The protonation constants of 1,3,5-trideoxy-1,3,5-tris(2-hydroxyl-benzyl)amino-cis-inositol (thci) in I = 1 M (NaClO4) were determined to be: pKa1 5.96 ± 0.03, pKa2 7.21 ± 0.01, pKa3 8.32 ± 0.07, pKa4 8.95 ± 0.06. The solvent extraction studies were consistent with the formation of the Ln(thci)3+ and complexes. The log of the stability constants (log β1 and log β2) at 25 °C in 1 M (NaClO4) at pH 4 for formation of these complexes are reported. Laser luminescence measurements of the 7F0-5D0 transition of Eu(III) complexed by thci indicated two species. The shifts in the peaks relative to that of Eu(aq)3+ were comparable to the values reported for other complexes of Eu(III) with organic ligands, but the intensities were greater. Luminescence lifetime measurements of the fluorescence spectra indicated that the complex has 5 inner sphere water molecules bound to the Eu(III) cation at pH 6.71-8.52. This was consistent with bidentate chelation of Eu(III) with each thci molecule. gaussian view energy calculations indicated bonding for M(III) to the amino and hydroxyl groups of the cyclohexanetriol and (2-hydroxybenzyl)amino moieties in the Ln(thci)3+ complex.  相似文献

Cystathionine β-synthase (CBS) catalyzes the pyridoxal-5′-phosphate-dependent condensation of l-serine and l-homocysteine to form l-cystathionine in the first step of the transsulfuration pathway. Although effective expression systems for recombinant human CBS (hCBS) have been developed, they require multiple chromatographic steps as well as proteolytic cleavage to remove the fusion partner. Therefore, a series of five expression constructs, each incorporating a 6-His tag, were developed to enable the efficient purification of hCBS via immobilized metal ion affinity chromatography. Two of the constructs express hCBS in fusion with a protein partner, while the others bear only the affinity tag. The addition of an amino-terminal, 6-His tag, in the absence of a protein fusion partner and in the absence or presence of a protease-cleavable linker, was found to be sufficient for the purification of soluble hCBS and resulted in enzyme with 86–91% heme saturation and with activity similar to that reported for other hCBS expression constructs. The continuous assay for l-Cth production, employing cystathionine β-lyase and l-lactate dehydrogenase as coupling enzymes, was employed here for the first time to determine the steady-state kinetic parameters of hCBS, via global analysis, and revealed previously unreported substrate inhibition by l-Hcys (Kil-Hcys = 2.1 ± 0.2 mM). The kinetic parameters for the hCBS-catalyzed hydrolysis of l-Cth to l-Ser and l-Hcys were also determined and the kcat/Kml-Cth of this reaction is only 2-fold lower than the kcat/Kml-SER of the physiological, condensation reaction.  相似文献

A complex trisaccharide β-d-GalpNAcA-(1 → 4)-β-d-GlcpNAc-(1 → 4)-d-ManpNAc (3) was prepared in a good yield (35%) in a transglycosylation reaction catalyzed by β-N-acetylhexosaminidase from Talaromyces flavus using p-nitrophenyl 2-acetamido-2-deoxy-β-d-galacto-hexodialdo-1,5-pyranoside (1) as a donor followed by the in situ oxidation of the aldehyde functionality by NaClO2. The disaccharide β-d-GlcpNAc-(1 → 4)-d-ManpNAc (2) was used as galactosyl acceptor. A disaccharide β-d-GalpNAcA-(1 → 4)-d-GlcpNAc (4; 39%) originated as a by-product in the reaction. Oligosaccharides comprising a carboxy moiety at C-6 are shown to be very efficient ligands to natural killer cell activation receptors, particularly to human receptor CD69. Thus, oxidized trisaccharide 3 is the best-known oligosaccharidic ligand to this receptor, with IC50 = 2.5 × 10−9 M. The presented method of introducing a β-d-GalpNAcA moiety into carbohydrate structures is versatile and can be applied in the synthesis of other complex oligosaccharides.  相似文献

d-Hydantoinase was covalently immobilized onto polystyrene anion exchange resin via glutaraldehyde. Immobilization conditions were optimized: the carrier as D-92 type polystyrene anion exchange resin, temperature as 25 °C, immobilization time as 12 h, and initial concentration of protein as 6 mg/ml. Under the optimized reaction conditions the activity of the free and immobilized d-hydantoinase was determined. The free and immobilized d-hydantoinase samples were characterized with their kinetic parameters, thermal, and storage stability. The Km and Vmax values were 14.985 mM and 0.6 mM/min for the free, and 27.030 mM and 1.187 mM/min for the immobilized, respectively. Operational stability of the immobilized d-hydantoinase was also detected in a circulating packed-bed reactor. The half-time of the immobilized d-hydantoinase was 11 days. Nearly 90% of activity of the immobilized d-hydantoinase was reserved for 100 days stored at 4 °C. The free and immobilized d-hydantoinases were also characterized under microwave irradiation. Results shown that the reactions catalyzed by both free and immobilized d-hydantoinase were accelerated under microwave irradiation. The half-time of the immobilized d-hydantoinase reduced to 16 min under microwave irradiation.  相似文献

Structural investigation of the polysaccharide was carried out using total acid hydrolysis, methylation analysis, periodate oxidation study, and NMR studies (1H, 13C, DQF-COSY, TOCSY, NOESY, ROESY, HMQC, and HMBC). A water-insoluble (1 → 3)-β-d-glucan (PCSG) isolated from the fresh sclerotium of Poria cocos was carboxymethylated to afford a water-soluble derivative coded as C-PCSG. The carboxymethylated (1 → 3)-β-d-glucan was fractionated to obtain eight fractions according to the nonsolvent addition method. The weight-average molecular mass (Mw), radius of gyration and intrinsic viscosity ([η]) of the fractions were determined by size-exclusion chromatography combined with laser light scattering (SEC-LLS) and viscometry in 0.2 M NaCl aqueous solution at 25 °C. The dependences of [η] and on Mw for C-PCSG were found to be , and (nm), respectively. Analysis of Mw and [η] in terms of the known theories for wormlike chain model yielded 633 nm−1 for molar mass per unit contour length (ML), 5.5 nm for persistence length (q), and 20.2 for characteristic ratio (C). These results indicated that C-PCSG exists as a relatively extended flexible chain in 0.2 M NaCl aqueous solution. Therefore, the introduction of the carboxymethyl groups into the β-glucan improved significantly the water solubility and enhanced the stiffness of the chains.  相似文献

The protonation constants of 1,3,5-triamino-2,4,6-trihydroxycyclohexane (taci), at 25 °C in I = 1.00 M (NaClO4) were determined to be: pKa1, 5.57 (0.08); pKa2, 7.45 (0.02); pKa3, 9.05 (0.04). The log of the stability constants, log β302, at 25°C in I = 1.00 M (NaClO4) for formation of were measured by potentiometry to be: Nd(III), 25.33 (0.09); Eu(III), 26.42 (0.06); Tm(III), 30.07 (0.10); Lu(III), 33.68 (0.07) ; Y(III), 28.59 (0.07). 1H NMR spectra were consistent with formation of a single complex from pcH 6 to 10. Laser fluorescence measurements of the 7Fo-5Do transition of Eu(III) complexed by taci indicated a single complexed species. The shift in this peak relative to that of Eu3+(aq) was significantly greater than the values reported for the complexes of other organic ligands with Eu(III). Luminescence lifetime measurements indicated two water molecules bound to each of the Eu(III) cations in the taci complex.  相似文献

Giardia lamblia arginine deiminase (GlAD), the topic of this paper, belongs to the hydrolase branch of the guanidine-modifying enzyme superfamily, whose members employ Cys-mediated nucleophilic catalysis to promote deimination of l-arginine and its naturally occurring derivatives. G. lamblia is the causative agent in the human disease giardiasis. The results of RNAi/antisense RNA gene-silencing studies reported herein indicate that GlAD is essential for G. lamblia trophozoite survival and thus, a potential target for the development of therapeutic agents for the treatment of giardiasis. The homodimeric recombinant protein was prepared in Escherichia coli for in-depth biochemical characterization. The 2-domain GlAD monomer consists of a N-terminal domain that shares an active site structure (depicted by an in silico model) and kinetic properties (determined by steady-state and transient state kinetic analysis) with its bacterial AD counterparts, and a C-terminal domain of unknown fold and function. GlAD was found to be active over a wide pH range and to accept l-arginine, l-arginine ethyl ester, Nα-benzoyl-l-arginine, and Nω-amino-l-arginine as substrates but not agmatine, l-homoarginine, Nα-benzoyl-l-arginine ethyl ester or a variety of arginine-containing peptides. The intermediacy of a Cys424–alkylthiouronium ion covalent enzyme adduct was demonstrated and the rate constants for formation (k1 = 80 s−1) and hydrolysis (k2 = 35 s−1) of the intermediate were determined. The comparatively lower value of the steady-state rate constant (kcat = 2.6 s−1), suggests that a step following citrulline formation is rate-limiting. Inhibition of GlAD using Cys directed agents was briefly explored. S-Nitroso-l-homocysteine was shown to be an active site directed, irreversible inhibitor whereas Nω-cyano-l-arginine did not inhibit GlAD but instead proved to be an active site directed, irreversible inhibitor of the Bacillus cereus AD.  相似文献

A novel polysaccharide designated EPS-1A with an average molecular weight around 40 kDa was fractionated and purified by anion-exchange and gel-filtration chromatography from the crude exopolysaccharide (EPS) isolated from fermentation broth of Cs-HK1, a Tolypocladium sp. fungus isolated from wild Cordyceps sinensis. The structural characteristics of EPS-1A were determined with various methods (e.g. GC, GC–MS, FT-IR, 1H NMR and 13C NMR) and through acid hydrolysis, methylation, periodate-oxidation and Smith degradation. The results suggested that EPS-1A was composed of glucose, mannose and galactose at 15.2:3.6:1.0 M ratio. EPS-1A was a slightly branched polysaccharide and its backbone was composed of (1 → 6)-α-d-glucose residues (77%) and (1 → 6)-α-d-mannose residues (23%). Branching occurred at O-3 position of (1 → 6)-α-d-mannose residues of the backbone with (1 → 6)-α-d-mannose residues and (1 → 6)-α-d-glucose residues, and terminated with β-d-galactose residues.  相似文献

An aerobic bacterium was isolated from activated sludge in a medium containing l-glutamate-N,N-diacetate (l-GLDA) as sole carbon and energy source. The isolate was identified as a Rhizobium radiobacter species. Besides l-GLDA, the strain utilized nitrilotriacetate (NTA) and proposed intermediates in l-GLDA metabolism such as glyoxylate and l-glutamate. l-GLDA-grown cells oxidized l-GLDA, l-glutamate but not iminodiacetate (IDA), and trans-ketoglutaconate, indicating removal of a carboxymethyl group as an initial degradation reaction. The removal of the first carboxymethyl group of l-GLDA is catalyzed by an NADH-dependent mono-oxygenase. The oxidative deamination of l-glutamate by a dehydrogenase resulting in the formation of oxoglutarate was also detected in cell-free extracts of R. radiobacter sp. A pathway for the metabolism of l-GLDA R. radiobacter sp. is proposed: First, l-GLDA leads to l-glutamate-N-monoacetate (l-GLMA) which in turn leads to l-glutamate. Then, l-glutamate leads to oxoglutarate, an intermediate of the TCA cycle.  相似文献

A water-soluble polysaccharide isolated from Dalbergia sissoo Roxb. leaves was purified and major homogeneous fraction obtained by GPC. Complete hydrolysis of the polysaccharide followed by paper chromatography and GLC analysis indicated the presence of l-rhamnose, d-glucuronic acid, d-galactose and d-glucose in molar ratio of 1:1:2:2.33, respectively. Partial hydrolysis of the polysaccharide furnished one tri-[I], one hepta-[II] and one nona-[III] saccharides. Hydrolysis of the oligosaccharide I, II and III followed by GLC analysis furnished d-glucose and l-rhamnose (2:1); l-rhamnose, d-galactose and d-glucuronic acid (1:3:3); and l-rhamnose, d-galactose and d-glucose (1:3:5), respectively. Methylation analysis and periodate oxidation of the oligosaccharide I indicated the presence of two non reducing glucose units linked to rhamnose by 1→2 and 1→4 linkages, respectively. Oligosaccharide II is a branched molecule with a main chain consisting of 1,3-linked β-d-galactopyranosyl (2 mol), 1,3,4 linked α-l-rhamnopyranosyl (1 mol) and 1,4,6 linked β-d-galactopyranosyl unit (1 mol) and non reducing β-d-glucuronic acid at the end along with side chains of β-d-glucouronopyranosyl units (2 mol). Oligosaccharide III is also a branched molecule with a main chain consisting of 1,3,4 linked α-l-rhamnopyranosyl (1 mol), 1,2,4 linked β-d-glucopyranosyl (1 mol), 1,3 and 1,4 linked β-d-galactopyranosyl (2 and 1 mol, respectively) having β-d-glucopyranosyl as a non reducing end.  相似文献

The kinetically controlled synthesis of N-benzyloxycarbonyl (Z)-dipeptides was investigated by the use of free amino acids as nucleophiles and a cysteine protease papain as catalyst. The coupling efficiency was significantly improved by the combined use of the carbamoylmethyl (Cam) ester of a Z-amino acid as acyl donor and frozen aqueous solution (ice, −16 or −24 °C) as reaction medium. The yield of peptide synthesis became high when both P1- and P1-positions were occupied by small non-polar amino acids (Z-Gly-Gly-OH, 76%; Z-Gly-Ala-OH, 75%; Z-Ala-Ala-OH, 72%). Similar results were observed by the use of ficin as catalyst instead of papain. Furthermore, this strategy was applied to the papain-catalyzed incorporation of a d-configured amino acid such as d-alanine into the resulting peptides. Although the coupling in aqueous solution (30 °C) afforded the desired Z-dipeptides in low yields, the freezing of reaction medium reduced significantly unfavorable hydrolysis of the acyl donors, resulting in improvement of the coupling efficiency (Z-Gly-d-Ala-OH, 80%; Z-Ala-d-Ala-OH, 45%; Z-d-Ala-Ala-OH, 22%).  相似文献

d-Amino acid oxidase is a FAD-dependent enzyme that catalyses the conversion of the d-enantiomer of amino acids into the corresponding α-keto acid. Substrate specificity of the enzyme from the yeast Rhodotorula gracilis was investigated towards aromatic amino acids, and particularly synthetic α-amino acids.A significant improvement of the activity (Vmax,app) and of the specificity constant (the Vmax,app/Km,app ratio) on a number of the substrates tested was obtained using a single-point mutant enzyme designed by a rational approach. With R. gracilis d-amino acid oxidase the complete resolution of d,l-homo-phenylalanine was obtained with the aim to produce the corresponding pure l-isomer and to use the corresponding α-keto acid as a precursor of the amino acid in the l-form.  相似文献

1. Retroperitoneal white adipose tissue (RpWAT) antioxidative defense was investigated in untreated, l-arginine-treated and Nω-nitro-l-arginine methyl ester (l-NAME)-treated rats kept at 4±1 °C (1, 3, 7, 12, 21 and 45 days) and compared to control rats at 22±1 °C.
2. Cold-acclimation-induced RpWAT weight decrease was accompanied by a decline in glutathione level and increased activity of manganese superoxide dismutase (MnSOD), glutathione S-transferase (GST), catalase, glutathione peroxidase and glutathione reductase at different time-points.
3. l-arginine accelerated RpWAT weight decrease, the increase in MnSOD and GST activities and the prolonged increase of catalase, MnSOD and GST activities. l-NAME delayed cold-induced catalase activity increase and tissue weight decrease. Prolonged l-NAME-treatment had a similar effect on RpWAT as l-arginine.
4. Results suggest the involvement of l-arginine/NO pathway in RpWAT oxidative metabolic augmentation induced by cold-acclimation.

The dapE-encoded N-succinyl-l,l-diaminopimelic acid desuccinylase (DapE) is a critical bacterial enzyme for the construction of the bacterial cell wall. A screen biased toward compounds containing zinc-binding groups (ZBG’s) including thiols, carboxylic acids, boronic acids, phosphonates and hydroxamates has delivered a number of micromolar inhibitors of DapE from Haemophilus influenzae, including the low micromolar inhibitor l-captopril (IC50 = 3.3 μM, Ki = 1.8 μM). In vitro antimicrobial activity was demonstrated for l-captopril against Escherichia coli.  相似文献

Twenty pregnane glycosides, tuberoside A1–L5, were isolated from the diethyl ether-soluble fraction of the MeOH extract from the aerial parts of Asclepias tuberosa (Asclepiadaceae). The pregnane glycosides were composed of 8,12;8,20-diepoxy-8,14-secopregnane as aglycon, and d-cymarose, d-oleandrose, d-digitoxose and/or d-glucose as the component sugars. The gene encoding α-methylserine aldolase was isolated from Bosea sp. AJ110407. Sequence analysis revealed that the predicted amino acid sequence encoded by the 1320-bp open reading frame was 65.0% similar to the corresponding sequence of the enzyme isolated from Ralstonia sp. AJ110405. The gene was expressed in Escherichia coli, and the recombinant enzyme was purified. Gel filtration revealed the molecular mass of the purified enzyme to be approximately 78 kDa, suggesting that the enzyme is a homodimer. The enzyme exhibited a specific peak at 429 nm in the spectrum and contained 1 mol pyridoxal 5′-phosphate per mole of the subunit. The Vmax value was 1.40 μmol min−1 mg−1, and the Km value was 1.5 mM for the reaction wherein formaldehyde was released from α-methyl-l-serine. This enzyme could also catalyze the reverse reaction, i.e., the synthesis of α-methyl-l-serine from l-alanine and formaldehyde. This activity was inhibited in the excess of formaldehyde; however, α-methyl-l-serine was efficiently produced from l-alanine in the presence of formaldehyde. This method was also applicable for producing α-ethyl-l-serine from l-2-aminobutyric acid.  相似文献

Insect β-N-acetyl-d-hexosaminidases are of particular interest due to their multiple physiological roles in many life processes. Chitinolytic β-N-acetyl-d-hexosaminidases, which function only in chitin degradation in insects, have long been regarded as species-specific target potentials in developing environmental friendly pesticides. Here the chitinolytic β-N-acetyl-d-hexosaminidase from the insect Ostrinia furnacalis was cloned and expressed in the yeast strain, Pichia pastoris, to meet the demands of biochemical studies and drug development. Enzymatic assay as well as Western blot confirmed that the high-level expression could be achieved after the induction of methanol for 120 h. Through the sequential combination of ammonium sulfate precipitation, metal chelating chromatography as well as anion exchange chromatography, 7.7 mg of the recombinant OfHex1 with high purity was obtained from 1 liter of culture supernatant. The recombinant OfHex1, characterized as a homodimer with molecular weight of 130 kDa, exhibited the same enzymatic activities as its native form, which could efficiently degrade the chitooligosaccharide substrate (GlcNAc)2 and release 4-methylumbelliferone (4MU) from substrates, 4MU-β-GlcNAc and 4MU-β-GalNAc. This work provides a low-costing and high-efficient purification procedure for the preparation of insect β-N-acetyl-d-hexosaminidases.  相似文献

d-Hydantoinase and d-carbamoylase genes from Agrobacterium radiobacter TH572 were cloned by polymerase chain reaction (PCR). The plasmid pUCCH3 with a polycistronic structure that is controlled by the native hydantoinase promoter was constructed to co-express the two genes and transformed into Escherichia coli strain JM105. To obtain the highest level of expression of the d-carbamoylase and avoid intermediate accumulation, the d-carbamoylase gene was cloned closer to the promoter and the RBS region in the upstream of it was optimized. This resulted in high active expression of soluble d-hydantoinase and d-carbamoylase that is obtained without any inducer. Thus, by the constitutive recombinant JM105/pUCCH3, d-p-hydroxyphenylglycine (d-HPG) was obtained directly with 95.2% production yield and 96.3% conversion yield.  相似文献

The effects of d-limonene concentration, enzyme loading, and pH on ethanol production from simultaneous saccharification and fermentation (SSF) of citrus peel waste by Saccharomyces cerevisiae were studied at 37 °C. Prior to SSF, citrus peel waste underwent a steam explosion process to remove more than 90% of the initial d-limonene present in the peel waste. d-Limonene is known to inhibit yeast growth and experiments were performed where d-limonene was added back to peel to determine threshold inhibition amounts. Ethanol concentrations after 24 h were reduced in fermentations with initial d-limonene concentrations greater than or equal to 0.33% (v/v) and final (24 h) d-limonene concentrations greater than or equal to 0.14% (v/v). Ethanol production was reduced when enzyme loadings were (IU or FPU/g peel dry solids) less than 25, pectinase; 0.02, cellulase; and 13, beta-glucosidase. Ethanol production was greatest when the initial pH of the peel waste was adjusted to 6.0.  相似文献
