目的探讨缺血预处理(IPC)对兔脊髓缺血再灌注损伤后水通道蛋白-4(AQP-4)表达的影响。方法日本大耳白兔72只,随机分为3组:假手术组(S组)、脊髓缺血再灌注损伤组(I/R组)和缺血预处理组(IPC组)。I/R组和IPC组阻断腹主动脉30min造成脊髓缺血再灌注损伤,IPC组在损伤前短暂阻断腹主动脉5min二次实施预处理,S组暴露肾动脉下腹主动脉但不阻断。分别于再灌注损伤后4h和24h进行神经功能评分,并取L4—6脊髓缺血节段,计算脊髓组织含水量,免疫组化法测定脊髓组织中AQP-4表达水平。结果与S组比较,I/R组神经运动功能评分降低,脊髓组织含水量增加,AQP-4表达增加(P〈0.05)。与I/R组比较,IPC组神经运动功能评分增高,脊髓组织含水量降低,AQP-4表达减少(P〈0.05)。结论IPC可抑制脊髓损伤后AQP-4的表达,进而减轻脊髓水肿,保护缺血再灌注损伤的脊髓。  相似文献

为了研究RNA干扰NF-κBp65对鼠肺缺血再灌注损伤肺组织中NF-κB、TNF-α表达的影响,并探讨减轻鼠肺损伤的保护机制。本研究构建了体外靶向NF-κBp65的短发夹RNA (short hairpin RNA, shRNA)重组表达载体,并采用QPCR检测NF-κBp65的沉默结果。供试的16只SD大鼠随机分为4组:假手术组4只、缺血再灌注+生理盐水组4只,缺血再灌注+NF-κBp65 sh RNA组4只,缺血再灌注+空载组4只,假手术组无缺血再灌注损伤,开胸游离左肺门180 min,缺血再灌注+生理盐水组左肺门阻断前术前24 h给予生理盐水处理,开胸游离左肺门,左肺缺血60 min,再灌注120 min,缺血再灌注+NF-κBp65 shRNA组左肺门阻断前术前24 h滴鼻给予NF-κB shRNA腺病毒(1×1010pfu, 100μL/只),开胸游离左肺门,左肺缺血60 min,再灌注120 min,缺血再灌注+空载组左肺门阻断前术前24 h滴鼻给予空载shRNA腺病毒(1×10~(10)pfu, 100μL/只),开胸游离左肺门,左肺缺血60 min,再灌注120 min。实验结束后每组分别留取左肺组织,留取左肺上叶肺组织测定肺湿/干重比(W/D),部分肺组织光镜下观察病理变化,ELISA法测量NF-κB和TNF-α的表达含量。研究结果表明:成功构建重组NF-κBp65载体,并获得稳定转染的NF-κBp65 shRNA细胞,与转染空载体的阴性对照组(negative control, NC)比较,NF-κBp65 sh RNA能明显使NF-κBp65基因沉默(p0.05),肺组织NF-κB、TNF-α的表达量及W/D值与假手术组比较,缺血再灌注+生理盐水组和缺血再灌注+空载组有明显升高(p0.05);而与缺血再和灌注+生理盐水组和缺血再灌注+空载组比较,缺血再灌注+NF-κBp65 shRNA组明显降低(p0.05),病理学检查表明:假手术组肺组织无明显的炎症损伤;缺血再灌注+NF-κBp65 shRNA组肺炎性损伤较缺血再灌注+生理盐水组和缺血再灌注+空载组明显减轻。本研究结果初步说明,RNA干扰NF-κBp65能明显减轻早期肺移植损伤,其机制可能与抑制NF-κB和TNF-α的表达从而减轻肺组织炎症损伤有关。  相似文献

探讨缺血后处理对兔脊髓缺血再灌注微循环损伤的影响.成年新西兰大白兔24只随机分为假手术组(C组),缺血再灌注损伤组(IR组),缺血后处理组(P组).IR组和P组采用Zivin改进法制备脊髓缺血再灌注模型,P组在缺血30 min后行复灌1 min/缺血1 min相同处理3次.采用激光多普勒检测缺血前,缺血时及再灌注各时点血流量值,在再灌注24 h时取兔脊髓组织作HE染色观察病理形态学,比色法检测脊髓组织一氧化氮(Nitric oxide,NO)的含量,放免法检测内皮素-1(Endothelin-1,ET-1)及免疫组化法检测血红素氧合酶(Hemeoxygenase-1,HO-1)的表达.研究发现与缺血前基础值相比,再灌注10 min时IR组与P组血流量均有增高,在再灌注30、60、120 min,IR组血流量值有不同程度的降低;与IR组相比,P组血流量值在再灌注各时点均有不同程度的增高.与IR组相比,P组NO含量与HO-1表达均有增加,ET-1含量明显减少,NO/ET-1显著高于IR组(P<0.05或0.01),且P组脊髓病理学损伤轻于IR组.结果表明缺血后处理可减轻兔脊髓缺血再灌注微循环损伤,改善脊髓血流量,...  相似文献

目的:探讨高压氧预处理对免脊髓缺血再灌注损伤的保护机制.方法:20只雄性新西兰大白兔,随机分为2组(每组n=10):对照组为常压空气组;HBO组为高压氧预处理组.采用肾下腹主动脉阻断法造成脊髓缺血再灌注损伤,观察两组再灌注后4h、12h、24h、48h时的神经功能评分;再灌注48 h时取出腰段脊髓组织(L5-7)测定脊髓抗氧化酶活性(SOD、CAT及GSH-px)及MAD含量.结果:再灌注4h、12h、24h、48h时,HBO组神经功能学评分均明显优于对照组(P<0.05).再灌注48h时,HBO组脊髓匀浆SOD、CAT及GSH-px活性明显高于对照组(P<0.05),脊髓匀浆MDA含量明显低于对照组(P<0.05).相关性分析发现,再灌注48 h时后肢运动神经功能学评分与脊髓SOD、CAT及GSH-px活性呈正相关(r=0.82,0.65,0.54,P<0.05),与脊髓MDA含量呈负相关(r=-0.69,P<0.05).结论:高压氧预处理对脊髓缺血再灌注损伤的保护机制可能与上调内源性抗氧化酶活性和清除自由基有关.  相似文献

目的:对现有的经腹部切口建立急性肾缺血再灌注损伤动物模型进行改良,探索建立急性肾缺血再灌注损伤模型的新方法。方法:实验组大鼠16例,经背部切口进入腹膜后间隙,游离钳夹双侧肾动脉45min后开放血流,建立急性肾缺血再灌注损伤模型;伪手术组8例,不夹闭肾动脉,余步骤与实验组相同;对照组8例无处理。术后通过建模成功率、组织病理检查、血肌酐和血尿素氮及氧化应激水平对模型进行评估。结果:实验组15只成功建立急性肾缺血再灌注损伤模型。术后1天病理检查显示实验组肾组织出现广泛损伤,术后实验组肾小管坏死评分、肾MDA水平、血肌酐及血尿素氮值明显高于对照组(P<0.05)。结论:经背部切口钳夹双侧肾动脉可建立稳定的大鼠急性肾缺血再灌注损伤模型。该造模方法简便易行,成功率高,且具备手术切口小、手术时间短及并发症少的优点,建立的模型适合于急性肾损伤的研究。  相似文献

目的:建立小肠急性缺血再灌注损伤模型,确定合适的肠系膜上动脉阻断时间.方法:将70只新西兰兔随机按不同的肠系膜缺血时间(0、15、30、45、60min)分为A、B、C、D、E5组,每组14只,取8只用于恢复血供2h后留取各组兔下腔静脉血标本及肠组织,检测血清中MDA含量的变化,光镜下观察小肠组织形态学变化并对小肠黏膜损伤程度进行评分.另6只用于术后24h、 48h及72h生存率的观察.结果:A、B、C组的术后生存率均>83.3%.C、D、E组的MDA含量及肠黏膜损伤评分与A组比较,差异均有显著性(F=12.13~280.24,p<0.01).结论:肠系膜缺血30min,再灌注2h是建立兔小肠急性再灌注损伤的合适时间.  相似文献

胆红素对大鼠肺脏缺血再灌注损伤保护作用的研究   总被引:3,自引:0,他引:3  
目的探讨胆红素对实验性大鼠肺缺血再灌注损伤的保护作用并探讨其发生的机制。方法60只健康Wistar大鼠随机分为3组:手术对照(C)组,缺血再灌注(IR)组,胆红素干预(B)组。每组分别于缺血第45min、再灌注30min、60min、120min4个时点,经左房放血处死大鼠,观察肺组织病理形态变化,检测血浆丙二醛(MDA)、超氧化物歧化酶(SOD)的含量,测定肺组织干/湿重(D/W)比值,TUNEL法测定肺组织中细胞凋亡指数(AI)。结果①肺组织病理变化:缺血再灌注后IR组肺组织损伤进行性加重,毛细血管充血、肺泡间隔炎性细胞浸润、肺泡腔内炎性细胞及炎性液体渗出显著,B组肺组织充血、水肿、炎性细胞浸润较IR组减轻。②血浆MDA含量:IR组在缺血再灌注后血浆MDA含量明显增加,较C组和B组同时点均显著增高(P〈0.01),而B组的MDA含量在缺血再灌注过程中的变化无显著性意义;③血浆SOD含量:经缺血再灌注后,IR和B组血浆SOD含量较C组同时点均显著下降(P〈0.05),B组减少的程度明显小于IR组(P〈0.05);④肺组织D/W比值:经缺血和再灌注后,IR组和B组的肺组织D/W比值都呈进行性下降(再灌注60、120minvs缺血45min,P〈0.01);B组下降幅度明显小于IR组(再灌注60、120min时,B组vsIR组,P〈0.05);⑤肺组织细胞AI的变化:IR组及B组缺血再灌注后均可见肺组织细胞凋亡现象,但与IR组相比,B组相同时点的肺组织细胞凋亡数显著减少(P〈0.05);结论胆红素对于实验性大鼠肺脏缺血再灌注损伤具有一定的保护作用,其作用机制与清除氧自由基、抗氧化及抗细胞凋亡有关。  相似文献

目的:研究缺血后处理对缺血再灌注心肌保护的相关蛋白的变化。方法:将6只新西兰大白兔随机分为两组(每组3只):心肌缺血再灌注对照组(I/R组)和缺血后处理组(P组)。两组均接受左冠状动脉前降支阻断30min,开放再灌注180min。缺血后处理组,结扎LAD30min,然后灌注30s,阻断30s,重复4次,继而再灌注直至180min,分别取各组缺血区心肌进行二维凝胶电泳,利用ImageMaster2D软件分析实验结果。结果:P组和I/R组对比,有11个蛋白表达发生了显著变化,其中表达增强的有7个蛋白,表达降低的有4个蛋白。结论:这些差异表达的蛋白可能在缺血后处理对心肌缺血再灌注损伤的保护中发挥了作用。  相似文献

目的:观察大鼠心肌缺血/再灌注损伤对血清和心肌组织瘦素(Leptin)表达的影响,探讨Leptin在心肌缺血/再灌注损伤中的作用。方法:建立大鼠心肌缺血/再灌注模型,检测血清乳酸脱氢酶(LDH)和Leptin浓度,并用HE染色和免疫组织化学观察心肌组织病理学及Lepfin表达水平。结果:缺血组、再灌注组血清LDH水平显著升高(P〈0.05),表明该模型制作成功,造成心肌局部一定程度的损伤。缺血组血清Leptin含量(6.34±2.49)ng/ml显著低于对照组(7.50±2.93ng/ml,P〈0.05);再灌注后Leptin水平缓慢恢复,于再灌注2h时Leptin达到(8.32±1.74)ng/ml,恢复到损伤前水平(8.38±2.56)ng/ml,且随再灌注时间延长有升高趋势。免疫纽化显示与假手术纽心肌Leptin蛋白表达水平相比,其他四组均有显著降低(P〈0.01),按缺血45min后再灌注1h组、缺血45min后再灌注3h组、单纯缺血45min组、缺血45min后再灌注2h组依次递减。结论:Leptin在心肌缺血/再灌注损伤后早期45min血中有明显减少,心肌组织中也明显表达下降。心肌组织病理损伤与Leptin的改变可能有一定的关系。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
