Pigments leached from Sinapis alba L. seeds and extracted from dark-grown seedlings are described and investigated by spectrophotometry and chromatography and their functions examined. Evidence is given that they are quinonoid, and it is suggested that they form part of a complex which absorbs ultraviolet light and utilizes it to produce growth energy at stages before visible light becomes available to the plant, and that this process may be the means by which mutations are introduced into species.  相似文献

Seeds of chickpea (Cicer arietinum L.) were exposed in batches to static magnetic fields of strength from 0 to 250 mT in steps of 50 mT for 1-4 h in steps of 1 h for all fields. Results showed that magnetic field application enhanced seed performance in terms of laboratory germination, speed of germination, seedling length and seedling dry weight significantly compared to unexposed control. However, the response varied with field strength and duration of exposure without any particular trend. Among the various combinations of field strength and duration, 50 mT for 2 h, 100 mT for 1 h and 150 mT for 2 h exposures gave best results. Exposure of seeds to these three magnetic fields improved seed coat membrane integrity as it reduced the electrical conductivity of seed leachate. In soil, seeds exposed to these three treatments produced significantly increased seedling dry weights of 1-month-old plants. The root characteristics of the plants showed dramatic increase in root length, root surface area and root volume. The improved functional root parameters suggest that magnetically treated chickpea seeds may perform better under rainfed (un-irrigated) conditions where there is a restrictive soil moisture regime.  相似文献

The effect of pH and temperature on the association equilibrium constant (Ka) for the binding of the trypsin inhibitor from white mustard (Sinapis alba L.) seeds (MTI) to bovine beta-trypsin (EC 3.4.21.4) has been investigated. On lowering the pH from 9 to 3, values of Ka for MTI binding to bovine beta-trypsin decrease thus reflecting the acid-pK and -midpoint shifts, upon inhibitor association, of two independent ionizable groups, and of a three-proton transition, respectively. At pH 8.0, values of thermodynamic parameters for MTI binding to bovine beta-trypsin are: Ka = 4.5 X 10(8)M-1, delta G0 = -11.6 kcal/mol, and delta S0 = +53 entropy units (all at 21 degrees C); and delta H0 = +4.1 kcal/mol (temperature independent between 5 degrees C and 45 degrees C). Binding properties of MTI to bovine beta-trypsin have been analyzed in parallel with those concerning macromolecular inhibitor association to serine (pro)enzymes.  相似文献

An attempt was made to assess the phytotoxicity of oxadiargyl on two crop plants, viz. sunflower and maize. The seed germination and early growth studies were carried out in hydroponic conditions with each variety of two crop plants, viz. sunflower (Morden) and maize (NAC-6004) supplemented with five concentrations of oxadiargyl. Seed germination and early growth of both the crop plants were significantly affected with increased concentration of oxadiargyl. The length of radicle and plumule was severely reduced along with increase in herbicidal concentration. Similarly, the calculated value of tolerance index and vigour index decreased drastically with increase in concentration of oxadiargyl. On the contrary, the per cent phytotoxicity also increased with increase in oxadiargyl concentration. Further, the pigment system of both the crop seedlings was severely affected by the herbicide oxadiargyl. These studies clearly indicate that oxadiargyl is phototoxic in nature.  相似文献

Disposal of industrial wastewater is a current issue of urbanization. However, this problem can be sorted out by using wastewater as an alternate source of irrigation after the addition of some amendment. In this way, the problem of disposal of wastewater not only will be resolved but also scarcity of irrigation water can be kept off in the future. The current research study was performed to evaluate the influence of different concentrations of wastewater along with canal water for enhancing growth and yield of field mustard. Plants were irrigated with different mixtures of canal water and wastewater (75:25, 50:50, 25:75, and 00:100) in addition to canal water as control. The results revealed that application of 50:50% waste and canal water improved plant height, the number of pods plant?1, pod length, root length, root dry weight, shoot dry weight, 100 grain weight, grain and biomass yield plant?1, and nitrogen, phosphorus, and potassium concentration in grain and straw up to 16%, 15%, 17%, 29%, 15%, 56%, 25%, 41%, 35%, 20%, 52%, 45%, 20%, 44%, and 42%, respectively, over positive control treatment. While, nutrient uptakes and agronomic efficiency of fertilizers also improved by the application of 50:50% canal and wastewater compared to positive control treatment. Furthermore, the concentration of heavy metals, predominantly Cr, Cu, Cd, and Pb, was reduced in grains by application of 50% canal water and 50% wastewater. The outcomes suggest that wastewater utilization along with canal water mixing might be an effective approach for enhancing growth and yield of field mustard.  相似文献

Seed germination and seedling growth as well as chlorophyll and carotenoid contents of mustard and gram seeds were inhibited or reduced significantly due to the treatment of different concentrations (100, 250, 500, 1000 and 2000 ng/ml) of aflatoxin B1. The range of inhibition in all the parameters was found to be directly influenced by the concentrations of toxin applied. Mustard and gram seedling germination inhibition assay can be used for aflatoxin B1.  相似文献

Traditional breeding methods based on inbreeding are difficult to implement in the case of Sinapis alba (white mustard) because this plant displays high levels of self-incompatibility. More rapid progress in breeding could be possible if biotechnological methods and in vitro cultures were used. However, white mustard is not readily amenable to biotechnological treatment. Seeds of traditional S. alba cultivars (e.g., Nakielska) are characterized by high levels of glucosinolates and erucic acid. However, a new Polish variety of white mustard (Bamberka) possesses low erucic acid content in the oil. The main goal of the study was elaboration of a plant regeneration system via in vitro culture of hypocotyl and cotyledon explants from low and high erucic acid-containing white mustard cultivars. In these experiments, a simple system for in vitro regeneration of white mustard was developed, with the aim to promote maximum formation of shoots within a short period of time. Traditional and improved cultivars of S. alba showed comparable capacity for shoot development from hypocotyl-derived and cotyledon-derived explants. The two types of cultivars were characterized by essentially equivalent shoot regeneration responses, being slightly higher in hypocotyl than the cotyledonary explants. A greater influence on shoot regeneration from hypocotyl explants was observed on medium supplemented with 4.4 μmol 6-benzylaminopurine, 0.57 μmol indole-3-acetic acid, and a low concentration of kinetin (4.6 μmol). This technique will allow for rapid generation of sufficient plant material for further use in a variety of white mustard breeding projects.  相似文献

Summary A procedure for the regeneration of fertile transgenic white mustard (Sinapis alba L.) is presented. The protocol is based on infection of stem explants of 7–9 day old plants with an Agrobacterium tumefaciens strain harboring a disarmed binary vector with chimeric genes encoding neomycin phosphotransferase and -glucuronidase. Shoots are regenerated from callus-forming explants within 3–4 weeks. Under selection, 10% of the explants with transgenic embryonic callus develop into fertile transgenic plants. Rooting shoots transferred to soil yield seeds within 14–16 weeks following transformation. Integration and expression of the T-DNA encoded marker genes was confirmed by histochemical glucuronidase assays and Southern-DNA hybridization using primary transformants and S1-progeny. The germination of Paulownia tomentosa Steud. (Empress tree) seeds can be induced either by red light (R) or by exogenously applied gibberellic acid (GA3). The R induced germination is completely suppressed by far red irradiation, abscisic acid (ABA) or by growth retardants such as ancymidol, tetcyclacis and paclobutrazol, though not by AMO 1618 or chloro-choline chloride (CCC). The inhibition caused by far red light (FR) and growth retardants can be overcome by GA3, while the ABA-induced inhibition can be reversed by fusicoccin (FC), which does not reverse the inhibition caused by FR or growth retardants. It is noteworthy that the germination of light insensitive wheat, corn, alfalfa and mung bean seeds is not inhibited by growth retardants.  相似文献

The temperature-dependent, primary dormancy of cv. Florida 683 celery seeds in darkness was partially broken by a 30 min light exposure on the third day of incubation at 20–22°C, resulting in c 50 percent germination after 20 days. This light stimulation was negated by including different inhibitors of gibberellin biosynthesis in the incubation medium. Subsequent addition of a solution of the gibberellins A4 and A7 or of the gibberellin-active compound (1-3-chlorophthalimido)-cyclohexane carboxamide (AC94,377) overcame the inhibitory effects on germination of these GA-biosynthesis inhibitors. PHA quantity and activity dynamics during early germination of bean seed were investigated. Electrophoretic characteristics, subunits composition and carbohydrate-binding specificity of lectin extracted from white kidney bean cv. Bilozerna were studied. It was shown that investigated lectin consisted of 2 subunits E and L with molecular weight 34 and 36 kDa, respectively, analogously to purified PHA ("Serva", Germany), and specifically bound N-acetyl-D-galactosamin and galactose. During germination both quantity and activity of PHA were dramatically decreasing in embryonic axes and in cotyledons, possibly, as a result of the lectin release from seeds to the environment. It is very likely that one of the defence mechanisms of germinating seeds is related with the releasing of lectins that are able to bind components of the bacterial cell wall and to inhibit their growth.  相似文献

The systemic insecticides namely phorate (Thimet 10 G) oxydemeton methyl (metasystox 25 EC) and dimethoate (Rogor 30 EC) decreased oil content in the developing seeds of Indian mustard (Brassica juncea L.) but showed an increase in the mature seeds. The inhibitory effect in the developing seed was accompanied by an increase in soluble sugars and a corresponding decrease in malate dehydrogenase and G6P dehydrogenase activity. The relative proportion of triacylglycerols and glycolipids decreased significantly while that of phospholipids and free fatty acids increased in the developing seeds. In the mature seeds, the proportion of triacylglycerols did not change appreciably from that in control. The erucic acid synthesis which was less at 10 and 20 days after fertilization (DAF) increased at 30 DAF with oxydemeton methyl and dimethoate; phorate was ineffective. In mature seeds, the proportion of erucic acid increased at the cost of linoleic and linolenic acids. All the insecticides appreciably decreased the rate of [1-14C]acetate incorporation into lipids both in vivo and in vitro experiments. In the in vivo experiment, the synthesis of polar lipids was enhanced at 10 and 20 DAF, and the higher doses of oxydemeton methyl and dimethoate at 30 DAF. On the other hand, the 14C-incorporation into triacylglycerols showed an opposite trend to that of polar lipids. In the in vitro experiment, oxydemeton methyl and dimethoate enhanced the synthesis of polar lipids at 10 and 20 DAF while these inhibited it at 30 DAF. The synthesis of triacylglycerols was inhibited by the use of these insecticides.  相似文献

The changes of chemical composition and functional properties of derooted sesame (Sesamum indicum L.) seeds (DSS) before, during, and after germination were investigated. Sesame seeds germinated in dark chambers maintained near 100% relative humidity at 35 degrees C without presoaking reached >99% germination rate in 4 days with the final moisture content stayed ca. 2% (w/w), characterizing sesame seeds as orthodox seeds that are suitable for long term storage at low temperature and humidity under defined environment. With noticeable reduction in fat content (23%), germinated DSS were found rich in linolenic acid, P, and Na, increasing from 0.38% (w/w), 445 mg/100 g, and 7.6 mg/100 g before germination to 0.81% (w/w), 472 mg/100 g, and 8.4 mg/100 g after germination, respectively. DSS after germination contained considerable amount of Ca (462 mg/100 g), higher than that of soybean. Germinated DSS presents an excellent source of sesamol (475 mg/100 g), a potent natural antioxidant, and alpha-tocopherol (32 mg/100 g), the most active form of vitamin E.  相似文献

The activity of wax-ester hydrolase in roots of white mustard (Sinapis alba L.) seedlings is located in a membranous fraction sedimenting at 15000 g. The enzyme which shows a high degree of hydrophobicity was solubilized with a synthetic detergent Triton X-100 and purified about 70-fold by acetone precipitation and gel permeation chromatography on Sepharose 6B. The purified enzyme preparation was active within a broad pH range of 5.8-8.5. Hydrolase activity with hexadecanyl palmitate as the substrate was stimulated by Triton X-100 and dithioerythritol. Of wax esters containing saturated fatty acids C2-C22 and saturated, primary alcohols C2-C24 the highest rate of hydrolysis was found with the esters containing palmitic acid (C16) and tetradecanol (C14). Data presented suggest that wax esters and steryl esters are either hydrolyzed by different specific enzymes or that two enzymes are present of different specificity towards the two substrates.  相似文献

The effect of various abiotic stresses on germination rate, growth and soluble sugar content in Sorghum bicolor (L.) Moench cv. CSH 6 seed embryos and endosperm during early germination was investigated. Under stress conditions germination, water potential and tissue water content decreased markedly. Subsequently, this reduction resulted in marked decreases in fresh weight both in embryos and endosperm. Conversely, a substantial increase in dry weight was observed. Furthermore, a considerable increase in the sugar contents in both embryo and endosperm was detected. The fructose level was always higher than glucose and sucrose in response to various stresses. However, as compared to the control the level of glucose and sucrose was higher in embryos and endosperm after stress treatments. Based upon these results a possible physiological role of sugars in the germination of sorghum seeds is discussed.  相似文献

Light-initiated germination levels of Rumex crispus L. seedswere reduced equally by imbibition in mannitol or polyethyleneglycol 6000 (PEG 6000) solutions of the same w, indicating thatthe effects of each were through w. Reduction of the water contentof the seeds with these osmotica decreased the effectivenessof the far-red absorbing form of phytochrome (Ptr) in causinggermination. However, reduced water content had no effect onthe slopes or saturation points of fluence/response curves whichindicates that has no effect on the number of Ptr receptor sites.The time during which a portion of the seeds were still photoreversibleby far-red light was increased by imbibition in PEG 6000, indicatinga direct effect of w on a reaction involving phytochrome. Oat seeds are susceptible to high temperature dormancy. Dormant grainsdo not germinate at 30 °C unless afterripened, dry, for severalweeks. Isolated embryos of dormant grains do germinate, especially ifGA3 is added to the germination medium. ABA inhibits germinationproportionally to the concentration applied and GA3 can overcome theABA inhibitory effect. Measurements of endogenous ABA and several GAs revealedthat the initial levels of ABA in dormant and non-dormant grains were quitesimilar. But, endogenous ABA in non-dormant seeds almost disappeared within thefirst 16 h of imbibition, while the amount in dormant grains haddecreased by less than 24%. The level of GA19 in non-dormant seedswas higher, and GA19 appears to be converted to GA20 within the first 16h. The GA20 was converted to GA1 at leastduring the first 48 h of the germination process. Bothphytohormones thus appear to be involved in the germination process ofnon-dormant seeds. ABA first declines, while GA1 is producedduring the first 16 h of imbibition to allow proper germination.Indormant grains the level of ABA remained high enough to prevent germinationduring at least a week and precursor GAs were not converted to GA1.  相似文献
