Extra- and intracellular reactions of 280 neurons of the pars principalis of the medial geniculate body (MGB) and of 408 auditory cortical neurons in area AI to stimulation of the inferior brachium of the midbrain and geniculocortical fibers were studied in cats immobilized with D-tubocurarine. Single electrical stimulation of the inferior brachium was shown to evoke a long and complex neuronal response in MGB in the form of excitation of some and inhibition of other neurons. The initial component of this response lasted 13 msec. Excitation of 72% of neurons participating in the response took place during the first 3 msec after the beginning of stimulation. In the same period 84% of IPSP arose. The inferior brachium was shown to contain a certain number of descending fibers. Some of them are axons of MGB neurons. Many fibers of the inferior brachium reach the auditory cortex without synaptic relay in MGB. Of all cells of MGB excited by stimulation of the inferior brachium monosynaptically, 76% are thalamocortical relay neurons; the rest are interneurons. Of the relay neurons of MGB 90% are excited monosynaptically, the rest by impulses passing through two or three synaptic relays in MGB. During stimulation of the inferior brachium, responses consisting of EPSP-IPSP and primary IPSP are recorded in many neurons of MGB. About 20% of primary IPSP arise monosynaptically, evidently in response to stimulation of inhibitory fibers of the inferior brachium. Most IPSP arise disynaptically, with the participation of an inhibitory interneuron located at the entrance to MGB. Inhibition observed in this case is direct afferent in nature.A. A. In experiments on immobilized cats, intra- and extracellular response in tonic type neurons to tones of differing frequencies and intensities were investigated, as well as the organizational pattern of receptive fields in these units. Tonic type neurons were encountered at different cortical layers, but mostly (93% of the total) were located at a depth of 1.0–2.2 mm. Minimum thresholds required for response in these neurons were on average 7.7 dB below that found in neurons generating a phasic reaction in response to a tone. "Tonic" differed from "phasic" neurons in their inferior frequency-discriminative ability, with a Q10 value averaging 4.1±0.4 as against 9.1±0.7 in phasic neurons. Size of receptive fields in tonic neurons (as revealed by occurrence of spike response in these units) was 3.5 times that observed in phasic cells. Length of action potentials in the majority (80%) of tonic neurons was about one and a half times to twice that found in phasic units. Tonic neurons also displayed a high degree of sensitivity to changes in the duration and intensity of acoustic stimulation.A. A. 在30只氨基甲酸乙酯麻醉的SD大鼠上记录神经元单位放电,观察短纯音诱发的皮层AI区神经元ON-OFF反应的特性及电刺激杏仁外侧核(lateral amygdaloid nucleus,LA)对ON-OFF反应以及调谐曲线的影响。实验证实,AI区神经元ON-OFF反应的模式与纯音刺激的强度、频率及作用时程有关;刺激LA可以抑制ON-OFF反应的放电频数,使反应的阈值升高,或使反应放电构型发生变化;此外,刺激LA能使ON-OFF神经元的调谐曲线变窄,Q10数值增大。研究结果不仅表明ON-OFF神经元能对纯音刺激的时程、强度和频率等多种信息进行编码,而且还证明杏仁外侧核可以在皮层水平参与听觉信息的调制,削弱或衰减某些听觉信息,导致整个调谐曲线上移变窄,从而提高AI区ON-OFF神经元的频率选择性能,有利于检测外界嘈杂环境中特定的听觉信息。  相似文献

The responses of the cortical secondary auditory area (AII) to the non-acoustic stimuli (electrical stimulation of the skin in the vibrissae area and light flash) and their combination with acoustic stimulation (sound click or tone) were studied in experiments on cats anesthetized by kalipsol using extra- and intracellular recording. Of the total number of neurons, 69% of the units generating spike responses to the acoustic stimulation responded to the non-acoustic stimulation too. The responses to the modal-nonspecific stimulation, as a rule, were weak and variable; they were mostly represented by a tonic change in the neuronal discharge frequency. The nonspecific stimulation evoked primary excitatory and inhibitory postsynaptic potentials in 77% and 20% of the examined neurons, respectively. We found that synaptic effects of the nonspecific and specific stimulations interact with each other, ensuring considerable modulation of the latter (mostly a significant facilitation resulting from the EPSP summation and suppression of an inhibitory component of the response to acoustic stimulation). 用双声刺激和多管电极方法在 6只大棕蝠 (bigbrownbat,Eptesicusfuscus)的 98个神经元上研究了锐化 (sharpening)蝙蝠听皮层 (primaryauditorycortex ,AC)神经元频率调谐的柱特征。结果发现 ,电极直插在 1个电极通道内连续记录到多个神经元时 ,它们锐化频率调谐的抑制性调谐曲线或抑制区基本相似。电极与AC表面呈 45°斜向推入使其跨越多个功能柱时 ,可观察到锐化频率调谐的抑制区构成也随电极进入不同的功能柱而发生相应的改变。两种不同的电极插入方式均证明锐化AC神经元频率调谐的神经抑制呈柱状组构。这些神经元组合起来排列在同一听觉功能柱内 ,构成AC频率分析的基本功能组构单位“微频率处理器”。实验中还观察到多峰频率调谐曲线神经元 ,它们在声通讯和声定位中不同波谱区域的时间匹配中起作用。此外 ,也有理由认为多峰调谐神经元亦被用于作为复杂波谱信息的“高级调谐预处理器” ,从而极大地提高了神经元对频率分析的能力。为研究锐化频率调谐的神经抑制机制 ,用多管电极电泳γ -氨基丁酸 (γ aminobutyricacid ,GA BA)能a受体拮抗剂荷包牡丹碱 (bicuculline ,Bic)至所记录的神经元 ,发现能大部分或几乎全部取消抑制区 ,从而表明在正常情况下GABA能抑制参与构成锐化AC神经元频率调谐的抑制区 ,  相似文献

Responses of 98 auditory cortical neurons to electrical stimulation of the medial geniculate body (MGB) were recorded (45 extracellulary, 53 intracellularly) in experiments on cats immobilized with tubocurarine. Responses of the same neurons to clicks were recorded for comparison. Of the total number of neurons, 75 (76%) responded both to MGB stimulation and to clicks, and 23 (24%) to MGB stimulation only. The latent period of extracellularly recorded action potentials of auditory cortical neurons in response to clicks varied from 7 to 28 msec (late responses were disregarded), and that to MGB stimulation varied from 1.5 to 12.5 msec. For EPSPs these values were 8–13 and 1–4 msec respectively. The latent period of IPSPs arising in response to MGB stimulation varied from 2.2 to 6.5 msec; for 34% of neurons it did not exceed 3 msec. The difference between the latent periods of responses to clicks and to MGB stimulation varied for different neurons from 6 to 21 msec. Responses of 11% of neurons to MGB stimulation, recorded intracellularly, consisted of sub-threshold EPSPs, while responses of 23% of neurons began with an EPSP which was either followed by an action potential and subsequent IPSP or was at once cut off by an IPSP; 66% of neurons responded with primary IPSPs. Neurons responding to MGB stimulation by primary IPSPs are distributed irregularly in the depth of the cortex: there are very few in layers III and IV and many more at a depth of 1.6–2 mm. Conversely, excited neurons are predominant in layer III and IV, and they are few in number at a depth of 1.6–2 mm. It is concluded that the afferent volley reaching the auditory cortex induces excitation of some neurons therein and, at the same time, by the principle of reciprocity, induces inhibition of others. This afferent inhibition takes place with the participation of inhibitory interneurons, and in some cells the inhibition is recurrent. The existence of reciprocal relationships between neurons in different layers of the auditory cortex is postulated.A. A. Summary Response characteristics of 130 single neurons in the superior olivary nucleus of the northern leopard frog (Rana pipiens pipiens) were examined to determine their selectivity to various behaviorally relevant temporal parameters [rise-fall time, duration, and amplitude modulation (AM) rate of acoustic signals. Response functions were constructed with respect to each of these variables. Neurons with different temporal firing patterns such as tonic, phasic or phasic-burst firing patterns, participated in time domain analysis in specific manners. Phasic neurons manifested preferences for signals with short rise-fall times, thus possessing low-pass response functions with respect to this stimulus parameter; conversely, tonic and phasic-burst units were non-selective and possessed all-pass response functions. A distinction between temporal firing patterns was also observed for duration coding. Whereas phasic units showed no change in the mean spike count with a change in stimulus duration (i.e., all-pass duration response functions), tonic and phasic-burst units gave higher mean spike counts with an increase in stimulus duration (i.e., primary-like high-pass response functions). Phasic units manifested greater response selectivity for AM rate than did tonic or phasic-burst units, and many phasic units were tuned to a narrow range of modulation rates (i.e., band-pass). The results suggest that SON neurons play an important role in the processing of complex acoustic patterns; they perform extensive computations on AM rate as well as other temporal parameters of complex sounds. Electric reactions of neurons of the specific (VPM) and nonspecific (CM PF) thalamic nuclei caused by noxious and electroacupuncture stimulation were studied in acute experiments on cats. About 18% of the investigated neurons demonstrated coupled reactions to afferent stimulation. The coupled reaction to either noxious or electroacupuncture stimulation had inhibitory or activating character. A hypothesis on the existence of functional units responsible for the effects of electroacupuncture analgesia with participation of inhibitory, relay neurons and interneurons is made.  相似文献

Responses of 74 cerebellar cortical neurons to electrical stimulation of the vagus nerve and its gastric branches and also of the limb nerves were recorded extracellularly in experiments on rats anesthetized with chloralose and pentobarbital. Phasic and tonic unit responses were similar to all types of stimulation, but fewer neurons responded to stimulation of the vagus nerve than to stimulation of the limb nerves. Monosensory neurons responding only to visceral stimulation, were not found. Among cells giving a phasic response, some had a short and others a long latent period. The latent period of responses of all types was longer to vagal stimulation than to stimulation of the limb nerves. The maximal frequency of rhythmic responses to vagal stimulation was 8/sec, compared with 16/sec for stimulation of the limb nerves. Tonic responses of the neurons were excitatory, inhibitory, and mixed. Responses of 92 neurons of the reticular (R) and 105 neurons of the ventral anterior (VA) thalamic nuclei to stimulation of the ventrobasal complex (VB) and the lateral (GL) and medial (GM) geniculate bodies were investigated in cats immobilized with D-tobocurarine. Altogether 72.2% of R neurons and 76.2% of VA neurons responded to stimulation of VB whereas only 15.0% of R neurons and 27.1% of VA neurons responded to stimulation of GM and 10.2% of R neurons and 19.6% of VA neurons responded to stimulation of GL. The response of the R and VA neurons to stimulation of the relay nuclei as a rule was expressed as excitation. A primary inhibitory response was observed for only two R and three VA neurons. Two types of excitable neurons were distinguished: The first respond to afferent stimulation by a discharge consisting of 5–15 spikes with a frequency of 250–300/sec; the second respond by single action potentials. Neurons of the first type closely resemble inhibitory interneurons in the character of the response. Antidromic responses were recorded from 2.2% of R neurons and 7.8% of VA neurons during stimulation of the relay nuclei. Among the R and VA neurons there are some which respond to stimulation not only of one, but of two or even three relay nuclei. If stimulation of one relay nucleus is accompanied by a response of a R or VA neuron, preceding stimulation of another nucleus leads to inhibition of the response to the testing stimulus if the interval between conditioning and testing stimuli is less than 30–50 msec.A. The temporal relations between simultaneously recorded neurons of the nucleus ventralis lateralis (VL) of cat thalamus were studied. The interaction and the functional connections between individual VL neurons are described. This was achieved with an application of cross correlation techniques. The response patterns of different individual neurons to somatic sensory and photic stimuli were also analyzed. For the purpose of classifying neurons as thalamocortical relay cells (T-C) and non relay cells (N-C) which do not project to the motor sensory cortex antidromic cortical stimulation was used. This stimulation was also used as conditioning one when proceeded peripheral stimuli. To analyze the nonspecific specific interactions upon single neurons conditioning photic stimuli were applied. The results show that T-C neurons are antidromically excited from a wide cortical areas and that the functional interaction between T-C neurons is mediated by a shared input from common sources. It is further postulated that N-C cells interposed between relay neurons subserve the functions of gating units modifying the neuronal network of lateral ventral nucleus of the thalamus.  相似文献

Unit responses of the nuclei pontis (NP) and reticular pontine nuclei (RPN) to stimulation of the frontobasal cortex (proreal, orbital, and basal temporal regions) and of the dorsal hippocampus were studied in cats. Stimulation of the various cortical structures was found to induce phasic and (less frequently) tonic responses in neurons of NP and RPN. The main type of unit response in RPN was primary excitation, whereas in NP it was primary inhibition. The largest number of responding neurons in the pontine nuclei was observed to stimulation of the proreal gyrus. In the cerebro-cerebellar relay system neurons of the reticular tegmental nucleus and ventromedial portion of NP showed the highest ability to respond. In the oral and caudal reticular pontine nuclei the regions of predominant influence of cortical structures were located in zones of these nuclei where neurons with rostral and (to a lesser degree) caudal projections were situated.M. The characteristics of extra- and intracellular responses of neurons in the AI region were studied in experiments with unanesthetized cats. It was established that auditory cortex neurons with similar best frequencies showed different forms of responses to tones of the corresponding frequency. About 40% of the auditory cortex neurons generated on responses to tone presentation. On — off and off responses were found in 27% of the neurons. Cortical neurons (27%) in which stimulation or inhibition of impulse discharge persisted throughout tone action were assigned to the tonic type group of cells. Approximately 6% of neurons in the AI region did not respond to a tone. During intracellular recording about 85% of the neurons responded to the turning on and/or off of a tone by generating an action potential followed by an IPSI. In 96% of the cortical neurons studied the IPSPs were a constant component of the intracellular responses to a tone. It is concluded that the inhibition of the impulse activity of the given neurons is of primarily a postsynaptic origin. Neurons showing one or another form of response differ from one another in the relative intensity and time characteristics of excitatory and inhibitory processes interacting on their postsynaptic membranes. In neurons of the phasic type inhibitory processes are dominant over excitatory, while excitatory processes are predominant in neurons of the tonic type.A. A. The effect of electrodermal stimulation of the contralateral forelimb on responses arising in neurons of the parvocellular part of the medial geniculate body (MGB) to clicks was studied in cats anesthetized with thiopental and immobilized with myorelaxin (suxamethonium). Neurons whose responses to clicks were inhibited by electrodermal stimulation were located in zones of the parvocellular part of MGB adjacent to the posterior ventral nucleus and magnocellular part of MGB. Electrodermal stimulation had no effect on unit responses in more lateral zones of the parvocellular part. Intracellularly recorded responses of most neurons to clicks were of the EPSP-spike-IPSP or EPSP-IPSP type, whereas those to electrodermal stimulation were of the IPSP type only. Inhibition of responses under the influence of electrodermal stimulation could arise both in the presence and absence of an IPSP in the neuron. The mechanisms of the inhibitory effect of electrodermal stimulation on responses arising in neurons of the parvocellular part of MGB to stimulation by clicks are discussed.A. A. Experiments on unanesthetized, immobilized cats showed different effects of individual hypothalamic nuclei on spontaneous unit activity in the sensomotor cortex. Compared with the posterior hypothalamic nucleus (PHN) and its anteromedial region (AMH), in response to stimulation of the lateral hypothalamic nucleus (LHN) changes in spontaneous activity were more frequently found. The ratio between activation and inhibitory responses was 36 and 36% for AMH, 51 and 30% for LHN, and 47 and 28% for PHN. An approximately equal number of sensomotor neurons (27–34%) gave tonic responses. Phasic changes in spontaneous activity were observed more often in response to stimulation of LHN, less frequently to stimulation of AMH and PHN. Responses of "nonpyramidal" neurons to stimulation of AMH and LHN were identical. "Pyramidal" units were more sensitive to LHN stimulation. Variations in hypothalamic effects depending on the type of response of sensomotor neurons to sensory stimuli were detected. Cells with tonic responses were more susceptible to influences of LHN and AMH than cells which responded by phasic changes in spontaneous activity to sensory stimuli. Extra- and intracellular responses of neurons in the primary somatosensory cortex to repetitive mechanical stimulation of the vibrissae at different frequencies were studied in unanesthetized curarized adult cats. Unlike responses to electrical stimulation of the combined afferent input (the infraorbital nerve) spike discharges of neurons in response to vibrissal stimulation can reproduce rather higher frequencies of stimulation and their initial character changes more often in the course of the repetitive series. Most cortical neurons were characterized by limitation of the area of their peripheral receptive fields with an increase in the frequency of adequate repetitive stimulation. A group of cortical neurons was distinguished by its ability to respond to high-frequency stimulation and to generate burst discharges. Comparison of the frequency characteristics of spike responses of these cells and of inhibitory synaptic action in other cortical neurons led to the conclusion that this group of cells thus distinguished may be inhibitory cortical neurons. The role of interaction between excitatory and inhibitory processes arising in cortical neurons during repetitive stimulation of different areas of their receptive fields is discussed.A. A. Cortical inhibition plays an important role in shaping neuronal processing. The underlying synaptic mechanisms remain controversial. Here, in vivo whole-cell recordings from neurons in the rat primary auditory cortex revealed that the frequency tuning curve of inhibitory input was broader than that of excitatory input. This results in relatively stronger inhibition in frequency domains flanking the preferred frequencies of the cell and a significant sharpening of the frequency tuning of membrane responses. The less selective inhibition can be attributed to a broader bandwidth and lower threshold of spike tonal receptive field of fast-spike inhibitory neurons than nearby excitatory neurons, although both types of neurons receive similar ranges of excitatory input and are organized into the same tonotopic map. Thus, the balance between excitation and inhibition is only approximate, and intracortical inhibition with high sensitivity and low selectivity can laterally sharpen the frequency tuning of neurons, ensuring their highly selective representation.  相似文献

We investigated the representation of four typical guinea pig vocalizations in the auditory cortex (AI) in anesthetized guinea pigs with the aim to compare cortical data to the data already published for identical calls in subcortical structures - the inferior colliculus (IC) and medial geniculate body (MGB). Like the subcortical neurons also cortical neurons typically responded to many calls with a time-locked response to one or more temporal elements of the calls. The neuronal response patterns in the AI correlated well with the sound temporal envelope of chirp (an isolated short phrase), but correlated less well in the case of chutter and whistle (longer calls) or purr (a call with a fast repetition rate of phrases). Neuronal rate vs. characteristic frequency profiles provided only a coarse representation of the calls’ frequency spectra. A comparison between the activity in the AI and those of subcortical structures showed a different transformation of the neuronal response patterns from the IC to the AI for individual calls: i) while the temporal representation of chirp remained unchanged, the representations of whistle and chutter were transformed at the thalamic level and the response to purr at the cortical level; ii) for the wideband calls (whistle, chirp) the rate representation of the call spectra was preserved in the AI and MGB at the level present in the IC, while in the case of low-frequency calls (chutter, purr), the representation was less precise in the AI and MGB than in the IC; iii) the difference in the response strength to natural and time-reversed whistle was found to be smaller in the AI than in the IC or MGB.  相似文献

Orientation tuning of 148 primary visual cortical neurons was studied in acute experiments on unanesthetized, curarized cats by analysis of their spike responses to flashes in a receptive field of a bar of light of optimal size. Orientation tuning of 88 neurons (59%) was found to be bimodal: Besides the principal preferred orientation there was a second, making an angle with the first. The second tuning maximum in some cases (64%) was exhibited only with a change in stimulus intensity or background brightness. Analysis of orientation tuning by the time-slice method, i.e., on the basis of individual cuts of the spike trace, showed double tuning to be present in 69% of cases only at certain moments after the beginning of stimulation. The results of analysis of the model showed that the double orientation tuning effect may be the result of the specific configuration of the receptive field, the use of a stimulus longer than the receptive field, the presence of a series of alternating excitatory and inhibitory zones in the receptive field, and also of end inhibitory zones on the narrow ends of the field. The unequal change in zones of the receptive fields in time explains the appearance of double orientation tuning in individual fragments of the spike trace. The functional role of double, "cross-wise" tuning in some primary visual cortical neurons and their role in the detection of the features of visual patterns are discussed.  相似文献

Summary The responses of 230 single neurons in the inferior colliculus of the horseshoebat to single tones have been studied, emphasizing systematic analysis of the effective frequency bands, dynamic properties and the time course of responses. Distribution of the units' best excitatory frequencies (BEF) is: low frequency neurons 23% (BEF 3–65 kHz); FM-frequency neurons 25% (BEF 65–81 kHz, i.e., frequencies occurring in the FM-part of the bat's echo signal); filter neurons 45% (BEF 81–88 kHz, i.e., frequencies occurring in the stabilized CF-part of the bat's echo=reference frequency (RF)); high frequency neurons about 7% (BEF > 88 kHz). Tuning curves show conventional shapes (Fig. 1), apart from those of filter neurons, which are extremely narrow. Accordingly, Q10dB-values (BEF divided by the bandwidth of the tuning curve at 10 dB above threshold) are 80–450 in filter neurons (Fig. 2). Response patterns (Fig. 3) are similar to those of Nucleus cochlearis units (transient, sustained, negative and complex responders) with an increased percentage of complex responders up to 38% and a decreased number of transient responders.All types of spike-count functions are found (Fig. 4); nonmonotonic ones dominating. Maximal spike counts are not at the BEF but a few kHz below. Distinct upper thresholds, especially at the BEF of filter neurons (Fig. 5) lead to abrupt changes in activity by slightly shifting stimulus frequency or intensity.The hallmark of inferior colliculus neurons is inhibition, disclosed by distinct inhibitory areas enfolding and overlapping excitatory ones (Figs. 3 and 5). Duration of inhibition varies with stimulus frequency, but is largely independent of stimulus duration (Fig. 6), whereas rebound of inhibition depends on stimulus duration building up periodic rebound activities, if stimulus duration is lengthened. In addition, there are neurons responding only periodically, regardless of stimulus frequency and intensity (Fig. 7).
