Hepatic endothelial lipase (HEL) activity is as high in the neonatal (1-day old) rat liver as in adults. Most of the HEL activity is located at the capillaries since 75% of the total activity is released by heparin or collagenase perfusion. The residual activity (non-releasable) is located in hepatocytes and not in hemopoietic cells, which are the major cell type in neonatal liver. Per mg of protein, the HEL activity is 50% higher in neonatal than in adult hepatocytes. We suggest that neonatal hepatocytes have an increased capacity to synthesize and secrete HEL activity, so maintaining a high activity in the whole organ. it might contribute to the hepatic uptake of cholesterol from circulating lipoproteins, in a period in which endogenous cholesterol synthesis is known to be inhibited in the liver.  相似文献

Liver lipoprotein lipase activity in neonatal (1- and 5-day-old) rats was 2-3-times than in the liver of adult rats. In mid-suckling (15-day-old) or weaned (30-day-old) animals, it was not significantly different from the low activity detected in adult rats. Starvation resulted in a 3-fold increase of lipoprotein lipase activity in the neonatal liver, but did not affect the activity in the liver of mid-suckling, weaned or adult rats. When isolated livers from both 1- and 5-day-old pups were perfused with heparin, a sharp peak of lipoprotein lipase activity appeared in the perfusate. In fed neonates, the peak area accounted for about 70% of the total (released + non-releasable) activity. In starved neonates, the proportion of heparin-releasable activity increased up to about 90%. These results indicate that neonatal rat liver lipoprotein lipase activity is markedly affected by changes in the nutritional status of the animal, and the effect is restricted to the vascular pool of the enzyme, as was reported in extrahepatic tissues from adult rats.  相似文献

L-Lysine-2-oxoglutarate reductase (EC 1.5.1.8, NADP+) in the liver of adult rats increased 4–5-times when the animals were treated with alloxan. In diabetic rats injection of insulin or adrenalectomy prevented the increase in enzyme activity. The activity of the similar enzyme in kidney was not changed by these treatments. The enzyme activity in primary cultured adult rat hepatocytes was also induced by addition of dexamethasone and glucagon together, and glucagon could be replaced by dibutyryl cyclic AMP. Insulin inhibited the induction. The hormonal induction was also inhibited by actinomycin D and by cycloheximide. During development of rats, fetal liver showed very low activity, but the activity appeared on day 1 after birth and then increased rapidly, reaching the adult level by day 5. The activity of the kidney enzyme increased more slowly and reached the adult level 1 month after birth. Intra-uterine injection of glucagon caused precocious induction of the liver enzyme in fetuses. These results indicate that the activity of L-lysine-2-oxoglutarate reductase in the adult liver and in part in neonatal liver also, is controlled by both glucagon and glucocorticoid.  相似文献

The interaction of glucagon with specific receptors has been studied in isolated intact neonatal and adult rat hepatocytes. The hormone binding measured directly with 125I-labelled glucagon was saturable and reversible. The 125I-labelled glucagon binding was inhibited by unlabelled homologous hormone at concentrations ranging from 0.5 nM to 50 μM. Two different binding models were assumed to analyse the binding data by a nonlinear least-squares procedure: (I) a single class of independent sites and (II) two classes of independent sites. The comparison of the fitted theoretical curves reveals that both binding models are in fact compatible with these data. Adult hepatocytes have a considerably higher affinity for glucagon than neonatal hepatocytes; the binding capacity of neonatal liver cells from 1–7-days-old rats proved to be markedly reduced compared with the cells from adult rats. The glucagon-induced intracellular cyclic AMP production was measured at various hormone concentrations under conditions identical to those for the determination of extracellular hormone binding. The correlation of both parameters indicates a direct connection between receptor-occupancy and adenylate cyclase stimulation. These results suggest that a decrease receptor concentration in neonatal hepatocytes is responsible for the decreased cyclic AMP production.  相似文献

Summary A technique for the organ culture of postnatal and adult rat liver has been developed. Liver slices, 0.3 mm thick, were maintained in Conway units at the interphase between medium and a 95% O2:5% CO2 atmosphere. Postnatal liver in culture for up to 72 h had healthy hepatocytes throughout the explants; if adult liver was used the upper 0.2 mm was healthy after 24 h. These slices incorporated tritiated orotate and leucine into trichloroacetic acid-precipitable material. Incorporation of orotate was shown to be spread over the entire slice of neonatal liver. Culturing did not alter the potassium ion content of postnatal liver. Tyrosine aminotransferase activity in liver slices from postnatal, adult, and adrenalectomized adult rats was stimulated by glucocorticoids and dibutyryl cyclic AMP. Cycloheximide and actinomycin D prevented this response. Further, cortisol exerted a permissive effect on the stimulation of tyrosine aminotransferase activity by dibutyryl cyclic AMP in slices from adrenalectomized rats. Induction of urea cycle enzymes by cortisol was demonstrated in cultures of liver from adrenalectomized adult animals. Deceased October 1, 1983. Lipoprotein lipase activity in neonatal-rat liver cell types.   总被引:5,自引:0,他引:5  
The lipoprotein lipase activity in the liver of neonatal (1 day old) rats was about 3 times that in the liver of adult rats. Perfusion of the neonatal liver with collagenase decreased the tissue-associated activity by 77%. When neonatal-rat liver cells were dispersed, hepatocyte-enriched (fraction I) and haemopoietic-cell-enriched (fraction II) populations were obtained. The lipoprotein lipase activity in fraction I was 7 times that in fraction II. On the basis of those activities and the proportion of both cell types in either fraction, it was estimated that hepatocytes contained most, if not all, the lipoprotein lipase activity detected in collagenase-perfused neonatal-rat livers. From those calculations it was also concluded that haemopoietic cells did not contain lipoprotein lipase activity. When the hepatocyte-enriched cell population was incubated at 25 degrees C for up to 3 h, a slow but progressive release of enzyme activity to the incubation medium was found. However, the total activity (cells + medium) did not significantly change through the incubation period. Cycloheximide produced a time-dependent decrease in the cell-associated activity. Heparin increased the amount of lipoprotein lipase activity released to the medium. Because the cell-associated activity was unchanged, heparin also produced a time-dependent increase in the total activity. In those cells incubated with heparin, cycloheximide did not affect the initial release of lipoprotein lipase activity to the medium, but blocked further release. The cell-associated activity was also decreased by the presence of cycloheximide in those cells. It is concluded that neonatal-rat hepatocytes synthesize active lipoprotein lipase.  相似文献

The activity of cytochrome P-450 dependent monooxygenase system from rat liver microsomes after induction by phenobarbital and 3-methylcholantrene in early neonatal period (3-16 days after birth) was studied. It was found that the total amount of cytochrome P-450 increases after injection of these inducers in neonatal rats of all age groups. In parallel, in the case of 3-methylcholantrene induction the benz(a)pyrene hydroxylase and 7-ethoxyresorufin deethylase activities increase; phenobarbital induction causes a rise in the benzphetamine-N-demethylase and benz(a)pyrene hydroxylase activities. Immunochemical analysis involving the use of antibodies specifically directed against cytochrome P-450 of adult rats revealed that the level of cytochrome P-450 in the case of 3-methylcholantrene induction increases from 5 to 50%, whereas that of cytochrome P-450 upon phenobarbital induction increases from 5 to 40% in liver microsomes of 3- and 16-day-old rats. The mode of inhibition of various substrates metabolism by antibodies in neonatal rat microsomes suggests that the 3-methylcholantrene-induced cytochrome P-448, like in adult rats, participates in the hydroxylation of benz(a)pyrene and O-deethylation of 7-etoxyresorufin. The participation of phenobarbital-induced cytochrome P-450 in the metabolism of benzphetamine and aldrin in neonatal rats is much lower than in the adult ones. The metabolism of benz(a)pyrene in phenobarbital-induced neonatal rat microsomes in all age groups is not inhibited by antibodies. The age-dependent differences in inhibition of metabolism and the increase in the benz(a)pyrene hydroxylase activity in phenobarbital-induced rats suggest that the spectrum of inducible forms of cytochrome P-450 in neonatal rats differ from that in adult animals.  相似文献

1. A method for the preparation of hepatocytes from livers of 11-15-day old rats is described. These cells in general behave similarly to hepatocytes made from adult rats with respect to stimulation of gluconeogenesis by glucagon and adrenaline and the effects of added oleate. 2. Significant differences in the behaviour of hepatocytes from neonatal and adult rats were nevertheless seen in certain situations, e.g. with alanine as gluconeogenic substrate, and appeared to be related to the redox state of the cells. 3. The importance of redox state upon gluconeogenesis was examined in more detail by determining the effects of oleate, ethanol and DL-3-hydroxybutyrate alone and in combinations. Major differences between neonatal and adult hepatocytes were again observed with alanine as substrate. 4. A discussion concludes that, while some relevant differences in the enzyme complements of neonatal and adult rat livers are known, it is the high capacity of the neonatal liver to generate reducing power by oxidation of fatty acid that can explain the observed differences.  相似文献

Neonatal hepatocytes are less active in uptake of bile acids than are mature hepatocytes. This phenomenon has been further investigated by transport studies with azidobenzamidotaurocholate (ABATC). Taurocholate, cholate and the photolabile ABATC were taken up by liver cells of adult rats by a sodium-dependent and by an additional sodium-independent mechanism. In the dark, ABATC inhibited the uptake of taurocholate and cholate. Taurocholate decreased the transport of ABATC in a competitive manner, both in the presence and absence of sodium. In neonatal hepatocytes the Vmax for taurocholate and for ABATC was similar but was lower than in mature liver cells. In contrast, the Km was similar for neonatal and mature hepatocytes. For identification of binding proteins in both kinds of cells ABATC was photolysed after preincubation with isolated hepatocytes. Under our experimental conditions (single ultraviolet flash) about 80% of the azido groups was converted to nitrene. The covalently binding nitrene derivative inhibited bile salt transport irreversibly. Photolabeling of intact hepatocytes or of isolated plasma membranes with ABATC resulted in radioindication of membrane proteins with 67, 60, 54, 50 and 43 kDa in mature plasma membranes but of proteins with masses of 67, 54, 43 and 37 kDa in neonatal basolateral membranes. The 50 kDa protein in largely lacking in membranes of 9-day-old rats. The process of photolabeling itself was sodium-independent when isolated cells were treated with ABATC. In contrast, the degree of labeling of intact hepatocytes was markedly reduced in the absence of sodium and chloride. 100-fold molar excess of taurocholate, benzamidotaurocholate (BATC), phalloidin or cyclosomatostatin protected isolated plasma membranes against coupling of ABATC. Photolabeling of hepatoma cells known to be deficient in bile salt transport did not result in radiomodification of membrane proteins.  相似文献

Maintenance of whole body cholesterol homeostasis is determined in part by the liver. Thus, changes in expression of hepatic parameters important in the regulation of cholesterol metabolism may play key roles in determining how homeostasis is maintained. The expression of hepatic lipoprotein uptake systems was studied during development using as a ligand very-low density lipoproteins rich in apolipoprotein E that had been obtained from hypercholesterolemic adult rats. These lipoproteins can serve as ligands for cell surface receptors recognizing apolipoproteins B and/or E. Uptake was lowest in freshly isolated fetal rat hepatocytes, increased substantially in hepatocytes from neonates and was intermediate in those from adults. Binding of these lipoproteins to liver membranes prepared from fetal, neonatal, suckling, weaned and adult rats was lowest in fetal preparations, while those from suckling, weaned and adult livers behaved similarly. Numbers of binding sites in neonatal liver membranes were similar to those in adult, but showed a different affinity. On the basis of this data, the ability of hepatocytes to recognize and remove apolipoprotein B/E-containing lipoproteins from the plasma appears to be a function of the differential expression or regulation of lipoprotein-uptake systems during development.  相似文献

The developmental profile of prostaglandin (PG)-synthesizing enzymes in liver was investigated in rats from the fetus to 2 years old. In the neonatal period, the activities of PGD2-(2.7 nmol/min/mg protein) and PGE2-(2.2 nmol/min/mg protein) synthesizing enzymes were predominant, whereas PGE2-synthesizing enzyme alone further increased in activity during adult to old ages (5.2-6.1 nmol/min/mg protein). In order to determine the sites of PGs production in rat liver, we prepared hepatocytes and non-hepatocytes by a collagenase digestion method. Regardless of the ages we examined, the major PG produced in the hepatocytes was proved to be PGE2, on the other hand, PGD2 was almost exclusively produced in the non-hepatocytes. These results suggest that each PG may have individual roles in the development of rat liver.  相似文献

Hepatocytes from neonatal rats of 0 to 3 days old grew actively in primary culture without added serum or growth factors. In these culture conditions, growth of hepatocytes decreased progressively with increase in age of the rats from which they were isolated, and hepatocytes from rats of 2 weeks old showed scarcely any growth. Actively growing hepatocytes were found to secrete a growth factor that promoted their growth and that of Swiss 3T3 cells, but not that of adult hepatocytes. This growth factor in conditioned medium of growing hepatocytes was heat- and acid-stable, but sensitive to trypsin, and had a molecular weight of over 10,000. It did not inhibit the binding of [125I]epidermal growth factor to its receptor, and its growth promoting activity was not inhibited by monoclonal antibody against insulin-like growth factor II. Therefore, it seems to be a new growth factor. These results, together with previous findings (Nakamura, T., Nagao, M., & Ichihara, A. (1987) Exp. Cell Res. 169, 1-14) demonstrated a reciprocal relation between growth and maturation of neonatal hepatocytes during development, like that of adult cells, but indicated that unlike growth of the latter, growth of neonatal cells is induced by an autocrine mechanism.  相似文献

Role of cell replication on aflatoxin B1 (AFB1) induced hepatocarcinogenesis was investigated in neonatal rats showing persistence of cell replication in the liver for 21 days of post natal life. Adult (8-10 weeks old) rats displaying no hepatocytic proliferation served as controls. Three doses of AFB1 were administered to both the groups at intervals of 48 hr with the doses starting on 10th day of age in the neonatal group. Appearance of phenotypically altered preneoplastic hepatocytes was quantitated in both the groups. A significantly higher incidence of preneoplastic foci was recorded in neonatal rats as compared to adult animals. The results suggest that presence of cell replication in neonatal rats at the time of AFB1 administration enhances the process of hepatocarcinogenesis.  相似文献

Beta-receptor number (measured by [3H]-CGP 12 177 binding) and beta-adrenergic response (measured by isoproterenol stimulated glucose liberation and isoproterenol stimulated adenylate cyclase activity) were compared in hepatocytes isolated from foetal (on day 22 of gestation), adult female and adult male rats. Beta-receptor numbers in crude membrane preparations of hepatocytes from adult female and adult male rats were found to be nearly equal (15.5 and 15.1 fmol/mg), but in crude membrane preparations of foetal rats beta-adrenergic receptor number was significantly higher (34.3 fmol/mg). Determination of number of beta-adrenergic surface receptors of intact hepatocytes showed relative high values in foetal rats (about 22,000/cell) and adult female rats (about 20,000/cell), but in male rats the number was less (about 6500/cell). Glucose liberation was stimulated by isoproterenol to the same extent in hepatocytes isolated from adult female and foetal rats (about 150% over basal), whereas no effect was found in hepatocytes isolated from adult male rats. Dose-response curves showed that in foetal rat hepatocytes glucose release was already increased by 10(-8) M isoproterenol, whereas in female rat hepatocytes at least 10(-6) M isoproterenol was required. Adenylate cyclase was stimulated by isoproterenol in lysates of hepatocytes from adult female rats by about 180% and from foetal rats by about 250%. No effects were observed using lysates of hepatocytes from adult male rats. We interpret the observed differences of beta-adrenergic responses between adult female and male rats as being primarily caused by different accessibility of the beta-receptor to the beta-agonist isoproterenol in intact hepatocytes.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

We have raised specific (rabbit anti-rat) polyclonal antibodies to hepatic mitochondrial carbonic anhydrase V (CA V) and used them to assay the amounts of protein expressed in liver mitochondria isolated from term-foetal, control or diabetic adult rats and in perivenous and periportal rat hepatocytes. The levels of CA V expressed in mitochondria isolated from the livers of adult male and female rats are similar and increase (about 2-fold) in mitochondria from adult diabetic rats when compared to those isolated from the livers of control rats. The level of enzyme in adult liver was higher than in the livers of term-foetal rats. CA V is expressed in both perivenous and periportal hepatocytes, but the level of expression is greater (approx. 40%) in perivenous cells. The implications and significance of these findings are discussed with reference to the roles and properties of the other carbonic anhydrase isoenzymes and the metabolic function of the mitochondrial isoenzyme.  相似文献

In this study, we explored the paradox that in suckling rats the serum concentration of LDL is high although the liver secretes only minimal quantities of VLDL, the presumed precursor of LDL. Freshly isolated hepatocytes and hepatocytes in primary culture obtained from adult (90 days old) and suckling (17 days old) rats were used to investigate the synthesis and secretion of apolipoprotein B (apoB) and lipids as well as the density profile of secreted apoB-containing lipoproteins. Furthermore, the effects of dexamethasone and oleate on apoB biogenesis were investigated in primary cultures of hepatocytes from adult and suckling rats. Hepatocytes from suckling rats were unable to assemble mature VLDL but secreted apoB as primordial lipoprotein particles in the LDL-HDL density range. Intracellular degradation of apoB was also reduced in hepatocytes from suckling rats compared with that in hepatocytes from adults. The immaturity in VLDL assembly and apoB degradation of hepatocytes from suckling rats could be overcome by treating the cultures with dexamethasone plus oleate or dexamethasone alone. The lower microsomal triacylglycerol transfer protein (MTP) mRNA concentrations in hepatocytes from suckling rats in comparison with hepatocytes from adult rats were not reflected in lower MTP activity levels. Furthermore, dexamethasone plus oleate treatment had no effect on MTP activity although VLDL assembly and secretion were clearly stimulated. We conclude that, during the suckling period of the rat, serum LDL is directly produced by the liver. This is a result of impaired hepatic VLDL assembly, which is a consequence of low triglyceride synthesis and an inefficient mobilization of bulk lipids in the second step of VLDL assembly.  相似文献

The contribution of the liver to glucose utilization is essential to maintain glucose homeostasis. Previous data from protein tyrosine phosphatase (PTP) 1B-deficient mice demonstrated that the liver is a major site for PTP1B action in the periphery. In this study, we have investigated the consequences of PTP1B deficiency in glucose uptake in hepatocytes from neonatal and adult mice. The lack of PTP1B increased basal glucose uptake in hepatocytes from neonatal (3-5 days old) but not adult (10-12 wk old) mice. This occurs without changes in hexokinase, glucokinase, and glucose 6-phosphatase enzymatic activities. By contrast, the glucose transporter GLUT2 was upregulated at the protein level in neonatal hepatocytes and livers from PTP1B-deficient neonates. These results were accompanied by a significant increase in the net free intrahepatic glucose levels in the livers of PTP1B(-/-) neonates. The association between GLUT2 and insulin receptor (IR) A isoform was increased in PTP1B(-/-) neonatal hepatocytes compared with the wild-type. Indeed, PTP1B deficiency in neonatal hepatocytes shifted the ratio of isoforms A and B of the IR by increasing the amount of IRA and decreasing IRB. Moreover, overexpression of IRA in PTP1B(-/-) neonatal hepatocytes increased the amount of IRA/GLUT2 complexes. Conversely, hepatocytes from adult mice only expressed IRB. Since IRA plays a direct role in the regulation of glucose uptake in neonatal hepatocytes through its specific association with GLUT2, we propose the increase in IRA/GLUT2 complexes due to PTP1B deficiency as the molecular mechanism of the increased glucose uptake in the neonatal stage.  相似文献

Morphological (light microscopical, immunohistological and electron microscopical) findings in the recipient liver of rats with streptozotocin-induced diabetes, obtained 9 months after intraportal injection of neonatal isologous pancreatic islets, are described and their significance discussed. - The results support the assumption of active ingrowth of nonmyelinated nerve fibers into the islet isografts. - The hepatocytes surrounding the islet isografts contain-obviously owing to the influence of unusually high and locally variable concentrations of insulin-a focally increased number of enlarged mitochondria, abundant glycogen and a smaller amount of neutral fat droplets. Furthermore, hepatocytes and cells looking like hepatocytes (hepato-cyte-like cells) with typically structured cytoplasmic beta(insulin)granules were found bordering the islet isografts. These results could be interpreted as an expression of arteficial or nonarteficial fusion of beta cells with hepatocytes, i.e. formation of hybrid cells (“in vivo hybridization”). Alternatively, they might reflect insulin uptake and storage in the hepatocytes. In addition, these findings suggest that contact between neonatal islet tissue and liver tissue could be a trigger for the in vivo transformation (modulation) of differentiated cells of similar embryonic development in the adult organism.  相似文献

1. The utilization of [1-14C]palmitate by hepatocytes prepared from fed and starved neonatal and adult rats has been examined by measuring isotopic incorporation into various products. 2. In cells from fed adult rats the principal products were esters (triglycerides and phospholipids) but ketone bodies were the main metabolic end products in cells from starved adult and fed and starved neonatal rats. Production of triglycerides exceeded that of phospholipids in fed adult cells whereas phospholipid formation always predominated in neonatal cells. 3. The high rate of fatty acid oxidation and hence NADH formation by neonatal cells is reflected by a lower acetoacetate--3-hydroxybutyrate ratio at the earlier stages of incubation of neonatal cells. 4. The addition of glycerol modified quantitatively the products of palmitate metabolism by adult hepatocytes but no such effects were observed with neonatal cells. 5. Compared with adult cells, neonatal hepatocytes showed very low rates of lipogenesis that were only enhanced a little by addition of lactate/pyruvate and did not show any effects of glucose concentration upon incorporation of tritium from 3H2O into lipids.  相似文献
