Summary The columnar epithelial cells of the gas gland in the swimbladder of the toadfish, Opsanus tau L., contain lamellar bodies that resemble the lamellar bodies found in epithelial cells of vertebrate lungs. Cytochemical assays indicate that swimbladder lamellar bodies are soluble in chloroform-methanol solution, react with tricomplex flocculation solution (indicating a phospholipid component), exhibit a positive reaction for cholesterol when exposed to digitonin, and contain acid phosphatase.The anterior chamber of the toadfish swimbladder is lined by an extracellular layer. Digitonin-cholesterol crystals are found in this layer when the swimbladder is treated with digitonin. A ruthenium red positive layer is also present in the anterior chamber of the toadfish swimbladder.The structure and cytochemistry of swimbladder lamellar bodies are compared with those of vertebrate lung lamellar bodies. Summary The swim bladder of the cyprinid Carassius auratus (goldfish) is a two-chambered organ connected to the esophagus by a pneumatic duct. The anterior chamber is lined by a single type of squamous epithelial cell. Two types of epithelial cells are present in the posterior chamber. Flattened cells with differences in the electron density of the cytoplasm line most of the chamber. Darker cells generally contain large amounts of glycogen. Cuboidal epithelial cells also occur in the posterior chamber. A glandular layer external to the muscularis in the posterior chamber is composed of large cells containing little glycogen, an extensive Golgi apparatus, and numerous mitochondria with single large granules. Capillaries and nerves are present in large numbers in this layer. Blood vessels form micro-retia mirabilia in the submuscular layer external to the glandular layer. Vessels are of two distinct types with wide lumina and flattened endothelium characterizing the venous vessels. Arterial vessels have smaller lumina, thick endothelial cells with prominent pinocytotic vesicles, and surrounding pericytes. Collagen is present in three forms in this swim bladder — large tactoids in the tunica externa of the anterior chamber, smaller tactoids in the lamina propria of the posterior chamber, and small fibrils in all other areas.Supported by a Young Investigator Pulmonary Research Grant # 1 R23 HL 19593-01 and by HL 23338-01 from the National Institutes of Health  相似文献

The wall of the stomach of the tigerfish is described and compared with that of other vertebrates. Light microscopic and ultrastructural characteristics of the stomach wall correspond to a large extent to those of other vertebrates, although some differences are found. The mucosa contains (1) surface epithelium characterized by narrow columnar cells with abundant mucous granules; (2) gastric glands consisting of pepsinogenic cells of variable height, containing tubulovesicles and bearing microvilli; (3) five granulated cell types located basally in the epithelium (types 1–5); and (4) lamina propria and muscularis mucosae. Connective tissue separating smooth muscle fibers of the muscularis mucosae constitutes a stratum compactum. The submucosa contains a loose connective tissue, a tunica muscularis of inner circular and outer longitudinal layers, and a serosa of mesothelium and subjacent connective tissue. Immunocytochemical tests with antisera to five polypeptides show gastrin/cholecystokinin (CCK), vasoactive intestinal polypeptide (VIP) immunoreactivities in some cells of the gastric glands, and somatostatin in cells lying among epithelial cells lining the gastric luminal surface or gastric pits.  相似文献

The swimbladder of the adult eel, Anguilla anguilla, with its bipolar countercurrent system, the rete mirabile, is a widely used model for swimbladder function, but very little is known about the development of this swimbladder. Our histological studies on the developing swimbladder revealed that during metamorphosis the swimbladder becomes present as a dorsal outgrowth of the esophagus. It is filled with surfactant, and gas was not detected in the swimbladder. In the young glass-eel, the epithelial (gas gland) cells of the swimbladder are columnar, but do not yet have the typical basolateral labyrinth established in adult animals. Few blood vessels are found in the swimbladder tissue, and the submucosa is present as a thick layer of connective tissue, giving a large diffusion distance between blood vessel and swimbladder lumen. Within the next 2 or 3 months of development, gas gland cells develop their typical basolateral labyrinth, and the thickness of the submucosa is significantly reduced, resulting in a short diffusion distance between blood vessels and the swimbladder lumen. The first filling of the swimbladder with gas is observed while the gas gland cells are still in a poorly differentiated status and it appears unlikely that these cells can accomplish their typical role in gas deposition. The presence of small gas bubbles in the swimbladder as well as in the ductus pneumaticus at the time of initial swimbladder inflation suggests that the swimbladder is filled by air gulping or possibly by taking up gas bubbles from the water.  相似文献

Development of the gastric connective tissue of Xenopus laevis during metamorphosis was investigated by electron microscopy. Throughout the larval period to stage 60, the layer of connective tissue underlying the gastric epithelium consists of immature fibroblasts surrounded by a sparse extracellular matrix. At the beginning of the transition from the larval to the adult epithelial form, at about stage 60, extensive changes occur in the connective tissue. The number of cells suddenly increses and different cell types appear. Numerous contacts between epithelial and connective tissue cells are established through random gaps in the thickened basal lamina. During stages 62–63, just after the beginning of the morphogenesis of adult-type glands, the basal lamina lining the glandular epithelium becomes thinner, and the number of contacts decreases rapidly except near the tips of the glands. After the glandular cells begin to produce zymogen granules at stage 64, contacts become rare. From stage 63, when the muscularis mucosae develops, until the completion of metamorphosis, the connective tissue consists mainly of typical fibroblasts. Outside the muscularis mucosae, the fibroblasts of the lamina propria are aligned in parallel with the curvature of the glands. These observations indicate that developmental changes in the connective tissue are closely related spatiotemporally to those of the epithelial transition from larval to adult form during metamorphic climax. Although some changes are similar to those in the intestine (Ishizuya-Oka and Shimozawa, '87b), others are specific to the gastric region, which suggests that connective tissue may have a role in organ-specific differentiation of the gastric epithelium.  相似文献

 This study presents histological and scanning electron microscopical findings on the structural differentiation, and the nervous and vascular supply of the digestive tracts of Nautilus pompilius and N. macromphalus, including the foregut, stomach, vestibulum, caecum, midgut and rectum. The stereoscopic reconstruction of the vestibulocaecal complex gives an idea how the digestive cycle between the stomach, vestibulum, caecum and proximal midgut could possibly proceed. All parts of the digestive tract are covered luminally by a columnar epithelium which contains numerous goblet cells. The epithelium is ciliated in the vestibulum, caecum, proximal midgut and the longitudinal groove of the rectum. On this lamina epithelialis mucosae borders the lamina propria mucosae, which consists of connective tissue and some muscle cells. In the stomach it is differentiated, forming a special bolster-like layer. The lamina propria mucosae is followed by the tunica muscularis, which consists of a stratum circulare and a stratum longitudinale in the foregut, vestibulum, caecum, midgut and rectum. In the stomach, midgut and rectum, the tunica adventitia, which consists of a thin layer of connective tissue, is located between the tunica muscularis and the cuboidal tunica serosa. Scanning electron microscopy of whole-mount preparations of the tela submucosa in the porcine small intestine, examined after trypsin digestion, fixation and HCl hydrolysis, visualized a clear differentiation of the submucosal plexuses, i.e., the plexus submucosus internus (Meissner) and the plexus submucosus externus (Schabadasch). The distinctive features refer to the topography, number, size and shape of the ganglia and the number and diameter of the nerve strands. The plexus of Meissner is closely apposed to the external surface of the lamina muscularis mucosae by the enveloping connective tissue and by connecting strands penetrating the lamina muscularis mucosae. Three distinctive subdivisions of connecting strands can be identified. Since the glial cells covering the ganglia and connecting strands have been preserved, neither individual neuronal cells nor axons can be observed.  相似文献

The digestive tract of Hoplosternum thoracatum consists of an esophagus, gastric area, anterior digestive intestine with elaborate folds, digestive intestine with decreasing folds and thin, smooth-surfaced respiratory intestine. The upper tract has a mucoid columnar lining which is gently folded, whereas the gastric area has numerous pits opening into the tubular secretory glands. Striated muscle comprises the anterior muscularis but is replaced by inner circular and outer longitudinal smooth muscle layers in the gastric region. The digestive intestinal mucosa is elaborately folded, consisting of columnar cells with prominent brush borders. Mucosa, submucosa, circular and longitudinal muscularis and serosa layers are present throughout the tract. Goblet cells occur in both the digestive and respiratory intestine. Major changes that appear in the respiratory intestine are a drastic reduction in mucosa epithelial thickness and the penetration of an elaborate capillary bed into the epithelium. The other basic layers are not significantly reduced in thickness. The air-blood barrier consists of the thin epithelium, basement lamina and very thin capillary endothelium. Regional cellular composition and ultrastructural features are correlated with respective digestive and respiratory functions.  相似文献

The structure and seasonal changes of the oviductal-cloacal junction remain poorly understood in most squamates. This study was undertaken to describe the histology of the oviductal-cloaca junction of a female viviparous snake Toluca lineata, during gestation, previtellogenesis, and vitellogenesis. The oviductal-cloacal junction exhibits a wider lumen and thicker layers of connective tissue, smooth muscle layers, and total wall width compared to the posterior vagina. The lining is characterized by thick, short longitudinal mucosal folds. The luminal epithelia differ morphologically from anterior to posterior portions of the oviductal-cloacal junction. The anterior portion is lined with a simple columnar epithelium composed of nonciliated cells. The middle portion is lined with stratified epithelium that contains an apical columnar cell layer that undergoes morphological changes coincident with the reproductive cycle. The posterior portion is lined with a stratified squamous epithelium. The connective tissue underlying the epithelium contains numerous ovoid cells having abundant acidophilic cytoplasmic granules—eosinophils. Copulation occurs during the previtellogenic stage, as evidenced by the presence of abundant spermatozoa in the lumen of the anterior portion and of a copulatory plug in the middle and posterior portion of the oviductal-cloacal junction. Development of the respiratory swimbladder of Pangasius sutchi   总被引:1,自引:0,他引:1  
The swimbladder of Pangusius sutchi first appears on the dorsal surface of the oesophagus at about 5 days after hatching. The swimbladder has double chambers when it is separated by a medial septum at 8–10 days. Alveoli start to develop and function in air-breathing at 12–14 days. Their number is increased by subdivision, and the respiratory portion grows towards the centre. Morphometric analysis shows that the swimbladder increases in respiratory surface, volume and surface area: volume ratio during development. On a histological basis, the development of the swimbladder is divided into three distinct periods: a blind tube, a double chamber and an alveolus period. It is characteristic that the flat epithelial cell arises from a primordial cuboidal cell and that a double capillary system is arranged in the interalveolar septa. Multilamellar bodies appear and a blood-air barrier is established when the swimbladder becomes functional.  相似文献

Prostaglandins and inhibitors of prostaglandin synthesis have striking regulatory effects on intestinal muscularis externa. We suggested earlier that a population of macrophage-like cells, located between the external muscle layers might release prostaglandins with a local effect on enveloping interstitial cells of Cajal, postulated pacemaker cells of the gut. To determine cellular production site(s) of prostaglandin we applied monoclonal antibodies against prostaglandin H synthase combined with the PAP technique to sections of rabbit and guinea-pig small intestine and kidney. In rabbit small intestine muscle cells in the circular muscle layer and in the muscularis mucosae were positive, longitudinal muscle negative. Vascular endothelial cells and serosal mesothelial cells were stained. In guinea-pig all muscle layers were unstained but endothelial and mesothelial cells were stained together with unidentified cells in the outermost submucosa. In rabbit kidney, positive staining of collecting ducts, interstitial cells, the parietal layer of Bowman's capsule and arterial endothelial cells was present. Furthermore, we found prostaglandin synthase antigenicity in the epithelial cells lining the loop of Henle, not described before. In guinea-pig medullary collecting ducts were stained and the papilla was lined by stained epithelial cells. The results show a species variation in the distribution of recognizable levels of prostaglandin H synthase. The impressive reaction in the mesothelium must be considered, when enzyme distribution is examined biochemically with fractionated tissue. Our findings do not support our hypothesis that macrophage-like cells are more potent sources of prostaglandins than smooth muscle cells.  相似文献

Amazona aestiva Linnaeus, 1758 is known as the blue-fronted parrot. These animals are considered seed dispersion, contributing to ecological balance. Despite its ecological importance, A. aestiva is listed as an imminent danger of becoming extinct, as it is one of the most illegally traded native species in Brazil, besides losing its habitat due to deforestation and forest fires. The cranial oesophagus mucosa is lined with a non-keratinized stratified squamous epithelium, and the basal layer presents melanocyte accumulation. No mucous glands were observed in the lamina propria/submucosa. The ingluvium two different muscular layer patterns were observed, one internal and circular and one very thick, external and longitudinal. The proventriculum submucosa consists of polygonal proventricular glands and contents into a broad central duct lined with simple columnar epithelium Periodic acid-reactive Schiff positive (PAS+) and Alcian blue positive (AB+). The 5-HT cells are dispersed in the proventricular glands located at base and apex. The ventricle mucous layer contains numerous branched folds lined by a simple columnar epithelium. The epithelium was negative reaction for PAS and positive for AB only on the fold surfaces. The 5-HT cells were scarce and observed only at the base of the ventricular gland, as well as in the adjacent connective tissue.  相似文献

Colonic organogenesis in rats was studied using light microscopic techniques for the demonstration of mucosubstances, glycogen, and connective tissue fibers. Crypts began as intraepithelial spaces which were in continuity with the colonic lumen. The cells forming the floors of these spaces invaded the nonsulfated acid glycosaminoglycan-rich mesenchyme as the basement membrane became discontinuous. As the diameter of the colon increased, the crypts lengthened and the lamina propria thickened until a layer of collagen and sulfated acid glycosaminoglycans formed at the bases of the crypts and the basement membrane was reestablished. The circular layer of the muscularis externa developed first, then the longitudinal layer, and finally the muscularis mucosae. Three types of mucous cells arose in these newly formed crypts. The initial epithelial cell type contained glycogen and gave rise to cells with apical coats of nonsulfated acid glycoproteins. This cell type was followed by the appearance of cells at the bases of the crypts containing nonsulfated acid glycoproteins. As the crypts lengthened, the goblet cells near the base contained nonsulfated and/or sulfated acid glycoproteins. Closer to and on the surface, the cells contained sulfated acid glycoproteins, a mixture of sulfated acid and neutral glycoproteins, or just neutral glycoproteins. Striated-border cells appeared intermingled with the mucous cells close to the bases of the crypts and continued onto the surface. A comparison was made between regeneration following placement of a surgical lesion in adult rats and events in organogenesis of the colon.  相似文献

Summary The fine structure of the wall of the rudimentary rear chamber in the swimbladder of the pond-loach was studied. The morphological features of a majority of the cells, as well as the presence of micro-rete mirabile, suggest that epithelial components in the wall of this organ represent a structural and functional equivalent of the gas gland.  相似文献

Whole-mount preparations of the porcine small intestine, consisting of the tela submucosa and the adjacent lamina muscularis mucosae, were used for scanning electron-microscopic investigation of the plexus submucosus externus (Schabadasch) after enzymatic digestion, fixation and HCI hydrolysis. The present results confirm previous light-microscopic data and provide irrefutable proof that within the submucosal plexus, considered by most authors as one ganglionated nerve plexus situated in the entirety of the tela submucosa, two distinct nerve meshworks can be distinguished, one lying close to the lamina muscularis mucosae, i.e., the plexus submucosus internus (Meissner), and the other, i.e., the plexus submucosus externus (Schabadasch), situated in the outer region of the tela submucosa against the circular smooth muscle layer. In addition to the distinct location of both plexuses, they are quite different with regard to the pattern and diameter of their nerve strands and the number and appearance of their ganglia.  相似文献

Among hatchery bred Sparus aurata , 5.4% of the offspring of a single parent pair showed severe skeletal deformations coupled with marked growth retardation and pathological changes in the swimbladder. This resulted from extreme proliferation, then hypertrophy, of the cuboidal epithelial cells of the gas gland and proliferation of the rete mirabile. This caused almost complete congestion of the swimbladder air space. The nature of the pathological changes in the swimbladder and the possibility of hereditary origin for both the skeletal and swimbladder deformations are discussed.  相似文献

The fine structure of the epithelial cells of the anterior lobe, or coagulating gland, of the mouse prostate has been investigated by electron microscopy. This organ is composed of small tubules, lined by tall, simple cuboidal epithelium surrounded by connective tissue and smooth muscle. The epithelial cells are limited by a distinct plasma membrane, which covers minute projections of the cytoplasm into the lumen. The cell membranes of adjacent cells are separated by a narrow layer of structureless material of low density. The cavities of the endoplasmic reticulum are greatly dilated, and the cytoplasmic matrix is reduced to narrow strands, in which the various organelles are visible. The content of the cavities of the endoplasmic reticulum appears as structureless material of lesser density than the cytoplasmic matrix. Material which may be interpreted as secretion products can be seen in the lumina of the tubules. The possible nature of the material inside the cisternal spaces and the secretory mechanisms in these cells is discussed.  相似文献

The filter chamber is a complex junction of anterior and posterior extremities of the midgut and Malpighian tubules. The sac-like anterior extremity, or filter chamber proper, comprises two cell types. These are large cuboidal cells which secrete a mucoprotein, and extremely thin cells which have regular tubular invaginations of the basal plasma membrane. The posterior extremity of the midgut and the internal Malpighian tubules coil round the filter chamber proper. They consist of thin epithelial cells identical in ultrastructure. The basal plasma membrane in these cells is formed into leaflets. A thin cellular sheath and thick muscle layers surround the filter chamber. The filter chamber proper is lined by the mucoprotein secretion of the cuboidal cells. This secretion appears to bind potassium ions. ATPase and alkaline phosphatase cannot be detected in the filter chamber epithelia. The structure and cytochemistry of the filter chamber suggests that water flows from filter chamber proper to midgut and Malpighian tubules by passive osmosis. This may be facilitated by ion binding in the filter chamber proper and by hydrostatic pressure engendered by contraction of the muscular coat. The Malpighian tubules appear to be structurally and chemically adapted for ion secretion by active transport and possibly for reabsorption in the Malpighian duct segment.  相似文献
