Summary

Development of the upper vas deferens and seminal vesicle, and the ecdysteroid titers of the cabbage armyworm (Mamestra brassicae) during pharate adult stage were investigated to provide evidence of endogenous control of the sperm movement through the reproductive tract. Apparently, development of the upper vas deferens is initiated when ecdysteroid titers increase after pupation and continues until eclosion. Sperm movement correlates with the decrease of ecdysteroid titers. When ecdysteroid titers remained at low levels, sperm release from the testis was observed one day before eclosion.  相似文献

Summary A highly specific recognition system, capable of foreseeing and distinguishing between two critical points in time, exists in Mamestra brassicae (Lepidoptera: Noctuidae). Both points in time, the onset of a drought period and the end of the growth season, require different growth patterns of the pupae. In order to minimize the likelihood of weather-induced mortality and to maximize fitness, individuals of M. brassicae must enter aestival dormancy or hibernal diapause, respectively, before the onset of drought or frost. This study is primarily concerned with aestival dormancy. Normally, the pupal period of dormancy-free developing individuals amounts to approximately 20 to 30 days. A modified pupal period of approximately 35 to 80 days is defined as aestival dormancy. The onset of aestival dormancy is triggered by day lengths exceeding an innate individaul-specific threshold. The results reported in this paper indicate that the photoperiodic response curve represents largely the genetic variability within a population with respect to the thresholds triggering aestival dormancy. This variability in thresholds is considered to reflect the frequency of correlation of a distinct day length with a certain onset of drought period in the past. Furthermore, the innate thresholds are characterized by a temperature dependent norm of reaction. Our results also indicate, that a strong genetical component is involved in variability of duration of the pupal period. This variability in duration of aestivation reflects the frequency of drought periods of a certain length in the past. The adaptive significance of both the variation in day length thresholds and duration of aestival dormancy is discussed with respect to the number of generations per season, and the synchronization of the individual life cycles with the seasonal changing environmental conditions.This study is dedicated to Prof. H.J. This study examines phenotypic plasticity in relation to rearing density in larvae of the moth, Mamestra brassicae. Larval phase, growth rate, weight at moulting and susceptibility to disease were quantified when reared at five densities. Larvae develop more quickly, but attain a smaller size and are more susceptible to disease, when reared at high than at intermediate densities. They also exhibit a higher degree of melanisation than larvae reared at intermediate densities, or singly. A review of the literature suggests that a switch to a rapidly developing dark phase at high densities is a widespread phenomenon within the Lepidoptera. Rapid development at the expense of attaining a large size, and increased melanisation, are interpreted as adaptive responses to reach pupation before food supplies are depleted, as is likely when larval density is high. High susceptibility to viral infection at high density may be a result of physiological stress associated with rapid development, or due to a shift in allocation of resources from resistance to development: larvae that developed quickly were more susceptible to infection. Larvae reared singly appeared to be less fit than larvae reared at intermediate densities: they exhibited many of the characteristics of larvae reared at high density, particularly low weight, a right-hand skew in their weight frequency distribution, and high susceptibility to disease. I hypothesise that expression of resistance may be phenotypically plastic with regard to environment. Contact with other larvae may, up to a point, stimulate both growth and resistance to infection, for the risk of infection will increase with the density of conspecifics.  相似文献

Summary In a geographically wide distribution the life cycles of different populations of the cabbage moth Mamestra brossicae are adapted to a remarkable diversity of climatic conditions. This is undoubtedly a proof of its success in adaptation. Some populations living in regions characterized by a drought period interrupting the growth season are capable of distinguishing between one critical day length signalling the onset of the drought period and another signalling the end of the growth season. This study, therefore, is primarily concerned with the geographical patterns in the variability of the adaptional responses of populations exposed to environmental conditions requiring different strategies and tactics in, synchronizing individual, life cycles. It is also a contribution to our understanding of evolutionary mechanisms maintaining median responses to photoperiodically inductive day lengths in geographically different populations. The populations investigated originated from regions differing in predictability of the incidence, onset and duration of a drought period: Freiburg (48.0°N, Southern Germany), Avignon (44.0°N, Southern France), and Argelès (42.5°N, Southern France). Geographical variation with respect to both onset and duration of a drought period consequently results in clinal variation of the variability of innate day length thresholds triggering aestival dormancy and of innate duration of aestivation. In this paper we considered the influence of geographically changing temperatures on aestival dormancy induction. Even in southern populations of M. brassicae a temperature dependent switch off-mechanism exists which prevents aestival dormancy under certain environmental conditions. The effective temperatures vary geographically, too. The courtship behavior of the cabbage moth, Mamestra brassicae(L.), was studied in moving air conditions in a wind tunnel, using video techniques. Quantitative analyses were undertaken to determine the behavioral sequence occurring in the courtship. Comparison of successful and unsuccessful courtship suggested that courtship success was more dependent on the behavior of the female than that of the male. In an attempt to elucidate the function of the male hair-pencils (HPs), courtships involving males without HPs were also studied. HP removal did not affect the overall courtship success rate of males, but detailed analysis showed significant changes infernale behavior during such courtships. HP removal also affected female behavior following pair formation, with females struggling more when paired with males without HPs. Consequently, it is proposed that the HP volatiles act as an arrestant for the female, both during courtship and after pair formation, to increase female acceptance and to prevent premature termination of copulation. Experiments were also conducted to test previous hypotheses for HP function. However, no evidence was found to suggest that the HP volatiles in M. brassicaeact to attract females, affect female calling behavior, or affect the behavior of other males. A further possible function of HPs in defense is discussed.  相似文献

Proteolytic activities in soluble protein extracts from Mamestra brassicae (cabbage moth) larval midgut were analysed using specific peptide substrates and proteinase inhibitors. Serine proteinases were the major activities detected, with chymotrypsin-like and trypsin-like activities being responsible for approximately 62% and 19% of the total proteolytic activity towards a non-specific protein substrate. Only small amounts of elastase-like activities could be detected. The serine proteinases were active across the pH range 7-12.5, with both trypsin-like and chymotrypsin-like activities maximal at pH 11.5. The digestive proteinases were stable to the alkaline environment of the lepidopteran gut over the timescale of passage of food through the gut, with 50% of trypsin and 40% of chymotrypsin activity remaining after 6h at pH 12, 37 degrees C. Soybean Kunitz trypsin inhibitor (SKTI) ingestion by the larvae had a growth-inhibitory effect, and induced inhibitor-insensitive trypsin-like activity. Qualitative and quantitative changes in proteinase activity bands after gel electrophoresis of gut extracts were evident in SKTI-fed larvae when compared with controls, with increases in levels of most bands, appearance of new bands, and a decrease in the major proteinase band present in extracts from control insects.  相似文献

The peritrophic matrix (PM) is essential for insect digestive system physiology as it protects the midgut epithelium from damage by food particles, pathogens, and toxins. The PM is also an attractive target for development of new pest control strategies due to its per os accessibility. To understand how the PM performs these functions, the molecular architecture of the PM was examined using genomic and proteomic approaches in Mamestra configurata (Lepidoptera: Noctuidae), a major pest of cruciferous oilseed crops in North America. Liquid chromatography‐tandem mass spectrometry analyses of the PM identified 82 proteins classified as: (i) peritrophins, including a new class with a CBDIII domain; (ii) enzymes involved in chitin modification (chitin deacetylases), digestion (serine proteases, aminopeptidases, carboxypeptidases, lipases and α‐amylase) or other reactions (β‐1,3‐glucanase, alkaline phosphatase, dsRNase, astacin, pantetheinase); (iii) a heterogenous group consisting of polycalin, REPATs, serpin, C‐Type lectin and Lsti99/Lsti201 and 3 novel proteins without known orthologs. The genes encoding PM proteins were expressed predominantly in the midgut. cDNAs encoding chitin synthase‐2 (McCHS‐2), chitinase (McCHI), and β‐N‐acetylglucosaminidase (McNAG) enzymes, involved in PM chitin metabolism, were also identified. McCHS‐2 expression was specific to the midgut indicating that it is responsible for chitin synthesis in the PM, the only chitinous material in the midgut. In contrast, the genes encoding the chitinolytic enzymes were expressed in multiple tissues. McCHS‐2, McCHI, and McNAG were expressed in the midgut of feeding larvae, and NAG activity was present in the PM. This information was used to generate an updated model of the lepidopteran PM architecture.  相似文献

Specialised phytophagous Lepidoptera such as Bombyx mori and Pieris brassicae have contact chemoreceptors that perceive ecdysteroids at very low concentrations. This sensory perception allows them to feed on substrates with a high content of phytoecdysteroids. We have evaluated if a polyphagous insect like Mamestra brassicae does possess contact chemoreceptor cells that are sensitive to these molecules. Electrophysiological recordings were performed from contact chemoreceptors located on the maxilla. These receptors were stimulated with some sugars, amino acids and salts and with three ecdysteroids. Our results demonstrate that a specific cell within the lateral sensilla responds to 20-hydroxyecdysone and ponasterone A but not to ecdysone.  相似文献

The activities of digestive protease within the midgut of Mamestra configurata (bertha armyworm) larvae were examined using specific substrates and protease inhibitors. The bulk of the activity was associated with serine proteases comprising trypsin-, chymotrypsin-, and elastase-like enzymes. At least 10-15 serine protease isozymes were detected using one-dimension gelatin gel electrophoresis. Cysteine or aspartic protease activities were not present; however, amino- and carboxypeptidase activities were associated with the midgut extract. Midgut proteases were active in the pH range of 5.0-12.0 with peaks at pH 7.5 and 11.0. In general, the middle region of the midgut exhibited a higher pH (approximately 8.0) than either the posterior or anterior regions (approximately 7.3-7.7). Moulting larvae possessed a neutral gut pH that was 0.5-1.5 units below that of feeding larvae. Degenerate PCR and expressed sequence tag (EST)-based approaches were used to isolate 30 distinct serine protease encoding cDNAs from a midgut-specific cDNA library including 8 putative trypsins, 9 chymotrypsins, 1 elastase, and 12 whose potential activities could not be determined. cDNAs encoding three amino- and two carboxypeptidases were also identified. Larvae feeding upon artificial diet containing 0.2% soybean trypsin inhibitor experienced a significant delay in development.  相似文献

The midgut protease profiles from 5th instar Mamestra configurata larvae fed various diets (standard artificial diet, low protein diet, low protein diet with soybean trypsin inhibitor [SBTI], or Brassica napus) were characterized by one‐dimensional enzymography in gelatin gels. The gut protease profile of larvae fed B. napus possessed protease activities of molecular masses of approximately 33 and 55 kDa, which were not present in the guts of larvae fed artificial diet. Similarly, larvae fed artificial diet had protease activities of molecular masses of approximately 21, 30, and 100 kDa that were absent in larvae fed B. napus. Protease profiles changed within 12 to 24 h after switching larvae from artificial diet to plant diet and vice versa. The gut protease profiles from larvae fed various other brassicaceous species and lines having different secondary metabolite profiles did not differ despite significant differences in larval growth rates on the different host plants. Genes encoding putative digestive proteolytic enzymes, including four carboxypeptidases, five aminopeptidases, and 48 serine proteases, were identified in cDNA libraries from 4th instar M. configurata midgut tissue. Many of the protease‐encoding genes were expressed at similar levels on all diets; however, three chymoptrypsin‐like genes (McSP23, McSP27, and McSP37) were expressed at much higher levels on standard artificial diet and diet containing SBTI as was the trypsin‐like gene McSP34. The expression of the trypsin‐like gene McSP50 was highest on B. napus. The adaptation of M. configurata digestive biochemistry to different diets is discussed in the context of the flexibility of polyphagous insects to changing diet sources. Aim of this study was to investigate relationships between the red palm weevil (RPW) Rhynchophorus ferrugineus (Olivier) and the entomopathogenic nematode Steinernema carpocapsae (EPN); particularly, the work was focused on the immune response of the insect host in naive larvae and after infection with the EPN. Two main immunological processes have been addressed: the activity and modulation of host prophenoloxidase‐phenoloxidase (proPO) system, involved in melanization of not‐self and hemocytes recognition processes responsible for not‐self encapsulation. Moreover, immune depressive and immune evasive strategies of the parasite have been investigated. Our results suggest that RPW possess an efficient immune system, however in the early phase of infection, S. carpocapsae induces a strong inhibition of the host proPO system. In addition, host cell‐mediated mechanisms of encapsulation, are completely avoided by the parasite, the elusive strategies of S. carpocapsae seem to be related to the structure of its body‐surface, since induced alterations of the parasite cuticle resulted in the loss of its mimetic properties. S. carpocapsae before the release of its symbiotic bacteria, depress and elude RPW immune defenses, with the aim to arrange a favorable environment for its bacteria responsible of the septicemic death of the insect target.  相似文献

【目的】明确自然变温环境对甘蓝夜蛾Mamestra brassicae生长发育和繁殖的影响,深入研究其对环境温度的适应性。【方法】在13~25℃(日平均19℃), 16~28℃(日平均22℃), 19~31℃(日平均25℃), 22~34℃(日平均28℃)和25~37℃(日平均31℃) 5个梯度变温条件下,以大豆Glycine max植株叶片为寄主材料饲养甘蓝夜蛾卵,测定其各虫态发育历期、发育速率、成虫繁殖力及发育起点温度和有效积温。【结果】变温范围为13~25℃时甘蓝夜蛾发育历期最长,世代发育历期为65.93 d,显著长于其他变温处理。且随温度升高,其发育历期缩短,变温范围为22~34℃时,该虫发育历期最短,世代发育历期为38.46 d,显著短于其他变温处理。在变温范围为25~37℃时,该虫不能正常完成个体发育。在日平均温度(T)19~28℃范围内(最大温差12℃),甘蓝夜蛾卵、幼虫及蛹期的发育速率随温度升高而加快,且各个虫态发育速率(V)拟合方程均符合线性方程模型:V卵期=0.125+0.048T, V幼虫期=0.023+0.012T, V蛹期0.027+0.013T, V成虫=0.073+0.47T。甘蓝夜蛾雌雄成虫的寿命随着日平均温度的升高而逐渐缩短,雌雄成虫寿命在日变温范围13~25℃时最长,分别为7.91 d和8.00 d;在变温范围22~34℃时最短,分别为3.00 d和3.57 d。甘蓝夜蛾卵、幼虫、蛹、成虫发育起点温度分别为7.98, 6.54, 9.36和10.78℃,有效积温依次为87.00, 607.36, 351.51和108.52 d·℃。16~28℃的变温范围更适合甘蓝夜蛾种群的生存与繁殖,其种群趋势指数I为117.81。【结论】甘蓝夜蛾属于偏低温适应性害虫,对高温环境适应能力较低。研究结果为进一步研究甘蓝夜蛾自然种群发生规律及其发生期、发生量预测预报提供了科学依据。  相似文献

Baculovirus infection in Lepidoptera can alter both larval mobility and feeding rates, which can in turn affect pathogen transmission and dispersal in the field. We compared the damage to cabbage plants in the field caused by healthy and nucleopolyhedrovirus-infected Mamestra brassicae L. (Lepidoptera: Noctuidae) larvae released as second and fourth instars. There was no significant difference in plant consumption by healthy and infected larvae for the first 4 days after release. From day 5 onwards, infected larvae caused significantly less defoliation. This pattern was similar for larvae at both larval instars. Defoliation was greater for fourth instars throughout the experiment.  相似文献

The synergistic enhancement of nucleopolyhedrovirus (NPV) infection by granuloviruses (GVs) is well documented; and a GV granule protein, named viral enhancin, has been identified as an active contributor to this effect. We detected the presence of two proteins with molecular mass of 93 and 108 kDa in granules of a GV isolated from Xestia c-nigrum (L.) (XecnGV) as candidates for enhancin, and we confirmed that at least the 108-kDa protein enhances the infectivity of Mamestra brassicae nucleopolyhedrovirus (MabrNPV). We tested the effect of virion-free proteins obtained from XecnGV granules (GVPs) on MabrNPV infection, and we made a comparison with an enhancing chemical, the stilbene-derived fluorescent brightener Tinopal. Bioassay was performed employing the diet contamination method, by using second instars of Mamestra brassicae (L.) (Lepidoptera: Noctuidae). The enhancing effects of GVPs (0.1 mg/g diet) and Tinopal (1 mg/g diet) were estimated to be 70.7-81.5-fold and 26.9-33.7-fold, respectively, as calculated from the LC50 values of MabrNPV with or without the additives. The additives reduced the lethal time of MabrNPV-infected larvae and they caused death at a younger instar. These results suggest that GVPs can enhance MabrNPV infection as effectively as Tinopal.  相似文献

We investigated the effects of a diamondback moth-resistant Chinese cabbage (Brassica campestris subsp napus variety pekinensis Makino), expressing the insecticidal protein CrylA(c) toxin derived from Bacillus thuringiensis, on the nontarget herbivore Mamestra brassicae (L.) (Lepidoptera: Noctuidae) and its parasitoid wasp Microplitis mediator (Haliday) (Hymenoptera: Braconidae). A decreased survival rate at neonate stage was observed in M. brassicae when reared on Bt cabbage, although overall development was not significantly affected. According to enzyme-linked immunosorbent assay test using CrylA(c) antibody, the Cry toxin was only detected in the alimentary canal, not in the hemolymph or remaining body parts of M. brassicae, indicating that the ingested Cry toxin is neither distributed inside the body nor transferred through the trophic level. As expected, no Cry toxin was found in the larvae and cocoons of M. mediator. In addition, no significant changes were observed in the parasitization rate, larval period, pupal period, cocoon weight, or adult emergence rate when M. mediator wasps were reared on the M. brassicae larvae fed with transgenic Chinese cabbage. In summary, no direct or indirect adverse effects of transgenic Chinese cabbage on the two nontarget insect species were observed, suggestive of low risk in herbivore-parasitoid food chain.  相似文献

The evaluation of Eurithia consobrina (Meigen) as a candidate for introduction against bertha armyworm, Mamestra configurata (Walker) (Lepidoptera: Noctuidae), in western Canada included studies of its host range, distribution, synchronization with host, constancy, abundance and life history in Europe. In addition, its diapause induction and its coldhardiness were compared with those of the target host M. configurata and its native tachinid parasitoid, Athrycia cinerea (Coq.). E. consobrina was found to fit criteria for a successful biocontrol agent reasonably well. It also had the potential to fill a largely unoccupied niche in the parasitoid complex of M. configurata. E. consobrina has a facultative diapause of the long-day type, similar to A. cinerea, but is less sensitive to diapause-inducing conditions. E. consobrina is at least as coldhardy as A. cinerea and more coldhardy than M. configurata. Laboratory exposures to temperatures of 0 C for 140 days apparently selected for greater coldhardiness, and later generations survived exposures of 140 days at -15 C without significant mortality. Adult E. consobrina were released at three locations in Manitoba in 1986 and 1987, but establishment has not been confirmed.  相似文献

【目的】本研究旨在筛选小菜蛾Plutella xylostella应对玫烟色棒束孢Isaria fumosorosea侵染的免疫应答及其网络调控基因,以进一步探讨小菜蛾对玫烟色棒束孢的防御机制。【方法】采用第二代高通量测序技术RNA-seq,对处理后12 h的感染玫烟色棒束孢和健康(平行对照)的小菜蛾4龄幼虫转录组进行了测序,通过生物信息学分析对得到的差异基因进行了功能注释和分类,对差异基因参与的信号通路进行了分析。【结果】在感菌和健康小菜蛾幼虫转录组两个表达谱里,分别获得了12 346 987和12 315 210个clean reads,有60.93%和61.26%的reads分别能比对到参考基因库里,其中完美匹配(perfect match)的比例分别为32.15%和32.73%。共得到351个显著差异表达基因(differentially expressed unigenes,DEUs),上调表达基因有275个,下调表达基因有76个,与免疫防御反应潜在相关的基因有156个。GO(Gene Ontology)富集分析有102个DEUs分布到46个GO term里,KEGG(Kyoto Encyclopedia of Genes and Genomes)pathway富集分析结果显示,有132个DEUs显著富集在13个代谢通路(pathway)里。【结论】这些差异表达基因中,大部分编码潜在的与免疫识别及调控相关的基因,集中在能量代谢、疾病反应和防御反应等相关通路。研究结果为挖掘与玫烟色棒束孢侵染相关的小菜蛾免疫应答候选基因提供了重要数据库,也为阐明小菜蛾对玫烟色棒束孢的免疫机制奠定理论基础。  相似文献

The interactions between the entomopathogenic bacterium Bacillus thuringiensis ssp. kurstaki and two entomopathogenic fungi Beauveria bassiana Balsamo (Vuillemin) (Hypocreales: Cordycipitaceae) and Metarhizium robertsii (Metchnikoff) Sorokin (Hypocreales: Clavicipitaceae) were examined on larvae of Sesamia nonagrioides (Lefebvre) (Lepidoptera: Noctuidae) in 8, 13 and 16 days post-treatment intervals. An overall positive interaction between the pathogens was observed and the larval mortality at 16 days was 56–100 % exposed to M. robertsii combined with B. thuringiensis subsp. kurstaki, whereas B. bassiana combined with B. thuringiensis ssp. kurstaki killed 54–100 % of exposed larvae. After 8 days, in 6 of the combinations, we found an additive relationship between the pathogens, whereas, a negative interaction was observed in 10 of them. In contrast, after 13 days, in 2 of the combinations the positive interaction could be considered as synergistic between pathogens, in 10 as additive, and in only 4 as negative. Finally, after 16 days, in 11 of the combinations we found an additive connection between the pathogens, wheras a negative interaction was seen in 5. Applying both pathogens simultaneously offers a method of Sesamia nonagrioides control that could be more effective than using each pathogen separately.  相似文献
