Summary There is a considerable sexual dimorphism as regards the development of the antenna inNeodiprion sertifer. In the male this is manifested in a larger antennal surface area and the possession of a great number of long single-walled sensilla (SW 1), which are located on the antennal branches. In the female the antennal branches are vestigial, and the single-walled sensilla, of a second type (SW 2), are shorter and present in lower numbers. Both sexes have in addition four types of sensilla in common:viz. a third type of single-walled sensilla (SW 3), terminal pore sensilla, double-walled sensilla, and poreless sensilla. These four sensillar types are characterized not only by their external appearance but also by their internal morphology. Protoplasts from pea (Pisum sativum L.) leaves and cultured soybean (Glycine max L.) cells were fused by means of polyethylene glycol and subsequently cultured for one week. Both agglutinated protoplasts and cultured fusion products were examined by electron microscopy. Agglutination occurred over large areas of the plasma membranes. The membrane contanct was discontinuous and irregularly spaced. Many cultured fusion products regenerated cell walls and divided to form cell clusters. Fusion of pea and soybean interphase nuclei occurred in some cells. The detection of heterochromatin typical of pea in the synkaryon, even after division, suggests the cells were hybrids. The cytoplasm of the cells from the fusion products contained both soybean leucoplasts and pea chloroplasts. The chloroplasts had apparently ceased dividing and some showed signs of degenerating. Summary A saline extract was prepared fromDrosophila eggs. When diluted to a concentration of 1% withDrosophila tissue culture medium, it did not support growth of cells from theDrosophila line D1 during the first few days of subculture as well as medium containing serum. When cells reached a stationary phase, however, the cell density in medium containing extract was greater than in medium containing serum. By altering the concentrations of the extract, and by adding bovine albumin, a medium was obtained in which D1 cells survived initial culturing, and which supported cell growth by day 4 as well as medium plus serum. The initial retardation of growth in medium containing egg extract might be due to the need of the cells to adapt to the new medium. At the present time fourDrosophila cell lines have been maintained in this medium for more than 16 passages. Preliminary experiments with primary embryonicDrosophila cells indicate that medium containing 2% extract and bovine albumin retards the differentiation of these cells. Summary In the posterior part of the mid-gut epithelium in the lancelet, Branchiostoma lanceolatum, fine-grained cells occur. The appearance of the these cells is conspicuous with a basal and an apical swelling with small secretory granules. At the end of the secretion cycle the granular content is released into the gut lumen. The secretion product seems to consist of proteins and probably has an enzyme function. The restricted localization, the fine granulation, and the characteristic shape are features, that make these cells distinguishable from other secretory cells in the lancelet intestinal epithelium. Summary Blastoderm explants fromBrachydanio rerio (Teleostei: Cyprinidae) high blastulas exhibited limited differentiation of optic structures in culture. A number of explants showed migration of pigmented retinal epithelial cells and formation of monolayers. The findings permit comparative studies in vitro on phenomena pertaining to pigmented retinal epithelial cell morphology, function, and differentiation. The expression of extA, an extensin gene from Brassica napus L. (oilseed rape) was examined in transgenic Nicotiana tabacum L. (tobacco) and untransformed Brassica juncea L. and B. napus tissues. Northern analysis showed that this gene maintained its normal pattern of expression when transferred to tobacco. In transgenic tobacco plants containing an extA promoter/-glucuronidase coding sequence fusion, expression of extA was detected in the external and internal phloem of the main stem. High expression levels were seen in cortical parenchyma cells at the point where the axillary flowering branch joined the main stem. Expression was greatest in regions where the maximum tensile stress would seem to be exerted on the main stem by the weight of the axillary branch. It was confirmed that this expression pattern was due to tensile stress by using weights to induce expression of the fusion gene in axillary flowering stalks. Summary The cytological changes in the pars distalis of pituitary of male and female green frog, Rana esculenta during the reproductive cycle have been studied by examining the form, cytoplasmic granulation and numerical proportions of the different cell types. Some sex differences were recorded, particularly in relative total number of cells in a median sagittal section of the pars distalis. No extensive cytological changes occur during the hibernation period, but there are striking alterations at the time of spring awakening, during spawning months and during replenishment of the spent gonads i.e. during late summer and autumn months. Important changes are to be observed in the acidophils type one (A1) and basophils type two (B2) and three (B3). Changes in the A1 cells during breeding season have been speculated as being in support of the spawning activity of the animal. The B2 cells exhibit most profound changes which correlate precisely with the morphological changes in gonads, and these cells are considered as the FSH-gonadotrops. Summary The innervation pattern of the intermediate lobe of the skate (Raja radiata) was studied with histological and fluorescence histochemical methods. Neurosecretory fibres, stained with i.a. pseudo-iso-cyanine, were found running in bundles in the central parts of the cell cords. They terminated partly around the perinuclear parts of the intermedia cells, partly around the apices of the cells close to the vascular walls.A catecholamine innervation of the intermedia was also established. Catecholaminecontaining fibres with the appearance of nerve terminals were found around the intermedia cell apices close to the vessels. In some specimens, catecholamine fibres also seemed to terminate at the perinuclear parts of the cells.Thus it is possible, judging solely from structural relations, that both the cell body (the synthesis pole) and the cell apex (the release pole) receive a dual innervation. Summary The turnip moth Agrotis segetum possesses seven different types of sensilla: four single-walled (SW), one double-walled (DW), one terminal-pore (TP), and one poreless sensilla (NP).The SW 1 and SW 2 sensilla have the same external appearance, being long and slender, but differ in the branching pattern of the sensory processes: unbranched and branched in SW 1 and SW 2, respectively. The SW 3 sensilla are shorter, sickle-shaped, and contain a large number of branches from the sensory processes. These three sensillar types are innervated by 2–3 sensory cells. The SW 4 sensilla are raisin-shaped and possess three profusely branched sensory processes. The DW sensilla are short and have apical slit-like pores. This sensillar type has 5–6 sensory processes. The TP sensilla possess five sensory processes, one of them terminates basally in a tubular body, the others in the apical part of the long cuticular bristle. The NP sensilla are stout and have apical conelike structures. Two of the sensory processes terminate in the apical part, the third proximally. The third sensory process has a lamellar pattern. Summary It has been previously demonstrated that the hepatopancreas of the snail, Helix pomatia, produce proteinaceous particles (termed b-granules), which are involved in the process of calcification. In calcium cells of this organ calcium spherites arise from these granules. The present study retraces the intracellular development of b-granules from Golgi vesicles and from Golgi saccules encircling the cytoplasmic areas up to the formation of calcium spherites. It is suggested that the mature b-granules are well-defined cytoplasmic units with calcifying capacity. The fine structure of b-granules and calcium spherites is described. The possible function of various cell organelles in the formation of the b-granules and spherites is discussed.This investigation was supported by grants from the Swedish Natural Science Research Council. Summary The cell wall regeneration on protoplasts derived from maize mesophyll cells was compared with wall regeneration on protoplasts derived from suspension cultured cells using light microscopy, transmission electron microscopy, and mass spectrometry. The time course of cell wall regeneration has shown that the mesophyll protoplasts regenerated walls much slower than the protoplasts derived from cultured cells. Moreover, cell wall materials on the mesophyll protoplasts were often unevenly distributed. Electron microscopy has further demonstrated that the mesophyll protoplasts have less organized and compact walls than the protoplasts from cultured cells. Chemical analysis revealed that the mesophyll protoplasts had a lower ratio ofβ-(1–3)-glucan toβ-(1–4)-glucan than protoplasts from cultured cells. An electrophoretically detectable variant of pyruvate kinase (EC 2.7.1.40) has been found in the house mouse Mus musculus. The variant was seen in all tissues examined except liver and red cells. The gene (Pk-3) determining this electrophoretic variation is inherited as an autosomal codominant located on chromosome 9. Our data confirm that the genetic determination of pyruvate kinase in liver and red cells is separate from that in other tissues. In addition, our results indicate that the muscle (M1) and kidney (M2) pyruvate kinase isozymes share at least one genetic determinant and may in fact be determined by the same structural gene.This work was supported by the Medical Research Council and by NIH Grants GM 20919 and RR 01183. Summary InNotodromas monachus, the three cups of the nauplius eye are formed by four pigment cells. The insides of the cups are lined with tapetal cells, which produce several layers of reflecting crystals. The reflecting crystals form a concave mirror in each cup upon which the retinular cells rest. The two-celled rhabdoms are few and perpendicular to the tapetal layer. The axons from the tripartite eye leave the retinular cells distally in three separate groups. The eye is thus of the inverse type. Large lens cells, with a low refractive index, are present in the open part of each cup. Distal to the lens cells, highly refractive lenses are formed in the cuticle. These lenses serve to decrease the effective curvature of the mirrors, thus enabling the reflectors to produce a focused image on the retina. The ventral cup differs by the lack of a cuticular lens and has degenerated-appearing cellular elements. Summary A cell line (SWT) was established from embryonic tissue of the red swordtailXiphophorus helleri. The SWT cells grew optimally, at 26°C to 30°C in Eagle's basal medium plus 10% fetal calf serum but failed to grow at 16°C and 37°C. After 50 subcultivations, the cells remained contact-inhibited and were pseudodiploid with a chromosomal modal number of 46. Virological studies demonstrated that SWT cells supported replication of the following viruses at the indicated temperatures: IPN virus (22°C), FV-3 (30°C), and VSV (33°C). The following mammalian, viruses failed to replicate at 33°C: vaccinia, poliovirus 2, herpes simplex, and reovirus 2. Although not replicating, reovirus induced interferon in there cells. The conductance of pores induced by Staphylococcus aureus -toxin in Lettre cells has been compared to that in bilayers composed of synthetic lipids or Lettre cell membrane constituents. Previously described characteristics of toxin-induced conductance changes in lipid bilayers, namely rectification, voltage-dependent closure, and closure at low pH or in the presence of divalent cations (Menestrina, 1986) are displayed also in bilayers prepared from Lettre cell membranes and in patch clamped Lettre cells. It is concluded that endogenous proteins do not affect the properties of -toxininduced channels significantly and that the relative lack of ion channels in Lettre cells makes them ideal for studies of pore-forming toxins by the patch clamp technique.Dr. Sviderskaya is on leave of absence from the Physiology Institute, University of St. Petersburg, RussiaWe are grateful to Dr. J.P. The relationship between the gas-exchange characteristics of spinach (Spinacia oleracea L.) leaves and the activation state of sucrose-phosphate synthase was examined at different intercellular partial pressures of CO2 at two different photon flux densities. There was a strong positive correlation between the activation state of sucrose-phosphate synthase and the assimilation rate. Summary A cell line designated SP-1 was established from tissue of the silver perch,Bairdiella chrysura. Cells were fibroblast-like and grew best at 26°C in Leibovitz medium (L-15) containing 15% fetal bovine serum and 0.150m sodium chloride. Passage 1 to passage 9 SP-1 cells contained a chromosome number of 48; at passages 27 and 50 the modal numbers were 51 and 54, respectively. Confirmation of the origin of SP-1 cells was made by the cytotoxic antibody dye-exclusion test. This cell line supported the growth of lymphocystis virus from the silver perch but was not found to replicate various other fish and mammalian viruses. Summary Synchronization ofPlasmodium falciparum cultured in vitro results in a one-step growth pattern that allows the study of stage-specific metabolic activities of the parasites. Lactic acid (LA) was selected as a metabolic marker, and the concentration of this end product found in spent media was correlated with the different erythrocytic stages of the parasites. When the medium was changed at 12 h intervals, cultures containing predominantly trophozoites produced 3.66±0.55 μmol LA per 12 h per 107 parasitized cells (n=26), an amount of LA that is about 8 to 20 times higher than that found in corresponding cultures containing predominantly ring forms. Depending on the stage of development, parasitized red blood cells produced between 5 and 100 times more LA than uninfected erythrocytes (3.72±0.62 μmol LA per 12 hours per 109 red blood cells) (n=41) when cultured under identical conditions. The intraerythrocytic development of the parasites was not impaired by exposure to extracellular concentrations of LA up to 12 mM over a 12 h period. The growth resulting in such cultures was described as uninhibited and was characterized by a multiplication index of 10 or higher. Above the threshold of 12 mM of LA, progressive inhibition of parasite development occurred. The stage-specific LA production reported can be used to predict the amount of LA that will have accumulated at the end of a subsequent 12 h incubation period during synchronized in vitro growth ofPlasmodium falciparum. Using these values, it is possible to establish an optimal medium exchange schedule, thereby assuring uninhibited growth and a correspondingly high parasite yield. J. W. Summary The appearance of the mucosal cell layer of the isolated urinary bladder of the toadBufo marinus, has been examined using stereoscopic and conventional transmission electron microscopy.Three cell types can be identified in surface view, these are granular cells, mitochondriarich cells and goblet cells. Cell boundaries between granular cells are clearly defined by membranous folds along their margins. Although no changes are seen in the stereoscopic electron micrographs when the granular cells are made permeable to water by vasopressin, the changes observed on transmission electron micrographs include swelling of cell bodies and nuclei, filling of intercellular channels with water, and the appearance beneath the mucosal cell membrane surface of electron dense granules.Differences between the appearance of the bladder mucosal cells by the two methods of electron microscopical examination are due largely to water loss when the tissue is freeze dried prior to stereoscopic examination.Grateful thanks are due to Dr. W. Summary Using the enzyme peroxidase and ionic lanthanum as tracers, paracellular uptake has been demonstrated in the foot of the slug Agriolimax reticulatus (Mü). Both tracers appeared to pass between adjacent foot epithelial cells and were demonstrated in the zonula adhaerens, the septate desmosomes, and the intercellular spaces which occur beneath the septate junctions. Ferritin, a somewhat larger tracer, was excluded from all these sites.Ionic lanthanum was not normally pinocytosed in short incubation times. The epithelial cells could be induced to endocytose this marker, however, when combined with a variety of proteins.
