Induction of stress ethylene production in the plant system is one of the consequences of salt stress which apart from being toxic to the plant also inhibits mycorrhizal colonization and rhizobial nodulation by oxidative damage. Tolerance to salinity in pea plants was assessed by reducing stress ethylene levels through ACC deaminase-containing rhizobacteria Arthrobacter protophormiae (SA3) and promoting plant growth through improved colonization of beneficial microbes like Rhizobium leguminosarum (R) and Glomus mosseae (G). The experiment comprised of treatments with combinations of SA3, G, and R under varying levels of salinity. The drop in plant biomass associated with salinity stress was significantly lesser in SA3 treated plants compared to non-treated plants. The triple interaction of SA3 + G + R performed synergistically to induce protective mechanism against salt stress and showed a new perspective of plant-microorganism interaction. This tripartite collaboration increased plant weight by 53%, reduced proline content, lipid peroxidation and increased pigment content under 200 mM salt condition. We detected that decreased ACC oxidase (ACO) activity induced by SA3 and reduced ACC synthase (ACS) activity in AMF (an observation not reported earlier as per our knowledge) inoculated plants simultaneously reduced the ACC content by 60% (responsible for generation of stress ethylene) in SA3 + G + R treated plants as compared to uninoculated control plants under 200 mM salt treatment. The results indicated that ACC deaminase-containing SA3 brought a putative protection mechanism (decrease in ACC content) under salt stress, apart from alleviating ethylene-induced damage, by enhancing nodulation and AMF colonization in the plants resulting in improved nutrient uptake and plant growth.  相似文献

The enzyme 1-aminocyclopropane-1-carboxylate deaminase catalyzes the degradation of 1-aminocyclopropane-1-carboxylic acid (ACC), the immediate precursor of the plant hormone ethylene, into α-ketobutyrate and ammonia. The enzyme has been detected in a limited number of bacteria and plays a significant role in sustaining plant growth and development under biotic and abiotic stress conditions by reducing stress-induced ethylene production in plants. We have screened 32 fluorescent Pseudomonas sp. isolated from rhizosphere and non-rhizosphere soils of different crop production systems for drought tolerance using polyethylene glycol 6000 (PEG 6000). Nine of these isolates were tolerant to a substrate metric potential of ?0.30 MPa (15 % PEG 6000) and therefore considered to be drought-tolerant. All of these drought-tolerant isolates were screened for ACC deaminase activity using ACC as the sole nitrogen source, and one (SorgP4) was found to be positive for ACC, producing 3.71?±?0.025 and 1.42?±?0.039 μM/mg protein/h of α-ketobutyrate under the non-stress and drought stress condition, respectively. The isolate SorgP4 also showed other plant growth-promoting traits, such as indole acetic acid production, phosphate solubilization, siderophore and hydrogen cyanide production. The ACC deaminase gene (acdS) from the isolate SorgP4 was amplified, and the nucleotide sequence alignment of the acdS gene showed significant homology with acdS genes of NCBI Genbank. The 16S rRNA gene sequencing analysis identified the isolate as Pseudomonas fluorescens. Both sequences have been submitted to the NCBI GenBank under the accession numbers JX885767 and KC192771 respectively.  相似文献

Our previous work showed that a consortium of three plant growth-promoting rhizobacterium (PGPR) strains (Bacillus cereus AR156, Bacillus subtilis SM21, and Serratia sp. XY21), termed as BBS for short, was a promising biocontrol agent. The present study investigated its effect on drought tolerance in cucumber plants. After withholding watering for 13 days, BBS-treated cucumber plants had much darker green leaves and substantially lighter wilt symptoms than control plants. Compared to the control, the BBS treatment decreased the leaf monodehydroascorbate (MDA) content and relative electrical conductivity by 40% and 15%, respectively; increased the leaf proline content and the root recovery intension by 3.45-fold and 50%, respectively; and also maintained the leaf chlorophyll content in cucumber plants under drought stress. Besides, in relation to the control, the BBS treatment significantly enhanced the superoxide dismutase (SOD) activity and mitigated the drought-triggered down-regulation of the expression of the genes cAPX, rbcL, and rbcS encoding cytosolic ascorbate peroxidase, and ribulose-1,5-bisphosphate carboxy/oxygenase (Rubisco) large and small subunits, respectively, in cucumber leaves. However, 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity was undetected in none of the culture solutions of three BBS constituent strains. These results indicated that BBS conferred induced systemic tolerance to drought stress in cucumber plants, by protecting plant cells, maintaining photosynthetic efficiency and root vigor and increasing some of antioxidase activities, without involving the action of ACC deaminase to lower plant ethylene levels.  相似文献

Plant growth-promoting bacteria (PGB) induce positive effects in plants, for instance, increased growth and reduced abiotic stresses susceptibility. The mechanisms by which these bacteria impact the host plant are numerous, diverse and often specific. Here, we studied the agronomical, molecular and biochemical effects of the endophytic PGB Bacillus subtilis B26 on the full life cycle of Brachypodium distachyon Bd21, an established model species for functional genomics in cereal crops and temperate grasses. Inoculation of Brachypodium with B. subtilis strain B26 increased root and shoot weights, accelerated growth rate and seed yield as compared to control plants. B. subtilis strain B26 efficiently colonized the plant and was recovered from roots, stems and blades as well as seeds of Brachypodium, indicating that the bacterium is able to migrate, spread systemically inside the plant, establish itself in the aerial plant tissues and organs, and is vertically transmitted to seeds. The presence of B. subtilis strain B26 in the seed led to systemic colonization of the next generation of Brachypodium plants. Inoculated Brachypodium seedlings and mature plants exposed to acute and chronic drought stress minimized the phenotypic effect of drought compared to plants not harbouring the bacterium. Protection from the inhibitory effects of drought by the bacterium was linked to upregulation of the drought-response genes, DREB2B-like, DHN3-like and LEA-14-A-like and modulation of the DNA methylation genes, MET1B-like, CMT3-like and DRM2-like, that regulate the process. Additionally, total soluble sugars and starch contents increased in stressed inoculated plants, a biochemical indication of drought tolerance. In conclusion, we show a single inoculation of Brachypodium with a PGB affected the whole growth cycle of the plant, accelerating its growth rates, shortening its vegetative period, and alleviating drought stress effects. These effects are relevant to grasses and cereal crops.  相似文献

The effect of water stress and subsequent rehydration on 1-aminocyclopropane-1-carboxylic acid (ACC) content, ACC synthase activity, ethylene production, and leaf abscission was studied in Cleopatra mandarin (Citrus reshni Hort. ex Tan.) seedlings. Leaf abscission occurred when drought-stressed plants were allowed to rehydrate, whereas no abscission was observed in plants under water stress conditions. In roots of water-stressed plants, a high ACC accumulation and an increase in ACC synthase activity were observed. Neither increase in ACC content nor significant ethylene production were detected in leaves of water-stressed plants. After rehydration, a sharp rise in ACC content and ethylene production was observed in leaves of water-stressed plants. Content of ACC in xylem fluid was 10-fold higher in plants rehydrated for 2 h after water stress than in nonstressed plants. Leaf abscission induced by rehydration after drought stress was inhibited when roots or shoots were treated before water stress with aminooxyacetic acid (AOA, inhibitor of ACC synthase) or cobalt ion (inhibitor of ethylene-forming enzyme), respectively. However, AOA treatments to shoots did not suppress leaf abscission. The data indicate that water stress promotes ACC synthesis in roots of Cleopatra mandarin seedlings. Rehydration of plants results in ACC transport to the shoots, where it is oxidized to ethylene. Subsequently, this ethylene induces leaf abscission.  相似文献

One of the proposed mechanisms through which plant growth-promoting rhizobacteria (PGPR) enhance plant growth is the production of plant growth regulators, especially cytokinin. However, little information is available regarding cytokinin-producing PGPR inoculation on growth and water stress consistence of forest container seedlings under drought condition. This study determined the effects of Bacillus subtilis on hormone concentration, drought resistance, and plant growth under water-stressed conditions. Although no significant difference was observed under well-watered conditions, leaves of inoculated Platycladus orientalis (oriental thuja) seedlings under drought stress had higher relative water content and leaf water potential compared with those of noninoculated ones. Regardless of water supply levels, the root exudates, namely sugars, amino acids and organic acids, significantly increased because of B. subtilis inoculation. Water stress reduced shoot cytokinins by 39.14 %. However, inoculation decreased this deficit to only 10.22 %. The elevated levels of cytokinins in P. orientalis shoot were associated with higher concentration of abscisic acid (ABA). Stomatal conductance was significantly increased by B. subtilis inoculation in well-watered seedlings. However, the promoting effect of cytokinins on stomatal conductance was hampered, possibly by the combined action of elevated cytokinins and ABA. B. subtilis inoculation increased the shoot dry weight of well-watered and drought seedlings by 34.85 and 19.23 %, as well as the root by 15.445 and 13.99 %, respectively. Consequently, the root/shoot ratio significantly decreased, indicative of the greater benefits of PGPR on shoot growth than root. Thus, inoculation of cytokinin-producing PGPR in container seedlings can alleviate the drought stress and interfere with the suppression of shoot growth, showing a real potential to perform as a drought stress inhibitor in arid environments.  相似文献

Ethylene inhibits nodulation in various legumes. In order to investigate strategies employed by Rhizobium to regulate nodulation, the 1-aminocyclopropane-1-carboxylate (ACC) deaminase gene was isolated and characterized from one of the ACC deaminase-producing rhizobia, Rhizobium leguminosarum bv. viciae 128C53K. ACC deaminase degrades ACC, the immediate precursor of ethylene in higher plants. Through the action of this enzyme, ACC deaminase-containing bacteria can reduce ethylene biosynthesis in plants. Insertion mutants with mutations in the rhizobial ACC deaminase gene (acdS) and its regulatory gene, a leucine-responsive regulatory protein-like gene (lrpL), were constructed and tested to determine their abilities to nodulate Pisum sativum L. cv. Sparkle (pea). Both mutants, neither of which synthesized ACC deaminase, showed decreased nodulation efficiency compared to that of the parental strain. Our results suggest that ACC deaminase in R. leguminosarum bv. viciae 128C53K enhances the nodulation of P. sativum L. cv. Sparkle, likely by modulating ethylene levels in the plant roots during the early stages of nodule development. ACC deaminase might be the second described strategy utilized by Rhizobium to promote nodulation by adjusting ethylene levels in legumes.  相似文献

Drought stress substantially impedes crop productivity throughout the world. Microbial based approaches have been considered a potential possibility and are under study. Based on our prior screening examination, two distinct and novel biofilm-forming PGPR strains namely Bacillus subtilis-FAB1 and Pseudomonas azotoformans-FAP3 are encompassed in this research. Bacterial biofilm development on glass surface, microtiter plate and seedling roots were assessed and characterized quantitatively and qualitatively by light and scanning electron microscopy. Above two isolates were further evaluated for their consistent performance by inoculating on wheat plants in a pot-soil system under water stresses. Bacterial moderate tolerance to ten-day drought was recorded on the application of individual strains with wheat plants; however, the FAB1 + FAP3 consortium expressively improved wheat survival during drought. The strains FAB1 and FAP3 displayed distinct and multifunctional plant growth stimulating attributes as well as effective roots and rhizosphere colonization in combination which could provide sustained wheat growth during drought. FAB1 and FAP3-induced alterations cooperatively conferred improved plant drought tolerance by controlling physiological traits (gs, Ci, E, iWUE and PN), stress indicators (SOD, CAT, GR, proline and MDA content) and also maintained physico-chemical attributes and hydrolytic enzymes including DHA, urease, ALP, protease, ACP and β glucosidase in the soil. Our findings could support future efforts to enhance plant drought tolerance by engineering the rhizobacterial biofilms and associated attributes which requires in-depth exploration and exploiting potential native strains for local agricultural application.  相似文献

Physiological parameters of mycorrhizal symbiosis by Helianthemum almeriense and Terfezia claveryi in orchards were characterized under water deficit conditions. Our orchard included 40 mycorrhizal and 40 nonmycorrhizal plants. Only mycorrhizal plants survived at the beginning of the experimental period, indicating dependency on fungal symbionts in roots for survival. Drought stress significantly affected the mycorrhizal colonization percentage which was 70% in nonirrigated mycorrhizal and 48% in irrigated mycorrhizal plants. No significant differences in plant growth were observed between nonirrigated and irrigated mycorrhizal plants before and after drought stress. Stomatal conductance was more sensitive to water stress than shoot water potential. It decreased more than two-fold under drought-stress compared to control mycorrhizal plants under irrigation/light saturating conditions, indicating important stomatal closure with water deficit. Plants’ water use efficiency improved with drought with stomatal conductance values below 0.3 mol?m?2?s?1. The ability to maintain open stomata and photosynthesis under drought increased carbon supply for growth, and ascocarp fruiting which requires current photosynthates. Basically, H. almeriense shows a conservative water use strategy based mainly on avoiding drought stress by reducing stomatal conductance as soil water potential decreases and atmospheric conditions dry. The results show that mycorrhizal H. almeriense plants maintain good physiological parameters with low soil matric potentials, thus making them an alternative agricultural crop in arid/semi-arid areas.  相似文献

One of the major mechanisms utilized by plant growth-promoting rhizobacteria (PGPR) to facilitate plant growth and development is the lowering of ethylene levels by deamination of 1-aminocyclopropane-1-carboxylic acid (ACC) the immediate precursor of ethylene in plants. The enzyme catalysing this reaction, ACC deaminase, hydrolyses ACC to α -ketobutyrate and ammonia. Several bacterial strains that can utilize ACC as a sole source of nitrogen have been isolated from rhizosphere soil samples. All of these strains are considered to be PGPR based on the ability to promote canola seedling root elongation under gnotobiotic conditions. The treatment of plant seeds or roots with these bacteria reduces the amount of ACC in plants, thereby lowering the concentration of ethylene. Here, a rapid procedure for the isolation of ACC deaminase-containing bacteria, a root elongation assay for evaluating the effects of selected bacteria on root growth, and a method of assessing bacterial ACC deaminase activity are described in detail. This should allow researchers to readily isolate new PGPR strains adapted to specific environments.  相似文献

Increased concentrations of atmospheric carbon dioxide (CO2) and drought stress have greatly influenced plant growth, the status of nitrogen (N) and phosphorus (P), and N:P ratios. We identified the plant biomass, N and P distributional patterns, and N:P stoichiometry of a grass species on the Loess Plateau in China under elevated CO2 concentration and drought stress conditions. Bothriochloa ischaemum, a C4 perennial herbaceous grass, was grown in pots at CO2 concentrations of 400 (ambient) and 800 (elevated) μmol mol?1 and at 60 ± 5 and 40 ± 5 % of field capacity. The elevated CO2 concentration significantly increased plant total biomass, N concentration, N and P content, allocation of biomass to roots, and allocation of N to shoots, and increased the N:P ratios of whole plants and the shoots, especially under well-watered conditions. Drought stress significantly decreased plant biomass and plant N and P content, especially under elevated CO2. Drought stress decreased the N:P ratios, but was only significant in the roots under ambient CO2. Drought stress may attenuate the stimulation of plant growth and N and P acquisition by CO2 enrichment, and projected elevated CO2 concentrations may partially offset the negative effects of increased drought by increasing the assimilation of N and P.  相似文献

The production of transgenic broccoli (Brassica oleracea) with increased shelf-life using an Agrobacterium rhizogenes-mediated co-transformation protocol is reported. An Agrobacterium rhizogenes Ri vector, pRi1855:GFP was constructed to allow expression of the green fluorescent protein to identify insertion of Ri TL-DNA into plant cells. The Brassica oleracea ACC synthase 1 and ACC oxidase 1 and 2 cDNAs in sense and antisense orientations were co-transformed into GDDH33, a doubled haploid calabrese-broccoli cultivar. Transformation efficiency was 3.26%, producing 150 transgenic root lines, of which 18 were regenerated into mature plants. The floral buds from T0 broccoli heads were assayed for post-harvest production of ethylene and chlorophyll levels. Buds from T0 lines transformed with ACC oxidase 1 and 2 constructs produced significantly less post-harvest ethylene at 20 °C than the untransformed plants and chlorophyll loss was significantly reduced over a 96 h post-harvest period. The T0 plants transformed with sense and antisense ACC synthase 1 had a significantly reduced 24 h post-harvest ethylene peak and delayed chlorophyll loss. A positive correlation between post-harvest bud ethylene production and chlorophyll loss was described by a regression. This demonstrates that the shelf-life of a very perishable vegetable may be increased up to 2 days at 20 °C by reducing post-harvest ethylene production.  相似文献

The ability for rhizobacteria and fungus to act as bioprotectants via induced systemic resistance has been demonstrated, and considerable progress has been made in elucidating the mechanisms of plant–biocontrol agent–pathogen interactions. Pseudomonas aeruginosa PJHU15, Trichoderma harzianum TNHU27, and Bacillus subtilis BHHU100 from rhizospheric soils were used singly and in consortium and assessed on the basis of their ability to provide disease protection by relating changes in ascorbic acid and hydrogen peroxide (H2O2) production, lipid peroxidation, and antioxidant enzymes in pea under the challenge of Sclerotinia sclerotiorum. Increased production of H2O2 24 h after pathogen challenge was observed and was 254.4 and 231.7–287.7 % higher in the triple consortium and singly treated plants, respectively, when compared to untreated challenged control plants. A similar increase in ascorbic acid content and ascorbate peroxidase activity was observed 24 and 48 h after pathogen challenge, respectively, whereas increased activities of catalase, guaiacol peroxidase, and glutathione peroxidase were observed 72 h after pathogen challenge. Similarly, lipid peroxidation reached a maximum at 72 h of pathogen challenge and was 61.2 and 11.2–32.1 % less in the triple consortium and singly treated plants, respectively, when compared to untreated challenged control plants. These findings suggest that the interaction of microorganisms in the rhizosphere enhanced protection from oxidative stress generated by pathogen attack through induction of antioxidant enzymes and improved reactive oxygen species management.  相似文献

The enzyme ACC deaminase encoded by acdS gene is known to play a significant role in sustaining plant growth and development under stress conditions by reducing stress induced ethylene production inside the plant. In the present investigation, Mesorhizobium ciceri isolates from chickpea plants growing under drought conditions were screened for the presence of acdS gene. Full length acdS gene from one of the isolates was successfully amplified. This is the first report showing presence of acdS gene in Mesorhizobium ciceri. Gene sequence analysis revealed that there were deletions at two positions in this gene as compared to M. loti acdS gene.  相似文献

The influence of arbuscular mycorrhizal fungi (AMF), Funneliformis mosseae and Rhizophagus intraradices, on plant growth, leaf water status, chlorophyll concentration, photosynthesis, nutrient concentration, and fractal dimension (FD) characteristics of black locust (Robinia pseudoacacia L.) seedlings was studied in pot culture under well-watered, moderate drought stress, and severe drought stress treatments. Mycorrhizal seedlings had higher dry biomass, leaf relative water content (RWC), and water use efficiency (WUE) compared with non-mycorrhizal seedlings. Under all treatments, AMF colonization notably enhanced net photosynthetic rate, stomatal conductance, and transpiration rate, but decreased intercellular CO2 concentration. Leaf chlorophyll a and total chlorophyll concentrations were higher in AM seedlings than those in non-AM seedlings although there was no significant difference between AMF species. AMF colonization improved leaf C, N, and P concentrations, but decreased C:N, C:P, and N:P ratios. Mycorrhizal seedlings had a larger FD value than non-mycorrhizal seedlings. The FD value was positively and significantly correlated to the plant growth parameters, photosynthesis, RWC, WUE, and nutrient concentration but negatively correlated to leaf/stem ratio, C:N and C:P ratios, and intercellular CO2 concentration. We conclude that AMF lead to an improvement of growth performance of black locust seedlings under all growth conditions, including drought stress via improving leaf water status, chlorophyll concentration, photosynthesis, and nutrient uptake. Moreover, FD technology proved to be a powerful non-destructive method to characterize the effect of AMF on the physiology of host plants during drought stress.  相似文献
