A relative biological effectiveness (RBE) not much larger than unity is usually assumed for soft x-rays (up to approximately 50 keV) that are applied in diagnostic radiology such as mammography, in conventional radiotherapy and in novel radiotherapy approaches such as x-ray phototherapy. On the other hand, there have been recent claims of an RBE of more than 3 for mammography and respective conventional x-rays. Detailed data on the RBE of soft x-rays, however, are scarce. The aim of the present study was to determine the effect of low-energy x-rays on chromosomal damage in vitro, in terms of micronucleus induction. Experiments were performed with 25 kV x-rays and a 200 kV x-ray reference source. The studies were carried out on primary human epidermal keratinocytes (HEKn), human fibroblasts (HFIB) and NIH/3T3 mouse fibroblasts. Micronucleus (MN) induction was assayed after in vitro irradiation with doses ranging from 1 to 5.2 Gy. Compared to the effect of 200 kV x-rays, 25 kV x-rays resulted in moderately increased chromosomal damage in all cell lines studied. This increase was observed for the percentage of binucleated (BN) cells with micronuclei as well as for the number of micronuclei per BN cell. Moreover, the increased number of micronuclei per micronucleated BN cell in human keratinocytes and 3T3 mouse fibroblasts suggests that soft x-rays induce a different quality of damage. For all cell lines studied the analysis of micronucleus induction by 25 kV soft x-rays compared to 200 kV x-rays resulted in an RBE value of about 1.3. This indicates a somewhat enhanced potential of soft x-rays for induction of genetic effects.  相似文献

The main aim of this study was to investigate occupational exposure to electric fields, and current densities and contact currents associated with tasks at air-insulated 110 kV substations and analyze if the action value of EU Directive 2004/40/EC was exceeded. Four workers volunteered to simulate the following tasks: Task (A) maintenance of an operating device of a disconnector at ground or floor level, Task (B) maintenance of an operating device of a circuit breaker at ground or floor level, Task (C) breaker head maintenance from a man hoist, and Task (D) maintenance of an operating device of a circuit breaker from a service platform. The highest maximum average current density in the neck was 1.8 mA/m(2) (calculated internal electric field 9.0-18.0 mV/m) and the highest contact current was 79.4 μA. All measured values at substations were lower than the limit value (10 mA/m(2)) of the EU Directive 2004/40/EC and the 2010 basic restrictions (0.1 and 0.8 V/m for central nervous system tissues of the head, and all tissues of the head and body, respectively) of the International Commission on Non-Ionizing Radiation Protection (ICNIRP).  相似文献

Low energy x-rays (Eph 50 keV) are widely used in diagnostic radiology and radiotherapy. However, data on their relative biological effectiveness (RBE) are scarce. Of particular importance for risk estimation are the RBE values of x-rays in the range which is commonly used in mammography (10–30 keV). We have determined clonogenic survival after low-energy x-ray irradiation for three cell lines: primary human epidermal keratinocytes (HEKn), mouse fibroblasts (NIH/3T3) and Chinese hamster fibroblasts (V79). Experiments were performed with a 25 kV x-ray tube and compared to 200 kV x-rays as a reference. Compared to the effect of 200 kV x-rays, irradiation with 25 kV x-rays resulted in a decreased survival rate in the murine fibroblasts, but not in the human epithelial cell line. The RBE value was calculated for 10% surviving fraction. For HEKn cells, RBE was 1.33±0.27, for NIH/3T3 cells 1.25±0.07 and for V79 cells 1.10±0.09, respectively. No consistently increased RBE was observed in the various cell lines. Nevertheless, a potential of increased cytogenetic changes has to be considered for risk estimation of low-energy x-rays.  相似文献

Experiments using the alkaline comet assay, which measures all single-strand breaks regardless of their origin, were performed to evaluate the biological effectiveness of photons with different energies in causing these breaks. The aim was to measure human lymphocytes directly for DNA damage and subsequent repair kinetics induced by mammography 29 kV X rays relative to 220 kV X rays, 137Cs gamma rays and 60Co gamma rays. The level of DNA damage, predominantly due to single-strand breaks, was computed as the Olive tail moment or percentage DNA in the tail for different air kerma doses (0.5, 0.75, 1, 1.5, 2 and 3 Gy). Fifty cells were analyzed per slide with a semiautomatic imaging system. Data from five independent experiments were transformed to natural logarithms and fitted using a multiple linear regression analysis. Irradiations with the different photon energies were performed simultaneously for each experiment to minimize interexperimental variation. Blood from only one male and one female was used. The interexperimental variation and the influence of donor gender were negligible. In addition, repair kinetics and residual DNA damage after exposure to a dose of 3 Gy were evaluated in three independent experiments for different repair times (10, 20, 30 and 60 min). Data for the fraction of remaining damage were fitted to the simple function F(d) = A/(t + A), where F(d) is the fraction of remaining damage, t is the time allowed for repair, and A (the only fit parameter) is the repair half-time. It was found that the comet assay data did not indicate any difference in the initial radiation damage produced by 29 kV X rays relative to the reference radiation types, 220 kV X rays and the gamma rays of 137Cs and 60Co, either for the total dose range or in the low-dose range. These results are, with some restrictions, consistent with physical examinations and predictions concerning, for example, the assessment of the possible difference in effectiveness in causing strand breaks between mammography X rays and conventional (150-250 kV) X rays, indicating that differences in biological effects must arise through downstream processing of the damage.  相似文献

This investigation studied the current densities in the neck and total contact currents in occupational exposure at 400 kV substations and power lines. Eight voluntary workers simulated their normal work tasks using the helmet–mask measuring system. In all, 151 work tasks with induced current measurements were made. Work situations were: tasks in 400 kV substations, tasks in 400–110 kV towers and the cutting of vegetation under 400 kV power lines. The average current density in the neck was estimated from the current induced in the helmet. The calculated maximum average current densities in the neck varied from 1.5 to 6.4 mA/m2 and the maximum total contact currents from 66.8 to 458.4 µA. The study shows that the maximum average current densities and the total contact currents (caused by electric field) in occupational exposure at 400 kV substations and power lines does not exceed the limit and action values (10 mA/m2 and 1 mA) of the new EU‐directive 2004/40/EC (live‐line bare‐hand works excluded). The amplitude contrast of frozen-hydrated biological samples was measured using the bacterial flagellar filament embedded in vitreous ice at an accelerating voltage of 300kV. From the mean radial amplitude spectra of overfocused images, amplitude contrast was estimated to be 6.9+/-1.9% and 2.7+/-1.0% of the whole contrast at the low spatial frequency range with and without energy filtering, respectively, and that of the carbon film to be 9.5+/-2.0% and 5.8+/-1.8%. Energy filtering effectively doubled the signal-to-noise ratio in the images of frozen-hydrated filaments, and substantially improved intensity data statistics of layer lines up to at least approximately 25A resolution in their Fourier transforms. It also markedly improved inter-particle fitting phase residuals of averaged data at resolutions up to approximately 15A. Using the energy filtered data recorded on a new high-performance, lens-coupled CCD camera the three-dimensional map of the flagellar filament was calculated at 8A by applying the amplitude contrast of 6.9%. The map and its mean radial density distribution validated the obtained value of the amplitude contrast.  相似文献

Neoplastic transformation of human CGL1-hybrid cells was examined after exposure to 29 kV x-rays (mammography x-rays) and conventional 220 kV x-rays. The study was designed to repeat, under well-defined irradiation and culture conditions, an earlier investigation by Frankenberg et al. (Radiat Res, 2002), and to assess the validity of the high RBE values of 29 kV x-rays that had been reported. The experiments with the two types of x-rays were performed simultaneously and shared the same controls. The transformation yields with both radiation qualities were fitted to the linear-quadratic dependence on absorbed dose, and a corresponding analysis was performed for the data earlier obtained by Frankenberg et al. The transformation yields in the present study exceed those in the earlier investigation substantially, and it appears that the difference reflects inadequate feeding conditions of the cell cultures in the early experiments. The standard error bands of the dose response curves are derived and are seen to be considerably more narrow in the present results. The lowest dose of the 29 kV x-rays was 1 Gy in both studies, and at this dose the RBE vs. the conventional x-rays has now been found to be 2 with a 95% confidence interval of 1.4-2.6. The previous result was about 3.2, but the 95% confidence is very broad for these data. The estimated limit at low doses is 3.4 in the present experiments with a confidence interval that extends from less than 2 to large values.  相似文献

The aim of the study was to analyze all values of electric currents from measured periods while performing tasks in forest clearing. The objective was also to choose and analyze measurement cases, where current measurements successfully lasted the entire work period (about 30 min). Two forestry workers volunteered to perform four forest clearing tasks under 400 kV power lines. The sampling frequency of the current measurements was 1 sample/s. The maximum values of the current densities were 1.0–1.2 mA/m2 (calculated internal EFs 5.0–12.0 mV/m), and the average values were 0.2–0.4 mA/m2. The highest contact current was 167.4 μA. All measured values during forest clearing tasks were lower than basic restrictions (0.1 V/m and 0.8 V/m) of the International Commission on Non‐Ionizing Radiation Protection. Summary The frequency of dicentric chromosomes induced by the irradiation of human lymphocytes inG 0 phase was determined with hard (150 kV) and soft (30 kV) X-rays. When a linear-quadratic dose-effect relation was used to fit the experimental data, a significant linear contribution was found for 30 kV X-rays. The RBE of 30 kV compared with 150 kV X-rays approaches the value 3 at a 30 kV X-ray dose of 20 rad and decreases with increasing dose. From the results it may be concluded that chromatids with primary breaks, undergoing second order reactions and thus forming dicentric chromosomes, are produced by traversals of low-energy electrons through chromatin material.  相似文献

The effectiveness of 14.1 MeV neutrons relative to 200 kV X-rays for the induction of the various kinds of dumpy mutation in mature sperm of Drosophila melanogaster was investigated. The estimated RBE values are: 0.52 for all complete mutations; 0.64 for the (olv, ov) types; 0.33 for the (ol, lv, o, v, c) types; 0.33 for all fractional mutations. These data lend support to the thesis that (1) complete dumpy mutations of the olv and ov types are more frequently associated with chromosomal aberrations than those of the ol, lv, o, v and c types, and (2) fractional mutations and complete mutations of the (ol, lv, o, v, c) types are most probably point mutational events.  相似文献

In a specific case, the magnetic field generated in a building by a nearby power line is usually easy to calculate, although the accuracy of these calculations is sensitive to the quality of source information. To be able to study public health dimensions of magnetic field exposure (e.g., risk of cancer), it is necessary to evaluate the size and exposure of the population at risk. Relatively little quantitative information on public exposure to power-frequency magnetic fields of high-voltage power lines is available. This report describes residential exposure to magnetic fields from 110 kV, 220 kV, and 400 kV power lines in Finland at the national level, including 90% of the total line length in 1989. A geographical information system (GIS) was used to identify the buildings located near the power lines. After determining the distances between the lines and the buildings, historical data on load currents of these lines were used to calculate the magnetic fields. The residential magnetic field histories were then linked to the residents by means of a computerized central population register. The data obtained on personal exposure have also been utilized in a nationwide epidemiological study on magnetic field exposure of power lines and risk of cancer. The methods of exposure assessment and results of the number of buildings near 110 kV, 220 kV, and 400 kV power lines, their average annual magnetic fields, and personal exposure to magnetic fields from these lines are described. We found that 15,600 residents lived in an average residential magnetic field ≥0.1 μT caused by power lines in 1989. The number of these residents increased fivefold during 1970-1989. We estimated that 0.3% of the population was exposed in their residences to an annual average magnetic flux density from 110 kV, 220 kV, and 400 kV power lines higher than 0.1 μT, the level that the background magnetic flux density in general does not exceed in Finnish homes. Thus, the problem of magnetic field exposure generated by high-voltage lines concerns only a relatively small fraction of the total population in Finland. However, the size and exposure of the population at risk remain somewhat arbitrary in practical multisource situations, as the biological interaction mechanism, the concept of harmful dose, and, in particular, the significance of the duration of exposure are unknown. © 1995 Wiley-Liss, Inc.  相似文献

Peripheral blood lymphocyte culture system was used to construct reference dose-response curves for 60Co gamma-rays and 110 kV X-ray-induced chromosome aberrations at 6 dose points ranging from 0.25 to 4.0 Gy. Qualitative and quantitative differences between these two types of radiation for the yield of induced aberrations and their distribution pattern were analysed. Experimental data of aberration yields were compared after fitting them to five different dose-response models. The yields of chromosome aberrations in particular dicentrics, gave a good fit to linear-quadratic besides linear and power models. In this model, single-track events predominated over double-track events for both the qualities of radiation used. The pattern of distribution was mainly Poisson for dicentrics but gave a conflicting result for acentrics which was in excess.  相似文献
