Seven seasonally anovular Merino ewes, isolated from rams, were bled every 10 min for 12 h. Rams were introduced the following day, after a further 2 h of sampling every 10 min, and samples were taken for the following 10 h. Three days after the introduction of rams, the ewes underwent laparoscopy, and four had ovulated.Radioimmunoassay of plasma samples revealed pulsatile rhythms in the concentration of LH, with the frequency of pulses varying between ewes, from about once hourly to once every 10 h. Upon the introduction of rams, LH secretion increased within 10 min in six ewes, of which four ovulated within 3 days. There is apparently a relationship between the pattern of LH secretion after introducing the rams, and the ovulatory response that can be expected.  相似文献

Two closed lines of sheep selected for high and low prolificacy were established in 1969. Rams within each line were selected on the basis of their dams' lambing records. Foundation ewes were assigned at random to selected rams, and no ewe selection was practiced until replacement yearlings were selected for mating. Selection of the rams used to mate the foundation ewes resulted in more lambs born per 100 ewes exposed to the ram (P<.10) and more lambs born per 100 ewes lambing from the high line rams (P<.05).Seven rams from each line were then used to estimate fertilization rate and embryonic survival rate when mated to an unrelated group of ewes. There was no difference in ovulation rate or fertilization rate of ewes mated to low and high line rams. Percent eggs recovered per CL was 80 and 92 (P<.01) for low and high line rams, respectively. At 35 days 82 and 86% of the ewes were pregnant, and 84 and 93% of the CL were represented by embryos in the pregnant ewes mated to low and high lines, respectively. Estimated embryonic survival rates were 81 and 89% in ewes mated to low and high line rams, respectively.  相似文献

The mean testis diameter of 20- to 25-week-old Blackface, Finn and Merino rams was ranked in the same order as the ovulation rates of females of their breeds. The removal of one testis at 12 or 16 weeks of age resulted in hypertrophy of the remaining testis. The relative increase in testis growth following hemicastration was greatest in the Merino rams (72%), least in the Finns (42%) and intermediate in the Blackfaces (57%), so that it was inversely related to their breed ovulation rates. This hypertrophy was associated with increases in the concentration of plasma LH in all breed types. The results indicate that differences in testis growth rate are associated with differences in gonadotrophic stimulation rather than in intrinsic growth potential, and it is postulated that these may arise from breed differences in sensitivity to negative feed-back from the testes.  相似文献

The effect on wool growth of thyroidectomy with subsequent thyroxine replacement at subnormal and supranormal levels has been investigated in Merino rams fed a restricted basal diet. Thyroidectomy without thyroxine replacement caused a greater than 60% reduction in wool growth. Provision of 30% of normal plasma thyroxine concentrations was sufficient to return wool growth to normal. Similarly, complete replacement of plasma thyroxine levels gave normal wool growth while increasing thyroxine concentrations to three times normal increased wool growth to levels slightly above normal. These results provide a possible explanation for the inability of other workers to correlate seasonal thyroxine patterns and annual wool growth variations.  相似文献

Two experiments were performed on seasonally anovular Merino ewes to determine the intervals between time of introduction of rams, the preovulatory surge of LH and the first ovulation. Ovulation was determined by laparoscopy and LH was measured by solid phase radioimmunoassay. In Experiment 1 the interval between the introduction of rams and the beginning of the LH surge was 27 ± 4 h (mean ± S.E., range 6–52 h), and in Experiment 2 probit analysis shows that 50% of teased ewes ovulated within 41 h of being exposed to rams.  相似文献

Ovulation was induced by rams in 74 of 91 seasonally anovular Merino ewes. The resulting corpora lutea (CL) were observed by laparoscopy and were found to either persist normally (3874), or regress prematurely (3674). In 32 of the latter ewes premature regression of the CL was followed by a second ovulation within 6 days of the introduction of rams.  相似文献

The objective was to determine if transrectal ultrasonography for determination of pregnancy in restrained ewes increases embryonic/fetal death or loss of pregnancy. Ten flocks (N=873 ewes) bred in either the estrous or anestrous season were randomized, into control (C) or examined (E) groups within flock. Examined ewes were placed in a tilting squeeze chute and scanned by one of three operators for pregnancy by transrectal ultrasonography once between Days 25 and 100 post-breeding. Control ewes were not subjected to handling in the squeeze chute or pregnancy diagnosis. Two operators counted embryos in E ewes in six flocks. There were no differences between E and C ewes in percent ewes lambing or lambing rate on a per flock basis. Prolificacy per flock was greater in E ewes (P=0.05; 1.53 versus 1.60, C and E, respectively) than in C ewes. Eighty-five percent of exposed ewes lambed in the estrous season, while only 62% of ewes lambed in the anestrous season (P=0.05). Overall prolificacy did not differ with season (1.60 versus 1.53), and there was no season by treatment interaction for any variable tested. Losses averaged 0.02 per E ewe exposed in the estrous season and 0.20 in the anestrous season (P<0.05). Examined ewes in this study had similar pregnancy and lambing rates to C ewes in the same flocks and prolificacy was slightly greater. Based on these data, the combination of transrectal ultrasonography with restraint is safe for pregnancy diagnosis in ewes.  相似文献

Chromosomal analysis was carried out on 48 Day 2-7 embryos collected from superovulated Merino ewes. Three embryos had abnormal chromosome complements (1 X 1N, 1 X 1N/2N, 1 X 3N), yielding an incidence of 6.25% abnormal embryos. It is concluded that superovulation does not cause an increase in the incidence of chromosomal abnormalities in embryos of Merino sheep.  相似文献

Samples of cervical mucus were collected from young and mature ewes at different stages of oestrus. The migration rate of spermatozoa (time taken to travel 410 mm) was not different in samples of mucus from young and mature ewes. Spermatozoa were unable to penetrate the mucus in 7 27 samples, five of which were collected about 16 hours after onset of oestrus. Migration rate in samples collected between 10 and 16 hours after onset of oestrus was significantly slower than in samples collected in the first five hours of oestrus. The transport of spermatozoa in young and mature ewes was also studied by observing distribution of spermatozoa in the female genital tract 26 to 27 hours after hand-mating. There were no significant differences between young and mature ewes either in mean sperm count or in variation of mean sperm numbers in all sections of the tract.  相似文献

Serial blood sampling before and after exposing four anovular Corriedale ewes to a group of rams and estrous ewes during the non-breeding season revealed a pattern of LH secretion similar to that previously observed in Merinos. Mean LH values doubled (P<0.001) from 0.24+/-0.06 microgL(-1) (mean+/-s.e.m.) before to 0.55+/-0.05 microgL(-1) after 2h of visual, auditory, and odor exposure to rams and estrous ewes in an indoor facility. A non-significant (P<0.17) increase of LH pulses per hour was also observed (0.7+/-0.3 pulses per hour before compared with 1.3+/-0.3 during stimulation). All four ewes had recently formed corpora lutea by five days after stimulation. Results are consistent with the pattern of sudden increase and sustained release of LH observed in other sheep breeds, particularly the Merino.  相似文献

To determine whether the high ovulation rate of the Booroola Merino ewe could be explained by FSH metabolism we have tested the proposition that FSH may have a longer half-life in the plasma of Booroola Merino ewes than in control ewes. The half-life of plasma FSH was determined by removal of the pituitary gland, to abolish FSH secretion into the peripheral circulation, and monitoring by repeated blood sampling the subsequent decline in plasma FSH concentrations. The half-life of FSH was similar in Booroola (103 +/- 14 (s.e.m.) min, N = 8) and control (116 +/- 8 min, N = 9) ewes. However, when ewes that had been ovariectomized at least 6 months earlier were hypophysectomized, the half-life of FSH was increased from 110 + 8 min in ovary-intact ewes (N = 11) to 1101 +/- 49 min (N = 6) (P less than 0.001) with no difference between the two Merino strains. We conclude that changes in the circulating half-life of FSH do not account for the high fecundity of the Booroola but that ovariectomy can alter the half-life of FSH secreted by the pituitary gland.  相似文献

The production of cervical mucus was determined at six-hour intervals during oestrus in young (1 (1 2 ) years old) and mature (3 to 7 years old) Merino ewes. The length of the cervix and the length of the cervical epithelium was also measured in similar ewes. Total weight of mucus secreted as well as secretion rate per unit of time was greater in mature ewes than young ewes. Production was maximal during the first six hours of oestrus and declined considerably after 12 hours in young and 18 hours in mature ewes. The cervix was significantly shorter in young ewes while the length of the cervical epithelium was not significantly different in young and mature. It was concluded that lower mucus production in young ewes is not related to lower mucus output per unit area.  相似文献

Animals heterozygous (I+) for the Inverdale prolificacy gene (FecX(I)) have an increased ovulation rate whereas those homozygous (II) for FecX(I) are infertile with "streak" ovaries and follicular development arrested at the primary (type 2 follicle) stage. The streak ovaries also contain small oocyte-free nodules with granulosa-like cells and often tumor-like structures. It has been hypothesized that these abnormal structures are of granulosa cell origin, and the aim of this study was to determine whether genes normally expressed in granulosa cells are also expressed in the nodules and tumor-like structures. The mRNAs encoding c-kit and its ligand stem cell factor (SCF), FSH receptor (FSH-R), follistatin, alpha-inhibin subunit, and the beta(A)- and beta(B)-activin/inhibin subunits were localized in ovaries of ewes with 0 (++), 1 (I+), or 2 (II) copies of the FecX(I) gene (n = 4-9 animals per genotype per gene) using in situ hybridization. Ontogeny of expression of all mRNAs examined was similar between ++ and I+ ewes. Expression of c-kit mRNA was observed in the oocyte of all follicular types present in ++, I+, and II ewes. Moreover, granulosa cells of type 2 (II) and type 2 and larger follicles (++, I+) expressed SCF mRNA. The mRNAs encoding FSH-R, follistatin, alpha-inhibin subunit, and beta(B)-activin/inhibin subunit were identified in type 3 and larger follicles of ++ and I+ ewes but not in follicles of II ewes that were only at the type 1, 1a, or 2 stages of development. However, the cells within the oocyte-free nodules of II ewes expressed all of these genes. The mRNAs encoding c-kit and beta(A)-activin/inhibin subunit were not observed in granulosa cells until antrum formation (type 5 follicles) or in the nodules of II ewes. Tumors from 4 ewes were obtained and classified as cystic, semisolid, or solid structures containing granulosa-like cells or as solid structures containing predominately fibroblast- and luteal-like cells. Often, two tumors were present on the same ovary. Tumors containing granulosa-like cells (n = 3-4 per gene) expressed the mRNAs encoding alpha-inhibin subunit, beta(A)-, and beta(B)-activin/inhibin subunits, follistatin, and the FSH-R but did not contain detectable amounts of mRNA for c-kit or SCF. Tumors composed predominately of fibroblast- and luteal-like cells expressed very low levels of SCF mRNA; of the other mRNAs examined, none were detected. Also, none of the genes examined were found to be expressed by the surface epithelium, theca externa, fibroblast, or vascular cells within the ovary of animals of any genotype. These findings are consistent with the hypothesis that the somatic cells in oocyte-free nodules and tumor-like tissue in II ewes originate from the granulosa cells of the small follicles.  相似文献

Four experiments were conducted at different locations in Western Australia to evaluate the effectiveness of immunizing young (maiden, 1 1/2 year old) and adult Merino ewes with Fecundin to improve their reproductive performances. The ovulation rates of immunized maiden ewes was increased (0.06-0.30) above that of untreated ewes in both experiments 1 and 2. However, there were no significant improvements in the marking percentages for the immunized ewes with the differences between the untreated and immunized ewes ranging from -16.4 to 5.8%. In comparison with untreated ewes immunization 6 and 2 weeks before the start of joining depressed ewe fertility by 26.1% whereas immunization 8 and 4 weeks before the start of joining did not significantly affect fertility. The availability of oat grain ad libitum prior to parturition and during early lactation did not improve the survival of lambs born to immunized ewes. In experiments 3 and 4 immunization of adult Merino ewes increased their ovulation rates (0.41-0.63) above untreated controls and tended to increase the proportion of pregnant ewes which had multiple pregnancies (from -2.3 to 34.2%). The responses at the end of lambing were variable (from -19.8 to 37.5% lambs marked) with high lamb mortalities occurring in some experiments. There was no adverse effect on the reproductive performances following consecutive annual immunizations over 3 years and the absence of treatment for 1 year did not prevent a response in the following year.  相似文献

Rams and ewes of the Romney Marsh (N = 6), Dorset Horn (N = 8) and Australian Merino (N = 8) breeds were subjected to 4 successive periods of alternating 6 h light/18 h dark ('short' days) and 18 h light/6 h dark ('long' days) preceded by 16 weeks of 12 h light/12 h dark. The initial period was of 32 weeks (16 weeks 'short' days; 16 weeks 'long' days) and the next 3 were of 24 weeks (12 weeks 'short' days; 12 weeks 'long' days). Rams of all breeds showed a cyclic pattern of growth and regression of testes associated with plasma testosterone concentration, influenced by the change in light regimen 15-19 weeks previously. Sexual behaviour was also cyclic but lagged by some 6-7 weeks. The changes were greatest in the Romneys and least in the Merinos in which a higher degree of sexual activity was evident even when the testes were regressed (P less than 0.001). This was the major breed difference. All ewes of the Romney and Dorset breeds showed marked seasonality related to the imposed light regimen, whereas only 1 of the 4 Merinos did so. The mean peak of ovarian activity in the former 2 breeds coincided with that of maximum sexual activity of rams housed with them; that is, some 6 weeks after maximum scrotal volume. The rams and ewes were subjected to serial blood sampling episodes for plasma LH and testosterone and tested for plasma LH release following GnRH administration. There was little variation between breeds in LH concentration. Testosterone concentration varied greatly in the ram, highest levels associated with the developed phase of the testes and with maximum LH pulse frequency. The LH response to GnRH changed with respect to the state of the gonads. Maximal responses were observed in the developing phase of testicular growth although this variation was greater in the Romney and Dorsets than in the Merinos (P less than 0.001). In the ewes, maximal responses were seen in the follicular phase (P less than 0.001), with no difference between the luteal and acyclic phases. There were no breed differences. The effects of daily administration of 10 mg of highly purified ovine growth hormone (GH) for a period of 4 weeks on wool growth have been measured in 12 Merino ewes fed either a calculated maintenance energy intake or 1.6 times this amount (six on each ration). Concentrations of hormones, glucose, urea, alpha-amino N and amino acids in the blood were monitored and faeces and urine collected for measurement of nitrogen balance. Wool growth rate decreased by 20% during the 4 weeks of GH treatment in sheep fed the high energy diet, largely because of reduced wool fibre diameter. This was followed by restoration of normal growth and then an increase of up to 20% above control levels, a response which persisted for 12 weeks following cessation of GH administration, and which was due to increases in both fibre length and diameter. GH administration caused marked increases in plasma concentrations of GH, insulin and somatomedin C, glucose and free fatty acids, all of which returned to basal levels following cessation of GH administration. No consistent changes in plasma concentration of T3, T4, cortisol, prolactin or alpha amino N were detected. Plasma urea and methionine levels decreased during GH treatment and returned to, or were raised above, basal levels after the GH treatment period. GH injection also resulted in a net retention of N during treatment, followed by a transient period of net N loss. The GH-induced changes in wool growth may be caused by a change in the partitioning of amino acids between the muscle mass and the skin. No other contributing factor(s) were identified.  相似文献

Ewes of the Préalpes-du-Sud breed (n=112) were mated with fertile rams and were used to investigate the effect of the presence of vasectomized rams during pregnancy on reproductive outcomes. Ewes in the control group (n=56) were isolated from rams during the whole period of pregnancy, whereas those in the experimental group (n=56) were kept with vasectomized rams from day 10 post-mating until lambing. At day 10 post-mating, a series of blood samples was collected every 15 min for 8 h from five control ewes and five experimental ewes to determine their patterns of LH secretion. The introduction of the ram was associated with a rapid increase of pulsatile LH release. The lag between the introduction of the ram and the onset of the first episodic LH release was less than 15 min. The mean(±sem) number of LH pulses/4 h after the introduction of the ram (2.8±0.4) was significantly higher (P<0.01) than that observed/4 h before the introduction of the ram (1.4±0.2). Although more ewes were pregnant in the control group (87.5%) than in the ram-exposed group (82.1%), the difference was not significant. The presence of rams did not affect gestation length (145.8 days), overall lamb mortality (3.5%) or birth weights of single (3.96 kg), twin (3.24 kg) or triplet (2.59 kg) lambs. The proportion of ewes with multiple births in the control group (69.4%) was significantly greater (P<0.05) than that in the ram-exposed ewes (47.8%). The ewes in the control group had significantly more (P<0.01) twin lambs born alive (72.3%) than the ewes in the ram-exposed group (50.0%). In conclusion, the presence of vasectomized rams during early pregnancy affected the incidence of multiple births but did not affect pregnancy rate or gestation length. The altered fertility of ewes exposed to vasectomized rams may reflect changes in embryonic loss during early pregnancy.  相似文献

Castrate rams and ovariectomized ewes were maintained in the presence of entire rams and ewes and subjected to successive periods of alternating 6 h light:18 h darkness ('short' days) and 18 h light:6 h darkness ('long' days) preceded by a period of 12 h light:12 h darkness ('constant' light days). Plasma concentrations of LH and prolactin were measured in the castrate animals in order to determine how LH and prolactin secretion responded to the artificial light regime and corresponding periods of elevated or depressed testicular and ovarian activity in the entire rams and ewes. There was no variation in mean plasma LH concentrations or LH pulse frequency with either the changes in photoperiod or the phases of gonadal activity in the entire animals. However, there was a highly significant (P less than 0.001) relationship between prolactin secretion and the artificial photoperiod in both castrate groups with high and low levels coinciding with long and short days respectively. In addition, there was a marginally significant (P less than 0.1) relationship between prolactin secretion in the castrate ram and the stage of testicular activity in the entire rams with elevated levels associated with regressed activity. Prolactin secretion in the ovariectomized ewes was significantly (P less than 0.05) related to the phase of ovarian development with high levels associated with acyclic activity. It is concluded that LH secretion and pituitary responsiveness to exogenous GnRH were not modified by the artificial light regime. However, the changing light pattern was physiologically 'perceived' by the castrate animals as indicted by a concomitant variation in plasma prolactin concentrations.  相似文献
