We recently reported that cultivation of oat (Avena sativa L.) without phosphate resulted in plasma membrane phosphoglycerolipids being replaced to a large extent by digalactosyldiacylglycerol (DGDG) (Andersson, M. X., Stridh, M. H., Larsson, K. E., Liljenberg, C., and Sandelius, A. S. (2003) FEBS Lett. 537, 128-132). We report here that DGDG is not the only non-phosphorous-containing lipid that replaces phospholipids but that also the content of glucosylceramides and sterolglycosides increased in plasma membranes as a response to phosphate starvation. In addition, phosphate deficiency induced similar changes in lipid composition in the tonoplast. The phospholipid-to-glycolipid replacement apparently did not occur to any greater extent in endoplasmic reticulum, Golgi apparatus, or mitochondrial inner membranes. In contrast to the marked effects on lipid composition, the polypeptide patterns were largely similar between root plasma membranes from well-fertilized and phosphate-limited oat, although the latter condition induced at least four polypeptides, including a chaperone of the HSP80 or HSP90 family, a phosphate transporter, and a bacterial-type phosphoesterase. The latter polypeptide reacted with an antibody raised against a phosphate deficiency-induced phospholipase C from Arabidopsis thaliana (Nakamura, Y., Awai, K., Masuda, T., Yoshioka, Y., Takamiya, K., and Ohta, H. (2005) J. Biol. Chem. 280, 7469-7476). In plasma membranes from oat, however, a phospholipase D-type activity and a phosphatidic acid phosphatase were the dominant lipase activities induced by phosphate deficiency. Our results reflect a highly developed plasticity in the lipid composition of the plasma membrane and the tonoplast. In addition, phosphate deficiency-induced alterations in plasma membrane lipid composition may involve different sets of lipid-metabolizing enzymes in different plant tissues or species, at different stages of plant development and/or at different stages of stress adjustments.  相似文献

The plasma membranes of oat normally resemble those of other eukaryotes in containing mainly phospholipids and sterols. We here report the novel finding that the galactolipid digalactosyldiacylglycerol (DGDG) can constitute a substantial proportion of oat plasma membrane lipids, in both shoots and roots. When oat was cultivated under severe phosphate limitation, up to 70% of the plasma membrane phosphoglycerolipids were replaced by DGDG. Our finding not only reflects a far more developed potential for plasticity in plasma membrane lipid composition than often assumed, but also merits interest in the context of the limited phosphate availability in many soils.  相似文献

Oat and rye plants were treated with either tetcyclacis (an experimental plant growth regulator), nuarimol (a fungicide) or gamma-ketotriazole (an experimental herbicide). These treatments reduced shoot growth and changed the lipid composition of the shoot plasma membranes. In oat, both tetcyclacis and nuarimol treatments increased plasma membrane cholesterol and increased the phosphatidylethanolamine/phosphatidylcholine (PE/PC) ratio, whereas gamma-ketotriazole treatment reduced cholesterol and the PE/PC ratio. In rye, all treatments reduced the PE/PC ratio. Generally, the sterol/phospholipid ratio was less in oat than in rye but the cholesterol/phospholipid ratio was greater. With all treatments in oat and rye, increases were observed in unsaturation of the phospholipid acyl chains. The fluidity of membranes was measured by steady-state fluorescence polarisation of the probe diphenylhexatriene; oat membranes were more fluid than rye. Membrane fluidity was greater in plasma membranes from plants treated with the xenobiotics than the controls. The results are discussed in the context of the effect of plasma membrane lipid composition on membrane fluidity, and it is concluded that there appears to be no overall simple relationship between membrane lipid composition and fluidity that holds for all treatments in both species.  相似文献

In a long-term experiment bean (Phaseolus vulgaris L.) seedlings were grown for 18 days in hydroponics in either phosphate-sufficient (+P) or phosphate-deficient (-P) nutrient solutions. Phosphate deprivation halved the phosphorous content of roots. In plasma membrane (PM) fractions isolated from -P roots the phospholipid (PL) level was reduced from 35 to 21 mol%, while PL composition and degree of unsaturation were hardly altered. Digalactosyldiacylglycerol (DGDG) accumulated up to 26% of total PM lipids, replacing PL to a large extent. Molecular species and fatty acid compositions of DGDG in root PM were different compared to DGDG present in the -P plastids. In a short-term study, bean seedlings were grown for 18 days in hydroponics with a complete nutrient solution containing phosphate and then incubated in a -P medium for increasing time ranging from 1 up to 96 h. At the end of the starvation period phosphorous content of -P roots was reduced by 30% compared to +P ones. An activation of phospholipase D and phospholipase C was observed after 1 and 2h of phosphate deprivation, respectively. Maximal phosphatidic acid accumulation was detected after 4h of phosphate deprivation, when also DGDG started to accumulate in PM of bean roots. The fatty acid composition of PLD-derived phosphatidylbutanol resembled that of phosphatidylcholine.  相似文献

Changes in lipid composition were investigated on maize roots and shoots under aluminum stress. After 4d exposure to 100 microM Al, root growth was inhibited while shoot growth was not affected. In roots, the decrease of the DBI (double bond index) of total fatty acids may signal a decrease in membrane fluidity. The total lipids (TL) decreased by 49%, but phospholipids (PL), phosphatidylcholine (PC) and phosphatidylinositol (PI) increased to approximately 3-fold. The MGDG increased to 2-fold but no significant change was found in the DGDG. The steryl lipids (SL) increased by 69%. The SL/PL ratio decreased from 2.64 to 1.52 and the MGDG/DGDG ratio increased from 0.45 to 1.06 in roots of Al-stressed plants. Al leads to oxidative stress in roots of treated plants as indicated by the increase of malondialdehyde (MDA) concentrations. In shoots, changes in fatty acid composition were associated with an increase of the DBI in all lipid classes except that of the DGDG decreased. The PG was the lipid class which shows the large variation of fatty acid composition. No significant changes were found either for TL, PL, SL or MDA concentrations in shoots of Al-treated plants. While PE levels did not show significant change, PI and PG increased and PC decreased. However, the Al caused 87% decrease in the GL levels. The MGDG and DGDG decreased to 19- and 8-fold, respectively. The deleterious effects of Al on polar lipids could be caused by a direct intervention of Al on plasma membrane and/or alteration of cell metabolism.  相似文献

The maintenance of membrane function is critical to the ability of plants to resist environmental stresses; specifically, the stability and appropriate fluidity of membranes are crucial to their normal function. We previously demonstrated that plants adapt to long term potassium (K+) deficiency by accumulation of membrane lipids in leaves and maintenance of the lipid composition in roots. In this study, which involved Arabidopsis thaliana and its K+ deficiency tolerant relative Crucihimalaya himalaica, we first calculated the double bond index (DBI) as an indicator of membrane fluidity. After exposure to long term K+ deficiency stress, the DBI of the total lipids in leaves of Athaliana and Chimalaica showed no significant changes, whereas the DBI of the total lipids in the roots of these species showed slight increases. Changes in lysophospholipids (lysoPLs) levels, and digalactosyldiacylglycerol/monogalactosyldiacylglycerol (DGDG/MGDG) and phosphatidylcholine/phosphatidylethanolamine (PC/PE) ratios, all of which strongly reflect membrane stability, were also studied in K+ stressed Athaliana and Chimalaica. After long term K+ deficiency, total lysoPLs levels increased in Athaliana and Chimalaica leaves, but showed no significant changes in roots. DGDG/MGDG and PC/PE ratios were higher in Chimalaica leaves and roots than in those of Athaliana. These results indicate that Chimalaica exhibits superior membrane stability compared with Athaliana. This may explain its superior growth and tolerance under K+ deficient conditions.  相似文献

The lipid composition of the inner and outer membranes of the chloroplast envelope isolated from winter rye (Secale cereale L. cv Puma) leaves was characterized before and after cold acclimation. In nonacclimated leaves the inner membrane contained high proportions of monogalactosyldiacylglycerols (MGDG, 47.9 mol% of the total lipids) and digalactosyldiacylglycerols (DGDG, 31.1 mol%) and a low proportion of phosphatidylcholine (PC, 8.1 mol%). The outer membrane contained a similar proportion of DGDG (30.0 mol%); however, the proportion of MGDG was much lower (20.1 mol%) and the proportion of PC was much higher (31.5 mol%). After 4 weeks of cold acclimation, the proportions of these lipid classes were significantly altered in both of the inner and outer membranes. In the inner membrane the proportion of MGDG decreased (from 47.9 to 38.4 mol%) and the proportion of DGDG increased (from 31.1 to 39.3 mol%), with only a slight change in the proportion of PC (from 8.1 to 8.8 mol%). In the outer membrane MGDG decreased from 20.1 to 14.8 mol%, DGDG increased from 30.0 to 39.9 mol%, and PC decreased from 31.5 to 25.4 mol%. Thus, both before and after cold acclimation, the proportion of MGDG was much higher in the inner membrane than in the outer membrane. In contrast, the proportion of PC was higher in the outer membrane than in the inner membrane. The relationship between the lipid composition of the inner and outer membranes of the chloroplast envelope and freeze-induced membrane lesions is discussed.  相似文献

植物维持膜的功能是其抵御胁迫的关键问题,而维持膜功能必须要保持膜的稳定性和合适的流动性。我们前期的研究发现植物主要是通过积累叶片膜脂和保持根部膜脂基本不变来适应长期缺钾。在本研究中,以拟南芥和其具有耐受缺钾胁迫特性的近缘种须弥芥为对象,研究了与膜的流动性密切相关的双键指数(double bond index,DBI)的变化,发现长期缺钾条件下,两种植物叶片中总的DBI保持不变,根部总的DBI略有降低。同时研究了与膜稳定性密切相关的溶血磷脂的含量和DGDG/MGDG以及PC/PE这两个比值的变化,发现长期缺钾后拟南芥和须弥芥叶片中溶血磷脂的总量呈上升趋势,根部溶血磷脂总量基本保持不变;无论在对照还是缺钾条件下,拟南芥溶血磷脂的总含量要高于须弥芥。须弥芥叶片具有更高的DGDG/MGDG值,根部具有更高的PC/PE值,说明长期缺钾条件下须弥芥膜的稳定性可能更好。这可能是须弥芥耐缺钾的原因之一。  相似文献

This study aimed to evaluate the effect of Cd exposure (100 μmol/L) on polar lipid composition, and to examine the level of fatty acid unsaturation in maize (Zea mays L.). In roots, the level of 16:0 and monounsaturated fatty acids (16:1 + 18:1) decreased in phosphatidylcholine (PC) and phosphatidylethanolamine (PE). In contrast, the proportion of unsaturated 18-C fatty acid species showed an opposite response to Cd. The content, on the other hand, of PC, PE, digalactosyldiacylglycerol (DGDG), and steryl lipids increased in roots (2.9-, 1.6-, 5.3-, and 1.7-fold increase, respectively). These results suggest that a more unsaturated fatty acid composition than found in control plants with a concomitant increase in polar lipids may favor seedling growth during Cd exposure. However, the observed increase in the steryl lipid (SL) : phospholipid (PL) ratio (twofold), the decrease in monogalactosyldiacylglycerol (MGDG) : DGDG ratio, as well as the induction of lipid peroxidation in roots may represent symptoms of membrane injury. In shoots, the unsaturation level was markedly decreased in PC and phosphatidylglycerol (PG) after Cd exposure, but showed a significant increase in sulfoquinovosyldiacylglycerol (SQDG), MGDG and DGDG. The content of PG and MGDG was decreased by about 65%, while PC accumulated to higher levels (4.4-fold increase). Taken together, these changes in the polar lipid unsaturation and composition are likely to be due to alterations in the glycerolipid pathway. These results also support the idea that the increase in overall unsaturation plays some role in enabling the plant to withstand the metal exposure.  相似文献

Substantial efforts have recently been made to demonstrate the importance of lipids and lipid-modifying enzymes in various membrane trafficking processes, including calcium-regulated exocytosis of hormones and neurotransmitters. Among bioactive lipids, phosphatidic acid (PA) is an attractive candidate to promote membrane fusion through its ability to change membrane topology. To date, however, the biosynthetic pathway, the dynamic location, and actual function of PA in secretory cells remain unknown. Using a short interference RNA strategy on chromaffin and PC12 cells, we demonstrate here that phospholipase D1 is activated in secretagogue-stimulated cells and that it produces PA at the plasma membrane at the secretory granule docking sites. We show that phospholipase D1 activation and PA production represent key events in the exocytotic progression. Membrane capacitance measurements indicate that reduction of endogenous PA impairs the formation of fusion-competent granules. Finally, we show that the PLD1 short interference RNA-mediated inhibition of exocytosis can be rescued by exogenous provision of a lipid that favors the transition of opposed bi-layer membranes to hemifused membranes having the outer leaflets fused. Our findings demonstrate that PA synthesis is required during exocytosis to facilitate a late event in the granule fusion pathway. We propose that the underlying mechanism is related to the ability of PA to alter membrane curvature and promote hemi-fusion.  相似文献

儿茶素是一种可以短时间内杀死植物细胞的植物毒素,由于具有强的植物毒性,儿茶素是开发除草剂的理想化合物,它可以诱导植物根系统的死亡。为了研究植物根细胞膜脂对化学胁迫的响应规律,我们运用高通量的脂类组学方法检测了拟南芥根中膜脂分子的组成,比较了儿茶素处理下拟南芥野生型(WS)及磷脂酶Dδ缺失突变体(PLDδ KO)根中膜脂分子的组成情况、膜脂含量、双键指数及碳链长度值。结果发现,儿茶素处理拟南芥根90min后,二半乳糖基二酰甘油(DGDG)、单半乳糖基二酰甘油(MGDG)、磷脂酰甘油(PG)、磷脂酰胆碱(PC)及磷脂酰肌醇(PI)的总含量在WS与PLDδ KO植株根中都显著下降,磷脂酰乙醇胺(PE)和磷脂酰丝氨酸(PS)在WS中下降,在PLDδ KO中上升。儿茶素处理导致PLDδ KO植株的PC/PE比值显著下降,WS植株PS碳链长度显著增加。上述结果说明儿茶素处理后,磷脂酶Dδ缺失突变体膜不稳定性增加,PLDδ KO植株对儿茶素胁迫更加敏感。  相似文献

Plasma membranes were isolated from roots of bean (Phaseolus vulgaris L.) plants cultured on phosphate sufficient or phosphate deficient medium. The phospholipid composition of plasma membranes was analyzed and compared with that of the microsomal fraction. Phosphate deficiency had no influence on lipid/protein ratio in microsomal as well as plasma membrane fraction. In phosphate deficient roots phospholipid content was lower in the plasma membrane, but did not change in the microsomal fraction. Phosphatidylcholine and phosphatidylethanolamine were two major phospholipids in plasmalemma and microsomal membranes (80 % of the total). After two weeks of phosphate starvation a considerable decrease (about 50 %) in phosphatidylcholine and phosphatidylethanolamine in microsomal membranes was observed. The decline in two major phospholipids was accompanied by an increase in phosphatidic acid and lysophosphatidylcholine content. The effect of alterations in plasma membrane phospholipids on membrane function e.g. nitrate uptake is discussed.  相似文献

BACKGROUND AND AIMS: Cell membranes are major targets of environmental stresses. Lipids are important membrane components, and changes in their composition may help to maintain membrane integrity and preserve cell compartmentation under water stress conditions. The aim of this work was to investigate the effects of water stress on membrane lipid composition and other aspects of lipid metabolism in the leaves of the model plant, Arabidopsis thaliana. METHODS: Arabidopsis thaliana (ecotype Columbia) plants were submitted to progressive drought stress by withholding irrigation. Studies were carried out in plants with hydration levels ranging from slight to very severe water deficit. Enzymatic activities hydrolysing MGDG, DGDG and PC were measured. Expression of several genes essential to lipid metabolism, such as genes coding for enzymes involved in lipid biosynthesis (MGDG synthase, DGDG synthase) and degradation (phospholipases D, lipoxygenase, patatin-like lipolytic-acylhydrolase), was studied. KEY RESULTS: In response to drought, total leaf lipid contents decreased progressively. However, for leaf relative water content as low as 47.5 %, total fatty acids still represented 61 % of control contents. Lipid content of extremely dehydrated leaves rapidly increased after rehydration. The time-course of the decrease in leaf lipid contents correlated well with the increase in lipolytic activities of leaf extracts and with the expression of genes involved in lipid degradation. Despite a decrease in total lipid content, lipid class distribution remained relatively stable until the stress became very severe. CONCLUSIONS: Arabidopsis leaf membranes appeared to be very resistant to water deficit, as shown by their capacity to maintain their polar lipid contents and the stability of their lipid composition under severe water loss conditions. Moreover, arabidopsis displayed several characteristics indicative of a so far unknown adaptation capacity to drought-stress at the cellular level, such as an increase in the DGDG : MGDG ratio and fatty acid unsaturation.  相似文献

The effects of membrane sterol level on the susceptibility of LM cell plasma membranes to exogenous phospholipases A2 has been investigated. Isolated plasma membranes, containing normal or decreased sterol content, were prepared from mutant LM cell sterol auxotrophs. beta-Bungarotoxin-catalyzed hydrolysis of both endogenous phospholipids and phospholipids introduced into the membranes with beef liver phospholipid exchange proteins was monitored. In both cases, phosphatidylcholine (PC) and phosphatidylethanolamine (PE) were degraded at similar rates in normal membranes, while PC hydrolysis was specifically accelerated in sterol-depleted membranes. Additional data suggest that this preferential hydrolysis of PC is not a consequence of the phospholipid head group specificity of the phospholipase, nor of a difference in the accessibility of PC versus PE to the enzyme. Analysis of the reaction products formed during treatment of isolated membranes with phospholipase A2 showed almost no accumulation of lysophospholipids. This was found to be due to highly active lysophospholipase(s), present in LM cell plasma membranes, acting on the lysophospholipids formed by phospholipase A2 action. A soluble phospholipase A2 was partially purified from LM cells and found to behave as beta-bungarotoxin with regard to membrane sterol content. These results demonstrate that the nature of phospholipid hydrolysis, catalyzed by phospholipase A2, can be significantly affected by membrane lipid composition.  相似文献

The two mammalian phosphatidylcholine (PC)-selective phospholipase D (PLD) enzymes remove the choline head group from PC to produce phosphatidic acid (PA). PA stimulates phosphatidylinositol(4)phosphate 5-kinases, can function as a binding site for membrane proteins, is required for certain membrane fusion or fission events and is an important precursor for the production of diacylglycerol (DAG). Both PA and DAG are lipids that favor negatively curved membranes rather than planar bilayers and can reduce the energetic barrier to membrane fission and fusion. Recent data provide a mechanistic explanation for the role PLDs play in some aspects of membrane traffic and provide an explanation for why some membrane fusion reactions require PA and some do not. PLDs also act as guanosine triphosphatase-activating proteins for dynamin and may participate with dynamin in the process of vesicle fission.  相似文献

Rat liver dolichyl-phosphomannose synthase (GDP mannose-dolicholphosphate mannosyltransferase; EC 2.4.1.83) was previously shown to catalyze optimal rates of mannosyl transfer to dolichyl-P when the polyprenol acceptor was incorporated into a phosphatidylethanolamine (PE) matrix that has a tendency to adopt a nonbilayer (hexagonal HII) phase [Jensen, J. W., & Schutzbach, J. S. (1985) Eur. J. Biochem. 153, 41-48]. The present investigations now further define the properties of the lipid environment that are essential for mannosyltransferase activity. Monogalactosyl diglyceride (MGDG), a glycoglycerolipid that prefers a nonbilayer-phase organization in isolation, was shown to provide a suitable lipid matrix for synthase activity. By comparison, the enzyme was not activated by digalactosyl diglyceride (DGDG), which forms stable bilayer structures upon hydration. Enzyme activity in MGDG/DGDG mixtures decreased as the proportion of DGDG in the dispersion was increased. Although bilayer-forming phospholipids supported low rates of mannosyl transfer, enzyme activity was stimulated by the addition of MGDG to either phosphatidylcholine (PC) or PE/PC (1:1) membranes. The incorporation of agents known to destabilize bilayer structures including dolichols, ubiquinone, dodecane, and cholesterol into PE/PC (1:1) membranes also increased the rate of mannosyl transfer. Enzyme activity in PC membranes was stimulated by the presence of gramicidin and also by greatly increased concentrations of the substrate, dolichyl-P. The results demonstrate that the enzyme does not have a requirement for PE and suggest that the physical state of the lipid matrix is an important determinant for reconstitution of the synthase and polyprenol phosphate substrate in a productive complex. The formation of an enzyme/lipid complex was demonstrated by sucrose density gradient centrifugation and could be correlated with the lipid requirements for enzyme activity.  相似文献

Phosphatidic acid (PA) and glucosylceramide (Cer), constituents of plant plasma membranes, were used in interaction studies with the major plasma membrane lipid components, phosphatidylcholine (PC) and phosphatidylethanolamine (PE). With molecular species combinations, representative for plant plasma membranes, packing conditions during compression of monolayers of PC/PE mixtures with different amounts of PA or Cer added, were investigated. In contrast to the behaviour of single PA or single Cer, which exhibited condensed compression curves, as compared with curves representative for phosphoglycerides, the triple mixtures of PC/PE with PA or Cer showed markedly expanded monolayer films. These data were evaluated as a spontaneous heterogeneous dispersion of PA and Cer in the PC/PE mixture. Membrane vesicles produced with different amounts of PA added to a PC/PE mixture of 1:1 (mol/mol) had an almost linear increase in permeability for glucose (chosen as a common polar low-molecular mass metabolite) with increasing percentage PA. The presence of PA in plasma membranes and its possible function are discussed in relation to recent reports on anionic protein-lipid interactions. PC/PE vesicles with different amounts of Cer added did not influence the permeability for glucose at 2.5 and 5 mol%, but did so, significantly, at 7.5 and 9 mol%.  相似文献

The plant galactolipids, monogalactosyldiacylglycerol (MGDG) and digalactosyldiacylglycerol (DGDG), are the most abundant lipids in chloroplast membranes, and they constitute the majority of total membrane lipids in plants. MGDG is synthesized by two types of MGDG synthase, type-A (MGD1) and type-B (MGD2, MGD3). These MGDG synthases have distinct roles in Arabidopsis. In photosynthetic organs, Type A MGD is responsible for the bulk of MGDG synthesis, whereas Type B MGD is expressed in non-photosynthetic organs such as roots and flowers and mainly contributes to DGDG accumulation under phosphate deficiency. Similar to MGDG synthesis, DGDG is synthesized by two synthases, DGD1 and DGD2; DGD1 is responsible for the majority of DGDG synthesis, whereas DGD2 makes its main contribution under phosphate deficiency. These galactolipid synthases are regulated by light, plant hormones, redox state, phosphatidic acid levels, and various stress conditions such as drought and nutrient limitation. Maintaining the appropriate ratio of these two galactolipids in chloroplasts is important for stabilizing thylakoid membranes and maximizing the efficiency of photosynthesis. Here we review progress made in the last decade towards a better understanding of the pathways regulating plant galactolipid biosynthesis.  相似文献
