The hyphomycetous fungus Aphanocladiutn album can grow over and around uredia of the rusts Puccinia coronata, P. hordei, P. graminis f.sp. avenae and P. recondita f.sp. triticina when host plants are kept under very humid conditions, but not on such plants not infected with rusts; uredia are adversely affected and telia develop in their vicinity. Plants inoculated with these rusts and with five isolates of A. album (one from a dead insect) showed: (1) much earlier development of telia on detached and non-detached rusted leaves inoculated with A. album than on corresponding leaves not thus inoculated; (2) telial induction by A. album in some isolates of rust species which hitherto had rarely or never produced telia; (3) precocious telial formation, in comparison with controls, when A. album spores were sprayed on leaves as much as 3 days before and 9 days after rust inoculation, and occasionally after uredia had already matured. As affected leaves remained green until the whole leaf became moribund, senescence is apparently not the factor inducing telia formation. The normal-appearing teliospores of some isolates were induced to germinate, whereas others did not. Rhamnus palaestina inoculated with basidiospores of one isolate of A. album-treated P. coronata f.sp. avenae produced pycnia and fertile aecia. The importance of A. album as a working tool in rust research and as a possible means for biological control of rust epiphytotics is discussed.  相似文献

Lemna perpusilla 6746 is induced to flower by a single longdark period, but the floral buds once formed disappear afterseveral days under 5000 lux/25?C. Such regression of floralbuds is prevented by lowering the light intensity or temperature,but if the light intensity and/or temperature are lowered beyondcritical levels, new floral buds form. If the cultures are subjectedto 100 lux/20?C, neither regression nor new formation of floralbuds occurs. Under such conditions, the number of floral frondsreaches maximum about 6 days after the inductive dark periodand reamins unchanged for at least 10 days, while the percentageof floral fronds rapidly decreases thereafter, owing to thedilution by newly developed vegetative fronds. When the cultures are subjected to various lengths of a singledark period (25?C) followed by 100 lux/20?C, flowering responsesrepresented by the number of floral fronds per flask show rhythmicfluctuation with a cycle length of about 24 hr. The present study evaluated the effects of exposure to light intensity in the morning on dim light melatonin onset (DLMO). The tested light intensities were 750 lux, 150 lux, 3000 lux, 6000 lux and 12,000 lux (horizontal illuminance at cornea), using commercial 5000 K fluorescent lamps. Eleven healthy males aged 21-31 participated in 2-day experiments for each light condition. On the first experimental day (day 1), subjects were exposed to dim light (<30 lux) for 3 h in the morning (09:00-12:00). On the same day, saliva samples were taken in dim light (<30 lux) every 30 min from 21:00 to 01:00 to determine the DLMO phase. The subjects were allowed to sleep from 01:00 to 08:00. On the second experimental day (day 2), the subjects were exposed to experimental light conditions for 3 h in the morning. The experimental schedule after light exposure was the same as on day 1. On comparing day 2 with day 1, significant phase advances of DLMO were obtained at 3000 lux, 6000 lux and 12,000 lux. These findings indicate that exposure to a necessary intensity from an ordinary light source, such as a fluorescent lamp, in the morning within one day affects melatonin secretion.  相似文献

油杉叶锈寄生在油杉叶上,作者研究了此菌的模式标本、同引模式标本和后来在模式地区采得的标本;并和柱锈属、金锈属、鞘锈属、栅锈属等11属锈菌作了比较研究。证明这种锈菌冬孢子堆为带状或扁平柱状,无细胞组织垫座,无被;冬孢子可互相撕离而不破裂,孢壁薄而无色,无柄。这些性状说明它不同于柱锈而近于金锈属。因此作者将其转归金锈属,改定为Chrysomyxa keteleeriae(Tai)Wang et Peterson新组合,(基础异名Cronartium keteleeriae Tai)。这种锈菌与其它生在云杉、铁杉叶上的4种短生金锈属锈菌一样,冬孢子堆都生在针叶上,窄带状到扁平柱状或圆形有短柄,鲜桔色或桔红色。虽然油杉叶锈冬孢子堆高达 6mm,但这只是同一属内的种间差别,也象Coleosporium crowellii Cumm。与同属内其它种的区别一样,属于种间的差别。云南粗杉金锈的冬孢子可以从10—20细胞组成的有柄孢子分离产生,正如鞘柄锈冬孢子的互相分离产生一样。冷杉金锈,特别是变形金锈的冬孢子的产生方式也是这样。生在同种油杉寄主枝上的Peridermium kunmingense Jen和油杉叶锈有无关系,需在接种试验后才能肯定。从寄主与寄主菌共同进化关系看,这种锈菌的寄主——油杉在分类系统上与金锈属寄主——云杉、铁杉等属近缘,传统分类学家多认为它同冷杉、银杉、黄杉等属同归冷杉亚科。但也有学者认为油杉有许多原始性状,与冷杉亚科无关。作者在油杉上发现金锈可能支持了传统植物分类系统。  相似文献

When mice infected with the Venezuelan equine encephalomyelitis (VEE) virus were exposed to 2500 lux with a 12 h light: 12 h dark photoperiod, the serum levels of melatonin (MLT) remained constantly elevated. In mice exposed to 400 lux low levels of serum MLT were detected during the day and high levels during the night. An increase in the survival rate of the infected mice from 6 to 13 days after virus inoculation was also observed. The significant increment in the concentration of serum MLT produced by the high intensity light could be responsible for the longer survival rate of mice infected with the VEE virus.  相似文献

Male guinea-pigs were inoculated by the testicular route with a suspension of chlamydo-arthrospores of the filamentous phase ofC. immitis. The following histopathological changes were observed: voluminous pyocytic foci as well as granulomatous mononuclear reaction was initially observed and granuloma with multinucleated giant cells were seen 20 days after the inoculation. The following changes of the microscopic aspect of the parasite were correlatively registered: the so-called primary infection type of sporangia appear in great number three days after the inoculation. This type of sporangium is characterized by its great diameter up to 98 µ, peripheral endospore formation leaving a large central vacuole which frequently contain residual protoplasm. The endospores are thin walled, polyhedral and get free through an ostiole. Sporangia completely filled with globose endospores (the cystic type of sporangia) with radiate acidophilic formations on the peridial wall were observed in the testicles of the guinea-pigs killed 6 days after the inoculation. Reduction in number and in the size of the parasite were seen after the 20th. day of the inoculation. Radiate formation of the cell wall of the parasite appeared simultaneously with precipitin antibodies and would be the expression of antigen-antibody reaction. All the intermediate types of sporangia between the primary infection type and the cystic type were observed after the 10th. day of the inoculation.  相似文献

Cowpeas, inoculated with one of five effective strains of Rhizobium isolated from African soils, were grown at root temperatures of 30 oC continuously or at 36, 38, 40, 42 and 44 oC for 5 h/day and returned to glasshouse ambient for the intermediate period. Growth was best at 30 and 36 oC; above 40 oC growth was poor and no nodules formed. At 40 oC two strains failed to nodulate. Symbiotic performance was not dependent only on nodule production as nodule efficiency varied inversely with temperature. The number of nodules formed by strain R5000 after exposure of inoculated seed or seedlings to 40,42 or 44 oC for 5 h/day on each of 3 or 6 days depended on the age of plant and the duration of exposure to stress. When exposed to 42 or 44 oC during the first 3 days after sowing nodulation was reduced from 18 to 1–3 nodules/plant even after a further 40 days growth at ambient (30 day, 20 oC night). Nodulation was unaffected when 10–15-day-old seedlings were exposed to the same conditions. Numbers of strain R5000 on seed declined rapidly following three daily exposures of 5 h at 39 and 42 oC; at 45 oC less than three bacteria survived on each seed. Other rhizobia of the cowpea group varied greatly in their toleration of high temperatures, some survived well at 45 oC whereas others behaved like R5000.  相似文献

We did this experiment to clarify the mechanism of granuloma formation and the killing functions of granuloma in nude mice against Blastomyces dermatitidis and Paracoccidioides brasiliensis infections. B. dermatitidis A-295 and P. brasiliensis B-1183 were the cultures used. Congenitally athymic nude (nu/ nu) mice and their heterozygous (nu/ +) littermates of BALB/ c background were the test animals. From culture A-295, 0.1% and 1% cell suspensions (wet weight) were prepared and from culture B-1183 0.2% and 2% cells suspensions were prepared. Ten nu/ + and 10 nu/ nu mice were allotted to each of four cell suspensions. For experimental blastomycosis each mouse was inoculated intravenously with 0.2 ml of the cell suspension of A-295 and for experimental paracoccidioidomycosis, with 0.15 ml of the cell suspension of B-1183. Two mice from each of the four groups were killed at 5, 8, 12, 18 and 25 days after inoculation, and histopathologic sections, stained with H&E or by PAS, were prepared from various internal organs.In the nu/ nu mice inoculated with B. dermatitidis A-295 granuloma was formed in the brain tissue after the 12th day. However, mononuclear cells, which formed the granuloma, did not kill the fungal cells, and the fungal cells continued to multiply in the granuloma. On the other hand, in the heart, kidney and fat tissue, their histopathological findings after the 18th day were clumps of fungal cells with slight cell reactions. In these organs the exertion of cell-mediated immunity was necessary for granuloma formation against the fungal infection.In the nu/ nu mice infected with P. brasiliensis B-1183, granuloma appeared in the brain and kidney after the 18th day and fungal cells continued to multiply within the granuloma as well as in those inoculated with culture A-295.These results show that the exertion of cell-mediated immunity plays an important role as the defense mechanisms of hosts against these fungal infections. However, PMNs also play an important role in the mouse's defense mechanisms against these fungal infections.We assume that the defense mechanisms of immunocompetent mice against B. dermatitidis or P. brasiliensis infection consist chiefly of two steps: in the first step phagocytosis by PMNs occurs and in the second step cell-mediated immunity enters into play.  相似文献

Summary The development of the vascular system in the neurohypophysis of the rat shows three periods of structural differentiation. The first period lasts until the 17th fetal day. During this period the superficial and the border plexuses are formed, and rapid vascularization of the neural lobe anlage takes place. The end of this period is marked by the formation of the perivascular space. The second period lasts from the 18th fetal day until the 10th day after birth. Its initial phase is characterized by the appearance of the first endothelial pores. During this period the formation of the internal capillaries proceeds, together with the maturation of the previously formed vascular elements. The third period lasts from the 10th postnatal day until the end of the first month after birth. At the end of this period the vascular net of the neural lobe has attained its fully developed state. The results of this study, together with data from previous investigations, lead to the conclusion that at the 18th fetal day the neural lobe displays signs of functional activity, and that the maturation of the vascular network in the neural lobe occurs more rapidly than the maturation of its other structural components.Research fellow of the Alexander von Humboldt Foundation.Dedicated to Prof. Dr. V. The humoral influence of cells of hemopoietic organs of chicken embryos of different terms on the development of the colony and cluster formation of mononuclears of the bone marrow of mice was studied in joint cultivation in two-compartment cylindrical diffuse microchambers. The process of formation of colonies and clusters is inhibited by cells of the yolk sac on the 2nd-4th day of the development, by cells of the liver on the 8th-12th day, of the spleen on the 13th-18th day and of the bone marrow--on the 15th day. The yolk sac cells were found to have most considerable inhibiting influence on proliferation and differentiation of cells on the 2nd day of the development of chicken embryo. The yolk sac cells on the 6th day stimulate the formation of colonies and clusters. The yolk sac, beginning from the 4th day of the development, and the liver release humoral factors promoting the formation of erythroid colonies. The erythroid colonies are formed but when cultivated on the vascular membrane of the chicken embryo; the erythroid colonies are not formed when cultivated in the abdominal cavity of mice. Local erythropoietinoid factors are not synthetized by the spleen and bone marrow cells. A supposition is put forward that a combination of the local inhibiting and erythropoietic effects promotes the erythroid differentiation of cells.  相似文献

Red clover mottle virus isolated in Czechoslovakia was studied in relation to its reaction to varying temperature on primary French bean leaves (Phaseolus vulgaris L.) on which it forms local necrotic lesions. The plants were kept 24 or 48 h before, or 24 or 48 h after inoculation at the temperatures 23, 25, 27, 30, 33 and 36°C. After such exposures the French beans were kept at a constant temperature of 25°C. The lesions were counted at various intervals. In the experiment the optimal temperature for the maximum number of lesions seems to be 36°C 48 h before inoculation. The temperature above 25°C applied 24 h after inoculation seems to have a decreasing effect upon the number of lesions formed by RCMV on primary leaves of French beans and the lesions appeared several hours later, especially at 30, 33 and 36°C. The temperatures 27, 30 and 33°C applied 48 h after inoculation have a further decreasing effect on the number of lesions. The temperature of 36°C applied 48 h after inoculation has an inactivating effect upon RCMV inoculated on French bean leaves and no lesions appeared 5 days after inoculation.  相似文献

Pileus formation in Favolus arcularius is induced by light,but no photoinduction occurred in young epileate stipes. Thestipes usually had to attain a length of about 5 mm to be photosensitive.Synchronous pileus formation could be induced by exposure tolight using epileate stipes which had been preincubated in darknessfor 48 to 72 hr. The pileus primordium formed about 24 hr afterthe start of illumination, however, continuous illuminationwas not necessary to produce this effect. A dark period givenbetween 1 and 8 hr after the start of illumination did not retardpileus formation. The photoinduction of pileus formation involvedtwo light-requiring processes, one occurring during the firsthour (the first light process) and the other from the 8th tothe 24th hr (the second light process). The photoresponse inthe first light process was saturated with 5 lux of light, buta light intensity below 1 lux was essentially ineffective. Onthe other hand, the reaction in the second light process couldbe started by less than 2 lux, and was accelerated by increasingthe light intensities up to about 150 lux. Morphological changes occurring during post-infection development and the influence of temperature on the life cycle of Heterodera lespedezae are reported. Morphological development was similar to that of H. schachtii. Each post-infection stage had a distinct styler, and fed actively. Detailed observations were made of cuticle formation and markings, esophageal glands, and reproductive system. Certain developmental phases, such as matrix deposition and oviposition, appeared to be correlated with color changes of the adult female body. The effect of temperature on nematode development was observed in a phytotron at day/night temperatures of 18/14, 18/18, 22/18, 26/22, 26/26, and 30/26 C; the optimum was 26/26 C. More time was required to complete the life cycle at the three lower temperatures than at the three higher temperatures.  相似文献

在模拟胁迫条件下(施加 CuSO4 的持续胁迫或加热40 ℃的瞬时胁迫),以大麦叶粉短期分解过程代表土壤功能,采用室内培养试验研究了土壤食真菌线虫(Aphelenchus avenae)与微生物的相互作用对土壤生态功能稳定性(抗性和恢复力)的影响.结果表明:无论施加胁迫与否,食真菌线虫的活动都有促进土壤微生物活性的趋势,尤其是在施加铜胁迫后第8 天开始到培养期结束,接种食真菌线虫导致土壤基础呼吸显著增加(P<0.05),但加热胁迫后食真菌线虫对土壤基础呼吸的促进作用仅在第8天有显著差异(P<0.05),反映了食真菌线虫对土壤微生物活性的影响程度与胁迫类型有关.在两种胁迫条件下,接种食真菌线虫对土壤功能的抗性没有影响,但都能促进胁迫条件下土壤功能的恢复.培养后期,两种胁迫条件下接种食真菌线虫处理真菌生物量低于未接种线虫处理,表明胁迫条件下食真菌线虫对真菌的取食可能限制甚至抑制了真菌生长,导致真菌对细菌的竞争压力减少,从而使细菌获得更大的生长优势,间接促进了细菌的生长.  相似文献

Chlorophyll formation in seven day old etiolated mung bean leaves was inhibited by CMU. The inhibition was reversed by feeding sucrose, or by leaving the cotyledons attached to the leaves. Photosynthesis appeared to contribute substrates for further chloroplast development soon after its commencement. When sucrose was fed in the presence of CMU at a range of light intensities, there was a distinct light induced promotion of chlorophyll formation at light intensities of 500–2000 lux. Treatment of the leaves with salicyl-aldoxime, an inhibitor of cyclic photophosphorylation indicated that this process could play an important part in chloroplast development.  相似文献

This clinical methods comparison study describes the difference between light levels measured at the wrist (Actiwatch-L) and at the eye (Daysimeter) in a postoperative in-patient population. The mean difference between the two devices was less than 10 lux at light levels less than 5000 lux. Agreement between the devices was found to decrease as eye-level light exposure increased. Measurements at eye level of 5000 lux or more corresponded to a difference between the devices of greater than 100 lux. Agreement between the eye- and wrist-level light measurements also appears to be influenced by time of day. During the day, the measurement differences were on average 50 lux higher at eye level, whereas at night they were on average 50 lux lower. Although the wrist-level monitor was found to underestimate light exposure at higher light levels, it was well tolerated by participants in the clinical setting. In contrast, the eye-level monitor was cumbersome and uncomfortable for the patients to wear. This study provides light-exposure data on patients in real conditions in the clinical environment. The results show that wrist-level monitoring provides an adequate estimate of light exposure for in-hospital circadian studies.  相似文献

During cold acclimation of potato plantlets ( Solanum commersonii Dun, PI 458317), there are two transitory increases in free ABA content corresponding to a three-fold increase on the 2nd day and a five-fold increase on the 6th day (Ryu and Li 1993). During this period, plantlets increased in cold hardiness from −5°C (killing temperature, control grown at 22/18°C, day/night) to −10°C by the 7th day of exposure to 4/2°C (day/night). This increase in free ABA was not found when cycloheximide (CHI), an inhibitor of cytoplasmic protein synthesis, was added to the culture medium 6 h before exposure to low temperatures. Plantlets treated with CHI did not acclimate to cold, maintaining a hardiness level (−5°C) similar to that of the 22/18°C-grown plantlets. When the CHI-treated plantlets were exposed to low temperatures for 3 days, transferred to CHI-free culture medium and grown at low temperatures, the plantlets showed a transitory increase in free ABA 2 days later. This increase was followed by the development of cold hardiness (−8°C). Application of CHI to the culture medium after 3 days of cold acclimation, when the first ABA peak and a partial development of cold hardiness (−8°C) had occurred, blocked the second transitory increase in free ABA and resulted in no further development of cold hardiness. These results suggest that de novo synthesis of proteins is required for these transitory increases in free ABA during cold acclimation of potato plantlets.  相似文献

The indirect immunofluorescence antibody test was performed on serial blood samples from eight young New Zealand White rabbits with experimental encephalitozoonosis. The test showed seroconversion in six of the eight infected rabbits by the 8th day after inoculation and in all rabbits by the 15th day. Antibody titers reached a peak by about the 36th day after inoculation and remained significantly elevated until the termination of the experiment at 84 days after inoculation. None of four sham-inoculated rabbits showed an immunofluorescence response by the 60th day after inoculation. Immunofluorescence and intradermal test responses were compared before infection and at the 60th day after inoculation in a total of 32 experimentally infected rabbits. Both tests were equally effective (100%) in detecting infected animals. Six of eight (first group) and 22 of 24 (second group) experimentally infected rabbits were confirmed histologically to have lesions compatible with encephalitozoonosis. No cross reactions were observed between Encephalitozoon cuniculi and Toxoplasma gondii, Eimeria perforans, or Eimeria stiedai by intradermal test or immunofluorescence test.  相似文献

Haploid rice plantlets were induced from cultured ovaries in our previously reported experiment. The present paper is an embryological study on this subject. Young flowers of two japonica cultivars were excised and cultured just in the same manner as before. Liquid medium used for float culture was N6+3% sucrose+0.125 ppm MCPA. The inoculated materials were checked to be at late uninuclcate pollen stage which corresponded mainly to the uuinuelcate embryo sac stage, but as well as some 2- or 4-nucleate embryo sacs. Samples were fixed at 23 days intervals in acetomethanol (1:3), stained in toto with diluted Ehrlich's hematoxylin and sectioned by paraffin method for microscopical observation. 4 days after inoculation most of the embryo sacs developed up to 8-nucleate stage with polarized differentiation of the egg apparatus, central cell and antipodals. From 7th day on, proembryos of various sizes and shapes appeared in the micropylar region of some embryo sacs; some consisted of meristcmatic ceils, others were highly vacuo]ated. One-celled as well as linear multicellular suspensors atypical of in vivo zygote proembryos were observed, ttowever, it was uncertain whether the proembryos originated from the egg cell, the synergids, or the differentiating egg apparatus as a whole. Another peculiar event occured during culture was the formation of endosperm-like free nuclei from the unfertilized polar nuclei in some embryo sacs. Sometimes the free nuclei were numerous and showed a tendency of cell formation in localized areas. 12–15 days after inoculation, the proembryos developed into mieroseopieal ealli with globular or pearlike shape, wbieh continued enlarging to visible size with naked eyes at about 18–24 th day. Further growth eventually led the ealli protruding out the ovary wall beyond 32–35 the day. These observations indicate that the embryo sue, similarly as the pollen, can be induced to embryogenesis in vitro. This may open a new way to study the mechanism controlling gametophytie and sporophytie developmental pathways of embryo sac and provide means for large-scale production of "embryo sac plants" in future.  相似文献

中国小麦条锈菌转主寄主小檗的鉴定   总被引:4,自引:0,他引:4  
用萌发的小麦条锈菌冬孢子接种采自陕西省境内的陕西小檗、少齿小檗和长穗小檗,3种小檗均产生了性孢子器和锈孢子器。用人工接种小麦条锈菌冬孢子在陕西小檗上产生的锈孢子器接种小麦铭贤169产生了典型的条锈菌夏孢子堆症状。特异性PCR和DNA序列分析表明,人工接种产生于小檗上的锈孢子、接种锈孢子于小麦上产生的夏孢子堆与小麦条锈菌DNA的ITS区序列完全一致。更为重要的是,用采自田间受锈菌侵染的小檗叶片产生的锈孢子接种小麦铭贤169,经培养在小麦铭贤169叶片上产生了典型的条锈病症状。从而证实,在自然条件下,在中国,小檗不仅可作为小麦条锈菌的转主寄主,而且小麦条锈菌可在小檗上完成其有性繁殖过程。这一发现对进一步揭示我国小麦条锈菌高度的群体遗传多样性与毒性变异机理、完善小麦条锈病的防治策略具有十分重要的理论和实际意义。  相似文献
