In the present study, we have evaluated the effects of increased UV-B radiation that simulates 17% ozone depletion, on fungal colonisation and concentrations of rutin, catechin and quercetin in common buckwheat (Fagopyrum esculentum) and tartary buckwheat (Fagopyrum tataricum). Induced root growth and reduced shoot:root ratios were seen in both of these buckwheat species after enhanced UV-B radiation. There was specific induction of shoot quercetin concentrations in UV-B-treated common buckwheat, whereas there were no specific responses for flavonoid metabolism in tartary buckwheat. Root colonisation with arbuscular mycorrhizal fungi significantly reduced catechin concentrations in common buckwheat roots, and induced rutin concentrations in tartary buckwheat, but did not affect shoot concentrations of the measured phenolics. Specific UV-B-related reductions in the density of microsclerotia were observed in tartary buckwheat, indicating a mechanism that potentially affects fungus-plant interactions. The data support the hypothesis that responses to enhanced UV-B radiation can be influenced by the plant pre-adaptation properties and related changes in flavonoid metabolism.  相似文献

Abstract

The proteins of the R2R3-MYB superfamily play key roles in the growth and development processes as well as defense responses in plants. However, their characteristics and functions have not been fully investigated in tartary buckwheat (Fagopyrum tataricum), a strongly abiotic resistant coarse cereal. In this article, eight tartary buckwheat R2R3-MYB genes were isolated with full-length cDNA and DNA sequences. Phylogenetic analysis of the members of the R2R3-MYB superfamily between Arabidopsis and tartary buckwheat revealed that the assumed functions of the eight tartary buckwheat R2R3-MYB proteins are divided into five Arabidopsis functional subgroups that are involved in abiotic stress. Expression analysis during abiotic stress and exogenous phytohormone treatments identified that the eight R2R3-MYB genes responded to one or more treatments. This study is the first comprehensive analysis of the R2R3-MYB gene family in tartary buckwheat under abiotic stress.

Two cultivars of common buckwheat (Fagopyrum esculentum), Pyra and Siva, were exposed to three treatments: water deficit (WD), foliar spraying by selenium (as Na2SeO4) (Se), and the combination of both. In WD-plants the stomatal conductance (g s) was significantly lower, while WD+Se-plants of Siva had significantly higher g s. None of the treatments resulted in significant differences of potential photochemical efficiency of photosystem 2 (PS2). A significantly higher actual photochemical efficiency of PS2 was obtained in Siva WD-plants and in Pyra Se-and WD-plants which was possibly due to improvement of plant water management during treatment. A significant interaction was observed between the effects of WD and Se on respiratory potential in Pyra. WD, Se, and the WD+Se combination resulted in shorter Pyra and Siva plants, with a reduced number of nodes. WD slightly negatively affected the yield per plant. The yield was highest in plants exposed to Se only. In Siva the number of seeds was triple while the average seed mass remained unchanged.  相似文献

Morphology, biomass accumulation and allocation, gas exchange, and chlorophyll fluorescence were compared for one-year-old seedlings of Salix psammophila and Artemisia ordosica, two dominant desert species, in response to two water supplies (equivalent to 315.0 mm for present precipitation in growing season and to 157.5 mm for future decreasing precipitation) during 105 d. For both species, photochemical efficiency of photosystem 2 (Fv/Fm), net photosynthetic rate, transpiration rate, stomatal conductance, biomass accumulation in different organs, tree height, number of leaves, and leaf area were reduced in response to the decrease in water supply. For both species, instantaneous water use efficiency was not affected by the water deficit. However, diurnal patterns of gas exchange and biomass allocation were affected in different ways for the two species, with notably a decrease in specific leaf area and an increase in root : shoot ratio for S. psammophila only. Overall, S. psammophila was more responsive to the decreasing precipitation than A. ordosica.  相似文献

Photosynthetic and growth characteristics of Mosla chinensis and M. scabra were compared at three irradiances similar to shaded forest understory, forest edge, and open land. At 25 % full ambient irradiance, M. chinensis and M. scabra had similar photosynthetic characteristics, but saturation irradiance, compensation irradiance, and apparent quantum yield of M. chinensis were higher than those of M. scabra at full ambient irradiance and 70 % full ambient irradiance. At the same irradiance treatment, specific leaf area and leaf area ratio of M. chinensis were lower than those of M. scabra. Photon-saturated photosynthetic rate and water use efficiency of M. chinensis, however, were not significantly higher than those of M. scabra, and the leaf area and total biomass were lower than those of M. scabra. As a sun-acclimated plant, the not enough high photosynthetic capacity and lower biomass accumulation may cause that M. chinensis has weak capability to extend its population and hence be concomitant in the community.  相似文献

The effects of supplemental UV-B radiation on crop growth, morphology, reproduction and physiology were studied in three cultivars of Fagopyrum esculentum Moench (buckwheat) originating from high elevation (Qinghai-Tibet plateau) and lower altitudes (The Sichuan Basin). Our results showed that common buckwheat was sensitive to UV-B stress. Plant growth, development, and reproduction were inhibited by elevated UV-B radiation. Plant lipid oxidation and polyphenol oxidase (PPO) activity increased with increasing UV-B radiation, along with the concentration of phenylpropanoid compounds, superoxide dismutase (SOD) activity and ascorbic acid (Asa) concentration were also enhanced at the lowest level of supplemental UV-B radiation but decreased at the higher level of enhanced UV-B. While, a cultivar originating from elevated locations had lower dry matter accumulation and was more tolerant to UV-B radiation than cultivars originating from lower elevations. The effects on leaf thickness and increased antioxidant capacity could be linked with the improved performance of cultivar originating from high elevation when exposed to enhanced UV-B radiation. We conclude that UV-B tolerance should be considered prior to introducing or breeding common buckwheat cultivars from lowland cultivation to regions at high elevation such as the Qinghai-Tibet plateau.  相似文献

Common buckwheat (Fagopyrum esculentum Moench) is well known as a weed suppressing crop due to its strong competitive and allelopathic characteristics. The possible allelopathic compounds in buckwheat include compounds from different groups, such as flavonoids, fatty acids, phenolic acids, etc. Less attention has been paid to other phenolic compounds, specifically eugenol, o-eugenol, coniferyl alcohol and 3,4,5-trimethoxyphenol as possible allelochemicals. The effects of eugenol, o-eugenol, coniferyl alcohol, and 3,4,5-trimethoxyphenol on germination and plant growth were tested on seven plant species. The results of our study showed that eugenols, coniferyl alcohol, or 3,4,5-trimethoxyphenol are minority components of the buckwheat plant. Eugenol reached the highest concentration (1.16μg/g DW in buckwheat leaves) from these compounds and they probably do not have a significant function in the allelopathy of common buckwheat. However, due to the inhibitory effects on germination and plant growth, eugenol could be utilized in the plant protection of sustainable agriculture.  相似文献

The morphological and genetic studies of the tlb mutant of common buckwheat Fagopyrum esculentum from the collection of the All-Russia Research Institute of Legumes and Groat Crops have demonstrated that gene TLB controls an important stage of flower development—it determines the lower boundary of simple perianths. The loss of TLB gene function leads to changes in the structure of the bract from scale-like to tepal-like. Gene TLB is assumed to limit, on the basal side, the region of the expression of genes determining the development of flower organs as petals.  相似文献

Calycanthus chinensis is an endangered plant of the national second-grade protection of China restricted in a small area in Zhejiang Province. We studied parameters of photosynthesis, chlorophyll (Chl) contents, and Chl fluorescence (minimum fluorescence, F0, maximum fluorescence, Fm, variable fluorescence, Fv, and Fv/Fm) of C. chinensis and Chimonanthus praecox. C. chinensis had lower compensation irradiance but higher saturation irradiance than C. praecox. Hence C. chinensis has more advantage in obtaining and utilizing photon energy and higher Chl content, and is more adaptive to higher temperature and propitious to thermal dissipation than C. praecox. In addition, C. chinensis produces abundant, well-preserved seed with a higher germination rate and a wider adaptability to temperature than C. praecox. Thus C. chinensis is prone to survival and viability, and gets rid of the endangered plant species of the national second-grade protection of China.  相似文献

This study describes two phenotypes of Arabidopsis thaliana (ecotype Columbia) developed in vitro under salt stress (75 mM NaCl). The phenotypes 01 and 02 appeared visibly distinguishable by rosette morphology and competence to produce flowers. Phenotype 01, sensible to salt stress, accumulated high quantities of Na+, showed a slight reduction in dry mass, and high protein and chlorophyll contents. Moreover, its anatomy exhibited some xeromorphic traits. Phenotype 02, clearly salt tolerant, showed a morphology similar to control plants, displaying typical phyllotactic rosette and flowering stalk production. Accumulation of Na+, protein and chlorophyll contents were close to control plants. Reversion experiments on NaCl free MS medium, showed a partially recovered phenotype 01. A threshold salt stress concentration that permits the simultaneous development of two phenotypes, was found.  相似文献

Tartary buckwheat (Fagopyrum tataricum Gaertn.) is highly nutritious and an excellent dietary source of flavonoid compounds. Chalcone synthase (CHS) is the first key enzyme involved in flavonoid biosynthesis. Here, three putative CHS genes (designated as FtCHS1 (GU172165), FtCHS2 (KT284884), and FtCHS3 (KT284885) were isolated from tartary buckwheat. Nucleotide sequence analysis indicated that FtCHS1 and FtCHS2 each contained one intron of 444 bp and 157 bp, respectively. FtCHS3 included two introns, one of 86 bp and another of 73 bp. The results of quantitative real-time PCR (qRT-PCR) showed the FtCHSs expression presented the same pattern in the stems and flowers, with FtCHS1>FtCHS3>FtCHS2. A different tendency was found in leaves, with FtCHS3>FtCHS2>FtCHS1. However, there was no direct correlation between the three CHS expression and total flavonoids. Furthermore, high-performance liquid chromatography (HPLC) performance reveals rutin is the most abundant flavonoid in all tissues, leaves should be the main location for quercetin storage in tartary buckwheat.  相似文献

Photosynthetic water use efficiency (PWUE), stomatal conductance (g s), and water potential were measured at two different positions in the tree crown of two emergent tropical tree species (Shorea beccariana Burck, Dryobalanops aromatica Gaertn. f.). The trees were about 50 m high, in a tropical rain forest in Sarawak, East Malaysia. In both species, g s at the upper crown position at midday was lower than at the lower crown position, even though both positions were exposed to full sunlight; the difference was greater in S. beccariana. Hydraulic limitation occurs in the upper crown position in both species. A midday depression was observed in the photon saturated photosynthetic rate in both species, especially at the upper crown. However, PWUE was markedly higher in the upper crown than the lower crown at midday, even though no morphological adjustment was observed in the leaves; this difference was greater in S. beccariana.  相似文献

The effect of arbuscular mycorrhizal fungi (AMF) inoculation and organic slow release fertilizer (OSRF) on photosynthesis, root phosphatase activity, nutrient acquisition, and growth of Ipomoea carnea N. von Jacquin ssp. fistulosa (K. Von Martinus ex J. Choisy) D. Austin (bush morning glory) was determined in a greenhouse study. The AMF treatments consisted of a commercial isolate of Glomus intraradices and a non-colonized (NonAMF) control. The OSRF was applied at 10, 30, and 100 % of the manufacturer’s recommended rate. AMF plants had a higher net photosynthetic rate (P N), higher leaf elemental N, P, and K, and generally greater growth than NonAMF plants. Total colonization levels of AMF plants ranged from 27 % (100 % OSRF) to 79 % (30 % OSRF). Root acid phosphatase (ACP) and alkaline phosphatase (ALP) activities were generally higher in AMF than non-AMF plants. When compared to NonAMF at 100 % OSRF, AMF plants at 30 % OSRF had higher or comparable ACP and ALP activity, higher leaf elemental P, N, Fe, Cu, and Zn, and a greater P N (at the end of the experiment), leading to generally greater growth parameters with the lower fertility in AMF plants. We suggest that AMF increased nutrient acquisition from an organic fertilizer source by enhancing ACP and ALP activity thus facilitating P acquisition, increasing photosynthesis, and improving plant growth.  相似文献

Lignin is closely related to the lodging resistance of common buckwheat (Fagopyrum esculentum Moench.). However, the characteristics of lignin synthesis related genes have not yet been reported. We investigated the lignin biosynthesis gene expression, activities of related enzymes, and accumulation of lignin monomers during branching stage, bloom stage, and milky ripe stage by real-time quantitative PCR, UVspectrophotometry, and gas chromatography-mass spectrometry in the 2nd internode of three common buckwheat cultivars with different lodging resistance. The results showed that lignin content and the activity of phenylalanine ammonia lyase (PAL), 4-coumarate: CoA ligase (4CL), cinnamyl alcohol dehydrogenase (CAD) and peroxidase (POD) were closely related to the lodging resistance of common buckwheat. Further, we studied gene expression of cinnamate 4-hydroxylase (C4H), caffeoyl-CoA O-methyltransferase (CCoAOMT), ferulate 5-hydroxylase (F5H), cinnamoyl-CoA reductase (CCR), and caffeic acid O-methyltransferase (COMT). The lignin biosynthesis genes were divided into three classes according to their expression pattern: 1) expression firstly increasing and then descending (PAL, 4CL, CAD, C4H, CCoAOMT, F5H, and CCR), 2) expression remaining constant during maturation (C3H), and 3) expression decreasing with maturation (COMT). The present study provides preliminary insights into the expression of lignin biosynthesis genes in common buckwheat, laying a foundation for further understanding the lignin biosynthesis.  相似文献

Scanning electron microscopy was used to study the structure of callus of Fagopyrum esculentum Moench. during induction of morphogenesis. It was found that transferring of callus on the medium lacking growth regulators induced several simultaneous developmental programs: embryoidogenesis, rhizogenesis, and gemmogenesis. The program of embryoidogenesis prevailed over the others. Various morphological structures were found to appear on the callus surface in the following order: trichome (on the 2nd day), roots (the 2nd or 3rd day), embryoids (the 6th to 8th day), and buds (the 10th to 14th day). It was shown that morphogenesis is preceded by loosening of the callus surface and by appearance of the secret on it. Common bean (Phaseolus vulgaris L.) varies in growth habit from aggressive climbing types to bush beans. Growth habit is determined by a combination of factors including determinate versus indeterminate growth, total plant height, degree of branching and internode length. Together these factors make up climbing ability. The objective of this research was to determine the quantitative trait loci (QTL) controlling climbing ability in a F5:8 recombinant inbred line population derived from an inter-gene pool cross of an aggressive indeterminate climbing bean with type IV growth habit (G2333) by an indeterminate bush bean of type IIb growth habit (G19839). The population was planted in four randomized complete block design experiments across environments that varied in altitude (from 1,000 to 1,750 masl) and soil fertility (low versus high phosphorus). QTL were identified for plant height, internode length and number of branches per plant on a genetic map covering all common bean linkage groups with a total length of 1,175 cM. In addition a scale was developed to evaluate overall climbing ability and was also used to identify QTL. A total of 7 QTL were found for plant height, 9 for climbing ability, 6 for internode length and 1 for branch number. The largest number and most significant QTL were found on the lower half of linkage group B04 suggesting a major pleiotropic locus for growth habit traits at this location of the genome that is distinct from previously characterized genes which control plant morphology of the crop.  相似文献

Green alga Zygnema was exposed to three concentrations of selenium and two levels of UV-B radiation. The combined effects of both treatments on energy availability; photochemical quantum yield and respiratory potential were studied. Our findings show that traces of selenium enhance metabolic process connected with photochemical quantum yield and mitochondrial respiration. Surprisingly, selenium does not diminish the effects of UV-B radiation; on the contrary, the combined action of UV-B radiation and traces of selenium leads to pronounced negative effects on photochemical quantum yield and the respiratory potential. Selenium is involved in the activation of energy resources in green alga Zygnema. The importance of selenium for activity of the mitochondria is possibly an evolutionary recollection from an endosymbiotic bacterium.  相似文献

Agronomic traits, photosynthetic pigments, gas exchange, and chlorophyll (Chl) fluorescence parameters of red stem buckwheat (Fagopyrum dibotrys Hara) mutants induced by γ-radiation were compared with green control at seedling stage. Plant height, number of first-class branches, and rhizome biomass were inhibited significantly (p<0.01). Chl a, Chl b, and Chl a+b contents decreased with elevated dose of γ-rays, while increasing carotenoid content indicated that buckwheat was capable of adjusting to the radiation damage. Decrease in net photosynthetic rate was the result of both stomatal and non-stomatal limitations. Fluorescence parameters, such as F0, Fm, Fv/Fm, Fv/F0, ΦPS2, electron transport rate, and photochemical quenching declined significantly (p<0.01) as compared with control due to photoinhibition, while non-photochemical quenching increased to enhance thermal dissipation. Lower parameters implied that leaf tissue was damaged significantly by high dose of γ-radiation and therefore leaf senescence was accelerated.  相似文献
