【目的】明确姜黄Curcuma longa根乙醇提取物对苜蓿蚜Aphis craccivora成蚜的生物活性和活性成分,并鉴定活性成分的结构,为利用姜黄根提取物研制新型植物源杀虫剂防治苜蓿蚜奠定基础。【方法】运用植物化学手段和生物活性跟踪方法,采用萃取、硅胶柱层析、葡聚糖凝胶层析,并结合薄层层析检测方法从姜黄根提取物中分离、纯化出对苜蓿蚜成蚜具有杀虫活性的化合物,并采用核磁共振和质谱对其进行结构鉴定。通过浸渍法测定姜黄根提取物对苜蓿蚜无翅成蚜的触杀活性,通过点滴法测定活性成分对苜蓿蚜无翅成蚜的触杀活性。【结果】姜黄根95%乙醇提取物对苜蓿蚜无翅成蚜具有触杀活性,处理后24 h的LC_(50)值为3 226.27 mg/L。从姜黄根中分离得到的活性化合物为芳姜黄酮,该物质对苜蓿蚜无翅成蚜表现出较好的触杀活性,处理后24 h的LC_(50)值为706.10 mg/L。【结论】姜黄根95%乙醇提取物对苜蓿蚜成蚜具有一定的杀虫活性,芳姜黄酮是主要的活性物质。  相似文献

影响麦长管蚜体色变化的主导因素   总被引:1,自引:0,他引:1  
研究麦长管蚜Macrosiphum avenae(Fabricius)体色变化生态主导因素,田间红色麦长管蚜种群对不同抗蚜性寄主的反应和自然条件下,不同体色麦长管蚜的生殖力以及后代种群体色变化情况。结果显示:在实验温度范围内,麦长管蚜种群中红体色蚜虫所占比例随温度升高而增加,在28,29,30,31℃时,红体色蚜虫所占比例分别为6.66%,38.30%,70.60%和65.24%。麦长管蚜体色变化过程中,温度起到重要的作用,而与光周期和寄主营养的关系甚微。红体色麦长管蚜在不同抗蚜性的品种上其种群消长情况存在差异。田间红绿体色麦长管蚜经2代观测,平均蚜量比值分别为9.96和15.85,生殖力差异不显著。在小麦抽穗期到乳熟期红体色麦长管蚜个体比例随着田间条件的改变逐代升高(分别由第1代的17.55%和14.70%增至第2代的29.80%和42.2%)。  相似文献

【目的】分析饥饿胁迫下麦长管蚜Sitobion avenae(Fabricius)的能量物质含量变化,以期从生理生化角度为蚜虫饥饿胁迫下的能量适应机制提供依据。【方法】以取食小麦苗的麦长管蚜为对照,测定饥饿胁迫下麦长管蚜有翅成蚜的鲜重、干重、含水量、可溶性糖、糖原、总脂和可溶性蛋白质含量变化,通过这些不同生化物质含量之间的相关关系,分析饥饿胁迫下麦长管蚜的能量代谢特点。【结果】麦长管蚜饥饿组鲜重和干重均低于羽化后同一时间取食个体,前者范围分别为5.30~8.73和1.67~3.10 mg/20头,后者分别为7.93~8.73和2.53~3.10 mg/20头;饥饿组和取食组含水量范围分别为63.16%~71.76%和63.25%~67.32%,除羽化后1 d外,饥饿组含水量均高于羽化后同一时间取食个体;饥饿组可溶性糖、糖原、总脂和可溶性蛋白质的含量分别在6.60~11.21,0.35~10.81,18.28~30.42和12.77~33.44μg/mg鲜重之间,均低于羽化后同一时间取食个体(分别为7.53~11.21,3.66~10.81,27.53~33.63和21.54~34.43μg/mg鲜重)。饥饿3 d恢复取食后1 d,鲜重、干重、含水量和糖原含量均增加,而可溶性糖、总脂和可溶性蛋白质的含量均降低。相关分析表明,饥饿组可溶性糖与糖原、总脂或者可溶性蛋白质的含量以及可溶性蛋白质与糖原或者总脂的含量存在正相关关系。【结论】饥饿胁迫下麦长管蚜的代谢变化与其糖原、总脂和可溶性蛋白质含量降低有密切关系,通过能量物质的综合利用来适应短期饥饿胁迫。  相似文献

外源化合物诱导后小麦对麦长管蚜和粘虫的抗虫性研究   总被引:6,自引:0,他引:6  
喷施外源化合物和昆虫取食均可诱导小麦的防御反应,如挥发物组成发生改变、某些次生物质含量增加或减少,进而影响昆虫和天敌的行为反应。本实验中小麦苗经茉莉酸或几丁质喷施诱导,可显著提高小麦中丁布的含量,但茉莉酸甲酯的诱导作用不明显。喷施茉莉酸及麦长管蚜或粘虫取食诱导小麦,会导致小麦挥发物的种类及含量与对照相比有显著不同,3种处理间小麦的挥发物也存在显著差异。用经茉莉酸处理的麦苗饲养麦长管蚜和粘虫,可显著抑制二者的生长发育速度,使体重减轻,并导致麦长管蚜繁殖力下降。经茉莉酸处理的麦苗的挥发物对粘虫寄生蜂有吸引作用,表明茉莉酸在诱导小麦产生直接抗虫性的同时,还能诱导其释放吸引天敌的挥发性化合物从而产生间接抗性。  相似文献

【目的】探讨禾谷缢管蚜 Rhopalosiphum padi (Linnaeus)鞣化激素基因的发育表达模式及功能。【方法】采用转录组测序得到禾谷缢管蚜鞣化激素基因bursicon-α 和 bursicon-β cDNA序列。通过实时荧光定量PCR方法,分析该基因的发育表达模式。利用RNA干扰(RNA interference, RNAi)介导 bursicon-α 和bursicon-β 沉默,分析鞣化激素的功能。【结果】序列分析结果显示,禾谷缢管蚜鞣化激素α亚基基因(bursicon-α)cDNA序列开放阅读框为480 bp,编码159个氨基酸残基;β亚基基因(bursicon-β)cDNA序列开放阅读框为417 bp,编码138个氨基酸残基。时序表达分析表明,鞣化激素两个亚基基因在禾谷缢管蚜整个发育期均有表达,以1龄若蚜期表达量最高;成蚜有翅个体中表达量显著高于无翅个体。RNAi介导的 bursicon-α 和 bursicon-β 沉默均能显著抑制禾谷缢管蚜成蚜表皮的黑化。【结论】研究结果表明,鞣化激素在禾谷缢管蚜体壁黑化中发挥着重要作用。该结果为进一步研究鞣化激素在蚜虫生长发育过程中的生理功能提供了基础资料。  相似文献

羧酸酯酶在昆虫对杀虫剂的解毒代谢过程中发挥重要作用,本研究旨在分析吡虫啉胁迫对麦长管蚜羧酸酯酶基因表达的影响。采用同源克隆方法克隆麦长管蚜羧酸酯酶基因cDNA片段,实时荧光定量PCR技术检测羧酸酯酶基因在不同吡虫啉剂量下的表达量变化。扩增所得麦长管蚜羧酸酯酶基因cDNA片段大小为392 bp,命名为SaEST 3(GenBank登录号KY 441614),该片段编码130个氨基酸残基,分子量14 kD,等电点4.93。序列同源性比对及生物信息学分析表明SaEST3推导的氨基酸序列与豌豆长管蚜、麦双尾蚜、夹竹桃蚜、桃蚜的羧酸酯酶氨基酸序列相似性较高,分别为94%、85%、80%和80%。实时荧光定量PCR结果显示,不同吡虫啉处理剂量下,SaEST3 m RNA的相对表达量均上调。克隆得到的基因片段为麦长管蚜羧酸酯酶基因片段,吡虫啉对麦长管蚜羧酸酯酶基因SaEST3表达有一定的影响。  相似文献

【背景】薇甘菊为危险性入侵植物;同时椰子坚蚜为椰子上的重要害虫,对其防治方面的研究较少。研究薇甘菊次生物质对椰子坚蚜的生物活性可为薇甘菊的综合利用及椰子坚蚜的防治提供参考。【方法】采用室内生物测定的方法研究了薇甘菊4种溶剂提取物对椰子坚蚜的胃毒和触杀活性。【结果】供试的4种提取物中,正丁醇提取物和丙酮提取物对椰子坚蚜的胃毒水平较高,LC50分别为12.93和12.48 mg·m L-1,显著高于环己烷提取物和石油醚提取物。而4种提取物对椰子坚蚜的触杀毒力差异不显著,环己烷提取物、正丁醇提取物、丙酮提取物和石油醚提取物对椰子坚蚜的LC50分别为15.28、12.28、11.40和16.62 mg·m L-1。【结论与意义】薇甘菊提取物在椰子坚蚜防控中具有一定的应用潜力。  相似文献

Summary Several members of the adipokinetic/hyperglycemic neurohormone family from several different invertebrate species have been prepared by solid-phase peptide synthesis and assayed by a modified in vivo hyperglycemic bioassay in Blaberus discoidalis cockroaches. The hypertrehalosemic hormone (HrTH) is the endogenous hypertrehalosemic factor for B. discoidalis and was the most potent peptide in the assay. The more divergent the sequence of a family member from Blaberus HrTH, the less potent was the bioanalog. Manduca adipokinetic hormone is the most divergent peptide of the family and was totally inactive in the bioassay. Locusta adipokinetic hormone I had reduced maximum activity in the assay, which suggests that Ser5 is an important residue for the transduction of the hyperglycemic response. The direct relation between bioanalog similarity to Blaberus HrTH sequence and potency suggests that the hormone and target cell receptor for HrTH have evolved to maintain an optimal fit.Abbreviations AKH adipokinetic hormone - HrGH hyperglycemic hormone - HrTH hypertrehalosemic hormone - RPCH red pigmentconcentrating hormone - CAH cardioacceleratory hormone. Three experimental groups of adult females (reproductive and diapausing brachypters, and macropters with reproductive arrest) of Pyrrhocoris apterus (Linnaeus) (Heteroptera: Pyrrhocoridae) from a temperate population were analysed for their adipokinetic responses. The adipokinetic response, expressed as an increase of haemolymph lipids after injection of adipokinetic hormone from Locusta migratoria (Lom-AKH-I), was assessed in relation to age, wing dimorphism and type of reproductive arrest. Two pmols of Lom-AKH-I were used for determination of adipokinetic responses. The increase of haemolymph lipids in all experimental groups of females induced by this dose of the hormone was comparable with that induced by crude extract of the bug’s own corpora cardiaca. The level of adipokinetic response after injection of 2 pmol of Lom-AKH-I was significantly higher in macropterous and diapausing brachypterous females than in reproductive brachypterous females. However, significantly higher contents of haemolymph lipids in control macropterous females than those found in the control reproductive and diapausing brachypterous females of the corresponding age revealed wing-morph-related differences in lipid metabolism. The observed wing morph- and diapause-related differences in the content of haemolymph lipids and adipokinetic response, respectively, are discussed in relation to the different roles of two wing morphs in the life history of this heteropteran.  相似文献

(Euphorbiaceae) and its various fractions in different solvent systems (chloroform, ethyl acetate and n-butanol) were screened for antibacterial and antifungal activities. Aminopeptidase activities were detected in extracts of the free-living nematodes Caenorhabditis elegans and Panagrellus redivivus using the aminoacyl substrate L-alanine-4-nitroanilide. The activities exhibited similarities in Km (C. elegans = 2.22 mM; P. redivivus = 2.09 mM) and specific activity (C. elegans = 1.38 +/- 0.43 mAU min(-1) x g(-1); P. redivivus, 1.23 +/- 0.18m AU min(-1) microg(-1). Each is inhibited competitively by amastatin (C. elegans IC50 = 0.46 microM; P. redivivus IC50 = 15.90 microM) and non-competitively by leuhistin (C. elegans IC50 = 3.00 microM; P. redivivus IC50 = 37.35 microM). The bioactive peptides adipokinetic hormone and substance P decrease the apparent aminopeptidase activities of each extract suggesting that the peptides compete with the Ala-pNA as substrates. With each extract, adipokinetic hormone appeared to be the more effective substrate. Digestion of adipokinetic hormone by C. elegans and P. redivivus extracts in the presence and absence of 1 mM amastatin produced distinct chromatographic profiles that suggest different digestion patterns for the two species. However, amastatin had clear effects on chromatographic profiles from each species indicating that an aminopeptidase is involved in the digestion of the peptide substrates. The data presented indicate that extracts of free-living nematodes are capable of metabolizing peptide hormones, and that this metabolism involves substrate-selective aminopeptidases.  相似文献

The methanol extract of the rhizomes of Gloriosa superba Linn (Colchicaceae) and its subsequent fractions in different solvent systems were screened for antibacterial and antifungal activities. Excellent antifungal sensitivity was expressed by the n-butanol fraction against Candida albicans and Candida glaberata (up to 90%) and against Trichophyton longifusus (78%) followed by the chloroform fraction against Microsporum canis (80%). In the antibacterial bioassay, the crude extract and subsequent fractions showed mild to moderate antibacterial activities. Chloroform fraction displayed highest antibacterial sensitivity against Staphylococcus aureous (88%) followed by the crude extract (59%). The total phenol content of the crude extract and fractions of the plant expressed no significant correlation with the antimicrobial activities.  相似文献

The effect of neuropeptides from the corpora cardiaca of the fruit beetle Pachnoda sinuata on proline metabolism has been investigated in vivo. Conspecific injections of a crude extract from corpora cardiaca cause an increase of the concentration of proline in the haemolymph by nearly 20% and a decrease of the concentration of alanine, the precursor in proline synthesis, by about 64% when compared with a water-injected group. Purification of an extract of corpora cardiaca on reversed-phase liquid chromatography revealed two distinct UV absorbance and fluorescence peaks that cause hyperprolinaemia in the fruit beetle. The major peak is the previously identified octapeptide Mem-CC; the second peak is also a peptide, but its primary sequence remains, as yet, unidentified. Synthetic Mem-CC elicited time- and dose-dependent increases/decreases of the concentrations of proline and alanine in the haemolymph respectively. Furthermore, the receptor for this peptide seems to be specific in P. sinuata: only peptides of the large family of adipokinetic hormones with an Asp, Asn or Gly residue at position 7 could elicit biological activity, whereas those with a Trp, Ser or Val residue at this position did not have any activity.  相似文献

From a crude extract of the sinus glands of the shrimp Penaeus (litopenaeus) schmitti a peptide with hyperglycemic activity in a homologous bioassay was isolated and characterized by a combination of automatic Edman degradation, enzymatic digestions, TLC of dansyl-amino acids, and mass spectrometry. Its M(r) is 8359.4 Da by MS, which coincides with the deduced sequence. Its N-terminus is free and its C-terminus is amidated. It has 6 Cys residues in conserved positions compared with other known CHHs. This is the first sinus gland hormone from an Atlantic Ocean shrimp characterized to date. It has a remarkable 90% sequence similarity to the Indo-Pacific shrimp P. (marsupenaeus) japonicus Pej-VII hyperglycemic hormone.  相似文献

In crustaceans, the X-organ-sinus gland (XO-SG) neurosecretory system is formed of distinct populations of neurons that produce two families of neuropeptides: crustacean hyperglycemic hormone and adipokinetic hormone/red pigment-concentrating hormone. On the basis of electrophysiological evidence, it has been proposed that γ-aminobutyric acid (GABA) regulates both electrical and secretory activity of the XO-SG system. In this work we observed that depolarizing current pulses to neurons located in the external rim of the X-organ induced repetitive firing that suppressed the spontaneous firing of previously active X-organ neurons. Picrotoxin reversibly blocked this inhibitory effect suggesting that the GABA released from the stimulated neuron inhibited neighboring cells. Immunoperoxidase in X-organ serial sections showed co-localization of GABA and glutamic acid decarboxylase (GAD) including the aforementioned neurons. Immunofluorescence in whole mount preparations showed that two subpopulations of crustacean hyperglycemic hormone-containing neurons colocalized with GABA. The expression of GAD mRNA was determined in crayfish tissue and X-organ single cells by RT-PCR. Bioinformatics analysis shows, within the amplified region, 90.4% consensus and 41.9% identity at the amino acid level compared with Drosophila melanogaster and Caenorhabditis elegans. We suggest that crustacean hyperglycemic hormone-GABA-containing neurons can regulate the excitability of other X-organ neurons that produce different neurohormones.  相似文献
