A new type of toxicity biphasically dependent on concentration was observed with diethyldithiocarbamate, a metal chelator utilized in medicine. As judged by cell survival and [3H]Urd incorporation, diethyldithiocarbamate was maximally toxic to T lymphocytes and polymorphonuclears at 2.5×10?5 M (first phase) and at higher than 2.5×10?3 M (second phase), but was not toxic at intermediate concentrations around 2.5×10?4 M. The response of chelator treated T lymphocytes to phytohemagglutinin was also biphasic. The first toxic phase was partially reversed by 2.5×10?5 M ZnCl2, while the second phase was partially reversed by 10?2 M CuCl2. This suggests that inhibition of Zn-metalloenzymes in the first phase and of Cu-metalloenzymes in the second may play a crucial role in the mechanism of toxicity. The second toxic phase may be in part due to the observed inhibition of superoxide dismutase rendering the cells susceptible to oxygen toxicity, like obligate anaerobes.  相似文献

Effect of heavy metal ions on the growth and the iron-oxidizing activity of Thiobacillus ferrooxidans were investigated.

Cupric, zinc, cadmium, and chromium ions had no effect on the growth and the iron-oxidizing activity of cell suspensions or cell-free extracts of the bacterium in high concentrations (10?3~10?2M). Lead ion delayed the start of the growth slightly in 10?3 M, but it did not inhibit the iron-oxidizing activity of the cells in the concentration. Tin and molybdenum oxide ions inhibited both of them in the concentration above 10?3 M.

Mercuric mercurous, and silver ions had the most harmful effect. In the concentration of 10?3 .M, each of the cations inhibited almost completely both the growth and the iron-oxidizing activity of the cells.

In the experiments with cell-free extracts it was observed that the activity of cytochrome oxidase (cytochrome a597) operating in the iron-oxidizing system of the bacterium was specifically inhibited with mercuric ion in the concentration above 5 × 10?4 M.  相似文献

The metamorphic response of pediveliger larvae of Mytilus galloprovincialis to the neurotransmitter blockers chlorpromazine, amitriptyline, rauwolscine, idazoxan, atenolol and butoxamine, and to tetraethylammonium chloride (TEA) was investigated through a series of bioassays. Chlorpromazine, amitriptyline and idazoxin inhibited larval metamorphosis induced by 10?4 M epinephrine. The concentration that inhibited metamorphosis by 50% (IC50) for chlorpromazine and amitriptyline was 1.6 × 10?6 M and 6.6 × 10?5 M, respectively. Idazoxan was less effective with an IC50 of 4.4 × 1013 M. Moreover, these three inhibitors showed no toxicity at any of the concentrations tested. The larval metamorphic response to K+ was not inhibited by 10?3 M tetraethylammonium chloride after 96 h. Thus, the neurotransmitter blockers chlorpromazine and amitriptyline are inhibitors of larval metamorphosis, and will be useful tools for antifouling studies.  相似文献

The early effects of penconazole (PCZ) at relatively high concentration (10?4 to 5 × 10?4 M) on changes in pH and in titratable acidity of the medium, transmembrane electrical potential difference (Em), electrolyte leakage and cell morphology were investigated in Egeria densa leaves. At the lowest (10?4 M) concentration and in the presence of a very low (10 μM) K+ concentration, triazole induced an early, moderate hyperpolarization of Em, associated with a decrease of net K+ uptake, suggesting some increase in the passive permeability to K+. This Em hyperpolarization was no longer detectable at high (2 mM) K+out concentration. At high PCZ concentrations (3 × 10?4 M and 5 × 10?4 M) the early hyperpolarization detectable in the presence of a low K+out concentration became transient, and was followed by a marked depolarization. PCZ, at these concentrations, suppressed acidification of the medium, stimulated electrolyte leakage and, in the mesophyll cells, induced some shrinking of the cytoplasm and its disconnection from the cell walls. These results are interpreted as due to an early effect of this triazole leading to the disorganization of the plasma membrane.  相似文献

Summary Different influences, like changes in salinity and temperature, the effects of heavy metals like copper and mercury, and the influences of nitrate and silicate deficiencies on the shape, generation time, and yield of the marine plankton diatomBiddulphia sinensis were tested in batch cultures. Those cells which show multiple or missing setae are classified as seta-aberrant, teratological forms. Sudden changes in salinity and temperature (30–15%. S; 12–20°C) lead to no more than 5% of aberrant cells (control cultures below 1%). Heavy metals (copper and mercury) cause a significant increase of the generation time, a distinct decrease of the cell yield, and (in the case of copper) up to 50% of seta-aberrant cells. The cells die at concentrations of copper above 10–5M.l–1, though EDTA was added as chelant. At concentrations below 10–5M.l–1 of copper, addition of EDTA as well as an increase of temperature from 12 to 20°C cause a reduction of toxicity. During batch growth the cells adapt to mercury but not to copper. At mercury concentrations above 10–8M.l–1 the cells die, even if EDTA is added. Unlike in copper-treated medium, EDTA does not reduce the toxicity of mercury. However, a detoxicating effect (20 instead of 12°C) can be observed due to higher temperature. Silicate deficiency, like the influence of copper and mercury leads to an elongation of the pervalvar axes. Particularly at a temperature of 20°C bended pervalvar axes and sometimes lateral evaginations of the frustule will be formed; this does not happen under the influence of copper and mercury. Multiple setae never appear under silicate deficiency. Under nitrate deficiency more than 10% of the cells are seta-aberrant. The suitability ofBiddulphia sinensis for test experiments in shipboard cultures is shown. Water samples were tested on the lack of nitrate, silicate, and phosphate. The percentage of seta-aberrant cells, the generation times, and the yields were determined. Simultaneous lack of phosphate and nitrate causes up to 30% of teratological cells. The seta-aberrant cells always appear during the exponential growth phase.  相似文献

The interactions of ganglioside GM1 with human and fetal calf sera were studied, the following main results being obtained: (a) GM1, upon incubation with both sera gave origin to two GM1-protein complexes, which also occurred after interaction of GM1 with the albumin fractions prepared from the same sera. Instead no complex formation occurred using the albumin-free fractions. Therefore GM1 appeared to specifically bind serum albumin and to form GM1-albumin complexes. (b) GM1 binding to serum albumin started at ganglioside concentrations surely micellar (above 10?6 M), was time and concentration dependent, and resulted in a relevant degree of GM1 complexation (up to 80% of total GM1 in human serum and up to 18% in fetal calf serum). (c) the binding kinetics appeared, in both serum and the correspondent albumin fraction, to be biphasic: in the first phase, occurring till about 2 · 10?4 M GM1, the ratio between bound and total GM1 increased linearly with increasing GM1 concentration; in the second phase, occurring above 2 · 10?4 M, the ratio remained practically constant. After these findings it should be expected that GM1, when present in serum containing systems, forms complexes with albumin. This should be appropriately considered when studying the effects of exogeneous GM1 in in vivo and in vitro (tissue cultures) systems.  相似文献

Mouse calvaria were maintained in organ culture for 96 h and endogenous prostaglandin production and active bone resorption (45 Ca release) measured. After a lag phase of 12 h, active resorption increased over the 96 h period. The amounts of prostaglandins released into the culture medium (measured by radioimmunoassay) were highest in the first 24 h of culture. Unless these were removed by preculturing for 24 h, or suppressed by indomethacin, no response to exogenous PGE2, PGF or prostaglandin precursors could be demonstrated. Bone resorption was stimulated after preculture by both PGE2 and PGF in a dose-dependent manner (10?18M – 10?5M), with PGE2 being the more potent. Collagen synthesis was unaffected by PGF, whereas PGE2 (10?5M) had an inhibitory effect. Eicosatrienoic acid did not stimulate bone resorption at lower concentrations (10?7M – 10?5M_, but was inhibitory at 10?4M. Arachidonic acid also inhibited resorption at 10?4M, but at lower concentrations (10?7M – 10?5M0 increased active resorption. This was concomitant with a rise in PGE2 and PGF levels, PGE2 production being significantly higher than PGF. The effects of PGE2 (10?8M) and PGF (10M appeared additive: there was no evidence of synergistic or antagonistic effects when varying ratios of PGE2 : PGF2α were employed.  相似文献

The polyphenol oxidase (LsPPO) from a wild edible mushroom Lactarius salmonicolor was purified using a Sepharose 4B-L-tyrosine-p-amino benzoic acid affinity column. At the optimum pH and temperature, the KM and VMax values of LsPPO towards catechol, 4-methylcatechol and pyrogallol were determined as 0.025 M & 0.748 EU/mL, 1.809 × 10? 3 M & 0.723 EU/mL and 9.465 × 10? 3 M & 0.722 EU/mL, respectively.

Optimum pH and temperature values of LsPPO for the three substrates above ranged between the pH 4.5–11.0 and 5–50°C. Enzyme activity decreased due to heat denaturation with increasing temperature. Effects of a variety of classical PPO inhibitors were investigated opon the activity of LsPPO using catechol as the substrate. IC50 values for glutathione, p-aminobenzenesulfonamide, L-cysteine, L-tyrosine, oxalic acid, β-mercaptoethanol and syringic acid were determined as 9.1 × 10? 4, 2.3 × 10? 4 M, 1.5 × 10? 4 M, 3.8 × 10? 7 M, 1.2 × 10? 4 M, 4.9 × 10? 4 M, and 4 × 10? 4 M respectively. Thus L-tyrosine was by far the most effective inhibitor. Interestingly, sulfosalicylic acid behaved as an activator of LsPPO in this study.  相似文献

Aims: To investigate the effect of lactic acid (LA), copper (II), and monolaurin as natural antimicrobials against Cronobacter in infant formula. Methods and Results: The effect of LA (0·1, 0·2 and 0·3% v/v), copper (II) (10, 50 and 100 μg ml?1) and monolaurin (1000, 2000, and 3000 μg ml?1) suspended into tween‐80? or dissolved in ethanol against Cronobacter in infant formula was investigated. Reconstituted infant formula and powdered infant formula were inoculated with five strains of Cronobacter spp. at the levels of c. 1 × 106 CFU ml?1 and 1 × 103 CFU g?1, respectively. LA at 0·2% v/v had a bacteriostatic effect on Cronobacter growth, whereas 0·3% v/v LA resulted in c. 3 log10 reduction. Copper (II) at the levels of 50 μg ml?1 and 100 μg ml?1 elicited c. 1 and 2 log10 reductions, respectively. The combination of 0·2% LA and 50 μg ml?1 copper (II) resulted in a complete elimination of the organism. Monolaurin exhibited a slight inhibitory activity against Cronobacter (c. 1·5 log10 difference) compared to the control when ethanol was used to deliver monolaurin. Conclusions: A complete elimination of Cronobacter was obtained when a combination of sublethal concentrations of LA (0·2%) and copper (II) (50 μg ml?1) was used. Significance and Impact of the Study: The use of the synergistic interactive combination of LA and copper (II) could be beneficial to control Cronobacter in the infant formula industry.  相似文献

Summary

A supernatant from eggs of the ruminant nematode Trichostrongylus colubriformis contained an enzyme that was similar to leucine aminopeptidase (LAP), based on hydrolysis of the substrate L-leucine β-naphthylamide to β-naphthylamine. A Michaelis-Menten constant (K m) of 0.155 mM was obtained. Rate of hydrolysis of 16 substrates revealed that L-phenylalanine and L-tyrosine β-naphthylamides were hydrolyzed most readily while seven additional substrates were hydrolyzed at lesser rates. The optimum pH for enzymatic activity was 6.75–7.5. Enzymatic activity was lost by heating the egg supernatant to 60°C for 5 min or freezing at 0°C for 28 days. Addition of millimolar concentrations of the chlorides of zinc, manganese and magnesium to the egg supernatant had no stimulatory effect on enzyme activity while 10 and 100 mM concentrations significantly reduced activity. Ethylenediamine tetraacetic acid at 10?4 M had no effect on enzymatic activity. Activity was inhibited by 10?4 M 1,10-phenanthroline, but the inhibition was reversed by zinc chloride at 10?3 M. Di-isopropylphosphofluoridate at 10?3 M reduced enzymatic activity moderately. Enzyme activity in egg supernatant increased 2.2-fold from 21 days to 60–90 days of a primary infection in the host while a 3.3-fold increase was found in primary versus secondary infections.  相似文献

Studies conducted in Port Philip Bay, Victoria, Australia are described that examined the effect of experimentally elevated copper concentrations on the recruitment of epifauna to settlement plates. Simultaneous measurement of the copper concentration using diffusive gradients in thin films (DGT) allowed direct comparisons to be made between the labile copper concentration measured at the settlement surface, and the biological effects observed. Copper concentrations created by the field dosing technique were between 20?–?30?μg?l?1 for the first 2 d, but then dropped considerably for the following 4 d (3?μg?l?1), and were indistinguishable from background for the final 7 d. The first 2 d of a copper pulse reduced the recruitment of barnacles, ascidians, serpulid worms, an encrusting bryozoan, and didemnid ascidians. The impacts occurred despite the copper pulse being much less than published LC50 values for similar species. The impacts were no longer obvious by day 7 or 14, having been obscured by either high mortality of early settlers, or large settlement events that took place after day 2. Thus the greatest impact of the pollution event occurred during the period of highest toxicant concentration. The value of this study lies in the correlation of toxicity effects with bio-available metal concentrations under realistic (natural, in situ) conditions.  相似文献

The development of new approaches to study the affinity between ligands and G‐protein‐coupled receptors proves to be of growing interest for pharmacologists, chemists, and biologists. The aim of this work was to determine the binding of seven drugs to β2‐adrenoceptors by frontal analysis using immobilized receptor stationary phase. The dissociation constants (Kd) were determined to be (3.16 ± 0.09) × 10?4 M for salbutamol, (4.29 ± 0.12) × 10?4 M for terbutaline, (6.19 ± 0.16) × 10?4 M for methoxyphenamine, (2.11 ± 0.07) × 10?4 M for tulobuterol, (1.82 ± 0.11) × 10?4 M for fenoterol, (9.75 ± 0.24) × 10?6 M formoterol, and (9.84 ± 0.26) × 10?5 M for clenbuterol. These results showed a good correlation with the data determined by radioligand binding assay. Further investigations revealed that the dissociation constant mainly attributed to the number of hydrogen bonds in the structures of ligands. This study indicates that affinity chromatography using immobilized receptor stationary phase can be used for the direct determination of drug‐receptor binding interactions and has the potential to become a reliable alternative for quantitative studies of ligand–receptor interactions. The effects of 5-HT and glutamate on dopamine synthesis and release by striatal synaptosomes were investigated and compared with the action of acetylcholine, which acts presynaptically on this system. 5-HT inhibited (28%) synthesis of [14C]dopamine from L-[U-14C]tyrosine, at 10-5M and above. This contrasts with the action of acetylcholine, which stimulated [14C]-dopamine synthesis by 24% at 10-4 M. Tissue levels of GABA were unaffected by either 5-HT or acetylcholine up to concentrations of 10-4 M. The inhibitory action of 5-HT (5 × 10?5 M and 2 × 10?4 M) on [19C]dopamine synthesis was completely abolished by methysergide (2 × 10?6 M). Higher concentrations of methysergide (10?4 M) or cyproheptadine (10?5 M) inhibited [14C]dopamine synthesis by 28% and 25%, respectively, when added alone to synaptosomes. However, only methysergide prevented the further inhibition of synthesis caused by 5-HT. At concentrations of 2 × 10?5 M and above, 5-HT stimulated [14C]dopamine release. This releasing action differed from that of acetylcholine, which occurred at lower concentrations (e.g., 10?6 M). Methysergide (up to 10?4 M) or cyproheptadine (2 × 10?4 M) did not reduce the 5-HT (5 × 10?5 M)-induced release of [14C]dopamine, but methysergide (10?4 M) showed a potentiation (49%) of this increased release. The stimulatory effects of 5-HT (2 × 10?5 M) and K+ (56 mM) on [14C]dopamine release were additive, indicating that two separate mechanisms were involved. However, when both agents were present the stimulatory effect of K+ (56 mM) on [14C]dopamine synthesis was not seen above the inhibitory effect of 5-HT. Glutamate (0.1-5 mM) did not affect [4C]dopamine release or its synthesis from L-[U-14C]tyrosine. It is concluded that 5-HT modulates the synthesis of dopamine in striatal nerve terminals through a presynaptic receptor mechanism, an action antagonised by methysergide. The releasing action of 5-HT apparently occurs through a separate mechanism which is also distinct from that involved in the response to K+ depolarisation.  相似文献

The effect of copper additions (Cu ranging from 0 to 30?µM) on the photosynthesis of three different microalgal biofilms was studied to identify the factors that cause sensitivity differences between benthic and pelagic algae. The response of biofilms which colonized artificial substrata in the River Meuse was compared with those of two laboratory-grown monospecific biofilms, one consisting of the diatom Synedra ulna, and the other composed of a filament-forming cyanobacterium, Oscillatoria sp. The photosynthetic yield ΦII (quantum efficiency of photosystem II) was studied with PAM (Pulse Amplitude Modulated) fluorimetry. S. ulna biofilms appeared to be the most sensitive to Cu, followed by the cyanobacteria, while natural biofilms, dominated by supposedly very sensitive diatom species such as Melosira varians and Diatoma vulgare, were the most resistant to Cu. In the highly productive biofilms, pH is suggested to play a role in lowering toxicity by helping the precipitation of cupric ions. Cu accumulation by the biofilms during the exposure period followed a linear relationship with Cu concentration, saturation not being observed; natural biofilms had an accumulation factor of 1–2.5?×?103 relative to the concentrations in the water, while the diatoms growing unattached to the substratum had a higher concentration factor, up to 4.9?×?103. It was concluded that the physical structure of the biofilm (package of cells and thickness), and not the species composition, was the main factor regulating the sensitivity of the biofilm to Cu toxicity during short-term exposures.  相似文献

A comparison has been made between the effects of 4-hydroxy-2,3-trans-nonenal (HNE) and 4-hydroxy-2,3-trans-octenal (HOE), two lipid peroxidation products, on the basal and GTPgammaS-stimulated activities of phosphoinositide-specific phospholipase C (PL-C) of rat polymorphonuclear leukocytes. PL-C activity was determined in vitro by measuring the hydrolysis of [3H] phosphatidylinositol-4,5-bis-phosphate (PtdIns-P2) added as exogenous substrate to neutrophil plasma membranes. PL-C was activated by concentrations of HNE ranging from 10?8 to 10?6 M both in the presence and in the absence of 2 × 10?5 M GTPgammaS; HOE stimulated the enzymatic activity between 10?11 and 10?8 M ; maximal stimulation was given by 10?11 M HOE plus GTPgammaS. The aldehyde concentrations able to accelerate PtdIns-P2 breakdown displayed a good correspondence with those which have been reported to stimulate the oriented migration of rat neutrophils. Pretreatment of neutrophils with pertussis toxin prevented the stimulation of PL-C by 10?11 M HOE and by HOE plus GTPgammaS. Our results suggest that the chemotactic action of HNE and HOE might depend on the activation of PL-C; furthermore a regulatory G protein appears to be involved in the acceleration of PtdIns-P2 turnover by HOE.  相似文献

Sodium selenite (Na2Se03) was tested for its sister-chromatid exchange (SCE)-inducing ability in human whole blood cultures and for the effect of its co-exposure with methyl methanesulfonate (MMS) or N-hydroxy-2-acetyl aminofluorene (N-OH-AAF) on SCE frequency. Long exposure times (77 h and 96 h) to 3.95 × 10-6 M Na2SeO3 resulted in cell death as measured by mitotic indices, but mitotic figures were present after exposure to higher concentrations for a shorter time (19 h). High Na2SeO3 concentrations (7.90 × 10?6 and 1.19 × 10?5 M) resulted in a three-fold increase in the SCE frequency above background level (6–7 SCEs/cell). Exposure of lymphocytes to 1 × 10?4 M MMS for the last 19 h of culture yielded an average SCE frequency of 30.17 ± 0.75 while a similar exposure to 2.7 × 10?5 M N-OH-AAF resulted in 13.61 ± 0.43 SCEs/cell. Simultaneous addition of the high Na2Se03 concentrations and MMS or N-OH-AAF to the cultures resulted in SCE frequencies that were 25–30% and 11–17%, respectively, below the sum of the SCE frequencies produced by the individual compounds.  相似文献

Amiloride in the mucosal fiuid (at concentrations of 5 · 10?6 M to 10?4 M) reversibly stimulates the HCO3?-dependent moiety of the short-circuiting current (Isc) in ouabain-treated turtle bladders bathed by Na-free Ringer solutions with or without Cl?.This effect is uniquely different from the known inhibitory effect of this agent on Na+ transport. Leukotrienes C4 (LTC4) and D4 (LTD4), major components of slow-reacting substances of anaphylaxis (SRS-A), caused dose-dependent contractions of rabbit coronary arteries in concentrations of 10?9 to 10?7 M and 10?10 to 10?7 M, respectively. The potency of LTC4 and LTD4, when compared with the concentration that elicits half of the contraction induced by 25 mM KCl, was 17 and 76 times, respectively, greater than that of histamine. In contrast, other blood vessels from rabbits were either unresponsive (renal artery and vein, mesenteric artery and thoracic aorta) or only weakly responsive (pulmonary artery and vein and portal vein) to both leukotrienes even at 10?7 M. The LTD4-induced coronary contraction was inhibited by FPL 55712 (10?7 and 10?6 M), a selective SRS-A inhibitor, in a dose-dependent manner, but not by diphenhydramine (10?7 M), a histamine H1-receptor blocker or by indomethacin (10?5 M), a prostaglandin synthetase inhibitor, suggesting that LTD4 has a direct effect on the coronary arteries. These results indicate that the leukotrienes may act as potent, selective coronary vasoconstrictors and that SRS-A responsive receptors exist in the rabbit coronary artery.  相似文献

The prawn Macrobrachium sintangense is likely to be subjected to occasional exposure to combined metal and saline stressors in its natural environment. This research evaluated the acute toxicity (96?h LC50) of cadmium (Cd) on the prawn M. sintangense, with respect to the osmoregulatory capacity (OC) of prawns and to document histological changes in the gills after exposure to sublethal Cd concentrations at different salinities. The 96?h LC50 of Cd to M. sintangense decreased with increasing salinity. The 96?h LC50 values were 89.12 (72.53–109.50), 681.26 (554.20–837.46) and 825.37 (676.99–1006.27) μg CdL?1 at 0, 10 and 20 ppt, respectively. The OC of prawns exposed to 30?μg?CdL?1 at 0 ppt and to 300?μg?CdL?1 at10 ppt decreased significantly compared with that of control prawns exposed to 0 and 10 ppt respectively. Swelling, hyperplasia and necrosis of gill lamellae resulting in the loss of marginal canals were observed in the gills of prawns exposed to 30?μg?CdL?1 at 0 ppt and to 300?μg?CdL?1 at 10 ppt for 7?days.  相似文献

The effects on platelet aggregation of α,β-methylene-adenosine-5′-diphosphate (Ado-PCP) have been investigated. Using human citrated platelet-rich plasma it has been shown that: (i) at concentrations of 10?3 M or higher Ado-PCP is able to induce platelet aggregation; (ii) the rate of Ado-PCP-induced aggregation increases on raising the pH of platelet-rich plasma above the pKa for the secondary phosphonyl dissociation of Ado-PCP; (iii) at concentrations from 1 · 10?4 to 5 · 10?4 M Ado-PCP does not cause platelet aggregation itself, but it inhibits ADP-induced aggregation. This inhibition is also observed in washed platelet suspensions. The data suggest that Ado-PCP acts at the same site on the platelet membrane as does ADP and that ADP to AMP transformation is not a prerequisite for the process of aggregation. The observed effect of pH on the rate of Ado-PCP induced aggregation suggests that the ionization state of a nucleotide terminal acid group is important in the process of aggregation.  相似文献
