The major β-1,4-endoglucanase (EG) of the thermophilic actinomycete, Thermomonospora curvata , contributed over 80% of the total EG activity recovered from cell-free culture fluid after growth on cellulose. The enzyme was purified to electrophoretic homogeneity by ammonium sulphate precipitation, ion-exchange chromatography and size exclusion HPLC. This monomeric enzyme had a specific activity of 750 IU mg−1 when assayed with 2.5% (w/v) carboxymethyl cellulose (CMC) at 70°C, pH 6.0. Highest activity was observed on CMC with a degree of polymerization of 3200. The EG was stable for 48 h at 60°C, pH 6.0 and had a half-life of 30 min at 80°C; temperature and pH optima were 70–73°C and 6.0–6.5, respectively. The mol. wt was 100000 and the pI was 4.0. The K m and V max values were 7.33 mg ml−1 and 833 μmol min−1, respectively. EG activity was inhibited by Fe2 +, Hg2 +, Ag+ and Pb2 +, and enhanced by dithiothreitol and Zn2 +. The first 12 amino acid residues at the N -terminus were: Asp-Glu-Val-Asp-Glu-Ile-Arg-Asn-Gly-Asp-Phe-Ser. Glutamic and aspartic acid constituted 24% of the total amino acid composition; no amino sugar was found.  相似文献

We investigated the combined effects of pressure, temperature, pH, initial spore concentration and the presence of nisin on the survival of spores of Bacillus coagulans. Spores were more sensitive to pressure both at lower pH and at higher treatment temperatures. An additional 1.5-log10 reduction in cfu ml-1 was observed when pH was lowered from 7.0 to 4.0 during pressurization at 400 Mpa and 45°C. A 4-log10 cfu ml-1 reduction was observed when the temperature was increased from 25°C to 70°C during pressurization at 400 Mpa. The spores were sensitive to nisin at concentrations as low as 0.2 IU ml-1. At least a 6-log10 reduction was generally achieved with pressurization at 400 Mpa in pH 4.0 buffer at 70°C for 30 min when plated in nutrient agar containing 0.8 IU ml-1 nisin.  相似文献

The solubility of carbon dioxide (CO2) in microbiological media at different pH values, water activities ( aw ), temperatures, buffering capacities and ratios of headspace to media volumes was determined by using a coulometer. Buffering capacity and ratio of headspace to media volume were shown to be the major factors influencing the solubility of CO2 in modified atmosphere model systems. The growth inhibitory effects of different dissolved CO2 concentrations (0–50 μmol ml-1) were determined for Pseudomonas fragi at 8°C and 22 C. Pseudomonas fragi was shown to be strongly affected by the CO2 concentration in the media. A carbon dioxide concentration of 40 μmol ml-1 was needed to inhibit Ps. fragi at 8°C. The importance of measuring dissolved CO2 concentrations in modified atmosphere packaging applications was shown and the coulometer proved to be an excellent tool for this purpose.  相似文献

Bifidobacterium adolescentis Int-57 isolated from human faeces produced extracellular amylase. The enzyme was purified from the culture supernatant fluids by ammonium sulphate precipitation, gel-filtration chromatography (Sephadex-G-75), ion-exchange chromatography (CM-cellulose) and FPLC. SDS-PAGE of the purified enzyme revealed a major band with an apparent molecular weight of 66 kDa. The pI was 5·2. Enzyme activity was optimal at 50°C, and at pH 5·5. The enzyme was stable at 20–40°C, and at pH 5–6 with a K m value of 2·4 g l−1 soluble starch. The activation energy was 42·3 kJ mol−1. The enzyme was significantly inhibited by maltose (10%), glucose (10%), Cu2+ (5 mmol l−1), Zn2+ (5 mmol l−1), N- bromosuccinimide (5 mmol l−1), EDTA (5 mmol l−1), I2 (1 mmol l−1) and activated by β-mercaptoethanol (10 mmol l−1).  相似文献

The effects of hyperoxia and change of temperature (range 20–30° C) on blood gases were studied in the teleost fish Piaractus mesopotamicus , native to several major river systems in Brazil. Large hyperoxia-induced increases of arterial P o2 ( P ao2) indicated that true branchial blood shunts are negligible in relation to total gill perfusion. This implies that blood gases will be influenced by ventilation rather than by shunts. Acute variations of temperature ( t ) were accompanied by changes of arterial blood pH (on the average Δ p HaΔt−1 of — 0·015 units °C−1), due mainly to alterations of P aco2: 2·4 mmHg at 20° C, 5·0 mmHg at 30° C. Concomitantly, P ao2 declined from 116 mmHg (20° C) to 89 mmHg (30° C). The data suggest that temperature-induced changes of acid-base status depend mainly on gill ventilation and that the decrease of P ao2 with higher temperature is a result of this regulation.  相似文献

Membrane-bound [NiFe]-hydrogenase from Hydrogenophaga sp. AH-24 was purified to homogeneity. The molecular weight was estimated as 100±10 kDa, consisting of two different subunits (62 and 37 kDa). The optimal pH values for H2 oxidation and evolution were 8.0 and 4.0, respectively, and the activity ratio (H2 oxidation/H2 evolution) was 1.61 × 102 at pH 7.0. The optimal temperature was 75 °C. The enzyme was quite stable under air atmosphere (the half-life of activity was c . 48 h at 4 °C), which should be important to function in the aerobic habitat of the strain. The enzyme showed high thermal stability under anaerobic conditions, which retained full activity for over 5 h at 50 °C. The activity increased up to 2.5-fold during incubation at 50 °C under H2. Using methylene blue as an electron acceptor, the kinetic constants of the purified membrane-bound homogenase (MBH) were V max=336 U mg−1, k cat=560 s−1, and k cat/ K m=2.24 × 107 M−1 s−1. The MBH exhibited prominent electron paramagnetic resonance signals originating from [3Fe–4S]+ and [4Fe–4S]+ clusters. On the other hand, signals originating from Ni of the active center were very weak, as observed in other oxygen-stable hydrogenases from aerobic H2-oxidizing bacteria. This is the first report of catalytic and biochemical characterization of the respiratory MBH from Hydrogenophaga .  相似文献

Of 22 commercial juice-processing enzyme preparations investigated, Clarex ML was found to exhibit the highest xylanase activity. The xylanase from Clarex ML was most active at 50–60°C and pH 5·0–5·5. The K m and V max values of the enzyme with oat-spelt xylan as the substrate were 8·6 mg ml−1 and 42 μmol xylose l−1 min−1, respectively. Xylobiose was the main product of enzymatic hydrolysis of xylan.  相似文献

The extracellular amylase activity from Myxococcus coralloides D was purified by Sephacryl S-200 gel filtration and by ion-exchange chromatography on DEAE-Sephadex A-25. The molecular weight was estimated by SDS-PAGE and by gel filtration as 22.5 kDa. The optimum temperature was 45°C. The pH range of high activity was between 6.5 and 8.5, with an optimum at pH 8.0. Activity was strongly inhibited by Hg2+, Zn2+, Cu2+, Ag+, Pb2+, Fe2+ and Fe3+, EDTA and glutardialdehyde, but was less affected by Ni2+ and Cd2+. Li+, Mg2+, Ba2+, Ca2+, N -ethylmaleimide, carbodiimide and phenyl methyl sulphonyl fluoride had almost no affect. The K m (45°C, pH 8) for starch hydrolysis was 2.0 times 10-3 gl-1. Comparison of the blue value-reducing curves with the time of appearance of maltose identified the enzyme produced by M. coralloides D as an α-amylase.  相似文献

Growth and proteinase production by Micrococcus sp. INIA 528 in a batch-operated laboratory fermentor were investigated, with trypticase soy broth as the basal medium for studies on optimum temperature, pH and medium composition. Maximum growth was recorded at 34°C and pH 715, whereas optimum temperature and pH for proteinase production were 31°C and pH 6.25. Maximum rate of enzyme production occurred during the late log and early stationary phases of growth. Addition of 5.0 g 1-1 yeast extract, 1.0 g 1-1 glucose, 1.0 g 1-1 MgSO4 or 1.0 g 1-1 K2HPO4 to basal medium resulted in a lower enzyme yield, but supplementation of basal medium with 2.5 g 1-1 (NH4)2SO4 increased enzyme production by 45%. A high initial biomass added to fresh broth supplemented with 2.5 g 1-1 (NH4)2SO4 only increased enzyme activity by 19%, compared to the maximum enzyme activity achieved with the standard inoculum.  相似文献

Standard metabolic rate (SMR), active metabolic rate (AMR) and critical oxygen saturation ( Scrit ) were measured in Atlantic cod Gadus morhua at 5, 10 and 15° C. The SMR was 35.5, 57.0 and 78.2 mg O2 kg−1 h−1 and Scrit was 16.5, 23.2 and 30.3%, at 5, 10 and 15° C, respectively. Previously reported SMR for Atlantic cod from arctic waters at 4° C was twice that measured at 5° C in the present study. A possible intraspecific latitudinal difference in the SMR is discussed. The AMR was 146.6, 197.9 and 200.4 mg O2 kg−1 h−1 and the critical swimming speed ( Ucrit ) was 1 6, 1.7 and 1.9 at 5, 10 and 15° C, respectively. The maximum oxygen consumption was found to be associated with exercise, rather than recovery from exercise as previously reported in another Study of Cod metabolism.  相似文献

Abstract A cellulase gene from Ruminococcus flavefaciens FD-1 had previously been cloned in Escherichia coli . The product of this gene, CelA, was purified from E. coli and characterised. This 39 kDa cellulase is antigenically related, and of similar mass, to a protein in R. flavefaciens . The enzyme has cellodextrinase activity with predominantly exo-type action. CelA activity was optimal at pH 6.5 and 41°C, and was inhibited in the presence of divalent metal cations. The K m and V max were determined as 0.68 mM and 1.89 μmol min−1 mg−1 of CelA, respectively. Cellobiose was the major end product of cellodextrin hydrolysis, and our results suggest that celluboise is competitive inhibitor of CelA.  相似文献

An improved method for purification of pectate lyases (PLI and PLII) from culture fluids of Pseudomonas fluorescens CY091 and Ps. viridiflava PJ-08-6 by using a phosphocellulose cation exchanger was described. Analysis of purified PLI and PLII by sodium dodecyl sulphate-polyacrylamide and isoelectric focusing gel electrophoresis revealed that both enzymes had been purified to near homogeneity. Optimal Ca2+ concentration required for PLI and PLII activity was determined to be 0·5 mmol l−1. The Ca2+ requirement could not be replaced by other metal cations such as Mg2+, Cu2+, Zn2+, Fe3+ and Co2+. Optimal pH for activity was determined to be between 8·5 and 9·0. The K m values for sodium polygalacturonate were 1·28 and 1·11 mg ml−1 for PLI and PLII, respectively. Both PLI and PLII were stable at low temperatures (25°C or below) for at least 1 month. However, at 37°C, the activity decreased 50% in 36 h. Optimal temperatures for activity were estimated to be 46° and 52°C for PLI and PLII, respectively. Thermal stability of both enzymes at elevated temperatures (48°C or higher) increased when CaCl2 or a positively charged molecule such as polylysine was present, but decreased when polygalacturonate or a negatively charged molecule such as heparin was present. PLI and PLII exhibit differential degrees of sensitivity to group-specific inhibitors, including iodoacetic acid and diethylpyrocarbonate. This result suggests that both sulphydryl and imidazole groups are important for the catalytic function of PLI and PLII.  相似文献

Rates of oxygen consumption were measured in the geothermal, hot spring fish, Oreochromis alcalicus grahami by stopped flow respirometry. At 37° C, routine oxygen consumption followed the allometric relationship: V o2=0.738 M 0.75, where V o2 is ml O2 h −1 and M is body mass (g). This represents a routine metabolic rate for a 10 g fish at 37° C of 0.415 ml O2 g−1 h −1 (16.4 μmol O2 g −1 h −1). Acutely increasing the temperature from 37 to 42° C significantly elevated the rate of O2 consumption from 0.739 to 0.970 ml O2 g −1 h −1 ( Q 10=l.72). In the field, O. a. grahami was observed to be 'gulping' air from the surface of the water especially in hot springs that exceeded 40° C. O. a. grahami may utilize aerial respiration when O2 requirements are high.  相似文献

At 14° C, standard metabolic rate (75·1 mg O2 h−1 kg−1), routine metabolic rate (108.8 mg O2 h−1 kg−1), active metabolic rate ( c . 380 mg O2 h−1 kg−1), critical swimming speed (Ucrit 1·7 BL s−1), heart rate 47 min−1), dorsal aortic pressure (3·2 kPa) and ventilation frequency (63 min−1) for triploid brown trout Salmo trutta were within the ranges reported for diploid brown trout and other salmonids at the same temperature. During prolonged swimming ( c . 80% U crit), cardiac output increased by 2·3-fold due to increases in heart rate (1·8-fold) and stroke volume (1·2-fold). At 18° C, although standard and routine metabolic rates, as well as resting heart rate and ventilation frequency increased significantly, active metabolic rate and certain cardiorespiratory variables during exercise did not differ from those values for fish acclimated to 14° C. As a result, factorial metabolic scope was reduced (2·93-fold at 18° C v . 5·13-fold at 14° C). Therefore, it is concluded that cardiorespiratory performance in triploid brown trout was not unusual at 18° C, but that reduced factorial metabolic scope may be a contributing factor to the mortality observed in triploid brown trout at temperatures near 18° C.  相似文献

Abstract. Cyperus longus L. , which has a widespread but disjunct distribution throughout Europe and extends northwards into Britain, was found to be a C4 species based upon its Kranz leaf anatomy, low CO2 compensation point and the labelling of malate as an early product of 14CO2 fixation. The photosynthetic characteristics of C. longus are similar to many other C4 species with a high maximum rate of photosynthesis (> 1.5 mg CO2 m −2 s −1) and a relatively high temperature optimum (30–35°C), but unlike many C4 species the rate of photosynthesis does not decline rapidly below the optimum temperature and a substantial rate (0.6 mgCO2 m−2s−1)occursat 15°C. Leaf extension is very slow at 15°C and shows a curvilinear response to temperatures between 15 and 25°C. Leaves extend at a rate of almost 4 cm d−1 at 25°C.  相似文献

The production and stability of pediocin N5p from Pediococcus pentosaceus , isolated from wine, were examined in grape juice medium. Maximum growth and higher titre (4000 U ml-1) were observed at a initial pH of 7·5 and 30°C. The activity of the inhibitory substance was stable between pH values from 2·0 to 5·0 at 4° and 30°C. At pH 10·0 it was completely inactivated. When submitted to 30 min at 80°, 100° and 115°C, maximal stability was observed at pH 2·0. Ethanol up to 10% did not affect pediocin activity at acid pH, nor did 40–80 mg 1-1 SO2, independently or combined with different ethanol concentrations, affect inhibitory activity.  相似文献

Abstract. Purified and crude phosphoenolpyruvate carboxylase from the CAM plant Kalanchoë daigremontiana Hamet et Perrier ( Bryophyllum diagremontianum ) was assayed at temperatures between 10 and 45° C. The optimum temperature of the enzyme activity changed with substrate availability and effector concentration in the assay. l -malate inhibited the enzyme activity and lowered the optimum temperature. Glucose-6-phosphate raised the optimum temperature to 43°C. K m values for phosphoenolpyruvate increased with assay temperature from 0.12 mol m-3 at 15° C to 0.36 molm−3 at 35° C. Inhibition by malate increased with temperature and acidity of the assay. In the crude enzyme 50% of control activity was inhibited by 1.65 mol m-3 malate at 15° C and by 0.5 mol m-3 at 35° C (at pH 7.0). With purification malate sensitivity was lost ( K i values for malate at least 10 times higher). The shift in optimum temperatures for PEP-carboxylase activity thus results from changes in the kinetic parameters with temperature and allosteric effectors. The often low optimum temperatures for CO2 fixation observed in nature may thus be the result of substrate and effector concentrations in the cytoplasm and the antagonistic effect of temperature on substrate affinity and effector efficiency on phosphoenolpyruvate carboxylase.  相似文献

Abstract β-xylosidase (EC 3.2.1.37) has been purified from Aspergillus nidulans mycelium grown on oat-spelt xylan as sole carbon source. Its pH optimum for activity was found to be 5.0 and the optimum temperature was 50 °C. Its molecular mass was estimated by gel filtration to be 180000. Using p-nitrophenyl-β-d-xylopyranoside as substrate, the K m and V max values have been found to be 1.1 mM and 25.6 μmol min−1(mg protein)−1, respectively. Enzyme activity was inhibited by Hg2+, Ag2+, and Cu2+ at a concentration of 1 × 10−3 M. The synthesis of β-xylosidase in A. nidulans is strongly induced by arabinose and xylose and is subject to carbon catabolite repression mediated by the cre A gene product.  相似文献

The photosynthetic and growth characteristics of Ceratophyllum demersum L. were investigated under laboratory conditions which simulated those encountered in the plants' normal environment. The carbon fixation rate of C. demersum measured with 14C at light and carbon saturation at pH 8.0 was 4.48 mg C (g ash-free dry weight)−1 h−1. It was lower at pH 6.5 than at pH 8.0. The light use efficiencies in quiescent plants and actively growing plants were 6.3 and 8.7 × 10−9 kg CO2 J−1, respectively, with corresponding maximum photosynthetic rates of 2.67 and 4.36 mg C (g ash-free dry weight)−1 h−1. Photorespiration in actively growing plants consumed 24% of the carbon fixed. Incubation with DCMU demonstrated that about one-third was refixed. The optimum temperature for carbon fixation was 25°C. The C3-photosynthetic pathway was the main operational route as indicated by the early photosynthetic products (largely C3-acids) and the absence of Krantz anatomy and the chlorophyll a:b ratio (2.7). The maximum relative growth rates ranged from 0.025 to 0.041 g ash-free dry weight (g ash-free dry weight)−1 day−1 in the field (Lake Vechten, 1 to 3 m depth classes).  相似文献

Effect of temperature on swimming performance of sea bass juveniles   总被引:1,自引:0,他引:1  
At four temperatures ( T= 15, 20, 25 and 28° C) swimming performance of Dicentrarchus labrax was significantly correlated with total length (23–43 mm L T); r2=0.623–0.829). The relative critical swimming speed ( RU crit= U crit L T−1), where U crit is the critical swimming speed, was constant throughout the L T range studied. The significant effect of temperature on the relative critical swimming speed was described binomially: RU crit=−0.0323T2+ 1.578 T −10.588 (r2=1). The estimated maximum RU crit (8.69 L T s−1) was achieved at 24.4° C, and the 90% performance level was estimated between 19.3 and 29.6° C.  相似文献
