Many species of bats secrete a wide variety of substances, frequently associated with olfactory communication. We characterized a seasonal phenomenon of dorsal sebaceous secretion in the Cura?aoan long-nosed bat, Leptonycteris curasoae, in Venezuela, and the lesser long-nosed bat, Leptonycteris yerbabuenae, in Mexico. The phenology of the sebaceous patch was determined, a histological analysis of the affected area was conducted using specimens of L. curasoae from Venezuela, and finally, a preliminary chemical characterization of the substance secreted was performed combining histochemical techniques with gas chromatography and mass spectrometry analyses. The sebaceous patch was detected exclusively in male adult specimens. Individuals presenting it had a variable area of fur covered with a fatty and odoriferous substance at the level of the interscapular zone. Occurrence of the sebaceous patch was cyclical and coincided with the mating season in Venezuela and Mexico. The following histological changes associated with occurrence of the patch were observed: increase of epidermis thickness and decrease of dermis and hypodermis thicknesses, increase in density of sebaceous glands, increase of percentage of skin covered by sebaceous glands, increase of size of sebaceous glands previous to secretion followed, and increase of the sebum volume within sebaceous glands previous to secretion. Several compounds tentatively identified as fatty acids, cholestanes and cholesterol were present in the sebaceous secretion. Based on the evidence obtained, we hypothesize that the sebaceous patch could be involved in olfactory communication, possibly related to mating behavior in these bats.  相似文献

Cell-based quantitative evaluation of the MTT assay   总被引:5,自引:0,他引:5  
OBJECTIVE: To analyze the bioreduction of 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) on a per cell basis and evaluate its modulation as a function of different stages of cell metabolism. STUDY DESIGN: Following MTT bioreduction, total optical density (TOD), cell area and specific activity (TOD/area) of V79 cells and cultured macrophages were recorded for individual cells by means of digital image analysis. The effect of different serum (0-10% vol/vol) or genistein (0-100 microM) concentrations was used to modulate the MTT-specific activity response. RESULTS: As cells in culture are heterogeneous in cell size, the contribution of each cell to the total amount of formazan formed per dish is variable. The production of formazan per cell as a result of MTT bioreduction was found to be proportional to cell size. CONCLUSION: Specific MTT-reducing activity was analyzed in phagocytes and nonphagocyte cells, revealing the utility of this variable in evaluating the MTT assay at the single-cell level.  相似文献

In vivo confocal Raman spectroscopy is a noninvasive optical method to obtain detailed information about the molecular composition of the skin with high spatial resolution. In vivo confocal scanning laser microscopy is an imaging modality that provides optical sections of the skin without physically dissecting the tissue. A combination of both techniques in a single instrument is described. This combination allows the skin morphology to be visualized and (subsurface) structures in the skin to be targeted for Raman measurements. Novel results are presented that show detailed in vivo concentration profiles of water and of natural moisturizing factor for the stratum corneum that are directly related to the skin architecture by in vivo cross-sectional images of the skin. Targeting of skin structures is demonstrated by recording in vivo Raman spectra of sweat ducts and sebaceous glands in situ. In vivo measurements on dermal capillaries yielded high-quality Raman spectra of blood in a completely noninvasive manner. From the results of this exploratory study we conclude that the technique presented has great potential for fundamental skin research, pharmacology (percutaneous transport), clinical dermatology, and cosmetic research, as well as for noninvasive analysis of blood analytes, including glucose.  相似文献

Generation of skin appendages in engineered skin substitutes has been limited by lack of trichogenic potency in cultured postnatal cells. To investigate the feasibility and the limitation of hair regeneration, engineered skin substitutes were prepared with chimeric populations of cultured human keratinocytes from neonatal foreskins and cultured murine dermal papilla cells from adult GFP transgenic mice and grafted orthotopically to full-thickness wounds on athymic mice. Non-cultured dissociated neonatal murine-only skin cells, or cultured human-only skin keratinocytes and fibroblasts without dermal papilla cells served as positive and negative controls respectively. In this study, neonatal murine-only skin substitutes formed external hairs and sebaceous glands, chimeric skin substitutes formed pigmented hairs without sebaceous glands, and human-only skin substitutes formed no follicles or glands. Although chimeric hair cannot erupt readily, removal of upper skin layer exposed keratinized hair shafts at the skin surface. Development of incomplete pilosebaceous units in chimeric hair corresponded with upregulation of hair-related genes, LEF1 and WNT10B, and downregulation of a marker of sebaceous glands, Steroyl-CoA desaturase. Transepidermal water loss was normal in all conditions. This study demonstrated that while sebaceous glands may be involved in hair eruption, they are not required for hair development in engineered skin substitutes.  相似文献

The study of the stimulating effect of the microgels (MGs) based on recombinant 1F9 spidroin on the regeneration of the deep skin wound in mice was carried out. The use of spidroin MGs was shown to increase significantly the quality of healing compared to the control. The introduction of the MG in the wound edges led to recovery of all the structural elements of the skin: the epidermis, the dermis, including vascular and nervous network, in the periphery of the wound underlying muscles, and skin appendages (sebaceous and sweat glands and hair follicles) was revealed.  相似文献

The histochemistry of glycoconjugates in the nasolabial skin of the goat has been studied by means of a series of selected methods of light microscopy. The epidermis of the nasolabial skin was found to contain neutral and acid glycoconjugates with different saccharide residues. The secretory epithelial cells and secretory substances of the sebaceous glands contained primarily neutral glycoconjugates, whereas those of the apocrine glands involved largely strongly acidic and neutral glycoconjugates. In the epithelial cells and secretory substances of the nasolabial eccrine glands, glycoconjugates involved were characteristically strongly neutral but weakly acidic in nature. From the present results, the histophysiological significance of glycoconjugates in the particular histologic structures of the nasolabial skin has been discussed with special reference to the functions of this particular skin type in the goat.  相似文献

Extracts of skin with sebaceous glands contain a substance which inhibits mitotic activity in sebaceous glands both in vivo and in vitro. This substance is neither the epidermal chalone nor the melanocyte chalone, both of which are also present in skin extracts. However, it resembles these other chalones in that it is water soluble, is precipitated by ethanol, is activated by the two stress hormones adrenalin and hydrocortisone, and is not species specific. It is present within the sebaceous glands, and it is evidently a sebaceous gland chalone.  相似文献

To test the hypothesis that the primer pheromone responsible for inducing the "male effect" is produced in the sebaceous gland androgen dependently, we examined the correlation between morphological changes of sebaceous glands and the pheromone activity in skin samples taken from castrated goats that had been treated with testosterone. Five castrated goats were implanted s.c. with testosterone capsules to maintain physiological levels of plasma testosterone for four weeks. Skin samples were obtained from the head region on Day 0 (the day of testosterone implant), Day 7, Day 14, Day 28 (the day of testosterone removal), Day 36, Day 42, and Day 56. Matched blood samples were also collected for measurement of testosterone concentration. The pheromone activity of the ether-extracts of the upper dermal layer containing sebaceous glands was assessed by its stimulatory effect on the hypothalamic GnRH pulse generator, which was monitored for changes of specific multiple unit activity (MUA) in ovariectomized estradiol-primed goats as described previously. The sebaceous gland enlarged during the testosterone treatment but reduced in size after testosterone removal. The pheromone activity first appeared in 2 out of 5 goats on Day 7 and in all the 5 goats by Day 28. Fourteen days after testosterone removal (Day 42), the pheromone activity was no longer detectable in any of the 5 goats. In short, the sebaceous gland size and the pheromone activity shifted almost in parallel. The present results provide strong support for the view that the primer pheromone is produced testosterone dependently in the sebaceous gland of the male goat.  相似文献

Using immunohistochemistry, the study clearly demonstrates three important β-glucan receptors (Ficolin/P35, MBL, Dectin-1; members of the lectin-complement pathway of innate immunity) in the integument of six marine and freshwater aquatic mammals (Northern fur seal, Common seal, Walrus, Coypu, Capybara, Otter), but only weakly in two dolphin species. Most of the non-dolphin mammals exhibited strong reactions, especially with regard to the skin glands (tubular apocrine glands, sebaceous glands), for L-Ficolin/P35 and MBL. Distinct reaction staining could also be observed in the epidermis and the outer epithelial sheath of primary hair follicles. Positive Dectin-1 staining was limited to secretory cells of the apocrine tubular glands, and to peripheral and central cells of sebaceous glands of the seals. The Capybara was the only animal to show a clear Dectin reaction in the epidermis (stratum granulosum). The findings are discussed with regard to the constant and high microbial challenge of the skin in the aquatic medium, and variations in hair density of the animals.  相似文献

The important role of epidermal appendages especially the sebaceous gland has only recently been recognized. In particular, it has been convincingly shown that normal development and maintenance of the sebaceous gland are required for skin homeostasis since atrophic sebaceous glands and disturbances in sebaceous lipid composition result in major defects of the physiological barrier and maintenance of the skin. Consequently, it is important to unravel the signaling network controlling proper sebaceous lineage differentiation in mammalian skin and to understand the underlying mechanisms leading to severe skin diseases, including abnormal proliferation and differentiation of the gland, defects of the lipid metabolism and barrier, as well as sebaceous tumor formation. Over the last years, results from transgenic and knock out mouse models manipulating distinct signaling pathways in the skin as well as the detailed analysis of human sebaceous gland-derived cell lines provided new insights into crucial mediators balancing proliferation and differentiation of the sebaceous gland. Here, we discuss our current knowledge of in vivo mechanisms of sebaceous gland development, maintenance and disorders and highlight recent contributions to the field of sebaceous gland biology.  相似文献

We examined the histological structure of 8 skin areas thought to contain cutaneous glands of potential importance in scent communication in 16 brown brocket deerMazama gouazoubira Fisher, 1814, using standard histological techniques. Frontal areas and preorbital sacs had scant glandular development. Sebaceous gland development was prominent in vestibular nasal glands and prepucial glands. Apocrine sudoriferous glands and sebaceous glands were well developed in tarsal glands, the caudal skin area and the interdigital glands of front and hind feet. The tail had a unique arrangement of apocrine sudoriferous glands. Anal glands had moderate glandular development, and metatarsal glands were absent. Several of these glandular areas may be important in the chemical communication among brocket deer.  相似文献

The objective was to investigate how nitrogen allocation patterns in plants are affected by their vertical position in the vegetation (i.e. being either dominant or subordinate). A garden experiment was carried out with Amaranthus dubius L., grown from seed, in dense stands in which a size hierarchy of nearly equally aged individuals had developed. A small number of dominant plants had most of their leaf area in the highest layers of the canopy while a larger number of subordinate plants grew in the shade of their dominant neighbours. Canopy structure, vertical patterns of leaf nitrogen distribution and leaf photosynthetic characteristics were determined in both dominant and subordinate plants. The light distribution in the stands was also measured. Average N contents per unit leaf area (total canopy nitrogen divided by the total leaf area) were higher in the dominant than in the subordinate plants and this was explained by the higher average MPA (leaf dry mass per unit area) of the dominant plants. However, when expressed on a weight basis, average N contents (LNCav; total canopy N divided by the total dry weight of leaves) were higher in the subordinate plants. It is possible that these higher LNCav values reflect an imbalance between carbon and nitrogen assimilation with N uptake exceeding its metabolic requirement. Leaf N content per unit area decreased more strongly with decreasing relative photon flux density in the dominant than in the subordinate plants showing that this distribution pattern can be different for plants which occupy different positions in the light gradient in the canopy. The amount of N which is reallocated from the oldest to the younger, more illuminated leaves higher up in the vegetation may depend on the sink strength of the younger leaves for nitrogen. In the subordinate plants, constrained photosynthetic activity caused by shading might have reduced the sink intensity of these leaves.  相似文献

The purpose of this study was to ascertain (1) whether human saliva contains irisin and whether its level correlates with serum irisin concentration, (2) whether salivary glands, eccrine glands and sebaceous glands in human skin produce irisin, (3) how the changes in saliva and serum irisin concentrations after the Turkish bath at 47 ± 3 °C compare with the changes caused by moderate exercise in obese and normal weight subjects. Seven obese male subjects and seven normal weight subjects were enrolled for Turkish bath. Seven obese male subjects and seven normal weight subjects were also enrolled for moderate outdoor exercise, and thirteen male normal weight subjects neither exercised nor showered at the Turkish bath. From each participant, 1.5 ml of saliva and 5 ml blood were collected simultaneously before and after the moderate exercise and Turkish bath. Salivary glands and eccrine and sebaceous glands in the skin were screened immunohistochemically for irisin while serum and saliva irisin were measured with an ELISA. Submandibular glands, eccrine glands and sebaceous glands in the human skin showed strong irisin immunoreactivity. Human saliva contained irisin and its level was significantly higher than the serum levels in both obese and normal weight subjects. However, irisin concentrations were more markedly increased in both saliva and serum samples from subjects who had showered at a Turkish bath than in obese subjects who had exercised or in normal weight subjects. Human skin fibroblast monolayers (S-126 cell line) were exposed to laser radiation (wavelength 670 nm, power density 40 mW/cm2). The energy densities were 2 J/cm2 and 12 J/cm2, respectively, and the irradiation was carried out at a temperature of 22°C. For fibroblast viability evaluation, the colorimetric assay (conversion of thiazolyl blue to formazan) was used. The experiments were carried out at 37°C, in the presence of 5% CO2, and at different time periods of incubation after irradiation (2, 4, 8 h and 1, 2, 3, 4, 5 days). The results indicated that there was a certain stimulating effect on the long-term proliferation of skin fibroblasts and that the stimulation proceeded in two stages, the first one 2 h and the second one 3 days post-irradiation. From an analytical model it was shown that for a given total amount of nitrogen in the canopy, there exists an optimal leaf area index (LAI), and therefore an optimal average leaf introgen content, at which canopy photosynthesis is maximal. If the LAI is increased above this optimum, increased light interception will not compensate for reduction in photosynthetic capacity of the canopy resulting from reduced leaf nitrogen contents. It was further derived from the model that the value of the optimal LAI increases with the photosynthetic nitrogen use efficiency (PNUE) and decreases with the canopy extinction coefficient for light (KL) and incident photon flux density (PFD) at the top of the canopy. These hypotheses were tested on dense stands of species with different photosynthetic modes and different architectures. A garden experiment was carried out with the C4 monocot sorghum ( Sorghum bicolor [L.] Moensch cv. Pioneer), the C3 monocot rice ( Oryza sativa L. cv. Araure 4), the C4 dicot amaranth ( Amaranthus cruentus L. cv. K113) and the C3 dicot soybean ( Glycine max [L.] Merr. cv. Williams) at two levels of nitrogen availability.
The C4 species had higher PNUEs than the C3 species while the dicots formed stands with higher extinction coefficients for light and had lower PNUEs than the monocots. The C4 and monocot species were found to have formed more leaf area per unit leaf nitrogen (i.e., had lower leaf nitrogen contents) than the C3 and dicot species, respectively. These results indicate that the PNUE and the extinction coefficient for light are important factors determining the amount of leaf area produced per unit nitrogen as was predicted by the model.  相似文献

The purpose of our study was to determine the effect of dilution and liquid-preservation of boar sperm on oxidoreductive capability of their mitochondria. The semen was diluted with BTS extender produced from water purified by destillation or by reverse osmosis. The spermatozoa were stored over a four-day period at 16-18 degrees C. The function of sperm mitochondria was assessed using the screening cytochemical test for NADH-dependent oxidoreductases (diaphorase/NADH, related to flavoprotein). Morphological assessment of cytochemical reaction was carried out using a light microscope. The intensity of the reaction was evaluated by means of a computer image analysing system (Quantimet 600S), measuring the integrated optical density (IOD) and mean optical density (MOD) of the reaction product (formazans) occurring in the sperm midpieces. In the non-diluted semen, intensive cytochemical reaction throughout the length of the sperm midpiece was observed. Furthermore, spermatozoa with the intensive reaction displayed the high optical density values. After dilution the semen with two variants of experimental extender, and as the conservation time expired, the cytochemical reaction was less intensive. Moreover, the absence of formazan deposits in various parts of the sperm midpiece was also noted. These morphological features corresponded to low values of optical density. These findings suggest that the dilution of semen and the time of sperm preservation may be critical factors that handicap energy metabolism of sperm mitochondria. The type of water used in preparing BTS extender does not have any significant effect on the oxidoreductive capability of sperm boar mitochondria.  相似文献

To examine the usefulness of the pika, Ochotona rufescens rufescens, as an experimental animal for skin irritability tests, the histological structure and enzymatic histochemical reactivity of pika skin were investigated. The pika had a hair cycle similar to that of the rabbit. The skin and epidermis of the pika trunk were 1.16mm and 29.5 microns thick, on the average, respectively. Both of them were the thickest in the dorsal region followed by the interscapular area, while they were the thinnest in the abdominal region. In the epidermis of the pika, the strata corneum, granulosum, spinosum and basale were rather clearly distinguished. The cell arrangement in the stratum basale was more compact than that in the rabbit. Dermal mast cells, which are distributed in the stratum reticulare in rabbits and guinea pigs, were distributed in the stratum papillare right beneath the epidermis. The mast cell of the pika in the TEM images had granules of low electron density and with relatively long microvilli and rather large mitochondria. The activities of the enzymes, SDH, MDH, LDH, beta HBDH, alpha GPDH, ALD, G6PDH and GPR, in the hair follicles and sebaceous glands of the pika were similar to those of the rabbit.  相似文献

Liposomes have frequently been used as models of biomembranes or vehicles for drug delivery. However, the systematic characterization of lipid vesicles by right angle light scattering and turbidity has not been carried out despite the usefulness of such studies for size estimation. In this study, liposomes of various sizes were prepared by sonication and extrusion. The mean cumulant radii of the vesicles were determined by dynamic light scattering. The lamellarities were estimated based on fluorescence quenching of N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)dipalmitoyl-L-alpha-phosph ati dylethanolamine by sodium dithionite. Right angle light scattering intensity and optical density at 436 nm per unit lipid concentration were measured as a function of vesicle radius. With a vesicle radius < or =100 nm, the optical parameters could be well explained by the Rayleigh-Gans-Debye theory in which the liposomes were modeled as homogeneous spheres with mean refractive indices determined by the volume fractions of lipids in vesicles.  相似文献

应用回归均匀设计构造试验方案,采用分期播种的方法,研究了不同冬前积温、种植密度对饲用黑麦单株分蘖动态、群体分蘖动态以及有效茎数动态变化的影响,并建立了模拟模型。结果表明,饲用黑麦单株分蘖数量同时受播期、播量的影响,生长前期主要受播期早晚的影响,生长后期主要受播种密度的制约,总体来看,播期越早、播量越小,其单株分蘖数量就越多,反之就越少,饲用黑麦不论是在生长前期、中期,还是后期,随着播期推迟,各播量群体分蘖数不断下降;随着播量增大,各播期群体总蘖数不断上升,饲用黑麦群体有效茎数取决于播期和播量的共同作用,播期早,要控制播量不宜过大,合理的群体结构主要依靠单株有效茎数的增长潜力来保证;播期晚,要防止播量过小,合理的群体结构主要依靠主茎数量的增加来实现。  相似文献

BACKGROUND AND AIMS: The production of axillary shoots (tillering) in spring wheat (Triticum aestivum) depends on intraspecific competition. The mechanisms that underlie this competition are complex, but light within the wheat canopy plays a key role. The main objectives of this paper are to analyse the effects of plant population density and shade on tillering dynamics of spring wheat, to assess the canopy conditions quantitatively at the time of tillering cessation, and to analyse the relationship between the tiller bud and the leaf on the same phytomer. METHODS: Spring wheat plants were grown at three plant population densities and under two light regimes (25 % and 100 % light). Tiller appearance, fraction of the light intercepted, and red : far-red ratio at soil level were recorded. On six sampling dates the growth status of axillary buds was analysed. KEY RESULTS: Tillering ceased earlier at high population densities and ceased earlier in the shade than in full sunlight. At cessation of tillering, both the fraction of light intercepted and the red : far-red ratio at soil level were similar in all treatments. Leaves on the same phytomer of buds that grew out showed more leaf mass per unit area than those on the same phytomer of buds that remained dormant. CONCLUSIONS: Tillering ceases at specific light conditions within the wheat canopy, independent of population density, and to a lesser extent independent of light intensity. It is suggested that cessation of tillering is induced when the fraction of PAR intercepted by the canopy exceeds a specific threshold (0.40-0.45) and red : far-red ratio drops below 0.35-0.40.  相似文献
