The galactopoietic effect of daily injections, for five day periods, of growth hormone was examined by measuring milk yield, mammary blood flow and arteriovenous differences of glucose and amino acids on 12 occasions in four goats. In 10 periods there were marked increases (mean 18.1%) in mammary blood flow (8 statistically significant) and less-marked increases (mean 8.0%) in milk yield (6 statistically significant). On 8 of the occasions on which it was measured the maximum blood plasma growth hormone concentration was increased to more than 8 ng/ml. There were no statistically significant changes in mammary arteriovenous concentration differences of glucose or amino acids in response to growth hormone injections. It is suggested that, contrary to the usual situation in which the rate of mammary blood flow appears to be regulated by the metabolic activity of the gland, the galactopoietic response to growth hormone may be a consequence of elevated blood flow, which increases the supply to the gland of rate-limiting metabolic substrates. 相似文献

Casein degradation was measured by a pulse-chase method in explants from freshly-prepared from goat mammary tissue. An initial increase in L-[5-3H]proline radioactivity incorporated in casein during the chase period was followed in some conditions by a decrease resulting from intracellular degradation of the major casein polypeptides. Differences in the proportion of casein degraded in explants cultured with or without prolactin suggested that the process was under hormonal control. Greater degradation of casein by explants obtained in late pregnancy and early lactation suggested that the process may affect net casein production in vivo. 相似文献

1. Non-invasive methods were developed for measuring mammary blood flow in lactating goats. 2. A Doppler principle ultrasound device was equipped with an external detector measuring maximal blood velocity (Vmax) and average blood velocity (Vav) was calculated as Vmax/2. Volume flow then depended on determination of the angle of insonation and the cross-sectional area of the milk vein (the caudal superficial epigastric or subcutaneous abdominal vein). 3. Blood velocities were measured on the milk vein of either side of the animal while clamping the pudendal veins manually. Blood velocities ranged from 7-34 cm/sec. 4. The milk vein diameter was measured by means of a slide gauge which, for clearly protruding veins, gave similar results to that measured by ultrasound scanning. In protruding veins the cross-section was circular. In non-protruding veins the cross-section was elliptical and the slide gauge significantly (P less than 0.01) overestimated the cross-sectional area. The milk vein diameter of either side measured in 10 lactating goats was 8.8 +/- 1.1 mm (means +/- SD). 5. Blood flow ranged from 90-675 ml/min in a dry and a high-yielding (3.4 l milk daily) goat, respectively. The reproducibility of the blood flow measurements was 12-16%. 6. 目的:研究半胱胺对空怀奶山羊乳腺发育的影响。方法:选6只空怀奶山羊进行自身前后对照试验。对照期饲喂基础日粮,试验期饲喂基础日粮加CS。结果:与对照期相比,试验期山羊乳腺组织的DNA含量明显升高,RNA含量明显下降,故RNA/DNA值显著下降。试验期血浆GH和PRL水平分别提高61.6%和31.6%。组织学观察发现,对照期乳腺的腺泡很少发育,几乎看不到乳导管;试验期乳腺有明显导管生长和少量腺泡发育。结论:半胱胺能够在一定程度上促进空怀奶山羊乳腺发育。 相似文献

The responses of the mammary microvasculature in lactating goats (n = 8) during feed withdrawal (18-20 h) and mammary engorgement (26-28 h of milk accumulation) were compared using an indicator-dilution technique with FITC-albumin and [(14)C]sucrose as the intravascular and diffusible indicators, respectively. Feed withdrawal and mammary engorgement caused a 50-60% decrease in mammary arterial flow and in the permeability-surface area product (PS) values for sucrose. Only feed withdrawal increased the mean transit time [from 17.3 to 30.0 s, SE of the difference (SED) = 2.16, P < 0.01] of FITC-albumin, whereas only mammary engorgement reduced sucrose extraction (0.63 to 0.51, SED = 0.04, P < 0.05). Mammary engorgement also caused a substantial reduction in the sucrose-accessible extravascular space from 92 to 44 ml (SED = 15.2, P < 0.01). In a separate experiment using five goats, milking after mammary engorgement did not immediately restore arterial flow or sucrose extraction, indicating that the effect of milk accumulation was not mediated simply via increased intramammary pressure. In conclusion, feed withdrawal resulted in slower flow in the capillary bed but apparently no change in capillary recruitment, whereas mammary engorgement caused capillary derecruitment. 相似文献

Lactating rats were fed with free access to an 8% protein-restricted diet (PR); the control group was fed a 23% protein diet (C). An energy-restricted (pair-fed) group was given the same food as the animals in the control group, but the amounts of food consumed by both PF and PR were about the same. The body weight and serum albumin concentration of PR and PF dams were significantly (P < 0. 05) lower than that of the controls. The PR group had a significant increase in serum-free triiodothyronine (FT3) concentration, 24-hr mammary gland and milk radioiodine (I131) uptake (67%, 278%, and 200%, respectively) as compared with the controls. On the other hand, those animals had a significantly lower serum-free thyroxine (FT4) concentration and 2- and 24-hr thyroid I131 uptake (67%, 64%, and 74%, respectively). Protein malnutrition during lactation did not alter thyroid or liver 5'-deiodinase activity significantly. However, PF dams had a significantly lower (25%) thyroid 5'-deiodinase activity. These data suggest that protein-restricted lactating dams had an adaptive change in the thyroid function, which could be important to increase the transference of iodine or triiodothyronine through the milk to their pups and prevent sequelae of neonatal hypothyroidism. 相似文献

The objective of this experiment was to test the hypothesis that the physiological state of lactation is accompanied by both an increase in total plasma lysine flux (rate of loss and replacement of lysine) and a net reduction in flux through the plasma lysine pool after accounting for lysine secreted in the milk. Eight lactating French Alpine does were primed and infused for three hours with solutions of alpha15N L-lysine HCl in 0.9% saline through indwelling jugular vein catheters. Enrichment of circulating plasma lysine by continuous intravenous infusion of alpha15N L-lysine was used to estimate whole body lysine flux. This procedure was repeated one month after cessation of milking. Total plasma lysine flux was similar for dry and lactating does (116.6 and 123.0 mmol/d, SEM 16.6 mmol), but 54.2 mmol/d lysine was secreted as milk protein during lactation. Direct measurement of lysine absorption from the lower tract and independent measurement of lysine degradation are needed to provide a more complete portrait of caprine lysine kinetics. 相似文献

Mammary epithelial cells dissociated from lactating mouse mammary glands form confluent monolayer cultures on collagen gel substrates. For these cultures, the substrate is more significant than the presence of lactogenic hormones in the maintenance of cell differentiation, as indicated by both morphological and biochemical criteria. Only cells cultured on floating collagen gels are able to maintain their lactose pool over several days in culture, although their ability to synthesize and secrete lactose becomes impaired. These cells are cuboidal in shape. In contrast, cells cultured on attached gels, which are constrained from changing shape and whose basolateral surfaces are inaccessible, lose their differentiation with time in culture. These flattened, dedifferentiated cells respond to the same hormonal environment by showing a mild proliferative response. Therefore, the response of cells to their hormonal milieu may be correlated with their shape: the squamous cells dedifferentiate and proliferate; the cuboidal cells maintain their differentiation and do not proliferate. 相似文献

With the aim of testing the hypothesis that regrouping decreases milk production in French Alpine goats that were lactating, a study was done using two groups (n = 8, 7). During their third month after parturition, four goats from each group were exchanged (first regrouping) between pens and left for 2 weeks, then the same two subgroups of four goats were taken back to their original pen for another 2 weeks (second regrouping). In the third regrouping, the two groups were all placed in the one pen. Milk production and social behaviour were measured daily before and after each regrouping. All regroupings led to an increase in aggressive behaviours that last by 1–2 days. Mean daily milk production decreased after first (2.82 ± 0.2 kg versus 2.53 ± 0.2 kg; P < 0.05) but not after second and third regrouping. It is concluded that aggressive behaviour increases after all regroupings, whereas milk production decreases only after the first regrouping, suggesting an important capacity of adaptation to a novel and stressful managements in the French Alpine goat. The study highlights the importance of considering effects of common practices in herd managements on social behaviour and production. 相似文献

It has been generally assumed in postruminal protein or AA infusion studies that casein or AA mixtures mimicking the essential AA (EAA) composition of milk approach ‘ideal protein’ status, but the differences in postabsorptive metabolism of individual EAA may bring the assumption into question. Graded amounts of caseinate sodium were abomasally infused into lactating goats, and concentrations in the plasma of carotid, mammary uptakes and utilization efficiencies of amino acids were monitored in the present study. The results indicated that arterial concentrations of all the amino acids, except for Arg were increased with the increase of caseinate sodium infused (P < 0.05). The increments in plasma concentration differed with individual EAA. Concentrations of Ile, Leu, and Lys increased about two-folds, and that of Met, Thr, Phe, and His increased 58.33, 32.5, 29.87, and 61.66%, respectively, when data of the 99 g/day and the control treatment was compared. Mammary extraction efficiencies of Arg, Thr, Lys, Leu, and Phe increased with graded casein infusion (P < 0.05), while those of the other EAA were not significantly changed (P > 0.05). Mammary extraction efficiencies of individual non-essential amino acids (NEAA) varied greatly, and ranged from 9.18 to 33.89% in the control treatment. Except for Pro, mammary extraction efficiency of NEAA increased with graded casein infusion. Mammary uptake to milk output ratios (U/O) of sequestered AA was generally increased with the increase of casein infused, but that of Thr, His, and Ala was not insignificantly changed (P > 0.05). Compared with the data of previous researchers, the U/O of Thr recorded in the present study was much lower (<0.42). We speculate that Thr may be extracted in forms other than FAA (such as peptide bond AA) by the mammary under conditions of the present study. Consistent with the results of other authors, mammary uptake of Arg was two times more than milk output even in the control treatment, which suggests that there may be a requirement of Arg other than milk protein synthesis in the mammary. 相似文献

The experiment has been performed in 124 non-inbred dogs by means of a complex of morphometrical methods in order to study compensatory-adaptive processes in the heart after pulmonary resection various in its volume. After 58% resection, and especially after 67-75% resection of the lung mass, a stable pulmonary arterial hypertension appears; it results in development of a chronic cor pulmonale and terminates in its decompensation. Application of extrapulmonary arterio-venous anastomosis, before 67-75% of the pulmonary parenchyma is resected, decreases hypertension in the pulmonary circulation, improves adaptive changes of the heart in the newly formed conditions of hemodynamics, preventing development of its failure. 相似文献

Luteal function and conception in lactating cows and some factors influencing luteal function after first insemination 总被引:1,自引:0,他引:1
The objective of this study was to investigate the types and incidence of luteal sub-function in lactating cows after artificial insemination (AI) and their relationship with pregnancy, and to clarify the relationship between luteal function and parity, body condition score (BCS), milk yield, and dietary intake. In 19 cows, milk samples were collected daily from AI to confirmation of pregnancy. Milk progesterone concentrations were determined by EIA. Based on peak progesterone concentration and the day of onset of luteal phase, 15 of 30 progesterone profiles (50%) were normal, with progesterone concentration reaching 1.0 ng/ml within 5 days after insemination and > or =2.0 ng/ml thereafter. In addition, 6 (20%) were insufficient, (progesterone concentration remained < 2.0 ng/ml), 5 (17%) were delayed (progesterone reached 1.0 ng/ml after 5 days), 2 (7%) were both delayed and insufficient, one (3%) was short (progesterone >1.0 ng/ml for only 7 days), and one (3%) remained basal. Cows with a normal profile had a higher (P < 0.05) pregnancy rate than those with an abnormal profile (87% versus 33%, respectively). The amount of progesterone secreted in milk after first AI, as indicated by progesterone area under curve (AUC), was negatively correlated with milk yield (r = -0.83, P < 0.01), dry matter intake (r = -0.81, P < 0.05), total digestible nutrients (r = -0.82, P< 0.05), and digestible crude protein (r = -0.79, P <0.05). Cows that produced more milk and consumed more dry matter had less progesterone during the luteal phase. In conclusion, abnormal luteal function was associated with reduced pregnancy rates and high milk production and increased dietary intake during breeding were associated with reduced progesterone concentrations. 相似文献
