A total of nine enzymes coded by 14 loci were assayed for each of six populations (from the north-eastern Atlantic and the Mediterranean) of two sea bream subspecies ( Diplodus sargus sargus and Diplodus sargus cadenati ). Diagnostic alleles were observed for each subspecies, although there were several common alleles. Estimates of variance in allele frequencies among samples ( F ST) revealed significant differences ( P  < 0.05) among both subspecies. Genetic divergence was found between Atlantic and Mediterranean samples: values for genetic distances were higher than 0.163. Furthermore, D. sargus cadenati populations displayed a higher mean weight and length than D. sargus sargus populations and significant differences in growth were found among subspecies and populations. Some studies on insects have found a relationship between habitat structure and investment in dispersal-related traits. In this study we compared the morphology of the butterfly Melitaea cinxia from five sites on the large Baltic island Öland that differed markedly in degree of fragmentation and size. Both wild-caught adults and individuals reared in a split-plot design were compared. We found significant site differences in size-adjusted thorax mass and total body mass. Male thorax mass was on average larger among the sites with the highest degree of habitat fragmentation. However, due to significant sex–site interactions, males and females may have adapted differently to the habitat fragmentation. Using museum specimens, we also analysed changes in morphology, finding an increase in size (measured as head and thorax width) over time. Thorax width appears to have increased among females and decreased among males. First record of fossil Mesozoic Ctenopoda (Crustacea, Cladocera)   总被引:1,自引:0,他引:1  
Pre-Pleistocene representatives of the crustacean order Ctenopoda Sars, 1865 are unknown. Here we describe Mesozoic fossil specimens of ctenopods from two localities in Mongolia: Khotont (Jurassic/Cretaceous boundary, about 145 Mya) and Khutel Khara (Lower Cretaceous, about 129 Mya). All specimens apparently belonged to the tribe Latonini Korovchinsky, 1986 of the subfamily Sidinae Baird, 1850. At the Khotont site, ctenopods were the most numerous microscopic animal fossils. We assigned these cladocerans to a new genus and species Archelatona zherikhini gen. nov., sp. nov. The standard model for the migration of the monarch butterfly in western North America has hitherto been movement in the autumn to overwintering sites in coastal California, followed by a return inland by most individuals in the spring. This model is based largely on observational and limited tagging and recovery data. In this paper we test the model by plotting many years of museum and collection records on a monthly basis on a map of the region. Our plots suggest a movement of Oregon, Washington and other north-western populations of summer butterflies to California in the autumn, but movement of more north-easterly populations (e.g. from Idaho and Montana) along two pathways through Nevada, Utah and Arizona to Mexico. The more westerly of these two pathways may follow the Colorado River south as indicated by museum records and seasonal temperature data. The eastern pathway may enter northern Utah along the western scarp of the Wasatch Mountains and run south through Utah and Arizona. Further analysis of distributions suggests that monarch butterflies in the American West occur primarily along rivers, and there are observations indicating that autumn migrants often follow riparian corridors. The genetic diversity and structure of four populations of the cycad Zamia loddigesii were studied throughout its range in Mexico. Allozyme electrophoresis of 15 loci was conducted. The mean number of alleles per locus was 1.80 ± 0.09, the percentage of polymorphic loci was 66.6 ± 5.4, and the expected heterozygosity was 0.266 ± 0.02. The results indicated that the genetic diversity was relatively higher, with respect to tropical tree species and other cycads. The genetic variation explained by differences among populations was 18%. On average, gene flow between paired populations was similar ( Nm  = 1.6) to other tropical forest trees and cycad species. Our results indicated that the geographical isolation among populations of Z. loddigesii generated allele loss, as well as a clinal variation in the frequencies of two loci ( MDH and MNR2 ), in relation to the latitudinal distribution of populations. The populations have become fragmented due to increasingly higher pressure of habitat conversion and disturbance. We studied population genetic variation and structure in the fire ant Solenopsis invicta using nuclear genotypic and mitochondrial DNA (mtDNA) sequence data obtained from samples collected throughout its native range. Geographic populations are strongly differentiated at both genomes, with such structure more pronounced in Brazil than in Argentina. Higher-level regional structure is evident from the occurrence of isolation-by-distance patterns among populations, the recognition of clusters of genetically similar, geographically adjacent populations by ordination analysis, and the detection of an mtDNA discontinuity between Argentina and Brazil coinciding with a previously identified landform of biogeographical relevance. Multiple lines of evidence from both genomes suggest that the ancestors of the ants we studied resembled extant northern Argentine S. invicta , and that existing Brazilian populations were established more recently by serial long-distance colonizations and/or range expansions. The most compelling evidence for this is the corresponding increase in F K (a measure of divergence from a hypothetical ancestor) and decrease in genetic diversity with distance from the Corrientes population in northern Argentina. Relatively deep sequence divergence among several mtDNA clades, coupled with geographical partitioning of many of them, suggests prolonged occupation of South America by S. invicta in more-or-less isolated regional populations. Such populations appear, in some cases, to have come into secondary contact without regaining the capacity to freely interbreed. There have been numerous attempts to resolve the taxonomy of the two-horned chamaeleons of East Africa. However, the high levels of intraspecific variation and reported sympatry of morphologically distinct taxa indicate that their current classification is unsatisfactory. Tissue samples were collected from specimens from most mountain massifs (excluding the Nguu) where two-horned chamaeleons are known to occur and were used to sequence partial 12S and 16S rRNA, as well as ND2 mtDNA genes. These specimens and further museum material were used to review morphological variation and to define discriminating characters for each taxon. Phylogenetic analyses of gene sequences show clear genetic divergence between allopatric populations, although two lineages occur in sympatry in the East Usambara Mountains, and is supported by divergent morphology. In light of these results a formal revision of the taxonomy of all East African two-horned chamaeleons is proposed and seven species are recognized ( Kinyongia boehmei , K. fischeri , K. matschiei , K. multituberculata , K. tavetana , K. uluguruensis and K. vosseleri ). These taxa form a monophyletic group except for K. uluguruensis , which seems to be more closely related to one-horned species. Most of these taxa have been previously described and subsequently reduced to synonyms. Two other previously described two-horned taxa are not recognized as valid: Chamaeleo tornieri is considered species inquiriendae , and C. fischeri werneri is placed in synonymy with K. multituberculata . Historical museum specimens are valuable for exploring population genetics and evolutionary questions because they can provide snapshots of morphological and genetic characteristics from populations over space and time. Unfortunately, DNA found in older museum specimens is frequently degraded, so obtaining genotypes from many individual samples necessary for rigorous molecular population genetic studies is challenging. Previous studies have varied greatly in their success at obtaining genotypes from older preserved insect material. Many well-intentioned collection curators have used research results showing poor preservation of DNA preserved in museum specimens to inform curatorial best practices, in some cases choosing not to allow DNA extraction by destructive sampling because, in their estimation, the likelihood of success would be low. Recent methodological advances in DNA extraction, amplification, and genotyping have allowed some researchers to include mid-19th century samples in molecular genetic analyses. Here we present a robust, high-throughput, and low-cost DNA extraction and genotyping protocol for historical insect specimens employing restriction digests of PCR products followed by high sensitivity electrophoresis. Using this technique, we obtained mitochondrial haplotypes for 100% of 48 New World Junonia butterfly specimens (Nymphalidae) ranging in age from pre-1813 to 1909 and show that the haplotype frequencies obtained are statistically indistinguishable from 20th-century and contemporary reference populations of Junonia (1632 specimens) matched by geographic region. As most extant insect specimens were collected after 1813, based on our findings we would expect that many or even most pinned specimens preserved in museum collections contain usable DNA for mitochondrial haplotyping.  相似文献

More than 20 populations of the cave-dwelling characid Astyanax occur within a restricted karst area in Mexico. The fish possess reduced eyes without lenses and visual cells. It is still an open question as to whether this condition evolved convergently after multiple entries of the surface ancestor into the different caves or whether a single cave ancestor, already characterized by reduced eyes, spread secondarily within them. In the crosses between specific populations, specimens appear that deviate considerably from those of the parents. They possess larger and better-developed eyes with histologically intact lenses and visual cells; they thus have the structural potential for vision. This indicates that in different cave populations, different mutations in the eye gene system have occurred. In cases where these non-functional rudimentary genes are recombined in hybrid specimens, gene expression may be restored. The glacial–interglacial cycles have caused severe range modifications of species' distributions. In Europe, thermophilic species had to retreat into geographically distinct southern refugia during glaciations. This process produced strong genetic imprints, which are still detectable by the present pattern of genetic differentiation and the distribution of regional diversity. To reveal the biogeographical imprints in the western Mediterranean, we analysed 26 populations of the butterfly Maniola jurtina spread over large areas of its European and North African distribution range. The samples were analysed using allozyme electrophoresis. We detected three genetic groups, divided into Western Europe, Central/Eastern Europe, and Italy with the Maghreb. The North African samples randomly cluster within the Italian samples. Even the population sampled in Morocco is genetically closely related to these samples and not to the geographically neighbouring Iberian ones. Parameters of genetic diversity showed similar values over the whole study area. The observed genetic pattern reflects possible glacial refugia in Europe located in the Iberian Peninsula and the Balkans. Systematic research on bivalved molluscs (Mollusca: Bivalvia = Pelecypoda) is briefly reviewed in an introduction to a series of papers focusing on seven of the larger branches of the bivalve tree. These are presented in an attempt to summarize current knowledge, to stimulate new research and to highlight needs for future research focus. We used mitochondrial cytochrome  b sequences (up to 778 bp) and starch gel electrophoresis (45 loci) to examine the phylogeographical history of 39 populations of the California newt, Taricha torosa . Phylogenetic and population genetic methods were integrated to infer history at deep and shallow time depths. Using a molecular clock, the subspecies T. t. torosa and T. t. sierrae were estimated to have diverged 7–13 Mya. Within T. t. torosa, genetically differentiated groups were identified along coastal California, in southern California, and in the southern Sierra Nevada. The coastal group exhibited isolation by distance, but a lack of genetic variation north of present-day Monterey was indicative of a recent range expansion. In southern California, a disjunct population in central San Diego County was genetically diverged from coastal populations to the north (Nei's genetic distance of 0.113). However, mtDNA and protein data were geographically discordant regarding the boundary between the coastal and southern Californian groups, and a biogeographical scenario was developed to account for this discordance. The southern Sierran clade of T. t. torosa was weakly diverged from coastal populations for mtDNA sequence variation, yet was strongly differentiated for allozyme variation (Nei's genetic distance of 0.17–0.20). Populations of T. t. sierrae exhibited substantial population structure, and showed a steeper pattern of isolation by distance than did coastal populations of T. t. torosa . According to current systematics, Festuca inops and F. gracilior are two distinct species. However, they are hardly distinguishable from each other on the basis of their morphological characters. Festuca inops is considered a diploid species endemic to Italy, while F. gracilior has a discontinuous distribution area, apparently related to chromosomal levels: diploid populations in Italy and south-east France, tetraploid populations in north-east Spain. The diploid populations of both taxa from Italy and south-east France are investigated in the present study. Nearly 1000 exsiccata were examined and morphometric analysis was carried out on macro- and micromorphological features of 119 specimens (including type-specimens) and on 20 natural populations (including loci classici ). All these data showed that the two species should be referred to a single taxon, for which the rank of species seems to be appropriate. This result is supported by karyological, ecological and chorological data and was confirmed by the results of ISSR analysis. Pleistocene climatic oscillations strongly influenced the genetic composition of many species which are often divided into several genetic lineages. In this context, we studied the allozymes of a common and widely distributed butterfly, the common blue Polyommatus icarus, over a large part of Europe. The species had a rather high genetic diversity within populations with a strikingly high mean number of alleles per locus (2.98). In contrast, differentiation between populations was very low ( F ST: 0.0187). Only a marginal trend of decline in genetic diversity from the south to the north was observed. Isolation-by-distance existed on a European scale ( r =  0.826), but not at a regional level. Regional differentiation between populations in western Germany was extremely low ( F ST: 0.0041). It is probable that P. icarus was widely distributed in the Mediterranean region during the last ice age and expanded into central Europe in the postglacial period without major genetic erosion. The evolution of viviparity in squamates has been the focus of much scientific attention in previous years. In particular, the possibility of the transition from viviparity back to oviparity has been the subject of a vigorous debate. Some studies have suggested this reversal is more frequent than previously thought. However, none of them provide conclusive evidence. We investigated this problem by studying the phylogenetic relationships between oviparous and viviparous lineages of the reproductively bimodal lizard species Zootoca vivipara . Our results show that viviparous populations are not monophyletic, and that several evolutionary transitions in parity mode have occurred. The most parsimonious scenario involves a single origin of viviparity followed by a reversal back to oviparity. Leiopotherapon unicolor is the most widespread freshwater fish species in Australia. A comprehensive allozyme and mitochondrial DNA 16S rRNA data set was assembled from 141 specimens of L. unicolor collected Australia-wide in order to test for cryptic speciation in this far-ranging species. Surprisingly, little genetic diversity was observed within L. unicolor and provided no evidence for the existence of cryptic species within this lineage. In contrast, a small sample set of L. aheneus used as the outgroup showed two highly divergent haplotypes strongly suggestive of cryptic speciation. L. unicolor has a number of ecological and life history attributes that may explain the lack of significant genetic divergence over substantial geographical distances. The occurrence of other widespread fish and crustacean species that also display only limited genetic diversity indicate that climate conditions more favourable to dispersal across central and northern Australia than is suggested by the extent of present-day aridity have occurred in the relatively recent geological past.  © 2006 The Linnean Society of London, Biological Journal of the Linnean Society , 2006, 87 , 537–552.  相似文献

Leptodactylus fuscus is a neotropical frog ranging from Panamá to Argentina, to the east of the Andes mountains, and also inhabiting Margarita, Trinidad, and the Tobago islands. We performed phylogenetic analyses of 12S rRNA, 16S rRNA, tRNA-Leu, and ND1 mitochondrial (mt) DNA sequences from specimens collected across the geographic distribution of L. fuscus to examine two alternative hypotheses: (i) L. fuscus is a single, widely distributed species, or (ii) L. fuscus is a species complex. We tested statistically for geographic association and partitioning of genetic variation among mtDNA clades. The mtDNA data supported the hypothesis of several cryptic species within L. fuscus. Unlinked mtDNA and nuclear markers supported independently the distinctness of a 'northern' phylogenetic unit. In addition, the mtDNA data divided the southern populations into two clades that showed no sister relationship to each other, consistent with high differentiation and lack of gene flow among southern populations as suggested by allozyme data. Concordance between mtDNA and allozyme patterns suggests that cryptic speciation has occurred in L. fuscus without morphological or call differentiation. This study illustrates a case in which lineage splitting during the speciation process took place without divergence in reproductive isolation mechanisms (e.g. advertisement call in frogs), contrary to expectations predicted using a biological species framework.  © 2006 The Linnean Society of London, Biological Journal of the Linnean Society, 2006, 87 , 325–341. Some aquatic taxa (fishes, snakes) exhibit a puzzling form of sexual dimorphism: males have a more rugose body surface than do conspecific females. As a first step towards understanding the biological significance of this phenomenon, the nature and correlates (sex, size, body condition, season, and latitude) of scale rugosity was quantified in preserved museum specimens of four species of sea snakes (Family Hydrophiidae): Astrotia stokesii , Emydocephalus annulatus , Hydrophis elegans , and Lapemis hardwickii . Males are more rugose than conspecific females in all four taxa, on ventral as well as dorsal scales. The position of rugosities on the body differs among species. For example, rugosities are found over the entire body in E. annulatus , but are concentrated anteriorly in L. hardwickii and posteriorly in H. elegans . Females possess rugosities that are similar to those of conspecific males, but smaller (in female E. annulatus , they are visible only with scanning electron microscopy analysis). The degree of male rugosity varies seasonally in at least two species ( E. annulatus , L. hardwickii ), being most pronounced during the winter breeding season. Missing data are frequent in morphometric studies of both fossil and recent material. A common method of addressing the problem of missing data is to omit combinations of characters and specimens from subsequent analyses; however, omitting different subsets of characters and specimens can affect both the statistical robustness of the analyses and the resulting biological interpretations. We describe a method of examining all possible subsets of complete data and of scoring each subset by the 'condition' (ratio of first eigenvalue to second, or of second to first, depending on context) of the corresponding covariance or correlation matrix, and subsequently choosing the submatrix that either optimizes one of these criteria or matches the estimated condition of the original data matrix. We then describe an extension of this method that can be used to choose the 'best' characters and specimens for which some specified proportion of missing data can be estimated using standard imputation techniques such as the expectation-maximization algorithm or multiple imputation. The methods are illustrated with published and unpublished data sets on fossil and extant vertebrates. Chromosome numbers and morphology in 22 populations belonging to 11 taxa of Aconitum subgenus Lycoctonum (Ranunculaceae) from China were studied. Some taxa were diploid, with 2 n  = 16, but four species and two varieties were found to be tetraploid, with 2 n  = 32. They are concentrated in the Hengduan Mountains region in south-west China, indicating that polyploidy could have played an important role in the speciation of the subgenus in this region, one of the areas of the world with a high concentration of endemic species.
