Over the past decade, molecular, biochemical and cellular markers have been extensively used in pollution monitoring of aquatic environments. Biochemical markers have been selected among early molecular events occurring in the toxicological mechanisms of main contaminants. This paper assesses the marine environment quality along the Tunisian coasts using a statistical approach. Clams (Ruditapes decussatus) were collected during the four seasons of 2003 on seven different sites from the Tunisian coasts. Oxidative stress was evaluated in gills using catalase activity (Cat), neutral lipids and malonedialdehyde accumulation. Glutathione S-transferase activity is related to the conjugation of organic compounds and was evaluated in both, gills and digestive glands. Acetylcholinesterase activity was evaluated as the biomarker of exposure to organophosphorous, carbamate pesticides and heavy metals. For each biomarker, a discriminatory factor was calculated and a response index allocated. For each site, a global response index was calculated as the sum of the response index of each biomarker. Discriminant analysis shows significant differences between sites and seasons compared with control sample. Faroua (site 1) and Menzel Jemile (site 2) seem to be the less polluted with respect to the other sites for all seasons. Gargour (site 6) shows the highest Multimarker Pollution Index during the four seasons, indicating higher contamination level.  相似文献

A multimarker approach was developed, establishing a scale for the classification of the water quality in European coastal sites (BIOMAR European programme). This study allows the evaluation of the temporal trends of this scale when applied to selected sites of European Mediterranean coast (BEEP Biological Effects of Environmental Pollution in Marine Coastal Ecosystems: European programme). Acetylcholinesterase activity (AChE) is highly sensitive to organophosphorus and carbamate insecticides and, to some extent, also to heavy metals. Catalase activity (CAT) and lipid oxidation (evaluated as malonedialdehyde) are markers of oxidative stress, glutathione S-transferase (GST) activity is related to conjugation of organic compounds and benzo(a)pyrene hydroxylase activity (BPH) is a marker of effect of certain planar organic compounds (e.g. polycyclic aromatic hydrocarbons, PAHs). These parameters were measured either in gills (AChE, GST) or digestive gland (BPH, GST, CAT, MDA). For each biomarker, a discriminatory factor was calculated (maximum variation range/confidence interval) and a response index was allocated. For each site, a Multimarker Pollution Index (MPI) was calculated as the sum of the response index of each of the five more discriminating biomarkers. As the result of our calculation method, the quality of the coastal environment at each site can be classified according to a five levels scale. Samples collected for five cruises in May 2001, 2002, 2003, and September 2001 and 2002 showed MPI evolutions. The results show that water quality can be classified from class 1 (clean areas in some sites of France, Italy and Spain) to class 4 (high pollution in main harbours). Results of the use of the biomarker scale in WP3 (Work Package Concernant Biomonitoring Programmes in Mediterranean Sea) during the BEEP programme make a strong contribution to the establishment of standardized strategies and methods for internationally agreed protocols for biomarker-based monitoring programmes. In comparison with scale pollution methodology used in the BIOMAR programme, the main contribution of BEEP was (1) to select from discriminatory analysis the biomarkers to be included in calculation of scale pollution; (2) to improve the use of the biomarker index in order to identify the main contaminants by analysis of individual contributions to the MPI; and (3) to apply methodology for temporal trends at sampled sites.  相似文献

Some areas fall into class 4, e.g. Carteau, Cortiou, Barcelona, Warnemunde, Swinemunde, Ebro delta. The global Biomarker Index was positively correlated with the level of PAHs in mussels in Baltic transects. A number of other contaminants or stressors may be present in the marine environment and the Biomarker Index appeared to be relevant to classify coastal environmental pollution.  相似文献

Contamination of mangrove ecosystems, including those of the Red Sea area, has caused serious concern globally. Spatial distribution of heavy metals and their bioaccumulation in one of the common mangrove plants of Saudi Arabia, Avicennia marina L., was evaluated in 8 stations at the Rabigh lagoon to assess the ecological risks due to heavy metal contamination. Among all the heavy metals, Fe concentration was recorded highest (8939.38 ± 312.63 mg/kg) at station S4. Contamination factor (CF) values for all heavy metals determined in this study were recorded in ascending order as Cu < Cr < Mn < Zn < Fe < Ni < Pb < Cd, with the pollution load index pattern recorded in descending order as S6 > S4 > S3 > S5 > S7 > S1 > S8 > S2. Bio-concentration factor (BCF) was <1 for all the heavy metals and there was a positive correlation between the antioxidants and lead (Pb), which can be a result of the ability of A. marina to exclude or detoxify this metal by its mechanism of exclusion or detoxification. A significant correlation existed between the heavy metals concentration in sediment and A. marina leaves at one combination or the other, except for Cu and Cd, which do not correlate with any other metal concentration. The information provided in the present study can be used in the monitoring and measurement of heavy metal pollution in marine ecosystems or other aquatic environments, to prevent several ecological risks to the mangrove ecosystem.  相似文献

A genetic stock identification (GSI) study was undertaken in a fishery for Atlantic salmon Salmo salar to determine the effects of restrictive fishery management measures on the stock composition of the fishery, and if accurate and precise stock composition estimates could be achieved on the small geographical scale where this fishery operates, using a suite of only seven microsatellite loci. The stock composition of the Foyle fishery was shown to comprise almost exclusively of Foyle origin fish in the 3 years after restrictive measures were introduced in 2007, compared to 85% the year before. This showed that the restrictive measures resulted in the Foyle fishery being transformed from a mixed‐stock fishery to an almost exclusively single‐stock fishery, and showed how GSI studies can guide and evaluate management decisions to successfully manage these fisheries. Highly accurate and precise stock composition estimates were achieved in this study, using both cBAYES and ONCOR genetic software packages. This suggests accurate and precise stock composition is possible even on small geographical scales.  相似文献

For Pacific salmon, the key fisheries management goal in British Columbia (BC) is to maintain and restore healthy and diverse Pacific salmon populations, making conservation of salmon biodiversity the highest priority for resource management decision‐making. Salmon status assessments are often conducted on coded‐wire‐tagged subsets of indicator populations based on assumptions of little differentiation within or among proximal populations. In the current study of southern BC coho salmon (Oncorhynchus kisutch) populations, parentage‐based tagging (PBT) analysis provided novel information on migration and life‐history patterns to test the assumptions of biological homogeneity over limited (generally < 100 km) geographic distances and, potentially, to inform management of fisheries and hatchery broodstocks. Heterogeneity for location and timing of fishery captures, family productivity, and exploitation rate was observed over small geographic scales, within regions that are, or might be expected to be, within the area encompassed by a single‐tagged indicator population. These results provide little support for the suggestion that information gained from tagged indicator populations is representative of marine distribution, productivity, and exploitation patterns of proximal populations.  相似文献

生态用地功能分类是其生态价值评估及管理的基础,与生态系统服务的权衡和协同关系联系密切;对小尺度区域进行生态用地功能分类研究有助于实现精细化生态资源管理。以重庆两江新区为例,选取该区8种重要的生态系统服务,兼顾生态与社会成分,基于层次聚类法得到6类生态系统服务簇,依据提供重要生态系统服务和维护关键生态过程的原则,对其重要性等级进行了划分;依据生态系统服务簇与土地利用类型之间的关联关系,将生态用地定义为提供重要生态系统服务、维护关键生态过程或生态脆弱的用地空间,并按重要性将其划分为3个等级。研究结果表明,基于生态系统服务簇的生态用地分类框架不仅能够最大限度涵盖生态用地的功能属性,而且能够在空间上量化不同生态系统服务的相对比例,有助于对用地类型与生态功能之间的对应关系形成深入系统的认识,也可为精细化生态管理提供参考。  相似文献

Stepwise synthetic assembly of polypeptide chains reversibly linked to polyethylene glycol represents a hybrid between traditional solution and solid-phase chemistries and combines the inherent advantages of both approaches. The technical simplicity and scalability of the liquid-phase peptide synthesis method renders it particularly attractive for multiple parallel syntheses, combinatorial approaches and the large-scale preparation of peptides. The versatile protection strategy based on the N alpha-fluorenylmethoxycarbonyl group commonly used in solid-phase peptide synthesis was adapted to the liquid-phase approach. Fluoride ions were used rather than the conventional organic base piperidine for the repetitive amino-deprotection step. Using a range of acid- and base-labile linkers between the polymer and the peptide, it was shown that free and fully side-chain protected peptides can be obtained using our version of the liquid-phase peptide synthesis method. Protocols for simultaneous multiple syntheses requiring a minimum of equipment are presented and the use of polyethylene glycol-bound peptides in biochemical binding and functional assay systems is demonstrated.  相似文献

土壤中水溶性盐的分析,是发展研究盐渍土盐分动态监测与预报技术的重要基础工作。针对目前干旱半干旱区广泛存在的绿洲土壤次生盐渍化问题,基于GIS和地统计学方法,研究了渭干河—库车河流域三角洲绿洲盐渍化土壤特征(土壤含盐量)的空间变异性。研究结果表明:渭-库绿洲土壤含盐量的空间变异性为中等变异,且随土壤深度增加而减弱。各层土壤的合理采样数为在95%置信水平,20%误差下的合理采样数目。高斯模型能够较好的拟合10-30cm及30-50cm层土壤含盐量的空间结构。各层土壤盐分空间变异性受到结构性与随机性因素共同作用的影响,呈强空间相关性。套合结构模型考虑到0-10cm层土壤盐分空间变异的尺度依赖性,能够更好的拟合0-10cm层土壤含盐量的空间结构。Kriging插值以及空间等值线分布趋势图能够直观的表现研究区内土壤盐分含量的空间分布状况和变化情况。研究为土壤特征变量空间变异分析与评价提供了普适性较强的实现方法,为构建盐渍土定量动态监测模型以及盐渍土的改良利用提供一定的科学依据。  相似文献

Phylogenetic relationships among 28 of the 68 species of the paleotropical genus Gaertnera (Rubiaceae) and two related genera were inferred from nucleotide sequence variation in four nuclear DNA (nDNA) markers: the internal transcribed spacers of nuclear rDNA (ITS), the large and small copies of phosphoenolpyruvate carboxylase (PepC-large and PepC-small), and triose phosphate isomerase (Tpi). Phylogenetic analysis of the combined nDNA dataset suggested that Gaertnera is monophyletic, but genetic variation among species was insufficient to reconstruct well-supported relationships within the genus. This was counter to expectations based on the very distinct morphologies and widespread distribution of the genus (West Africa to Sulawesi). Molecular clock analyses suggested variable dates of origin for Gaertnera depending upon the calibration method used. The most plausible calibration implies that Gaertnera migrated to Africa during the early Tertiary, possibly via a boreotropical land bridge and suggests that Gaertnera started to radiate 5.21 +/- 0.14 million years ago. This implies that range expansion in the group has occurred via a number of long-distance dispersal events rather than vicariance. The molecular clock estimate in turn estimated an unusually rapid lineage diversification rate within the radiation of 0.717-0.832 species/million years, comparable to those estimated for radiations on oceanic islands. Although low interspecific competition levels may have contributed to the diversification of Gaertnera on Mauritius, the mechanisms driving the rapid radiation of the group in other parts of its range remain elusive.  相似文献

Phylogenetic relationships of the orchid genus Aerides (Epidendroideae, Vandeae, Aeridinae) from Southeast Asia were inferred from DNA sequences of one nuclear (nrITS) and two plastid (matK, trnL–trnLF) regions of 48 taxa (21 Aerides, 25 other Aeridinae, 2 outgroup). Analyses of the combined datasets with parsimony, maximum likelihood and Bayesian methods revealed that Aerides is monophyletic and consists of three well-supported subclades which are only partly in accordance with previous sectional delimitations based on floral characters. The two different flower types in Aerides (hidden versus open spur entrance) seem to have evolved at least twice in geographically distinct areas. The phylogeny presented here is yet another example in Orchidaceae where floral morphology cannot be relied on to reconstruct phylogenetic history but rather is the result of pollinator-driven selection. The Aerides subclades are characterized by three different length classes of the mutation-rich P8 region in the trnL intron. To our knowledge, this is the first time that the P8 region was studied in orchids. The matK gene has been assumed to be a pseudogene in orchids due to occasional occurrence of frameshift indels, low transition/transversion (ts:tv) ratios and low substitution rates at the 3rd codon position. However, matK does not appear to be a pseudogene in Aerides and a comparison with data from other angiosperms suggests that ts:tv ratios and low substitution rates have been overestimated as arguments for a pseudogene status of matK in orchids.  相似文献

Background

Quantitative trait locus (QTL) mapping is an efficient approach to discover the genetic architecture underlying complex quantitative traits. However, the low density of molecular markers in genetic maps has limited the efficiency and accuracy of QTL mapping. In this study, specific length amplified fragment sequencing (SLAF-seq), a new high-throughput strategy for large-scale SNP discovery and genotyping based on next generation sequencing (NGS), was employed to construct a high-density soybean genetic map using recombinant inbred lines (RILs, Luheidou2 × Nanhuizao, F5:8). With this map, the consistent QTLs for isoflavone content across various environments were identified.

Results

In total, 23 Gb of data containing 87,604,858 pair-end reads were obtained. The average coverage for each SLAF marker was 11.20-fold for the female parent, 12.51-fold for the male parent, and an average of 3.98-fold for individual RILs. Among the 116,216 high-quality SLAFs obtained, 9,948 were polymorphic. The final map consisted of 5,785 SLAFs on 20 linkage groups (LGs) and spanned 2,255.18 cM in genome size with an average distance of 0.43 cM between adjacent markers. Comparative genomic analysis revealed a relatively high collinearity of 20 LGs with the soybean reference genome. Based on this map, 41 QTLs were identified that contributed to the isoflavone content. The high efficiency and accuracy of this map were evidenced by the discovery of genes encoding isoflavone biosynthetic enzymes within these loci. Moreover, 11 of these 41 QTLs (including six novel loci) were associated with isoflavone content across multiple environments. One of them, qIF20-2, contributed to a majority of isoflavone components across various environments and explained a high amount of phenotypic variance (8.7% - 35.3%). This represents a novel major QTL underlying isoflavone content across various environments in soybean.

Conclusions

Herein, we reported a high-density genetic map for soybean. This map exhibited high resolution and accuracy. It will facilitate the identification of genes and QTLs underlying essential agronomic traits in soybean. The novel major QTL for isoflavone content is useful not only for further study on the genetic basis of isoflavone accumulation, but also for marker-assisted selection (MAS) in soybean breeding in the future.

资源型城市作为我国重要战略保障基地,面临较为突出的不可持续问题,定量化开展可持续发展水平监测评估是推动其绿色转型发展、激活再生动力的重要途径。基于联合国可持续发展目标体系(SDGs),结合行星边界理论,从资源型城市可持续发展特征出发,建立了由2个系统、4个支柱、12个领域和48项指标组成的资源型城市可持续发展评价指标体系。借鉴全球SDGs监测评估实践经验,考虑指标属性,综合确定各指标的阈值,并采用改进的离差标准化、障碍度分析及耦合协调度分析等方法建立综合评价技术体系。以拥有“有色金属之乡”称号的郴州市国家可持续发展议程创新示范区为评价对象,对其2005—2019年连续15年的可持续发展水平进行实证分析。结果表明:(1)郴州市可持续发展水平整体上呈现稳步上升趋势,得分由0.398上升到0.610。(2)4个支柱中,自然资源支柱的得分进步幅度最大,高达71.3%,说明郴州市已经逐步提高对资源利用效率的重视,同时意识到挖掘替代资源的重要性,资源保障与利用能力不断提升;而发展动力支柱得分始终处于较弱或中等水平,已成为制约郴州市可持续发展和绿色转型的主要因素,其中科技创新驱动是该支柱发展关键障碍...  相似文献

With the aime of developing a sensitive mutagen screening system, teh responses of 15 different chemical mutagens at 5 independent genetic loci in Chinese hamster ovary (CHO) cells have been determined. The genetic markers which have been employed include resistance to thioguanine (Thgr), ouabain (OuaR), the protein syntheis inhibitor emetime (Emtr, the plyamine synthesis inhibitor methylglyoxal bisguanylhydrazone (Mbgr) and the nucleoside analog 5,6-dichlororibofuranosyl benzimidazole (DrbR). The optimal selection conditions for all of these genetic markers in CHO cells have been described. The chemicals whose response was investigated in these studies include direct-acting alkylating agents (ethyl methanesulfonate, methyl methanesulfonate, β-propiolactone, ethyleneimine,N-nitrosomethylurea and 4-nitroquinolineN-oxide), DNA intercalating and cross-linking agents (ICR-170, acridine orange, ethidium bormide, mitomycin C and actinomycine D), polycyclic hydrocarbons (benzo[a]pyrene (B(a)P) and 7,12-dimethylbenz[a]anthracene (DMBA)) and aromatic amines (benzidine and β-naphthylamine). Simultaneous examination of the response of the set of genetic markers to these chemicals revealed that although all of these chemicals caused a dose-dependent increase in the frequency of mutations at many of the above genetic loci, the magnitude of the mutagenic response at different genetic loci varied greatly depending upon the chemical. Of the genetic loci examined, no one single locus showed higher response to all of the above chemicals, instead, depending upon the chemical, specific loci were found to be more responsive than other. The polycyclic hydrocarbons and aromatic amines were weakly mutagenic in this system at several genetic loci even without any exogenous microsomal activation, although in the presence of a rat liver S9 fraction similar toxic and mutagenic effects of B(a)P and DMBA were observed at 5–20-fold lower concentrations. These results indicate that CHO cells may possess significant capacity for the metabolic activation of many procarnicogens, and also underscore the merits of measuring the mutagenic response at multiple genetic loci in mutagen screening studies.  相似文献

Juvenile and adult scale characteristics were used to compare two juvenile groups of Atlantic salmon in a large subarctic river in northern Scandinavia: individuals that have migrated from the main stem into small tributaries and those which remain in the main stem. Body size and scale measurements indicated enhanced growth in migratory parr as compared to their resident main stem counterparts. Analysis of adult salmon scale characteristics using maximum likelihood estimators revealed that 20% of the adults had been in the tributaries before the end of their second year of life, and more than 30% more had moved into the tributaries in the third year. Tributary fish matured at a smaller size and younger age (one-sea-winter salmon) than those rearing in the main stem which included a higher proportion of multi-sea-winter salmon. In addition, when smolt ages and ages at maturity were compared, older female smolts often resulted in smaller spawners and younger smolts, larger spawners. Small female spawners were more likely to survive to become repeat spawners.  相似文献

Many northern lake‐rich regions are undergoing pronounced hydrological change, yet inadequate knowledge of the drivers of these landscape‐scale responses hampers our ability to predict future conditions. We address this challenge in the thermokarst landscape of Old Crow Flats (OCF) using a combination of remote sensing imagery and monitoring of stable isotope compositions of lake waters over three thaw seasons (2007–2009). Quantitative analysis confirmed that the hydrological behavior of lakes is strongly influenced by catchment vegetation and physiography. Catchments of snowmelt‐dominated lakes, typically located in southern peripheral areas of OCF, encompass high proportions of woodland/forest and tall shrub vegetation (mean percent land cover = ca. 60%). These land cover types effectively capture snow and generate abundant snowmelt runoff that offsets lake water evaporation. Rainfall‐dominated lakes that are not strongly influenced by evaporation are typically located in eastern and northern OCF where their catchments have higher proportions of dwarf shrub/herbaceous and sparse vegetation (ca. 45%), as well as surface water (ca. 20%). Evaporation‐dominated lakes, are located in the OCF interior where their catchments are distinguished by substantially higher lake area to catchment area ratios (LA/CA = ca. 29%) compared to low evaporation‐influenced rainfall‐dominated (ca. 10%) and snowmelt‐dominated (ca. 4%) lakes. Lakes whose catchments contain >75% combined dwarf shrub/herbaceous vegetation and surface water are most susceptible to evaporative lake‐level drawdown, especially following periods of low precipitation. Findings indicate that multiple hydrological trajectories are probable in response to climate‐driven changes in precipitation amount and seasonality, vegetation composition, and thermokarst processes. These will likely include a shift to greater snowmelt influence in catchments experiencing expansion of tall shrubs, greater influence from evaporation in catchments having higher proportions of surface water, and an increase in the rate of thermokarst lake expansion and probability of drainage. Local observations suggest that some of these changes are already underway.  相似文献
