Three new aromatic compounds, identified as 1-(3′,4′-methylenedioxy-phenyl)-10-(3″-hydroxyphenyl)-decane (1), 1-(3′,4′-methylenedioxy-phenyl)-12-(3″-hydroxyphenyl)-dodecane (2), and 1-(3′,4′-methylenedioxy-phenyl)-12-(3″-hydroxyphenyl)-6Z-dodecylene (3), along with six known compounds (4–9) were isolated from the 95% EtOH extract of Homalomena occulta. Their structures were elucidated by chemical and spectral methods Compounds 4–9 were isolated for the first time from this plant. Compounds 1–3 exhibited inhibitory activity against BACE1, with IC50 values of 0.82–1.09 μmol/L.  相似文献

Synthetic biology and metabolic engineering rely on computational search tools for predictions of novel biosynthetic pathways to industrially important compounds, many of which are derived from aromatic amino acids. Pathway search tools vary in their scope of covered reactions and compounds, as well as in metrics for ranking and evaluation. In this work, we present a new computational resource called ARBRE: Aromatic compounds RetroBiosynthesis Repository and Explorer. It consists of a comprehensive biochemical reaction network centered around aromatic amino acid biosynthesis and a computational toolbox for navigating this network. ARBRE encompasses over 33′000 known and 390′000 novel reactions predicted with generalized enzymatic reactions rules and over 74′000 compounds, of which 19′000 are known to biochemical databases and 55′000 only to PubChem. Over 1′000 molecules that were solely part of the PubChem database before and were previously impossible to integrate into a biochemical network are included into the ARBRE reaction network by assigning enzymatic reactions. ARBRE can be applied for pathway search, enzyme annotation, pathway ranking, visualization, and network expansion around known biochemical pathways and products of lignin degradation to predict valuable compound derivations. In line with the standards of open science, we have made the toolbox freely available to the scientific community on git (https://github.com/EPFL-LCSB/ARBRE) and we provide the web-version at http://lcsb-databases.epfl.ch/arbre/. We envision that ARBRE will provide the community with a new computational resource and comprehensive search tool to predict and rank pathways towards industrially important aromatic compounds.  相似文献

Abstract: An enzyme immunoassay using a double-antibody solid-phase technique for myelin basic protein (MBP) has been developed. Antisera were prepared by immunizing rabbits with the purified MBP from chick brain. The conjugation of MBP with horseradish peroxidase was performed by the periodate oxidation method in triethanolamine-acetate buffer (pH 8.5). The sample, antiserum, and conjugate were incubated at 4°C for 16 h, after which the insoluble second antibody was added and the reaction mixture was incubated at 4°C for 3 h. The peroxidase activity of the insoluble conjugate was assayed fluorometrically with hydrogen peroxide and 3-( p -hydroxyphenyl)propionic acid as substrates. The method had an analytical range from 50 pg to 1 ng (from 2.3 × 10−15 to 4.5 × 10−14 mol). The within-assay coefficient of variation (CV) was between 4 and 11% and the between-assay CV for 200 and 400 pg of MBP was 5.5 and 7.1%, respectively. A weak cross-reactivity was observed between chick MBP and bovine MBP, while no reactivity was shown with calf thymus histone. The MBP content of the brain during development increased markedly from the 3rd embryonic week to the 3rd post-hatch week (from 0.01 to 2.4 mg/g of fresh tissue), and the adult level was 3.2 mg/g of fresh tissue.  相似文献

Peroxynitrite (ONOO(-)), a reactive nitrogen species, is capable of nitrating tyrosine residue of proteins. Here we show in vitro evidence that plant phenolic compounds can also be nitrated by an ONOO(-)-independent mechanism. In the presence of NaNO(2), H(2)O(2), and horseradish peroxidase (HRP), monophenolic p-coumaric acid (p-CA, 4-hydroxycinnamic acid) was nitrated to form 4-hydroxy-3-nitrocinnamic acid. The reaction was completely inhibited by KCN, an inhibitor for HRP. The antioxidant ascorbate suppressed p-CA nitration and its suppression time depended strongly on ascorbate concentration. We conclude that nitrogen dioxide radical (NO(2)(radical)), but not ONOO(-), produced by a guaiacol peroxidase is the intermediate for phytophenolic nitration.  相似文献

Effects of immobilization pH and pore characters of mesoporous silicas (MPSs), MCM-41, SBA-15, and MCF, were simultaneously investigated for the immobilization of horseradish peroxidase (HRP; EC 1.11.1.7). MCM-41 and SBA-15 were rod-like with respective average pore diameters of 32, and 54 Å, while that of MCF with spherical cell and frame structure was 148 Å. Moreover, the MPSs synthesized were of identical surface functional groups and similar contents of free silanol groups. At immobilization pH 6 and 8 almost 100% HRP loadings were obtained and insignificant leaching were observed for all types of supports at pH 6. However, MCF was found to give both the highest enzyme loading and leaching at pH 10. Maximum and minimum HRP activities were obtained at respective immobilization pH 8, and 6. Activities of immobilized HRP increased with support pore diameters in the order: MCM-41 < SBA-15 < MCF. HRP immobilized at pH 8 gave the highest storage stability (both at 4 °C and room temperature), and in opposition to pH 6. In addition, HRP immobilized in MCF was found to be the most stable under storage. The finding should be useful for the creation of biocatalysts and biosensors.  相似文献

Monitoring of microwave emission from aqueous solution of horseradish peroxidase (HRP) in the process of the enzyme functioning was carried out. For the monitoring, a system containing HRP, luminol and Н2О2 was employed. Microwave emission measurements were carried out in the 3.4-4.2 GHz frequency range using the active and passive modes (active-mode and passive-mode measurements). In the active mode, excitation of the solution in the pulsed electromagnetic field was accomplished. In the passive mode, no excitation was induced. It appears that the passive-mode measurements taken in the course of the peroxidase reaction in the enzyme system have shown a 0.5 °С increase of the microwave signal. Upon the active-mode measurements, taken in the same reaction conditions, the forced excitation of the solution has also led to the increase (by 2 °С) of the level of the microwave signal – i.e. to its 4-fold enhancement compared to the signal obtained in passive-mode measurements.  相似文献

Reaction of silver nitrite with Ru(salen)(PPh3)(Cl) (salen2− = N,N′-ethylenebis(salicylideneiminato dianion) using [NEt4]OH as a phase transfer agent led to the formation of a stable, N-coordinated nitrite ion complex (RuIII-NO2) that was characterized by single crystal X-ray diffraction. This reaction also resulted in nitration of the two phenolic rings of the salen ligand, which was unexpected given the basic conditions. With sodium nitrite, the Ru(III)-nitro complex was formed, but no ring nitration was observed. Thus, in a non-acidic medium, the combination of nitrite with redox active metal centers may provide a route to the nitration of phenolic derivatives. Possible mechanisms of this unusual reaction having potential relevance to aromatic nitrations in biological systems are discussed. This is also the first report of the crystal structure of a Ru(III)-nitro complex, a species that has been suggested to be unstable and prone to rapid disproportionation.  相似文献

Microperoxidase-11 (MP-11) which has been immobilised in a matrix of chitosan-embedded gold nanoparticles on the surface of a glassy carbon electrode catalyzes the conversion of aromatic substances. This peroxide-dependent catalysis of microperoxidase has been applied in an enzyme electrode for the first time to indicate aromatic compounds such as aniline, 4-fluoroaniline, catechol and p-aminophenol. The electrode signal is generated by the cathodic reduction of the quinone or quinoneimine which is formed in the presence of both MP-11 and peroxide from the substrate. The same sensor principle will be extended to aromatic drugs.  相似文献

 The well-known and easily available horseradish peroxidase (HRP) catalyzes the H2O2-dependent oxidative 4-dechlorination of the pollutant 2,4,6-trichlorophenol, which is recalcitrant to many organisms except those producing ligninases. UV-visible spectroscopy and gas chromatography-mass spectrometry identified the oxidized reaction product as 2,6-dichloro-1,4-benzoquinone. NMR and IR spectroscopic data further supported the above characterization. Experimental evidence for the elimination of HCl from the substrate was acquired by detecting the decrease in pH of the reaction mixture, and by observing the presence of the β-chlorocyclopentadienone cation fragment in the mass spectrum of 2,6-dichloro-1,4-benzoquinone. Consequently, nucleophilic attack by water on the 2,4,6-trichlorocyclohexadienone cation was proposed to give the final product. Our results indicate an oxidative dechlorination pathway catalyzed by HRP for 2,4,6-trichlorophenol, similar to that by extracellular lignin peroxidases. The relative catalytic efficiency of HRP seems higher than that of lignin peroxidases. The HRP-H2O2 catalytic system could be utilized in the degradation of polychlorinated phenols for industrial and biotechnological purposes. Summary Rumen contents from three fistulated Japanese native goats fed Lucerne hay cubes (Medicago sativa) and concentrate mixture were collected to prepare the suspensions of mixed rumen bacteria (B), mixed protozoa (P) and a combination of the two (BP). Microbial suspensions were anaerobically incubated at 39°C for 12h with or without 1 MM ofl-phenylalanine (Phe). Phe, tyrosine (Tyr) and other related compounds in both supernatant and microbial hydrolysates of the incubations were analyzed by HPLC. Tyr can be produced from Phe not only by rumen bacteria but also by rumen protozoa. The production of Tyr during 12h incubation in B (183.6 mol/g MN) was 4.3 times higher than that in P. One of the intermediate products between Phe and Tyr seems to bep-hydroxyphenylacetic acid. The rate of the net degradation of Phe incubation in B (76.O mol/g MN/h) was 2.4 times higher than in P. In the case of all rumen microorganisms, degraded Phe was mainly (>53%) converted into phenylacetic acid. The production of benzoic acid was higher in P than in B suspensions. Small amount of phenylpyruvic acid was produced from Phe by both rumen bacteria and protozoa, but phenylpropionic acid and phenyllactic acid were produced only by rumen bacteria.  相似文献

Fee (1990) Control ofEscherichia coli superoxide dismutases (sodA andsodB) genes by the ferric uptake regulation (fur) locus,J. Bacteriol. 172, 1930–1938]. Previous work from other laboratories has shown that a variety of metal chelators and of redox-active aromatic compounds can dramatically induce expression ofsodA. Here we show that non-redox-active, non-metal-chelating aromatic compounds also enhance expression of a chromosomalsodA gene fusion and that these effects are strongly modulated by the Fur phenotype (Fur±) and by the availability of iron in the culture medium. Unsaturated fatty acids are nitrated endogenously to produce nitrated lipids. Recent studies have shown that these nitrated lipids have high chemical reactivity and profound biological implications. We report an efficient, enzymatic synthesis of nitrated derivatives of the oleic acid. The methyl oleate could react with NO and horseradish peroxidase (HRP)/H2O2/NO based nitrating systems to give various nitration products which could be isolated by silica gel column and TLC fractionation, respectively. As reacting directly with NO, the obtained products contain (E)-methyl 9-nitrooctadec-9-enoate (1), (E)-methyl 10-nitrooctadec-9-enoate (2), (E)-methyl 9-nitrooctadec-10-enoate (3) and (E)-methyl 10-nitrooctadec-8-enoate (4), characterized by extensive IR, NMR and GC–MS analysis. Whereas the products of the reaction between the methyl oleate and NO with the presence of HRP/H2O2 were mainly composed of (E)-methyl 9-nitrooctadec-9-enoate and (E)-methyl 10-nitrooctadec-9-enoate. The improving selectivity of the products is attributed to the HRP catalysis system.  相似文献

The surfaces of cells of Desulfovibrio desulfuricans,Desulfovibrio vulgaris and a new strain, Desulfovibrio sp. `Oz-7' were used to manufacturea novel bioinorganic catalyst via the reduction of Pd(II) to Pd(0) at the cell surface usinghydrogen as the electron donor. The ability of the palladium coated (palladised) cells to reductivelydehalogenate chlorophenol and polychlorinated biphenyl species was demonstrated. Dried, palladisedcells of D. desulfuricans, D. vulgaris and Desulfovibrio sp. `Oz-7'were more effective bioinorganic catalysts than Pd(II) reduced chemically under H2 orcommercially available finely divided Pd(0). Differences were observed in the catalyticactivity of the preparations when compared with each other. Negligible chloride release occurredfrom chlorophenol and polychlorinated biphenyls using biomass alone.  相似文献

The conjugation of a ligand to an enzyme is often a necessary step in the development of enzyme-linked immunoassays. Such conjugation is typically accomplished by reacting an amine with a carboxyl functional group in the presence of an activator such as a carbodiimide. However, one enzyme's free carboxyl groups often react with another's free amino groups and a large amount of cross-linking between enzyme molecules occurs; few discrete enzyme molecules conjugated only to the ligand of interest are produced. Hence, it is necessary to carry out laborious chromatographic purification steps or to make an activated ligand such as an N-hydroxysuccinimide ester. This too can be a difficult task because N-hydroxysuccinimide esters are not stable in protic solvents and many biological ligands that would be of interest are poorly soluble in organic solvents. This difficulty may limit the quantity and yield of product. We describe a method that eliminates enzyme-enzyme cross-linking by blocking the solvent-accessible carboxyl groups of horseradish peroxidase and alkaline phosphatase, with dialysis being the only purification step necessary. We are consequently able to produce enzyme-ligand conjugates in high purity and in large quantity with little effort and in a relatively short period of time.  相似文献

The last step of sulfur assimilation is catalyzed by O-acetylserine(thiol)lyase (OASTL) enzymes. OASTLs are encoded by a multigene family in the model plant Arabidopsis thaliana. Cytosolic OASA1 enzyme is the main source of OASTL activity and thus crucial for cysteine homeostasis. We found that nitrating conditions after exposure to peroxynitrite strongly inhibited OASTL activity. Among OASTLs, OASA1 was markedly sensitive to nitration as demonstrated by the comparative analysis of OASTL activity in nitrated crude protein extracts from wild type and different oastl mutants. Furthermore, nitration assays on purified recombinant OASA1 protein led to 90% reduction of the activity due to inhibition of the enzyme, as no degradation of the protein occurred under these conditions. The reduced activity was due to nitration of the protein because selective scavenging of peroxynitrite with epicatechin impaired OASA1 nitration and the concomitant inhibition of OASTL activity. Inhibition of OASA1 activity upon nitration correlated with the identification of a modified OASA1 protein containing 3-nitroTyr(302) residue. The essential role of the Tyr(302) residue for the catalytic activity was further demonstrated by the loss of OASTL activity of a Y302A-mutated version of OASA1. Inhibition caused by Tyr(302) nitration on OASA1 activity seems to be due to a drastically reduced O-acetylserine substrate binding to the nitrated protein, and also to reduced stabilization of the pyridoxal-5'-phosphate cofactor through hydrogen bonds. This is the first report identifying a Tyr nitration site of a plant protein with functional effect and the first post-translational modification identified in OASA1 enzyme.  相似文献

Ten different Pseudomonas strains isolated from contaminated soils were tested for expression of active dioxygenases. Of these, two different clusters, related to strain origin were observed. The first included two P. fluorescens strains and two P. aeruginosa strains isolated from soils polluted with polyaromatic hydrocarbons and the second two P. cepacia strains and four P. chlororaphis strains from soils with polyphenols. All the isolates showed catechol 1,2-dioxygenase basal activity, while other dioxygenases (catechol 2,3-dioxygenase, protocatechuate 2,3-, 3,4- and 4,5-dioxygenases) were detected only after growth in the presence of suitable inducers (benzoate, catechol, salicylate, phenol). Significant induction of catechol 1,2-dioxygenase, the major activity of the tested strains, was also observed when combining starvation with the presence of high molecular weight aromatic hydrocarbons with recalcitrant structures (fluoranthene, chrysene, benzanthracene, pyrene).  相似文献
