Predictions in integrated pest management on the compatibility of an insecticide with biological control often are based on incomplete screening tests. While measuring levels of mortality from direct insecticide exposure is a very common screening method, possible sublethal effects as a result of either direct or indirect insecticide exposure remain relatively unknown. The impact of sublethal effects on the success of biological control can be as deleterious as mortality. Here, we report the reduced host foraging ability and longevity of the parasitoid Microplitis croceipes Cresson (Hymenoptera: Braconidae) after feeding on extrafloral nectar from cotton (Gossypium hirsutum L., Malvaceae) plants that were treated with systemic insecticides. The insecticides used in this study are regularly applied in cotton-growing areas in the United States. For all tested insecticides, longevity of M. croceipes females that fed on nectar from cotton was affected for at least 10 days after plants were treated with insecticides. Moreover, the parasitoid's host foraging ability was severely affected for periods ranging from 2 days (imidacloprid) to 18 days (aldicarb) after insecticide application. The consequences of these sublethal effects on the success of biological control are discussed.  相似文献

Birds of prey that are poisoned by cholinesterase inhibitors (e.g. organophosphate and carbamate insecticides) are often cared for at animal shelters, rehabilitation centres and wildlife diagnostic facilities. Plasma cholinesterase (ChE) activity is a recognized method of assessing exposure to these insecticides, but standard blood-handling protocols are difficult to follow in non-laboratory settings. The primary objective of this study was to expand upon a method for storing human blood on filter paper without the need for complicated equipment or refrigeration, and to test its utility for measurement of ChE activity in avian blood. ChE activity from whole blood, plasma, and dried blood spots was analysed from 169 wild birds and comparisons made among sample types. ChE activity measured in whole blood haemolysates and dried blood spots were significantly correlated (r = 0.74, p < 0.001), as was ChE activity measured in plasma and dried blood spots (r = 0.68, p < 0.001). This study demonstrated that monitoring pesticide exposure in birds could be conducted using elementary blood sampling, preserving and shipping techniques.  相似文献

小峰熊蜂Bombus hypocrita是我国优势熊蜂种群之一, 因其易于饲养、 群势较强且授粉性能优良而成为我国设施农业常用优良授粉蜂种, 但常受到以乙酰胆碱酯酶(AChE)为靶标酶的有机磷和氨基甲酸酯类杀虫剂的危害。 为合理规避这两类杀虫剂对熊蜂的危害, 同时也为完善熊蜂授粉配套技术和保护野生熊蜂资源提供理论基础, 本研究利用正交试验对小峰熊蜂头部乙酰胆碱酯酶活性的测定条件进行了优化, 并明确了6种常用有机磷和氨基甲酸酯类杀虫剂对乙酰胆碱酯酶活性的影响。结果表明: 各测定因素对小峰熊蜂乙酰胆碱酯酶活性测定影响的大小顺序依次为: 酶浓度>pH>温度>底物浓度>反应时间; 小峰熊蜂头部乙酰胆碱酯酶活性的最适反应条件为: 酶浓度0.25 g 蛋白质/L, 底物浓度0.8 mmol/L, pH值7.5, 温度40℃, 反应时间5 min。毒死蜱、 三唑磷、 丙溴磷、 异丙威、 仲丁威和残杀威6种杀虫剂对小峰熊蜂头部乙酰胆碱酯酶离体抑制作用均呈现明显的剂量-效应关系, 其抑制中浓度IC50分别为0.39, 1.79, 0.42, 0.04, 0.43和0.63 mmol/L。这6种杀虫剂对小峰熊蜂AChE抑制作用的强弱依次为: 异丙威>毒死蜱>三唑磷>仲丁威>残杀威>丙溴磷, 即小峰熊蜂对异丙威最敏感, 而对丙溴磷的敏感性最弱。  相似文献

The target enzymes, acetylcholinesterase (for phosphamidon and carbaryl) and Mg2+ ATPase (for DDT and fenvalerate) have been assayed during exposure and reclamation of these insecticides in M. monoceros. Toxicity of these insecticides are in the order: fenvalerate greater than DDT greater than carbaryl greater than phosphamidon. Reclamation studies show that fenvalerate is rapidly degradable while DDT is slowly degradable. It is suggested that pyrethroid, organophosphate and carbamate insecticides may be preferred over organochlorine compounds in farm operations.  相似文献

Background  

Pyrethroids and pyrethrins are widely used insecticides. Extensive applications not only result in pest resistance to these insecticides, but also may lead to environmental issues and human exposure. Numerous studies have shown that very high exposure to pyrethroids might cause potential problems to man and aquatic organisms. Therefore, it is important to develop a rapid and efficient disposal process to eliminate or minimize contamination of surface water, groundwater and agricultural products by pyrethroid insecticides. Bioremediation is considered to be a reliable and cost-effective technique for pesticides abatement and a major factor determining the fate of pyrethroid pesticides in the environment, and suitable esterase is expected to be useful for potential application for detoxification of pyrethroid residues. Soil is a complex environment considered as one of the main reservoirs of microbial diversity on the planet. However, most of the microorganisms in nature are inaccessible as they are uncultivable in the laboratory. Metagenomic approaches provide a powerful tool for accessing novel valuable genetic resources (novel enzymes) and developing various biotechnological applications.  相似文献

Several fruit fly species (Diptera: Tephritidae) are invasive pests that damage the quality of fruits in horticultural crops and cause significant value losses worldwide. Management of fruit flies mainly depends on conventional insecticides. Unfortunately, the application of synthetic insecticides has caused environmental pollution, risks for humans and animals, and development of resistance. Furthermore, controlling fruit flies by applying synthetic insecticides is challenging because fruit containing third instars often fall from the tree – subsequently the larvae leave the decaying fruits and pupate in the soil. Consequently, both larvae and pupae are protected from surface-applied insecticides in fruits and soil. So, there is a pressing need for more eco-friendly and selective control measures with new modes of action. Among such measures are entomopathogenic fungi (EPFs) and nematodes (EPNs). I gathered knowledge on past and present research about EPFs and EPNs as biocontrol agents against fruit flies to investigate approaches that may improve their capacities. I also highlighted several recommendations that may help future field studies on the suppression of fruit fly populations by EPFs and EPNs.  相似文献

A common method of adult mosquito control consists of residual application on surfaces and aerial spraying often using pyrethroids. However, not all insects that contact insecticides are killed. Sublethal exposure to neurotoxic compounds can negatively affect sensory organs and reduce efficiency of host location. Flight tracks of host-seeking female Culex quinquefasciatus, Anopheles albimanus, and Aedes aegypti in a wind tunnel were video-recorded to compare activation of host-seeking and patterns of flight orientation to host odors. During host-seeking flights, all three mosquito species differed significantly in flight duration, velocity, turn angle, and angular velocity. Mosquitoes were then exposed to sublethal levels (LD(25) ) of pyrethroid insecticides to evaluate the effects of the neurotoxicants 24 hours post-exposure. Significant reductions in time of activation to flight and flight direction were observed in mosquitoes exposed to deltamethrin and permethrin. Additionally, pesticide-treated Cx. quinquefasciatus mosquitoes flew significantly slower, spent more time in flight, and turned more frequently than untreated controls.  相似文献

We present a novel portable immunochromatographic electrochemical biosensor (IEB) for simple, rapid, and sensitive biomonitoring of trichloropyridinol (TCP), a metabolite biomarker of exposure to organophosphorus insecticides. Our new approach takes the advantage of immunochromatographic test strip for a rapid competitive immunoreaction and a disposable screen-printed carbon electrode for a rapid and sensitive electrochemical analysis of captured HRP labeling. Several key experimental parameters (e.g. immunoreaction time, the amount of HRP labeled TCP, concentration of the substrate for electrochemical measurements, and the blocking agents for the nitrocellulose membrane) were optimized to achieve a high sensitivity, selectivity and stability. Under optimal conditions, the IEB has demonstrated a wide linear range (0.1-100 ng/ml) with a detection limit as low as 0.1 ng/ml TCP. Furthermore, the IEB has been successfully applied for biomonitoring of TCP in the rat plasma samples with in vivo exposure to organophosphorus insecticides like Chlorpyrifos-oxon (CPF-oxon). The IEB thus opens up new pathways for designing a simple, rapid, clinically accurate, and quantitative tool for TCP detection, as well as holds a great promise for in-field screening of metabolite biomarkers, e.g., TCP, for humans exposed to organophosphorus insecticides.  相似文献

Pesticide pollution can alter parasite transmission, but scientists are unaware if effects of pesticides on parasite exposure and host susceptibility (i.e. infection risk given exposure) can be generalised within a community context. Using replicated temperate pond communities, we evaluate effects of 12 pesticides, nested in four pesticide classes (chloroacetanilides, triazines, carbamates organophosphates) and two pesticide types (herbicides, insecticides) applied at standardised environmental concentrations on larval amphibian exposure and susceptibility to trematode parasites. Most of the variation in exposure and susceptibility occurred at the level of pesticide class and type, not individual compounds. The organophosphate class of insecticides increased snail abundance (first intermediate host) and thus trematode exposure by increasing mortality of snail predators (top–down mechanism). While a similar pattern in snail abundance and trematode exposure was observed with triazine herbicides, this effect was driven by increases in snail resources (periphytic algae, bottom–up mechanism). Additionally, herbicides indirectly increased host susceptibility and trematode infections by (1) increasing time spent in susceptible early developmental stages and (2) suppressing tadpole immunity. Understanding generalisable effects associated with contaminant class and type on transmission is critical in reducing complexities in predicting disease dynamics in at‐risk host populations.  相似文献

Modifications of metabolic pathways are important in insecticide resistance evolution. Mutations leading to changes in expression levels or substrate specificities of cytochrome P450 (P450), glutathione-S-transferase (GST) and esterase genes have been linked to many cases of resistance with the responsible enzyme shown to utilize the insecticide as a substrate. Many studies show that the substrates of enzymes are capable of inducing the expression of those enzymes. We investigated if this was the case for insecticides and the enzymes responsible for their metabolism. The induction responses for P450s, GSTs and esterases to six different insecticides were investigated using a custom designed microarray in Drosophila melanogaster. Even though these gene families can all contribute to insecticide resistance, their induction responses when exposed to insecticides are minimal. The insecticides spinosad, diazinon, nitenpyram, lufenuron and dicyclanil did not induce any P450, GST or esterase gene expression after a short exposure to high lethal concentrations of insecticide. DDT elicited the low-level induction of one GST and one P450. These results are in contrast to induction responses we observed for the natural plant compound caffeine and the barbituate drug phenobarbital, both of which highly induced a number of P450 and GST genes under the same short exposure regime. Our results indicate that, under the insecticide exposure conditions we used, constitutive over-expression of metabolic genes play more of a role in insect survival than induction of members of these gene families.  相似文献

【目的】评价新烟碱类杀虫剂对地熊蜂Bombus terrestris工蜂的毒性和生态风险性,为温室施用新烟碱类杀虫剂提供科学依据。【方法】分别采用饲喂法和接触法测定了噻虫嗪、噻虫胺、啶虫脒、吡虫啉、烯啶虫胺、呋虫胺、噻虫啉和氟吡呋喃酮8种新烟碱类杀虫剂对地熊蜂成年工蜂的急性经口和急性接触毒性。同时评估了8种新烟碱类杀虫剂对地熊蜂工蜂的生态风险性。【结果】8种杀虫剂经饲喂法测定,噻虫胺对地熊蜂成年工蜂的毒性最高,24 h和48 h的LD 50值分别为0.0433和0.0330μg a.i./蜂;氟吡呋喃酮毒性最低,24 h和48 h的LD 50值分别为72.4119和67.9079μg a.i./蜂。接触法测定的毒性与饲喂法测得的结果一致,噻虫胺的毒性最高,24 h和48 h的LD 50值分别为0.0220和0.0192μg a.i./蜂;氟吡呋喃酮的毒性最低,24 h和48 h的LD 50值分别为141.7641和130.3062μg a.i./蜂。生态风险评估表明,啶虫脒、噻虫啉和氟吡呋喃酮对地熊蜂成年工蜂的经口毒性和接触毒性均表现为低风险,吡虫啉、烯啶虫胺和呋虫胺的毒性表现为中等风险。噻虫嗪和噻虫胺对地熊蜂成年工蜂的经口毒性表现为中等风险,而接触毒性则表现为高风险。【结论】检测的8种新烟碱类杀虫剂中啶虫脒、噻虫啉与氟吡呋喃酮对地熊蜂成年工蜂的毒性为低毒,而噻虫嗪、噻虫胺、吡虫啉、烯啶虫胺、呋虫胺这5种杀虫剂均为高毒。在设施蔬菜花期使用地熊蜂授粉时,建议禁用这5种中、高风险的新烟碱类杀虫剂,以避免对熊蜂授粉的危害,而另3种低风险药剂可根据田间试验情况合理施用。  相似文献

The number of honey bee colonies in the United States has declined to half of its peak level in the 1940s, and colonies lost over the winter have reached levels that are becoming economically unstable. While the causes of these losses are numerous and the interaction between them is very complex, the role of insecticides has garnered much attention. As a result, there is a need to better understand the risk of insecticides to bees, leading to more studies on both toxicity and exposure. While much research has been conducted on insecticides and bees, there have been very limited studies to elucidate the role that bee genotype and age has on the toxicity of these insecticides. The goal of this study was to determine if there are differences in insecticide sensitivity between honey bees of different genetic backgrounds (Carniolan, Italian, and Russian stocks) and assess if insecticide sensitivity varies with age. We found that Italian bees were the most sensitive of these stocks to insecticides, but variation was largely dependent on the class of insecticide tested. There were almost no differences in organophosphate bioassays between honey bee stocks (<1-fold), moderate differences in pyrethroid bioassays (1.5 to 3-fold), and dramatic differences in neonicotinoid bioassays (3.4 to 33.3-fold). Synergism bioassays with piperonyl butoxide, amitraz, and coumaphos showed increased phenothrin sensitivity in all stocks and also demonstrated further physiological differences between stocks. In addition, as bees aged, the sensitivity to phenothrin significantly decreased, but the sensitivity to naled significantly increased. These results demonstrate the variation arising from the genetic background and physiological transitions in honey bees as they age. This information can be used to determine risk assessment, as well as establishing baseline data for future comparisons to explain the variation in toxicity differences for honey bees reported in the literature.  相似文献

Laboratory toxicity studies were conducted in southeastern Queensland, Australia, to determine the acute lethal effects of a 1-h pulse exposure of selected insecticides to adult and juvenile (<72 h old) crimson-spotted rainbowfish, Melanotaenia duboulayi (Castlenau). In addition, to its ecological significance, this native fish is a predator of mosquitoes. Two organophosphate (OP) compounds (temephos and pirimiphos-methyl), an entomophathogenic bacterium (Bacillus thuringiensis variety israelensis [Bti]) and two insect growth regulators (IGRs) (s-methoprene and pyriproxyfen) were evaluated. Although none of the five insecticides were acutely toxic to adult M. duboulayi under the test conditions, temephos and pirimiphos-methyl were found to be toxic to juveniles, with 24 h pulse exposure LC50 values of 27 and 15 microg/liter (ppb), respectively. Of the two OPs, pirimiphos-methyl was the most toxic, with a lethal dose ratio (pulse exposure LC50 temephos/ pulse exposure LC50 pirimiphos-methyl) of 1.8 (95% CL 0.5-6.4). These pulse exposure LC50 values represented 40 and 4.5% of the estimated environmental concentrations (EEC) for a 15 cm deep water body, respectively. Bti and the two IGRs had no acute toxic effects at up to 10 and 12.5 times the EEC, respectively. Accordingly, in the interests of environmental conservation and integrated pest management (IPM), preference should be given to the latter three insecticides for control of mosquito larvae in juvenile M. duboulayi habitat.  相似文献

Currently, there is an elevated interest in reducing feeding damage caused by the Nantucket pine tip moth, Rhyacionia frustrana (Comstock), a common regeneration pest of loblolly pine, Pinus taeda L. The toxicity of several insecticides was tested in a laboratory against four common R. frustrana parasitoids. There were no differences in parasitoid mortality between the control and indoxacarb treatments. However, the pyrethroids, permethrin and lambda-cyhalothrin, caused significantly more mortality initially (up to 240 min exposure time) than other insecticides. Spinosad was less toxic than the pyrethroids initially, but the spinosad related mortality increased with time until it reached a level similar to the pyrethroids. For the most part, spinosad and the pyrethroids caused more mortality than the control and indoxacarb treatmtents within the 1-d sample period. These results may have important implications for decisions concerning which insecticides are best suited for reducing pest damage while conserving natural enemies in timber and agricultural systems. Large-scale field trials are required to further define the impacts of these insecticides on natural enemies.  相似文献

The effect of exposure of Aedes aegypti larvae to sub-lethal doses of the pyrethroid insecticide permethrin, the organophosphate temephos, the herbicide atrazine, the polycyclic aromatic hydrocarbon fluoranthene and the heavy metal copper on their subsequent tolerance to insecticides, detoxification enzyme activities and expression of detoxification genes was investigated. Bioassays revealed a moderate increase in larval tolerance to permethrin following exposure to fluoranthene and copper while larval tolerance to temephos increased moderately after exposure to atrazine, copper and permethrin. Cytochrome P450 monooxygenases activities were induced in larvae exposed to permethrin, fluoranthene and copper while glutathione S-transferase activities were induced after exposure to fluoranthene and repressed after exposure to copper. Microarray screening of the expression patterns of all detoxification genes following exposure to each xenobiotic with the Aedes Detox Chip identified multiple genes induced by xenobiotics and insecticides. Further expression studies using real-time quantitative PCR confirmed the induction of multiple CYP genes and one carboxylesterase gene by insecticides and xenobiotics. Overall, this study reveals the potential of xenobiotics found in polluted mosquito breeding sites to affect their tolerance to insecticides, possibly through the cross-induction of particular detoxification genes. Molecular mechanisms involved and impact on mosquito control strategies are discussed.  相似文献

BackgroundThe current body of research on insecticide use in Peru deals primarily with application of insecticides offered through Ministry of Health-led campaigns against vector-borne disease. However, there is a gap in the literature regarding the individual use, choice and perceptions of insecticides which may influence uptake of public health-based vector control initiatives and contribute to the thousands of deaths annually from acute pesticide poisoning in Peru.MethodsResidents (n = 49) of the Alto Selva Alegre and CC districts of peri-urban Arequipa participated in seven focus group discussions (FGD). Using a FGD guide, two facilitators led the discussion and conducted a role-playing activity. this activity, participants insecticides (represented by printed photos of insecticides available locally) and pretended to “sell” the insecticides to other participants, including describing their qualities as though they were advertising the insecticide. The exercise was designed to elicit perceptions of currently available insecticides. The focus groups also included questions about participants’ preferences, use and experiences related to insecticides outside the context of this activity. Focus group content was transcribed, and qualitative data were analyzed with Atlas.ti and coded using an inductive process to generate major themes related to use and choice of insecticides, and perceived risks associated with insecticide use.ResultsThe perceived risks associated with insecticides included both short- and long-term health impacts, and safety for children emerged as a priority. However, in some cases insecticides were reportedly applied in high-risk ways including application of insecticides directly to children and bedding. Some participants attempted to reduce the risk of insecticide use with informal, potentially ineffective personal protective equipment and by timing application when household members were away. Valued insecticide characteristics, such as strength and effectiveness, were often associated with negative characteristics such as odor and health impacts. “Agropecuarios” (agricultural supply stores) were considered a trusted source of information about insecticides and their health risks.ConclusionsIt is crucial to characterize misuse and perceptions of health impacts and risks of insecticides at the local level, as well as to find common themes and patterns across populations to inform national and regional programs to prevent acute insecticide poisoning and increase community participation in insecticide-based vector control campaigns. We detected risky practices and beliefs about personal protective equipment, risk indicators, and safety levels that could inform such preventive campaigns, as well as trusted information sources such as agricultural stores for partnerships in disseminating information.  相似文献

Quantitative studies concerning total and differential haemocyte counts and abnormalities were performed under laboratory conditions for larvae, pupae and adults collected from a wild Apis dorsata colony. Haemolymph samples were observed immediately, thirty and sixty minutes after field recommended concentration exposure of five different insecticides. Total haemocyte counts were significantly higher for larvae and pupae but less for adult bees, however, differential haemocyte counts insignificantly different. Exposure of insecticides showed variable response for tested insecticides with immediate increased change with ethofenprox, diafenthiuron and imidacloprid but decreased for all tested insecticides after sixty minutes. For differential haemocyte counts, plasmatocytes and granulocytes increased with exposure of insecticides. Immune response of haemocytes against insecticides showed different degrees of abnormalities like agglutination, denucleation and cell shape distortion. Such studies may help in possible identification of insect defense mechanisms against their exposure to external hazards for instance insecticide exposure.  相似文献

Abstract Field doses of six selected insecticides were tested against the immature (pupae) and mature (adult) stages of Diadegma semiclausum (Hellén) and Oomyzus sokolowskii (Kurdjumov), parasitoids of the diamondback moth, Plutella xylostella (L.). Effects of contact toxicity (direct spraying) of the six insecticides on emergence of parasitoids were found negligible on both species except permethrin which caused 37.5% mortality. All adults of both parasitoid species died 24 hours after exposure to chlorfenapyr, emamectin benzoate and permethrin. In contrast, the three insect growth regulators (IGRs), chlorfluazuron, flufenoxuron and teflubenzuron, were found harmless to both species, and adult mortality of both parasitoid species was 0–16.7%. However, parasitism by the females of both parasitoid species was severely impaired when the females were offered the three IGR diluted solutions for 24 hours. Effects of oral toxicities of the IGRs on longevity of both parasitoids after 12 hours exposure were found to be significantly different between males and females. Compatibility of tested insecticides with D. semiclausum and O. sokolowskii and integration of compatible insecticides with these parasitoids in integrated pest management programs of crucifers are discussed.  相似文献
