Lakes lying within the blast zone of Mount St. Helens showed dramatic increases in heterotrophic bacterial numbers after the eruption of 18 May 1980. The total microscopic counts of bacteria in some of the most severely affected lakes were more than 107 cells per ml, an order of magnitude above the counts in outlying control lakes. Likewise, the numbers of viable bacteria reached levels of more than 106 cells per ml, compated with fewer than 104 cells per ml in control lakes. The CPS medium used for enumeration provided growth of up to 81.5% of the bacteria during sampling of one of the blast zone lakes. The high numbers of bacteria and the efficacy of the viable enumeration procedure are evidence that the lakes have been transformed rapidly from oligotrophy to eutrophy due to the eruption and its aftermath. Organic material leached from the devastated forest vegetation is thought to be responsible for the enrichment of heterotrophs. Total coliform bacteria were found in all of the blast zone lakes, and some lakes contained fecal coliform bacteria. Klebsiella pneumoniae was the predominant total coliform and was also identified as one of the fecal coliform bacteria, although Escherichia coli was the predominant species in that category. Our data indicate that bacterial populations peaked in the outer blast zone lakes in the summer of 1980 and in most of the inner lakes during the summer of 1981.  相似文献

Despite the destruction and total rearrangement of much of the area north of Mt St. Helens, many different species of algae became established in the aquatic systems located in the blast zone within 3–4 months after the eruption of May 18, 1980. Initial sites of intense algal activity were found in benthic regions associated with warm springs and in the littoral and phytoplanktonic communities of some small lakes. In the summers of 1980–81, overall phytoplankton numbers and activity were low in the large lakes closest to the crater (e.g. Spirit Lake). However, diatoms, blue-green and green algal isolates from these lakes were obtained in laboratory cultures on a reduced inorganic medium used to enumerate metal and sulfur oxidizing bacteria. Species of Nodularia, Pseudanabaena, Anabaena, Oscillatoria, Nitzschia, Tribonema, Chlamydomonas, Chlorella, and microflagellates (predominantly cryptomonads) were among the more common forms found in preserved samples or isolated in cultures.  相似文献

Survival ofLegionella pneumophila SG 1 in seawater and river water was assessed using plate counts on buffered charcoal yeast extract agar amended with α-ketoglutarate (BCYEα) and [3H]thymidine-labeling. The [3H]thymidine-labeling method for assessing survival ofL. pneumophila in aquatic environments was compared with viable counts, direct fluorescent microscopy (DFA), and acridine orange direct counts (AODC). Protozoa were isolated from the samples employed in the study and identified by characteristic trophozite and cyst morphology. Selective filtration employing 2.0 μm Nucleopore filters was used to determine the effect of grazing on survival ofL. pneumophila in seawater and river water.Legionella viability as measured by plate counts (CFU/ml), declined to a greater extent than cell lysis, assessed by thymidine, DFA, and AODC counts, suggesting thatL. pneumophila survives in aquatic habitats to a greater extent than revealed through culturable counts.  相似文献

Dead fir needles were collected from standing trees of Abies amabilis in five sites at the margin of the blast zone and located progressively farther from the eruption crater of Mount St. Helens. Scanning electron microscope techniques were used to determine patterns of cuticular melting for these needles and for Abies grandis needles which were heated at specific temperatures for 2 min. Comparisons between cuticular appearances of oven-heated needles and needles from Mount St. Helens were made to determine air temperatures at the collection sites at the time of the eruption. Air temperatures at these sites are estimated to have ranged from about 50 C to about 250 C. Analysis of cuticular sulfur content showed these needles adsorbed little or no volcanic S02. Conifer needles provided a record of maximum air temperatures during the eruption, and helped reveal the pattern of heat distribution from the eruption cloud. This technique may prove useful for ecological studies in other heat-stressed habitats.  相似文献

Mt. St. Helens, a volcanic peak in the Cascade Range in southern Washington erupted violently on May 18, 1980, causing enormous damage to both terrestrial and aquatic ecosystems. The initial explosion evaporated, scoured or buried all springs and streams in the blast impact area. Ash fall and erosion from defoliated hillsides subsequently filled most of the lotic habitats with organic debris and volcanic ash. Recolonization of springs and streams by algae occurred quickly in areas where erosion through the ash progressed down to bedrock. Within 15 months or less of the eruption, algal communities were established throughout the blast impact area. However, as a result of the initial and continued disturbance these communities remained in an early successional stage. Floral assemblages were highly variable except that they were composed mostly of diatoms, with Achnanthes minutissima dominating most lotic sites. Springs showed the most rapid development toward stable floras.  相似文献

Abstract. 1. The 1980 eruption of Mount St Helens (Washington, U.S.A.) created a 60‐km2 region of primary successional habitat. Since colonising in 1981, the spatial spread of the legume Lupinus lepidus at Mount St Helens, Washington, U.S.A., has afforded intriguing opportunities to study the effect of trophic dynamics on primary succession. 2. Insect herbivory on this lupine has exhibited striking spatial structure for over a decade, with inverse density‐dependent damage patterns occurring over both small (10–100 m) and large (1–10 km) spatial scales. 3. Hypothesising that lupine nutritional chemistry might underlie the spatial patterns in herbivory, the distribution of elemental macronutrients (nitrogen, phosphorus) across the landscape was characterised. 4. Samples of soil and lupine tissue (roots and leaves) were collected from sites along both local and regional gradients in lupine density. On both large and small spatial scales, lupine leaves from low‐density conditions were significantly more nutrient rich. 5. In addition, in a laboratory growth study native lepidopteran herbivores that specialise on lupines (Gelechiidae: Filatima sp.) performed better when fed leaves from low‐density, high‐nutrient lupines than on diets of low‐nutrient lupine leaves from high‐density areas a few metres away. 6. These data suggest that spatial heterogeneity in lupine nutrient chemistry may underlie the remarkable herbivory gradients witnessed at Mount St Helens.  相似文献

1. The vulnerability of softwater, oligotrophic lakes to eutrophication has caused the disappearance of many, if not most, of the unique isoetid plant communities. We tested whether the presence or disappearance of the isoetid Littorella uniflora (L.) could be predicted from environmental parameters, soil types and land use in the catchment area, and atmospheric nitrogen deposition. 2. We found that the topographic catchment area of a lake was an irrelevant unit to study effects of soil type and land use. Instead, using a GIS‐generated buffer zone around the lakes it proved feasible to classify 472 lakes into historical (if L. uniflora had disappeared) or recent (if L. uniflora was still present) Littorella lakes, based on soil type and land use. Our analysis showed that aeolian sand deposits and heath in the buffer zone favoured the presence of L. uniflora, whereas moraine clay and agriculture were strongly linked to the disappearance of L. uniflora. 3. However, in order to understand fully the presence or disappearance of L. uniflora, environmental data were needed in addition to soil types, land use and nitrogen deposition, and the use of discriminant analysis allowed us to classify 96% of the investigated lakes correctly into recent or historical sites. Alkalinity, total phosphorus, total nitrogen, aeolian sand deposits and heath were the most important parameters explaining the presence or disappearance of L. uniflora. Our analysis also indicated that eutrophication, rather than acidification, has likely caused the disappearance of L. uniflora from 218 of the 472 lakes investigated. 4. Our findings have widespread implications for the conservation or restoration of isoetid habitats and we recommend applying a wide buffer zone around lakes, with restrictions on farming and traditional forestry activities. In addition, our buffering concept may prove a useful tool for aquatic ecologists to investigate relationships between catchment features and organisms (plants, insects and amphibians) with aquatic as well as terrestrial life forms.  相似文献

To determine the effects of tephra (volcanic aerial ejecta) on forest understory plants, six sites were chosen along a tephra depth gradient (23 to 150 mm) northeast of Mount St. Helens, USA. All sites were in old forests beyond the limits of direct blast damage from the volcanic eruption. At each site, 150 one m2 plots were permanently marked; all tephra was removed from 50 of these in 1980. Cover and density of plant species were recorded during 1980, 1981, and 1982.Tephra 23 mm deep had almost no effect on cover and density of vascular plants, and reduced bryophyte cover for only two years. Tephra 45 mm deep destroyed almost all bryophytes. Although damaged by 45 mm tephra, deciduous herbs recovered by 1982, but some evergreen species did not. Tephra 75 mm deep reduced herb cover in 1982 to 32% and density to 26% of that in cleared plots. At two sites with an average tephra depth of 150 mm, almost all herbs were eliminated except in microsites where tephra was thin, but shrub abundance was greatly reduced only where snow had been present during tephra deposition. Almost all cover was contributed by plants established previous to the eruption; seedling cover never exceeded 0.2%. Refugia with thin tephra, resulting from erosion, were vital to the survival of many species, especially bryophytes.Nomenclature of vascular plants follows Hitchcock & Cronquist (1973); moss nomenclature follows Lawton (1971).We thank T. The vertical distribution and abundance of Gammarus lacustris in the pelagic zone of two fishless meromictic lakes, L. Shira and L. Shunet, in Southern Siberia (Russia), was studied with the underwater video recording system and using vertical hauls. In both lakes, during summer stratification, Gammarus was distributed non-homogenously, with a stable peak in the metalimnion. The average depth of Gammarus population in the pelagic zone was significantly correlated with the depth of the thermocline. Gammarus abundances obtained using vertical plankton hauls with net were quite comparable with those obtained from video records. The peak abundance of Gammarus in the pelagic zone of the lakes observed with underwater video amounted up to 400 individuals m−2, while the peak animal densities in the metalimnion reached 50 ind. m−3. The data are compared with previously published abundances of Gammarus in the littoral of Lake Shira. Both littoral and pelagic can be equally important habitats for amphipods in meromictic lakes. The absence of fish in the pelagic zone, high oxygen concentration, low water temperature, increased seston concentration, elevated water density in the metalimnion and the anoxic hypolimnion can be the most probable combination of factors that are responsible for the peak of Gammarus in the metalimnion of these lakes.  相似文献

Three detection methods for Legionella species in water samples from cooling towers and a river were examined. Direct counting of bacteria stained with fluorescent antibody (FA) for L. pneumophila (serogroups 1 to 6) could detect the cell of 104 to 106 cell/100 ml in all 14 samples, while colony counting method detected 10 to 103 CFU/100 ml only in 8 samples from cooling towers. Polymerase chain reaction (PCR) assay with primers to amplify 16S ribosomal DNA sequence of most Legionella species (LEG primer) detected legionellae in 13 samples, while species-specific primers for L. pneumophila detected the DNAs from 3 samples. In laboratory examination, LEG primers could amplify DNAs of 29 species of genus Legionella with high sensitivity, even from 1 cell of L. pneumophila GIFU 9134. The PCR assay with LEG primers was specific and sensitive methods to be satisfied the survey of legionellae. Thus, PCR assay is a suitable method to detect and monitor Legionella species in an environment.  相似文献

Influence of disturbance on insect communities in Pacific Northwest streams   总被引:2,自引:2,他引:0  
Coniferous forests of the Pacific Northwest provide a unique setting for stream ecology research because of the great age of the forests and the important role of wood debris in structuring aquatic systems. The composition and diversity of the insect community in Mack Creek, a stream in a 450 yr conifer forest, was compared with that in Grasshopper Creek where it flowed through a recent clearcut, and at Quartz Creek, which had a 40 yr deciduous canopy. Of the 256 taxa identified, Mack Creek had the highest species richness (196) and evenness. The open site had 191 taxa but high dominance of a few grazer taxa. The deciduous-canopy site had 165 taxa with abundant detritivores. Despite differences in density, the biomass of emergence was similar at the three sites, ranging from 1.53 to 1.65 g m–2 yr–1.The importance of disturbance in structuring stream communities was demonstrated in phenomenological studies after a debris torrent at Quartz Creek, and by monitoring stream recovery following the eruption of Mt. St. Helens in 1980. At Quartz Creek, the debris torrent eliminated the fauna from a 300 m reach, but there was rapid recovery. Emergence density in the same year was similar to that of the control site. The major shift in populations was a decrease in detritivores and moss associates and an increase in grazers, especially Baetis mayflies.At Ape Creek on Mt. St. Helens, over 200 taxa were recorded by 1987 but most occurred in very low densities. This site is reset by winter freshets and by infilling with glacial fines in the summer so the fauna continues to be dominated by weedy, or early successional species. At Clearwater Creek, the presence of wood debris as a stable substrate and limited inputs of fine sediment after 1980 have hastened population recovery. A decade after the eruption this site can be characterized as being in the mid-stages of succession with high insect productivity from an algal-based food web. With further growth of the riparian vegetation I predict a shift towards a detritus-based food web and fauna more similar to Mack Creek than it is at present.  相似文献

Legionella pneumophila, a micro-organism encountered in aquatic environments, can cause serious intracellular infections among humans. Since the bacterium is ubiquitous in aquatic habitats, it appears to be impossible to prevent L. pneumophila from entering man-made water systems. However, many questions concerning the survival and/or growth in the environment, the partners and opponents of L. pneumophila remain unanswered. This review focuses on the factors governing the ecology of L. pneumophila, since there is considerable divergence and even contradiction in literature on its environmental requirements. A key question to be resolved is the discrepancy between the fastidious nature of L. pneumophila in axenic cultures (e.g. 400 mg l−1 L-cysteine and 250 mg l-1 ferric iron) and the nutritionally poor environments in which it is commonly detected. It is assumed that dense microbial communities, as occurring in sediments and biofilms – but not likely in surface and drinking water, – can provide the necessary growth requirements for L. pneumophila. However, most of the studies concerning L. pneumophila have led to the general opinion that the organism can only multiply in the aquatic environment as a parasite in certain protozoa. The discovery of the non-classical siderophore legiobactin also indicates that the iron requirement for survival and autonomous growth is not as high as has been assumed. It thus appears that in order to control Legionella in the environment, focus should be on the eradication of microbial hotspots in which L. pneumophila resides.  相似文献

Biofilms are a major source of human pathogenic Legionella pneumophila in aquatic systems. In this study, we investigated the capacity of L. pneumophila to colonize floating biofilms and the impact of Acanthamoeba castellanii on the replication of biofilm-associated Legionella. Biofilms were grown in Petri dishes and consisted of Aeromonas hydrophila, Escherichia coli, Flavobacterium breve, and Pseudomonas aeruginosa. Six hours following inoculation, Legionella were detected in floating biofilms in mean concentrations of 1.4 × 104 cells/cm2 (real-time polymerase chain reaction) and 8.3 × 102 CFU/cm2 (culture). Two-way analysis of variance tests and fluorescent in situ hybridization clearly proved that increased biofilm-associated L. pneumophila concentrations were the result of intracellular replication in A. castellanii. Forty-eight hours after the introduction of A. castellanii in the Petri dishes, 90 ± 0.8% of the amoebae (infection rate) were completely filled with highly metabolic active L. pneumophila (mean infection intensity).  相似文献

Mice are highly resistant to infection byLegionella pneumophila compared with guinea pigs. In the present study injection of mice with moderate doses of cyclophosphamide before and at the time of challenge injection with viable legionellae made the animals much more susceptible to these organisms. The doses of cycloplosphamide used for the short time suppressed the mice in terms of their ability to form antibody and develop cellular immunity toLegionella antigens as measured by the splenic blastogenic assay. Vaccination of mice with a sublethal dose ofLegionella (106 organisms) or with 107 killed bacteria in complete Freund's adjuvant protected the cyclophosphamide-treated mice from infection with liveLegionella. It seems apparent that a vaccine prepared from live organisms or killed bacteria in adjuvant may be protective for immunosuppressed mice.  相似文献

 Mt. St. Helens is an active volcano on the western flank of the Cascade: range in the southwestern Washington State, USA. It is located at lat. 46˚00′—46˚30′N, long 121˚52′—122˚40′W.We had investigated the alpine vegetation at Pine Creek, Butte Camp on Mt. St. Helens for three times, totaling ten days during August and eptember of 1983.The volcano had erupted sending out much pyroclastic, pumice and lava, which devdstated the vegetation around the crater. They not only mechanically destroyed plants tissue, but also buried entirely or partly of the plants.The radiant heat from the volcanic eruption caused the melting of ice and snow, creating huge mudflows, which estroyed and buried vegetation. The high temperture from direct radiant heat incinerated and scorched all the plants. On the other hand, the higher the elevation is, the stronger the wind and the lower the temperture; also snow was present on the alpine range, and this provided a lee for plants, and consequently some plants survived, and new ones row out of the ground. The alpine vegetation on Mt. St. Helens bears both the characteristic of alpine plants and the brand of volcanic activities: vegetation is sparse, species composition of plants limited, structure of community simple, production low and vegetation propagation being the main type of vegetative reproduction.The different types of lpine vegetation grow in different habitats: The community of Luekea pectinata grow in places covered by ice and snow; the community of Polygonum newberryi, Eriogonum pyroleafolium in localities covered by mudflow; the community of Phyllodace empetriformis on slopes of steep rocks and the community of grasses and sedges on low hills and plains. Hese are the four main communities on Mt. St. Helens. The recovery and succession of the alpine vegetation has been discussed. There are two types of succession, the primary and the secodary, with the secondary being the main one.Finally, the paper has analysed the reason why no forest occurs, on Mt. St. Helens. We think that it is a result of the ctive period of the volcano and that the climate conditions are favouable for forest. Also, we have suggested that some herbaceous plants be sown as soon as possible for they can accelerate the recovery of vegetation, and prevent the runoff of soil and water.  相似文献

Legionella species are ubiquitous bacteria in aquatic environments. To examine the effect of anthropogenic impacts and physicochemical characteristics on the Legionellaceae population, we collected water from two sites in the Itanhaém River system in the Atlantic Forest of Brazil. One sample was collected from an upstream pristine region, the other from a downstream estuarine region moderately affected by untreated domestic sewage. Cultures on a selective medium failed to isolate Legionella species. Culture-independent methods showed that water from the estuarine aquatic habitat contained DNA sequences homologous to the 16S ribosomal DNA gene of Legionella pneumophila and non-pneumophila species. In pristine water, only two sequences related to L. pneumophila were detected. The results suggest that salinity and anthropogenic factors, such as wastewater discharge, favor a diversity of Legionella species, whereas pristine freshwater selects for Legionella pneumophila.  相似文献

Ryan Lake, a 1.6-hectare basin lake near the periphery of the tree blowdown area in the blast zone 19 km north of Mount St. Helens, was studied from August to October 1980 to determine the microbial and chemical response of the lake to the eruption. Nutrient enrichment through the addition of fresh volcanic material and the organic debris from the surrounding conifer forest stimulated intense microbial activity. Concentrations of such nutrients as phosphorus, sulfur, manganese, iron, and dissolved organic carbon were markedly elevated. Nitrogen cycle activity was especially important to the lake ecosystem in regulating biogeochemical cycling owing to the limiting abundance of nitrogen compounds. Nitrogen fixation, both aerobic and anaerobic, was active from aerobic benthic and planktonic cyanobacteria with rates up to 210 nmol of N2 cm−1 h−1 and 667 nmol of N2 liter−1 h−1, respectively, and from anaerobic bacteria with rates reaching 220 nmol of N2 liter−1 h−1. Nitrification was limited to the aerobic epilimnion and littoral zones where rates were 43 and 261 nmol of NO2 liter−1 day−1, respectively. Potential denitrification rates were as high as 30 μmol of N2O liter−1 day−1 in the anaerobic hypolimnion. Total bacterial numbers ranged from 1 × 106 to 3 × 108 ml−1 with the number of viable sulfur-metal-oxidizing bacteria reaching 2 × 106 ml−1 in the hypolimnion. A general scenario for the microbial cycling of nitrogen, carbon, sulfur, and metals is presented for volcanically impacted lakes. The important role of nitrogen as these lakes recover from the cataclysmic eruption and proceed back towards their prior status as oligotrophic alpine lakes is emphasized.  相似文献

We studied the ability ofLegionella to multiply in potable water samples obtained from investigations of nosocomial legionellosis. AutochthonousLegionella multiplied in three of 14 hospital water samples after incubation at 35°C and 42°C. All three samples were from hot water tanks. Multiplication did not occur when a selected sample was filtered through a 0.45-m membrane and reinoculated with indigenousLegionella. We isolated bothLegionella pneumophila and one or more species of free-living amoebae, primarity members of theHartmannellidae, from each of these hot water tank samples. Amoebae from a total of six hot water tank samples were used for cocultivation studies withL. pneumophila. All amoebae supported multiplication ofLegionella in coculture at 35°C. Four of six isolates of amoebae supported multiplication oflegionella at 42°C, while none supported multiplication at 45°C. Gimenez staining and electron microscopy showed thatLegionella multiplied intracellularly in amoebae. Control of these amoebae in potable water may prevent colonization and multiplication ofLegionella in domestic hot water systems.  相似文献

Abstract. Primary succession on Mount St. Helens, Washington State, USA, was studied using long-term observational and experimental methods. Distance from potential colonists is a major factor that impedes early primary succession. Sites near undisturbed vegetation remain low in plant cover, but species richness is comparable to intact vegetation. Sites over 500 m from sources of potential colonists have as many species, but mean species richness is much lower than in undisturbed plots. Cover is barely measurable after 11 growing seasons. Highly vagrant species of Asteraceae and Epilobium dominate isolated sites. Sites contiguous to undisturbed communities are dominated by large-seeded species. For a new surface to offer suitable conditions to invading plants, weathering, erosion and nutrient inputs must first occur. The earliest colonists are usually confined to specific microsites that offer some physical protection and enhanced resources. Primary succession on Mount St. Helens has been very slow because most habitats are isolated and physically stressful. Well-dispersed species lack the ability to establish until physical processes ameliorate the site. Species capable of establishment lack suitable dispersal abilities. Subsequently, facilitation may occur, for example through symbiotic nitrogen fixation, but these effects are thus far of only local importance. Lupinus lepidus usually facilitates colonization of other species only after it dies, leaving behind enriched soil lacking any competitors. Experiments and fine-scale observations suggest that successional sequences on Mount St. Helens are not mechanistically necessary. Rather, they result from local circumstances, landscape effects and chance.  相似文献
