Chaetosphaeria tortuosa is described as the newly discovered teleomorph of Menispora tortuosa, based on specimens from Canada and the Czech Republic, and single spore isolations from both morphs. The fungus produces superficial, more or less globose, papillate, dark brown to black smooth perithecia (200–)220–250 × (220–)230–260 μm. The asci are unitunicate, 8-spored, cylindrical-fusiform, (110–)120–133(–145) × 12–14 with a distinct apical, nonamyloid annulus 1–1.5 μm high, 3.5–4 μm wide. The ascospores are fusiform, 19–24 × 5–6 μm, hyaline, 3-septate, smooth, and 2-seriate in the ascus. The morphology of the teleomorph and anamorph are similar to that of C. ovoidea (anamorph: M. glauca), differing in dimensions of asci and ascospores, and in the disposition and morphology of the phialides of the anamorphs. The generic concept and phylogeny of Menispora is briefly discussed, and a key to the 11 species currently accepted in the genus is provided.  相似文献

Resin glycosides are secondary metabolites exclusive to the convolvulaceous plants. In this study, crypthophilic acids A–C (13), the first resin glycosides occurring in another family (Scrophulariaceae), and the other constituents of Scrophularia cryptophila were examined for in vitro antiprotozoal and antimycobacterial potentials. Except for crypthophilic acid B (2), all tested compounds exhibited growth-inhibitory effect against Trypanosoma brucei rhodesiense, with l-tryptophan (6) and buddlejasaponin III (7) being the most potent ones (IC50's 4.1 and 9.7 μg/ml). In contrast, the activity towards Trypanosoma cruzi was poor, and only crypthophilic acid C (3), 6 and 7 were trypanocidal at concentrations above 40 μg/ml. With the exception of 2 and 6, all compounds were active against Leishmania donovani. Harpagide (4) and 3 emerged as the best leishmanicidal agents (IC50's 2.0 and 5.8 μg/ml). Only compounds 3, 6 and 7 showed antimalarial activity against Plasmodium falciparum with IC50 values of 4.2, 16.6 and 22.4 μg/ml. Overall the best and broadest spectrum activity was presented by compounds 3 and 7, as they inhibited all four parasitic protozoa. None of the isolates had significant activity against Mycobacterium tuberculosis (MICs >100 μg/ml) or were toxic towards mammalian (L6) cells. This is the first report of antiprotozoal activity for natural resin glycosides, as well as for harpagide (4), acetylharpagide (5), tryptophan (6) and buddlejasaponin III (7).  相似文献

Efforts have been made to evaluate the microbial and decomposition efficiency of three different vermireactors: (i) polyculture (introducing equal numbers of anecic and epigeic earthworms), (ii) monoculture (anecic) and (iii) monoculture (epigeic), designed by using earthworms of two different ecological categories i.e. anecic (Lampito mauritii Kinberg) and epigeic (Eisenia fetida (Savigny)). The microbial load of vermireactors was measured through substrate-induced respiration rate (SIR), microbial biomass N content and rate of dehydrogenase activity, while mineralization rate was evaluated measuring some chemical parameters of the substrate. Earthworms caused a decrease (as compared to initial value) in pH (41.9–80.7%), organic C (10.3–14.2%) and C:N ratio (41.9–80.7%) and an increase in total N (29.1–58.8%), NH4-N (876.1–1485.7%), NO3-N (29081.8–56792.6%), available P (16–19.4%) and exchangeable K (9.8–13.5%) contents of the substrate. The mineralization efficiency of the reactors was in the order: polyculture (epigeic + anecic) > monoculture (anecic) > monoculture (epigeic). The polyculture reactor showed the maximum rate of SIR (2.91 ± 0.2 mg COg−1 substrate), microbial biomass N (3108.1 ± 289.2 mg N g−1 substrate), and dehydrogenase activity (2453.3 ± 379.8 μg g−1 substrate 24 h), while in the monoculture (epigeic) the lowest values of the same parameters were observed. It is concluded that the observed differences among reactors were due to different feeding behaviour and niche structures of epigeic and anecic earthworms. Data suggests that burrowing earthworms in waste-decomposing-system not only enhance the microbial efficiencies, but at the same time also accelerate the organic matter mineralization in a vermireactor. However, most of the previous studies were based on monoculture reactors (using epigeic earthworms) which have been recommended for waste decomposition operations, but this study revealed that polyculture vermicomposting (adding of burrowing worms with epigeic earthworms in vermicomposting system) might be beneficial for rapid decomposition of organic wastes.  相似文献

A technically standardised bioassay method was designed, evaluated and used to assess virulence and host range of hypocrealean fungi against aphids. A track mounted sprayer was used to apply conidia because hand held versions of the same sprayer can be used for field applications, thereby allowing the outcome from laboratory experiments to predict activity in the field accurately. Eighteen fungal isolates were assessed in single concentration bioassays against the black bean aphid Aphis fabae Scopoli. Isolates comprised commercially available mycoinsecticides (based on Beauveria bassiana and Lecanicillium longisporum) and isolates of B. bassiana, Lecanicillium spp., Paecilomyces fumosoroseus and Metarhizium anisopliae. Aphid mortality was in excess of 80% for 15 isolates, and HRI 1.72 (L. longipsorum), Z11 (P. fumosoroseus), Mycotech strain GHA (B. bassiana) and ARSEF 2879 (B. bassiana) were studied further. Multiple concentration bioassays identified HRI 1.72 as the most virulent isolate against A. fabae with significantly smaller LC50 and LT50 values compared to other isolates. A precise LC50 value (2.95 × 102 conidia ml−1) was calculated for HRI 1.72 using a second multiple concentration assay with smaller concentrations of conidia. The four isolates were applied at a single concentration (1 × 108 conidia ml−1) against Myzus persicae, A. fabae, Acyrthosiphon pisum, Metopolophium dirhodum, Sitobion avenae and Rhopalosiphum padi. A ranking of aphid susceptibility was obtained, such that S. avenae > M. persicae, A. pisum, A. fabae > R. padi. Results indicate the importance of standardising bioassay methods to reduce bioassay variability without compromising the ability to use the bioassay to investigate fungus–host interactions under varying abiotic and biotic conditions.  相似文献

The nitrogen uptake and growth capabilities of the potentially harmful, raphidophycean flagellate Heterosigma akashiwo (Hada) Sournia were examined in unialgal batch cultures (strain CCMP 1912). Growth rates as a function of three nitrogen substrates (ammonium, nitrate and urea) were determined at saturating and sub-saturating photosynthetic photon flux densities (PPFDs). At saturating PPFD (110 μE m−2 s−1), the growth rate of H. akashiwo was slightly greater for cells grown on NH4+ (0.89 d−1) compared to cells grown on NO3 or urea, which had identical growth rates (0.82 d−1). At sub-saturating PPFD (40 μE m−2 s−1), both urea- and NH4+-grown cells grew faster than NO3-grown cells (0.61, 0.57 and 0.46 d−1, respectively). The N uptake kinetic parameters were investigated using exponentially growing batch cultures of H. akashiwo and the 15N-tracer technique. Maximum specific uptake rates (Vmax) for unialgal cultures grown at 15 °C and saturating PPFD (110 μE m−2 s−1) were 28.0, 18.0 and 2.89 × 10−3 h−1 for NH4+, NO3 and urea, respectively. The traditional measure of nutrient affinity—the half saturation constants (Ks) were similar for NH4+ and NO3 (1.44 and 1.47 μg-at N L−1), but substantially lower for urea (0.42 μg-at N L−1). Whereas the α parameter (α = Vmax/Ks), which is considered a more robust indicator for substrate affinity when substrate concentrations are low (<Ks), were 19.4, 12.2 and 6.88 × 10−3 h−1/(μg-at N L−1) for NH4+, NO3 and urea, respectively. These laboratory results demonstrate that at both saturating and sub-saturating N concentrations, N uptake preference follows the order: NH4+ > NO3 > urea, and suggests that natural blooms of H. akashiwo may be initiated or maintained by any of the three nitrogen substrates examined.  相似文献

Mt. Teide (Tenerife, Spain) is a high volcanic island mountain with an unusually low tree line elevation (2000–2100 m). While searching for the causes of this tree line depression, we analysed the concentrations of total N, available P, and salt-exchangeable Ca, K and Mg in three soil horizons, and the foliar N, P, Ca, K and Mg concentrations in six abundant plant species (trees, shrubs, forbs) along a transect from 1400 (1600) to 3100 m a.s.l. The objective of the study was to detect altitudinal trends in soil and plant nutrient status below and above the tree line. For characterising elevational changes in microclimate and hydrology, we also conducted measurements of air and soil temperatures (Ta and Ts), atmospheric water vapour saturation deficit (D), potential evaporation (E) and soil moisture (θ) on 3–4 occasions along the transect.Ta and Ts did not linearly decrease with elevation but were highest at or immediately below the tree line and were relatively low in the closed Pinus canariensis forest at 1600 m. Ts reached maxima at about 70 °C near the tree line. The absence of a linear temperature decrease with elevation was caused by a stable temperature inversion at about 2000 m and by canopy shading in the forest canopy below the tree line. In June, the topsoil (10 cm) dried out almost completely between 1800 and 3100 m, but remained moist in the pine forest at 1600 m. This gradient reflects the transition from the montane cloud belt to the dry alpine belt higher upslope. The subsoil (30 cm) contained >30 vol% of soil water at all elevations even in June except for the uppermost site (3100 m). Potential evaporation increased with elevation despite a decrease in D. We assume that this is mainly due to the air pressure-dependent increase with elevation in the diffusion coefficient for water vapour in air.The concentrations of N and ‘available’ P (after Olsen) in the mineral topsoil were by far smaller than in alpine soils of other humid mountains which is thought to be a consequence of a very dry and biologically inactive topsoil in the semi-arid alpine belt of Mt. Teide. In contrast to many other mountains, foliar N, P and cation concentrations in the plants did not increase with elevation but either remained unchanged, or decreased as in the tree line species P. canariensis. Nevertheless, P. canariensis probably is not limited by nutrient deficiency at the tree line despite rather low N and P needle concentrations. Rather, drought and heat stress effects on seedling establishment are thought to be the causes of the tree line depression.  相似文献

A series of experiments was conducted to examine effects of four strains of the estuarine dinoflagellate, Pfiesteria shumwayae, on the behavior and survival of larval and adult shellfish (bay scallop, Argopecten irradians; eastern oyster, Crassostrea virginica; northern quahogs, Mercenaria mercenaria; green mussels, Perna viridis [adults only]). In separate trials with larvae of A. irradians, C. virginica, and M. mercenaria, an aggressive predatory response of three strains of algal- and fish-fed P. shumwayae was observed (exception, algal-fed strain 1024C). Larval mortality resulted primarily from damage inflicted by physical attack of the flagellated cells, and secondarily from Pfiesteria toxin, as demonstrated in larval C. virginica exposed to P. shumwayae with versus without direct physical contact. Survival of adult shellfish and grazing activity depended upon the species and the cell density, strain, and nutritional history of P. shumwayae. No mortality of the four shellfish species was noted after 24 h of exposure to algal- or fish-fed P. shumwayae (strains 1024C, 1048C, and CCMP2089) in separate trials at ≤5 × 103 cells ml−1, whereas higher densities of fish-fed, but not algal-fed, populations (>7–8 × 103 cells ml−1) induced low (≤15%) but significant mortality. Adults of all four shellfish species sustained >90% mortality when exposed to fish-fed strain 270A1 (8 × 103 cells ml−1). In contrast, adult M. mercenaria and P. viridis exposed to a similar density of fish-fed strain 2172C sustained <15% mortality, and there was no mortality of A. irradians and C. virginica exposed to that strain. In mouse bioassays with tissue homogenates (adductor muscle, mantle, and whole animals) of A. irradians and M. mercenaria that had been exposed to P. shumwayae (three strains, separate trials), mice experienced several minutes of disorientation followed by recovery. Mice injected with tissue extracts from control animals fed cryptomonads showed no response. Grazing rates of adult shellfish on P. shumwayae (mean cell length ±1 standard error [S.E.], 9 ± 1 μm) generally were significantly lower when fed fish-fed (toxic) populations than when fed populations that previously had been maintained on algal prey, and grazing rates were highest with the nontoxic cryptomonad, Storeatula major (cell length 7 ± 1 μm). Abundant cysts of P. shumwayae were found in fecal strands of all shellfish species tested, and ≤45% of the feces produced viable flagellated cells when placed into favorable culture conditions. These findings were supported by a field study wherein fecal strands collected from field-collected adult shellfish (C. virginica, M. mercenaria, and ribbed mussels, Geukensia demissa) were confirmed to contain cysts of P. shumwayae, and these cysts produced fish-killing flagellated populations in standardized fish bioassays. Thus, predatory feeding by flagellated cells of P. shumwayae can adversely affect survival of larval bivalve molluscs, and grazing can be depressed when adult shellfish are fed P. shumwayae. The data suggest that P. shumwayae could affect recruitment of larval shellfish in estuaries and aquaculture facilities; shellfish can be adversely affected via reduced filtration rates; and adult shellfish may be vectors of toxic P. shumwayae when shellfish are transported from one geographic location to another.  相似文献

The pollen morphology of the taxa belonging to the generaAetheorhiza Cass.,Launaea Cass.,Reichardia Roth andSonchus L. in the Iberian Peninsula has been studied with light and electron microscopy. The pollen is 3(-4)-zonocolporate and echinolophate (without polar lacunae, but in general with prelacunae), with equatorial ridges and 15–20 lacunae: 3–4 poral, 6–8 abporal and 6–8 paraporal. Small to medium size, P × E = 19–36 × 23–42 µm; sometimes two different sizes have been found. Exine 3–9 µm thick and ornamentation microreticulate and echinate. Removal of phosphorus (P) by Ceratophyllum demersum L. and associated epiphytic periphyton was quantified by measuring the disappearance of soluble reactive P (SRP) from microcosms during 1-h in situ incubations conducted over a 1-year period. Initial P concentrations in these incubations ranged from 30 to >10,000 μg P L−1. Phosphorus removal was proportional to initial P concentrations and was weakly correlated with solar irradiance and water temperature. Removal rates (0.6–32.8 mg P m−2 d−1) and kv coefficients (0.68–1.93 h−1) from experiments run at low initial P concentrations (up to 200 μg P L−1) were comparable to results reported for other macrophytes. Removal rates from experiments run at the highest (>10,000 μg P L−1) initial P concentrations (5300 and 11,100 mg P m−2 d−1) most likely represented luxury nutrient consumption and were not thought to be sustainable long term. We were unable to determine a Vmax for P removal, suggesting that the nutrient-storage capability of the C. demersum/periphyton complex was not saturated during our short-term incubations. Based on N:P molar ratios, the marsh was P limited, while the C. demersum/periphyton complex was either N limited or in balance for N and P throughout this study. However, despite its tissue stoichiometry, the C. demersum/periphyton complex always exhibited an affinity for P. It appeared that the biochemical mechanisms, which mediate P removal, at least on a short-term basis, were more influenced by increases in ambient P levels than by tissue nutrient stoichiometry.  相似文献

Karlodinium veneficum is a common member of temperate, coastal phytoplankton assemblages that occasionally forms blooms associated with fish kills. Here, we tested the hypothesis that the cytotoxic and ichthyotoxic compounds produced by K. veneficum, karlotoxins, can have anti-grazing properties against the heterotrophic dinoflagellate, Oxyrrhis marina. The sterol composition of O. marina (>80% cholesterol) renders it sensitive to karlotoxin, and does not vary substantially when fed different algal diets even for prey that are resistant to karlotoxin. At in situ bloom concentrations (104–105 K. veneficum ml−1), grazing rates (cells ingested per Oxyrrhis h−1) on toxic K. veneficum strain CCMP 2064 were 55% that observed on the non-toxic K. veneficum strain MD5. At lower prey concentrations typical of in situ non-bloom levels (<103 cells ml−1), grazing rates (cells ingested per Oxyrrhis h−1) on toxic K. veneficum strain CCMP 2064 were 70–80% of rates on non-toxic strain MD5. Growth of O. marina was significantly suppressed when fed the toxic strain of K. veneficum. Experiments with mixed prey cultures, where non-toxic strain MD5 was fluorescently stained, showed that the presence of toxic strain CCMP 2064 inhibited grazing of O. marina on the co-occurring non-toxic strain MD5. Exogenous addition of a sub-lethal dose (100 ng ml−1) of purified karlotoxin inhibited grazing of O. marina by approximately 50% on the non-toxic K. veneficum strain MD5 or the cryptophyte S. major. These results identify karlotoxin as an anti-grazing compound for those grazers with appropriate sterol composition (i.e., desmethyl sterols). This strategy is likely to be an important mechanism whereby growth of K. veneficum is uncoupled from losses due to grazing, allowing it to form ichthyotoxic blooms in situ.  相似文献

In this study we investigated the ability of Chara intermedia to acclimate to different irradiances (i.e. “low-light” (LL): 20–30 μmol photons m−2 s−1 and “high-light” (HL): 180–200 μmol photons m−2 s−1) and light qualities (white, yellow and green), using morphological, photosynthesis, chlorophyll fluorescence and pigment analysis.Relative growth rates increased with increasing irradiance from 0.016 ± 0.003 (LL) to 0.024 ± 0.005 (HL) g g−1 d−1 fresh weight and were independent of light quality. A growth-based branch orientation towards high-light functioning as a mechanism to protect the plant from excessive light was confirmed. It was shown that the receptor responsible for the morphological reaction is sensitive to blue-light.C. intermedia showed higher oxygen evolution (up to 10.5 (HL) vs. 4.5 (LL) nmol O2 mg Chl−1 s−1), photochemical and energy-dependent Chl fluorescence quenching and a lower Fv/Fm after acclimation to HL. With respect to qP, the acclimation of the photosynthetic apparatus depended on light quality and needed the blue part of the spectrum for full development. In addition, pigment composition was influenced by light and the Chl a/Car and Antheraxanthin (A) + Zeaxanthin (Z)/Violaxanthin (V) + A + Z (DES) ratios revealed the expected acclimation behaviour in favour of carotenoid protection under HL (i.e. decrease of Chl a/Car from 3.41 ± 0.48 to 2.30 ± 0.35 and increase of DES from 0.39 ± 0.05 to 0.87 ± 0.03), while the Chl a/Chl b ratios were not significantly affected. Furthermore it was shown that morphological light acclimation mechanisms influence the extent of the physiological modifications.  相似文献

The occurrence of Alexandrium taylori and Alexandrium peruvianum is reported for the first time in Malaysia waters. The Malaysian A. taylori isolates were pyriform in shape with a transdiameter range of 36–40 μm and a cell length range of 33–37 μm. The first apical plate (1′) was pentagonal with two distinctive anterior margins. No direct connection between 1′ and the apical pore complex was observed. The posterior sulcal plate (S.p.) was large, elongated and oblique to the right with anterior projections. The ventral pore (vp) was relatively large and situated at a confluence point of 1′, the second apical (2′) and the fourth apical (4′) plates. Cells of A. peruvianum were slightly anteriorly and posteriorly compressed. S.p. had an irregular pentagonal shape, with the anterior margin divided into 2 portions. 1′ was boomerang-shaped with a large and truncated ventral pore in the middle right margin. The anterior right margin of 1′ was straight. The sixth precingular plate (6″) was wider than long. The anterior sulcal plate (S.a.) was triangular and lacked a left portion extension. In laboratory cultures, both A. taylori and A. peruvianum produced paralytic shellfish toxins, with GTX4 and GTX6 as the predominant toxin, respectively. This is the first report of PSP toxins production for both species as well as the occurrences in Malaysia waters.  相似文献

Interspecific ecophysiological differences in response to different light environments are important to consider in regeneration behavior and forest dynamics. The diurnal changes in leaf gas exchange and chlorophyll fluorescence of two dipterocarps, Shorea leprosula (a high light-requiring) and Neobalanocarpus heimii (a low light-requiring), and a pioneer tree species (Macaranga gigantea) growing in open and gap sites were examined. In the open site, the maximum net photosynthetic rate (Pn), photosystem II (PSII) quantum yield (; F/Fm), and relative electron transport rate (r-ETR) through PSII at a given photosynthetic photon flux density (PPFD) was higher in S. leprosula and M. gigantea than in N. heimii, while non-photochemical quenching (NPQ) at a given PPFD was higher in N. heimii. The maximum values of net photosynthetic rate (Pn) in M. gigantea and S. leprosula was higher in the open site (8–11 mol m–2 s–1) than in the gap site (5 mol m–2 s–1), whereas that in N. heimii was lower in the open site (2 mol m–2 s–1) than in the gap site (4 mol m–2 s–1), indicating that N. heimii was less favorable to the open site. These data provide evidence to support the hypothesis that ecophysiological characteristics link with plants regeneration behavior and successional status. Although Pn and stomatal conductance decreased at midday in M. gigantea and S. leprosula in the open site, both r-ETR and leaf temperature remained unchanged. This indicates that stomatal closure rather than reduced photochemical capacity limited Pn in the daytime. Conversely, there was reduced r-ETR under high PPFD conditions in N. heimii in the open site, indicating reduced photochemical capacity. In the gap site, Pn increased in all leaves in the morning before exposure to direct sunlight, suggesting a relatively high use of diffuse light in the morning.  相似文献

Phytochemical investigation of the rhizome of Dioscorea dumetorum has led to the isolation by several chromatographic steps on normal and reversed phase silica gel of a new ecdysteroid, (20R)-5β,11α,20-trihydroxyecdysone (1), and two known ecdysteroids, ajugasterone C (2) and herkesterone (3). Their structures were determined by spectroscopic methods including 1D- and 2D-NMR (COSY, TOCSY, HSQC and HMBC). This is the first report on the occurrence of phytoecdysteroids in the Dioscoreaceae family. These compounds were devoid of antifungal activity against three Candida species (Candida albicans, Candida glabrata and Candida tropicalis, MIC > 200 μg/ml).  相似文献

The Delaware Inland Bays (DIB) have experienced harmful algal blooms of dinoflagellates and raphidophytes in recent years. We used quantitative polymerase chain reaction (QPCR) techniques to investigate the community dynamics of three DIB dinoflagellates (Karlodinium veneficum, Gyrodinium instriatum, and Prorocentrum minimum) and one raphidophyte (Heterosigma akashiwo) at a single site in the DIB (IR-32) in summer 2006 relative to salinity, temperature and nutrient concentrations. We also carried out complementary laboratory culture studies. New primers and probes were developed and validated for the 18S rRNA genes in the three dinoflagellates. K. veneficum, H. akashiwo, and G. instriatum were present in almost all samples throughout the summer of 2006. In contrast, P. minimum was undetectable in late June through September, when temperatures ranged from 20 to 30 °C (average 25.7 °C). Dissolved nutrients ranged from 0.1 to 2.8 μM PO43− (median = 0.3 μM), 0.7–30.2 μM NOx (median = 12.9 μM), and 0–19.4 μM NH4+ (median = 0.7 μM). Dissolved N:P ratios covered a wide range from 2.6 to 177, with a median of 40. There was considerable variability in occurrence of the four species versus nutrients, but in general P. minimum and H. akashiwo were most abundant at higher (>40) N:P ratios and dissolved nitrogen concentrations, while K. veneficum and G. instriatum were most abundant at low dissolved N:P ratios (<20) and dissolved nitrogen concentrations < 10 μM. The semi-continuous laboratory competition experiment used mixed cultures of K. veneficum, P. minimum, and H. akashiwo grown at dissolved N:P ratios of 5, 16, and 25. At an N:P of 16 and 25 P. minimum was the dominant alga at the end of the experiment, even at a temperature that was much higher than that at which this alga was found to bloom in the field (27 °C). P. minimum and H. akashiwo had highest densities in the N:P of 25. K. veneficum grew equally well at all three N:P ratios, and was co-dominant at times at an N:P of 5. H. akashiwo had the lowest densities of the three algae in the laboratory experiment. Laboratory and field results showed both interesting similarities and significant differences in the influences of important environmental factors on competition between these harmful algal species, suggesting the need for more work to fully understand HAB dynamics in the DIB.  相似文献

The diatom Eucampia zodiacus Ehrenberg is one of the harmful diatoms which indirectly cause, through nutrient depletion, discoloration of Porphyra thalli. The effect of temperature on light-limited growth of E. zodiacus was examined at 13 irradiance levels (5–350 μmol m−2 s−1) in combination with five temperatures (8.0–25.0 °C). The results showed that all the parameters of growth-irradiance curves, such as the maximum growth rate (μm), half saturation constant (Ks), threshold value of irradiance (I0) and saturation irradiance for growth (S), increased with increasing temperature. On the basis of the relationship between temperature and growth-irradiance curves and seasonal fluctuation of the light environment in Harima-Nada, the effect of irradiance on the population dynamics of E. zodiacus during the period from October to March was evaluated using two indices, depth of the threshold irradiance for growth (Dt) and depth where a half of its maximum growth rate is attained (Dk). Dt and Dk remained almost stable from October to December, but gradually increased in early March. This indicates that the range of depth at which E. zodiacus was able to grow increased markedly in early spring when E. zodiacus blooms in Harima-Nada. As the vegetative cells of E. zodiacus tend to distribute in relatively deeper water layers, where growth is limited by irradiance, the increase in the depth range over which E. zodiacus is able to grow is concluded to be an important factor allowing development of its blooms.  相似文献

A family of 10 competing, unstructured models has been developed to model cell growth, substrate consumption, and product formation of the pyruvate producing strain Escherichia coli YYC202 ldhA::Kan strain used in fed-batch processes. The strain is completely blocked in its ability to convert pyruvate into acetyl-CoA or acetate (using glucose as the carbon source) resulting in an acetate auxotrophy during growth in glucose minimal medium. Parameter estimation was carried out using data from fed-batch fermentation performed at constant glucose feed rates of qVG=10 mL h–1. Acetate was fed according to the previously developed feeding strategy. While the model identification was realized by least-square fit, the model discrimination was based on the model selection criterion (MSC). The validation of model parameters was performed applying data from two different fed-batch experiments with glucose feed rate qVG=20 and 30 mL h–1, respectively. Karenia brevis (formerly Gymnodinium breve) is a toxic marine dinoflagellate generally restricted to the Gulf of Mexico and is the main causative organism in fish kills, shellfish intoxications and respiratory distress in humans following bloom events. K. mikimotoi is a morphologically similar co-occurring species which is toxic in other parts of the world oceans, but has not been recognized as a major contributor in toxicity of blooms within the Gulf of Mexico. Recently there has been increasing evidence of the simultaneous production of a variety of bioactive compounds in addition to potent neurotoxins (brevetoxin) in Karenia brevis isolates. These compounds are potentially ichthyotoxic and have been shown to cause hemolysis in several bioassays [Eshbach, E., Scharsack, J., John, U., Medlin, L., 2001. Current monitoring methods include assays which are highly sensitive in brevetoxin detection and yet may not target other harmful compounds.By adapting protocols developed by Eshbach et al. [Eshbach, E., Scharsack, J., John, U., Medlin, L., 2001. Improved erythrocyte lysis assay in microtitre plates for the sensitive detection and efficient measurement of haemolytic compounds from ichthyotoxic algae. J. Appl. Toxicol. 21, 513–519], Red drum (Sciaenops ocellatus) erythrocytes were used to create a modified bioassay to detect hemolytic activity of crude algal extracts. Red drum was selected because it is endemic to coastal areas throughout the Gulf of Mexico and is sensitive to Karenia blooms, and thus makes this species a valid ecological target. Preliminary data has shown this method is sensitive for use in assessing hemolysis induced by laboratory cultures down to levels of 1 × 103 cells mL−1. Results showed an unexpectedly high level of hemolytic activity among K. mikimotoi clones, with one Texas strain inducing significantly higher hemolysis compared to Florida K. brevis isolates. Using this approach, future research efforts will examine the difference in production of hemolytic compounds among various Karenia clones.  相似文献
