Separate and combined action of nitrosomethylurea (NMU) and heat shock (HS) on germination of seeds of the inbred line 3629 and derived nuclear and plastome mutant lines, n-chlorina-1 and en-chlorina-5, respectively, were studied. NMU at a concentration of 0.015%, as well as HS (40 degrees C), had no effect on the spontaneous level of chromosome aberrations in root meristem of sunflower seedlings. However, at a concentration of 0.03%, the mutagen considerably increased the frequency of chromosome rearrangements. Pretreatment with heat for 30 min enhanced the cytogenetic effect of NMU (0.03%) on line 3629 seedlings. In the nuclear mutant, the reaction of root meristematic cells did not depend on the additional heat treatment. The combined action of HS and NMU at early germination stages had no effect on the survival of line 3629 plants and plastome mutant en-chlorina-5; the survival of n-chlorina-1 plants decreased. The highest frequency of M1 plants with chlorophyllic abnormalities (green revertants, lethals, and variegated forms) was observed in n-chlorina-1 line.  相似文献

The influence of increased temperature on the intensity of fission and level of chromosome aberrations (ChA) in root meristeme cells of two lines of sunflower seedlings was studied. Within the first hour of seed germination, the temperature of 40 degrees C reduced proliferation activity of root meristeme cells line 3629 for almost the next 15 h of growth. Heat stress (HS) blocks the beginning of cell fission in the plastome mutant en-chlorina-5. However, subsequent by mitosis activity was seen to increase. The influence of HS on the ChA level is ambiguous: the effect depends on the duration of warm-up influence and the used sunflower lines. So, ChA level does not change inbred line 3629 even after 6 h HS. In mutant en-chlorina-5 the ChA level increases after 1 h HS and decreases after 3 and 6 h HS. The study of HS modification effect has shown that preliminary warm-up influence (40 degrees C for 1 h) reduces differences in reactions of cells 3629 of line on the action of different nitrosomethylurea (NMU) concentrations. Preliminary warm-up influence does not change sensitivity of en-chlorina-5 cells to NMU. The combined action of the increased temperature and NMU induce a powerful cytostatic effect.  相似文献

The influence of high temperatures on sunflower chlorophyll mutants was studied. The tolerance criteria were the level of chromosome aberrations and the mitotic index in the root apical meristem of seedlings, the level of nucleus-free cells in the epidermis of cotyledonous leaves, and the intensity of chlorophyll accumulation after the action of heat shock (HS). In addition, the frequency of plants with an altered content of pigments in M1 and M2 was analyzed. The results indicated that the plastomic mutant en-chlorina-5 is more tolerant to temperature stress as compared to other sunflower lines.  相似文献

The effects of heat shock (HS) (40°C for 1 h) on the level of malondialdehyde (MDA), the terminal product of lipid peroxidation, superoxide dismutase (SOD) activity, catalase activity, and total peroxidase activity (TPA) were studied in root meristems and chloroplasts of several sunflower (Helianthus annuusL.) lines that carried nuclear or plastome chlorophyll mutations. HS either lowered or did not affect the MDA level in the root meristem and in the chloroplasts from the first true leaf, as compared to the untreated plants. In both treatments, the root and leaf enzyme activities varied in the sunflower lines. In the root meristem, catalase was the most sensitive to HS, whereas, in the chloroplasts from HS-treated sunflower lines, HS activated either TPA or SOD.  相似文献

The biosynthesis of heme, a plant tetrapyrrole, was studied in the leaves of a chlorophyll-deficient plastome mutant of the sunflower (Helianthus annuus L, line 2-24, albina form). In the light, the content of 5-aminolevulinic acid (ALA) in white mutant leaves was, on the average, ten times less than in that of the wild-type form (line 3629). Chlorophyll content in mutant leaves comprised only 0.3% of that of control plants. The activities of Fe-chelatase and ALA dehydratase in the heme synthesis were either comparable to or even higher than those in the wild-type leaves. A normal respiration rate in white mutant leaves, the equal content of phytochrome apoproteins in plants of both types, and the lack of noticeable morphogenetic differences realized through the phytochrome system can indicate that mutant and wild-type leaves are similar in their levels of phytochrome and the cytochromes of mitochondrial respiration. Nevertheless, in the mutant, the content of heme noncovalently bound by apoproteins amounted to only one third of its content in the wild-type plants. It seems that a dramatic decrease in the capability of white leaves for chlorophyll biosynthesis and for the formation of the photosynthetic apparatus is responsible for a low demand for chloroplast cytochromes, which is the major cause of a reduced heme content in the mutant.  相似文献

A unique property of Drosophila melanogaster l(1)ts403 strain with the defect in heat shock protein system (HSP) is high frequency of losses and non-disjunction of sex chromosomes induced by heat shock (HS) (37 degrees C, 1 h). This effect was shown in only 6-14-th stages of oocytes. Anoxia was not effective in induction of these mutations. Successive action of anoxia and HS decreased loss frequency and non-disjunction in comparison with the only action of HS. These findings agree with the data in literature indicating that HSP synthesis was increased in the l(1)ts403 mutant when first anoxia and then HS were administered, in contrast to the action of HS only. The role of HSP in the recovery of HS-induced disruptions (chromosomal proteins and meiotic division apparatus) which can lead to chromosome non-disjunction and losses is discussed.  相似文献

The effect of hyberbaric oxygenation on mutagenicity of nitrosomethylurea (NMU) was examined. It was shown that in the regimes studied, hyperbaric oxygenation enhances the NMU mutagenic effect both on the nuclear and on the plastid genetic material of sunflower, but do not increase intensity of free-radical reactions and do not induce plastid mutations and chromosome aberrations per se. At high concentrations inducing chromosome aberrations (0.03%), NMU was shown to enhance the level of free-radical processes. Possible mechanisms of the increase of NMU-induced mutagenesis by hyperbaric oxygenation are discussed.  相似文献

Chlorophyll (Chl) deficiency in leaves of a plastome sunflower (Helianthus annuus L.) en:chlorina-5 mutant is due to the formation of smaller chloroplasts with a markedly reduced membrane system, as compared to the parent 3629 line. Abnormalities in the structure of the photosynthetic apparatus in the mutant can be mainly attributed to changes in the formation of photosystem I and its light-harvesting complexes. Chl deficiency in en:chlorina-5 correlated with its lower capability of synthesizing the first specific Chl precursor, 5-aminolevulinic acid (ALA) in the light. Light-independent stages of Chl biosynthesis in the mutant had the same efficiency as in leaves of the parent line. ALA formation in darkness and its conversion into protochlorophyllide did not depend on the extent of photosynthetic membrane development and photosynthetic activity.  相似文献

Genetic approaches using Arabidopsis thaliana aimed at the identification of mutations affecting events involved in auxin signalling have usually led to the isolation of auxin-resistant mutants. From a selection screen specifically developed to isolate auxin-hypersensitive mutants, one mutant line was selected for its increased sensitivity to auxin (x 2 to 3) for the root elongation response. The genetic analysis of sax1 (hypersensitive to abscisic acid and auxin) indicated that the mutant phenotype segregates as a single recessive Mendelian locus, mapping to the lower arm of chromosome 1. Sax1 seedlings grown in vitro showed a short curled primary root and small, round, dark-green cotyledons. In the greenhouse, adult sax1 plants were characterized by a dwarf phenotype, delayed development and reduced fertility. Further physiological characterization of sax1 seedlings revealed that the most striking trait was a large increase (x 40) in ABA-sensitivity of root elongation and, to a lesser extent, of ABA-induced stomatal closure; in other respects, hypocotyl elongation was resistant to gibberellins and ethylene. These alterations in hormone sensitivity in sax1 plants co-segregated with the dwarf phenotype suggesting that processes involved in cell elongation are modified. Treatment of mutant seedlings with an exogenous brassinosteroid partially rescued a wild-type size, suggesting that brassinosteroid biosynthesis might be affected in sax1 plants. Wild-type sensitivities to ABA, auxin and gibberellins were also restored in sax1 plants by exogenous application of brassinosteroid, illustrating the pivotal importance of the BR-related SAX1 gene.  相似文献

In higher plants, shoots show negative gravitropism and rootsshow positive gravitropism. To elucidate the molecular mechanismsof root and hypocotyl gravitropism, we segregated the secondmutation from the original phyB-1 mutant line which impairedboth root and hypocotyl gravitropism and characterized thisnovel mutation named rhg (for root and hyzypocotyl gravitropism).The rhg is a single recessive nuclear mutation and it is mappedon the lower part of the chromosome 1. Analyses on the gravitropicresponses of the rhg mutant indicate that root and hypocotylgravitropism are severely impaired but inflorescence stem gravitropismis not affected by the rhg mutation. In the rhg mutant seedlings,amyloplasts (statoliths for gravity-perception) were presentin the presumptive statocytes of roots and hypocotyls. Phototropismby roots and hypocotyls was not impaired in the rhg mutant.These results suggest that the RHG gene product probably actson the gravity-perception and/or the gravity-signal transductionin root and hypocotyl gravitropism. This is the first reportabout the genetic locus specifically involved in both root andhypocotyl gravitropism but not inflorescence stem gravitropism,supporting our hypothesis that the mechanisms of gravitropismare genetically different between hypocotyls and inflorescencestems. Oenothera plants homozygous for the recessive plastome mutator allele (pm) show chloroplast DNA (cpDNA) mutation frequencies that are about 1,000-fold higher than spontaneous levels. The pm-encoded gene product has been hypothesized to have a function in cpDNA replication, repair and/or mutation avoidance. Previous chemical mutagenesis experiments with the alkylating agent nitroso-methyl urea (NMU) showed a synergistic effect of NMU on the induction of mutations in the pm line, suggesting an interaction between the pm-encoded gene product and one of the repair systems that corrects alkylation damage. The goal of the experiments described here was to examine whether the pm activity extends to the repair of damage caused by non-alkylating mutagens. To this end, the intercalating mutagen, 9-aminoacridine hydrochloride (9AA) was tested for synergism with the plastome mutator. A statistical analysis of the data reported here indicates that the pm-encoded gene product is not involved in the repair of the 9AA-induced mutations. However, the recovery of chlorotic sectors in plants derived from the mutagenized seeds shows that 9AA can act as a mutagen of the chloroplast genome.Communicated by R. Hagemann  相似文献

Carrot (Daucus carota L.) is an important horticultural crop with significant health benefits, providing provitamin A carotenoids in the human diet. Carotenoids primarily serve as photoprotectants in leaves during photosynthesis where they accumulate in chloroplasts. Carotenoids can also accumulate in chromoplasts, non-chlorophyll-containing plastids, in non-photosynthetic organs such as the storage roots of carrot. Therefore, plastid development is closely associated with carotenoid accumulation. The biosynthesis of chromoplasts, or more specifically the conversion of chloroplasts to chromoplasts, has been studied in a number of carotenoid-accumulating plant species, but the presence of the plastome had not been confirmed in non-pigmented carrot storage root. In this study, the plastome was confirmed to occur in similar relative abundance (plastome–nuclear genome ratio) in yellow and orange carrot storage roots while dark orange storage roots had significantly higher plastome content than white cultivated carrots. In the leaf tissue of these same plants, the relative abundance of the plastome was similar across genotypes but was lower than the ratio of plastid to nuclear genome in the root tissue of pigmented storage rooted carrot. This study is the first quantification of the ratio of plastome to nuclear genome in storage root and confirms the presence of the plastome in both pigmented and non-pigmented carrot storage root tissue.  相似文献

以西南地区具有代表性的16种绿肥植物为受体材料,采用培养皿药膜法研究了铁核桃(Juglans sigillata)根水浸提液对受体种子发芽率及幼苗鲜重、干重的化感效应;并进一步研究了铁核桃根、叶水浸提液和胡桃醌对化感效应存在明显差异的4种绿肥植物(绿豆、红三叶、白三叶、花生)种子萌发与幼苗生长以及抗氧化酶特性的影响,以筛选适宜中国西南地区核桃园种植的绿肥植物,探讨核桃根和凋落物对绿肥作物的化感作用机制。结果表明:(1)铁核桃根水浸提液对绿豆的发芽率没有影响,但对绿豆幼苗鲜重和干重有显著抑制作用,而对其他15种绿肥的发芽率和鲜重、干重均有抑制作用。(2)胡桃醌显著抑制绿豆种子萌发,而铁核桃根或叶水浸提液对绿豆种子萌发没有影响。(3)铁核桃根或叶水浸提液以及胡桃醌对绿肥植物幼苗生长的化感效应趋势一致,但核桃根或叶水浸提液的化感效应强于胡桃醌。(4)绿豆幼苗在铁核桃根或叶水浸提液以及胡桃醌处理下,超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、过氧化物酶(POD)的活性均高于其他3种(红三叶、白三叶、花生)受体幼苗,表明绿豆清除活性氧能力高,细胞受损害程度较低,受化感作用影响最弱。研究认为,绿豆为适宜中国西南地区幼龄核桃园种植的间作绿肥植物。  相似文献

Summary Oenothera plants homozygous for a recessive plastome mutator allele (pm) showed spontaneous mutation frequencies for plastome genes that are 200-fold higher than spontaneous levels. Mutations occurred at high frequencies in plants grown in the field, in a glass-house, or as leaf tip cultures under fluorescent light, indicating that the plastome mutator activity is UV-independent. However, the chlorotic sectors became visible at an earlier stage of development when seedlings were irradiated, compared to seedlings that were not exposed to UV. These results imply that the rate of sorting-out was increased by the irradiation treatment, possibly due to a decrease in the effective number of multiplication-competent plastids, or a reduction in the extent of cytoplasmic mixing. Nitroso-methyl urea treatment of seeds had a dramatic effect on mutation frequency in both wild-type and plastome mutator samples. When the background mutation rates were low, the combination of the plastome mutator nucleus and the chemical mutagenesis treatment resulted in a synergistic effect, suggesting that the plastome mutator may involve a cpDNA repair pathway.  相似文献
