We present a method for predicting the wall stress in a class of cerebral aneurysms. The method hinges on an inverse formulation of the elastostatic equilibrium problem; it takes as the input a deformed configuration and the corresponding pressure, and predicts the wall stress in the given deformed state. For a membrane structure, the inverse formulation possesses a remarkable feature, that is, it can practically determine the wall tension without accurate knowledge of the wall elastic properties. In this paper, we present a finite element formulation for the inverse membrane problem and perform material sensitivity studies on idealized lesions and an image-based cerebral aneurysm model.  相似文献

Wall shear stress estimates from laminar boundary layer theory were found to agree fairly well with the magnitude of shear stress levels along coronary artery constrictions obtained from solutions of the Navier Stokes equations for both steady and pulsatile flow. The relatively simple method can be used for in vivo estimates of wall shear stress in constrictions by using a vessel shape function determined from a coronary angiogram, along with a knowledge of the flow rate.  相似文献

Complex blood flow in large arteries creates rich wall shear stress (WSS) vectorial features. WSS acts as a link between blood flow dynamics and the biology of various cardiovascular diseases. WSS has been of great interest in a wide range of studies and has been the most popular measure to correlate blood flow to cardiovascular disease. Recent studies have emphasized different vectorial features of WSS. However, fixed points in the WSS vector field have not received much attention. A WSS fixed point is a point on the vessel wall where the WSS vector vanishes. In this article, WSS fixed points are classified and the aspects by which they could influence cardiovascular disease are reviewed. First, the connection between WSS fixed points and the flow topology away from the vessel wall is discussed. Second, the potential role of time-averaged WSS fixed points in biochemical mass transport is demonstrated using the recent concept of Lagrangian WSS structures. Finally, simple measures are proposed to quantify the exposure of the endothelial cells to WSS fixed points. Examples from various arterial flow applications are demonstrated.  相似文献

Wall shear stress in normal left coronary artery tree   总被引:1,自引:0,他引:1
Despite the fact that the role of wall shear stress (WSS) as a local mechanical factor in atherogenesis is well established, its distribution over the entire normal human left coronary artery (LCA) tree has not yet been studied. A three-dimensional computer generated model of the epicardial LCA tree, based on averaged human data set extracted from angiographies, was adopted for finite-element analysis of the Navier-Stokes flow equations treating blood as non-Newtonian fluid. The LCA tree includes the left main coronary artery (LMCA), the left anterior descending (LAD), the left circumflex artery (LCxA) and their major branches. In proximal LCA tree regions where atherosclerosis frequently occurs, low WSS appears. Low WSS regions occur at bifurcations in regions opposite the flow dividers, which are anatomic sites predisposed for atherosclerotic development. On the LMCA bifurcation, at regions opposite to the flow divider, dominant low WSS values occur ranging from 0.75 to 2.25 N/m2. High WSS values are encountered at all flow dividers. This work determines, probably for the first time, the topography of the WSS in the entire normal human LCA epicardial tree. It is also the first work determining the spatial WSS differentiation between proximal and distal normal human LCA parts. The haemodynamic analysis of the entire epicardial LCA tree further verifies the implications of the WSS in atherosclerosis mechanisms.  相似文献

Stenotic artery hemodynamics are often characertised by metrics including oscillatory shear index (OSI) and residence time (RT). This analysis was conducted to clarify the link between the near-wall flow behaviour and these resultant flow metrics. A computational simulation was conducted of a stenosed femoral artery, with an idealised representative geometry and a physiologically realistic inlet profile. The overall flow behaviour was characterised through consideration of the axial flow, which was non-dimensionalised against mean flow velocity. The OSI and RT metrics, which are a useful indicator of likely atherosclerotic sites, were explained through a discussion of the WSS values at different time points, the velocity behaviour and velocity profiles, with a particular focus on the near-wall behaviour which influences wall shear stress and the transient evolution of the wall shear stress. While, the stenosis throat experiences high values of wall shear stress, the smooth flow through this contracted region results in low variation in wall shear stress vectors and limited opportunity for any particle stasis. However, regions were noted distal and proximal (though to a lesser extent), where the change in recirculation zones over the cycle created highly elevated regions of both OSI and RT.  相似文献

BACKGROUND: Static deformation analysis and estimation of wall stress distribution of patient-specific cerebral aneurysms can provide useful insights into the disease process and rupture. METHOD OF APPROACH: The three-dimensional geometry of saccular cerebral aneurysms from 27 patients (18 unruptured and nine ruptured) was reconstructed based on computer tomography angiography images. The aneurysm wall tissue was modeled using a nonlinear, anisotropic, hyperelastic material model (Fung-type) which was incorporated in a user subroutine in ABAQUS. Effective material fiber orientations were assumed to align with principal surface curvatures. Static deformation of the aneurysm models were simulated assuming uniform wall thickness and internal pressure load of 100 mm Hg. RESULTS: The numerical analysis technique was validated by quantitative comparisons to results in the literature. For the patient-specific models, in-plane stresses in the aneurysm wall along both the stiff and weak fiber directions showed significant regional variations with the former being higher. The spatial maximum of stress ranged from as low as 0.30 MPa in a small aneurysm to as high as 1.06 MPa in a giant aneurysm. The patterns of distribution of stress, strain, and surface curvature were found to be similar. Sensitivity analyses showed that the computed stress is mesh independent and not very sensitive to reasonable perturbations in model parameters, and the curvature-based criteria for fiber orientations tend to minimize the total elastic strain energy in the aneurysms wall. Within this small study population, there were no statistically significant differences in the spatial means and maximums of stress and strain values between the ruptured and unruptured groups. However, the ratios between the stress components in the stiff and weak fiber directions were significantly higher in the ruptured group than those in the unruptured group. CONCLUSIONS: A methodology for nonlinear, anisotropic static deformation analysis of geometrically realistic aneurysms was developed, which can be used for a more accurate estimation of the stresses and strains than previous methods and to facilitate prospective studies on the role of stress in aneurysm rupture.  相似文献

High levels of wall shear stress on the surface of valvular cusps can cause mechanical damage to the blood cells and the cusp surfaces. The shear stresses are also responsible for mechanical failure of prosthetic heart valves. Quatitative measurements of wall shear stress in the vicinity of the leaflets are thus essential for diagnosis of suspected complications and provide important information for the design and fabrication of bioprosthetic heart valves. For this purpose we measured the velocity distribution along the inside wall of the cusps of a tri-leaflet heart valve with a two colour laser Doppler anemometer system. The wall shear stresses on the cusp surface were computed and found to range from 80 to 120 N/m2 during the ejection phase. Wall shear stresses of up to 180 N/m2 were measured in loci of cusp flexure and the accelerated boundary layer. The results of this study show a correlation between the high shear stress loci and the clinically (animal) observed regions of cusp calcification.  相似文献

Wall shear stress distribution in the carotid siphon, which is a multiple curved segment of the internal carotid artery, is investigated numerically under physiological flow conditions. The computer simulation of flow through the model segment is based on the time-dependent, three-dimensional Navier-Stokes equations, solved numerically with a finite element method. The study shows the behavior of the wall shear stress-vector field and identifies the zones of high and low wall shear stress values during the cardiac cycle.  相似文献

The strength and direction of blood flow into and within a cerebral aneurysm are important issues in developing effective interventional strategies to stabilize the aneurysm. We tested the hypothesis that there are significant major hemodynamic features that are common to many aneurysm flows of the type studied here. This was investigated by performing computational fluid dynamic simulations of flow near 7 cerebral aneurysms using geometrical data obtained from clinical CT scans. Our numerical simulations of flow across the ostium plane of an aneurysm show that in many cases there is relatively stable flow structure that is maintained over the phase of the pulsatile flow cycle. The two main features of this flow are (1) quasi-permanent regions of flow influx and efflux across the ostium plane exist, separated by a “virtual boundary”, and (2) a helical vortex flow pattern within the aneurismal sac with swirl in two orthogonal cross-sectional planes. These numerical observations are consistent with in vitro experimental data from ultrasound color-Doppler velocimetry and other numerical and experimental studies. The observed flow patterns are found to occur in different types of aneurysms (bifurcation and sidewall), and can persist even after flow parameters are perturbed beyond the normal range of physiological flow conditions. These results suggest that in many cases, major aspects of the behavior of aneurismal hemodynamics for important classes of aneurysms can be learned from an analysis of steady, non-pulsatile flow, which is simpler and faster to simulate than time-dependent, pulsatile flow. An understanding of this fluid dynamical behavior may also prove useful in the design of stents, coils, and various other endovascular flow diverting devices.  相似文献

The effect of luminal shear stress was studied in cerebral arteries and arterioles. Middle cerebral arteries (MCA) and penetrating arterioles (PA) were isolated from male Long-Evans rats, mounted in a tissue bath, and pressurized. After the development of spontaneous tone, inside diameters were 186 +/- 5 microm (n = 28) for MCA and 65 +/- 3 microm (n = 37) for PA. MCA and PA constricted approximately 20% with increasing flow. Flow-induced constriction persisted in MCA and PA after removal of the endothelium. After removal of the endothelium, the luminal application of a polypeptide containing the Arg-Gly-Asp amino acid sequence (inhibitor of integrin attachment) abolished the flow-induced constriction. Similarly, an antibody specific for the beta(3)-chain of the integrin complex significantly inhibited the flow-induced constriction. The shear stress-induced constriction was accompanied by an increase in vascular smooth muscle Ca(2+). For example, a shear stress of 20 dyn/cm(2) constricted MCA 8% (n = 5) and increased Ca(2+) from 209 +/- 17 to 262 +/- 29 nM (n = 5). We conclude that isolated cerebral arteries and arterioles from the rat constrict to increased shear stress. Because the endothelium is not necessary for the response, the shear forces must be transmitted across the endothelium, presumably by the cytoskeletal matrix, to elicit constriction. Integrins containing the beta(3)-chain are involved with the shear stress-induced constrictions.  相似文献

Numerical predictions of blood flow patterns and hemodynamic stresses in Abdominal Aortic Aneurysms (AAAs) are performed in a two-aneurysm, axisymmetric, rigid wall model using the spectral element method. Homogeneous, Newtonian blood flow is simulated under steady conditions for the range of Reynolds numbers 10 < or =Re < or =2265. Flow hemodynamics are quantified by calculating the distributions of wall pressure (p(w)), wall shear stress (tau(w)), Wall Shear Stress Gradient (WSSG). A correlation between maximum values of hemodynamic stresses and Reynolds number is established, and the spatial distribution of WSSG is considered as a hemodynamic force that may cause damage to the arterial wall at an intermediate stage of AAA growth. The temporal distribution of hemodynamic stresses in pulsatile flow and their physical implications in AAA rupture are discussed in Part II of this paper.  相似文献

In continuing the investigation of AAA hemodynamics, unsteady flow-induced stresses are presented for pulsatile blood flow through the double-aneurysm model described in Part I. Physiologically realistic aortic blood flow is simulated under pulsatile conditions for the range of time-average Reynolds numbers 50< or =Re(m) < or =300. Hemodynamic disturbance is evaluated for a modified set of indicator functions which include wall pressure (p(w)), wall shear stress (tau(w)), Wall Shear Stress Gradient (WSSG), time-average wall shear stress (tau(w)*), and time-average Wall Shear Stress Gradient WSSG*. At peak flow, the highest shear stress and WSSG levels are obtained at the distal end of both aneurysms, in a pattern similar to that of steady flow. The maximum values of wall shear stresses and wall shear stress gradients are evaluated as a function of the time-average Reynolds number resulting in a fourth order polynomial correlation. A comparison between numerical predictions for steady and pulsatile flow is presented, illustrating the importance of considering time-dependent flow for the evaluation of hemodynamic indicators.  相似文献

Thrombosis is a common complication following the surgical implantation of blood contacting artificial organs. Platelet transport, which is an important process of thrombosis and strongly modulated by flow dynamics, has not been investigated under the shear stress level associated with these devices, which may range from tens to several hundred Pascal. The current research investigated platelet transport within blood under supra-physiological shear stress conditions through a micro flow visualization approach. Images of platelet-sized fluorescent particles in the blood flow were recorded within microchannels (2 cm x 100 microm x 100 microm). The results successfully demonstrated the occurrence of platelet-sized particle margination under shear stresses up to 193 Pa, revealing a platelet near-wall excess up to 8.7 near the wall (within 15 microm) at the highest shear stress. The concentration of red blood cells was found to influence the stream-wise development of platelet margination which was clearly observed in the 20% Ht sample but not the 40% Ht sample. Shear stress had a less dramatic effect on the margination phenomenon than did hematocrit. The results imply that cell-cell collision is an important factor for platelet transport under supra-physiologic shear stress conditions. It is anticipated that these results will contribute to the future design and optimization of artificial organs.  相似文献

The shear stress at the wall has been of interest as one of the possible fluid dynamic factors that may be damaging in the region of prosthetic valves. The purpose of this study was to measure the axial wall shear stresses in the region of a 29 mm tissue annulus diameter porcine stent mounted prosthetic aortic valve (Hancock, Model 242). Studies were performed in an in vitro pulse duplicating system. The axial wall shear stress was calculated from velocities obtained near the wall with a laser Doppler anemometer. The largest axial wall shear stress was 29 dyn cm-2 and it occurred at the highest stroke volume used (80 ml). At a stroke volume of 50 ml, the largest axial wall shear stress was 17 dyn cm-2 and at a stroke volume of 35 ml, it was 15 dyn cm-2. Stresses of these magnitudes are far below those reported to be damaging to the endothelial surface. These stresses may be high enough, however, to affect platelet function.  相似文献

An experimental investigation of the wall shear stress distribution downstream of a backward-facing step is carried out. The wall shear stress distribution was determined by measuring the deformation of a gel layer, attached to the wall downstream of the step. Speckle pattern interferometry was applied to measure the deformation of the gel layer. The measured deformation, combined with the properties of the gel layer, served as an input for a finite element solid mechanics computation to determine the stress distribution in the gel layer. The wall shear stress, required to generate the measured deformation of the gel layer, was determined from these computations. A Newtonian buffer solution and a non-Newtonian red blood cell suspension were used as measuring fluids. The deformation of the gel layer was determined for a Newtonian buffer solution to evaluate the method and to obtain the properties of the gel layer. Subsequently, the wall shear stress distribution for the non-Newtonian red blood cell suspension was determined for three different flow rates. The inelastic non-Newtonian Carreau-Yasuda model served as constitutive model for the red blood cell suspension. Using this model, the velocity and wall shear stress distribution were computed by means of a finite element fluid mechanics computation. From the comparison between the numerical and the experimental results, it can be concluded that wall shear stresses, induced by the red blood cell suspension, can be modeled accurately by employing a Carreau-Yasuda model.  相似文献

Hepatitis C viral (HCV) RNA includes an internal ribosome entry segment (IRES) that extends some 30 nt into the coding region and promotes internal initiation of translation at the authentic initiation codon at nt 342. The 5'-boundary of this IRES was mapped by in vitro translation and transfection assays and was found to lie between nt 42 and 71. Within these IRES boundaries there are, in most HCV strains, three AUG triplets upstream of the authentic initiation site. Although the first, 5'-proximal, of these is absolutely conserved, a mutational analysis showed that it is not a functional initiation codon. In particular, the G residue could be substituted provided compensatory mutations were made to maintain base pairing. The other two upstream AUGs are not absolutely conserved, and mutation of the third (5'-distal) had little effect on IRES activity. When an additional AUG codon was introduced by single-site mutation just upstream of the authentic initiation codon, it was found to be used when most of the IRES had been deleted to generate an RNA translated by the scanning ribosome mechanism, but was not used in the background of the full-length IRES when internal initiation is operative. These results argue that the IRES promotes direct ribosome entry immediately at, or indeed very close to, the authentic initiation codon, and that the upstream AUGs do not serve as ribosome entry sites.  相似文献

In this paper, we introduce a method to construct a Reduced-Order Model (ROM) to study the physiological flow and the Wall Shear Stress (WSS) conditions in Abdominal Aortic Aneurysms (AAA). We start the process by running a training case using Computational Fluid Dynamics (CFD) simulations with time-varying flow parameters, such that these parameters cover the range of parameters that we would like to consider in our ROM. We use the inflow angle as the variable parameter in the current study. Then we use the snapshot Proper Orthogonal Decomposition (POD) to construct the reduced-order bases, which are subsequently enhanced using a QR-factorization technique to satisfy the relevant fluid boundary conditions. The resulting ROM enables us to study the flow pattern and the WSS distribution over a range of system parameters computationally very efficiently. We have used this method to show how the WSS varies significantly for an AAA with a simplified geometry, over a range of inflow angles usually considered mild in clinical terms. We have validated the ROM results with CFD results. This approach enables comprehensive analysis of the model system across a range of inflow angles and frequencies without the need to re-compute the simulation for small changes.
