组织纤溶酶原激活剂(tPA)是一种较理想的溶血栓药物,本研究采用其突变体-长效组织纤溶酶原激活剂(LAtPA)的cDNA作为目的基因,将它插入羊β-乳球蛋白(BLG)基因起始密码之前,使LAtPA的转录、翻译受控于BLG基因的5'、3'序列,再将所构建的BLG-LAtPA用显微注射方法建立转基因鼠,经点杂交筛选和Southern印迹鉴定,获得2只LAtPA基因整合阳性鼠,并在阳性母鼠乳汁中检测到有  相似文献

以矮牵牛(Petunia hybrida L.)栽培品种为材料,取开放前的花蕾分离mRNA,反转录合成cDNA,以cDNA为模板,通过PCR扩增,对获得的目的片段进行序列分析。结果表明,分离的目的片段含有686个核苷酸(含有起始密码和终止密码)。核苷酸序列与文献报道相比,同源率为99.6%,只有3个碱基发生改变,5’端的MADS盒区域完全相同。将得到的矮牵牛花同源异型基因fbp2的cDNA(yfbp2)与CaMV355启动子和NOS3’终止子融合,构建了表达载体pBBP2。表达载体通过农杆菌(Agrobacterium tumefaciens)LBA4404(pAL4404)介导转化烟草(NicotianatabacumL.)叶片,在含有100mg/L卡那霉素的抗性培养基上再生成株。对抗性株进行总DNASouthern杂交和总RNA的点杂交,证明目的基因已导入烟草细胞中,整合到烟草基因组上,并且在烟草细胞中转录。同源异型基因fbp2导入烟草后导致烟草花型改变,在雄蕊上产生了花瓣。  相似文献

根据已发表的序列,分别在鸡贫血病毒(CAV)环形基因组DNA(全长2.3kb)的EcoRI位点和BamHI位点的两侧选择适当序列合成两对引物,用PCR技术,从斑点杂交检测到病毒核酸的CAV感染的MDCCRP1细胞基因组DNA中,分别扩增出包含EcoRI和BamHI分割开的病毒基因组两部分(1.5kb和0.8kb)约1.5kb和约1.25kb的两个片段。再将其中相应序列拼接克隆进pUC18载体,获得包含CAV全基因组序列DNA片段的克隆质粒pCAV2.4。酶切分析表明,该质粒具有预期的BamHI位点、PstI位点、HindⅢ位点,而预期的EcoRI位点消失。重组质粒插入DNA片段的两端序列分析表明,质粒pCAV2.4是包含CAV全基因组序列的重组质粒,插入DNA片段序列中的EcoRI位点序列发生了一个碱基突变。  相似文献

以赤霉素(GA)处理后的G2豌豆为材料,构建了一个2.0×106的cDNA文库,用随机筛选法从该cDNA文库中得到一个完整的cDNA.其中1115bp全序列分析表明,它编码了豌豆5S核糖体RNA(5SribosomalRNA,5SrRNA),基因内部存在着与核糖体蛋白L5及5SrRNA结合蛋白(5SrRNABindingProtein)同源的区段,这些同源区段是蛋白与RNA结合的位点.基因内部还存在着大量的重复序列.  相似文献

通过构建可富集人胰岛素基因的λ噬菌体文库,克隆了人胰岛素基因组基因.首先从中国人血液白细胞中提取到人基因组DNA,用EcoRⅠ和BglⅡ对基因组DNA进行全酶切,经0.4%琼脂糖凝胶电泳,特异回收9.5kb左右的DNA片段.将该片段与λEMBL3/BamHⅠ臂连接,构建成一个特殊的人基因组λ噬菌体文库(富集文库),效价为2×104.同时采用PCR方法及用引物Ⅰ:5′GGACAGGCTACATCAGGAAGAGG3′,引物Ⅱ:5′CTGCGTCTAATTGCAGTAGTTC3′,从人基因组DNA中扩增出一段含胰岛素基因的1.36kbDNA片段,做为放射性标记探针,对文库进行了噬菌斑原位杂交筛选,从1×104个噬菌斑中筛选到一个含人胰岛素基因组基因的阳性克隆,并进一步完成了亚克隆和该基因1732bpDNA序列的测定.结果该基因的1732bpDNA序列包括部分5′端和3′端与国外发表的人胰岛素α型等位基因的序列相同  相似文献

将抗癌胚抗原(CEA)单链抗体基因插入家蚕杆状病毒转移载体pBacPAKHis, 与修饰的家蚕核型多角体病毒BmBacPAKDNA共转染家蚕细胞, 经同源重组得到含有在多角体蛋白基因启动子控制下的抗CEAScFv 基因的重组病毒BmBacScFv。用重组病毒分别感染家蚕细胞和幼虫, 在两者中均得到了高效表达, 产物分子量为28kD, 前者占细胞总蛋白的6 % , 后者为0 .3 mg/ 蚕。目的基因在家蚕细胞和幼虫中表达产物经Ni2+IDASepharose6B亲和柱纯化, 前者纯度可达90% 以上, 后者纯度较低; 纯化后的融合蛋白具有CEA 结合活力, 其亲和常数分别为5 .4×108/mol·L- 1 和2.3 ×108/mol·L-1 , 略低于其亲本单抗E7B10 2.7 ×109/mol·L- 1 。  相似文献

将牛生长激素基因cDNA 与Patatin ClassI启动子及NOS3终止子连接,构建了表达载体pPBGT. 用直接法将表达载体转入农杆菌(Agrobacterium tum efaciens) LBA4404(pRAL4404)菌株, 用此菌株转化马铃薯(Solanum tuberosum )得到再生植株. 经NPTⅡ活性检测,总DNA PCR和Southern blot证明目的基因已整合到马铃薯基因组中.RNA 点杂交和Western blot表明牛生长激素基因已在转基因马铃薯块茎中转录和表达  相似文献

以玫瑰香葡萄(Vitisvinifera L.)为材料,克隆了含有叶绿体rbc L基因的3.1 kb Bam HⅠ片段,构建了该基因的限制性酶切图谱,测定了该基因的核苷酸序列。所测的核苷酸序列总长度为2004 bp,其中基因的编码区为1428 bp,编码一个含475 个氨基酸的蛋白质,其分子量约为53 kD;测定基因的5上游含启动子的部分共358 bp,包括- 10 区(TAAAAT)、- 35区(TTGCGC)和SD 序列(GGAGG);基因的3下游区共218 bp,含有3 个转录茎环终止结构。玫瑰香葡萄rbc L基因编码区的核苷酸序列与烟草、矮牵牛、菠菜、苜蓿、水稻和玉米之间的同源性分别为91.5% 、91.4% 、90.2% 、89.8% 、86.3% 和84.5% ;推导出的氨基酸序列的同源性分别为92.2% 、91.6% 、92.2% 、93.7% 、93.5% 和90.1% 。  相似文献

人GM—CSF cDNA的克隆和在大肠杆菌中的表达   总被引:3,自引:0,他引:3  
从诱导的人胚肺细胞HFL株中提取总RNA.经RT-PCR反应获取了人GM-CSFcDNA,DNA序列测定表明其顺序与文献报道完全一致。为了获得高效表达,应用PCR改造了人GM-CSF的cDNA5’端核苷酸序列,并将改造的人GM-CSF基因插入含T7启动子的质粒pET-11d构建成表达质粒pETC-5,将此质粒转化大肠杆菌株BL21(DE3)得到表达菌株BLEC4。表达菌株用0.5mol/LIPTG诱导2小时后,产生大量重组蛋白并形成包涵体。SDS—PAGE电泳图谱扫描结果表明,rhGM-CSF产量占菌体总蛋白量的16%。ELISA和TF-1细胞培养测定表明,初步纯化和复性的rhGM-CSF具有天然的hGM-CSF生物活性。  相似文献

The objective of this study was to examine whether the ICSI-mediated gene transfer method using in vitro matured oocytes and frozen sperm head could actually produce transgenic pigs. We also aimed at examining whether transgenic pigs can be cloned from somatic cells of a transgenic pig generated by the ICSI-mediated method. A bicistronic gene constituted of the human albumin (hALB) and enhanced green fluorescent protein (EGFP) genes was introduced into pig oocytes by the ICSI-mediated method. Transfer of 702 embryos produced by the ICSI-mediated method into five gilts resulted in 4 pregnancies. When three of the recipients, which had received total 312 of the embryos were autopsied, 32 including 1 transgenic fetuses were obtained. One of the recipients gave birth to three live piglets including one transgenic pig, showing a strong green fluorescence in the eyeballs, oral mucous membrane and subcutaneous tissues. Fluorescent microscopy revealed uniform GFP expression in all cell lines established from kidney, lung and muscle of the founder transgenic pig obtained. Nuclear transfer of these cells resulted in stable in vitro development of cloned embryos into the blastocyst stage, ranging from 12.9 to 19.8%. When 767 of the nuclear transfer embryos were transferred to 5 recipients, all became pregnant and gave birth to a total of six live transgenic-clones. The transgene copy number and integrity in the founder pig were maintained in the primary culture cells established from the founder as well as in the clones produced from these cells. Our study demonstrates that the ICSI-mediated gene transfer is an efficient and practical method to produce transgenic pigs, using frozen sperm heads and in vitro matured oocytes. It was also shown that combination of ICSI-mediated transgenesis and nuclear transfer is a feasible technology of great potential in transgenic pig production.  相似文献

We report here the production of transgenic quails using a replication-defective pantropic retroviral vector based on Moloney murine leukemia virus (MoMLV) pseudotyped with vesicular stomatitis virus G protein (VSV-G). The retroviral vector was injected into laid quail embryos at the blastodermal stage, and the embryos were incubated to hatch to produce G(0) transgenic quails. Among 134 embryos subjected to viral injection, 37 hatched. The viral vector sequence was detected in the tissues of all G(0) quails. The germ-line transmission efficiency of G(0) quails mated with nontransgenic quails was more than 80% on average. Southern blot analysis revealed that the G(1) transgenic progeny had one to three copies of the transgene. The expression of vector-encoded neomycin-resistance gene under the control of the Rous sarcoma virus (RSV) promoter was observed in several tissues including heart and muscle of both G(1) and G(2) transgenic offspring. Due to the high frequency of germ-line transmission, this method may markedly facilitate the production of transgenic avian.  相似文献

根据已克隆的鲢鱼(Hypophthalmichthysmolitrix)微囊藻毒素去毒酶cDNA全序列设计特异引物,利用PCR方法获得鲢鱼微囊藻毒素去毒酶基因编码区,将该编码区与绿色荧光蛋白连接,分别构建融合表达载体pEGFP-N1-sGST和双顺反子表达载体pIRES2-EGFP-sGST。利用脂质体转染法将融合表达载体pEGFP-N1-sGST转染Hela细胞,60h后检测到绿色荧光基因表达;通过显微注射,将双顺反子表达载体pIRES2-EGFP-sGST注入斑马鱼(Daniorerio)受精卵,获得了转鲢鱼微囊藻毒素去毒酶基因斑马鱼,从而构建了微囊藻毒素去毒酶转基因模型。上述2种转基因模型的成功构建为进一步研究鲢鱼、鳙鱼(Aristichthysnobilis)、罗非鱼(Oreochromisnilotica)等淡水鱼类微囊藻毒素去毒酶基因调控元件、去毒分子机理及研发转基因鲢鱼、鳙鱼、罗非鱼等微囊藻毒素高效生物去毒器奠定了基础。  相似文献

Human ALR gene sequence was amplified by PCR from human total DNA and inserted into pIRES(2)-EGFP vector. The bicistronic eukaryotic expression vector, pIRES-EGFP/ALR, expressing EGFP, Neo(r) and ALR genes was constructed. Sheep fetal fibroblast cells (sEFCs) were transfected with pIRES-EGFP/ALR by the induction of lipofectAMINE(TM). The positive cell clones were selected with medium containing G418 (800 μg/mL). The fluorescence of transgenic cells was examined with a confocal laser scanning microscope. The expression of ALR gene was tested by PCR, RT-PCR and immuno-histochemical staining. The transgenic cells were used as donors for nuclear transfer to enucleated ovine oocytes. Transgenic embryos were tested by confocal laser scanning microscope and immuno-histochemical staining. Results showed that the EGFP and ALR genes linked with IRES were coexpressed simultaneously in sFFCs; the blastocysts formed by nuclear transfer using tranfected donor cells are all transgenic blastocysts. EGFP, ALR and Neo(r) gene were all expressed in the transgenic embryos. In conclusion that a method to construct the positive embryos before pre-implantation which stably express ALR gene by the indication of EGFP expression has been successfully established. The application of this method can simplify the procedure of testing the targets and contribute to the efficiency increasing of transgenic domestic animal production.  相似文献

A novel technique of chimeric somatic cell cloning was applied to produce a transgenic rabbit (NT20). Karyoplasts of transgenic adult skin fibroblasts with Tg(Wap-GH1) gene construct as a marker were microsurgically transferred into one, previously enucleated, blastomere of 2-cell non-transgenic embryos, while the second one remained intact. The reconstructed embryos either were cultured in vitro up to the blastocyst stage (Experiment I) or were transferred into recipient-females immediately after the cloning procedure (Experiment II). In Experiment I, 25/102 (24.5%) embryos formed blastocysts from whole embryos and 46/102 (44.12%) embryos developed to the blastocyst stage from single non-operated blastomeres, while the reconstructed blastomeres were damaged and degenerated. Thirteen (12.7%) embryos did not exceed 3- to 4-cell stages and 18 (17.7%) embryos were inhibited at the initial 2-cell stage. Out of 14 blastocysts which were subjected to molecular analysis, the transgene was detected in the cells of 4 blastocysts. In Experiment II, 163/217 (75.0%) embryos were transferred into 9 pseudopregnant recipient-rabbits (an average of 18 embryos per recipient). Four recipient-females (44.4%) became pregnant and delivered a total of 24 (14.7%) pups. Molecular analysis confirmed that two pups (1.2%), one live and one stillborn, showed a positive transgene signal. Live transgenic rabbit NT20 appeared healthy and anatomically as well as physiologically normal. The results of our experiments showed that transgenic adult skin fibroblast cell nuclei, which have been introduced into the cytoplasmic microenvironment of single enucleated blastomeres from 2-cell stage rabbit embryos, are able to direct the development of chimeric embryos not only to the blastocyst stage but also up to term.  相似文献

The fat-1 gene was isolated from roundworm Caenorhabditis elegans, and built into pIRES2-EGFP expression vectors driven by cytomegalovirus (CMV) promoter or cytomegalovirus enhancer and chickenβ-actin (CAG) promoter. Both CMV- and CAG-driven expression vectors were transfected to sheep fetal fibroblast cells. Positive transfected cells were used as donors for somatic cell nuclear transfer (SCNT) and the cloned embryos were transferred into the oviducts of synchronized recipient sheep. Two lambs derived from CMV vector and three lambs derived from CAG vector developed to term. Although Southern analyses using tissues from the two lambs derived from CMV vectors indicated integration of fat-1 gene into the genome, fat-1 mRNAs were not detected by RT-PCR. However, there was fat-1 expression (detected by RT-PCR) in tissues from transgenic lambs driven by CAG vectors. To investigate potential mechanisms involved in the two transgene models, methylation state of the vector promoters were examined. In CMV-driven transgenics, CMV promoters had almost no methylation in transfected cells and the resultant cloned embryos, whereas high methylations were detected in tissues and organs in transgenic lambs. In the CAG-driven transgenics, there were almost no methylations in transgenic cells and transgenic cloned embryos, and cloned lambs expressed fat-1 mRNA (detected by RT-PCR). Moreover, although SV40 promoters which drove neo/kan marker gene in CMV vectors were highly methylated in tissues from transgenic lambs, they were without methylation in cells and embryos. Therefore, we concluded that highly methylated CMV promoters induced the silence of fat-1 transgene expression in sheep. Furthermore, CAG promoter, but not CMV promoter was suitable for generation of fat-1 transgenic sheep.  相似文献

根据已发表的小麦(T.aestivum)转录因子CBF1基因序列(GenBank Accession No.AF376136),设计引物从小麦品种‘京花1号’叶片中克隆出该基因,用拟南芥RD29B基因为启动子构建含CBF1基因的逆境诱导表达载体pBAC127F(6 967 bp),以‘99-92’、‘5-98’、‘104’和‘轮选987’等冬小麦品种(系)的幼穗和幼胚为材料,基因枪转化该表达载体。经筛选与植株再生,共获得14株转基因植株及其后代株系。这14个株系经PCR分析和点杂交检测,最终确认了5-98-40、5-98-41这2个株系为转基因株系,结果表明拟南芥RD29B启动子调控下的转录因子CBF1基因已稳定整合到转基因植株中。  相似文献

In an attempt to produce transgenic cloned cows secreting alpha 1-antitrypsin (alpha1-AT) protein into milk, bovine cumulus cells were transfected with a plasmid containing an alpha1-AT gene and green fluorescent protein (GFP) reporter gene using Fugene 6 as a lipid carrier. The GFP-expressing cells were selected and transferred into enucleated bovine oocytes. Couplets were fused, chemically activated and cultured. Developmental competence was monitored and the number of inner cell mass (ICM) and trophectoderm (TE) cells in blastocysts were counted after differential staining. The percentage of blastocysts was lower (P < 0.05) in transgenic cloned embryos compared to non-transgenic cloned embryos (23% versus 35%). No difference in the numbers of ICM and TE cells between the two groups of embryos was observed. One or two GFP-expressing blastocysts were transferred into the uterus of each recipient cow. Out of 49 recipient cows, three pregnancies were detected by non-return estrus and rectal palpation. However, the pregnancies failed to maintain to term; two fetuses were aborted at Day 60 and 150, respectively, and one fetus at Day 240. The genomic DNA from the aborted fetus was amplified by polymerase chain reaction (PCR) to investigate integration of the transgene in the fetus. The expected PCR product was sequenced and was identical to the sequence of alpha1-AT transgene. In conclusion, the present study demonstrated that developmental competence of cloned embryos derived from transgenic donor cells was lower than embryos derived from non-transfected donor cells. Although we failed to obtain a viable transgenic cloned calf, integration of alpha1-AT gene into the fetus presents the possibility of producing transgenic cloned cows by somatic cell nuclear transfer.  相似文献

Production of transgenic founder goats involves introducing and stably integrating an engineered piece of DNA into the genome of the animal. At LFB USA, the ultimate use of these transgenic goats is for the production of recombinant human protein therapeutics in the milk of these dairy animals. The transgene or construct typically links a milk protein specific promoter sequence, the coding sequence for the gene of interest, and the necessary downstream regulatory sequences thereby directing expression of the recombinant protein in the milk during the lactation period. Over the time period indicated (1995–2012), pronuclear microinjection was used in a number of programs to insert transgenes into 18,120, 1- or 2- cell stage fertilized embryos. These embryos were transferred into 4180 synchronized recipient females with 1934 (47%) recipients becoming pregnant, 2594 offspring generated, and a 109 (4.2%) of those offspring determined to be transgenic. Even with new and improving genome editing tools now available, pronuclear microinjection is still the predominant and proven technology used in this commercial setting supporting regulatory filings and market authorizations when producing founder transgenic animals with large transgenes (> 10 kb) such as those necessary for directing monoclonal antibody production in milk.  相似文献

Fetal-derived fibroblast cells were transduced with replication defective vectors containing the enhanced green fluorescent protein (EGFP). The transgenic cells were treated with colchicine, which theoretically would synchronize the cells into G2/M stage, and then used as donor nuclei for nuclear transfer. The donor cells were transferred into the perivitalline space of enucleated in vitro matured porcine oocytes, and fused and activated with electrical pulses. A total of 8.3% and 28.6% of reconstructed oocytes showed nuclear envelope breakdown and premature chromosome condensation 0.5 and 2 hr after activation, respectively. Percentage of pronuclear formation was 62.5, 12 hr after activation. Most (91.4%) of the 1-cell embryos with pronuclei did not extrude a polar body. Most (77.2%) embryos on day 5 were diploid. Within 2 hr after fusion, strong fluorescence was detectable in most reconstructed oocytes (92.3%). The fluorescence in all NT embryos became weak 15 hr after fusion and disappeared when culture to 48 hr. But from day 3, cleaved embryos at the 2- to 4-cell stage started to express EGFP again. On day 7, 85.8% of cleaved embryos expressed EGFP. A total of 9.4% of reconstructed embryos developed to blastocyst stage and 71.5% of the blastoctysts expressed EGFP. After 200 reconstructed 1-cell stage embryos were transferred into four surrogate gilts, three recipients were found to be pregnant. One of them maintained to term and delivered a healthy transgenic piglet expressing EGFP. Our data suggest that the combination of transduction of somatic cells by a replication defective vector with the nuclear transfer of colchicine-treated donors is an alternative to produce transgenic pigs. Furthermore, the tissues expressing EGFP from descendents of this pig may be very useful in future studies using pigs that require genetically marked cells.  相似文献
