Intraerythrocytic parasites of Plasmodium vinckei and Plasmodium berghei were separated according to their developmental stages using discontinuous Percoll gradients. Contaminating nucleated blood cells such as leukocytes were removed by elutriation centrifugation. The stages were unequivocally identified in smears using a newly developed DNA-specific staining procedure with mithramycin and fluorescence microscopy. This stain can also be used to detect parasites in human blood of very low parasitemias. The combination of methods described has many possible applications in immunologic and biochemical parasite research.  相似文献

The Australian platypus, Ornithorhynchus anatinus, is one of three extant genera of the order monotremata. Given the divergent evolutionary lineage of monotremes in relation to more commonly studied animals, it was of interest to determine first, whether platypuses possess endogenous biological pacemakers and, second, general parameters of aquatic activity rhythms under artificial and natural light–dark (LD) cycles. Using a novel recording device, aquatic activity rhythms were measured in three platypuses: a paired male and female studied together, and a single female studied in isolation from other platypuses. Under a constant photic environment, some evidence was found for persistent and free-running rhythmicity, indicating the presence of an endogenous circadian pacemaker in the platypus. Under artificial LD cycles the paired animals exhibited a nocturnal pattern of entrainment, although in the single female considerable variability in entrained phase-relations was found under natural LD cycles. Evidence for a circadian pacemaker in the hypothalamic region of platypuses is also discussed.  相似文献

Cytomegaly and intranuclear inclusion bodies were observed in the renal collecting duct epithelium in three of four wild caught platypuses (Ornithorhynchus anatinus) from New South Wales using light and electron microscopy during routine pathological studies. Non-enveloped, spherical virions measuring about 80 nm in diameter were present in the nucleus and cytoplasm of affected cells as well as in the lumen of the renal tubule. A single enveloped virion measuring about 150 nm in diameter was found. There was no serological evidence of infection with cytomegalovirus (AD169 antigen) or adenovirus (mammalian and avian group antigens) in any of the platypuses. Although the identity of the virus was not confirmed, it was probably an adenovirus based on morphological grounds. The infection appeared to have little effect on the host.  相似文献

The platypus, Ornithorhynchus anatinus is a unique, iconic mammal endemic to Australia. Despite being listed as ‘common’ throughout its range, platypus abundance is poorly understood. Dependence on aquatic habitats in Australia renders this species potentially vulnerable to a variety of processes including drought, climate change and habitat loss. To assist with understanding population processes, 180 individuals from Tasmania and Victoria were characterised across thirteen microsatellite loci. Large genetic differences were evident between Tasmanian and Victorian O. anatinus. Within Tasmania, high levels of allelic diversity were detected with genetic differentiation identified among some populations. Similarly, allelic diversity was high within Victorian platypuses, along with significant genetic differentiation among populations. The large genetic differences found between Tasmanian and mainland platypuses indicate long-term isolation and it is likely that the lack of past/present catchment connectedness contributes to differentiation found between populations within these regions. Understanding patterns of genetic differentiation within and between catchments will help guide future conservation management decisions for platypus.  相似文献

We investigated the diving behaviour, the time allocation of the dive cycle and the behavioural aerobic dive limit (ADL) of platypuses (Ornithorhynchus anatinus) living at a sub-alpine Tasmanian lake. Individual platypuses were equipped with combined data logger-transmitter packages measuring dive depth. Mean dive duration was 31.3 s with 72% of all dives lasting between 18 and 40 s. Mean surface duration was 10.1 s. Mean dive depth was 1.28 m with a maximum of 8.77 m. Platypuses performed up to 1600 dives per foraging trip with a mean of 75 dives per hour. ADL was estimated by consideration of post-dive surface intervals vs. dive durations. Only 15% of all dives were found to exceed the estimated ADL of 40 s, indicating mainly aerobic diving in the species. Foraging platypuses followed a model of optimised recovery time, the optimal breathing theory. Total bottom duration or total foraging duration per day is proposed as a useful indicator of foraging efficiency and hence habitat quality in the species.  相似文献

The fungal disease mucormycosis has affected Tasmanian platypuses for nearly three decades. We investigated the influences of mucormycosis on the hematologic, plasma biochemical, and other indicators of health in free-living platypuses across 18 Tasmanian river catchments. Live trapping enabled sampling of 161 (apparently) healthy and six ulcerated, mucormycosis-affected platypuses in 75 rivers and streams between January 2008 and June 2009. There were no obvious differences in any hematologic or biochemical measures between healthy and mucormycosis-affected platypuses. However, multivariate analysis revealed that ulceration was associated with living at higher altitudes, low tail fat content (high tail fat index), and low trypanosome load. There was evidence of overall lymphocytosis and monocytosis in animals from areas currently affected by mucormycosis, which suggests that some level of immune response to the introduced fungus is now widespread in disease-affected catchments. Animals from currently, historically, and possibly disease-affected catchments had lower neutrophil counts, mean cell volumes, plasma alkaline phosphatase, alanine aminotransferase, and aspartate aminotransferase levels, and higher plasma gamma-glutamyl transpeptidase and platelet counts compared to animals from catchments with no evidence of infection. Reference intervals were generated for all hematologic and biochemical measurements. Since this is the most comprehensive, systematic, and large-scale assessment of the health of the Tasmanian platypus to date, these references intervals should act as the standard against which future studies of platypuses in Tasmania should be compared.  相似文献

Pathological findings associated with helminth and arthropod parasites of four wild platypuses (Ornithorhynchus anatinus) are described. Fourth-stage larval rhabditoid nematodes (?Cylindrocorporidae) found partially embedded in the stratum corneum were associated with mild acanthosis, orthokeratotic hyperkeratosis and inconstant subacute dermatits. A filarioid nematode (?Lemdaninae) was found in hypodermal lymphoid tissue. A larval trichostrongyloid nematode was found in the dermis, with no evidence of host response. An undescribed species of trombiculid mite ("chigger") was found in the pelage and attached to superficial stratum corneum. Focal inclusions of eosinophilic material within stratum corneum, interpreted as mite gel saliva produced during formation of the "stylostome," were also observed. The tick Ixodes ornithorhynchi punctured the epidermis causing adnexal trauma, dermal haemorrhage and chronic active dermatitis. The digenean Mehlisia ornithorhynchi was present in the small intestine of one animal and was associated with mild catarrhal enteritis.  相似文献

Giardia sp. was found in the white stork (Ciconia ciconia) in The Netherlands for the first time. The Giardia sp. trophozoites that were found in the feces of a 6-wk-old white stork, were examined by light microscopy. The parasites closely resembled Giardia ardeae that had been isolated by others from several species of wading birds belonging to the order Ciconiiformes, sharing a deeply notched adhesive disk, a single caudal flagellum, and a single round median body. Serologically, the parasites did not react with anti-Giardia intestinalis monoclonal antibodies. Although no signs of intestinal disease were observed in the stork chick, the presence of parasites in all stages of development and the huge number of parasites show that the stork chick was experiencing an active infection with G. ardeae type parasites.  相似文献

DAPI is a fluorescent dye which appears to complex specifically with DNA. We have used this probe to detect and identify malarial infections by fluorescence microscopy. Experiments were conducted using Plasmodium berghei yoeli--infected mouse blood, P. lophurae--infected duck blood, and P. vivax--infected human blood. Infected avian blood was used to detect parasites within nucleated erythrocytes. Control blood smears from uninfected hosts revealed fluorescence only in the leukocytes of mammalian blood or in nuclei of leukocytes and erythrocytes of avian blood. Cytoplasmic staining of red blood cells was absent in all controls. In contrast, the cytoplasm of infected red blood cells was stippled with fluorescence centers. Ring forms, trophozoites, segmenters, and merozoites frequently were observed. This simple procedure can be applied directly to routine clinical analysis, as well as experimental procedures, DAPI can also be used to stain other parasites, including nuclei in microfilariae.  相似文献

We measured the energy requirements of platypuses foraging, diving and resting in a swim tank using flow-through respirometry. Also, walking metabolic rates were obtained from platypuses walking on a conventional treadmill. Energy requirements while foraging were found to depend on water temperature, body weight and dive duration and averaged 8.48 W kg(-1). Rates for subsurface swimming averaged 6.71 W kg(-1). Minimal cost of transport for subsurface swimming platypuses was 1.85 J N(-1)m(-1) at a speed of 0.4 m s(-1). Aerobic dive limit of the platypus amounted to 59 s. Metabolic rate of platypuses resting on the water surface was minimal with 3.91 W kg(-1) while minimal RMR on land was 2.08 W kg(-1). The metabolic rate for walking was 8.80 W kg(-1) and 10.56 W kg(-1) at speeds of 0.2 m s(-1) and 0.3 m s(-1), respectively. A formula was derived, which allows prediction of power requirements of platypuses in the wild from measurements of body weight, dive duration and water temperature. Platypuses were found to expend energy at only half the rate of semiaquatic eutherians of comparable body sizes during both walking and diving. However, costs of transport at optimal speed were in line with findings for eutherians. These patterns suggest that underwater locomotion of semiaquatic mammals have converged on very similar efficiencies despite differences in phylogeny and locomotor mode.  相似文献

Tissues from barred tiger salamanders (Ambystoma tigrinum mavortium), Great Plains toads (Bufo cognatus) and New Mexico spadefoots (Spea multiplicata) collected from 16 playa wetlands in Texas during 1999 and 2000 were examined by light microscopy. Digenean cysts were primarily distributed subcutaneously throughout the specimens and occasionally coelomic invasion was noted. The parasites within the cysts were 1.5-2 mm in diameter, with a thin (c. 10 microm wide) eosinophilic-staining tegument, two suckers (oral and ventral), posteriorly located primordial genitalia and paired digestive caeca. These digeneans were identified as the metacercariae of Clinostomum attenuatum. This is the first record of Clinostomum attenuatum in these amphibian species.  相似文献

Twenty-nine (64.4%) of 45 reindeer, Rangifer tarandus, examined over a two-year period were infected with trypanosomes. Trypomastigotes and dividing epimastigotes were found in the blood of fawns, cows, and bulls. Morphometric analysis of bloodstream trypomastigotes from reindeer and comparison of these parasites with similar stages of trypanosomes from elk, mule deer, and white-tailed deer from the contiguous United States proved them conspecific; the trypanosomes from these members of the Cervidae are identified as Trypanosoma cervi Kingston & Morton, 1975. This is the first report of trypanosomes from reindeer. No pathogenic effects are known to be caused by these parasites.  相似文献

  1. The platypus is a cryptic mammal that inhabits freshwater streams and rivers of eastern Australia. Tracking the movements of wild platypuses has been notoriously difficult due to the animals' morphology and methodological limitations. Knowledge of fine‐scale movements and interactions among individuals remain particularly poorly understood, as do responses to changes in hydrology.
  2. We tracked movements of 15 platypuses (six females, nine males) downstream of the Jindabyne Dam on the Snowy River, using externally attached acoustic transmitters (September–November 2017), to assess spatio‐temporal activity patterns among individuals and changes in movement and activity before and after an environmental flushing flow. As the study took place during the breeding season, we expected to observe overlap in area of activity among males and females, but not among males due to increased territoriality during these months. We also anticipated that a large flow event would impact their activity and foraging behaviour, possibly displacing platypuses downstream.
  3. Overlaps in area of activity and temporal co‐occurrence within a pool varied among individuals, with two resident males exhibiting some spatial overlap of activity and varying temporal co‐occurrence, despite tracking during the breeding season. All six tracked females were captured in the same pool and appeared to be residents, possibly highlighting preferences for certain habitats during the breeding months.
  4. We found no evidence that the movements of adult platypuses were affected by an environmental flushing flow, with no significant changes to area of activity, number of detections, or daily range of movements. However, foraging duration increased in the week after the flow, possibly associated with increased prey availability.
  5. These findings suggest that territoriality between males during and after the breeding season may depend on platypus density and resource availability and that pools with high resource availability may support several breeding females.

ABSTRACT. During routine histopathology of 180 juvenile hard clams, Mercenaria mercenaria , from a site in Virginia, USA, in 2007, we discovered a single individual heavily infected with a parasite resembling a haplosporidian, some members of which cause lethal bivalve diseases. Scanning electron microscopy of spores and sequencing of small subunit ribosomal DNA confirmed a new species: Minchinia mercenariae n. sp. Further sampling of clams at the site found prevalences up to 38% using polymerase chain reaction (PCR). No parasites were found in routine histological screening of the same individuals, but re-examination of clams judged positive by in situ hybridization (ISH) revealed very faintly staining plasmodia. No unusual mortalities have occurred among the sampled groups. Analysis of clams from Massachusetts to Florida by PCR failed to detect the parasite, but a haplosporidian found in a clam from New Jersey in 2001 was subsequently identified by ISH as M. mercenariae . No other haplosporidians have been reported in thousands of hard clams from the US east coast examined histologically since the mid-1980s. The discovery underscores critical questions about how to assess the risks associated with parasites in groups known to be lethal, but that themselves are not considered a problem.  相似文献

The present work demonstrates the expression of receptors for the Fc portion of rodent Ig by the murine parasite Trypanosoma musculi. By using a rosette assay adapted to the parasite morphology and by flow cytometry analysis, three distinct receptors were identified. A receptor binding rabbit or rat polyclonal IgG and mouse monoclonal IgG1, IgG2a, and IgG2b was found on parasites purified from the blood and the peritoneal cavity of infected mice and on parasites maintained in culture conditions. This IgG receptor was degraded by pepsin. A separate receptor, binding only mouse monoclonal IgG3 was observed on cultured parasites. A receptor binding rabbit, rat, and mouse IgM was found on cultured and peritoneal parasites, but not on blood parasites. This receptor did not bind IgG or IgA but it bound mouse and rat IgE as well as IgM. It was degraded by trypsin. IgG and IgM/IgE receptors were co-expressed on single parasites. They were not of host origin but synthesized by trypanosomes as shown by reexpression in vitro after proteolytic degradation. Their expression was variable with the development of trypanosomes both in vitro and in vivo.  相似文献

Mature exoerythrocytic forms (EEF) of Plasmodium falciparum from the chimpanzee were examined by light- and transmission electron microscopy from a liver biopsy taken on Day 6 after sporozoite inoculation. Infectivity of the sporozoites obtained from whole mosquitoes which were membrane fed on cultured gametocytes was about 4-6%. In comparison, salivary gland sporozoites added to human hepatocytes in vitro had only a developmental percentage of 0.02 to 0.05% at Day 5. The EEF found in the liver biopsy were not all at the same stage of development. Immature compact parasites were seen simultaneously with stages with fully formed merozoites, indicating a rapid final maturation or asynchrony. At Day 7.5, large numbers of rings were already seen in the peripheral blood, indicating a duration of the liver development of P. falciparum in the chimpanzee of about 5.5-6 days. The process of merogony at the fine structural level was comparable to that described for rodent and other primate parasites in vivo. Compared to the fine structure of EEF in vitro in cultured human hepatocytes, the parasites described here were much more advanced in development. There appeared to be some cell infiltration with collagen deposition around the intracellular parasite; however, no marked degeneration of EEF was observed.  相似文献

采用血细胞计数、光学显微及电子显微技术对二龄史氏鲟外周血细胞的数目、形态及结构进行了研究。二龄史氏鲟红细胞的数目为47.75×104个/mm3,白细胞数目为2.9万个/mm3,其中淋巴细胞所占比率最高。史氏鲟的外周血中除正常红细胞外,还有处于分裂状态及未成熟的红细胞。史氏鲟外周血中的白细胞有四种类型,分别为淋巴细胞、粒细胞、血栓细胞和单核细胞。其中粒细胞有两种,即嗜中性粒细胞和嗜酸性粒细胞。嗜中性粒细胞含有多种形状的核,其中分叶的核数目较多,粒细胞及淋巴细胞均类似于哺乳动物。对史氏鲟外周血细胞细微结构的观察显示:红细胞中具有少量的细胞器;淋巴细胞结构典型;单核细胞较粒细胞稍小且具有较多线粒体;血栓细胞具有梭形和圆形两种,胞质较少,其中梭形的血栓细胞胞质几乎透明;对粒细胞的颗粒按照形状和电子密度进行了分类。  相似文献

A survey was carried out on the prevalence of blood parasites in birds in the Mascarene Islands. Smears from 357 birds of 25 species in 12 families were examined, of which 150 (42%) were found to harbour blood parasites. The most common parasites were Leucocytozoon ; a new species, L. zosteropis , is described from the Grey White-eye Zosterops borbonica mauritiana. This parasite was observed in smears from 68 birds of three species: Z. borbonica, Z. chloronothos and Z. olivacea. Other species of Leucocytozoon identified were L. fringillinarum from fodies, sparrows and a bulbul and L. marchouxi from two doves.
Haemoproteus was found only in domestic pigeons Columba livia and identified as H. columbae. Plasmodium relictum, P. vaughani and an unidentified species with elongate gametocytes were found in Zosterops , and Plasmodium sp. of low infection observed in other hosts. Trypanosoma mayae is redescribed from the House Sparrow Passer domesticus and the Mauritius Fody Foudia rubra , and considered to be a valid species. A new species of trypanosome, Trypanosoma phedinae , is described from the Malagasy Swallow Phedina b. borbonica. Other birds were found to harbour low infections of unidentified species of trypanosomes. A small number of birds were infected with Atoxoplasma , haemogregarines and Rickettsia-like organisms. An unidentified organism with a predilection for eosinophils was observed in several Mascarene Swiftlets Collocalia francica.
The results are discussed in relation to the possible effects of the parasites on the birds of the Mascarene Islands and comparisons made with the results of similar surveys on other Indian Ocean Islands.  相似文献
