole breaking through the seed coat to the appearance of the first and second leaves was examined. After emergence of the cotyledonary sheath/petiole, a root emerged from it, and additional roots appeared after 1–2 months. The plumule emerged from the cotyledonary sheath/petiole after the seedling had three roots. Seeds dispersed in autumn, and germinate slowly in the next spring after exposure to low temperature even though dark condition as buried seed. We compared the seed germination characteristics among species in the Asparagoideae or with other recent taxonomy, and found that seedlings of Paris verticillate and Trillium apetalon, which belong to Melanthiaceae, and Streptopus streptopoides Var. japonica and Clintonia udensis, which belong to Liliaceae (linear cotyledon), were different from Asparagaceae, showing a globose cotyledon in the APG II.  相似文献

The soybean callus assay was used to study the effect of high oxygen tensions on the cytokinin levels of Leucadendron daphnoides Meisn. seed, where dormancy is apparently due to the restricting effect of the seed coat on oxygen diffusion to the embryo. High oxygen tensions led to a six-fold increase in germination compared to seed incubated in air and resulted in significant increases in butanol soluble cytokinins prior to visible germination. It is suggested that the primary effect of oxygen is to increase the rate of respiration and thus, to provide the energy required for the synthesis of butanol soluble cytokinins which leads to cotyledon expansion and subsequent radicle elongation. Present indications are that untreated seeds remain dormant due to low concentrations of butanol soluble cytokinins in their embryos.  相似文献

As the hypocotyl and epicotyl emerge from the micropylar collar, the seminal adventitious roots become apparent and the cotyledonary sheath surrounding the epicotyl emerges as a coleoptile-like structure. Subsequent growth results in the elongation of the first leaves beyond the cotyledonary sheath and the rapid elongation of the adventitious root system. The vascular transition occurs between the cotyledon and the radicle. The sheath is considered to be only analogous to the coleoptile of the grasses.  相似文献

Dormancy-breaking and seed germination studies in genus Lilium reveal that the majority of Lilium spp. studied have an underdeveloped embryo at maturity, which grows inside the seed before the radicle emerges. Additionally, the embryo, radicle or cotyledon has a physiological component of dormancy; thus, Lilium seeds have morphophysiological dormancy (MPD). A previous study suggested that seeds of Lilium polyphyllum have MPD but the study did not investigate the development of the embryo, which is one of the main criteria to determine MPD in seeds. To test this hypothesis, we investigated embryo growth and emergence of radicles and epicotyls in seeds over a range of temperatures. At maturity, seeds had underdeveloped embryos which developed fully at warm temperature within 6 weeks. Immediately after embryo growth, radicles also emerged at warm temperatures. However, epicotyls failed to emerge soon after radicle emergence. Epicotyls emerged from >90% seeds with an emerged radicle only after they were subjected to 2 weeks of cold moist stratification. The overall temperature requirements for dormancy-breaking and seed germination indicate a non-deep simple epicotyl MPD in L. polyphyllum.  相似文献

Background and AimsInternal seed morphological traits such as embryo characteristics and nutritive tissue can vary considerably within a plant lineage. These traits play a prominent role in germination processes and the success of seedling establishment, and are therefore under high selective pressure, especially in environments hostile to seedlings, such as arid, saline or highly dynamic habitats. We investigated the relationships of seed internal morphology and germination characteristics of 84 species of Amaranthaceae s.l., a family with numerous lineages that have adapted to stressful growing conditions.MethodsWe used seed cross-sections to assess embryo type and the ratios of embryo to seed surface and radicle to cotyledon length. Furthermore, seed mass, mean time to germination, habitat preferences and further plant traits such as C3 or C4 photosynthesis and life form were compiled for each species. Data were analysed using phylogenetic comparative methods.Key resultsWe found embryo type (λ = 1), log seed mass (λ = 0.86) and the ratio of embryo to seed size (λ = 0.78) to be evolutionarily stable, with an annular embryo as ancestral in the family. Linked to shifts to the three derived embryos types (spiral, horseshoe-shaped and curved) is an increase in the ratio of root to cotyledon length and a reduction of nutritive tissue. We observed stabilizing selection towards seeds with relatively large embryos with longer radicles and less nutritive tissue that are able to germinate faster, especially in lineages with C4 photosynthesis and/or salt tolerance.ConclusionsWe conclude that the evolutionary shift of nutrient storage from perisperm to embryo provides an ecological advantage in extreme environments, because it enables faster germination and seedling establishment. Furthermore, the evolutionary shift towards a higher ratio of root to cotyledon length especially in small-seeded Amaranthaceae growing in saline habitats can provide an ecological advantage for fast seedling establishment.  相似文献

The in vitro germination of excised embryos can break dormancy rapidly and shorten the time required to produce seedlings, speeding up olive breeding programmes as well as rootstock production. In this study, the in vitro germination potential of four Sicilian olive cultivars was evaluated during two years of experiments, using explants with three different morphological configurations that represent three different degrees of embryo exposure: (1) intact stoneless seeds containing the embryo, the endosperm and the seed coat (Emb+En+SC), (2) seeds without the seed coat (Emb+En) and (3) naked, isolated embryos (seed coat and endosperm both removed: Emb). Differences were found in the germination percentages and timing due to both genotype and explant type. The root and shoot meristems, the radicle-hypocotyl axis, the provascular tissues and embryo storage reserves were identified as embryo anatomical structures which could influence germination capacity. Observation of these structures, however, indicated similar germination potential among cultivars, suggesting possible differences in other dormancy factors. In spite of variation in cultivar performance, after 60 days of in vitro culture all cultivars demonstrated the highest germination of naked embryos (explant type 3) and lowest for stoneless seeds (explant type 1); stoneless seeds without the seedcoat (explant type 2) showed intermediate germination percentages.  相似文献

Somatic embryogenesis was obtained from cotyledon and mature zygotic embryo callus cultures of Terminalia chebula Retz. Callus cultures of cotyledon and mature zygotic embryo were initiated on induction medium containing Murashige and Skoog (MS) nutrients with 1.0 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D) either 0.01 or 0.1 mg/l Kinetin and 30 g/l sucrose. Induction of somatic embryogenesis, proliferation and development was obtained through different culture passages. Embryogenic cotyledon callus with globular somatic embryos was obtained on MS basal medium supplemented with 50 g/l sucrose. Globular somatic embryos were observed from mature zygotic embryo callus on induction medium. Different stages of somatic embryo development from cotyledon and mature zygotic embryo calluses were observed on MS basal medium supplemented with 50 g/l sucrose after 4 weeks of culture. Histological studies have revealed the developmental stages of somatic embryos. A maximum of 40.3±1.45 cotyledonary somatic embryos/callus was obtained from mature zygotic embryo compared to 7.70±0.37 cotyledonary somatic embryos/callus initiated from cotyledons. Germination of somatic embryos and conversion to plants were achieved. Highest frequency of germination (46.66±0.88) of somatic embryos was obtained on MS basal medium containing benzyladenine (0.5 mg/l) with 30 g/l sucrose.  相似文献

The soybean callus assay was used to study the effect of stratification on the cytokinin levels of the embryo dormant seed of Protea compacta R.Br. and the seed of Leucadendron dapbnoides Meisn., where dormancy is coat imposed. Chilling the seed for 30 days increased germination significantly, and resulted in a simultaneous increase in the butanol soluble cytokinins of both species. It would appear as if these compounds are either synthesized or released from a bound form in embryo dormant seed. In contrast, an interconversion from water soluble to butanol soluble cytokïnins appears to account for the increase where dormancy is coat imposed. The results also indicate that for germination to take place a threshold concentration of cytokinin may be required. It is suggested that the increase in butanol soluble cytokinins may lead to the breaking of dormancy, probably by increasing radicle elongation and/or cotyledon expansion.  相似文献

Seeds of Acer pseudoplatanus L. are usually considered to show only testa-imposed dormancy, but a transient embryo dormancy has also been identified at the time of fruit dispersal. Even embryos that did not show full dormancy at this stage possessed low germinative vigour. Removal of embryo dormancy and the development of increased germination potential did not require the low temperatures necessary for the removal of testa-imposed dormancy in this species. Germination rates of embryos from freshly harvested or briefly stored fruits were accelerated by removal of cotyledonary tissue, the most rapid responses occurring in isolated embryonic axes following complete removal of both cotyledons. Longer storage reduced this effect because of the increases in germinative vigour of whole embryos. Abscisic acid (ABA) reinforced embryo dormancy in embryos from freshly harvested or briefly stored fruits and also reduced germination rates in similarly derived isolated embryonic axes. This response to ABA also became progressively less marked as the storage period was extended. Loss of embryo dormancy was correlated with a reduction in endogenous abscisic acid levels in both whole embryos and cotyledons, suggesting that endogenous ABA contributes to the regulation of embryo dormancy in these seeds. There are no indications, however, that endogenous ABA is directly implicated in the low temperature processes associated with the removal of testa-imposed dormancy. The relevance of embryo dormancy in the intact seed of A. pseudoplatanus is discussed.  相似文献

利用抑制物生物测定法和酸蚀技术研究了血皮槭种子休眠的原因。血皮槭种子吸水是一个非常缓慢的过程,在140 h以后种子含水量才能达到68%左右。酸蚀处理种子3 h,虽然没有加快种子的吸水速率,但能较好得使果皮变薄,也不影响种子的生活力。种子的各部位(果皮、种皮、子叶、胚根)均含有抑制物质,对小白菜种子的发芽率及胚根生长有很强的抑制作用,子叶各种处理水浸提液的抑制作用最强,果皮和种皮次之。血皮槭种子休眠主要由种壳机械障碍和种胚生理休眠两重因素导致,因此如何克服致密果壳而使激素能接触生理休眠的种胚是打破其种子休眠的关键技术。  相似文献

Both germination and seedling establishment in palm trees are strongly influenced by the morphoanatomy of the fruits, although the interactions of these processes with ecophysiological aspects are not yet well understood. The present work evaluated structural and physiological aspects of seed germination in Attalea vitrivir, a species living under the seasonal climate of the Cerrado (Brazilian savanna) biome. We studied morphology, anatomy and histochemistry of the fruits and seedlings, the effects on germination of the pericarp, of diaspore storage conditions, germination temperature, removal of the operculum, and of gibberellic acid (GA3) application, and characterized the imbibition process of the seeds. Germination depends on a series of complex interactions between structures and physiological processes. The pericarp protects the seed and also causes physical dormancy that, when interacting with the temperature regime, can delay germination until subsequent rainy periods. Some seeds demonstrated non-profound physiological dormancy associated with restricted germination imposed by the operculum. Germination and initial development result from the elongation of the cotyledon cells and are affected by the activities of two distinct meristems in the proximal region of the embryo. The haustorium develops an invaginated secretory epithelium and aerenchyma, and actively participates in seed reserve mobilization. The adaptation of A. vitrivir to the Cerrado environment, with a strongly seasonal climate, is favored by its diaspore structure, by the abundance of endosperm reserves that allow the seedlings to survive for a long period above soil level, and by a seedling development pattern that protects the vegetative axis by deep burial.  相似文献

Associations of recalcitrance and dormancy are rare, and little information is available concerning the structure of seeds demonstrating this type of behavior or their ecological implications. Mauritia flexuosa is a palm tree associated with swampy environments in the Amazon rainforest and areas of Cerrado (neotropical savanna), the latter biome having marked climatic seasonality. We describe the structures and physiological aspects of the seeds and seedlings of this species to examine its adaptations to swampy environments and its germination control mechanisms, as well as the relationship between recalcitrance and dormancy in terms of reproductive success. Morphoanatomy and histochemistry of the seeds and seedlings were evaluated using standard methodologies, and the effects of temperature and operculum removal on germination were investigated. Differentiated tracheal elements in the embryo are associated with recalcitrance, and presence of numerous stomata on the embryo and the abundance of secondary metabolite compounds in the seedling indicate their adaptations to swampy environments. The inability of the embryo to overcome the resistance of the adjacent tissues determines dormancy of the physiological type, thus the removal of the operculum is an efficient method for promoting propagation. Cellular elongation in the cotyledon promotes seedling protrusion. Mobilization of endosperm reserves is achieved by symplastic flux and is associated with overcoming of dormancy by weakening the tissues adjacent to the embryo. The association between recalcitrance, which favors adaptation to swampy environments, and dormancy, which favors dispersal, is crucial for the reproductive success of this species and its broad geographical distribution.  相似文献
