The invasion of NW Europe by a single clone of male-sterile Fallopia japonica var. japonica by purely vegetative reproduction has demonstrated that reproduction by seed and the genetic diversity associated with it may not be an essential feature of plant invasions, at least in the short term. What is less well known is that a significant proportion of the Japanese Knotweed s.l. involved is not F. japonica var. japonica, but the hybrid between it and F. sachalinensisF. × bohemica. This hybrid is able to backcross to either parent with the potential to replace the missing male F. japonica; by the same process, the hybrid is generating the genetic diversity so conspicuously lacking in F. japonica. In terms of understanding the population structure in a particular country, it is important to be able to identify hybrids and putative back-crosses. We bring together a mixture of published and unpublished information to provide a comprehensive section on morphological and anatomical aids to identification. Regeneration of plants from stem and rhizome fragments varies by taxon, as do responses to control techniques, underlining the importance of correct identification of these plants. In this paper we look closely at this group of taxa, with special emphasis on the role played by hybridization, with an examination of the genetic make up of seedlings produced in the wild or by artificial hybridizations, and the implications that this will have on the future directions of the invasion process.  相似文献

The marine microalga Fibrocapsa japonica Toriumi and Takano (Raphidophyceae) produces haemolysins, neurotoxins and reactive oxygen species (ROS). To quantify potential effects of such bioactive compounds on surrounding organisms the marine bacterium Vibrio fischeri was exposed to F. japonica culture samples. Inhibition of V. fischeri ‘s natural luminescence, indicative of impaired metabolism, was related to the number of F. japonica cells added. The effect was fast, within 15 min. It was caused by one, possibly several, excreted substances that were less active after heating. Freezing of culture supernatant partly inactivated these substances, but ROS-scavenging enzymes had no effect. Light enhanced the V. fischeri luminescence inhibition in two ways. The direct effect of light on the action of F. japonica luminescence inhibiter(s) could be described by a saturation curve with maximum effect above 20 μmol photons m−2 s−1. Light also had an indirect effect: biomass production, dependent on light availability, was closely related to the amount of inhibiting compound(s) produced by the alga. Algal growth rate, rather than its cell density, determined the bacterial luminescence inhibition per F. japonica cell, resulting in a 5-fold stronger inhibition at maximum growth rates compared to cells that barely grew during the stationary growth phase. The bioassay with F. japonica and V. fischeri has allowed quantification of the negative effects on bacteria in the microalgal microenvironment. The results presented here suggest that at favourable growth conditions F. japonica releases bioactive compounds that improve its competitive abilities.  相似文献

Habitat patch colonization dynamics and distribution patterns were analysed at a landscape scale in four invasive Fallopia (Polygonaceae) species. Fallopia sachalinensis and F. aubertii were uncommon and population expansion was not evident during the three consecutive years of study. The two most widespread species, F. japonica and F. × bohemica displayed similar habitat selection patterns with ruderal and natural/semi-natural forests favoured. The highest densities of F. japonica and F. × bohemica individuals were at the edge of preferred habitat patches with different patterns of edge selection. Linear network played an important role in species invasion, with 71% of all F. japonica and F. × bohemica occurring within a 10 m buffer of total linear networks (roads, railways, and rivers). However, the buffer represented only 14.5% of the total landscape surface. The rate of population increase was higher for F. japonica (75.8% and 35.2%, in 2002 and 2003, respectively) than for F. × bohemica (63.6% and 0% in 2002 and 2003, respectively) and was largely the result of intra-patch dynamics with low inter-patch colonization. The total surface area occupied by Fallopia clones in the landscape grew by 34.7% over 2 years of the study, with comparable area growth means for F. japonica and F. × bohemica (34.9% and 34.7%, respectively). The hypothesis that F. × bohemica exhibits higher invasive dynamics due to both clonal and sexual reproduction was not supported by our results.  相似文献

We have examined morphological and chromosomal variation inFallopia sect.Reynoutria in Korea to clarify their taxonomic identities and to determine whether their morphological variability is associated with ploidy levels. Principal components analysis (PCA) of individuals from 21 populations, using major distinguishing characters, revealed the presence of four major entiries of sect.Reynoutria in Korea; these includeF. sachalinensis, F. japonica var.japonica, F. forbesii, and the Nonsan population consisting of presumed hybrids. Based on morphology, it is hypothesized that the Nonsan population was probably derived from multiple hybridization events involving the three named taxa. The results also indicate thatF. forbesii is distinct fromF. japonica var.japonica. Polyploidy is more prevalent in sect.Reynoutria than has been previously recognized.Fallopia sachalinensis in Korea occurs as dodecaploids with 2n=132; our count is the first dodecaploid count for the species, and represents the highest chromosome number known in the genus.Fallopia japonica var.japonica occurs as tetraploids (2n=44), hexaploids (2n=66), and octoploids (2n=88), whileF. forbesii occurs as hexaploids (2n=66) and octoploids (2n=88); our counts appear to be the first reported chromosome numbers forF. forbesii. Morphological analysis indicates that there is no apparent correlation between the ploidy levels in these taxa and the morphological characters that we have considered in this study except that the tetraploids ofF. japonica var.japonica tend to have somewhat thicker leaves.  相似文献

Invasive, non-native, white grubs (Coleoptera: Scarabaeidae) cause significant damage in urban landscapes. Although the lack of natural enemies in their new home is often suggested as an important factor in the establishment and spread of invasive species, the potential of incumbent generalist parasites and pathogens to delay their establishment and spread has not been explored. We compared the susceptibility of the introduced Popillia japonica and the native Cyclocephala borealis to 16 species and strains of entomopathogenic nematodes isolated from within or outside the geographic ranges of the two scarabs. We found large variation in the virulence of the species/strains of nematodes with over 50% mortality of P. japonica produced by Heterorhabditis zealandica strain X1 and H. bacteriophora strain GPS11 and of C. borealis by H. zealandica and H. bacteriophora strains KMD10 and NC1. Heterorhabditis indica and H. marelatus caused less than 20% mortality of both scarab species. When considered as a group the nematode species and strains from within and outside the geographic ranges of either P. japonica or C. borealis did not differ in virulence towards either scarab species. Dose response studies with selected nematode species and strains against P. japonica and two additional non-native species Anomala (Exomala) orientalis and Rhizotrogus majalis and the native C. borealis indicated that R. majalis was the least susceptible and P. japonica and A. orientalis were as susceptible as the native C. borealis. Heterorhabditis zealandica was significantly more virulent than any other species or strain against P. japonica with a LC50 of 272 IJs/grub. The LC30 and LC50 values for H. zealandica were also the lowest among the four nematode species/strains tested against A. orientalis and C. borealis. The LC50 values for H. zealandica and H. megidis (UK strain) were significantly lower for the native C. borealis than the introduced A. orientalis. H. zealandica also showed the highest penetration efficiency and the lowest encapsulation in P. japonica and C. borealis grubs. Results suggest that the introduction of the exotic H. zealandica into the front-line states with respect to the movement of P. japonica and A. orientalis should be explored as a tactic to delay their establishment and spread. The results also suggest that the manipulation of the indigenous H. bacteriophora populations may help in delaying spread and mitigating losses caused by the invasive grub species.  相似文献

We compared the virulence of the entomopathogenic nematodes Steinernema scarabaei, Heterorhabditis zealandica, and Heterorhabditis bacteriophora (GPS11 and TF strains) against third instars of the Japanese beetle, Popillia japonica, the oriental beetle, Anomala (=Exomala) orientalis, the northern masked chafer, Cyclocephala borealis, the European chafer, Rhizotrogus majalis, and the Asiatic garden beetle, Maladera castanea, in laboratory and greenhouse experiments. The virulence of the nematode species relative to each other differed greatly among white grub species. H. bacteriophora and H. zealandica had similar modest virulence to P. japonica, A. orientalis, C. borealis, and M. castanea. But against R. majalis, H. zealandica showed low virulence with a clear concentration response whereas H. bacteriophora caused only erratic and very low mortality. In contrast, S. scarabaei had modest virulence against C. borealis, but was highly virulent against R. majalis, P. japonica, A. orientalis, and M. castanea with R. majalis being the most susceptible and M. castanea the least susceptible.  相似文献

Garden experiments focused on vegetative regeneration were carried out with four invasive taxa of the genus Reynoutria (R. japonica var. japonica, R. japonica var. compacta, R. sachalinensis and a hybrid between R. sachalinensis and R. japonica var. japonica, R. ×bohemica). Regeneration ability of stems and rhizomes, timing of shoot emergence and biomass production were studied under the following treatments: laid horizontally on the soil surface; placed upright; buried in the soil; floating in water. Two different soils (sand and garden loam) representing contrasting nutrient levels were applied. Differences were found in the capability and speed of regeneration, as well as in the quality of shoots produced. Regeneration from stems was less efficient than that from rhizomes in all taxa except R. sachalinensis. R. ×bohemica exhibits higher regeneration potential (61%) than all other taxa and can be considered as the most successful taxon of the Czech representatives of the genus Reynoutria in terms of regeneration and establishment of new shoots. High regeneration capacity was also exhibited by R. japonica var. compacta (52%). Other taxa showed generally lower regeneration rates (R. japonica var. japonica 39% and R. sachalinensis 21%), but under some treatments the percentage of regenerated segments was high, too. R. japonica var. japonica rhizomes regenerated successfully in all three soil treatments but not in the water. An opposite pattern was found for its stems: they regenerated well if exposed to water treatment but in the soil, they did not regenerate at all. Particular taxa responded to the soil type in a contrasting way. R. sachalinensis and R. ×bohemica regenerated better in loam while the opposite was true in R. japonica var. japonica. R. japonica var. compacta produced the tallest and R. ×bohemica the heaviest and most robust shoots. It is concluded that rhizomes are more crucial than stems for the spread of knotweeds through fragmentation and clonal growth, suggesting the importance of soil disturbance.  相似文献

We examined the floral morph of tetraploid Ophiorrhiza japonica Blume var. amamiana Hatus. and diploid O. japonica var. japonica to elucidate the association of distyly and ploidy levels. Chloroplast DNA phylogeny was reconstructed to determine the number of tetraploidization events and floral morph shifts in O. japonica. All individuals of O. japonica var. amamiana proved to be long-homostylous, whereas O. japonica var. japonica was distylous with typical long- and short-styled flowers. Distyly is related to the ploidy level. The bagging treatment of flowers indicated that O. japonica var. amamiana is self-compatible and potentially automatically self-pollinating. In cpDNA sequencing analysis, no haplotype was shared between the two varieties. The cpDNA haplotype network displayed the monophyly of O. japonica var. amamiana, suggesting a single origin of this variety. Hence, both tetraploidization and the breakdown of distyly to homostyly in O. japonica var. amamiana likely occurred just once. Because O. japonica var. amamiana having the morphological and cytological entity is recognized as a single lineage and clearly separated from O. japonica var. japonica, this variety can be considered to be a distinct species. We therefore propose to raise O. japonica var. amamiana to the rank of species. We studied the influence of the root-crown weevil Ceutorhynchus scrobicollis on its host plant Alliaria petiolata, a European biennial herb that is currently invading much of temperate North America. Varying timing of attack and herbivore densities in a common garden allowed to assess seasonality of plant response, density-dependence of impact, and the effect of intraspecific competition on C. scrobicollis recruitment (number of F1 generation adults emerged). Data collected in the common garden were compared with data collected at field sites. C. scrobicollis is a common weevil in Europe, frequently attaining high attack levels on its host plant. In the common garden, weevil attack decreased plant survival by up to 43%, reduced plant height by 54%, increased the number of shoots by up to four–fold and delayed seed ripening, but had no significant negative effect on seed production. Plants infested in spring allocated less biomass to aboveground plant parts, and remained smaller than plants attacked in autumn, indicating that the latter were able to partly compensate for weevil attack. Increasing weevil density rarely had an effect on A. petiolata performance, and did not increase F1 recruitment, suggesting strong intraspecific competition. At field sites, C. scrobicollis attack is spread over a long time period, which probably alleviates intraspecific competition. In summary, attack by the root-crown feeding weevil, C. scrobicollis, can substantially reduce growth and survival of A. petiolata. If introduced as a biological control agent into North America, C. scrobicollis is likely to decrease the fitness and competitive superiority of A. petiolata.  相似文献

Fusarium Head Blight of small grain cereal is a disease of growing concern in Europe. Along with Microdochium nivale, several species of Fusarium may be associated with the disease, including species that are potentially toxigenic. This paper describes the results of a large scale survey of the variety and frequency of different Fusarium species and M. nivale in France. A total of 749 soft wheat, durum wheat and barley samples were collected and analyzed from 2000 to 2002. The most frequent species isolated were F.graminearum, F. avenaceum and F. poae. The frequency of F. poae seems to have increased while M.nivale and F. culmorum appear less frequent than previously described in France. Other Fusarium species detected in decreasing prevalence were F. tricinctum, F. equiseti, F. acuminatum, F. sambucinum, F.sporotrichioides, F. moniliforme, F. heterosporum, F. subglutinans and F. oxysporum. All the most frequent pathogenic species and also the less pathogenic ones were frequently associated with individual fields. The implications of these associations for the protection of cereals crops and for contamination by mycotoxins are discussed.  相似文献

Allozyme variation was examined in three diploid taxaChionographis japonica var.japonica, var.kurokamiana, andC. koidzumiana and three tetraploid taxaC. japonica var.kurohimensis, ssp.hisauchiana, and ssp.minoensis. Results show thatC. japonica var.kurokamiana is genetically closer toC. koidzumiana than to var.japonica. In the tetraploid taxa, fixed heterozygosities were found at several loci, and this supports the hypothesis that these taxa are allotetraploids. Furthermore, the tetraploid taxa have many unique alleles not found in the diploid taxa. This suggests that sufficient time has passed since the origin of tetraploids for new mutations to have been fixed.  相似文献

We present the isolation and functional analysis of a transformer2 homologue Mdtra2 in the housefly Musca domestica. Compromising the activity of this gene by injecting dsRNA into embryos causes complete sex reversal of genotypically female individuals into fertile males, revealing an essential function of Mdtra2 in female development of the housefly. Mdtra2 is required for female-specific splicing of Musca doublesex (Mddsx) which structurally and functionally corresponds to Drosophila dsx, the bottom-most regulator in the sex-determining pathway. Since Mdtra2 is expressed in males and females, we propose that Mdtra2 serves as an essential co-factor of F, the key sex-determining switch upstream of Mddsx. We also provide evidence that Mdtra2 acts upstream as a positive regulator of F supporting genetic data which suggest that F relies on an autocatalytic activity to select and maintain the female path of development. We further show that repression of male courtship behavior by F requires Mdtra2. This function of F and Mdtra2 appears not to be mediated by Mddsx, suggesting that bifurcation of the pathway at this level is a conserved feature in the genetic architecture of Musca and Drosophila.Edited by D. As dominant herbivores and notorious pests in their native Neotropics, introduced leaf-cutting ants have the potential for ecological and economic harm. Although a large-scale invasion of leaf-cutting ants has not occurred, an isolated introduction in the Caribbean islands of Guadeloupe provides useful insight into the progress of such an invasion. Since being first detected in 1954, Acromyrmex octospinosus has colonized virtually all available land area, defying an aggressive control campaign and damaging agriculture. I attempted to reconstruct the origins and spread of the invasion, as well as screen for the presence of garden pathogens, which could be used for biological control. Mitochondrial sequencing of the A. octospinosus complex throughout the Caribbean showed that the probable source of the invasion lies on Trinidad and Tobago or northeast South America. Using historical records and field surveys, the invasion’s rate of spread was estimated at 0.51 km/year. Microsatellite genotyping further confirmed the limited dispersal abilities of A. octospinosus, showing the presence of isolation by distance (even in a relatively small geographic area) and suggested ubiquitous local inbreeding. Although the invasion likely resulted from the introduction of a single colony, microsatellites showed a high level of genetic variation in the introduced population, likely as a consequence of multiple mating by the queen. A survey showed that the specialized fungus garden pathogen Escovopsis exists on the islands, suggesting that the successful spread of the ants was not due to escape from this parasite. Given that chemical control has failed in the past and that biological control using specialized garden pathogens seems improbable, only vigorous quarantine and inspection programs may prevent wide-scale leaf-cutting ant invasions in the future.  相似文献

The seaweed Laminaria japonica (Phaeophyceae) has a two-generation life cycle consisting of haploid gametophytes and diploid sporophytes. Female and/or male gametophytes were transformed using particle bombardment and the histological LacZ assay was performed on sporophytes generated by either parthenogenesis or inbreeding. Female gametophyte-targeted transformation resulted in similar lower efficiencies in both parthenogenetic and zygotic sporophytes, and only a chimeric expression pattern was observed. Male gametophyte-targeted transformation led to a higher efficiency, with 3.5% of the zygotic sporophytes stained completely blue (all-blue), implying the integration of lacZ at the one-cell stage. Polymerase chain reaction analysis using primers specific for a lacZ-vector juncture fragment and subsequent blotting indicated the presence of the introduced gene in the sporophytes. On a moderately protected intertidal sand flat in west Kyushu, Japan, most of the population of the cirolanid isopodEurydice nipponica Bruce & Jones (89%–100%) was found in the zones occupied by the thalassinidean ghost shrimpCallianassa japonica Ortmann in July, August and December 1980.C. japonica later extensively expanded its habitat, and in July 1984, when almost the whole sand flat had been densely populated byC. japonica, the range of the distribution ofE. nipponica was the same as that ofC. japonica with a density about 10 times greater than in July 1980. The occurrence ofE. nipponica has previously been recorded from several exposed sandy beaches as well as their adjacent subtidal areas of well oxygenated sands along the coast of Kyushu, but not on more protected shores like the present sand flat. It is suggested thatC. japonica, through its bioturbating activities, produces sediment characteristics approximating those of the exposed sandy beaches which are the preferred habitat ofE. nipponica. It is supposed thatE. nipponica is a facultative commensal ofC. japonica. In a field experiment to excludeC. japonica and to detect its positive influence onE. nipponica, the densities ofE. nipponica were found to be lower in the experimental plots than in the intact plots. The genus Reynoutria is represented by four taxa in the Czech Republic – R. japonica var. japonica and compacta, R. sachalinensis and R. × bohemica. Using isoenzyme analysis, we determined the degree of genotype variability in all taxa and compared clones of R. japonica var. japonica from the Czech Republic with those from Great Britain. While the rarely occurring tetraploid variety R. japonica var. compacta possesses low variability, the octoploid female clone of R. japonica var. japonica is genetically uniform in the 93 clones sampled and belongs to the same genotype that is present in the whole Europe. R. japonica var. japonica can be fertilized by the pollen of tetraploid R. sachalinensis and a hexaploid hybrid R. × bohemica is produced. In R. sachalinensis, 16 genotypes were found in the 50 clones sampled. R. × bohemica is genetically the most diverse taxon in the study area, with 33 genotypes recorded among 88 clones sampled.  相似文献

Because susceptibility of white grub species to entomopathogenic nematodes differs, we compared the virulence of Photorhabdus temperata and Xenorhabdus koppenhoeferi, the symbiotic bacteria of the nematodes Heterorhabditis bacteriophora and Steinernema scarabaei, respectively, to the three white grub species, Popillia japonica, Rhizotrogus majalis, and Cyclocephala borealis. Both bacteria were pathogenic to all three grub species even at 2 cells/grub. However, the median lethal dose at 48 h post injection and median lethal time at 20 cells/grub showed that P. temperata was more virulent than X. koppenhoeferi to C. borealis. Although H. bacteriophora is less pathogenic than S. scarabaei to R. majalis and P. japonica, their symbiotic bacteria did not differ in virulence against these two grub species, and they also showed similar growth patterns both in vitro and inside R. majalis larvae at 20 °C. We then tested the pathogenicity of oral- and intrahemocoel-introduced H. bacteriophora to R. majalis to determine whether nematodes are able to successfully vector the bacteria into the hemolymph. Hemocoel injected H. bacteriophora was pathogenic to R. majalis indicating successful bacterial release, but orally introduced H. bacteriophora were not. Dissection of grubs confirmed that the orally introduced H. bacteriophora were unable to penetrate into the hemolymph through the gut wall. We conclude that the low susceptibility of R. majalis to H. bacteriophora is not due to the symbiotic bacteria but rather to the nematode’s poor ability to penetrate through the gut wall and the cuticle to vector the bacteria into the hemolymph.  相似文献

A two-parameter statistical model was used to predict the solubility of 96 putative virulence-associated proteins of Flavobacterium psychrophilum (CSF259-93) upon over expression in Escherichia coli. This analysis indicated that 88.5% of the F. psychrophilum proteins would be expressed as insoluble aggregates (inclusion bodies). These solubility predictions were verified experimentally by colony filtration blot for six different F. psychrophilum proteins. A comprehensive analysis of codon usage identified over a dozen codons that are used frequently in F. psychrophilum, but that are rarely used in E. coli. Expression of F. psychrophilum proteins in E. coli was often associated with production of minor molecular weight products, presumably because of the codon usage bias between these two organisms. Expression of recombinant protein in the presence of rare tRNA genes resulted in marginal improvements in the expressed products. Consequently, Vibrio parahaemolyticus was developed as an alternative expression host because its codon usage is similar to F. psychrophilum. A full-length recombinant F. psychrophilum hemolysin was successfully expressed and purified from V. parahaemolyticus in soluble form, whereas this protein was insoluble upon expression in E. coli. We show that V. parahaemolyticus can be used as an alternate heterologous expression system that can remedy challenges associated with expression and production of F. psychrophilum recombinant proteins.  相似文献

Resistance to Fusarium head blight (FHB) caused by Fusarium graminearum Schwabe in wheat (Triticum aestivum L.) was identified in disomic chromosome substitution and translocation lines, into which chromosome 7el2 had been introgressed from wheatgrass, Thinopyrum ponticum. In this study, two chromosome substitution lines with different origins (designated as el1 and el2) and with different reactions to infection by F. graminearum were crossed to develop a segregating mapping population. The objectives of this study were to determine the effectiveness of this type II resistance and map it on chromosome 7el2. Type II resistance to FHB was characterized in the F2, F2:3 families, F4:5 plants and F5:6 recombinant inbred lines developed by single-seed descent; and the population was characterized in the F2 and F5 with DNA markers along the long arm of 7el. Composite interval mapping revealed a FHB resistance QTL, designated Qfhs.pur-7EL, located in the distal region of the long arm of 7el2 and delimited with flanking markers XBE445653 and Xcfa2240. Additive effects of Qfhs.pur-7EL reduced the number of diseased spikelets per spike following inoculation of one floret in four experiments by 1.5–2.6 and explained 15.1–32.5% of the phenotypic variation in the populations. Several STS-derived and EST-derived PCR or CAPS markers were developed in this chromosomal region, and showed the specificity of 7el2 compared to an array of wheat lines possessing other sources of FHB resistance. These markers are useful in an effort to shorten the chromosome segment of 7el2 and to use for marker-assisted introgression of this resistance into wheat.  相似文献
