To examine the effects of ovarian hormones on the behavior of female Macaca fascicularis and their male partners, daily 1-hr behavior tests were conducted while ovariectomized females were (1) untreated, (2) given estradiol benzoate (EB) (5 μg subcutaneously [s.c.]/day), (3) given estradiol benzoate together with increasing doses of progesterone (P) (5 mg, 10 mg, and 20 mg. s.c./day), and (4) given testosterone propionate (TP) (0.25 mg s.c./day) (six pairs, 540 tests). Weekly blood samples were analyzed by radioimmunoassay for plasma hormone levels (81 samples). Estrogen treatment produced plasma estradiol levels similar to those of intact females during the late follicular phase of the menstrual cycle. Additional progesterone at the lowest dose produced plasma progesterone levels similar to or somewhat higher than those during the midluteal phase, while higher doses produced supraphysiological levels. Androgen treatment resulted in plasma levels well above the physiological range. Hormone treatments produced highly significant effects on the sexual, social, and aggressive interactions of the pairs. As in rhesus monkeys, estrogen increased male and female sexual activity, and increasing doses of additional progesterone reversed these effects. Unlike in rhesus monkeys, testosterone propionate increased both female sexual motivation (invitations) and also male sexual activity and ejaculatory performance. The direction of the hormone-dependent changes in grooming and aggressive interactions confirmed earlier results with intact females and indicated that aggressive interactions and male grooming times were highest, and female grooming times were lowest, when copulatory activity was at its height.  相似文献

The effects of estrogen and progesterone on the social and sexual behavior of brown lemmings, Lemmus trimucronatus, were investigated. The behavior of hormone-treated and untreated ovariectomized females and sexually vigorous males was observed in six consecutive daily 5-min dyadic encounters. Sexual receptivity, as measured by lordosis, and other social behaviors including nasonasal contact, boxing postures, allogrooming, perineal investigation, and male mounting increased following 48 hr of exposure to daily injections of 0.5 μg estradiol benzoate (EB). Lordosis in EB-primed females was not facilitated or inhibited by short-term (4 hr) exposure to 0.5 mg progesterone (P). Long-term (greater than 24 hr) exposure to P apparently inhibited lordosis and other social behaviors in EB-treated females, although males continued to attempt to mount these females. In EB-treated females a dramatic increase in threat-leaps, directed by the female toward the male, was observed within 4 hr of P injection. Threat-leaps declined when P was withdrawn. Threat-leaps were also observed in ovariectomized females after prolonged exposure to P only (0.5 mg/day). Vaginal perforation and cornification were first apparent 48 hr after EB injection. P-alone treated ovariectomized females also showed vaginal perforation but cornified cells were infrequent and these animals did not show lordosis.  相似文献

The effects of repeated estradiol benzoate (1.0 or 3.3 μg) and progesterone (0.5 mg) injections and mating experiences (10 min or 4 hr) were examined in ovariec-tomized female Mongolian gerbils (Meriones unguiculatus) paired with sexually vigorous but unfamiliar males. Estradiol benzoate alone for 2 days did not elicit female sexual responses. Estradiol benzoate for 2 days followed by progesterone facilitated lordosis in the female. In females injected daily with 1.0 μg estradiol benzoate, progesterone exposure for approximately 1 day produced a significant inhibition in subsequent lordosis, although these females continued to respond to the male with lordosis quotients of approximately 60. A 4-hr mating experience produced a nearly complete elimination of sexual activity in tests observed 24 hr later with a fresh male. Footstomping was positively associated with sexual activity in both sexes, although females footstomped more often in the 1.0 rather than the 3.3-μg estradiol benzoate condition. Fighting was most frequent in tests in which lordosis quotients were low. Interactions among copulatory experiences and inhibitory effects of progesterone summed to terminate female sexual receptivity and alter other social interactions in the gerbil.  相似文献

Social and sexual incentive motivation, defined as the intensity of approach to a social and a sexual incentive, respectively, were studied in female Swiss Webster mice. In the first experiment, the social incentive was a castrated mouse of the same strain as the females, whereas the sexual incentive was an intact male mouse of the same strain. Ovariectomized females were first tested after oil treatment and then after administration of estradiol benzoate + progesterone in doses sufficient to induce full receptivity. The hormones increased sexual incentive motivation while leaving social incentive motivation unaffected. This suggests that sexual incentive motivation in the female mouse is dependent on ovarian hormones. In the next experiment, ovariectomized females were tested with an intact, male estrogen receptor α knockout and its wild type as incentives, first without hormones and then when fully receptive. There were no differences in incentive properties between the wild type and the knockout. In a similar experiment, we used an intact male estrogen receptor β knockout and its corresponding wild type as incentives. The wild type turned out to be a more attractive social incentive than the knockout, while they were equivalent as sexual incentives. Finally, an intact male oxytocin knockout and its wild type were used as incentives. The knockout turned out to be a superior incentive, particularly a superior sexual incentive. The fact that the estrogen receptor β and oxytocin knockouts have incentive properties different from their wild types may be important to consider in studies of these knockouts' sociosexual behaviors.  相似文献

In a three year study, the interactions of social, spatial and hormonal variables on the behavior of rhesus monkeys were investigated. Observations were made on (1) an oppositelysexed pair alone in a cage; (2) the pair alone in a large room; and (3) the pair together with three ovariectomized females in the same large room. Each of four males was tested with the same group of four ovariectomized females (16 pairs, 480 1-hr tests). Each female served in turn as the focal female, namely, the hormone-treated one, and received first estradiol benzoate (EB) alone and then in combination with progesterone (P). The presence of three other females (the social variable) had a marked effect on all aspects of the behavior of the focal pair, and reduced the male's ejaculations as well as other sexual and affiliative interactions with the focal female. This was due to an inhibitory influence exerted by the dominant female on the interactions of subordinate females with the male. Comparing data from two groups of females showed that female dominance rank, and not male sexual preference, was the critical factor, and that mate competition between female rhesus monkeys was operating. Decreasing the space available for the interactions of the pair appeared to imitate a bonding effect in the male, increasing male grooming and tending to increase male sexual activity. In the group setting, treating the female with EB increased sexual activity, and additional progesterone decreased it in all three test settings. The effects of progesterone were most clearly apparent when the pair was tested alone in a large area.  相似文献

The induction of sexual receptivity and its maintenance after copulation in ovariectomized female golden hamsters (Mesocricetus auratus) was found to be a function of the levels of ovarian hormones administered. Various combinations of estradiol benzoate (between 0.6 and 666 μg) and progesterone (between 0.05 and 5.0 mg) were administered in two experiments. Although some animals responded at 0.6 μg, higher levels of estradiol benzoate (1–6 μg or more) were more effective in inducing levels of lordosis equivalent to those seen in intact females in natural estrus. After mating, a depression in lordosis was observed in both ovariectomized and intact females. However, in ovariectomized females (excluding animals that did not respond initially) the duration of postcopulatory receptivity was a function of the level of progesterone administered. High levels of progesterone tended to prolong slightly the duration of postcopulatory receptivity.  相似文献

Octodon degus, a social hystricomorph rodent, responds to olfactory cues from a gonadally intact female entrained to a light-dark cycle (LD) by accelerating reentrainment of running wheel activity following a 6-h phase advance of the LD cycle. In this study, we examined the role of ovarian hormones in the production of and responsiveness to olfactory social cues in females. Experiment 1: intact females were sequentially phase-advanced 6 h with photic cues alone, or in the presence of an intact female donor, ovariectomized (OVX) donor, a castrated male, or a castrated male with testosterone replacement. Acceleration of reentrainment occurred only in the presence of the intact female donor while reentrainment was delayed by OVX donors. Experiment 2: OVX females undergoing a 6-h phase advance did not accelerate reentrainment in the presence of an intact female donor compared to reentrainment with photic cues alone. Thus, ovarian hormones are necessary for both the production of and responsiveness to olfactory cues. Experiment 3: OVX females implanted with estrogen-filled Silastic capsules did not accelerate reentrainment following the 6-h phase advance in the presence of an intact donor, whereas animals implanted with a combination of estrogen- and progesterone-filled capsules (Experiment 4) reduced the length of time needed to reentrain in the presence of an intact donor. Therefore, combined progesterone and estrogen are sufficient for responsiveness to the effective olfactory cue in intact donor females. These data clarify that the sex difference in sensitivity to non-photic odor effects on circadian reentrainment is caused by both the testosterone's inhibitory effects (Octodon degus. J. Biol. Rhythms 18 (2003) 43-50) and the enhancing effects of progesterone and estrogen.  相似文献

These experiments were designed to test the effects of chronic estradiol treatment on aggression and sexual behavior in female hamsters. Isolated female hamsters were ovariectomized and tested for their behavioral responses to a group-housed, ovariectomized female hamster (aggression test) and a group-housed, intact male hamster (sexual behavior test). Following these baseline tests, the experimental females were implanted sc with Silastic capsules containing different concentrations of estradiol (100, 25, 10, or 0%) diluted with cholesterol and retested 3, 7, 10, and 14 days after implantation. High levels of aggression were observed on the baseline test, with no changes in aggression toward an intruder female observed for any implant group on subsequent tests. Despite these high levels of aggression toward another female, most of the estradiol-treated females (80% at 14 days) were sexually responsive in the presence of a male. There was no effect of Silastic estradiol concentration on sexual behavior, even though a range of serum estradiol levels (39–105 pg/ml) resulted. Lordosis latencies decreased and lordosis durations increased over the extent of estradiol treatment. Seventeen days after Silastic implantation, all females were injected with progesterone and retested. Estradiol-treated females showed an extreme reduction in aggression toward a stimulus female, as well as a further stimulation of sexual behavior after progesterone treatment. High levels of aggression in cholesterol-treated females (0% estradiol) were maintained even after progesterone injection, and these females never displayed any sexual responsivity. These results suggest that sexual behavior in the female hamster is sensitive to estradiol alone, whereas the inhibition of aggression requires the combination of estradiol plus progesterone.  相似文献

Novel male mice can accelerate reproductive maturation in proximal developing females, an effect mediated by the chemistry of the males' urine. Exogenous estrogens can similarly accelerate female sexual development. In Experiment 1, adult male mice were housed across wire grid from either empty compartments or those containing post-weanling females. Proximity of females caused males to urinate more, progressively over days of exposure, with most urination directed towards females' compartments. Male urine collected after 5 days in these conditions was analyzed by enzyme immunoassay for 17β-estradiol, testosterone, and creatinine. Urinary creatinine of isolated males significantly exceeded that of female-exposed males. Unadjusted urinary steroids also trended toward higher levels in isolates, but creatinine-adjusted estradiol and testosterone of female-exposed males significantly exceeded that of isolated males. In Experiment 2, measurement of water consumption indicated significantly greater drinking by female-exposed as opposed to isolated males. In Experiment 3, males were housed in isolation or beside post-weanling intact (sham-operated) females, ovariectomized females, or intact (sham-operated) males. Male water consumption was elevated in all conditions involving social contact. Urinary creatinine was significantly lower in female-exposed males compared to isolated controls, while unadjusted testosterone was significantly lower in males in all social conditions. Again, creatinine-adjusted estradiol in female-exposed males significantly exceeded that of isolates. These data indicate that adult males drink and urinate more, have more dilute urine, and have a higher ratio of estradiol to creatinine when they are near developing females. These dynamics increase females' exposure to urinary steroids and other urinary constituents that can hasten sexual maturity.  相似文献

The mechanisms involved in the control of precocious sexual receptivity were studied in 4-day cyclic female Wistar rats injected with 10 μg estradiol benzoate (EB) and caged with a male during the night from diestrus II to proestrus. Early mating frequencies were compared in intact females, in animals ovariectomized on the morning of diestrus I, in adrenalectomized and in adrenalectomized-ovariectomized females. No change in early sexual receptivity occurred either in ovariectomized, or in adrenalectomized animals. On the contrary, a significant decrease of precocious mating frequencies was noted in adrenalectomized-ovariectomized females. The role played by the ovary in the control of precocious receptivity was supposed to be due to the secretion of progesterone which has been evidenced on the late afternoon of diestrus II in estrogen treated females.Concerning the mechanisms by which the adrenals may compensate for the ovaries in the control of early sexual receptivity in estrogen-primed females it was observed that notwithstanding an inhibitory action exerted by EB on the adrenal progesterone secretion, a low rate of progesterone was maintained in the peripheral plasma which was compatible with early mating in ovariectomized animals.  相似文献

The neural mechanisms controlling sexual behavior are sexually differentiated by perinatal actions of gonadal hormones. We recently observed using female mice deficient in alpha-fetoprotein (AFP-KO) and which lack the protective actions of AFP against maternal estrogens, that exposure to prenatal estrogens completely defeminized their potential to show lordosis behavior in adulthood. Therefore, we determined here whether mate preferences were also affected in female AFP-KO mice. We observed a robust preference for an estrous female over an intact male in female AFP-KO mice, which were ovariectomized in adulthood and subsequently treated with estradiol and progesterone, whereas similarly treated WT females preferred the intact male over the estrous female. Gonadally intact WT males preferred the estrous female over the male, but only when visual cues were blocked by placing stimulus animals behind opaque partitions. Furthermore, when given the choice between an intact male and a castrated male, WT females preferred the intact male, whereas AFP-KO females showed no preference. Finally when given the choice between an estrous female and an ovariectomized female, WT males preferred the estrous female whereas AFP-KO females preferred the ovariectomized female or showed no preference depending on whether they could see the stimulus animals or not. Taken together, when AFP-KO females are tested under estrous conditions, they do not show any male-directed preferences, indicating a reduced sexual motivation to seek out the male in these females. However, they do not completely resemble males in their mate preferences suggesting that the male-typical pattern of mate preferences is not solely organized by prenatal estrogens.  相似文献

Intact male squirrel monkeys were paired with ovariectomized females given either oil vehicle, estradiol benzoate (10 μg) alone, or estradiol benzoate combined with progesterone (5 mg). Affiliative behaviors, including social contacts, female grasping of the male, male and female approaches, and following by the male were observed more frequently, and social distance decreased when females received estradiol as compared to oil. Furthermore, when progesterone was combined with estradiol, some of the effects seen with estradiol alone, such as grasping by the female and following by the male, were reversed. These results support the hypothesis that seasonal changes between sex-segregated and sex-integrated forms of social organization reported for this species are influenced by endocrine conditions of the female.  相似文献

The present study investigates the effects of the antiestrogen ICI 182,780 (ICI) on the display of sexual behaviors in female rats. ICI 182,780 is a pure anti-estrogen and when given systemically, ICI is thought to act only in the periphery, and is not believed to cross the blood brain barrier. The present study examines the effects of ICI on sexual receptivity and on paced mating behavior following treatment with estradiol benzoate (EB) and progesterone (P) (Experiment 1) or with EB alone (Experiment 2). In Experiment 1, ICI (250.0 microg) did not affect the display of receptivity or paced mating behavior induced by EB and P. In contrast, in Experiment 2 female rats receiving EB alone displayed a decrease in the level of sexual receptivity following treatment with 500.0 and 750.0 microg ICI (but not 250.0 microg ICI). In addition, in Experiment 2 EB-treated female rats receiving 250.0 microg ICI spent more time away from the male rat following an intromission and were more likely to exit from the male compartment following a mount. Last, ICI had potent antiestrogenic effects on vaginal cytology (Experiment 2) and on the uterus (Experiments 1 and 2). The present study supports a role for peripheral estrogen receptors in sexual receptivity and paced mating behavior and suggests that estrogen receptor activation may decrease the aversive sensation associated with sexual stimulation.  相似文献

The effect of daily injections of estradiol benzoate (1 or 10 micrograms) and of progesterone (10 mg) on chin marking activity, sexual receptivity, and emission of nipple-search pheromone in ovariectomized rabbits was investigated. Both estradiol treatments resulted in a significant increase in all three measures over baseline and control group levels within 1-3 days, and withdrawal in a return to pretreatment levels within 2 weeks (Experiment I). In contrast, the administration of progesterone to such estradiol-primed does resulted in an almost immediate suppression of chin marking and lordosis, but in marked enhancement of pheromone emission and aggressive behavior (Experiment II). However, progesterone given alone to nonprimed does had no effect on any of these measure (Experiment III). The response profiles resulting from these treatments correspond well to patterns reported for intact does during estrus (= estradiol alone), pregnancy (= estradiol plus progesterone), and at parturition (= progesterone withdrawal).  相似文献

Mating terminates behavioral estrus in the female lizard, Anolis carolinensis. Postcopulatory sexual inhibition was not observed in females receiving estradiol benzoate (EB) in 10-mm Silastic implants (0.025-in. i.d. × 0.047-in. o.d.). To determine the role of the ovaries in mating-induced inhibition, intact and ovariectomized females received either a 6-mm EB implant or a 0.8-μg EB injection. Ovariectomized females remained sexually receptive after copulation while intact females were no longer receptive. Progesterone was implicated in the regulation of postcopulatory sexual receptivity. Several models are proposed to explain these results, and the adaptive significance of coition-induced sexual inhibition is discussed.  相似文献

The cycling female rat spontaneously regulates (paces) the amount and timing of cervical-vaginal stimulation received during mating. Female pacing of coital contacts increases the ability of vaginal intromissions to induce luteal function and to abbreviate the period of behavioral estrus. In the experiments reported here, we examined whether paced mating results in alterations in serum steroid concentrations, which might contribute to these processes. In Experiment 1, estradiol (E2), progesterone (P), testosterone (T), 5 alpha-dihydrotestosterone (DHT), and 5 alpha-androstane-3 alpha, 17 beta-diol (3 alpha-androstanediol, 3 alpha-diol) were measured in sera obtained from independent groups of animals between 0 min and 12 h after mating onset. Levels of 3 alpha-diol were significantly higher at 30 min in females pacing coital stimulation (Paced) than in females receiving mounts-without-intromission (Mounts-Only) or solitary exposure to the test chamber. Serum T, E2, P, and DHT did not differ between these groups at any time. P and 3 alpha-diol levels declined significantly for all groups across the 12-h post-treatment period. In Experiment 2, serum 3 alpha-diol measured over 3h after mating was compared in females receiving Paced stimulation with females receiving temporally unregulated coital stimulation (Non-Paced), or Mounts-Only. At 20 min, serum 3 alpha-diol concentrations were significantly higher in Paced than in Non-Paced and Mounts-Only females. In Experiment 3, Paced and Non-Paced stimulation did not differentially effect the proportion of females becoming pseudopregnant/pregnant. The selective increase in serum 3 alpha-diol in females that pace matings is discussed with regard to the known inhibitory effects of 5 alpha-reduced androgens on sexual receptivity.  相似文献

Zearalenone is a resorcylic acid lactone compound that is produced by fungal infection of edible grains and is believed to influence reproduction by binding to estrogen receptors. In order to study the potential estrogenic effects of this compound in the brain, we examined the effects of zearalenone on the expression of neuronal progestin receptors and feminine sexual behavior in female rats. Ovariectomized rats were treated with zearalenone (0.2, 1.0, or 2.0 mg), estradiol benzoate, or vehicle daily for 3 days. They were then either perfused, and progestin receptors visualized by immunocytochemistry, or injected with progesterone and tested for sexual receptivity with male rats. Progestin receptor-containing cells were counted in the medial preoptic area and ventromedial hypothalamus. The two highest doses of zearalenone increased the concentration of neuronal progestin receptors, as did 10 microg of estradiol. The highest dose of zearalenone (2 mg) also induced progestin receptor staining density comparable to that of 10 microg of estradiol benzoate. In behavioral tests, ovariectomized animals treated with 2 mg of zearalenone followed by progesterone showed levels of sexual receptivity comparable to females treated daily with estradiol benzoate (2 microg) followed by progesterone. These studies suggest that, although structurally distinct and less potent than estradiol, zearalenone can act as an estrogen agonist in the rat brain.  相似文献

Heterosexual interactions of pairs of stumptail macaques (Macaca arctoides) were studied in relation to the female menstrual cycle and after ovariectomy. Five intact male and 10 tubal-ligated female macaques were observed in laboratory pair tests of 20-mins duration, and data were obtained on various male and female behaviors. Each male was tested with the same two females during four 40-day observation periods. Males were tested daily and females were tested every other day. After two 40-day testing periods, one female partner of each male was ovariectomized and the other was sham-operated. Blood was collected regularly from the females during the course of observation and serum levels of estradiol and progesterone were determined by radioimmunoassay. The midcycle peak of estradiol was observed to occur approximately 18 days prior to menses. A distinct secondary peak in estradiol was observed to occur during the luteal phase of all cycles examined. Of 28 different male and female behaviors studied only female presentation to male sexual contact showed a significant midcycle peak related to the endogenous estradiol surge. After ovariectomy a significant decrease in the frequency of several male copulatory behaviors was observed, but most males continued to copulate regularly with their spayed partners throughout the period of this study. Thus, the pattern of copulatory behavior observed in stumptail macaques over the cycle of the female partner and subsequent to ovariectomy differs from that observed in other macaque species studied in the laboratory. It is concluded that cyclical fluctuations in the level of ovarian hormones are not significantly related to measures of sexual interactions in laboratory tests of this species, although the maintenance of copulation and associated behaviors at high levels depends to some degree upon the ovary.  相似文献

Two experiments were undertaken to examine the stimulation of home-cage and/or maternal aggressiveness by a hormonal treatment stimulating short-latency maternal behavior. Nonpregnant ovariectomized rats were treated with a 16-day regimen providing pregnancy levels of estrogen (E, 5-mm Silastic capsule) and progesterone (P, daily injection of 4 mg) followed by E and P withdrawal, with or without a terminal injection of estradiol benzoate (EB, 5 micrograms/kg). In Experiment 1, hormonally treated and control females were exposed continuously to pups and tested for aggression toward male intruders on the fifth day of pup exposure. Females receiving E/P/Oil and E/P/EB were highly aggressive whether or not they had yet shown maternal behavior, whereas vehicle-treated females were nonaggressive. In Experiment 2, hypophysectomized (HYPX) and Sham-HYPX females received either E/P/EB or a control treatment and were tested with male intruders (a) immediately preceding and (b) on the fifth day of continuous pup exposure. HYPX and Sham-HYPX females treated with E/P/EB were almost equally aggressive both preceding and following pup exposure (during which they initiated maternal care), whereas HYPX and Sham-HYPX vehicle-treated females were nonaggressive at both tests. In contrast, maternal behavior latencies were reduced by E/P/EB only among Sham-HYPX females. The results establish that an E/P/EB-treatment which elicits short-latency maternal responses also increases aggressiveness toward intruders. Pituitary products, although involved in the mediation of maternal responsiveness, do not contribute significantly to the stimulation of female aggressiveness by ovarian hormones.  相似文献

Advances in magnetic resonance imaging are driving the development of higher-resolution machines equipped with high-strength static magnetic fields (MFs). The behavioral effects of high-strength MFs are largely uncharacterized, although in male rats, exposure to 7 T or above induces locomotor circling and leads to a conditioned taste avoidance (CTA) if paired with a novel taste. Here, the effects of MFs on male and female rats were compared to determine whether there are sex differences in behavioral responses and whether these can be explained by ovarian steroid status. Rats were given 10-min access to a novel saccharin solution and then restrained within a 14-T magnet for 30 min. Locomotor activity after exposure was scored for circling and rearing. CTA extinction was measured with two-bottle preference tests. In experiment 1, males were compared with females across the estrous cycle after a single MF exposure. Females circled more and acquired a more persistent CTA than males; circling was highest on the day of estrus. In experiment 2, the effects of three MF exposures were compared among intact rats, ovariectomized females, and ovariectomized females with steroid replacement. Compared with intact rats, ovariectomy increased circling; estrogen replacement blocked the increase. Males acquired a stronger initial CTA but extinguished faster than intact or ovariectomized females. Thus the locomotor circling induced by MF exposure was increased in females and modulated by ovarian steroids across the estrous cycle and by hormone replacement. Furthermore, female rats acquired a more persistent CTA than male rats, which was not dependent on estrous phase or endogenous ovarian steroids.  相似文献
