The reactions of 1 nm particles of plutonium-238 dioxide and curium-244 dioxide with rat lung fluid have been studied both in vitro and in vivo. The plutonium-238 particles are positively charged and combine by electrostatic attraction with the negative pulmonary surfactant which mediates the transfer of radioactivity to the blood. In contrast the curium-244 particles are negative and are assumed to diffuse passively through the alveolar walls. The results emphasise that electrostatic charge is an important factor governing the reaction of 1 nm actinide oxide particles with macromolecules.  相似文献

A major factor influencing the movement of plutonium-238 from the lungs to blood after the intubation of oxide suspensions is the presense of 0.001 micrometer diameter particles. In a polydisperse suspension of particles this fraction increases with time, due it is thought, to fragmentation of larger particles induced by alpha decay. The rate of this process could account for the greater transportability in vivo of plutonium-238 relative to plutonium-239 when the oxides are inhaled. In blood, 0.001 micrometer diameter plutonium-238 oxide particles undergo a rapid reaction to form a low molecular weight species before plutonium is complexed with transferrin and citrate ions. The filtration of this species through the kidneys may explain the observed enhanced urinary excretion of plutonium relative to administered plutonium citrate. The mechanism of urinary excretion and relationship between cumulative urinary excretion and body content for plutonium-238 is similar to that previously observed for plutonium-239, even though different methods of preparation of the oxides were used.  相似文献

Bystander effects from ionizing radiation have been detailed for a number of cell systems and a number of end points. We wished to use a cell culture/ex vivo rat model of respiratory tissue to determine whether a bystander effect detected in culture could also be shown in a tissue. Examination by immunofluorescence techniques of tracheal cell cultures after exposure to very low doses of alpha particles revealed a large proportion of cells with proliferating cell nuclear antigen (PCNA) bound in their nuclei. PCNA was selected as an end point because it is involved in both DNA repair and the changes in cell cycle that are typical of many reported bystander effects. Maximum response can be detected in up to 28% of the cells in sub-confluent cultures with a dose of only 2 mGy. At this dose less than 2% of the cell nuclei have experienced a particle traversal and less than 6% of the cells have experienced an alpha-particle traversal through either their nucleus or some part of their cytoplasm. The hypothesis that this bystander response in nontargeted cells is mediated through secreted factor(s) is presented, and supporting evidence was found using partial irradiation and co-culture experiments. Examination of the effect with excised pieces of trachea demonstrated a response similar to that seen in culture.  相似文献

Studies with pure LICAM(C) and with its methyl esters showed that all the substances caused increased renal retention of plutonium-238 in the kidneys of mice and rats but were equally effective in reducing plutonium retention in bone and liver.  相似文献

Plutonium-214 citrate solution at pH 6-5 was injected intravenously or intra-peritoneally into hamsters and rats at a dose of 50 MBq kg-1 (1-35 mCi kg-1). The animals were killed 1 day or 1 week later, and tissues were removed for autoradiography and radiochemical analysis. Plutonium-241 was distributed in rats in the same way as plutonium-239, and is a suitable isotope for high-resolution tissue-section autoradiography. Plutonium deposits in cells consisted of a nuclear and a cytoplasmic component. In the hamster kidney cells, the amount associated with the nucleus was about 55 per cent of the total cellular plutonium at 24 hours after injection. Six days later, it was only about 30 per cent. Plutonium deposits were also characterized in hepatocytes, in the interstitial cells of the testes, in the cells of ovarian follicles, in chondrocytes and in bone cells, including osteoblasts and osteocytes. In bone there appeared to be both an extracellular and intracellular deposit. No evidence was found of substantial incorporation of plutonium into the mineral phase of bone.  相似文献

A particulate fraction from rat brain catalyzes the incorporation of [(14)C]choline, [(14)C]ethanolamine, and l-[(14)C]serine into phosphatidylcholine, phosphatidylethanolamine, and phosphatidylserine, respectively. The reaction appears to be energy-independent since Mg(2+), CTP, ATP, and NaF have no stimulatory action. The incorporation is inhibited by EDTA and activated by Ca(2+). The pH optimum for the incorporation of choline is 9.5, for ethanolamine it is 9.0, and for l-serine it is 8.5. Tris, bicine, and imidazole buffers are inhibitory. The incorporations are inhibited by a variety of structurally related alcohols and are stimulated by isoserine (alpha-hydroxy,beta-aminopropionic acid).  相似文献

A simple one-step procedure is described for the isolation of lamellar bodies and their membranous content in high yield from rat lung. A linear sucrose gradient, ranging from 0.9M – 0.2M sucrose, is poured over a sample of homogenate in IM sucrose and the lamellar body fraction if floated to isopycnic equilibrium by high speed centrifugation. When the same procedure is applied to fluid drained from the lungs of newborn lambs or to human amniotic fluid collected late in pregnancy, a fraction is obtained which appears to consist of the membranous content of lamellar bodies in the process of unravelling.  相似文献

Male rats were exposed to single doses (0-30 Gy) of 60Co gamma rays to the right hemithorax. Half of each dose group consumed only control powdered chow after irradiation, and half consumed feed containing 0.10% (w/w) pentoxifylline (50 mg/kg/day). The severity of epilation and desquamation in the field of the radiation port was scored weekly. Two months after irradiation the animals were killed, and pulmonary endothelial function was monitored by the activity of lung angiotensin converting enzyme (ACE) and plasminogen activator (PLA), and by production of prostacyclin (PGI2) and thromboxane (TXA2). The amount of hydroxyproline (HP) in the lung served as an index of pulmonary fibrosis. Radiation produced a dose-dependent decrease in ACE and PLA activity in the right lung and an increase in the production of PGI2 and TXA2. This endothelial dysfunction was accompanied by an increase in wet weight and in protein and HP content in the irradiated lung. Pentoxifylline spared only the increase in lung wet weight and protein content, and actually elevated the radiation-induced hyperproduction of PGI2 and TXA2. The severity of the epilation and desquamation reactions increased with increasing radiation dose and time but was independent of diet. These data indicate that pentoxifylline, despite some promising pharmacological actions, has no beneficial effect on acute radiation reactions in rat lung and skin.  相似文献

The behavior of neutron-irradiated, simulated Chernobyl UO2 particles containing 141Ce, 144Ce, 95Zr, 95Nb, and 103Ru in the gastrointestinal tract was investigated to obtain basic information for dosimetric and risk analyses of nuclear accidents. After the UO2 particles were administered to rats intragastrically, the distribution and retention of specific radionuclides were studied by using whole-body autoradiography and gamma-spectrometric analysis of tissues. None of the radionuclides were detected in liver, kidney, muscle, bone, brain, blood, and urine. Approximately 98% of the total administered radioactivity was excreted in feces within 3 days. A two times greater intestinal retention (about 6%) of 95Nb than for the other radionuclides was observed 1 day after administration. The results indicate that this kind of relatively insoluble particulate material is not absorbed or retained significantly in the epithelial cells of the intestinal wall. Fallout particles containing high-energy beta sources, 106Ru and 144Ce, result in a very high radiation dose (up to several Gy/day) in the vicinity of a hot particle. Niobium-95 with low average beta energy (0.043 MeV (100%)) does not increase the total dose to the GI tract significantly despite its longer retention in the intestine. Evaluation of the biological effects of these particles in the GI tract by using a dosimetric model based on uniform distribution of activity may be misleading.  相似文献

A filtration-method on Millipore-membranes for the assay of the base-exchange reaction was described. Its advantage over the usual procedure based upon the extraction and the washing of lipids was discussed with the viewpoint of processing many samples, which would be indispensable for purifying the enzyme.The reaction showed an absolute dependency for calcium ion with different optimal concentrations for each of the three bases, a sensitivity to inhibition by high ionic strength, and a pH optimum around 9.0. Exogenously added phospholipid, asolectin, gave a slight stimulation for ethanolamine and l-serine incorporation at a low concentration while choline incorporation was essentially inhibited at all concentrations examined. In heat-denaturation experiments with the particulate and soluble the incorporation of choline into lipid was more sensitive than that of ethanolamine and l-serine. A developmental study showed that brain particles sedimenting between 10,000 and 35,000g prepared from rats aged 22–27 days readily incorporated ethanolamine, l-serine, and choline into their corresponding phosphatidyl compound.Several procedures for solubilization of the “base-exchange” enzyme were examined. The most effectively solubilized preparation was obtained by the use of an ionically balanced detergent, Miranol H2M. This preparation showed a marked dependency on exogenously added phospholipids for its maximal enzymic activity, had a pH optimum at around 7.2, and had an absolute requirement for Ca2+. This particular detergent at a concentration of 1% (wv) solubilized approximately 50% of the protein, and about 30% of the phospholipids, 40% of the cerebrosides, and only 11% of the cholesterol originally present in the particles. The relative proportions of different phospholipids solubilized by the detergent were, however, similar to those present in the original particles.The base-exchange reaction catalyzed by the solubilized enzyme was found to be highly sensitive to ionic strength, and the inhibitory effect of a specific monovalent cation paralleled its ionic size. Substantial differences in the Km value for each of the substrates with only slight differences in V were observed.The choice of solubilizing agents in relation to these properties and to the maintenance of the activity of the base-exchange reaction was discussed.  相似文献
