A sand culture experiment was conducted to study ameliorative role of 28-homobrassinolide (HBL) in Brassica juncea seedlings raised from the seeds treated with water, or 10−10, 10−8 and 10−6 M 28-homobrassinolide (HBL) and grown in the presence of copper (50, 100 and 150 mg kg−1 sand) and sampled at 30 days after sowing. The plants grown in the presence of copper exhibited a significant decline in growth, chlorophyll and photosynthetic parameters. However, the activity of antioxidant enzymes: catalase (E.C. 1.11.1.6), peroxidase (E.C. 1.11.1.7) and superoxide dismutase (E.C. 1.15.1.1) and the content of proline increased in the plants grown under copper stress and/or raised from treatment with HBL. However, H2O2 content increased significantly in copper-treated plants and decreased in plants given HBL treatment. Treatment of seeds with HBL improved the growth, photosynthetic parameters and antioxidant enzymes and also improved in the plants grown under copper stress. The elevated antioxidant enzyme and proline might be responsible to overcome the toxic effects of copper in B. juncea.  相似文献

In the view of physiological role of H2O2, we investigated whether exogenous H2O2 application would affect short-term cold response of tomato and induce acclimation. Pretreatments were performed by immersing roots into 1 mM H2O2 solution for 1 h when transferring seedlings from seedling substrate to soil (acclimated group). Cold stress (3 °C for 16 h) caused significant reduction in relative water content (RWC) of control and non-acclimated (distilled water treated) groups when compared with unstressed plants. H2O2 promoted maintenance of relatively higher RWC under stress. Anthocyanin level in leaves of acclimated plants under cold stress was significantly higher than that of unstressed control and non-acclimated plants. Malondialdehyde (MDA) levels demonstrated low temperature induced oxidative damage to control and non-acclimated plants. MDA remained around unstressed conditions in acclimated plants, which demonstrate that H2O2 acclimation protected tissues against cold induced lipid peroxidation. H2O2 acclimation caused proline accumulation in roots under cold stress. Ascorbate peroxidase (APX) activity in roots of cold stressed and unstressed H2O2 acclimated plants increased when compared with control and non-acclimated plants, with highest increase in roots of acclimated plants under cold stress. CAT levels in roots of acclimated plants also increased, whereas levels remained unchanged in unstressed plants. Endogenous H2O2 levels significantly increased in roots of control and non-acclimated plants under cold stress. On the other hand, H2O2 content in roots of acclimated plants was significantly lower than control and non-acclimated plants under cold stress. The results presented here demonstrated that H2O2 significantly enhanced oxidative stress response by elevating the antioxidant status of tomato.  相似文献

Heavy metal contaminated agricultural soils are one of the most important constraints for successful cultivation of crops. The current research was conducted to evaluate the role of potassium (K) on plant growth and amelioration of cadmium (Cd) stress in Gladiolus grandiflora under greenhouse conditions. G. grandiflora corms were sown in media contaminated with 0 (C), 50 (Cd50) and 100 (Cd100) mg Cd kg?1 soil. The plants growing in Cd-contaminated media exhibited reduced gas exchange attributes, chlorophyll (Chl) contents, vegetative and reproductive growth as compared to control. The plants raised in Cd contaminated media showed reduced nutrition yet higher Cd contents. However, supplementation of 60 mg Kg?1 K in treated plants (C+K, Cd50+K and Cd100+K) improved quantity of total soluble protein and proline (Pro) along with activity of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT), peroxidase (POD) and ascorbate peroxidase (APX) under Cd stress. Similarly, K supplementation reduced the level of malondialdehyde (MDA) and hydrogen peroxide (H2O2) in treated plants. Potassium supplemented plants exhibited better vegetative and reproductive growth. The improved stress tolerance in K supplemented plants was attributed to the reduced quantity of MDA and H2O2, enhanced synthesis of protein, proline, phenols, flavonides and improved activity of antioxidant enzymes. The present research supports the application of K for alleviation of Cd stress in G. grandiflora.  相似文献

The alleviative effects of exogenous calcium on copper phytotoxicity were investigated in Elodea canadensis plants. There was a significant accumulation of Cu in the plants after their exposure to 0.01 mM Cu accompanied by many symptoms of toxicity. Increased uptake of Cu severely reduced content of photosynthetic pigments, soluble proteins, and free proline. The total antioxidant capacity (T-AOC), reduced glutathione (GSH), and non-protein thiol (NP-SH) were severely suppressed in Cu-stressed plants resulting in a rapid increase in content of superoxide anion (O2 ·?), hydrogen peroxide, lipid peroxidation, and cell death. Simultaneous application of Ca markedly increased the content of photosynthetic pigments, soluble proteins, free proline, T-AOC, GSH, and NP-SH, and reduced oxidative damage as indicated by lowered content of MDA, O2 ·?, and H2O2; and decreased cell death. Furthermore, application of Ca reduced Cu uptake and effectively reversed the Cu-induced nutrient imbalance.  相似文献

We investigated individual and combined effects of salinity, soil boron (B), silicon (Si) and salicylic acid (SA) on the activities of major antioxidant enzymes (superoxide dismutase, SOD; catalase, CAT and ascorbate peroxidase, APX) and non-enzymatic antioxidants (AA), proline, chlorophyll, anthocyanin, H2O2 concentration, stomatal resistance (SR), lipid peroxidation (MDA), membrane permeability (MP), and the uptake of sodium (Na), chloride (Cl), boron and Si of spinach plants. In general, salinity significantly increased H2O2 and proline concentrations, antioxidant activity, membrane permeability, lipid peroxidation and SR of the spinach plants, indicating that they were stressed, whereas application of B only increased proline concentration. However, plant fresh weights did not decline with either treatment. The application of Si decreased H2O2 and increased the activity of SOD and CAT. The application of SA increased SOD activity. Neither SA nor Si had any effect on the proline concentration, or MP. However, application of Si increased chlorophyll concentration and decreased lipid peroxidation (MDA concentration). Si treatment had no effect on SR. The concentration of B in the tissues, which was strongly increased by B treatment, was decreased by NaCl. As a result of salinity, concentrations of Na+ and Cl ions were increased in the plant tissues, and application of Si slightly increased these concentrations. These results indicate that exogenous Si application increases stress tolerance of spinach, a plant that is naturally reasonably resistant to combined salinity and B toxicity, by the enhancement of antioxidant mechanisms that reduce membrane damage. Exogenous SA has a less obvious effect, although the levels of salinity and boron stress applied were not sufficient in this experiment to reduce plant fresh weight.  相似文献

The current research was performed to evaluate the effect of Bacillus megaterium MCR-8 on mitigation of nickel (Ni) stress in Vinca rosea grown on Ni-contaminated soil (50, 100, and 200 mg Ni kg?1 soil). The treated plants exhibited reduced growth, biomass, gas exchange capacity, and chlorophyll (Chl) content under Ni stress. The inoculated plants growing in Ni-contaminated media exhibited relatively higher growth, total soluble protein, and proline contents. Similarly, bacterial inoculation improved the activity of antioxidant enzymes including superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX) under Ni stress. The Ni stress alleviation in inoculated plants was attributed to the reduced level of malondialdehyde (MDA) and hydrogen peroxide (H2O2), enhanced synthesis of protein, proline, phenols, and flavonides in conjunction with improved activity of antioxidant enzymes. The growth-promoting characteristics of microbe such as 1-aminocyclopropane-1-carboxylate deaminase (ACCD) and phosphate solubilization activity, siderophore, and auxin production capability also improved the growth and stress mitigation in inoculated plants. Furthermore, the inoculated plants exhibited higher value for bioconcentration factor (BCF), translocation factor (TF), and resulted in higher loss of Ni content from soil. The current results exhibited the beneficial role of B. megaterium MCR-8 regarding stress alleviation and Ni phytoextraction by V. rosea.  相似文献

Salinity and waterlogging are two stresses which in nature often occur simultaneously. In this work, effects of combined waterlogging and salinity stresses are studied on the anatomical alteration, changes of enzymatic antioxidant system and lipid peroxidation in Mentha aquatica L. plants. Seedlings were cultured in half-strength Hoagland medium 50 days after sowing, and were treated under combination of three waterlogging levels (well drained, moderately drained and waterlogging) and NaCl (0, 50, 100, 150 mM) for 30 days. Moderately drained and waterlogging conditions induced differently aerenchyma formation in roots of M. aquatica salt-treated and untreated plants. Moreover, stele diameter and endodermis layer were also affected by salt stress and waterlogging. Salt stress significantly decreased growth, relative water content (RWC), protein level, catalase (CAT) and polyphenol oxidase (PPO) activities, and increased proline content, MDA content, H2O2 level and activities of superoxide dismutase (SOD), peroxidase (POX), and ascorbate peroxidase (APX). Waterlogging in salt-untreated plants increased significantly growth parameters, RWC, protein content, antioxidant enzyme activity, and decreased proline content, H2O2 and MDA levels. In salt-treated plant, waterlogging caused strong induction of antioxidant enzymes activities especially at severe stress condition. These results suggest M. aquatica is a waterlogging tolerant plant due to significant increase of antioxidant activity, membrane stability and growth under water stress. High antioxidant capacity under waterlogging can be a protective strategy against oxidative damage, and help to salt stress alleviation.  相似文献

The role of 28-homobrassinolide (HBL) in countering nickel-induced oxidative damage through overexpression of antioxidant enzymes and proline in Vigna radiata has been investigated. Two varieties of V. radiata, one sensitive to Ni (PDM-139) and the other tolerant to Ni (T-44), were sown in the soil fed with different levels (0, 50, 100 or 150 mg kg−1) of Ni, and at 29-day stage, foliage of plants was applied with deionized water (control), 10−8 or 10−6 M of HBL. The plants were sampled at 45-day stage of growth to assess various physiological as well as biochemical characteristics. The remaining plants were allowed to grow up to maturity to study the yield characteristics. The growth traits, leghemoglobin, nitrogen and carbohydrate content in the nodules, leaf chlorophyll content, photosynthesis efficiency, leaf water potential, activities of nitrate reductase, carbonic anhydrase and nitrogenase decreased proportionately with the increasing concentrations of nickel, whereas electrolyte leakage, various antioxidant enzymes viz. catalase, peroxidase and superoxide dismutase and accumulation of proline increased at 45-day stage. However, the exogenously applied HBL to the nickel-stressed or non-stressed plants improved growth, nodulation and photosynthesis and further enhanced the various antioxidant enzymes viz. catalase, peroxidase and superoxide dismutase and accumulation of proline. The deleterious impact of Ni on the plants was concentration dependent where HBL applied to the foliage induced overexpression of antioxidant enzyme and accumulation of proline (osmolyte) which could have conferred tolerance to Ni up to 100 mg kg−1, resulting in improved growth, nodulation, photosynthesis and yield attributes.  相似文献

The effects of thiamin (Thi) applied as seed soaking or foliar spray on some key physiological parameters were investigated in two differentially salt-responsive maize (Zea mays L.) cultivars, DK 5783 and Apex 836 F1, exposed to saline stress in two different experiments. An initial experiment (germination experiment) was designed to identify appropriate doses of Thi which could lessen the deleterious effects of salt on plants and screen all available maize cultivars for their differential tolerance to salt stress (100 mM NaCl). The seeds of nine maize cultivars were soaked for 24 h in solutions containing six levels of Thi (25, 50, 75, 100, 125 and 150 mg l?1). Based on the results obtained from the germination experiment, maize cultivar DK 5783 was found to be the most salt tolerant and Apex 836 as the most sensitive cultivar. Also, of six Thi levels used, two levels (100 and 125 mg l?1) were chosen for subsequent studies. In the second experiment (glasshouse experiment), two maize cultivars, DK 5783 (salt tolerant) and Apex 836 (salt sensitive) were subjected to saline regime (100 mM NaCl) and two levels of Thi (100 and 125 mg l?1) applied as foliar spray. Salt stress markedly suppressed shoot and root dry mass, total chlorophylls (“a” + “b”), leaf water potential and maximum fluorescence yield (Fv/Fm) in the plants of both maize cultivars, but it increased proline accumulation, leaf osmotic pressure, malondialdehyde (MDA) and hydrogen peroxide (H2O2) concentrations, electrolyte leakage (EL) as well as activities of some key antioxidant enzymes, superoxide dismutase (SOD; EC. 1.15.1.1), peroxidase (POD; EC. 1.11.1.7) and catalase (CAT; EC. 1.11.1.6). Salt-induced reduction in plant growth parameters was higher in the salt-sensitive cultivar, Apex 836, which was found to be associated with relatively increased EL, and MDA and H2O2 levels, and decreased activities of the key antioxidant enzymes. Application of Thi as seed soaking or foliar spray partly mitigated the deleterious effects of salinity on plants of both maize cultivars. The most promising effect of Thi on alleviation of adverse effects of salt stress on maize plants was found when it was applied as foliar spray at 100 mg l?1. Thiamin application considerably reduced tissue Na+ concentration, but improved those of N, P, Ca2+ and K+ in the salt-stressed maize plants. Exogenously applied thiamin-induced growth improvement in maize plants was found to be associated with reduced membrane permeability, MDA and H2O2 levels, and altered activities of some key antioxidant enzymes such as CAT, SOD and POD as well as increased photosynthetic pigment concentration under saline regime.  相似文献

The effects of Cd, in combination with salicylic acid (SA) and sodium nitroprusside (SNP), on ryegrass seedlings were studied. Exposure of plants to 0.1 mM CdCl2 for 2 weeks resulted in toxicity symptoms such as chlorosis and necrotic spots on leaves. The addition of 0.2 mM SA or 0.1 mM SNP slightly alleviated the toxic effects of Cd. After application of both SA and SNP, these symptoms significantly decreased. Treatment with Cd resulted in a decrease of dry weight of roots and shoots, chlorophyll content, net photosynthetic rate (P n), transpiration rate (T r), and the uptake and translocation of mineral elements. In Cd-treated plants, levels of lipoxygenase activity and malondialdehyde, hydrogen peroxide (H2O2), and proline contents significantly increased, whereas the activities of antioxidant enzymes, such as superoxide dismutase, guaiacol peroxidase, catalase, and ascorbate peroxidase, decreased in both roots and shoots. The results indicated that Cd caused physiological stresses in ryegrass plants. The Cd-stressed plants exposed to SA or SNP, especially to SA + SNP, exhibited improved growth compared with Cd-stressed plants. Application of SA or SNP, especially the combination SA + SNP, considerably reduced root-to-shoot translocation of Cd and increased the activities of antioxidant enzymes in both roots and shoots of Cd-stressed plants. The interaction of SA and SNP increased chlorophyll content, P n and T r in leaves, and the uptake and translocation of mineral elements, and decreased lipid peroxidation and H2O2 and proline accumulation in roots and shoots. These results suggest that SA or SNP, and, in particular, their combination counteracted the negative effects of Cd on ryegrass plants.  相似文献

The effect of exogenously applied hydrogen peroxide on salt stress tolerance was investigated in Panax ginseng. Pretreatment of ginseng seedlings with 100 μM H2O2 increased the physiological salt tolerance of the ginseng plant and was used as the optimum concentration to induce salt tolerance capacity. Treatment with exogenous H2O2 for 2 days significantly enhanced salt stress tolerance in ginseng seedlings by increasing the activities of ascorbate peroxidase, catalase and guaiacol peroxidase and by decreasing the concentrations of malondialdehyde (MDA) and endogenous H2O2 as well as the production rate of superoxide radical (O2 ?). There was a positive physiological effect on the growth and development of salt-stressed seedlings by exogenous H2O2 as measured by ginseng dry weight and both chlorophyll and carotenoid contents. Exogenous H2O2 induced changes in MDA, O2 ?, antioxidant enzymes and antioxidant compounds, which are responsible for increases in salt stress tolerance. Salt treatment caused drastic declines in ginseng growth and antioxidants levels; whereas, acclimation treatment with H2O2 allowed the ginseng seedlings to recover from salt stress by up-regulation of defense-related proteins such as antioxidant enzymes and antioxidant compounds.  相似文献

Cucumber seedlings were pretreated with 3 μM 5-aminolevulinic acid (ALA) followed by cultivation at normal (25/18 °C) or high (42/38 °C) day/night temperature to investigate the protective effects of ALA on heat stress in plants. Heat elevated the contents of malondiadehyde (MDA), superoxide radical (O2 .−) and hydrogen peroxide (H2O2) in leaves of all plants but less in ALA-pretreated plants. Heat treatment resulted in higher antioxidant enzyme activities and proline and soluble sugar contents and weaker growth inhibition in ALA-pretreated plants than in those treated with heat alone. These results indicate that ALA pretreatment increased the tolerance of seedlings to heat stress.  相似文献

Nitric oxide (NO) is a stress factor or a signal molecule involved in various plant physiological and developmental processes. In the present study, the generation of reactive oxygen species and the metabolism of proline due to different sodium nitroprusside (SNP, an NO donor) concentrations were investigated in callus from halophyte Nitraria tangutorum Bobr. Treatment with SNP led to significant increases of hydrogen peroxide (H2O2) content and cell viability but notable reductions in hydrogen radical level and lipid peroxidation degree, and superoxide onion (O2 ?) content also enhanced in 100 μM SNP-treated calli. Using a chemical inhibitor for plasma membrane (PM) NADPH oxidase diphenylene iodonium (DPI), we found low O2 ? generation in untreated and 25 μM SNP-treated calli, whereas in those treated with 100 μM SNP O2 ? level exhibited a very little alteration, comparable to the absence of DPI. These suggest a high activity of PM NADPH oxidase in untreated calli. H2O2 scavenging enzymes (catalase, peroxidase [POD] and ascorbate peroxidase) and H2O2 forming enzymes (superoxide dismutase [SOD], cell wall-POD and diamine oxidase [DAO]) stimulated significantly in calli treated with different SNP concentrations while glutathione reductase activity decreased. In addition, a reduction in proline content was observed in SNP-treated calli. Moreover, different SNP concentrations stimulated proline dehydrogenase (PDH) and ornithine δ-aminotransferase but inhibited r-glutamyl kinase (GK). In conclusion, our results suggest that the increasing H2O2 generation was associated with the stimulation of SOD, cell wall-POD and DAO, and that the reduction of proline content might be the consequence of increased PDH activity and decreased GK activity in N. tangutorum Bobr. calli under SNP treatment.  相似文献

The paper mainly studied the effects of ultraviolet‐B (UV‐B) radiation, nitrogen, and their combination on photosynthesis and antioxidant defenses of Picea asperata seedlings. The experimental design included two levels of UV‐B treatments (ambient UV‐B, 11.02 KJ m−2 day−1; enhanced UV‐B, 14.33 KJ m−2 day−1) and two nitrogen levels (0; 20 g m−2 a−1 N) – to determine whether the adverse effects of UV‐B are eased by supplemental nitrogen. Enhanced UV‐B significantly inhibited plant growth, net photosynthetic rate (A), stomatal conductance to water vapor (Gs), transpiration rate and photosynthetic pigment, and increased intercellular CO2 concentration, UV‐B absorbing compounds, proline content, malondialdehyde (MDA) content, and activity of antioxidant enzymes (peroxidase (POD), superoxide dimutase, and glutathione reductase). Enhanced UV‐B also reduced needle DW and increased hydrogen peroxide (H2O2) content and the rate of superoxide radical (O2) production only under supplemental nitrogen. On the other hand, supplemental nitrogen increased plant growth, A, Gs, chlorophyll content and activity of antioxidant enzymes (POD, ascorbate peroxidase, and catalase), and reduced MDA content, H2O2 content, and the rate of O2 production only under ambient UV‐B, whereas supplemental nitrogen reduced activity of antioxidant enzymes under enhanced UV‐B. Carotenoids content, proline content, and UV‐B absorbing compounds increased under supplemental nitrogen. Moreover, significant UV‐B × nitrogen interaction was found on plant height, basal diameter, A, chlorophyll a, activity of antioxidant enzymes, H2O2, MDA, and proline content. These results implied that supplemental nitrogen was favorable for photosynthesis and antioxidant defenses of P.asperata seedlings under ambient UV‐B. However, supplemental nitrogen made the plants more sensitive to enhanced UV‐B, although some antioxidant indexes increased.  相似文献

Nitric oxide (NO) and glutathione (GSH) regulate a variety of physiological processes and stress responses; however, their involvement in mitigating Cu toxicity in plants has not been extensively studied. This study investigated the interactive effect of exogenous sodium nitroprusside (SNP) and GSH on Cu homeostasis and Cu-induced oxidative damage in rice seedlings. Hydroponically grown 12-day-old seedlings were subjected to 100 μM CuSO4 alone and in combination with 200 μM SNP (an NO donor) and 200 μM GSH. Cu exposure for 48 h resulted in toxicity symptoms such as stunted growth, chlorosis, and rolling in leaves. Cu toxicity was also manifested by a sharp increase in lipoxygenase (LOX) activity, lipid peroxidation (MDA), hydrogen peroxide (H2O2), proline (Pro) content, and rapid reductions in biomass, chlorophyll (Chl), and relative water content (RWC). Cu-caused oxidative stress was evident by overaccumulation of reactive oxygen species (ROS; superoxide (O2 ?–) and H2O2). Ascorbate (AsA) content decreased while GSH and phytochelatin (PC) content increased significantly in Cu-stressed seedlings. Exogenous SNP, GSH, or SNP?+?GSH decreased toxicity symptoms and diminished a Cu-induced increase in LOX activity, O2 ?–, H2O2, MDA, and Pro content. They also counteracted a Cu-induced increase in superoxide dismutase (SOD), ascorbate peroxidase (APX), glutathione reductase (GR), monodehydroascorbate reductase (MDHAR), and glyoxalase I and glyoxalase II activities, which paralleled changes in ROS and MDA levels. These seedlings also showed a significant increase in catalase (CAT), glutathione peroxidase (GPX), dehydroascorbate reductase (DHAR), glutathione S-transferase (GST) activities, and AsA and PC content compared with the seedlings stressed with Cu alone. Cu analysis revealed that SNP and GSH restricted the accumulation of Cu in the roots and leaves of Cu-stressed seedlings. Our results suggest that Cu exposure provoked an oxidative burden while reduced Cu uptake and modulating the antioxidant defense and glyoxalase systems by adding SNP and GSH play an important role in alleviating Cu toxicity. Furthermore, the protective action of GSH and SNP?+?GSH was more efficient than SNP alone.  相似文献

Nitric oxide (NO) is a plant signaling compound known to mitigate key physiological processes and salicylic acid (SA) is considered to be a signaling molecule that plays a key role in growth, development, and defense responses in plants under stress conditions. This work investigated the effects of sodium nitroprusside (SNP, a donor of NO) and SA on salt-tolerance of cotton (Gossypium hirsutum L.) seedlings by examining growth, photosynthetic performance, total osmoregulation substance content, antioxidative enzymes and H+-ATPase enzyme subjected to 100 mM NaCl. Addition of 100 mM NaCl inhibited the growth and photosynthetic parameters of cotton seedlings, and dramatically increased the electrolyte leakage, the plant contents of proline, lipid peroxidation (malondialdehyde), hydrogen peroxide (H2O2) and Na. Furthermore, antioxidant enzyme activities were restrained. Foliar applications of 0.1 mM SNP or/and 0.1 mM SA led to increase in the growth rate and photosynthesis, including photosystem II, net photosynthetic rate and transpiration rate, improvement of reactive oxygen species-scavenging enzymes activities and reduction of H2O2 accumulation in cotton seedlings induced by NaCl. In addition, membrane transport and function were facilitated by decreasing leaf electrolyte leakage, improving ion absorption and activating the osmotic-regulated substances metabolic. Further investigation also showed that SNP and SA alleviated the inhibition of H+-ATPase in plasma membrane induced by NaCl. The present study showed that foliar application of SNP and SA alone mitigated the adverse effect of salinity, while the combined application proved to be even more effective in alleviating the adverse effects of NaCl stress.  相似文献

The development of nanotechnologies has increased the amount of manufactured metal oxide nanoparticles in the environment. In the view of nanoparticle dispersion to the environment, assessment of their toxicity becomes very crucial. Aluminum oxide (Al2O3) nanoparticles have wide range of use in industry as well as personal care products. The aim of this study was to evaluate the dose dependent effects of 13-nm-sized Al2O3 nanoparticles on wheat correlating with the appearance of enzymatic and non-enzymatic antioxidant defense response. Wheat roots were exposed to different concentrations of Al2O3 nanoparticles (5, 25 and 50 mg mL?1) for 96 h. The effects of Al2O3 nanoparticles were studied using different parameters such as H2O2 content, superoxide dismutase and catalase activity, lipid peroxidation, total proline, photosynthetic pigment and anthocyanin content. The results indicated that while Al2O3 nanoparticles caused a dose dependent increase in H2O2 content, superoxide dismutase activity, lipid peroxidation and proline contents, the catalase activity was decreased in compare the control. Moreover, total chlorophyll, chlorophyll a, carotenoids and anthocyanin contents reduced in the highest concentration 50 mg mL?1. In conclusion, Al2O3 nanoparticles caused oxidative stress in wheat after 96 h.  相似文献

It has been widely suggested that selenium (Se) deficiency play an important role in the pathophysiology of epilepsy. It has been reported that Se provides protection against the neuronal damage in patients and animals with epilepsy by restoring the antioxidant defense mechanism. The neuroprotective effects of topiramate (TPM) have been reported in several studies but the putative mechanism of action remains elusive. We investigated effects of Se and TPM in neuronal PC12 cell by evaluating Ca2+ mobilization, lipid peroxidation and antioxidant levels. PC12 cells were divided into eight groups namely control, TPM, Se, H2O2, TPM + H2O2, Se + H2O2, Se + TPM and Se + TPM + H2O2. The toxic doses and times of H2O2, TPM and Se were determined by cell viability assay which is used to evaluate cell viability. Cells were incubated with 0.01 mM TPM for 5 h and 500 nM Se for 10 h. Then, the cells were exposed to 0.1 mM H2O2 for 10 h before analysis. The cells in all groups except control, TPM and Se were exposed to H2O2 for 15 min before analysis. Cytosolic Ca2+ release and lipid peroxidation levels were higher in H2O2 group than in control, Se and TPM combination groups although their levels were decreased by incubation of Se and TPM combination. However, there is no difference on Ca2+ release in TPM group. Glutathione peroxidase activity, reduced glutathione and vitamin C levels in the cells were lower in H2O2 group than in control, Se and TPM groups although their values were higher in the cells incubated with Se and TPM groups than in H2O2 groups. In conclusion, these results indicate that Se induced protective effects on oxidative stress in PC12 cells by modulating cytosolic Ca2+ influx and antioxidant levels. TPM modulated also lipid peroxidation and glutathione and vitamin C concentrations in the cell system.  相似文献

Drought is the major constraint in arid regions throughout the world and identification of drought-resistant plants is therefore of crucial importance. Since young seedling stage is especially sensitive to water stress, the present work analyzed the physiological behavior of seedling from Acacia arabica issued from a dry area, grown under controlled environmental conditions and subjected to progressive soil drying. Although soil gravimetric water content (g H2O g?1 soil dry weight) dropped from 80 % to less than 35 %, most plants remained alive until the end of the water stress. Seedlings were able to efficiently close their stomata to reduce water losses and accumulated high amounts of proline. Despite osmotic adjustment, turgor pressure decreased in stressed plants and could explain the stress-induced inhibition of plant growth. Decrease in net photosynthesis was related to stress-induced decrease in stomatal conductance and not to any impact on chlorophyll concentration or fluorescence-related parameter: both PSII efficiency and photochemical quenching remained unaffected by water stress while drought-induced increase in non-photochemical quenching should be regarded as a strategy to avoid over-energisation of the photosynthetic apparatus. Instantaneous water use efficiency increased in stressed plants comparative to controls. Oxidative stress estimated by malondialdehyde concentration was recorded only at the end of the treatment, suggesting that stressed plants remained able to cope with reactive oxygen species. Water stress induced an increase in anthocyanins, while aglycone flavonols decreased. Those compounds were not involved in the management of oxidative stress. It is concluded that A. arabica is a promising drought-resistant plant species for rehabilitation of dry areas.  相似文献

Abstract

The effects of exogenous silicon (Si) on leaf relative water content (RWC), and the growth, Si concentrations, lipid peroxidation (MDA), lipoxygenase (LOX) activity, proline and H2O2 accumulation, non-enzymatic antioxidant activity (AA) and the activity of some antioxidant enzymes (superoxide dismutase, SOD; catalase, CAT; ascorbate peroxidase, APX) in shoots of ten chickpea cultivars grown under drought were investigated. Drought stress decreased the growth of all the cultivars while applied Si improved the growth at least five of the 10 chickpea cultivars. Silicon applied to the soil at 100 mg kg?1 significantly increased Si concentrations of the cultivars and counteracted the deleterious effects of drought in 5 of the ten chickpea cultivars by increasing their RWC. In most cultivars tested H2O2, proline and MDA content and LOX activity were increased by drought whereas application of Si decreased their levels. APX activity was increased by drought but it was depressed by Si. In general, SOD and CAT activities of the cultivars were decreased by drought. Depending on cultivars, the CAT activity was decreased, and increased or unchanged in response to applied Si, while the SOD activity of the cultivars increased or unchanged by Si. The non-enzymatic antioxidant activity of the cultivars was also increased by Si. These observations implied an essential role for Si in minimizing drought stress-induced limitation of the growth and oxidative membrane damage in chickpea plants.  相似文献
